A chlorophyll fluorescence (ChlF) assessment was carried out on oak seedlings (Quercus ilex L. Quercus pubescens Willd. Quercus frainetto Ten.) of Italian and Greek provenance, during the years 2017 and 2018, in a common garden in central Italy planted in 2017. This trial aimed to test the relative performances of the oak species in the perspective of assisted migration as part of the actions for the adaptation of forests to climate change. The assessment of the photosynthetic performance of the tree species included the analysis of the prompt chlorophyll fluorescence (PF) transient and the modulated reflection (MR) at 820 nm, leaf chlorophyll content, leaf gas exchange (net photosynthesis, stomatal conductance), plant growth (i.e. height) and mortality rate after 2 years from the beginning of the experiment. The assessment of the performance of the three oak species was carried out 'in vivo'. Plants were generated from seeds and exposed to several environmental factors, including changing seasonal temperature, water availability, and soil biological and physical functionality. The results of PF indicate a stable functionality of the photosynthetic system PSII (expressed as F V/ F M) across species and provenances and a decline in photochemistry functionality at the I–P phase (Δ V IP) in Q. frainetto , thus indicating a decline of the content of PSI in this species. This result was confirmed by the findings of MR analysis, with the speed of reduction and subsequent oxidation of PSI (V RED and V OX) strongly correlated to the amplitude of Δ V IP. The photosynthetic rates (net photosynthesis, P N) and growth were correlated with the parameters associated with PSI content and function, rather than those related to PSII. The low performance of Q. frainetto in the common garden seems to be related to early foliar senescence with the depletion of nitrogen, due to suboptimal climatic and edaphic conditions. Chlorophyll fluorescence allowed discrimination of populations of oak species and individuation of the less (or/and best) suitable species for future forest ecology and management purposes. Flavescence dorée ( FD) is among the major grapevine diseases causing high management costs; curative methods against FD are unavailable. In FD-infected plants, decrease in photosynthesis is usually recorded, but deregulation in stomatal control of leaf gas exchange during FD infection and recovery is unknown. We measured the seasonal time course of gas exchange rates in two cultivars ('Barbera' and 'Nebbiolo') during the term of 1 year when grapevines experienced a water stress and another with no drought, with difference in gas exchange rates in response to FD infection and recovery as assessed by symptom observation and phytoplasma detection through PCR analysis. Chlorophyll fluorescence was also evaluated at the time of maximum symptom severity in 'Barbera', the cultivar showing the most severe stress response to FD infection, causing the highest damage in vineyards of north-western Italy. In FD-infected plants, net photosynthesis and transpiration gradually decreased during the season, more during the no drought year than during drought. During recovery, healthy ( PCR negative) plants infected 2 years before, but not those infected an year before, regained the gas exchange performances to the level as measured before infection. The relationships between stomatal conductance and the residual leaf intercellular CO2 concentration ( ci) discriminated healthy versus FD-infected and recovered plants; at the same ci, FD-infected leaves had higher non-photochemical quenching than healthy ones. We conclude that metabolic, not stomatal, leaf gas exchange limitation in FD-infected and recovered grapevines is the basis of plant response to FD disease. In addition, we also suggest that such response is dependent upon water stress, by showing that water stress superimposes on FD infection in terms of stomatal and metabolic non-stomatal limitations to carbon assimilation. The study was carried out in a four-year-old super-high density olive grove in Central Italy to compare leaf gas exchanges of Spanish Arbequina and Italian Maurino olive cultivars. Overall, from mid July to mid November, Maurino had a slightly higher maximum light-saturated net photosynthetic rate ( P) than Arbequina. The lowest and the highest P values were recorded at the end of July and in mid November, respectively. Current-season leaves showed similar or slightly higher P values than one-year-old leaves. During the day Maurino always had slightly higher values or values similar to Arbequina, with the highest P being in the morning. Maurino had similar or higher dark respiration rate ( R) values compared to Arbequina. During the day, in both cultivars the R was lower at 9:00 than in the afternoon. The pattern of the photosynthetic irradiance-response curve was similar in the two genotypes, but the apparent quantum yield ( Y) was higher in Maurino. In both cultivars intercellular CO concentration ( C) tended to increase when P decreased. The increase in C corresponded to a decrease in stomatal conductance ( g). The transpiration rate ( E) increased from mid July to the beginning of August, then decreased in September and increased again in November. Particularly in the morning, the current-season leaves showed similar or slightly higher E values than the one-year-old leaves. During the day, in both cultivars and at both leaf ages, E was higher in the afternoon. No effects on leaf gas exchanges due to the presence or absence of fruit on the shoot were found. Overall, there was satisfactory physiological adaptation for Arbequina to the conditions of Central Italy and for Maurino to the superintensive grove conditions. [ABSTRACT FROM AUTHOR]

The development of isidia in thalli of Pseudevernia furfuracea from the Carnic Alps (North-eastern Italy), and the effects of these structures on CO2 gas exchanges were investigated. The ontogenetic events were studied by comparison of sections stained with different histochemical tests and SEM observations. A high cell turnover rate in both symbiotic partners is the first sign of isidium development, followed by an increased aplanosporogenesis of algae and growth of neighbouring medullary hyphae which become oriented upwards. Large nuclei and an intense cytoplasm activity characterize the mycobiont cells. The surface of very young isidia shows an irregular structure of spherical to ovoid protruding tips of perpendicular cortical hyphae, that are later organised in a pseudomeristematic area similar to that observed in the apex of growing lobes. CO2 gas exchange measurements carried out in the laboratory confirmed the high metabolic activity of isidia. At optimal water content and favourable light conditions, isolated isidia had rates of gross photosynthesis and dark respiration that were twice those of non-isidiate lobes. Isolated isidia also had a very low CO2 saturation point, probably because of their favourable surface/volume ratio, and a high light saturation, probably linked to their high content of photosynthetic pigments. The different roles played by isidia in the biology of Pseudevernia furfuracea, and particularly their rejuvenating effect on aged lobes, are discussed, and the presence of thalloconidia is briefly mentioned. Isoprene basal emission (the emission of isoprene from leaves exposed to a light intensity of 1000 µmol m−2 s−1 and maintained at a temperature of 30 °C) was measured in Phragmites australis plants growing under elevated CO2 in the Bossoleto CO2 spring at Rapolano Terme, Italy, and under ambient CO2 at a nearby control site. Gas exchange and biochemical measurements were concurrently taken. Isoprene emission was lower in the plants growing at elevated CO2 than in those growing at ambient CO2. Isoprene emission and isoprene synthase activity (IsoS) were very low in plants growing at the bottom of the spring under very rich CO2 and increased at increasing distance from the spring (and decreasing CO2 concentration). Distance from the spring did not significantly affect photosynthesis making it therefore unlikely that there is carbon limitation to isoprene formation. The isoprene emission rate was very quickly reduced after rapid switches from elevated to ambient CO2 in the gas-exchange cuvette, whereas it increased when switching from ambient to elevated CO2. The rapidity of the response may be consistent with post-translational modifications of enzymes in the biosynthetic pathway of isoprene formation. Reduction of IsoS activity is interpreted as a long-term response. Basal emission of isoprene was not constant over the day but showed a diurnal course opposite to photosynthesis, with a peak during the hottest hours of the day, independent of stomatal conductance and probably dependent on external air temperature or temporary reduction of CO2 concentration. The present experiments show that basal emission rate of isoprene is likely to be reduced under future elevated CO2 levels and allow improvement in the modelling of future isoprene emission rates.
