This article describes the development of female gametophyte inSedum rupestreL. New embryological information about the processes of megasporogenesis and megagametogenesis provided in this paper expand the current knowledge about the embryology of the studied species.S.rupestreis characterized by monosporic megasporogenesis and the formation ofPolygonum–type embryo sac.The process of megasporogenesis is initiated by one megaspore mother cell, resulting in the formation of a triad of cells after meiosis and cytokinesis. The functional megaspore, which is located chalazally, is a mononuclear cell present next to the megaspore in the centre of the triad. Only one of the two non-functional cells of the triad is binucleate, which occur at the micropylar pole. In this paper, we explain the functional ultrastructure of the female gametophytic cells inS. rupestre. Initially, the cytoplasm of the gametophytic cells does not differ from each other; however, during differentiation, the cells reveal different morphologies. The antipodals and the synergids gradually become organelle-rich and metabolically active. The antipodal cells participate in the absorption and transport of nutrients from the nucellar cells towards the megagametophyte. Their ultrastructure shows the presence of plasmodesmata with electron-dense material, which is characteristic of Crassulaceae, and wall ingrowths in the outer walls. The ultrastructure of synergid cells is characterized by the presence of filiform apparatus and cytoplasm with active dictyosomes, abundant profiles of endoplasmic reticulum and numerous vesicles, which agrees with their main function—the secretion of pollen tube attractants. Reported data can be used to resolve the current taxonomic problems within the genusSedumser.Rupestria.

Yet, information is scarce on their possible involvement in the ovule, a sporophytic organ that hosts the female gametophyte. Apple flowers have a prolonged lapse of time between pollination and fertilization, offering a great system to study the developmental basis of glycoprotein secretion and their putative role during the last stages of the progamic phase and early seed initiation. For this purpose, the sequential pollen tube elongation within the ovary was examined in relation to changes in arabinogalactan proteins (AGPs) in the tissues of the ovule before and after fertilization. To evaluate what of these changes are developmentally regulated, unpollinated and pollinated flowers were compared. AGPs paved the pollen tube pathway in the ovules along the micropylar canal, and the nucellus entrance toward the synergids, which also developmentally accumulated AGPs at the filiform apparatus. Glycoproteins vanished from all these tissues following pollen tube passage, strongly suggesting a role in pollen–ovule interaction. A newly released, late ripening plum cultivar 'Pozna Plava' sets fruit poorly, although it produces high quality fruit. This study aimed to evaluate which factors in the reproductive process could be related to the lack of fruit set. In two consecutive years, establishment of a suitable polleniser and the stage of ovule development at anthesis as well as initial and final fruit set have been studied. In addition to this, the impact made by temperature fluctuations on the interaction between male gametophytes and female sporophytes was also analysed. Growth of the pollen tubes in the style and penetration into the nucellus as well as fruit set were more effective in cross-pollination than in open and self-pollination. A relative delay in ovule development was observed, and most ovules had an embryo sac with eight nuclei. Considering the results of the quantitative parameter study of pollen tube growth in the ovary as well as the results of the stage of ovule development, a conclusion can be made that this cultivar is characterised by an extremely short effective period of pollination.

La reproduction est un événement crucial pour la vie des plantes. Ce processus nécessite la formation du pollen et des ovules. Les cellules germinales vont subir la méiose puis une succession de mitoses, deux pour le pollen et trois pour l’ovule, ce qui va leur permettre d’acquérir leurs structures finales. Une fois formés, ces deux gamètes doivent se rencontrer. Pour cela, le grain de pollen va germer sur le stigmate puis former le tube pollinique. Le tube pollinique en croissance va traverser les différents tissus femelles et ainsi tracter les deux cellules spermatiques jusqu’à l’ovule, permettant la reproduction. Un important réseau de signalisation cellulaire est nécessaire pour permettre ces événements. Les cascades des Mitogen-Activated Protein Kinases (MAPKs) sont l’un des réseaux de signalisation les plus étudiés chez les plantes. Ces kinases sont impliquées dans de très nombreux processus développementaux tels que la formation de l’embryon ou des stomates. Pour autant, leurs rôles restent encore peu caractérisés pendant de la fécondation. Ce projet a pour objectif de mieux comprendre le rôle que jouent les MAPK lors de la formation des gamètes mâles et femelles ainsi que lors de la croissance des tubes polliniques. Plusieurs membres de la superfamille des MAPKs ont été caractérisés pour leurs rôles dans la reproduction sexuée des végétaux. De précédents travaux dans le laboratoire de Daniel P. Matton, ont démontré l’implication de deux MAPK Kinases Kinases (MAP3K), la Solanum chacoense Fertilization-Related Kinase 1 (ScFRK1) et ScFRK2. Ces deux kinases sont nécessaires pour le développement de l’ovule et du pollen chez S. chacoense, une espèce de pomme de terre sauvage diploïde. Dans un premier temps, nous avons étudié la fonction d’une nouvelle ScFRK, la ScFRK3. Ce troisième membre de la classe des FRKs chez S. chacoense, est, elle aussi, impliquée dans le développement des gamétophytes mâles et femelles. Du patron d’expression jusqu’à l’établissement d’une voie de signalisation potentielle, en passant par la caractérisation phénotypique des mutants, plusieurs expériences ont été réalisées dans le but de comprendre le rôle de ScFRK3 lors de la formation des gamètes chez Solanum chacoense. Nous montrons que la ScFRK3 est impliquée dans la formation du pollen ainsi que celui des ovules. Nous avons ensuite poursuivi nos recherches en affinant le phénotypage du mutant de surexpression ScFRK2. En effet, les précédentes études ont permis de montré que la surexpression de ScFRK2 conduit le primordium ovulaire à la formation de structures carpéloïdes. Pour autant, les ensembles des primordius ovulaires ne sont pas devenu des strutures capéloïde. Nous montrons ici que seulement 10 % des ovules dans l'ovaire sont devenu 4 des carpéloïde. Notre étude montre qu’en plus des structures carpéloïdes, un grand nombre d'ovule n'ont pas de sac embryonnaire à l'anthèse ce qui explique le faible nombre de graines par fruit. L’analyse du développement des sacs embryonnaires monte que la surexpression de ScFRK2 entraine l’arrêt au stade mégaspore fonctionnelle. Ce phénotype est similaire à ce qui a pu être observé dans les lignées ARN interférant pour la ScFRK1 et la ScFRK3. De précédentes études faites chez Arabidopsis thaliana semblent montrer que les membres de la superfamille des MAPK ne sont pas essentiels pour la croissance du tube pollinique. Pour comprendre le rôle que jouent les MAPK dans l’élongation du tube pollinique, nous avons utilisé un inhibiteur des MAP Kinase Kinase (MKK), appelé U0126. La présence de cette drogue dans le milieu de croissance des grains de pollen provoque une diminution de la germination et de l’élongation du tube pollinique. L’utilisation de la méthode semi-in vivo montre une perte de la polarisation de la croissance des tubes polliniques causée par l’inhibition des MKK. La présence de l’inhibiteur conduit à la diminution de la quantité de filaments d’actine ainsi qu’à leur désorganisation à l’apex du tube. L’exocytose est aussi affectée par l’inhibition des MKK. Les cascades MAPK sont nécessaires à la croissance polarisée du tube pollinique chez Arabidopsis thaliana. Pour finir, nous avons voulu identifier certains membres de la superfamille des MAPK impliqués dans la croissance du tube pollinique. Nous nous sommes intéressés en premier lieu aux orthologues de la famille ScFRK chez Arabidopsis thaliana. Les AtMAP3K19-20-21 sont les orthologues les plus proches de ScFRK3. Ces AtMAP3K sont exprimées lors du développement des grains de pollen et lors de l’élongation de tube pollinique. L’analyse du pollen des différentes lignées mutantes montre qu’en leur absence, le pollen ne présente aucun problème de développement contrairement à ScFRK3. Par contre, les doubles mutants et le triple mutant pour les AtMAP3K19-20-21 montrent une diminution de la capacité de germination. L’élongation du tube pollinique est affectée lors de la mutation d’au moins une des AtMAP3K. Ces deux études démontrent que les MAPK sont essentielles dans la formation et l’élongation du tube pollinique., Reproduction is a crucial event for plant life. This process requires the formation of pollen and ovules. The germ cells will undergo meiosis and then a succession of mitosis, two for the pollen and three for the ovule, which will allow them to acquire their final structures. Once formed, these two gametes must meet each other. For this, the pollen grain will germinate on the stigmas to form the pollen tube. The growth of the pollen tube will pass through the different female tissues and thus pull the two sperm cells to the ovum for reproduction. An important cellular signaling network is necessary to allow these events to occur. The Mitogen-Activated Protein Kinase (MAPKs) cascades are one of the most studied signaling networks in plants. These kinases are involved in a wide range of developmental processes such as embryo formation and stomata. However, their roles remain poorly characterized during fertilization. The aim of this project is to better understand the role played by MAPKs during the formation of male and female gametes as well as during the growth of pollen tubes. Several members of the MAPK superfamily have been characterized for their role in the sexual reproduction of plants. Previous work in Daniel P. Matton's laboratory has demonstrated the involvement of two MAPK Kinases (MAP3K), Solanum chacoense Fertilization-Related Kinase 1 (ScFRK1) and ScFRK2. These two kinases are necessary for egg and pollen development in S. chacoense, a diploid wild potato species. In a first step, we studied the functionality of a new ScFRK, ScFRK3. This third member of the class of FRKs in S. chacoense, is also involved in the development of male and female gametophytes. From the expression pattern to the establishment of a potential signaling pathway, through the phenotypic characterization of mutants, several experiments have been performed in order to understand the role of ScFRK3 in the formation of gametes in S. chacoense. We show that ScFRK3 is involved in the formation of pollen as well as that of the embryonic sac. We then continued our research by refining the phenotyping of the overexpression mutant ScFRK2. Indeed, previous studies have shown that ScFRK2 overexpression leads the ovular primordium to the formation of carpeloid structures. However, the sets of ovular primordia have not become capeloid structures. We show here that only 10% of the eggs in the ovary have become carpeloid. Our study shows that in addition to the carpeloid structures, a large number of ova do not have an embryonic sac in the anthesis, which explains the low number of seeds per fruit. The analysis of the development of the embryonic sacs shows that 7 overexpression of ScFRK2 leads to the cessation of the functional megaspore stage. This phenotype is similar to what has been observed in interfering RNA lines reducing expression of ScFRK1 and ScFRK3. Previous studies in Arabidopsis thaliana suggest that members of the MAPK superfamily are not essential for pollen tube growth. To understand the role that MAPKs play in pollen tube elongation, we used a MAP Kinase Kinase (MKK) inhibitor called U0126. The presence of this drug in the growth medium of pollen grains causes a decrease in germination and elongation of the pollen tube. The use of the semi in vivo method shows a loss of polarization of the pollen tube growth caused by the inhibition of MKK. The presence of the inhibitor leads to a decrease in the number of actin filaments and their disorganization at the apex of the tube. Exocytosis is also affected by MKK inhibition. We show in this chapter that MAPK cascades are necessary for polarized pollen tube growth in Arabidopsis thaliana. Finally, we wanted to identify some members of the MAPK superfamily involved in pollen tube growth. We were first interested in the ScFRK family orthologs in Arabidopsis thaliana. AtMAP3K19-20-21 are the closest orthologs to ScFRK3. These AtMAP3K are expressed during the development of pollen grains and during the elongation of the pollen tube. Pollen analysis of the different mutant lines shows that in their absence the pollen does not present any development problems unlike ScFRK3. On the other hand, double mutants and triple mutant for AtMAP3K19-20-21 show a decrease in germination capacity. Pollen tube elongation is affected when at least one of the AtMAP3Ks is mutated. These two studies demonstrate that MAPKs are essential for the formation and elongation of the pollen tube and that AtMAP3K19-20-21 participate in these biological processes.

In most angiosperms, the female gametophyte is hidden in the mother tissues and the pollen tube enters the ovule via a micropylar canal. The mother tissues play an essential role in the pollen tube guidance. However, in Utricularia, the female gametophyte surpasses the entire micropylar canal and extends beyond the limit of the integument. The female gametophyte then invades the placenta and a part of the central cell has direct contact with the ovary chamber. To date, information about the role of the placenta and integument in pollen tube guidance in Utricularia, which have extra-ovular female gametophytes, has been lacking. The aim of this study was to evaluate the role of the placenta, central cell and integument in pollen tube pollen tube guidance in Utricularia nelumbifolia Gardner and Utricularia humboldtii R.H. Schomb. by studying the production of arabinogalactan proteins. It was also determined whether the production of the arabinogalactan proteins is dependent on pollination in Utricularia. In both of the examined species, arabinogalactan proteins (AGPs) were observed in the placenta (epidermis and nutritive tissue), ovule (integument, chalaza), and female gametophyte of both pollinated and unpollinated flowers, which means that the production of AGPs is independent of pollination, however, the production of some AGPs was lower after fertilization. There were some differences in the production of AGPs between the examined species. The occurrence of AGPs in the placental epidermis and nutritive tissue suggests that they function as an obturator. The production of some AGPs in the ovular tissues (nucellus, integument) was independent of the presence of a mature embryo sac.

Knowledge on the reproductive biology of cassava, relevant to breeders and molecular geneticists, is still limited. Therefore, different studies were carried out to determine the duration of stigma receptivity and the rate of pollen tube growth. Inflorescences were covered for up to 3 days after the first opening of the bracts (e.g. anthesis day) to prevent open pollination. Results indicate that fruit and seed set are drastically reduced when flowers were covered for 2 or 3 days. However, fruits and seeds were obtained even from flowers that had been covered for 3 days after anthesis, although at low frequency. The rate of pollen tube growth was assessed in many combinations of female and male progenitors crossed through controlled pollinations and collecting the pistils at varying hours after pollination (HAP). Pollen tube growth is fast during the first 6 HAP reaching the tip of the nucellar beak. The growth slows down thereafter, taking 10 additional hours to reach the end of the beak. The growth of pollen tubes slows down even further until they enter the embryo sac. Only 10% of samples showed pollen tubes entering the embryo sac between 48 and 66 HAP. Although several tubes may reach the nucellar beak, only one was observed entering the embryo sac. Results, across the different experiments, were highly variable suggesting that the timeline of fertilization is influenced both by genotypic and environmental factors as well as the manual manipulation of inflorescences and cyathia.

We developed a fertilization method in which pollen tubes entered into embryo sacs without any need to contact surrounding female sporophytic cells by using Torenia fournieri (Torenia) plants under the condition of hindering movement of the virus from a stigma, which is the first infection site leading to systemic infection. When RBDV-infected Torenia pollen grains were used for the developed fertilization method, the virus was transmitted to the seeds by pollen tubes germinating from them. On the other hand, no seeds were infected with the virus when Torenia plants were pollinated with healthy Torenia pollen grains in combination with RBDV-infected raspberry pollen grains, which caused the virus infection in the stigma by penetration of their pollen tubes arrested in its style. Our results indicate that vertical transmission of RBDV by pollen occurs in the transport of the virus into embryo sacs by pollen tubes reaching the embryo sacs.

Background In rice, the pistil is the female reproductive organ, and it consists of two stigmas and an ovary. The stigma is capable of receiving pollen grains and guiding pollen tube growth. The ovary holds the embryo sac, which is fertilized with male gametes to produce seed. However, little is known about the gene function and regulatory networks during these processes in rice. Results Here, using the RNA-Seq technique, we identified 3531 stigma-preferential genes and 703 stigma-specific genes within the rice pistils, and we verified 13 stigma-specific genes via qRT-PCR and in situ hybridization. The GO analysis showed that the transport-, localization-, membrane-, communication-, and pollination-related genes were significantly enriched in the stigma. Additionally, to identify the embryo sac-preferential/specific genes within the pistils, we compared a wild-type ovary with a mutant dst (defective stigma) ovary and found that 385 genes were down-regulated in dst. Among these genes, 122 exhibited an ovary-specific expression pattern and are thought to be embryo sac-preferential/specific genes within the pistils. Most of them were preferentially expressed, while 14 of them were specifically expressed in the pistil. Moreover, the rice homologs of some Arabidopsis embryo sac-specific genes, which played essential roles during sexual reproduction, were down-regulated in dst. Additionally, we identified 102 novel protein-coding genes, and 6 of them exhibited differences between the stigma and ovary in rice as determined using RT-PCR. Conclusions According to these rice ovary comparisons, numerous genes were preferentially or specifically expressed in the stigma, suggesting that they were involved in stigma development or pollination. The GO analysis indicated that a dry rice stigma might primarily perform its function through the cell membrane, which was different from the wet stigma of other species. Moreover, many embryo sac-preferential/specific genes within the pistils were identified and may be expressed in female rice gametophytes, implying that these genes might participate in the process of female gametophyte specialization and fertilization. Therefore, we provide the gene information for investigating the gene function and regulatory networks during female gametophyte development and fertilization. In addition, these novel genes are valuable for the supplementation and perfection of the existing transcriptome in rice, which provides an effective method of detecting novel rice genes. Planar morphogenesis, a distinct feature of multicellular organisms, is crucial for the development of ovule, progenitor of seeds. Both receptor-like kinases (RLKs) such as STRUBBELIG (SUB) and auxin gradient mediated by PIN-FORMED1 (PIN1) play instructive roles in this process. Fine-tuned intercellular communications between different cell layers during ovule development demands dynamic membrane distribution of these cell-surface proteins, presumably through vesicle-mediated sorting. However, the way it’s achieved and the trafficking routes involved are obscure. We report that HAPLESS13 (HAP13)-mediated trafficking of SUB is critical for ovule development. HAP13 encodes the μ subunit of adaptor protein 1 (AP1) that mediates protein sorting at the trans-Golgi network/early endosome (TGN/EE). The HAP13 mutant, hap13-1, is defective in outer integument growth, resulting in exposed nucellus accompanied with impaired pollen tube guidance and reception. SUB is mis-targeted in hap13-1. However, unlike that of PIN2, the distribution of PIN1 is independent of HAP13. Genetic interference of exocytic trafficking at the TGN/EE by specifically downregulating HAP13 phenocopied the defects of hap13-1 in SUB targeting and ovule development, supporting a key role of sporophytically expressed SUB in instructing female gametogenesis., Author Summary Ovules, being the progenitors of seeds, hold an important role in crop production. Ovule is a complex organ connecting two generations, i.e. diploid sporophytic cells and haploid female gametophytes (FG). Fertilization of female gametes leads to the development of embryo and endosperm while the sporophytic cells of ovules become seed coats. Because of its importance for plant reproduction, the development of ovules is tightly controlled. The integument cells are divided in a polar way so that integument cell layers fully enclose a mature FG, leaving an opening only for pollen tube entrance. The integuments also instruct FG development, synchronizing the two generations. Receptor-like kinases, especially STRUBBELIG (SUB), are crucial both for the establishment of ovular polarity and the intercellular communication between sporophytic and gametophytic cells. However, how SUB is post-translationally regulated, as would be expected for cell-surface proteins, is unclear. We report here that the dynamic trafficking of SUB via a subpopulation of endosomes is crucial for its functionality during ovule development. The membrane association of SUB was impaired when the recycling route from the trans-Golgi network/early endosome to the plasma membrane was disrupted in hap13-1, a mutant of HAPLESS13. We further demonstrate that defects of SUB targeting in the outer integuments compromised FG development, suggesting an intriguing intercellular communication. Our results highlight the importance of vesicular trafficking in the establishment of ovular polarity and in the communications between spatially separated cells.

Brachiaria are forage grasses widely cultivated in tropical areas. In vitro pollination was applied to accessions of Brachiaria spp. by placing pollen of non-dehiscent anthers on a solid medium near isolated ovaries. Viability and in vitro germination were tested in order to establish good conditions for pollen development. Comparing sexual to apomictic plants, apomictic pollen has more abortion after meiosis during the microspore stage and a lower viability and, of both types, only some plants have sufficient germination in a high sugar concentration. Using in vitro pollination with the sexual plant, the pollen tube penetrates into the nucellus and micropyle, but the embryo sac degenerates and collapses. In the apomictic B. decumbens, in vitro pollination leads to the transfer of the sperm nuclei into the egg cell and the central cell. The results are discussed according to normal fertilization and barriers in sexual and apomictic plants.

The embryo sac, the female gametophyte of flowering plants, plays important roles in the pollination and fertilization process. Maize (Zea mays L.) is a model monocot, but little is known about the interactions between its embryo sac and the pollen tube. In this study, we compared the transcript profiles of mature embryo sacs, mature embryo sacs 14–16 h after pollination, and mature nucelli. Comparing the transcript profiles of the embryo sac before and after the entry of the pollen tube, we identified 3,467 differentially expressed transcripts (3,382 differentially expressed genes; DEGs). The DEGs were grouped into 22 functional categories. Among the DEGs, 221genes were induced upon the entry of the pollen tube, and many of them encoded proteins involved in RNA binding, processing, and transcription, signaling, miscellaneous enzyme family processes, and lipid metabolism processes. Genes in the DEG dataset were grouped into 17 classes in a gene ontology enrichment analysis. The DEGs included many genes encoding proteins involved in protein amino acid phosphorylation and protein ubiquitination, implying that these processes might play important roles in the embryo sac–pollen tube interaction. Additionally, our analyses indicate that the expression of 112 genes encoding cysteine-rich proteins (CRPs) is induced during pollination and fertilization. The CRPs likely regulate pollen tube guidance and embryo sac development. These results provide important information on the genes involved in the embryo sac–pollen tube interaction in maize.

Sexual reproduction in plants is intimately connected to the activity of the cytoskeletal apparatus in reproductive cells. Because of the ease with which the pollen tube can be studied, it has become a model for studying many aspects of cell physiology related to the cytoskeleton, such as movement of organelles and vesicles and cell division. However, information about cytoskeletal proteins is still insufficient for determining cytoskeletal functions during reproduction, especially in terms of cell-cell interactions. One reason may be that cytological and biochemical research on the cytoskeleton of pollen and the embryo sac has not been complemented by sufficient research activity at genetic and molecular levels, and few laboratories are currently involved in this work. This might be because of problems in identifying appropriate applied applications of the work that might attract more investigation.

Previously considered as toxic by-products of aerobic metabolism, reactive oxygen species (ROS) are emerging as essential signaling molecules in eukaryotes. Recent evidence showed that maintenance of ROS homeostasis during female gametophyte development is crucial for embryo sac patterning and fertilization. Although ROS are exclusively detected in the central cell of mature embryo sacs, the study of mutants deficient in ROS homeostasis suggests that controlled oxidative bursts might take place earlier during gametophyte development. Also, a ROS burst that depends on pollination takes place inside the embryo sac. This oxidative response might be required for pollen tube growth arrest and for sperm cell release. In this mini-review, we will focus on new insights into the role of ROS during female gametophyte development and fertilization. Special focus will be made on the mitochondrial Mn-Superoxide dismutase (MSD1), which has been recently reported to be essential for maintaining ROS homeostasis during embryo sac formation. The definitive version, with the figures, is available at: http://aob.oxfordjournals.org/, Background and Aims: Recent studies of reproductive biology in ancient angiosperm lineages are beginning to shed light on the early evolution of flowering plants, but comparative studies are restricted by fragmented and meagre species representation in these angiosperm clades. In the present study, the progamic phase, from pollination to fertilization, is characterized in Annona cherimola, which is a member of the Annonaceae, the largest extant family among early-divergent angiosperms. Beside interest due to its phylogenetic position, this species is also an ancient crop with a clear niche for expansion in subtropical climates. Methods: The kinetics of the reproductive process was established following controlled pollinations and sequential fixation. Gynoecium anatomy, pollen tube pathway, embryo sac and early post-fertilization events were characterized histochemically. Key Results: A plesiomorphic gynoecium with a semi-open carpel shows a continuous secretory papillar surface along the carpel margins, which run from the stigma down to the obturator in the ovary. The pollen grains germinate in the stigma and compete in the stigma-style interface to reach the narrow secretory area that lines the margins of the semi-open stylar canal and is able to host just one to three pollen tubes. The embryo sac has eight nuclei and is well provisioned with large starch grains that are used during early cellular endosperm development. Conclusions: A plesiomorphic simple gynoecium hosts a simple pollen–pistil interaction, based on a support–control system of pollen tube growth. Support is provided through basipetal secretory activity in the cells that line the pollen tube pathway. Spatial constraints, favouring pollen tube competition, are mediated by a dramatic reduction in the secretory surface available for pollen tube growth at the stigma–style interface. In angiosperms, the female gametophyte has a secluded life; it is protected by several concentric layers that envelop each other. The embryo sac is surrounded by the nucellus, which in turn is wrapped by the integuments forming the ovule, which is nested in the ovary. These wrappings are not hermetic, but contain little ”gates” the pollen tube must traverse on its way towards the embryo sac. Information is emerging that shows that the ovary and ovule provide signals orienting and directimng the pollen tube on the right course. There are three main bodies of evidence supporting this hypothesis. One relates to developmental changes in the female tissues and how they affect pollen tube growth. The second refers to defective ovule mutants, which induce defective pollen tube guidance. And the third relates to the possible molecules involved in this signalling. Here, information gathered along these three main lines of evidence is reviewed. All converge to the conclusion that different checkpoints exist all along the pollen tube pathway. The female gametophyte (embryo sac or megagametophyte) plays a critical role in sexual reproduction of angiosperms. It is the structure that produces the egg cell and central cell which, following fertilization, give rise to the seed's embryo and endosperm, respectively. In addition, the female gametophyte mediates a host of reproductive processes including pollen tube guidance, fertilization, and the induction of seed development. Several major events occur during megagametogenesis, including syncitial nuclear divisions, cellularization, nuclear migration and fusion, and cell death. While these events have been described morphologically, the molecules regulating them in the female gametophyte are largely unknown. We discuss a genetic screen based on reduced seed set and segregation distortion to identify mutations affecting megagametogenesis and female gametophyte function. We report on the isolation of four mutants ( fem1, fem2, fem3 , and fem4 ) and show that the four mutations map to different locations within the genome. Additionally, we show that the fem1 and fem2 mutations affect only the female gametophyte, while the fem3 and fem4 mutations affect both the female and male gametophyte. We analyzed female gametophyte development in these four mutants as well as in the gfa2, gfa3, gfa4, gfa5 , and gfa7 mutants. We found that the fem2, fem3, gfa4 , and gfa5 mutants abort development at the one-nucleate stage, while the fem1, fem4, gfa2, gfa3 , and gfa7 mutants are affected in processes later in development such as polar nuclei fusion and cellularization. The establishment of a genetic screen to identify mutants and the development of a rapid procedure for analyzing mutant phenotypes represent a first step in the isolation of molecules that regulate female gametophyte development and function.

Key message The composition of homogalacturonans (HGs) in the ovule and the female gametophyte cell walls was shown to be rearranged dynamically during sexual reproduction ofH. orientalis. Abstract In angiosperms, homogalacturonans (HGs) play an important role in the interaction between the male gametophyte and the pistil transmitting tract, but little is known about the participation of these molecules at the final stage of the progamic phase and fertilization. The aim of our study was to perform immunocytochemical localization of highly (JIM7 MAb) and weakly (JIM5 MAb) methyl esterified and Ca2+-associated HG (2F4 MAb) in the ovule and female gametophyte cells of Hyacinthus orientalis before and after fertilization. It was found that pollination induced the rearrangement of HG in (1) the micropylar canal of the ovule, (2) the filiform apparatus of the synergids, and (3) the region of fusion between sperm cells and their target cells. Fertilization led to further changes in pectin composition of these three regions of the ovule. A new cell wall was synthesized around the zygote with a characteristic pattern of localization of all examined HG fractions, which we called “sporoderm-like”. The developing endosperm prepared for cellularization by synthesizing highly methyl-esterified HG, which was stored in the cytoplasm. Pollination- and fertilization-induced changes in the composition of the HG in the micropyle of the ovule and the apoplast of female gametophyte cells are discussed in the context of: (1) micropylar pollen tube guidance, (2) preparation of the egg cell and the central cells for fusion with sperm cells, and (3) the polyspermy block.

Calreticulin expression is upregulated during sexual reproduction of Hyacinthus orientalis , and the protein is localized both in the cytoplasm and a highly specialized cell wall within the female gametophyte. Several evidences indicate calreticulin (CRT) as an important calcium (Ca2+)-binding protein that is involved in the generative reproduction of higher plants, including both pre-fertilization and post-fertilization events. Because CRT is able to bind and sequester exchangeable Ca2+, it can serve as a mobile intracellular store of easily releasable Ca2+ and control its local cytosolic concentrations in the embryo sac. This phenomenon seems to be essential during the late progamic phase, gamete fusion, and early embryogenesis. In this report, we demonstrate the differential expression of CRT within Hyacinthus female gametophyte cells before and during anthesis, during the late progamic phase when the pollen tube enters the embryo sac, and at the moment of fertilization and zygote/early endosperm activation. CRT mRNA and the protein localize mainly to the endoplasmic reticulum (ER) and Golgi compartments of the cells, which are involved in sexual reproduction events, such as those in sister synergids, the egg cell, the central cell, zygote and the developing endosperm. Additionally, immunogold research demonstrates selective CRT distribution in the filiform apparatus (FA), a highly specific component of the synergid cell wall. In the light of our previous data showing the total transcriptional activity of the Hyacinthus female gametophyte and the results presented here, we discuss the possible functions of CRT with respect to the critical role of Ca2+ homeostasis during key events of sexual plant reproduction. Moreover, we propose that the elevated expression of CRT within the female gametophyte is a universal phenomenon in the cells involved in double fertilization in higher plants.
