Glechon spathulata Benth. (Lamiaceae) is a native plant in Brazil popularly known as "mangerona-do-campo". This plant is a perennial sub-shrub with aromatic leaves used in South America as a condiment and in traditional medicine for different purposes, such as inflammations, dyspepsia, diaphoretic, expectorant and antiseptic in catarrhal affections of the respiratory tract, colds, bronchitis and laryngitis. In this study, morpho-anatomical parameters of leaves of this plant were determimed, by macro and microscopic analysis, aiming to help its diagnosis as a pharmaceutical ingredient. The leaves are sub-sessile with spatulate shape and subrevolute margin, finely crenulate in the middle-upper. The uniseriate epidermis shows straight to slightly sinuous cells, diacytic stomata and non-glandular and glandular trichomes. The mesophyll is dorsiventral, displaying an uniseriate palisade parenchyma and a little compact spongy parenchyma, with an average of 4 cell layers. The vascular bundles are collateral closed. There was no presence of crystals. These morphological and anatomical features, when taken together, contribute to quality control of plant leaves of G. spathulata as a pharmaceutical ingredient., Colegio de Farmacéuticos de la Provincia de Buenos Aires

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Plant belonging to the family Rhamnaceae, the Scutia buxifolia is a small tree. In Brazil occurs in Paraná, Santa Catarina and Rio Grande do Sul and in these places, is popularly known as coronilha . By infusing water in the bark and leaves, S. buxifolia it is used as a cardiotonic, antihypertensive and diuretic substance. This study aimed to evaluate the acute and subacute toxicity of the ethyl acetate fraction of the stem bark of S. buxifolia in male and female mice. As methodology guidelines of the Organization for Economic Cooperation and Development (OECD-guidelines 423 and 407). In the analysis of acute toxicity, a single dose of 2000 mg/kg of S. buxifolia was administered orally to both sexes mice. Mortality, biochemical and hematological parameters were evaluated. The study of subacute toxicity, the plant was administered orally to the animals, males and females, at doses of 100, 200 and 400 mg/kg/day for 28 days. Behavioral changes, body weight gain, food intake, biochemical, hematological and histological parameters were analyzed. In acute exposure, no behavioral changes were found. However, caused a decrease in platelet levels in mice and male and female reduction in leukocyte levels only in females. In subacute toxicity study, no behavioral changes were observed in body weight gain and food intake in mice of both sexes. There was a reduction in glucose levels in male mice treated with 200 and 400 mg/kg S. buxifolia and activity of aspartate aminotransferase (AST) was increased in treated male mice with S. buxifolia at the highest dose. In relation hematological parameters, male mice showed a reduction in hemoglobin (HGB) and hematocrit (HCT), when treated with 400 mg/kg of plant fraction. The females showed no change in biochemical and hematological parameters. Histopathological examination of liver tissue showed small changes that were consistent with the biochemical changes observed. Histopathological changes were also observed in the kidneys of animals treated with 400 mg/kg of plant fraction. As the evaluation of acute toxicity by oral administration ethyl acetate fraction from the bark of S. buxifolia of can be classified as safe (category 5), according to the OECD guide. However, the changes observed after subacute administration with high doses of extract, ethyl acetate fraction of the stem bark of S. buxifolia suggest that repeated administration of the plant fraction can cause liver, kidney and hematologic adverse effects. Planta pertencente à família Rhamnaceae, a Scutia buxifolia é uma árvore de pequeno porte. No Brasil ocorre no Paraná, Santa Catarina e Rio Grande do Sul e, nestes locais, é conhecida popularmente como coronilha. Através da infusão em água da casca do tronco e das folhas, a S. buxifolia é utilizada com propriedades hipotensora, diurética e cardiotônica. Este trabalho teve como objetivo avaliar a toxicidade aguda e subaguda da fração acetato de etila da casca do caule de S. buxifolia em camundongos machos e fêmeas. Como metodologia foi utilizada as diretrizes da Organização para a Cooperação e Desenvolvimento Econômico (OCDE diretrizes 423 e 407). Na análise da toxicidade aguda, uma única dose de 2000 mg/kg de S. buxifolia foi administrada oralmente a camundongos de ambos os sexos. Mortalidade, parâmetros bioquímicos e hematológicos foram avaliados. No estudo de toxicidade subaguda, a fração da planta foi administrada oralmente aos animais, machos e fêmeas, em doses de 100, 200 e 400 mg/kg/dia durante 28 dias. Alterações comportamentais, ganho de peso corporal, consumo alimentar e parâmetros bioquímicos, hematológicos e histológicos foram analisados. Na exposição aguda, não foram encontradas alterações comportamentais. Entretanto, causou diminuição nos níveis de plaquetas nos camundongos machos e fêmeas e redução nos níveis de leucócitos, somente nas fêmeas. No estudo de toxicidade subaguda, não foi observada alteração comportamental, no ganho de peso corporal e na ingestão alimentar em camundongos de ambos os sexos. Houve redução nos níveis de glicose nos camundongos machos tratados com 200 e 400 mg/kg de S. buxifolia e a atividade de aspartato aminotransferase (AST) foi aumentada nos camundongos machos tratados com S. buxifolia na dose mais alta. Em relação aos parâmetros hematológicos, os camundongos machos apresentaram uma redução nos níveis de hemoglobina (HGB) e do hematócrito (HCT), quando tratados com 400 mg/kg da fração da planta. As fêmeas não mostraram mudança nos parâmetros bioquímicos e hematológicos. O exame histopatológico do tecido hepático mostrou pequenas alterações que foram consistentes com as variações bioquímicas observadas. Alterações histopatológicas também foram observadas no rim de animais tratados com 400 mg/kg dasfração da planta. Conforme avaliação da toxicidade aguda, a administração por via oral da fração acetato de etila da casca do caule de S. buxifolia pode ser classificada como segura (categoria 5), de acordo com o guia da OCDE. No entanto, as alterações observadas após a administração subaguda com altas doses de extrato, fração acetato de etila, da casca do caule da S. buxifolia, sugerem que a administração repetida da fração dessa planta pode causar efeitos adversos hepáticos, renais e hematológicos.

Fundação de Amparo a Pesquisa do Estado de São Paulo Pithecoctenium echinatum belongs to the family Bignoniaceae, popularly known as comb monkey used in folk medicine for inflammations, rheumatism, skin infections, headaches, cleanses the blood and calming. The objective of this study was to characterize morphological and anatomical fruit of P. echinatum; to determine the physico-chemical quality of the plant drug, isolating chemical compounds of secondary metabolism, as well as to investigate antimicrobial, antioxidant and anti-inflammatory, and analyzing biochemical parameters of liver and kidney toxicity in the crude extract (CE) of seeds of P. echinatum. This species has bicarpelar ovary with the presence of trichomes, crystals and emergencies. It has parietal placentation. The rudiment seminal anatropous, unitegmic and tenuinucellate. The ring nectary has stomatal complexes and trichomes. The epicarp has cutin with the presence of lenticels, estomatíferos and complex emergencies. The outer mesocarp has parenchyma cells, stone cells, sclerified cells and crystals. The endocarp is represented by a single cell layer is not sclerified. The seed is winged and exotestal. The endothelium and the endosperm are the membranaceous embryo. The mesophyll has chemical compounds such as alkaloids, fixed oils, volatile oils, scarce presence of starch and consists of pectic walls. In physico-chemical characterization, using pharmacopoeia tests was used dried vegetable drug, noting that no significant variation of the samples collected in different locations. The hydroethanolic extract 70% seeds of P. echinatum presented secondary metabolic compounds such as steroids and/or triterpenes, flavonoids, alkaloids, anthraquinones, tannins, catechin, phenolic hydroxyl heterosides cardiotonic and volatile oils. Assay polyphenols, flavonoids and tannins were determined by spectroscopic in the CE and fractions. P. echinatum showed antioxidant potential (IC50 131.84 mg/ml) in acetate fraction by the DPPH method and antimicrobial activity to P. aeruginosa, P. zophii and C. albicans by the microdilution method (MIC). From the CE the seeds of P. echinatum we performed the isolation and identification of hesperetin and diometin. These flavonoids were isolated from P. echinatum first. The compounds were determined and analyzed by HPLC / MS and NMR and their spectra compared with the literature. The anti-inflammatory activity of the CE was assayed by the induction of inflammation for cotton rolls and serum taken from rats submitted to the bioassay, in which were administered orally by 300mg/kg/day of CE for 6 consecutive days and compared to nimesulide (5mg/kg/day) and negative control (propylene glycol 20% v/v). The results showed that the CE of P. echinatum showed significant inhibition of inflammation of 24.7%, similar to the inhibition obtained by the group of animals treated with nimesulide 21.4% compared to controls. The hematological parameters showed an increase in the number of leukocytes and decreased of corpuscular medium volume, while AST, ALT and creatinine showed no significant difference in the use of CE compared with the control group propylene glycol. Therefore, the physico-chemical parameters established for P. echinatum enable quality control of drugs of this kind, and this presented antimicrobial, antioxidant and anti-inflammatory activities. Furthermore, no evidence of a hepatotocidade and nephrotoxicity in the experimental conditions tested. Pithecoctenium echinatum pertence à família Bignoniaceae, conhecida popularmente como pente-de-macaco e é utilizada na medicina popular para inflamações, reumatismo, infecções cutâneas, dor de cabeça, depurativo do sangue e calmante. O objetivo deste trabalho foi realizar a caracterização morfo-anatômica do fruto de P. echinatum; determinar parâmetros físico-químicos de qualidade da droga vegetal; isolar compostos químicos do metabolismo secundário, bem como investigar atividade antimicrobiana, antioxidante e anti-inflamatória, além de analisar parâmetros bioquímicos de toxicidade hepática e renal no extrato bruto das sementes de P. echinatum. Esta espécie possui ovário bicarpelar com presença de tricomas, emergências e cristais. Possui placentação parietal. O rudimento seminal é anátropo, unitegumentado e tenuinucelado. O anel nectarífero possui complexos estomáticos e tricomas. O epicarpo é cutinizado com presença de lenticelas, emergências e complexos estomatíferos. O mesocarpo externo possui células parenquimáticas, células pétreas, células esclerificadas e cristais. A semente é alada e exotestal. O endotélio e o endosperma constituem o envoltório membranáceo do embrião. O mesofilo cotiledonar possui compostos químicos como alcaloides, óleos fixos e voláteis, presença escassa de amido e é constituído de paredes pécticas. Na caracterização físico-química, por meio de testes farmacopeicos foi utilizado droga vegetal seca, constatando que não houve variação significativa das amostras coletadas em locais diferentes. O extrato hidroetanólico 70% (EB) das sementes de P. echinatum apresentou compostos do metabolismo secundário como esteróides e/ou triterpenos, flavonoides, alcaloides, antraquinonas, taninos catéquicos, oxidrila fenólica e heterosídeos cardiotônicos e óleos voláteis. Por doseamento espectroscópico foram determinados polifenóis, flavonoides e taninos no EB e frações. P. echinatum apresentou potencial antioxidante (CE50 131,84 μg/ml) na fração acetato pelo método do DPPH e atividade antimicrobiana à P. aeruginosa, P. zophii e C. albicans pelo método de microdiluição em caldo (CIM). A partir do EB das sementes de P. echinatum foi realizado o isolamento e a identificação de dois flavonoides diometina e hesperitina, agliconas flavona e flavanona, respectivamente. Estes flavonoides foram isolados de P. echinatum pela primeira vez. Os compostos foram determinados e analisados por CLAE/MS e RMN e seus espectros comparados com a literatura. A atividade anti-inflamatória do EB foi avaliada pelo método da indução da inflamação por cilindros de algodão, e realizadas dosagens séricas dos ratos submetidos ao bioensaio, nos quais foram administrados a três grupos de ratos, por via oral, 300mg/kg/dia do extrato bruto, 5mg/kg/dia de nimesulida e propilenoglicol 20% v/v, por 6 dias. Os resultados obtidos mostraram que o extrato bruto de P. echinatum apresentou inibição da inflamação significativa de 24,7%, semelhante à inibição obtida pelo grupo de animais tratados com nimesulida de 21,4%, em relação ao grupo controle. Os parâmetros hematológicos mostraram que houve aumento no número de leucócitos e diminuição no volume corpuscular médio (VCM), enquanto que AST, ALT e creatinina não apresentaram diferença significativa no uso do extrato de P. echinatum, quando comparados com o grupo controle propilenoglicol. Portanto, os parâmetros físico-químicos estabelecidos para P. echinatum possibilitam controle de qualidade da droga dessa espécie, e esta, apresentou atividades antimicrobiana, antioxidante e antiinflamatória. Além disso, não foram observados sinais de hepatotocidade e nefrotoxicidade nas condições experimentais testadas.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior The quality of plant-derived drugs must be ensured through tests of authenticity (organoleptic features, macro and microscopic identification), purity and integrity (foreign matter, total ash, acid insoluble ash, loss on drying, microbiological contaminants and heavy metals) and also by qualitative and quantitative determinations of active (s) principle (s) and / or chemical (s) marker (s) characteristic (s) of the species. Many plant species are used in traditional medicine without scientific evidence of its pharmacological properties. The purpose of this work lies in the botanical characterization, physico-chemical and anti-inflammatory activity of Glechon spathulata. Among the macroscopic features of G. spathulata leaves stand out the spatulate form and subrevolute margin, finely crenulate in the middle-upper. Microscopically, the uniseriate epidermis shows straight to slightly sinuous cells, diacytic stomata, and non-glandular and glandular trichomes, dorsiventral mesophyll. Vascular bundles are collateral closed, and no crystals were identified. These features help to the botanical quality control of G. spathulata, and when analyzed in conjunction with its organoleptic properties, allow to ensure the authenticity of the sample. Furthermore, the procedures which measure the extractives content, particle size, volatile oil content, swelling index, bitterness and foaming index, and the chromatographic profile of crude ethanolic extract (CEE), its fractions and essential oil by thin layer chromatography (TLC), allow to verify the authenticity besides to reveal information about purity and integrity of this species. The chromatographic fingerprinting of CEE of G. spathulata by high performance liquid chromatography (HPLC) was carried out in order to determine the nature of the main compounds, and the presumptive identification of the flavonoids rutin and luteolin, also it allowed the verification of the major compound rutin. Besides the authenticity of the extract obtained from aerial parts, the chromatographic fingerprinting of G. spathulata enabled to monitor its quality and to characterize its phenolic compounds. Therefore, the significant anti-inflammatory activity observed by the oral treatment with CEE of G. spathulata in chronic inflammation in rats is probably exerted by polyphenolic compounds. The performed analyses were based on techniques described in literature, official textbooks and Brazilian legislation and were useful in establishing some parameters for the quality control of aerial parts of G. spathulata as raw material for the pharmaceutical and / or food industries. A qualidade dos derivados de drogas vegetais deve ser assegurada através de testes de autenticidade (caracterização organoléptica, identificação macro e microscópica), pureza e integridade (matéria estranha, cinzas totais, cinzas insolúveis em ácido, perda por dessecação, contaminantes microbiológicos e metais pesados), e ainda por determinações qualitativa e quantitativa do (s) princípio (s) ativo (s) e/ou marcador (es) químico (s) característico (s) da espécie. Muitas espécies vegetais são utilizadas na medicina tradicional sem comprovação científica das suas propriedades farmacológicas. O objetivo deste trabalho reside na caracterização botânica, físico-química e atividade anti-inflamatória de Glechon spathulata. Entre as características macroscópicas das folhas de G. spathulata destacam-se a forma espatulada e a margem subrevoluta, crenada no meio-superior. Microscopicamente, destacam-se a epiderme uniestratificada com células retas a pouco sinuosas, estômatos diacíticos, tricomas tectores e glandulares, mesofilo dorsiventral, feixes vasculares do tipo colateral fechado com bainha parenquimática, que na nervura central é incompleta e constituída por esclerênquima, bem como a ausência de cristais. Estas características contribuem no controle botânico de qualidade de G. spathulata, quando analisadas em conjunto com suas propriedades organolépticas, permitem a autenticidade da amostra. Por outro lado, os teores extrativos, a granulometria, conteúdo de óleos voláteis, índices de intumescência, amargor e espuma, bem como o perfil cromatográfico do extrato bruto (EB), suas frações e do óleo essencial através da cromatografia em camada delgada (CCD), além de permitir a autenticidade revelam informações sobre a pureza e integridade desta espécie. O perfil cromatográfico do EB de G. spathulata estabelecido através da cromatografia líquida de alta eficiência (CLAE) permitiu determinar a natureza dos principais compostos presentes, bem como a identificação presuntiva dos compostos flavonóides rutina e luteolina, evidenciando como composto majoritário a rutina. Além da autenticidade do extrato obtido a partir das partes aéreas, o perfil cromatográfico de G. spathulata permite monitorar a sua qualidade e caracterizar os compostos fenólicos presentes, cujas ações farmacológicas são bem descritas na literatura. Desta forma, a significativa atividade anti-inflamatória verificada pelo tratamento via oral com o EB de G. spathulata na inflamação crônica em ratos, é provavelmente exercida pelos compostos polifenólicos presentes. As análises realizadas basearam-se em técnicas descritas na literatura, em compêndios oficiais e na legislação brasileira, e permitiram estabeler parâmetros úteis para o controle de qualidade das partes aéreas de G. spathulata como matéria-prima para a indústria farmacêutica e/ou de alimentos.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior The main groups of secondary metabolites of Glechon spathulata Benth. were evaluated by a phytochemical screening and the volatile oil obtained by hydrodistillation was analyzed by GC-MS. Volatile oil, ethanolic crude extract and hexanic, chloroformic, ethyl acetate and butanolic fractions of G. spathulata were submitted to the lethality test against Artemia salina Leach., to the antimicrobial activity by bioauthographic assay and broth microdilution. Antioxidant activity by DPPH in thin-layer chromatography (TLC) and spectrophotometric assay were also evaluated. Flavonoids, tannins, sterols and/or triterpenoids, saponins, anthocyanic pigments, volatile and fixed acids were found in G. spathulata. The GC of the oil revealed the presence of 9 constituents.
