Many studies have investigated the photoprotective and photosynthetic capacity of plant leaves, but few have simultaneously evaluated the dynamic changes of photoprotective capacity and photosynthetic maturation of leaves at different developmental stages. As a result, the process between the decline of photoprotective substances and the onset of photosynthetic maturation during plant leaf development are still poorly understood, and the relationship between them has not been quantitatively described. In this study, the contents of photoprotective substances, photosynthetic pigment content and photosynthetic capacity of leaves at different developmental stages from young leaves to mature leaves were determined by spatio-temporal replacement in eight dominant tree species in subtropical evergreen broadleaved forests. The correlation analysis found that the data sets of anthocyanins, flavonoids, total phenolics and total antioxidant capacity were mainly distributed on one side of the symmetry axis (y = x), while the data sets of flavonoids, total phenolics and total antioxidant capacity were mainly distributed on both sides of the symmetry axis (y = x). In addition, the content of photoprotective substances in plant leaves was significantly negatively correlated with photosynthetic pigment content and photosynthetic capacity but was significantly positively correlated with dark respiration rate (Rd). When chlorophyll accumulated to ~50% of the final value, the photoprotective substance content and Rd of plant leaves reached the lowest level, and anthocyanins disappeared completely; in contrast, the photosynthetic capacity reached the highest level. Our results suggest that anthocyanins mainly play a light-shielding role in the young leaves of most plants in subtropical forests. In addition, 50% chlorophyll accumulation in most plant leaves was the basis for judging leaf photosynthetic maturity. We also believe that 50% chlorophyll accumulation is a critical period in the transition of plant leaves from high photoprotective capacity (high metabolic capacity, low photosynthetic capacity) to low photoprotective capacity (low metabolic capacity, high photosynthetic capacity).

Photoprotection strategies that have evolved in plants to cope with high light (HL) stress provide plants with the ability to resist HL. However, it has not been clearly confirmed which photoprotection strategy is the major HL resistance mechanism. To reveal the major photoprotection mechanism against short-term high light (STHL), the physiological and biochemical responses of three Arabidopsis mutants (Col, chi and ans) under STHL were analyzed in this study. After STHL treatment, the most serious photosynthetic pigment damage was observed in chi plants. At the same time, the degrees of membrane and Rubisco damage in chi was the highest, followed by Col, and ans was the smallest. The results showed that ans with high antioxidant capacity showed higher resistance to STHL treatment than Col containing anthocyanins, while chi with no anthocyanin accumulation and small antioxidant capacity had the lowest resistance. In addition, the gene expression results showed that plants tend to synthesize anthocyanin precursor flavonoids with antioxidant capacity under STHL stress. To further determine the major mechanism of photoprotection under STHL, we also analyzed Arabidopsis lines (Col, CHS1, CHS2 and tt4) that had the same anthocyanin content but different antioxidant capacities. It was found that CHS2 with high antioxidant capacity had higher cell viability, smaller maximal quantum yield of PSII photochemistry (Fv/Fm) reduction and less reactive oxygen species (ROS) accumulation under HL treatment of their mesophyll protoplasts. Therefore, the antioxidant capacity provided by antioxidant substances was the major mechanism of plant photoprotection under STHL treatment.

In eukaryotes, the STRUCTURAL MAINTENANCE OF CHROMOSOME 5/6 (SMC5/6) complex is critical to maintaining chromosomal structures around double-strand breaks (DSBs) in DNA damage repair. However, the recruitment mechanism of this conserved complex at DSBs remains unclear. In this study, using Arabidopsis thaliana as a model, we found that SMC5/6 localization at DSBs is dependent on the protein scaffold containing INVOLVED IN DE NOVO 2 (IDN2), CELL DIVISION CYCLE 5 (CDC5), and ALTERATION/DEFICIENCY IN ACTIVATION 2B (ADA2b), whose recruitment is further mediated by DNA-damage-induced RNAs (diRNAs) generated from DNA regions around DSBs. The physical interactions of protein components including SMC5–ADA2b, ADA2b–CDC5, and CDC5–IDN2 result in formation of the protein scaffold. Further analysis indicated that the DSB localization of IDN2 requires its RNA-binding activity and ARGONAUTE 2 (AGO2), indicating a role for the AGO2–diRNA complex in this process. Given that most of the components in the scaffold are conserved, the mechanism presented here, which connects SMC5/6 recruitment and small RNAs, will improve our understanding of DNA repair mechanisms in eukaryotes.

In the high-light environment, young leaves accumulate anthocyanins as a photoprotection strategy. However, anthocyanin biosynthesis-related enzymes gene sequence is still unknown in the leaves of subtropical forest plants. There are thus few reports on the relationship between the expression level of these genes and photoprotection. In this study, Machilus chinensis and Castanopsis chinensis were taken as plant material in a subtropical forest. Non-full-length nucleotide sequences of chalcone synthase, dihydroflavonol 4-reductase, anthocyanin synthase, and β-tubulin gene were obtained by homologous and electronic cloning. The expression of those genes was verified and analyzed, and some physiological indicators were measured. The anthocyanin content and anthocyanin synthesis-related gene expression in the young leaves of C. chinensis were significantly higher than that of M. chinensis. Although young leaves of M. chinensis did not accumulate anthocyanins, they showed higher antioxidants and nonphotochemical quenching (NPQ). This study indicates that anthocyanins, antioxidants, and NPQ together mediate the positive effects on photoadaptation in young leaves.

Differences in plant shade tolerance constitute a major mechanism driving the succession of forest communities in subtropical forests. However, the indirect effects of differences in light requirements on the growth of mid- and late-successional tree species are unclear, and this potential growth effect has not been explained at the transcriptome level. Here, a typical mid-successional dominant tree species, Schima superba Gardn. et Champ, and a typical late-successional dominant tree species, Cryptocarya concinna Hance were used as materials and planted under 100% full light (FL) and 30% FL (low light, LL) to explore the responses of tree species in different successional stages of subtropical forests to different light environments. Transcriptome sequencing was used to analyze the expression changes in genes related to growth and photoprotection under different light environments. The young leaves of S. superba accumulated more malondialdehyde (MDA) and superoxide radicals (${\mathrm{O}}_2^{{{}^{\bullet}}^{-}}$) under LL. A lower hormone content (auxin, cytokinin, gibberellin) in the young leaves, a weaker photosynthetic capacity in the mature leaves and significant downregulation of related gene expression were also found under LL, which resulted in the total biomass of S. superba under LL being lower than that under FL. The young leaves of C. concinna had less MDA and ${\mathrm{O}}_2^{{{}^{\bullet}}^{-}}$, and a higher hormone contents under LL than those under FL. There was no significant difference in photosynthetic capacity between mature leaves in contrasting light environments. Although the biomass of C. concinna under LL was less than that under FL, the height of C. concinna under LL was higher than that under FL, indicating that C. concinna could grow well under the two light environments. Our results describing the acclimatization of light at the physiological, molecular and transcriptome levels are important for a complete understanding of successional mechanisms.

Light initiates plant photosynthesis, and plant growth depends on light. In order to explore the effects of long-term dark treatment on photosynthesis and leaf growth of plants, this study compared and analyzed the growth state, pigment content, antioxidant capacity, photosynthetic capacity and chlorophyll fluorescence of the leaves of Syzygium rehderianum Merr. and L.M. Perry under full sunlight (natural full sunlight) and a dark treatment. The results showed that the photosynthesis and development of leaves were significantly inhibited after the dark treatment day 3. After the dark treatment day 6, the length and width of the leaves and the synthesis of photosynthetic pigments were significantly lower than those in leaves under full sunlight. Compared with those under full sunlight, the net photosynthetic rate (Pn), maximal quantum yield of PSII photochemistry (Fv/Fm), nonphotochemical quenching and actual photochemical efficiency (Yield) of young and mature leaves were significantly reduced after the dark treatment day 6. These results indicated that the timing of long-term dark treatment caused stress to the leaves. It directly affected the photosynthesis of leaves by affecting the absorption of light energy, inhibiting the synthesis of photosynthetic pigments and destroying the activity of the photosystem II (PSII) reaction center, which ultimately affected the growth and development of plant leaves.

The hybrid rice (Oryza sativa L.) cultivar Peiai64S/E32, which is a hybrid of E32 and Peiai64S, has a super high yield compared with that of its parents. Changes in photosynthetic pigments and chlorophyll (Chl) fluorescence parameters were investigated in the flag leaves during senescence. The content of Chl and carotenoids in the flag leaves of Peiai64S/E32 was more stable than those of their parents during senescence. Furthermore, Peiai64S/E32 had a more stable PSII activity and a lower nonphotochemical quenching and deepoxidation state than that of its parents in the later stage of senescence. These results demonstrated that Peiai64S/E32 functioned longer than its parents did during the leaf senescence and grain-filling period. This occurred because there was more excitation energy to drive photosynthetic CO2 fixation in Peiai64S/E32, as the pigments supplying the energy survived longer in the high-yielding rice compared to the parents.

Low temperature is an important environmental factor that affects plant growth and development. To determine the adaptability of different organs of Paederia scandens (Lour.) Merr. to low temperature, the chlorophyll (Chl) fluorescence parameters as well as the Chl, malondialdehyde (MDA), soluble sugar, and anthocyanin contents of leaves and stems were measured under low temperature. The results confirmed that the Chl fluorescence parameters and Chl content of the leaves and stems of P. scandens tended to decrease consistently, while the MDA content increased, and the change range of the stems was much lower than that of the leaves. The soluble sugar and anthocyanin contents rapidly increased in the leaves and stems to cope with low temperature. Our results suggest that stems are more tolerant than leaves during winter and may continue to grow during that time, which could provide theoretical guidance for clonal propagation of plants to study stem tolerance in the future.

Anthocyanins are water-soluble pigments in plants known for their photoprotective role against photoinhibitory and photooxidative damage under high light (HL). However, it remains unclear whether light-shielding or antioxidant activity plays a major role in the photoprotection exerted by anthocyanins under HL stress. To shed light on this question, we analyzed the physiological and biochemical responses to HL of three Arabidopsis thaliana lines (Col, chi, ans) with different light absorption and antioxidant characteristics. Under HL, ans had the highest antioxidant capacity, followed by Col, and finally chi; Col had the strongest light attenuation capacity, followed by chi, and finally ans. The line ans had weaker physiological activity of chloroplasts and more severe oxidative damage than chi after HL treatment. Col with highest photoprotection of light absorption capacity had highest resistance to HL among the three lines. The line ans with high antioxidant capacity could not compensate for its disadvantages in HL caused by the absence of the light-shielding function of anthocyanins. In addition, the expression level of the Anthocyanin Synthase (ANS) gene was most upregulated after HL treatment, suggesting that the conversion of colorless into colored anthocyanin precursors was necessary under HL. The contribution of anthocyanins to flavonoids, phenols, and antioxidant capacity increased in the late period of HL, suggesting that plants prefer to synthesize red anthocyanins (a group of colored antioxidants) over other colorless antioxidants to cope with HL. These experimental observations indicate that the light attenuation role of anthocyanins is more important than their antioxidant role in photoprotection.

Aims Natural hybridization between invasive and native species, as a form of adaptive evolution, threatens biodiversity worldwide. However, the potential invasive mechanisms of hybrids remain essentially unexplored, especially insights from soil chemical properties and soil microbial communities. Methods In a field experiment, soil microbial community, potassium-solubilizing bacteria, phosphorus-solubilizing bacteria, enzyme activities, and light-saturated photosynthetic rate were measured in invasive Sphagneticola trilobata and its hybrid with native Sphagneticola calendulacea in 2 years. Important Findings In general, soil dissolved organic carbon and the biomass of phosphorus-solubilizing bacteria were significantly higher under the hybrid treatment than S. trilobata and S. calendulacea. However, there were no significant differences in acid phosphatase, total PLFAs, bacterial PLFAs, fungi PLFAs, cellulase, and urase in these treatments. The hybrids had significantly higher light-saturated photosynthetic rate, photosynthetic nitrogen-, phosphorus-, potassium- use efficiencies than the invasive S. trilobata, but no significant difference with S. calendulacea. The total biomass and root biomass of hybrids were higher than S. calendulacea. Our results indicate that the hybrids species have a higher invasive potential than S. calendulacea, which may aggravate the local extinction of S. calendulacea in the future.

Phenotypic plasticity affords invasive plant species the ability to colonize a wide range of habitats, but physiological plasticity of their stems is seldom recognized. Investigation of the stem plasticity of invasive plant species could lead to a better understanding of their invasiveness. We performed pot experiments involving defoliation treatments and isolated culture experiments to determine whether the invasive species Mikania micrantha exhibits greater plasticity in the stems than do three non-invasive species that co-occur in southern China and then explored the mechanism underlying the modification of its stem photosynthesis. Our results showed that the stems of M. micrantha exhibited higher plasticity in terms of either net or gross photosynthetic rate in response to the defoliation treatment. These effects were positively related to an increased stem elongation rate. The enhancement of stem photosynthesis in M. micrantha resulted from the comprehensive action involving increases in the Chl a/b ratio, D1 protein and stomatal aperture, changes in chloroplast morphology and a decrease in anthocyanins. Increased plasticity of stem photosynthesis may improve the survival of M. micrantha under harsh conditions and allow it to rapidly recover from defoliation injuries. Our results highlight that phenotypic plasticity promotes the invasion success of alien plant invaders.

FTL10 is an early-flowering mutant of rice (Oryza sativa L.) with a premature senescent phenotype. Early leaf senescence can cause negative effects on rice yield. Moreover, rice leaves are damaged under high-light conditions, which promote rice senescence. Artificial shading can reduce the amount of light absorbed by rice leaves. The aim of this study was to investigate the effects of shading at noon (11:30–14:00) on the senescence and yield of FTL10. The results showed that shading improved the total antioxidant capacity of rice leaves, reduced the accumulation of reactive oxygen species (ROS) and reduced the expression of genes related to senescence. In the shaded group, the degradation rate of chlorophyll and Rubisco proteins, which are related to photosynthesis, was relatively slow, and the photosynthetic rate was relatively high. Compared with those under the natural growth conditions, the proportion of photosynthetic electron allocated to photorespiration in the shaded group rice leaves was lower, and the proportion allocated to carbon fixation was higher. The yield data showed that the single-spike weight and yield per plant of rice significantly increased after shading. Therefore, our research shows that shading at noon could delay FTL10 senescence and increase yields.

Ascorbic acid (AsA) is an important antioxidant protecting plant against environmental stresses. L-galactono-1,4-lactone dehydrogenase (GLDH) is a key enzyme in AsA synthesis pathway. To investigate the roles of AsA in mitigating high light (HL) damage, Arabidopsis GLDH mutants SALK_060087 and SALK_008236 with altered GLDH-expression were screened for homozygotes. No homozygotes were identified from SALK_060087, but most individuals of the SALK_008236 line (GLDH-236OE) were GLDH-overexpressing homozygous mutants accumulating more AsA than wild type (WT). An investigation of the physiological responses to HL demonstrated that the chlorophyll fluorescence parameters were significantly higher in GLDH-236OE than that in WT after 14-d HL. The degradation of photosynthetic pigment in WT was more severe than that in GLDH-236OE. GLDH-236OE accumulated more AsA, anthocyanins, flavonoids, and phenolics, while WT accumulated more reactive oxygen species (ROS) during HL. Our results suggest that GLDH-236OE have lesser sensitivity and higher tolerance to HL due to a higher capacity to eliminate ROS, absorb extra light, and dissipate thermal energy.

Anthocyanin accumulation is the main factor underlying why young plant leaves turn red, and plant growth follows the principle of maximizing the economic efficiency of energy. There is a need to verify the role of young plant leaves turning red and confirm whether anthocyanin accumulation overconsumes the energy of the plant. We compared the different pigment contents, antioxidant capacities, leaf mass per area, photosynthetic characteristics, dark respiration rates, light compensation points (LCPs) and electron flow distribution of young and mature Schima superba leaves grown under full sunlight and 30% full sunlight conditions. We then examined the correlations between anthocyanins and total antioxidant capacity, photosynthetic pigments, dark respiration rates, LCPs by using linear regression. Finally, we analyzed Pearson correlation coefficients and used principal component analysis to evaluate the interactions of these functional indicators. The young leaves of S. superba accumulated anthocyanins in full sunlight but not in 30% full sunlight. Anthocyanins substantially contributed to the total antioxidant capacity (accounting for 33.29%) in juvenile S. superba leaves. Young leaves containing more anthocyanin accumulated less reactive oxygen species after high light (HL) treatment. However, juvenile leaves accumulating anthocyanins showed 56.97% higher dark respiration rates, 24.79% higher LCPs and 5.32% higher allocations of electron flow to photorespiration and lower organic substance accumulation than did those without anthocyanins. These results suggest that young S. superba leaves must consume more energy to accumulate anthocyanins to adapt to HL environments. Therefore, plants sacrifice growth rate to ensure survival, which is a trade-off for their ability to respond to external environments.

Theoretical and experimental studies have demonstrated that temperature is an important environmental factor that affects the regional distribution of plants. However, how to modify the distribution pattern of plants in different regions is a focus of current research. Obtain the information of cold tolerance genes from cold tolerance species, cloning genes with real cold tolerance effects is one of the most important ways to find the genes related to cold tolerance. In this study, we investigated whether transferring the VHA-c gene from Antarctic notothenioid fishes into Arabidopsis enhances freezing tolerance of Arabidopsis. The physiological response and molecular changes of VHA-c overexpressing pedigree and wildtype Arabidopsis were studied at −20 °C. The results showed that the malondialdehyde (MDA) and membrane leakage rates of WT plants were significantly higher than those of VHA-c8 and VHA-c11 plants, but the soluble sugar, soluble protein, proline and ATP contents of WT plants were significantly lower than those of VHA-c8 and VHA-c11 plants under −20 °C freezing treatment. The survival rate, VHA-c gene expression level and VHA-c protein contents of WT plants were significantly lower than those of VHA-c8 and VHA-c11 plants under −20 °C freezing treatment. Correlation analysis showed that ATP content was significantly negatively correlated with MDA and membrane leakage rate, and positively correlated with soluble sugar, soluble protein and proline content under −20 °C freezing treatment. These results demonstrated that overexpression of the VHA-c gene provided strong freezing tolerance to Arabidopsis by increasing the synthesis of ATP and improved the adaptability of plants in low temperature environment.

In the process of biological invasion, hybridization between invasive species and native species is very common, which may lead to the formation of hybrids with a stronger adaptability. The hybrid of Wedelia trilobata (an alien invasive species) and Wedelia chinensis (an indigenous congener) has been found in South China. In our previous study, we found that the hybrid showed heterosis under cadmium stress. However, the results of this experiment demonstrated that the leaves of the hybrid had no heterosis in controlling water loss. The results showed that the water loss rate of W. trilobata was the slowest, that of W. chinensis was the fastest, and that of the hybrid was in the middle. Compared with W. chinensis and the hybrid, W. trilobata accumulated more abscisic acid (ABA) in leaves to control water loss. After the leaves were detached, W. chinensis leaves suffered the most serious damage, the lowest maximum photochemical efficiency, the most serious membrane lipid peroxidation, and the largest accumulation of malondialdehyde and reactive oxygen species. Compared with W. chinensis and its hybrid, the leaves of W. trilobata could accumulate more antioxidant enzymes and antioxidants, and the total antioxidant capacity was the strongest. The results demonstrate that the ability of the hybrid to reduce water loss was lower than that of W. trilobata, but higher than that of W. chinensis. They showed that the drought resistance of the hybrid may be higher than that of W. chinensis, and it might threaten the survival of W. chinensis.

This study compared the response to methyl viologen (MV)-induced photooxidation in wild-type barley (wt), and both its chlorina f104-nuclear gene mutant (that restricts Chl a and b synthesis) and its f2-nuclear gene mutant (that inhibits all Chl b synthesis). Without MV, the f2 mutant showed the highest sensitivity to high light, with Fv/Fm being reduced by 80% after 80 min of irradiation. There was little difference in response to high light without MV between f104 and wt. After vacuum infiltration with 100 μM MV and exposure to high light, f104 exhibited the highest sensitivity while f2 was the most tolerant to the photooxidation effects. 77K fluorescence spectral analysis indicated that PSII of f104 was especially damaged, as evidenced by the appearance of a new Chl a emission band around 700 nm at the expense of the F685 and F695 bands from the PSII core-inner antenna. With MV, chlorophyll degraded more rapidly in f104 than in either f2 or wt. During MV treatment, zeaxanthin content increased significantly during the initial period of exposure (0–20 min) in all strains, but decreased sharply in f104 after longer exposure time (20–80 min). β-Carotene, on a chlorophyll basis, was not much changed under high light without MV, but with MV it decreased significantly, mostly in f104, intermediately in f2 and least in wt. We conclude that the light-sensitive chlorosis phenotype of f104 is exacerbated by MV-induced photooxidation.

Increasing amounts of experimental evidence show that anthocyanins provide physiological protection to plants under stress. However, the difference in photoprotection mediated by anthocyanins and other photoprotective substances in different seasons is still uncertain. To determine the relationship between anthocyanin accumulation and the photoprotective effects in different seasons, Castanopsis chinensis and Acmena acuminatissima, whose anthocyanin accumulation patterns differ in different seasons, were used as materials to explain how plants adapt to different seasons; as such, their physiological and biochemical responses were analyzed. Young leaves of C. chinensis and A. acuminatissima presented different colors in the different seasons. In summer, the young leaves of C. chinensis were purplish red, while those of A. acuminatissima were light green. In winter, the young leaves of C. chinensis were light green, while those of A. acuminatissima were red. Compared with the young red leaves, the young light green leaves that did not accumulate anthocyanins had higher flavonoid and phenolics contents, total antioxidant capacity, non-photochemical quenching (NPQ), and relative membrane leakage, and a slower recovery rate in the maximum photochemical efficiency (Fv/Fm) after high-light treatment. In addition, the net photosynthesis rate (Pn), transpiration rate (Tr), stomatal conductance (gs), and the effective quantum yield of PSII (ΦPSII) of the young leaves in winter were significantly lower than those in summer, while the activities of catalase (CAT, EC 1.11.1.6), peroxidase (POD, EC 1.11.1.7), and superoxide dismutase (SOD, EC 1.15.1.1) were significantly higher than those in summer. These data indicate that to adapt to seasonal changes anthocyanins, other antioxidative substances and antioxidative enzymes, as well as components involved in the safe dissipation of excitation energy as heat need to cooperate with one another.

Anthocyanins, a protective substance in plant leaves, can accumulate in large quantities under low-temperature induction. In order to explore the effect of anthocyanins in Mikania micrantha leaves, the Rubisco, photosynthesis, pigments, and antioxidative capacity in mature leaves (ML) and young leaves (YL) of M. micrantha were investigated in winter. YL were red on both the adaxial and abaxial surfaces, while ML was red on the abaxial surfaces and green on the adaxial surfaces. Compared with ML, the relative expression of the genes related to anthocyanin synthesis and anthocyanin content were significantly higher in YL. Antioxidants such as flavonoids and total phenols were found in higher quantities, and the total antioxidant capacity was also significantly higher in YL. However, in ML, the Rubisco and chlorophyll content related to photosynthesis were significantly higher. The stomata of ML displayed a larger aperture than YL, and the stomatal conductance and photosynthetic rate were significantly higher in ML. The results suggested that M. micrantha leaves could better adapt to the winter environment through changing the distribution of anthocyanins in leaves of different maturity.

Light adaptability is a major driver of succession and evolution in natural forest plant communities. However, the relationship between plants and light is contradictory; light provides energy for plant growth, but excessive light negatively affects plant growth. To reveal the pattern of plant adaptation to diurnal variation in light, photoprotection strategies were analyzed in young and mature leaves of six dominant species at two successional stages. Mature leaves of both mid- and late-successional dominant species exhibited a higher net photosynthetic rate (Pn) than young leaves. In particular, the mature leaves of the mid-successional tree species had higher high-light tolerance because of a higher Pn, water use efficiency (WUE) and electron transfer rate (ETR). In contrast, the mature leaves of the late-successional tree species had a higher restoration ability after high-light stress because of higher thermal energy dissipation and faster stomatal regulation. Young leaves of both the mid- and late-successional dominant species exhibited a higher photoprotection capability than mature leaves. The young leaves of the mid-successional tree species had higher high-light tolerance because of their highest WUE and anthocyanin/chlorophyll (Anth/Chl) and higher flavonoid/chlorophyll (Flav/Chl), total phenolics/chlorophyll (Phen/Chl) and total antioxidant capacity/chlorophyll (TAC/Chl) ratios. The young leaves of the late-successional tree species had a stronger restoration ability due to highest Flav/Chl, Phen/Chl, and TAC/Chl ratios and a higher moisture content and non-photochemical quenching coefficient (qN, NPQ). This study shows that in a high-light environment, when leaves are at the same developmental stage, mid-successional tree species better tolerate high-light, while late-successional tree species have a greater restoration ability after high-light stress.

Leaf senescence is an important factor that affects crop yield traits and is regulated by various factors. Here, we propose the photo–carbon imbalance hypothesis to explain the mechanism of rice leaf senescence. The main idea of this hypothesis is that carbon assimilation decreases faster than the absorption of light energy in photosynthesis during the late stages of rice growth, which ultimately results in leaf senescence. Our results indicate that endogenous ascorbic acid (Asc) plays an important role in leaf senescence by affecting the expression of senescence genes, thereby influencing photosynthetic capacity and consequently grain yield. The effects of exogenous Asc and methyl jasmonate (MeJA) on photosynthetic capability implied that the balance between photoreaction and carbon assimilation is regulated by exogenous antioxidants or accelerators of senescence. The results of the shading treatments indicated that shading will mitigate the photo–carbon imbalance and improve photosynthetic capacity, resulting in increased yields. Increasing antioxidant concentrations can enhance the reactive oxygen species (ROS) scavenging capacity, whereas shading reduces excess light energy, which may help to restore the photo–carbon balance.

The chlorophyll fluorescence response of desiccation-tolerant cyanobacterial crusts from shaded and non-shaded habitats on sub-tropical inselberg rocks, Guangdong province, China to rehydration under different light intensities and temperature was studied. It was shown that chlorophyll fluorescence change showed the same trend under different light-temperature conditions. After rehydration in the dark following a term of desiccation, the potential quantum yield of PSII (Fv/Fm) and the effective quantum yield of PSII (Yield) of each sample recovered quickly, following activation after 1 minute. Moreover, the two samples could adapt to a wide range of light intensity and temperature while 250 mmol m s light and 30 °C were fitted for rehydration. However, Lower light caused more inhibition in non-shaded habitats, but higher lights caused more inhibition in shaded habitats. Further, samples from non-shaded habitats suffered more damage under temperature treatments than did samples from shaded habitats. Although extreme light and temperature inhibited photosynthetic activity of the samples to some extent, cyanobacterial crusts were protected from damage under high light through photochemical fluorescence quenching (qP). In conclusion, cyanobacteria as pioneers on such bare rocks can recover photosynthesis capability after a term of desiccation, and can adapt well to different light intensities and temperatures.

The content of cytochrome (cyt) bf complex is the main rate-limiting factor that determines light- and CO2-saturated photosynthetic capacity. A study of the half-life of the cyt f content in leaves was conducted whereby Pisum sativum L. plants, grown in moderately high light (HL), were transferred to low light (LL). The cyt f content in fully-expanded leaves decreased steadily over the 2 weeks after the HL-to-LL transfer, whereas control leaves in HL retained their high contents. The difference between the time courses of HL-to-LL plants and control HL plants represents the time course of loss of cyt f content, with a half-life of 1.7 days, which is >3-fold shorter than that reported for tobacco leaves at constant growth irradiance using an RNA interference approach (Hojka et al. 2014). After transfer to LL (16 h photoperiod), pea plants were re-exposed to HL for 0, 1.5 h or 5 h during the otherwise LL photoperiod, but the cyt f content of fully-expanded leaves declined practically at the same rate regardless of whether HL was re-introduced for 0, 1.5 h or 5 h during each 16 h LL photoperiod. It appears that fully-expanded leaves, having matured under HL, were unable to increase their cyt f content when re-introduced to HL. These findings are relevant to any attempts to maintain a high photosynthetic capacity when the growth irradiance is temporarily decreased by shading or overcast weather.

Low temperature (LT) is one of the major factors that limit crop production and reduce yield. To better understand the cold-tolerance mechanism in the plantains, a sensitive cultivar Williams (Musa acuminata AAA cv. Williams) and a tolerant cultivar Cachaco (Musa paradisiaca ABB cv. Dajiao) were used. LT resulted in increased malondialdehyde (MDA) content, elevated contents of hydrogen peroxide (H2O2) and superoxide radical (O 2 ·− ), and decreased photochemical efficiency (Fv/Fm) and net photosynthetic rate (P N), but cv. Cachaco showed better LT tolerance than cv. Williams. After LT treatment for 120 h, total scavenging capability (DPPH· scavenging capability) in Williams showed a significant decrease but no significant alternations was found in Cachaco. Ascorbate peroxidase (APX) and peroxidase (POD) displayed a significant increase but superoxide dismutase (SOD) showed no significant alternations and catalase (CAT) showed a significant decrease in Cachaco after 120 h of LT treatment. All the four antioxidant enzymes above showed a significant decrease in Williams after 120 h of LT treatment. Our results suggest that higher activities of APX, POD, SOD, and DPPH· scavenging capability to a certain extent can be used to explain the higher cold tolerance in the plantain, which would provide a theoretical guidance for bananas production and screening cold-resistant variety.

Alterations in fluidity of thylakoid membranes isolated from spinach chloroplasts in response to sodium bisulfite (NaHSO3), hydrogen peroxide (H2O2), sodium dodecyl sulfate (SDS), bovine serum albumin (BSA), and free linoleic acid (LA) were investigated by means of a fluorescence polarization study with 1,6-diphenyl-1,3,5-hexatriene as the fluorescence probe. A decrease in fluidity and an increase in microviscosity of membrane were caused by NaHSO3 and H2O2 treatment. In contrast, SDS and BSA were found to increase thylakoid membranes fluidity and decrease microviscosity, in which the corresponding correlation coefficients were −0.9995 to −0.9516 (SDS) and −0.9359 (BSA), respectively. No changes in thylakoid membranes fluidity induced by free LA were found until its concentration above 5 mM where the polarization value (P value) declined (increased fluidity). The results suggest that the changes in thylakoids membrane fluidity might depend on the characteristics, mechanism and extent of the interactions between membrane components and compounds added.

Leaf discs of Alocasia macrorrhiza were treated with various stress factors, including two photo-oxidants, methyl viologen (MV) or riboflavin (RB); three pollutants, sodium bisulphite (NaHSO3), or the heavy metals lead or cadmium; or an osmotic medium, polyethylene glycol 6000. The in situ localisation sites for O2 − generation were identified using specific dye nitro blue tetrazolium as a probe. The level of superoxide production was determined by scanning the blue-stained formazan area and was defined as the percentage of pixels from the stained portion versus the total number of pixels in the entire leaf disc area. All stress factors induced the generation of O2 − in a time- or concentration-dependent pattern. Although superoxide production also was enhanced by longer time periods in untreated discs (control), the degree to which this occurred was less than that measured in leaves treated with either MV or RB. Generation sites were primarily found in the chloroplasts of stomatal guard cells and in the plasma membrane of the epidermis and mesophyll cells, indicating that they were most responsive to stress conditions. Nevertheless, the site of O2 − generation varied among these stress factors.

Pigment combinations are regulated during leaf ontogenesis. To better understand pigment function, alterations in chlorophyll, carotenoid and anthocyanin concentrations were investigated during different leaf development stages in six subtropical landscape plants, namely Ixora chinensis Lam, Camellia japonica Linn, Eugenia oleina Wight, Mangifera indica L., Osmanthus fragrans Lowr and Saraca dives Pierre. High concentrations of anthocyanin were associated with reduced chlorophyll in juvenile leaves. As leaves developed, the photosynthetic pigments (chlorophyll and carotenoid) of all six species increased while anthocyanin concentration declined. Chlorophyll fluorescence imaging of ΦPSII (effective quantum yield of PSII) and of NPQ (non-photochemical fluorescence quenching) and determination of electron transport rate-rapid light curve (RLC) showed that maximum ETR (leaf electron transport rate), ΦPSII and the saturation point in RLC increased during leaf development but declined as they aged. Juvenile leaves displayed higher values of NPQ and Car/Chl ratios than leaves at other developmental stages. Leaf reflectance spectra (400–800 nm) were measured to provide an in vivo non-destructive assessment of pigments in leaves during ontogenesis. Four reflectance indices, related to pigment characters, were compared with data obtained quantitatively from biochemical analysis. The results showed that the ARI (anthocyanin reflectance index) was linearly correlated to anthocyanin concentration in juvenile leaves, while a positive correlation of Chl NDI (chlorophyll normalized difference vegetation index) to chlorophyll a concentration was species dependent. Photosynthetic reflectance index was not closely related to Car/Chl ratio, while a structural-independent pigment index was not greatly altered by leaf development or species. Accordingly, it is suggested that the high concentration of anthocyanin, higher NPQ and Car/Chl ratio in juvenile leaves are important functional responses to cope with high radiation when the photosynthetic apparatus is not fully developed. Another two leaf reflectance indices, ARI and Chl NDI, are valuable for in vivo pigment evaluation during leaf development.

The adaptations and responses of photosynthesis to long- and short-term growth light gradient treatments were investigated in five subtropical forest plants, namely Pinus massoniana Lamb., Schima superba Gardn. et Champ., Castanopsis fissa (Champ. ex Benth.) Rehd. et Wils., Acmena acuminatissima (BI.) Merr et Perry, and Cryptocarya concinna Hance. With diurnal changes in sunlight and air temperature, the de-epoxidation state and lutein content in the five woody plants under three light intensities first increased and then decreased during the day. However, maximal photochemical efficiency (Fv/Fm; where Fm is the maximum fluorescence yield and Fv is variable fluorescence) and the photochemical quantum yields of photosystem (PS) II (ΦPSII) of the species examined changed in the opposite manner, with those in plants grown under 100% natural light changing the most. After long-term treatment (21 months), anti-oxidant capacity (1, 1-diphenyl-2-picrylhydrazyl radical (DPPH·)-scavenging capacity) and utilization of excitation energy showed differences in modulation by different light intensities. It was shown that A. acuminatissima and C. concinna, as dominant species in the late succession stage of a subtropical forest in Dinghu mountain, South China, were better able to adapt to different light environments. However, P. massoniana, the pioneer species of this forest, exhibited less adaptation to low light intensity and was definitely eliminated by the forest succession process.

Schima superba and Pinus massoniana distributed over large areas in southern China both are dominant species at Dinghushan Biosphere Reserve. In the present study, the changes of chlorophyll fluorescence and xanthophyll cycle in the leaves of S. superba and P. massoniana exposed to simulated acid rain (SAR) were measured. When exposed to high light, the PSII photochemistry efficiency (F v/F m), efficiency of energy conversion in PSII (ΦPSII) and photochemical quenching (qP) of both S. superba and P. massoniana all decreased when acidity of SAR increased. Regarding non-photochemical quenching (qN), S. superba exposed to SAR had higher value than control plants, but there was no significant difference between the respective seedlings of P. massoniana. As for xanthophyll cycle of the two plant species, the leaves of S. superba exposed to SAR showed a higher content of carotenoids and a higher ability to convert violaxanthin to zeaxanthin than leaves of P. massoniana, which was consistent with S. superba exhibiting a stronger resistance to high light than P. massoniana. Although both species were susceptible to acid rain as shown by our results, P. massoniana was more susceptible compared to S. superba. These results provide an insight into how to protect the forest ecosystem at Dinghushan Biosphere Reserve.

The effects of simulated SO2 treatment on the photosynthetic apparatus were investigated in five subtropical forest plants, namely Pinus massoniana Lamb., Schima superba Gardn. et Champ., Castanopsis fissa (Champ. ex Benth.) Rehd. et Wils., Acmena acuminatissima (Bl.) Merr et Perry, and Cryptocarya concinna Hance. After leaf sections had been immersed in 0, 20, 50, and 100 mmol/L NaHSO3 for 20 h, total chlorophyll (Chl) content, Chl a/b, maximal photochemical efficiency, and the photochemical quantum yields of photo-system II of all five woody plants were reduced to different degrees, whereas lutein content (Chl base) was increased. Two protective mechanisms, namely the xanthophyll cycle (deepoxidation) and an anti-oxidant system (1,1-diphenyl-2-picrylhydrazyl radical-scavenging capacity), showed differences in the degree of modulation under simulated SO2 treatment. Compared with control (distilled water treatment), the revised normalized difference vegetation index, a leaf reflectance index, was lowered with increasing concentrations of NaHSO3. Cryptocarya concinna, a dominant species in the late succession stage of subtropical forests in South China, exhibited less sensitivity to NaHSO3. Conversely, Pinus massoniana, the pioneer heliophyte species, was most susceptible to NaHSO3 treatment. It is suggested that SO2 pollution may accelerate the succession of subtropical forest. The chemical composition, mineral elements, vitamins, free fatty acids and amino acid content of the edible red alga Gracilaria lemaneiformis Bory, grown in the sea near Nan'ao island, Guangdong Province, were analyzed in the present study. Gracilaria lemaneiformis Bory showed a total sugar content of 14.65%. The protein content was 21%, of which approximately 41% was determined to be essential amino acids (EAA). The major amino acid components were glutamic acid, leucine, arginine, and alanine. Of the EAA assayed, methionine and cysteine appeared to be the most limiting amino acids compared with the EAA pattern provided by Food and Agricultural Organization of the United Nations. The total lipids content was 0.87% and comprised a high composition of unsaturated fatty acids (61%), mainly as linoleic acid and oleic acid, and a little amount of polyunsaturated fatty acid; palmitic acid was the main component (39%) of saturated acids. Relatively high levels of vitamin C, iodine, phosphorus, and zinc were also present in G. lemaneiformis. The nutritional composition between G. lemaneiformis and Nostoc flagelliforme, a rare alga that is widely eaten in Chinese society, was compared. The results suggest that N. flagelliforme can be substituted for by G. lemaneiformis, not only because of their similar shape, but also because of their approximate nutritional composition. Gracilaria lemaneiformis may possibly serve as a potential healthy food in human diets in the future. (Managing editor: Ping He)

Two new yellow rice chlorophyll (Chl) b-less (lack) mutants VG28-1 and VG30-5 differ from the other known Chl b-less mutants with larger amounts of soluble protein and ribulose-1,5-bisphosphate carboxylase/oxygenase small sub-unit and smaller amounts of Chl a. We investigated the altered features of Chl-protein complexes and excitation energy distribution in these two mutants, as compared with wild type (WT) rice cv. Zhonghua 11 by using native mild green gel electrophoresis and SDS-PAGE, and 77 K Chl fluorescence in the presence of Mg2+. WT rice revealed five pigment-protein bands and fourteen polypeptides in thylakoid membranes. Two Chl b-less mutants showed only CPI and CPa pigment bands, and contained no 25 and 26 kDa polypeptides, reduced amounts of the 21 kDa polypeptide, but increased quantities of 32, 33, 56, 66, and 19 kDa polypeptides. The enhanced absorption of CPI and CPa and the higher Chl fluorescence emission ratio of F685/F720 were also observed in these mutants. This suggested that the reduction or loss of the antenna LHC1 and LHC2 was compensated by an increment in core component and the capacity to harvest photon energy of photosystem (PS) 1 and PS2, as well as in the fraction of excitation energy distributed to PS2 in the two mutants. 77 K Chl fluorescence spectra of thylakoid membranes showed that the PS1 fluorescence emission was shifted from 730 nm in WT rice to 720 nm in the mutants. The regulation of Mg2+ to excitation energy distribution between the two photosystems was complicated. 10 mM Mg2+ did not affect noticeably the F685/F730 emission ratio of WT thylakoid membranes, but increased the ratio of F685/F720 in the two mutants due to a reduced emission at 685 nm as compared to that at 720 nm.

We compared the responses of wild type (WT) and three mutants including npq1 (lutein-replete and violaxanthin deepoxidase-deficient), lut2 (lutein-deficient), and lut2-npq1 (double mutant) to high irradiance (HI, 2 000 μmol m−2 s−1) at both low (LT, 5 °C) and room (25 °C) temperature. Xanthophyll-dependent energy dissipation was highest in the WT, followed by the lut2, npq1, and npq1-lut2. At 25 °C the relative stress tolerance expressed by Fv/Fm was consistent with the energy dissipation capacity for the first 2 h of treatment. After 3–4 h, the Fv/Fm levels in lut2 and npq1 converged. Under combined LT and HI the relative tolerance sequence was in contrast to the energy dissipation capacity being WT > npq1> lut2 > lut2-npq1. There were little or no significant change in the contents of xanthophylls and carotenes or the chlorophyll (Chl) a/b ratio in any of the materials. Thus lutein (L) substitution possibly alters the conformation/organisation of L binding proteins to enhance damage susceptibility under HI at LT. The enhanced vulnerability is not compensated for the energy dissipation capacity in the lut2 background at LT.

Two yellow rice mutants VG28-1 and VG30-5 were obtained during the tissue culture process from a rice plant (cv. Zhonghua No.11 japonica) with inserted maize Ds transposon element. Absorption spectra and pigment composition showed that two mutants had no chlorophyll (Chl) b and lower Chl a content in comparison to the wild type (WT). Net photosynthetic rate (PN), total electron transport rate (JF), photochemical quenching (qp), quantum yield of PS2 dependent non-cyclic electron transport (ΦPS2), fraction of Prate, and leaf area were lower but Fv/Fm and apparent quantum yield (AQY) remained at similar levels as in the WT plant. Xanthophyll cycle pool size (V+A+Z) on a Chl basis, and de-epoxidation state were enhanced in the mutants. The mutants had larger amounts of soluble protein and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), especially the small subunit of RuBPCO, than WT. The characteristics of two rice mutants differed somewhat from the other common Chl b-less mutants originating from mutagenic agent treatments.

The daily changes in the behavior of xanthophyll cycle and antioxidant systems in flag leaves of superhigh-yield hybrid rice were investigated in relation to various developing stages. Dark-adapted Fv/Fm decreased with the increasing incident light intensity on leaf surface in the morning and then minimized at midday; Deepoxidation State showed an opposed daily pattern to Fv/Fm at different developing stage. As compared with increased deepoxidation state maximum value, the relative content of xanthophyll cycle pigments remained almost constant during development. The daily changes in activities of superoxide dismutase, ascorbate-peroxidase and glutathione reductase and the content of ascorbate and glutathione displayed a similar pattern, where they increased from 8:00 and reached maximum at midday, however, a lower daily fluctuation of superoxide dismutase activity was observed in senescent leaves. The enhanced contribution of xanthophyll cycle and Mehler-ascorbate peroxidase reaction to photoprotection in old leaves could be partially due to the altered leaf posture. In conclusion, daily changes of xanthophyll cycle and antioxidant systems in leaves of rice at various developing stages were dependent on leaf age, leaf angle and intensity of solar irridiance.

In the present study, both electron spin resonance (ESR) and chemical detection confirmed that lutein [extracted from alfalfa (Medicago sativa L.)], the most abundant xanthophyll in thylakoids of chloroplasts, could serve as an antioxidant to scavenge reactive oxygen species (ROS) in vitro. Lutein exhibited a greater capacity for scavenging hydroxyl (OH·) and superoxide (O2·–) radicals than β-carotene at the same concentration, whereas the opposite trend was observed in the capacity for scavenging singlet oxygen (1O2). The capacity of lutein for scavenging ROS from high to low is OH· > O2·– > 1O2. We hypothesise that lutein plays an important photoprotective role in scavenging O2·– and OH· under severe stress. This hypothesis is consistent with our previous report that the lut2 (lutein-deficient) Arabidopsis mutant is more susceptible to damage than the npq1 (lutein-replete but violaxanthin de-epoxidase-deficient) Arabidopsis mutant under severe stress during exposure to high light intensity at low temperature (Peng and Gilmore 2003).
