As part of a whole egg, egg white proteins are embedded in a lipid matrix that could modify their presentation to the immune system and their allergenic properties. The present study examines the impact of the main egg lipid components, triacylglycerides and phospholipids, in the early events of sensitization to egg. To this end, BALB/c mice were exposed intragastrically to egg lipids and egg lipid fractions, alone and in mixtures with egg white proteins, and Th2-promoting and proinflammatory effects were investigated. Our results highlight that the egg lipid fraction is responsible for Th2 adjuvant effects and point at a different influence of triacylglycerides and phospholipids on the bioavailability and immunomodulating properties of egg white proteins. While triacylglycerides promote type 2 responses at the small intestine level, phospholipids reduce the solubility of EW proteins and induce Th2 skewing in lymphoid intestinal tissues, which may have a direct impact on the development of egg allergy.

Impaired glucose metabolism and mitochondrial decay greatly increase with age, when cognitive decline becomes rampant. No pharmacological or dietary intervention has proven effective, but proper diet and lifestyle do postpone the onset of neurodegeneration and some nutrients are being investigated. We studied insulin signaling, mitochondrial activity and biogenesis, and synaptic signaling in the hippocampus and cortex following dietary supplementation with bioactive phospholipid concentrates of krill oil (KOC), buttermilk fat globule membranes (BMFC), and a combination of both in aged rats. After 3 months of supplementation, although all groups of animals showed clear signs of peripheral insulin resistance, the combination of KOC and BMFC was able to improve peripheral insulin sensitivity. We also explored brain energy balance. Interestingly, the hippocampus of supplemented rats-mainly when supplemented with BMFC or the combination of KOC and BMFC-showed an increase in intracellular adenosine triphosphate (ATP) levels, whereas no difference was observed in the cerebral cortex. Moreover, we found a significant increase of brain-derived neurotrophic factor (BDNF) in the hippocampus of BMFC+KO animals. In summary, dietary supplementation with KOC and/or BMFC improves peripheral and central insulin resistance, suggesting that their administration could delay the onset of these phenomena. Moreover, n-3 fatty acids (FAs) ingested as phospholipids increase BDNF levels favoring an improvement in energy state within neurons and facilitating both mitochondrial and protein synthesis, which are necessary for synaptic plasticity. Thus, dietary supplementation with n-3 FAs could protect local protein synthesis and energy balance within dendrites, favoring neuronal health and delaying cognitive decline associated to age-related disrepair.

During many years, the milk fat has been unfairly undervalued due to its association with higher levels of cardiovascular diseases, dyslipidaemia or obesity, among others. However, currently, this relationship is being re-evaluated because some of the dairy lipid components have been attributed potential health benefits. Due to this, and based on the increasing incidence of cancer in our society, this review work aims to discuss the state of the art concerning scientific evidence of milk lipid components and reported anticancer properties. Results from the in vitro and in vivo experiments suggest that specific fatty acids (FA) (as butyric acid and conjugated linoleic acid (CLA), among others), phospholipids and sphingolipids from milk globule membrane are potential anticarcinogenic agents. An improved HPLC-ELSD method has been developed for the analysis of the lipid classes of buttermilk and milk from different species, focused in the phospholipids fraction without a prior fractionation step and in a single run. The total lipid profile analysis showed the major and minor lipid compounds as cholesterol esters, triacylglycerides, cholesterol, diacylglycerides, free fatty acids, monoacylglycerides, and also the polar compounds as glucosylceramide, lactosylceramide, phosphatidyl-ethanolamine, phosphatidylinositol, phosphatidylserine, phosphatidylcholine, sphingomyelin and lysophosphatidylcholine. The identification and quantification of the different compounds, using calibration curves made with individual standards and the low coefficients of variation obtained in the inter- and intra-assays showed the suitability of the developed method. Glycerophospholipids and sphingolipids participate in a variety of indispensable metabolic, neurological, and intracellular signaling processes. In this didactic paper we review the biological roles of phospholipids and try to unravel the precise nature of their putative healthful activities. We conclude that the biological actions of phospholipids activities potentially be nutraceutically exploited in the adjunct therapy of widely diffused pathologies such as neurodegeneration or the metabolic syndrome. Thermal processing of milk promotes oxidation reactions and changes in organoleptic characteristics. Application of High Pressure Processing (HPP) is a promising alternative but little is known about the impact on milk compounds, mainly lipids. This research aims to study the possible alteration of the composition of milk lipids and fatty acids as result of HPP using 250–900 MPa. Thus, two cow milk batches were assayed and the composition in tri-., di-., monoacylglycerols, cholesterol, cholesterol esters, free fatty acids, phospholipids and fatty acids analysed. It was found differences (p < 0.05) among batches in the distribution of triacylglycerols (CN34, CN50 and CN54) and also in the concentration of diacylglycerols, monoacylglycerols, cholesterol, phosphatidylethanolamine and phosphatidylinositol and unsaturated fatty acids (C18:1 10t, C18:1 9c, C18:1 11c, C18:1 16t+14c, C18:2 9c,12c, C18:2 9c,11t and C20:1 9c). Data from the current study showed that processing by HPP up to 900 MPa did not produce significant changes to the lipid classes or fatty acid composition of milk fat. [ABSTRACT FROM AUTHOR]
