Toxic heavy metals and metalloids in agricultural ecosystems are crucial factors that limit global crop productivity and food safety. Industrial toxic heavy metals and metalloids such as cadmium, lead, and arsenic have contaminated large areas of arable land in the world and their accumulation in the edible parts of crops is causing serious health risks to humans and animals. Plants have co-evolved with various concentrations of these toxic metals and metalloids in soil and water. Some green plant species have significant innovations in key genes for the adaptation of abiotic stress tolerance pathways that are able to tolerate heavy metals and metalloids. Increasing evidence has demonstrated that phytohormone abscisic acid (ABA) plays a vital role in the alleviation of heavy metal and metalloid stresses in plants. Here, we trace the evolutionary origins of the key gene families connecting ABA signaling with tolerance to heavy metals and metalloids in green plants. We also summarize the molecular and physiological aspects of ABA in the uptake, root-to-shoot translocation, chelation, sequestration, reutilization, and accumulation of key heavy metals and metalloids in plants. The molecular evolution and interaction between the ABA signaling pathway and mechanisms for heavy metal and metalloid tolerance are highlighted in this review. Therefore, we propose that it is promising to manipulate ABA signaling in plant tissues to reduce the uptake and accumulation of toxic heavy metals and metalloids in crops through the application of ABA-producing bacteria or ABA analogues. Our previous studies showed that high salt tolerance in Tibetan wild barley accessions was associated with HvHKT1;1, a member of the high-affinity potassium transporter family. However, molecular mechanisms of HvHKT1;1 for salt tolerance and its roles in K+/Na+ homeostasis remain to be elucidated. Functional characterization of HvHKT1;1 was conducted in the present study. NaCl-induced transcripts of HvHKT1;1 were significantly higher in the roots of Tibetan wild barley XZ16 relative to other genotypes, being closely associated with its higher biomass and lower tissue Na+ content under salt stress. Heterologous expression of HvHKT1;1 in Saccharomyces cerevisiae (yeast) and Xenopus laevis oocytes showed that HvHKT1;1 had higher selectivity for Na+ over K+ and other monovalent cations. HvHKT1;1 was found to be localized at the cell plasma membrane of root stele and epidermis. Knock-down of HvHKT1;1 in barley led to higher Na+ accumulation in both roots and leaves, while overexpression of HvHKT1;1 in salt-sensitive Arabidopsis hkt1-4 and sos1-12 loss-of-function lines resulted in significantly less shoot and root Na+ accumulation. Additionally, microelectrode ion flux measurements and root elongation assay revealed that the transgenic Arabidopsis plants exhibited a remarkable capacity for regulation of Na+, K+, Ca2+ and H+ homeostasis under salt stress. These results indicate that HvHKT1;1 is critical in radial root Na+ transport, which eventually reduces shoot Na+ accumulation. Halophyte species implement a "salt-including" strategy, sequestering significant amounts of Na+ to cell vacuoles. This requires a reduction of passive Na+ leak from the vacuole. In this work, we used quinoa (Chenopodium quinoa) to investigate the ability of halophytes to regulate Na+-permeable slow-activating (SV) and fast-activating (FV) tonoplast channels, linking it with Na+ accumulation in mesophyll cells and salt bladders as well as leaf photosynthetic efficiency under salt stress. Our data indicate that young leaves rely on Na+ exclusion to salt bladders, whereas old ones, possessing far fewer salt bladders, depend almost exclusively on Na+ sequestration to mesophyll vacuoles. Moreover, although old leaves accumulate more Na+, this does not compromise their leaf photochemistry. FV and SV channels are slightly more permeable for K+ than for Na+, and vacuoles in young leaves express less FV current and with a density unchanged in plants subjected to high (400 HIM NaCl) salinity. In old leaves, with an intrinsically lower density of the FV current, FV channel density decreases about 2-fold in plants grown under high salinity. In contrast, intrinsic activity of SV channels in vacuoles from young leaves is unchanged under salt stress. In vacuoles of old leaves, however, it is 2- and 7-fold lower in older compared with young leaves in control- and salt-grown plants, respectively. We conclude that the negative control of SV and FV tonoplast channel activity in old leaves reduces Na+ leak, thus enabling efficient sequestration of Na+ to their vacuoles. Stomatal guard cells play a key role in gas exchange for photosynthesis while minimizing transpirational water loss from plants by opening and closing the stomatal pore. Foliar gas exchange has long been incorporated into mathematical models, several of which are robust enough to recapitulate transpirational characteristics at the whole-plant and community levels. Few models of stomata have been developed from the bottom up, however, and none are sufficiently generalized to be widely applicable in predicting stomatal behavior at a cellular level. We describe here the construction of computational models for the guard cell, building on the wealth of biophysical and kinetic knowledge available for guard cell transport, signaling, and homeostasis. The OnGuard software was constructed with the HoTSig library to incorporate explicitly all of the fundamental properties for transporters at the plasma membrane and tonoplast, the salient features of osmolite metabolism, and the major controls of cytosolic-free Ca2+ concentration and pH. The library engenders a structured approach to tier and interrelate computational elements, and the OnGuard software allows ready access to parameters and equations 'on the fly' while enabling the network of components within each model to interact computationally. We show that an OnGuard model readily achieves stability in a set of physiologically sensible baseline or Reference States; we also show the robustness of these Reference States in adjusting to changes in environmental parameters and the activities of major groups of transporters both at the tonoplast and plasma membrane. In guard cells, activation of anion channels ( Ianion) is an early event leading to stomatal closure. Activation of Ianion has been associated with abscisic acid (ABA) and its elevation of the cytosolic free Ca2+ concentration ([Ca2+]i). However, the dynamics of the action of [Ca2+]i on Ianion has never been established, despite its importance for understanding the mechanics of stomatal adaptation to stress. We have quantified the [Ca2+]i dynamics of Ianion in Vicia faba guard cells, measuring channel current under a voltage clamp while manipulating and recording [Ca2+]i using Fura-2 fluorescence imaging. We found that Ianion rises with [Ca2+]i only at concentrations substantially above the mean resting value of 125 ± 13 nm, yielding an apparent Kd of 720 ± 65 nm and a Hill coefficient consistent with the binding of three to four Ca2+ ions to activate the channels. Approximately 30% of guard cells exhibited a baseline of Ianion activity, but without a dependence of the current on [Ca2+]i. The protein phosphatase antagonist okadaic acid increased this current baseline over twofold. Additionally, okadaic acid altered the [Ca2+]i sensitivity of Ianion, displacing the apparent Kd for [Ca2+]i to 573 ± 38 nm. These findings support previous evidence for different modes of regulation for Ianion, only one of which depends on [Ca2+]i, and they underscore an independence of [Ca2+]i from protein (de-)phosphorylation in controlling Ianion. Most importantly, our results demonstrate a significant displacement of Ianion sensitivity to higher [Ca2+]i compared with that of the guard cell K+ channels, implying a capacity for variable dynamics between net osmotic solute uptake and loss. As an important organic compound, chiral alcohols are the key chiral building blocks to many single enantiomer pharmaceuticals. Asymmetric reduction of the corresponding prochiral ketones to produce the chiral alcohols by biocatalysis is one of the most promising routes. Asymmetric reduction of different kinds of non-natural prochiral ketones catalyzed by various plants tissue was studied in this work. Acetophenone, 4′-chloroacetophenone and ethyl 4-chloroacetoacetate were chosen as the model substrates for simple ketone, halogen-containing aromatic ketone and β-ketoesters, respectively. Apple ( Malus pumila), carrot ( Daucus carota), cucumber ( Cucumis sativus), onion ( Allium cepa), potato ( Soanum tuberosum), radish ( Raphanus sativus) and sweet potato ( Ipomoea batatas) were chosen as the biocatalysts. It was found that these kinds of prochiral ketoness could be reduced by these plants tissue with high enantioselectivity. Both R- and S-form configuration chiral alcohols could be obtained. The e.e. and chemical yield could reach about 98 and 80% respectively for acetophenone and 4′-chloroacetophenone reduction reaction with favorable plant tissue. Asymmetric reduction of the prochiral aromatic ketone catalyzed by yeast cells is one of the most promising routes to produce its corresponding enantiopure aromatic alcohol, but the space-time yield does not meet people's expectations. Therefore, the toxicity of aromatic ketone and aromatic alcohol to the yeast cell is investigated in this work. It has been found that the aromatic compounds are poisonous to the yeast cell. The activity of yeast cell decreases steeply when the concentration of acetophenone (ACP) is higher than 30.0 mmol/L. Asymmetric reduction of acetophenone to chiral S-α-phenylethyl alcohol (PEA) catalyzed by the yeast cell was chosen as the model reaction to study in detail the promotion effect of the introduction of the resin adsorption on the asymmetric reduction reaction. The resin acts as the substrate reservoir and product extraction agent in situ. It has been shown that this reaction could be remarkably improved with this technique when the appropriate kind of resin is applied. Understanding the physiological and molecular response of crop genotypes could be useful in eco-toxicological evaluation with cadmium (Cd) and could be a strategy to solve heavy metal contamination in agriculture. This study corroborates unique patterns of Cd accumulation and molecular mechanisms adopted by plants to acquire Cd tolerance and counteractive effects of zinc (Zn) against Cd toxicity. Two rice (Oryza sativa) genotypes (Heizhan 43 and Yinni 801) differing in cadmium tolerance and its accumulation in plant tissues were investigated hydroponically using two Cd levels [Cd 10 (10 μM L−1) and Cd 15 (15 μM L−1)] and two Zn levels [Zn 25 (25 μM L−2) and Zn 50 (50 μM L−1)] and their combinations. Cadmium toxicity rendered substantial reduction in plant height, biomass, chlorophyll contents and photosynthesis as compared to the control plants after 15 days of treatment. Supplementation of Zn evidently ameliorated Cd toxicity by minimizing the reduction in plant growth, chlorophyll contents and photosynthetic attributes (Pn , gs , Ci , and Tr). Comparatively, lower accumulation of Cd in Yinni 801 under combined treatments revealed a preferential uptake of Zn in this genotype. A cross-talk among Cd, Zn, Fe, Ca and K correlated with fluctuating gs , Ci and Tr. Both genotypes also differed in morphological alterations of cell membrane, chloroplasts and appearance of enlarged plastoglobuli along with distorted mitochondria. An increased ascorbate peroxidase activity in roots of Yinni 801 presented a defensive strategy. Relative expression of Cd and Zn ion transporter genes also confirmed the genotypic background of phenotypic divergence. The OsLCT1 and OsHMA2 expression was significant in Heizhan 43, indicating possible translocation of Cd from shoot to grains contrary to Yinni 801, which accumulated Cd in shoot and showed stunted growth. Zn supplementation promises tolerance to Cd in Yinni 801 by differential expression of putative genes for Cd translocation with minimum ultrastructural modifications by maintaining physiological functions in contrast to Heizhan 43. • Reduced Cd accumulation in shoots of Yinni 801 genotype by Zn caused Cd tolerance. • Elemental cross-talk indicates that K+ and Ca2+ regulate photosynthesis. • Zn competes with Cd by sharing the same transporters such as OsIRT1 in rhizosphere and OsHMA2 a xylem loading transporter. • Suppression of Cd transporter genes reduced its active translocation. Panax ginseng is one of the most valuable medicinal plants in the world, and wild-forest (WG) and artificial-forest (AG) ginseng are very popular in the ginseng market, with ginsenosides constituting a majority of the bioactives. Research on the biochemical and physiological patterns of metabolic accumulation in different tissues of ginseng cultivated under various conditions is relatively scarce. We profiled metabolites using GC/MS and LC/MS to explore the bioactive component changes and interrelationships that occur in 7 tissues of WG and AG. In total, 149 primary metabolites and 46 secondary compounds were found in aboveground and belowground tissues. Metabolite changes associated with primary and secondary biochemistry were observed, and the levels of ginsenoside F2 and other compounds showed a significant correlation by statistical analysis in ginseng under both cultivation methods, as observed for secondary compounds and C and N metabolites. In addition, the number of secondary components was higher in the aboveground parts than in the belowground parts, showing a different pattern, and the same accumulation pattern of compounds involved in C and N metabolism was observed in individual plant tissues, but the high rate of photosynthesis and energy metabolism in WG provided energy for the biosynthesis of secondary compounds. Furthermore, artificial neural network models explained the variation in the secondary compounds very well via the combination of several different metabolites from WG and AG. Finally, C and N metabolism plays a key role in secondary compound biosynthesis in specific tissues and cultivation conditions and highlights large-scale metabolite patterns in WG and AG.
