Common bean (Phaseolus vulgaris L.) is a staple food in Brazil with both nutritional and socioeconomic importance. As an orphan crop, it has not received as much research attention as the commodity crops. Crop losses are strongly related to virus diseases transmitted by the whitefly Bemisia tabaci, one of the most important agricultural pests in the world. The main method of managing whitefly-transmitted viruses has been the application of insecticides to reduce vector populations. Compared to chemical vector control, a more sustainable strategy for managing insect-borne viruses is the development of resistant/tolerant cultivars. RNA interference has been applied to develop plant lines resistant to the whitefly in other species, such as tomato, lettuce and tobacco. Still, no whitefly-resistant plant has been made commercially available to date. Common bean is a recalcitrant species to in vitro regeneration; therefore, stable genetic transformation of this plant has been achieved only at low frequencies (B. tabaci vATPase (Bt-vATPase) gene, with stable expression of siRNA. Northern blot analysis revealed the presence of bands of expected size for siRNA in leaf samples of the line Bt-22.5, while in the other line (11.5), the amount of siRNA produced was significantly smaller. Bioassays were conducted with both lines, but only the line Bt-22.5 was associated with significant mortality of adult insects (97% when insects were fed on detached leaves and 59% on the whole plant). The expression of the Bt-vATPase gene was 50% lower (p Aphis craccivora, the leafminer Liriomyza sp. and the whitefly parasitoid Encarsia formosa. The results presented here serve as a basis for the development of whitefly-tolerant transgenic elite common bean cultivars, with potential to contribute to the management of the whitefly and virus diseases.

Anthracnose, caused by the fungus Colletotrichum lindemuthianum (Sacc. and Magn.), is one of the most important diseases affecting the common bean (Phaseolus vulgaris L.) in Brazil. Fusarium wilt is a serious soil disease affecting common bean cultivation, especially crops under a central pivot irrigation system. Our objective was to identify bean lines that combine resistance to Fusarium wilt in the field with other desirable traits. Twenty-eight randomized block trials with three replicates were conducted in the rainy, winter, and dry seasons from 2009 to 2012 in the states of Goiás, Federal District, and Paraná. The trials were composed of six elite lines (one carioca and five black grain type) selected in a previous study as resistant to Fusarium wilt under controlled conditions and five cultivars (three carioca two black grain type). Variance analysis demonstrated variability for reaction to Fusarium wilt and anthracnose, yield, architecture, and lodging tolerance. The interaction among the lines/cultivars and environments was significant for all traits. Five black bean lines were resistant to Fusarium wilt (grades 4.0). The Carioca line (CNFC 15872) was susceptible to fusarium wilt (5.6). Thus, these lines should not be indicated as suitable cultivars. Cultivars BRS Esplendor and BRS Notável exhibited excellent yield (2144 and 2200 kg.ha-1, respectively), high stability, erect architecture (3.4 and 4.1), lodging tolerance (3.3 and 4.0), anthracnose resistance (1.0 and 1.7), and high fusarium wilt resistance (2.0 and 2.6). Diallel analysis and field tests have not been used in selecting for resistance to fusarium wilt in common beans (dry beans, Phaseolus vulgaris). Our goal was to select parents and segregating populations of dry bean (common bean) with black seed coat that are resistant to fusarium wilt and have high yield and high seed weight. Eight common bean cultivars and elite lines with some level of resistance to fusarium wilt based on previous evaluations, were used to make crosses in a complete diallel scheme. The 28 populations obtained were evaluated with two controls in trials in the F3 (winter/2012), F4 (winter/2013), and F5 (winter/2014) generations in randomized blocks, with three replications, in plots of two 4-meter rows. The trials were conducted in Santo Antônio de Goiás, GO, Brazil, in a field infested by Fusarium oxysporum f. sp. phaseoli in the winter growing season. Data were obtained on reaction to fusarium wilt, yield, and 100 seed weight. Individual and joint analyses of variance and diallel analysis were conducted on these data regarding reaction to fusarium wilt. Mean values were clustered using the Scott-Knott test. Genetic variability was detected for the three traits. There were significant differences among the general combining abilities of the parents and specific combining abilities of the populations for reaction to fusarium wilt, and the additive and non-additive effects were significant. The CNFP 15867 line is recommended for performing new crosses, because it exhibited a positive estimate of general combining ability (0.39), which indicates that it contributes to obtaining more resistant populations. The populations BRS Esplendor x BRS Expedito and BRS Expedito x CNFP 15867 were the most promising for extraction of lines, because they present high resistance to fusarium wilt (-2.6 and -2.8) and high grain yield (2212 and 2270 kg.ha-1), and acceptable seed weight (21.1 and 20.5 g/100grains). Common beans are a key source of protein and are consumed daily by most of the Brazilian population. More than 70% of what is consumed in this country is classified as carioca beans, based on seed qualities and appearance. We evaluated progenies of carioca common beans of the Brazilian agriculture research agency (Embrapa) recurrent selection program. This recurrent selection program is based on resistance to common bean bacterial blight (CBB), seed yield, and other important agronomic traits. Selection also considers the genetic representativeness of the parents and the genetic diversity among phenotypically selected progenies. Initially, 60 superior progenies were selected based on resistance to CBB and on seed type, in two locations (Santo Antônio de Goiás, GO and Ponta Grossa, PR). These progenies were evaluated in the C0S0:2 generation in field trials in six locations during the rainy growing season. A randomized block experimental design was used with two replications, in plots of two 3-m rows spaced at 0.5 m. The most promising C0S0:3 progenies for combined agronomic performance in the different environments were selected and were then evaluated by means of 24 microsatellite molecular markers for the purpose of determining the genetic representativeness of their parents and the genetic diversity among them. The phenotypic data was subjected to analysis of variance for each trait in each of the environments, and then joint analyses were performed. To estimate genetic diversity among the progenies, the Rogers-W genetic distance was used, and a dissimilarity matrix was used to construct a dendrogram of genetic distances through the UPGMA method. Among the 60 C0S0:2 progenies that were evaluated, those that stood out for resistance to CBB also stood out for seed yield and for resistance to other diseases. Various progenies were superior, exhibiting large genetic distances between each other and in relation to the parents. This indicates a possibility of direct and indirect gains from the recurrent selection program. This molecular information will help direct selection of individuals for future recombination cycles. The main goal of this work was to select resistance sources to common bean anthracnose by field phenotyping and DNA marker-assisted screening. Fifty-five common bean genotypes, including differential varieties, characterized resistance sources, elite lines, cultivars and controls, were evaluated in a field inoculation trial and screened with SCAR markers linked to resistance genes that are important in Brazil. The field trial was carried out in Santo Antônio de Goiás, GO, Brazil, during the fall/winter growing season of 2014, using artificial inoculation with a mixture of six races of Colletotrichum lindemuthianum, selected based on their high virulence and prevalence in Brazil. Amplification reactions with the SCAR markers previously identified as linked to important anthracnose resistance genes on Brazil followed standard procedures. Twenty-eight of the 58 genotypes were resistant to anthracnose (mean severity score ≤ 3.5). Ten of these 28 resistant genotypes stood out because they presented a mean anthracnose severity score of 1.0. Four of the six SCAR markers tested shown to be useful for the assisted selection of their respective target genes (SH18 and SAS13 for Co-42, SAB03 for Co-5, and SAZ20 for Co-6). Two carioca seeded elite lines were highlighted by the phenotypic and molecular screening: K10 (Co-34, Co-42, Co-5 and Co-6) and K13 (Co-4²). The phenotypic and molecular characterization of candidate resistance sources to common bean anthracnose based on their disease reaction in field inoculation trials and on the analysis with molecular markers linked to resistance genes has shown to be a useful strategy. The availability of high-density molecular markers in common bean has allowed to explore the genetic basis of important complex agronomic traits with increased resolution. Genome-Wide Association Studies (GWAS) and Regional Heritability Mapping (RHM) are two analytical approaches for the detection of genetic variants. We carried out GWAS and RHM for plant architecture, lodging and productivity across two important growing environments in Brazil in a germplasm of 188 common bean varieties using DArTseq genotyping strategies. The coefficient of determination of G · E interaction (c2 int) was equal to 17, 21 and 41%, respectively for the traits architecture, lodging, and productivity. Trait heritabilities were estimated at 0.81 (architecture), 0.79 (lodging) and 0.43 (productivity), and total genomic heritability accounted for large proportions (72% to 100%) of trait heritability. At the same probability threshold, three marker?trait associations were detected using GWAS, while RHM detected eight QTL encompassing 145 markers along five chromosomes. The proportion of genomic heritability explained by RHM was considerably higher (35.48 to 58.02) than that explained by GWAS (28.39 to 30.37). In general, RHM accounted for larger fractions of the additive genetic variance being captured by markers effects inside the defined regions. Nevertheless, a considerable proportion of the heritability is still missing (42% to 64%), probably due to LD between markers and genes and/or rare allele variants not sampled. RHM in autogamous species had the potential to identify larger-effect QTL combining allelic variants that could be effectively incorporated into whole-genome prediction models and tracked through breeding generations using marker-assisted selection. The common bean (Phaseolus vulgaris L.) is one of the most important legumes for direct human consumption (Lin et al. 2008). Anthracnose, caused by Colletotrichum lindemuthianum is the most widespread, recurrent and devastating disease of the common bean in Latin America and Africa (Pastor-Corrales and Tu 1989). The genetic mapping is carried out using segregating populations. Obtaining segregating populations for genetic mapping have been standard practice in the common bean research (Gepts et al. 1993; Blair et al. 2006). These populations have led to detailed studies and promising to greater efficiency in the genetic breeding of the crop worldwide, but have as limitation, the small size (Sanglard et al. 2013). Common bean is an economically, nutritionally, and socially important crop. It is grown in distinct regions and different seasons around the world by subsistence level farmers with low-technology input as well as by farmers that use high input technologies. One important factor that can limit the bean growing and drastically affect grain yields is the high number of destructive pathogens that attack and cause serious damage to the crop. Among them is bean rust, incited by the fungus Uromyces appendiculatus. This disease is distributed throughout the world, but it effectively causes major production problems in humid tropical and subtropical regions. In Brazil, rust causes major losses in south, southeast, and central regions of the country. Bean rust control by resistant cultivars is an easy and economical strategy to be used in association to other rust management practices. Pyramiding of different race-specific resistance genes in association with other genes conferring adult plant resistance, slow rusting, and reduced pustule size can prolong the lifespan of a common bean cultivar by creating a more durable resistance complex against the rust pathogen. This review manuscript presents an overview on bean rust and reports some breeding efforts aiming to develop rust resistant cultivars in Brazil.
