We report the migration of platinum ligand unit {Pt(en)} 2 (en = ethylenediamine) on a short peptide during collision-induced dissociation fragmentation combined with the characterization of the same species by 2D [ 1 H, 15 N] HSQC (Heteronuclear Single Quantum Coherence) NMR spectroscopy. The NMR spectrum showed that the cysteine is platinated while the MS/MS (Tandem mass spectrometry) showed the platination at glutamic acid. The human immunodeficiency virus (HIV) nucleocapsid protein (NCp7) plays significant roles in the virus life cycle and has been targeted by compounds that could lead to its denaturation or block its interaction with viral RNA. Herein, we describe the interactions of platinum(II) and gold(III) complexes with NCp7 and how the reactivity/affinity of potential inhibitors can be modulated by judicious choice of ligands. The interactions of [MCl(N 3 )] n+ (M = Pt 2+ (n = 1) and Au 3+ (n = 2); N 3 = tridentate chelate ligands: bis(2-pyridylmethyl)methylamine (Mebpma, L 1 ) and bis(2-pyridylmethyl)amine (bpma, L 2 ) with the C-terminal zinc finger of NCp7 (ZF2) were investigated by electrospray ionization-mass spectroscopy (ESI-MS). Mass spectra from the incubation of [MCl(Mebpma)] n+ complexes (PtL 1 and AuL 1 ) with ZF2 indicated that they were more reactive than the previously studied diethylenetriamine-containing analogues [MCl(dien)] n+ . The initial product of reaction of PtL 1 with ZF2 results in loss of all ligands and release of zinc to give the platinated apopeptide {PtF} (F = apopeptide). This is in contrast to the incubation with [PtCl(dien)] + , in which {Pt(dien)}-peptide adducts are observed. Incubation of the Au 3+ complex AuL 1 with ZF2 gave Au x F n+ species (x = 1, 2, 4, F = apopeptide) again with loss of all ligands. Furthermore, the formally substitution-inert analogues [Pt(N 3 )L] 2+ (L = 4-methylpyridine (4-pic), 4-dimethylaminopyridine (dmap), and 9-ethylguanine (9-EtGua)) were prepared to examine stacking interactions with N-acetyltryptophan (N-AcTrp), the Trp-containing ZF2, and the "full" two-finger NCp7 itself using fluorescence quenching titration. Use of bpma and Mebpma gave slightly higher affinity than analogous [Pt(dien)L)] 2+ complexes. The dmap-containing complexes (PtL 1 a and PtL 2 a) had the greatest association constants (K a ) for N-AcTrp and ZF2 peptide. The complex PtL 1 a had the highest K a when compared with other known Pt 2+ analogues: [Pt(dien)(9-EtGua)] 2+ < [Pt(bpma)(9-EtGua)] 2+ < [Pt(dien)(dmap)] 2+ < PtL 2 a < PtL 1 a. A K a value of ca. 40.6 ± 1.0 × 10 3 M -1 was obtained for the full NCp7 peptide with PtL 1 a. In addition, the mass spectrum of the interaction between ZF2 and PtL 1 a confirms formation of a 1:1 PtL 1 a/ZF2 adduct. The reactivity of selected complexes with sulfur-containing amino acid N-acetylcysteine (N-AcCys) was also investigated by 195 Pt and 1 H NMR spectroscopy and ESI-MS. The precursor compounds [PtCl(N 3 )] + PtL 1 and PtL 2 reacted readily, whereas their [Pt(N 3 )L] 2+ analogues PtL 1 a and PtL 2 a were inert to substitution.

cis-DDP presents reactivity towards the transcription factor Sp1-F3, as opposed to previous observations for Sp1-F2. Replacing the ammine ligands with the chelating ethylenediamine increases the reactivity giving a unique dinuclear {Pt(en)}2-bis(cysteine)-bridged product, confirmed by study of the binding sequence ACPECP.

The interactions of polynuclear platinum complexes with human serum albumin were studied. The compounds examined were the "non-covalent" analogs of the trinuclear BBR3464 as well as the dinuclear spermidine-bridged compounds differing in only the presence or absence of a central -NH(2)-(+) (BBR3571 and analogs). Thus, closely-related compounds could be compared. Evidence for pre-association, presumably through electrostatic and hydrogen-bonding, was obtained from fluorescence and circular dichroism spectroscopy and Electrospray Ionization Mass Spectrometry (ESI-MS). In the case of those compounds containing Pt-Cl bonds, further reaction took place presumably through displacement by sulfur nucleophiles. The implications for protein pre-association and plasma stability of polynuclear platinum compounds are discussed.

This paper reports on the chemistry of platinum complexes containing bidentate pyridine-carboxylate (pyAc = pyridin-2-yl-acetate and picEt = pyridine-2-ethylcarboxylate, ethylpicolinate) (N,O) ligands. The pyridine-2-acetate and ethylpicolinate ligands form six- and five-membered chelates, respectively, upon formation of the Pt-carboxylate bond. In all reactions with picEt with various platinum complex starting materials, spontaneous de-esterification of the pendant carboxylate ester occurs to give directly the chelates K[PtCl(2)(pic-N,O)]-trans-[Pt(pic-N,O)(2)] and SP-4,2-[PtCl(pic-N,O)(NH(3))] without any evidence of intermediates. The de-esterification is solvent dependent, and molecular modeling was used to explain this reaction. The reactions of the geometric isomers of [PtCl(pyAc-N,O)(NH(3))] with 5'-guanosine monophosphate, 5'-GMP, and N-acetyl-l-methionine, AcMet, were investigated by NMR spectroscopy. The objective was to ascertain by model chemistry the feasibility of formation of ternary DNA-Pt-protein adducts in biology. Model nucleotide and peptide compounds were formed in situ by chloride displacement giving [PtL(pyAc-N,O)(NH(3))](+) (L = 5'-GMP or AcMet). Competitive reactions were then examined by addition of the complementary ligand L. Sulfur displacement of coordinated 5'-GMP was slow. For SP-4,3-[Pt(AcMet)(NH(3))(PyAc-N,O)](+), a rapid displacement of the sulfur ligand by 5'-GMP was observed, giving SP-4,2-[Pt(5'-GMP-N7)(pyAc-N,O)(NH(3))](+).

Research has been conducted on platinum antitumor compounds which react with sulfur-containing tripeptide glutathione. The authors report that the products of the reactions have been characterized by multinuclear ((super)1 H, (super)1 H- (super)15 N gradient heteronuclear single-quantum coherence, and (super)195 Pt) NMR spectrosccopy and high-performance liquid chromatography.

We have successfully synthesized a 3D-NbO x supported Pt composite as oxygen reduction reaction (ORR) catalyst. 3D-NbO x support is prepared by using KIT-6 hard template, and 3D-NbO x supported Pt composite is prepared by reducing H 2 PtCl 6 and depositing Pt nanoparticles onto 3D-NbO x framework. Physical tests show that 3D-NbO x has an ordered framework, and that in Pt/3D-NbO x composite, ultra fine Pt nanoparticles are distributed along with 3D-NbO x framework. Electrochemical tests indicate that Pt/3D-NbO x -C composite exhibits comparable ORR activity and significantly higher durability compared with Pt/C catalyst. These findings demonstrate that 3D-NbO x is a suitable Pt support, facilitating catalytic performance enhancement. [ABSTRACT FROM AUTHOR]
