Menthae Haplocalycis Herba has been utilized for food and medicinal purposes in China for thousands of years. It has various efficacies, including dispelling wind and heat and relieving sore throat. M. Haplocalycis Herba has been also widely used in food, cosmetics, spices, and other fields. Exploring the constituents and detecting the metabolites of M. Haplocalycis are of great significance to clarify the effective substances. However, the in vivo metabolites of M. Haplocalycis Herba water extract are still unclear. Herein, a sensitive and specific method, ultra-high performance liquid chromatography with linear ion trap-Orbitrap mass spectrometry, established in this assay was used to study the metabolism of M. Haplocalycis Herba water extract in rat plasma, urine, and feces. We characterized and identified 9, 50, and 34 metabolites in plasma, urine, and feces, respectively. Seven metabolic pathways, including phase Ⅰ (isomerization, demethylation, hydroxylation, and dehydration) and phase Ⅱ (sulfation and glucuronidation) were mainly involved in the metabolism. It is the first systematic study on the metabolism of M. Haplocalycis Herba water extract in vivo, which enrich current understanding of the metabolic behavior of M. Lonicerae Japonicae Flos (LJF), a kind of traditional Chinese medicines (TCMs), has functions of detoxifying and evacuating heat. In the study, a method based on ultra-high performance liquid chromatography coupled with linear ion trap-Orbitrap mass spectrometry (UHPLC-LTQ-Orbitrap MS) was developed for the chemical constituent analysis of organic acids, flavonoids, iridoids and new-generated compounds in sulfur-fumigated LJF (SF-LJF). Based on the accurate mass measurement (< ± 5 ppm), chromatographic behavior and diagnostic product ions (DPIs), 113 constituents were unambiguously or tentatively characterized from SF-LJF extract, including 46 chlorogenic acids, 19 flavonoids, 29 iridoid glycosides and 19 newly-generated compounds (including 17 sulfur-containing derivatives). In addition, 5-CQA (5-caffeoylquinic acid, chlorogenic acid) was chosen to be sulfur-fumigated for the result validation. It was found that the most significant change of LJF after sulfur fumigation was the occurrence of sulfate or sulfite esterification reactions, which resulted in the emergence of many new sulfur-containing components. Our results demonstrated that the established method was a useful and efficient analytical tool to comprehensively characterize the material basis of SF-LJF, and also an excellent guidance of quality control about LJF., (Copyright © 2020 China Pharmaceutical University. Liriope (Liliaceae) species have been used as folk medicines in Asian countries since ancient times. From Liriope plants (8 species), a total of 132 compounds (except polysaccharides) have been isolated and identified, including steroidal saponins, flavonoids, phenols, and eudesmane sesquiterpenoids. The crude extracts or monomeric compounds from this genus have been shown to exhibit anti-tumor, anti-diabetic, anti-inflammatory, and neuroprotective activities. The present review summarizes the results on phytochemical and biological studies on Liriope plants. A rapid and sensitive UHPLC-HR-MSn method was developed for the identification of chemical constituents in capillary wormwood extract. ACQUITY UHPLC HSS T3 chromatography column (2.1 mm×100 mm, 1.7 μm) was used with 0.1% formic acid-acetonitrile solution as the mobile phase in gradient elution. The extract was detected by ESI-LTQ-Orbitrap equipped with an ESI ion source in a negative mode. Based on the accurate mass measurements, retention time, mass fragmentation patterns and literature reports, a total of 50 compounds including 21 flavonoids, 22 phenolic acids, 6 coumarins and 1 other compound were tentatively screened and characterized. Aim: To investigate the chemical constituents of Dioscorea zingiberensis C. H. Wright., Methods: The compounds were isolated by various chromatographic techniques, and the structures of the new steroidal saponins were elucidated by extensive 1D- and 2D-NMR, MS, and IR spectral analysis., Results: The 70% EtOH extract of the rhizomes of Dioscorea zingiberensis afforded two new steroidal saponins, zingiberenosides A (1) and B (2), along with eight known analogues, 3β, 26-dihydroxy-25(R)-furosta-Δ(5, 20(22))-diene-3-O-α-L- rhamnopyranosyl-(1→2)-O-β-D-glucopyranoside (3), methyl parvifloside (4), deltoside (5), methyl deltoside (6), zingiberensis new saponin (7), deltonin (8), progenin III (9) and diosgenin-diglucoside (10)., Conclusion: Two new steroidal saponins were isolated from Dioscorea zingiberensis and their structures determined., (Copyright © 2014 China Pharmaceutical University. Four new steroidal saponins, named ophiopogonin P-S (1-4), together with eleven known ones (5-15) were isolated from the tuberous roots of Ophiopogon japonicus. Their structures were elucidated by spectroscopic and chemical analysis. Compounds 2-15 were evaluated for their cytotoxic activity against five human tumor cell lines (HepG2, HLE, BEL7402, BEL7403 and Hela). Compounds 2, 5, 6, 8 and 9 were cytotoxic for all cell lines tested. Compounds 7, 11 and 15 showed selective cytotoxicity against some of the cell lines. Eleven new triterpene saponins, ilexpublesnins C-M (1-11), along with ten known analogues were isolated from the roots of Ilex pubescens. Their structures were elucidated on the basis of extensive spectroscopic analysis, including 1D and 2D NMR experiments. Compounds 1, 2, 10, and 11 contain a 24-aldehyde, which is rare for triterpene saponins from Ilex. These compounds were evaluated in vitro for their cytotoxic effects on human cancer cell lines HepG2, HLE, BEL7402, BEL7403, BEL7405, MCF-7, and HeLa. A sensitive HPLC-DAD-ESI-MS/MS method was established to screen and identify the polymethoxylated flavonoids (PMFs) in the peels of 'Shatangju' mandarin ( Citrus reticulata Blanco). Eight PMF standards, including four polymethoxylated flavones, two polymethoxylated flavanones, and two polymethoxylated chalcones, were first to be analyzed in positive mode by CID-MS/MS. On the basis of the ESI-MS(n) characteristics of PMFs and the results of EIC-MS/MS experiment, 32 PMFs including 24 flavones and 8 flavanones or chalcones were screened from the complex extract of the peels of 'Shatangju' mandarin. Among them, 10 PMFs were hydroxylated polymethoxyflavonoids (OH-PMFs), and the rest were all permethoxylated PMFs. This was the first systematic report of the presence of PMFs in the peels of 'Shatangju' mandarin, especially for polymethoxylated flavanones and chalcones. Meanwhile, the contents of the three main PMFs and total flavonoids in the peels of 'Shatangju' were determined by HPLC and UV spectrophotometry, respectively. The results indicated that the developed analytical method could be employed as an effective technique for the characterization of PMFs.

Abstract: Two new homoisoflavonoids, named ophiopogonone E (1) and ophiopogonanone H (2), together with thirteen known ones (3–15) were isolated from the tuberous roots of Ophiopogon japonicus. Their structures were elucidated by spectroscopic and chemical analyses. Compounds 7 and 15 were isolated from the genus for the first time. In addition, compounds 2–15 were evaluated for their effects on the inhibition of NO production induced by lipopolysaccharide in the murine microglial cell line BV-2. Compounds 2, 4, 6, 7, 10, 11 showed potent inhibitory effects on NO production with IC50 values of 20.1, 17.0, 7.8, 5.1, 19.2 and 14.4μM respectively. [Copyright &y& Elsevier]

Abstract: A sensitive HPLC-DAD–ESI-MS/MS method was established to screen and identify the polymethoxylated flavonoids (PMFs) in the leaves of Murraya paniculata (L.) Jack. 16 PMF standards were first to be analyzed in positive mode by the CID-MS/MS. For polymethoxylated flavones, the fragments of [M+H−n ×15]+ produced by loss of one or more methyl radicals from the protonated molecule, as well as [M+H−16]+, [M+H−28]+, [M+H−29]+, [M+H−31]+, [M+H−33]+, [M+H−43]+, [M+H−44]+, [M+H−46]+ and [M+H−61]+ fragment ions were detected, which could be taken as their diagnostic characters. For polymethoxylated flavanones and chalcones, their [M+H]+ ions usually underwent RDA cleavage fragmentation of the C-ring prior to the similar loss of diagnostic fragment ions as polymethoxylated flavones, which could be adopted as a shortcut to distinguish them from ordinary flavones rapidly. For the PMF glycosides, the neutral loss of the similar fragments with polymethoxylated flavones from their [aglycone+H]+ could be adopted as a simple method to screen them out from complex mixture. Based on these characterizations of PMFs and the results of EIC-MS/MS experiment, 70 PMFs including 45 flavones, 17 flavanones or chalcones and 8 PMFs glycosides were screened out from the complex extract of the leaves of M. paniculata. Among them, 16 compounds were unambiguously identified by comparison with reference substances.
