Coronatine (COR) is a phytotoxin produced by Pseudomonas syringae. Its structure is similar to Jasmonates, which play a number of diverse roles in plant defense. Both have the COI1 plant receptor, so coronatine can manipulate plant hormone signaling to access nutrients and counteract defense responses. In addition to the hormone system, coronatine affects plant nitrogenous metabolism and chloroplast ultrastructure. In this study, we first examined a typical nitrogen-losing phenotype, and used the polyacrylamide gel approach to demonstrate soluble total protein patterns in a time-course experiment under different temperature conditions. We then employed dimensional gel electrophoresis technology (2-DE) and MALDI-TOF-MS to sequester and identify the sensitive proteins. We found a total of 27 coronatine sensitive proteins, 22 of which were located in the chloroplast and 6 of which were directly involved in photosynthesis. Finally, we measured levels of chlorophyll and photosynthetic performance to reveal the phenotypic effect of these proteins. Taken together, these results demonstrated that coronatine enhanced heat tolerance by regulating nitrogenous metabolism and chloroplast ultrastructure to maintain photosynthetic performance and reduce yield loss under heat stress.

Coronatine (COR), a structural and functional mimic of jasmonates, is involved in a wide array of effects on plant development and defence responses. This study was conducted to explore the role of exogenously applied COR in alleviating the adversities of drought stress in soybean. COR treatment markedly increased the activities of antioxidant enzymes and proline content, and reduced the accumulation of malondialdehyde and hydrogen peroxide under drought stress. Thus, COR-treated plants had higher leaf relative water content and lower electrolye leakage, which led to higher chlorophyll content, activities of RuBPCase and PEPCase, and net photosynthetic rate compared to control plants exposed to drought. COR also increased maximal efficiency of PS II photochemical reaction and photochemical quenching coefficient, but decreased non-photochemical quenching coefficient. These beneficial effects led to enhanced photosynthetic performance and the translocation of assimilated (14)C which promoted growth and accumulation of dry biomass in COR-treated soybean plants subjected to drought. Interestingly, COR application did not affect the growth and biomass accumulation under well-watered condition. A hydroponic culture experiment was conducted to determine genotypic variation in photosynthetic rate and the associated physiological changes in response to potassium (K) deficiency in cotton (Gossypium hirsutum L.) seedlings with contrasting two cotton cultivars in K efficiency. The K-efficient Liaomian18 produced 66.7% more biomass than the K-inefficient NuCOTN99(B) under K deficiency, despite their similar biomass under K sufficiency. Compared with NuCOTN99(B), Liaomian18 showed 19.4% higher net photosynthetic rate (P(n), per unit leaf area) under K deficient solutions and this was associated with higher photochemical efficiency and faster export of soluble sugars from the phloem. The lower net P(n) of NuCOTN99(B) was attributed to higher capacity for nitrate assimilation and lower export of soluble sugars. Furthermore, NuCOTN99(B) showed 38.4% greater ETR/P(n) than Liaomian18 under K deficiency, indicating that more electrons were driven to other sinks. Higher superoxide dismutase (SOD) and lower catalase (CAT) and ascorbate peroxidase (APX) activities resulted in higher levels of reactive oxygen species (ROS; e.g. O(2)(-)and H(2)O(2)) in NuCOTN99(B) relative to Liaomian18. Thus, the K inefficiency of NuCOTN99(B), indicated by lower biomass and net P(n) under K deficiency, was associated with excessively high nitrogen assimilation, lower export of carbon assimilates, and greater ROS accumulation in the leaf., (Crown Copyright © 2012. Silicon (Si) may be involved in metabolic, physiological, and/or structural activity in higher plants exposed to abiotic and biotic stresses. This has not yet been determined due to the absence of direct evidence that it is part of the molecule of an essential plant constituent or metabolite. The aim of this study was to investigate the effect of silicon on soybean seedlings under drought and ultraviolet-B (UV-B) radiation stresses. The relative leaf water content (RWC), which was the main factor resulting in reduced growth in response to drought, increased 19.0% and 30.0% with Si application under drought and drought+UV-B stresses, respectively. Under UV-B radiation, the anthocyanin and phenol levels decreased 91.5% and 10.0% in the treatment of Si. Ultraviolet-B radiation and drought stress caused great membrane damage, as assessed by lipid peroxidation and osmolyte leakage, but Si application significantly reduced the membrane damage. Catalase (CAT), peroxidase (POD), superoxide dismutase (SOD) and hydrogen peroxide were observed under stress conditions. Proline increased primarily in drought-stressed seedlings and may be the drought-induced factor with a protective role in response to UV-B and silicon. Photosynthesis (P(N)) increased following Si application by 21.0%, 18.3% and 21.5% under UV-B radiation, drought and the combination, respectively. The physiological and biochemical parameters measured indicated that the UV-B light had more adverse effects on growth of soybean seedlings than drought, but the data also showed that Si could alleviate seedling damage under these stress conditions., (Copyright (c) 2010 Elsevier GmbH. This study investigated whether uniconazole confers drought tolerance to soybean and if such tolerance is correlated with changes in photosynthesis, hormones and antioxidant system of leaves. Soybean plants were foliar treated with uniconazole at 50 mg L-1 at the beginning of bloom and then exposed to water deficit stress at pod initiation for 7 d. Uniconazole promoted biomass accumulation and seed yield under both water conditions. Plants treated with uniconazole showed higher leaf water potential only in water-stressed condition. Water stress decreased the chlorophyll content and photosynthetic rate, but those of uniconazole-treated plants were higher than the stressed control. Uniconazole increased the maximum quantum yield of photosystemand ribulose-1,5-bisphosphate carboxylase/oxygenase activity of water-stressed plants. Water stress decreased partitioning of assimilated 14C from labeled leaf to the other parts of the plant. In contrast, uniconazole enhanced translocation of assimilated 14C from labeled leaves to the other parts, except stems, regardless of water treatment. Uniconazole-treated plants contained less GA3, GA4 and ABA under well-watered condition than untreated plants, while the IAA and zeatin levels were increased substantially under both water conditions, and ABA concentration was also increased under water stressed condition. Under water-stressed conditions, uniconazole increased the content of proline and soluble sugars, and the activities of superoxide dismutase and peroxidase in soybean leaves but not the malondialdehyde content or electrical conductivity. These results suggest that uniconazole-induced tolerance to water deficit stress in soybean was related to the changes of photosynthesis, hormones and antioxidant system of leaves.

The role of melatonin in plant growth and response to environmental stress has been widely demonstrated. However, the physiological and molecular regulation of salt tolerance in wheat seedlings by melatonin remains unclear. In this study, we investigated changes in phenotype, physiology, photosynthetic parameters, and transcript levels in wheat seedlings to reveal the role of melatonin in the regulation of salt tolerance in wheat. The results indicate that the application of exogenous melatonin significantly alleviates growth inhibition, reactive oxygen species accumulation, and membrane oxidative damage induced by salt stress in wheat. Additionally, exogenous melatonin increased antioxidant enzyme activity and regulated photosynthetic gas exchange. Transcriptomic data showed a significant up-regulation of genes encoding light-harvesting chlorophyll protein complex proteins in photosynthesis and genes related to chlorophyll and carotenoid biosynthesis under the influence of melatonin. These results suggest that exogenous melatonin improves salt tolerance in wheat seedlings by enhancing the antioxidant, photoprotective, and photosynthesis activities. [ABSTRACT FROM AUTHOR]

The aim of this study was to investigate the potential of silicon (Si) for alleviating Ultraviolet-B (UV-B) radiation stress based on changes in biomass, physiological attributes and photosynthetic characteristics of two soybean (Glycine maxL.) cultivars, Kenjiandou 43 (‘K 43’) and Zhonghuang 35 (‘ZH 35’). The cultivars were raised with and without Si in the greenhouse, and then subjected to ambient, ambient + 2.7 kJ m−2d−1and ambient + 5.4 kJ m−2d−1of UV-B radiation. Depending on cultivar, plants suffered severe growth limitations under UV-B radiation, but the application of Si alleviated the adverse effects on growth and development by increasing the stem length, net photosynthetic rate (PN) and leaf chlorophyll content. Concurrently, it decreased the stomatal conductance (Sc) and intercellular carbon dioxide (CO2) concentration (Ci). In response to the UV-B radiation stress, the antioxidant enzyme activities of superoxide dismutase (SOD) increased by 41.2–72.7%, peroxidases (POD) by 49.5–85.7%, malodialdehyde (MDA) by 6.7–20.4% and soluble protein by 4.2–7.6%. The overall results indicated that media treatment with Si might improve soybean growth under elevated UV-B radiation through positive changes in biomass and some physiological attributes that were dependent on cultivar. Depletion of the ozone layer leads to increasing UV-B radiation on the earth's surface, which may affect weeds and their responses to herbicides. However, the effect of increased UV-B radiation on weeds and the interaction of weeds and herbicides are still obscure. The objective of this study was to compare glyphosate efficacy on velvetleaf that was grown under with and without increased UV-B radiation. Leaf area, dry weight and net photosynthesis of velvetleaf seedlings were adversely affected by increased UV-B radiation. Leaf cuticle wax significantly increased by 28% under increased UV-B radiation. Glyphosate efficacy on velvetleaf, evaluated by shoot dry weight, was significantly decreased by increased UV-B radiation. Exposure to increased UV-B radiation significantly decreased 14C-glyphosate absorption from 49% to 43%, and also resulted in less 14C-glyphosate translocation out of treated leaves and less glyphosate accumulation in newly expanded leaves. The decrease in glyphosate efficacy was due to changes in absorption and distribution, which were attributed to increased cuticle wax and decreased photosynthesis caused by increased UV-B radiation. These results suggest that the responses of weeds to herbicides may be affected by increased UV-B radiation, to the extent that higher rates may be required to achieve the desired effects. With the aim to determine whether coronatine (COR) alleviates drought stress on wheat, two winter wheat ( Triticum aestivum L.) cultivars, ChangWu134 (drought-tolerant) and Shan253 (drought-sensitive) were studied under hydroponic conditions. Seedlings at the three-leaf stage were cultured in a Hoagland solution containing COR at 0.1 μM for 24 h, and then exposed to 20% polyethylene glycol 6000 (PEG-6000). Under simulated drought (SD), COR increased the dry weight of shoots and roots of the two cultivars significantly; the root/shoot ratio also increased by 30% for Shan253 and 40% for ChangWu134. Both cultivars treated with COR under SD (0.1COR+PEG) maintained significantly higher relative water content, photosynthesis, transpiration, intercellular concentration of CO2 and stomatal conductance in leaves than those not treated with PEG. Under drought, COR significantly decreased the relative conductivity and malondialdehyde production, and the loss of 1,1-diphenyl-2-picrylhydrazyl scavenging activity in leaves was significantly alleviated in COR-treated plants. The activity of peroxidase, catalase, glutathione reductase and ascorbate peroxidase were adversely affected by drought. Leaves of plants treated with COR under drought produced less abscisic acid (ABA) than those not treated. Weed tolerance of UV-B radiation varies with species, and the radiation could affect weed ecology and management. Variations in growth, photosynthesis and defense system among four important agronomic weeds, Abutilon theophrasti Medik, Amaranthus retroflexus L., Digitaria sanguinalis (L.) Scop and Chloris virgata Swartz, under increased UV-B radiation (ambient and increased radiation at 2.7, 5.4 and 10.8 kJ-m−2-d−1) were studied in the greenhouse experiment. After 2 weeks of radiation, the shoots’dry mass decreased with increasing UV-B radiation except for D. sanguinalis. The reduction in biomass was the result of changes in morphology and physiology. Higher levels of UV-B treatment decreased the leaf area, plant height, net photosynthetic rate and chlorophyll contents, while it increased the contents of wax and UV-B absorbing compound in all species, except for A. retroflexus, which did not increase significantly. The activity of superoxide dismutase, catalase, ascorbate peroxide and the content of ascorbic acid changed differently among the weed species as UV-B radiation increased. D. sanguinalis was the most tolerant and A. retroflexus the most sensitive to increased UV-B radiation. The results also show that the two grass species (D. sanguinalis and C. virgata) were more tolerant to UV-B radiation than the two broad-leafed species (A. theophrasti and A. retroflexus). The UV-B absorbing compound and leaf wax played important roles against UV-B damages in the two grass weeds. Coronatine (COR) is a phytotoxin produced by Pseudomonas syringae . Its structure is similar to those of jasmonates (JAs), which play diverse roles in multiple plant biotic and abiotic defenses. However, the biological activity of COR is 1000 times greater than the activity of JA. In addition to being involved in the JA pathway, COR affects plant photosynthetic efficiency. In this study, we examined wheat blade pretreatment with COR. Blades treated with COR remained green longer than those of control plants under drought stress conditions, resulting in less yield loss with COR treatment. To investigate the mechanism of COR in drought resistance further, we employed two-dimensional gel electrophoresis technology and matrix-assisted laser desorption/ionization mass spectrometry to sequester and identify key proteins. Six COR-inducible proteins that are located in the chloroplast and involved directly in photosynthesis were found. The wheat homologue of protein gi|326509937 is degradation of periplasmic proteins 1 (DEGP1) in Arabidopsis , which is a response to photosystem II reparation, and was maintained at a low level with COR treatment. Finally, we measured levels of chlorophyll and photosynthetic performance to reveal the phenotypic effect of COR. Abstract: The objective of this study was to determine the roles of shoot and root in the regulation of premature leaf senescence induced by potassium (K) deficiency in cotton (Gossypium hirsutum L.). Two contrasting cultivars (CCRI41, more sensitive to K deficiency; and SCRC22, a less sensitive cultivar) were selected for self- and reciprocal-grafting, using standard grafting (one scion/one rootstock), Y grafting (two scions/one rootstock) and inverted Y grafting (one scion/two rootstocks) at the seedling stage. Standard grafting was studied in the field in 2007 and 2008. There were no obvious differences in senescence between CCRI41 and SCRC22 scions while supplied with sufficient K. However, SCRC22 scions showed significantly greater K content, SPAD values (chlorophyll content), soluble protein content and net photosynthetic rates than CCRI41 scions while grown in K deficient solution or soil, regardless of rootstock cultivars, grafting types, growth stage and growth conditions. Also, SCRC22 scions had greater yield and less variation in boll weight either between upper- and lower sympodials, or between proximal and distal fruit positions from the main stem in the field under K deficiency, probably owing to reduced leaf senescence. Although the effect of rootstocks on leaf senescence under K deficiency was significant in some cases, the scion cultivars explained the highest percentage of variations within grafting treatments. The shoot-to-root feedback signal(s), rather than high shoot demand for K nutrition, was involved in the shoot regulation of premature senescence in cotton plants, achieved possibly by altering root K uptake.
