1. Phenotypic and genetic factors in motives for smoking.

Abstract

Underlying phenotypic and genetic factors involving motives for smoking were assessed using data from 1756 twins, including 390 complete pairs. Respondents were aged 50 or older and included both current and former smokers. The Motives for Smoking Questionnaire (MSQ) hypothesizes eight correlated but distinct motives for smoking, which correspond to pharmacological and nonpharmacological second-order factors. This paper concentrates on the pharmacological motives. Principal-factor analysis recovered five categories. Items constituting Sedative, Stimulative, and Addictive reasons for smoking merged into one factor, whereas Automatic smoking, also considered to have pharmacological origins, formed a separate factor. Twin analysis was used to test for genetic effects and to determine whether these factors, although combined phenotypically, were affected by distinct genetic or environmental influences. Genetic effects accounted for a significant amount of the variance in the four hypothesized pharmacological motives for smoking, as did shared environment. The separate factor for Automatic smoking in the phenotypic analysis was due to environmental effects, and shared environment was a greater influence on Automatic smoking for women than men. One common effect explained the genetic variance; no genetic influence uniquely affected any specific motive. Therefore, individual differences in the hypothesized pharmacological motives for smoking appear to originate from a common genetic pathway.

Abstract

The "Virginia 30,000" comprise 29,698 subjects from the extended kinships of 5670 twin pairs. Over 80 unique correlations between relatives can be derived from these kinships, comprised of monozygotic (MZ) and dizygotic (DZ) twins and their spouses, parents, siblings, and children. This paper describes the first application of a fairly general model for family resemblance to data from the Virginia 30,000. The model assesses the contributions of additive and dominant genetic effects in the presence of vertical cultural inheritance, phenotypic assortative mating, shared twin and sibling environments, and within-family environment. The genetic and environmental effects can be dependent on sex. Assortment and cultural inheritance may be based either on the phenotype as measured or on a latent trait of which the measured phenotype is an unreliable index. The model was applied to church attendance data from this study. The results show that the contributions of genes, vertical cultural inheritance, and genotype-environment covariance are all important, but their contributions are significantly heterogeneous over sexes. Phenotypic assortative mating has a major impact on family resemblance in church attendance.

Abstract

Four inbred strains of mice, DBA/2J, C57BL/10J, BALB/cJ, and SJL/J, were mated in a diallel cross. The cerebella of the F1 generation were examined for the presence (Type I) or absence (Type II) of an intraculminate fissure between vermian lobule IV and vermian lobule V (the ventral and dorsal lobules of the culmen). One strain (DBA/2J) consistently expressed the Type I pattern. Another strain (SJL/J) expressed predominantly the Type II pattern. The other two strains (C57BL/10J and BALB/cJ) and many of the hybrids exhibited variability in their expression of the foliation patterns. The results were analyzed using biometrical genetic procedures and showed significant additive and dominance genetic effects and a maternal effect. Correlations of these cerebellar anatomical variants with the development of behavior are discussed.

Abstract

Grayson (see the preceding paper) discusses some circumstances in which estimates of genetic and environmental parameters derived from the study of twins reared together may be biased and documents in those circumstances what the magnitude of the biases may be. As Grayson suggests, the points he makes have been made previously by various authors and issues such as the power to detect dominance have been analyzed at some length. This paper draws attention to some other sources of variation which Grayson does not consider but which have been considered by other writers and which might have somewhat different consequences for the estimation of shared environmental effects. The classical twin study has never been an end in itself, but it is the nucleus of a systematic genetic approach to the study of human behavior.

Abstract

Eating disorders and substance use disorders frequently co-occur. Twin studies reveal shared genetic variance between liabilities to eating disorders and substance use, with the strongest associations between symptoms of bulimia nervosa (BN) and problem alcohol use (genetic correlation [rg], twin-based=0.23-0.53). We estimated the genetic correlation between eating disorder and substance use and disorder phenotypes using data from genome-wide association studies (GWAS). Four eating disorder phenotypes (anorexia nervosa [AN], AN with binge-eating, AN without binge-eating, and a BN factor score), and eight substance-use-related phenotypes (drinks per week, alcohol use disorder [AUD], smoking initiation, current smoking, cigarettes per day, nicotine dependence, cannabis initiation, and cannabis use disorder) from eight studies were included. Significant genetic correlations were adjusted for variants associated with major depressive disorder (MDD). Total sample sizes per phenotype ranged from ~2,400 to ~537,000 individuals. We used linkage disequilibrium score regression to calculate single nucleotide polymorphism-based genetic correlations between eating disorder and substance-use-related phenotypes. Significant positive genetic associations emerged between AUD and AN (rg=0.18; false discovery rate q=0.0006), cannabis initiation and AN (rg=0.23; qwith binge-eating (rg=0.27; q=0.0016).
