Background: Effects on maize were assessed of dual inoculation with arbuscular mycorrhizal fungi (AMF) and dark septate endophytes (DSE) isolated from other plant species., Methods: Suspensions of DSE isolated from Stipa krylovii were prepared at different densities (2, 4, and 8 × 10 5  CFU mL - 1 ) and inoculated separately (AMF or DSE) or together (AMF + DSE), to explore their effects on maize growth., Results: Inoculation with AMF or medium and high densities of DSE and combined inoculation (AMF + DSE) increased plant above-ground growth and altered root morphology. Differences in plant growth were attributable to differences in DSE density, with negative DSE inoculation responsiveness at low density. AMF promoted plant above-ground growth more than DSE and the high density of DSE promoted root development more than AMF. Combined inoculation might lead to synergistic growth effects on maize at low density of DSE and competitive effects at medium and high DSE densities., Conclusions: AMF and DSE co-colonized maize roots and they had positive effects on the host plants depending on DSE density. These findings indicate the optimum maize growth-promoting combination of AMF and DSE density and provide a foundation for further exploration of potentially synergistic mechanisms between AMF and DSE in physiological and ecological effects on host plants., (© 2021. Understanding the diversity and community structure of arbuscular mycorrhizal fungi (AMF) is important for potentially optimizing their role in mining phosphorus (P) in agricultural ecosystems. Here, we conduct a comprehensive study to investigate the vertical distribution of AMF in a calcareous field and their temporal structure in maize-roots with fertilizer P application over a three-year period. The results showed that soil available-P response to P fertilization but maize yields did not. Phosphorus fertilization had no-significant effect on richness of AMF except at greater soil-depths. High P-supply reduced root colonization while optimum-P tended to increase colonization and fungal richness on all sampling occasions. Crop phenology might override P-supply in determining the community composition of active root inhabiting fungi. Significant differences in the community structure of soil AMF were observed between the controls and P treatments in surface soil and the community shift was attributable mainly to available-P, N/P and pH. Vertical distribution was related mainly to soil electrical conductivity and Na content. Our results indicate that the structure of AMF community assemblages is correlated with P fertilization, soil depth and crop phenology. Importantly, phosphorus management must be integrated with other agricultural-practices to ensure the sustainability of agricultural production in salinized soils.

The diversity of arbuscular mycorrhizal fungi (AMF) in sedges on the Tibetan Plateau remains largely unexplored, and their contribution to soil aggregation can be important in understanding the ecological function of AMF in alpine ecosystems. Roots of Kobresia pygmaea C.B. Clarke and Carex pseudofoetida Kük. in alpine Kobresia pastures along an elevational transect (4149-5033 m) on Mount Mila were analysed for AMF diversity. A structural equation model was built to explore the contribution of biotic factors to soil aggregation. Sedges harboured abundant AMF communities covering seven families and some operational taxonomic units are habitat specific. The two plant species hosted similar AMF communities at most altitudes. The relative abundance of the two sedges contributed largely to soil macroaggregates, followed by extraradical mycorrhizal hyphae (EMH) and total glomalin-related soil protein (T-GRSP). The influence of plant richness was mainly due to its indirect influence on T-GRSP and EMH. There was a strong positive correlation between GRSP and soil total carbon and nitrogen. Our results indicate that mycorrhization might not be a major trait leading to niche differentiation of the two co-occurring sedge species. A rhizobox experiment was conducted to investigate degradation of decabromodiphenyl ether (BDE-209) in the rhizosphere of ryegrass and the influence of root colonization with an arbuscular mycorrhizal (AM) fungus. BDE-209 dissipation in soil varied with its proximity to the roots and was enhanced by AM inoculation. A negative correlation (P < 0.001, R(2) = 0.66) was found between the residual BDE-209 concentration in soil and soil microbial biomass estimated as the total phospholipid fatty acids, suggesting a contribution of microbial degradation to BDE-209 dissipation. Twelve and twenty-four lower brominated PBDEs were detected in soil and plant samples, respectively, with a higher proportion of di- through hepta-BDE congeners in the plant tissues than in the soils, indicating the occurrence of BDE-209 debromination in the soil-plant system. AM inoculation increased the levels of lower brominated PBDEs in ryegrass. Plant cells have been reported to play an important role in the uptake of organic contaminants. This study was undertaken to provide an insight into the role of the root cell walls and their subfractions on sorption of phenanthrene to roots of wheat (Triticum aestivum L.). Root cell walls were isolated and further sequentially fractioned by removing pectin, hemicellulose one, and hemicellulose two. They were characterized by elemental analysis, Fourier transform infrared spectroscopy, and solid-state (13)C NMR. Root cell walls had a greater proportion of aromatic carbon and exhibited a lower polarity than the bulk roots. There was a stepwise increase in aromatic carbon content and a decrease in polarity following the sequential fractionation. The sorption affinity of phenanthrene increased gradually following the sequential extraction of root cells. A significant positive correlation between the sorption affinity K(OC) values and the aromatic carbon contents (r(2) = 0.896, p < 0.01) and a negative correlation between the sorption affinity K(OC) values and polarity ((O + N)/C) of root cell fractions (r(2) = 0.920, p < 0.01) were obtained. Improved modeling was achieved for phenanthrene sorption by involving the contribution of root cell walls as a source of root carbohydrates instead of using root lipids alone, which further confirms the significant contribution of root cell walls to phenanthrene sorption on wheat roots. The results provide evidence for the importance of the root cell walls in the partitioning of phenanthrene by plant roots.

Triolein-embedded cellulose acetate membrane (TECAM) was buried in 15 field-contaminated soils in parallel with the cultivation of wheat to predict bioavailability of naphthalene, phenanthrene, pyrene, and benzo[a]pyrene to wheat roots, and the method was compared with chemical extraction methods. Although a good linear relationship was found between PAH concentrations in chemical extractants and wheat roots, the percentage of PAH in soil removed by chemical extraction was much higher than the corresponding percentage removed by wheat roots. In contrast to chemical extraction, a nearly 1:1 relationship was found between the amount of each PAH taken up by TECAMs and wheat roots (r(2) = 0.798-0.925, P < 0.01). Furthermore, the uptake of PAHs by TECAMs and wheat roots had the same pathway of passive transport via the soil solution. Moreover, TECAM caused minimal disturbance to the soil and was easy to deploy. Therefore, TECAM is believed to be a useful tool to predict bioavailability of PAHs to wheat roots grown in contaminated soils.

The interactive impacts of an arbuscular mycorrhizal fungus (AMF; Glomus intraradices) and an earthworm species ( Aporrectodea trapezoides) on N uptake by maize growing during wheat straw decomposition was studied with a modified up-down two-compartment root containers incorporating an air gap system. Soil was sterilized and root compartments inoculated with AMF inoculum in each treatment. Treatments comprised hyphal compartments (HC) with or without AMF hyphae and with or without earthworms. Wheat straw labeled with N was added to the HC. We determined maize yield, N content of shoots and roots, concentration of inorganic N in soil, percent of N transfer, and soil microbial biomass and community composition as indicated by phospholipid fatty acid profiles of HC soil at harvest. Earthworms enhanced N mineralization by 47.7 % from wheat straw compared to the control but did not directly increase N mineralization or transfer N from mycorrhizae to plant. AMF hyphae increased shoot and root N uptake by 117.9 and 87.4 %, respectively. Interaction between the AMF and earthworms increased shoot and root N uptake by 22.0 and 36.2 %, respectively, (compared to hyphae alone) and decreased soil N content by 28.9 % (compared to earthworms alone). The AMF hyphae assisted maize in the uptake of N mineralized from wheat straw by earthworms. Interactions between earthworms and AMF may help plants to take up N mineralized from crop residues. Nutrient and water management is crucially important in shallow-rooted vegetable production systems characterized by high input and high environmental risk. A 2-year field experiment on greenhouse cucumber double-cropping systems examined the effects of root zone nitrogen management and planting of sweet corn as a catch crop in the summer fallow period on cucumber yield and soil Nmin dynamics compared to conventional practices. Cucumber fruit yields were not significantly affected by root zone N management and catch crop planting despite a decrease in N fertilizer application of 53% compared to conventional N management. Soil Nmin content to a depth of 0.9 m decreased markedly and root zone (0–0.3 m) soil Nmin content was maintained at about 200 kg N ha−1. Root zone N management efficiently and directly reduced apparent N losses by 44% and 45% in 2005 and 2006, respectively. Sweet corn, the summer catch crop, depleted Nmin residue in the soil profile of 1.8 m at harvest of winter–spring season cucumber by 304–333 kg N ha−1, which contributed 19–22% reduction in N loss. A pot experiment investigated the response of two maize inbred lines with contrasting root morphology and phosphorus (P) efficiency to inoculation with Glomus mosseae or Glomus etunicatum compared with non-mycorrhizal controls. Soil phosphorus was supplied at rates of 10, 50, and 100 mg P kg -1 soil. Root length, specific root length, and specific phosphorus uptake of maize line 178 (P-efficient) were significantly higher than of line Hc (P-inefficient). Percentage of root length colonized showed the opposite trend regardless of soil P supply level. The two maize lines did not differ significantly in growth response to mycorrhizal colonization. Root colonization rate decreased with increasing soil phosphorus supply. The beneficial effect of the two AM fungi on plant growth and P uptake was greatest at low soil P level and the responses were negative at high P supply. Abstract: A greenhouse pot experiment was conducted to investigate the colonization of alfalfa roots by the arbuscular mycorrhizal (AM) fungus Glomus etunicatum and application of the non-ionic surfactant Triton X-100 on DDT uptake by alfalfa and depletion in soil. Mycorrhizal colonization led to an increase in the accumulation of DDT in roots but a decrease in shoots. The combination of AM inoculation and Triton X-100 application enhanced DDT uptake by both the roots and shoots. Application of Triton X-100 gave much lower residual concentrations of DDT in the bulk soil than in the rhizosphere soil or in the bulk soil without Triton X-100. AM colonization significantly increased bacterial and fungal counts and dehydrogenase activity in the rhizosphere soil. The combined AM inoculation of plants and soil application of surfactant may have potential as a biotechnological approach for the decontamination of soil polluted with DDT. [Copyright &y& Elsevier]

The influence of sulfur (S) supply and mycorrhizal colonization on the growth and pungency of spring onion (Allium fistulosum L.) seedlings grown for four months in Perlite and nutrient solution containing three levels of sulfate [0.1, 1.75, and 4 mM sulfate (SO42-)] were investigated. Plants were inoculated with Glomus etunicatum, Glomus intraradices, or Glomus mosseae, and uninoculated controls were included. Shoot and root dry weights of mycorrhizal and control plants supplied with 4 mM SO42+ were higher than with 0.1 or 1.75 mM SO42-. Enzyme produced pyruvic acid (EPY) and plant sulfur concentrations increased with increasing SO42- supply. The EPY and plant S concentrations were usually higher in mycorrhizal plants than controls irrespective of S supply, and shoot total S concentrations and EPY were strongly correlated. Arbuscular mycorrhizal colonization appeared to make a substantial contribution to the sulfur status of spring onion, and may thus have had a strong influence on its flavor characteristics. Effects of benzo[a]pyrene (B[a]P) on ryegrass ( Lolium perenne L.) growth, plant accumulation and dissipiation of B[a]P in a red sandy soil (Hapli-Udic Argosol) were studied in a pot experiment. The plants were grown for 61 days in soil spiked with B[a]P at 0, 12.5, 25 and 50 mg kg−1. Control pots without plants were also set up. Soil extractable B[a]P, plant shoot and root biomass, and concentrations of B[a]P in plant shoots and roots were determined. Ryegrass biomass was increased by addition of B[a]P and root B[a]P concentrations were significantly correlated with B[a]P application rate, but no such correlation was found for shoot B[a]P concentrations. This indicates that B[a]P enhanced the growth of the ryegrass. The extractable B[a]P concentration in the planted soil was significantly lower than that in the unplanted control soil at the rate of 50 mg B[a]P kg−1. This indicates that ryegrass may help to dissipate B[a]P in soil at concentrations over 50 mg kg−1 soil although the mechanism for this is not understood. [ABSTRACT FROM AUTHOR]

Field observations have indicated that Fe deficiency chlorosis symptoms in peanut are more severe and widespread in monoculture than intercropped with maize in calcareous soils of northern China. Here we report a pot experiment that investigated the mechanisms underlying the marked improvement in Fe nutrition of peanut grown in mixture with maize. Iron deficiency chlorosis occurred in the young leaves of peanut in monoculture and was particularly obvious at the flowering stage, while the young leaves of peanut grown in mixture with maize remained green throughout the experiment. The chlorophyll and HCl-extractable Fe concentrations in young leaves of peanut grown in mixture were much higher than those in monoculture, indicating that maize may have markedly improved the peanut Fe nutrition. Growth in mixture was associated with greatly altered root morphology and microbial populations in the rhizosphere of peanut. Visual observation of peanut roots in monoculture showed that they were larger in diameter and shorter than those in mixture. Moreover, peanut roots in mixture with maize produced more lateral roots and had increased root length compared with plants in monoculture. Peanut grown together with maize showed obvious rhizodermal transfer cells in the subapical root zone, but cells with cell wall ingrowths were poorly developed in peanut in monoculture. Mixed culture resulted in a significantly decreased abundance of bacteria in the rhizosphere of peanut compared with monoculture, and electron microscope observations indicated that this was associated with a thicker mucigel layer on the root surface of peanut in mixture with maize. Iron plaque is ubiquitously formed on the root surfaces of rice. However, little is known about the role of iron plaque in Cd movement from soil to the plant aboveground parts. A pot experiment was conducted to investigate the influence of iron plaque in Cd uptake and accumulation by rice seedlings in soil. Rice seedlings were pre-cultivated in solution culture for 16 days. Two seedlings were transplanted in a nylon bag containing no substrate but surrounded by soil amended with Fe and Cd combined at rates of 0, 1, or 2 g Fe kg−1 and 0, 2.0, or 10 mg Cd kg−1 soil. Fe was added to induce different amounts of iron plaque, and Cd to simulate Cd-polluted soils. Plants were grown for a further 43 days and then harvested. The length of the longest leaf and SPAD values of the newly mature leaves were measured during plant growth. Fe and Cd concentrations were determined in dithionite–citrate–bicarbonate (DCB) soil extracts and in plant roots and shoots. Shoot and root dry weights were significantly affected by Fe supply level but not by added Cd. Root dry weight declined with increasing Fe supply but shoot dry weight decreased at 2 g Fe kg−1 and increased at 1 g Fe kg−1 (except at 2 mg Cd kg−1). The length of the longest leaf and SPAD values of the newly mature leaves were significantly affected by plant growth stage and added Fe and Cd. Fe tended to diminish the negative effect of Cd on these two parameters. Cd concentrations in DCB extracts increased with increasing Cd and Fe supply. In contrast, external Fe supply markedly reduced shoot and root Cd concentrations and there was generally no significant difference between the two Fe supply levels. Shoot and root Cd concentrations increased with increasing Cd addition. Root Cd concentrations were negatively correlated with root Fe concentrations. The proportion of Cd in DCB extracts was significantly lower than in roots or shoots. The results indicate that enhanced Fe uptake by plants can diminish the negative effects of Cd to some extent and that iron plaque on root surfaces is of little significance in affecting uptake and accumulation of Cd by rice plants. Uptake and acropetal translocation of 14 priority polycyclic aromatic hydrocarbons (PAHs) by wheat (Triticum aestivum L.) grown in 15 field-contaminated soils were investigated in a growth chamber. PAH concentrations in roots correlated positively with the corresponding concentrations in soils and negatively with the contents of soil organic carbon (p < 0.01). No clear linear relationship was found between log RCF (root concentration factor, μg g-1 root/μg g-1 soil on dry weight basis) and log Kow of these PAHs. Four-ring PAHs had the highest tendency to be taken up by roots. PAH concentrations in shoots correlated well with their concentrations in soils and roots. Furthermore, distribution profiles of PAHs in shoots were fairly similar to those in soils. Acropetal translocation of 10 PAHs (with log Kow varying from 3.45 to 5.78) was also implicated by Rt (ratio of PAH from root-to-shoot translocation to the total accumulation in shoots) ranging from 53.6 to 72.6%. A negative linear relationship was found between log Rt and log Kow of these PAHs (p < 0.01), and acropetal translocation of PAHs depended on their chemical properties. [ABSTRACT FROM AUTHOR]

Abstract: The effects of an arbuscular mycorrhizal (AM) fungus (Glomus etunicatum) on atrazine dissipation, soil phosphatase and dehydrogenase activities and soil microbial community structure were investigated. A compartmented side-arm (‘cross-pot’) system was used for plant cultivation. Maize was cultivated in the main root compartment and atrazine-contaminated soil was added to the side-arms and between them 650 or 37μm nylon mesh was inserted which allowed mycorrhizal roots or extraradical mycelium to access atrazine in soil in the side-arms. Mycorrhizal roots and extraradical mycelium increased the degradation of atrazine in soil and modified the soil enzyme activities and total soil phospholipid fatty acids (PLFAs). Atrazine declined more and there was greater stimulation of phosphatase and dehydrogenase activities and total PLFAs in soil in the extraradical mycelium compartment than in the mycorrhizal root compartment when the atrazine addition rate to soil was 5.0mg kg−1. Mycelium had a more important influence than mycorrhizal roots on atrazine degradation. However, when the atrazine addition rate was 50.0mg kg−1, atrazine declined more in the mycorrhizal root compartment than in the extraradical mycelium compartment, perhaps due to inhibition of bacterial activity and higher toxicity to AM mycelium by atrazine at higher concentration. • Ground fissures altered root system spatial distribution and endogenous hormone levels. • Mycorrhiza increased fine root percentage in root-damaged plants. • Mycorrhiza may change damaged root system morphology by regulating hormone levels. Ground subsidence caused by coal mining can produce large amounts of surface ground fissures which may damage the surrounding plant roots. Here, a device was used that simulates root damage and we observed the growth of maize roots in real time, the morphological responses of roots and leaves to fissure-induced root damage stress, and the effects of arbuscular mycorrhizal fungi (AMF). Time series of root and leaf growth and hormonal changes in mycorrhizal and control plants with damaged root systems were observed for the first time. Indole-3-acetic acid (IAA) and cytokinin (CTK) levels in roots and leaves decreased significantly one week after ground fissuring. Abscisic acid (ABA) levels increased and the growth of roots and leaves was inhibited. Arbuscular mycorrhizal fungi (AMF) reversed these trends and significantly increased the percentages of root tips and fine roots compared with uninoculated controls. IAA and CTK levels declined significantly in roots and leaves six weeks after ground fissuring but ABA levels remained unchanged. AMF inoculation significantly increased both IAA and CTK levels but decreased ABA levels. The spatial distribution of roots was affected by fissures in addition to the changes in hormones. The negative impacts of ground fissures on maize growth were alleviated by AMF inoculation at harvest with increased shoot and root biomass. The results suggest that the ameliorative effects of AMF on root damage stress may be related to the regulation of hormone levels and an increase in the percentage of fine roots which was conducive to the growth of roots and leaves and increased plant resistance to environmental stress.
