Polychlorinated naphthalenes (PCNs) are toxic and can accumulate through the food chain. PCNs have been detected in different categories of foods. Intake of animal-derived foods is an important pathway for human exposure to PCNs. However, information on PCNs in meat from farmed animals is scarce. In this study, PCNs were assessed in beef sourced from local markets in Beijing and six provinces in China. The mean PCN concentrations in beef samples from the seven regions varied from 41.2 to 88.7 pg/g wet weight (ww). The homologue profiles of PCNs in the specimens were similar, with tri- and di-CNs being dominant. The mean concentration of PCNs in the flank (74.7 pg/g ww) was higher than that in the round (58.2 pg/g ww) or shank (53.6 pg/g ww), likely because the former contained a higher proportion of lipids than the latter. Significantly different PCN distributions in beef and dairy cow milk were identified using machine learning. The toxic equivalencies (TEQs) of PCNs in all beef samples ranged from 0.0003 to 0.022 pg TEQ/g ww. PCNs contributed to approximately 4.6% of the total TEQ values of polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans, polychlorinated biphenyls, and PCNs in the beef. Polychlorinated biphenyls (PCBs) can be metabolized into hydroxylated PCBs (OH-PCBs) that exhibit greater toxicity than their parent compounds. In particular, 2,2',4,5,5'-pentachlorobiphenyl (PCB 101) is commonly found in chicken feeds and breeding environments, although information on the biotransformation of this PCB in chickens is lacking. In this study, the hydroxylation metabolization of PCB 101 was assessed based on in vitro trials with Sanhuang chicken liver microsomes and in vivo experiments with Hy-Line Brown hens. The para -substituted metabolite 4'-OH-PCB 101 is the predominant metabolite of PCB 101. 4'-OH-PCB 101 is preferentially retained in the chicken bloodstream and partly distributed into different tissues of laying hens. The blood-brain barrier can effectively prevent the OH-PCB from entering the brain, and the adipose tissue contains a relatively low residue concentration of the OH-PCB. The laying hen can deplete the OH-PCB via laying eggs and excrement. The ratio of 4'-OH-PCB 101/PCB 101 in egg yolk is about 1:2. These results first provide definite evidence for the previous hypothesis of the PCB 101 metabolism by chickens. They could assist in predicting the environmental fate of PCBs, and in the risk assessment of PCBs and OH-PCBs in chicken-based foodstuffs.

A controlled feeding experiment was conducted to investigate the toxicokinetic of polychlorinated dibenzo-p-dioxins/dibenzofurans (PCDD/Fs) in laying hens. The laying hens were fed with fly ash contaminated diets to replicate the typical environment-feed-chicken transfer chain in China. Levels of PCDD/Fs rapidly increased in the pectoralis, adipose tissue, liver and blood of laying hens with daily ingestion of dioxin-associated diets during the 14-days exposure period, and then a gradual decrease was observed in the 28-days depletion period. The depletion rates (k d ) of the toxic equivalent of PCDD/Fs (TEQ PCDD/Fs ) were 0.043, 0.031 and 0.030 day -1 for pectoralis, liver and adipose tissue in the high-exposure group, respectively. The k d of individual PCDD/Fs in liver increased with the numbers of chlorine and n-octanol/water partition coefficient (logK OW ), indicating that lower chlorinated congeners had higher half-lives in liver. Decreasing ratios of liver to adipose tissue for PCDD/Fs (L/AT) throughout the experiment suggested a tendency of equilibrium partitioning between liver and adipose tissue. Congener-specific sequestration of PCDD/Fs in liver was revealed by the positive correlation between L/AT ratios and logK OW . Physiological bioconcentration factors of PCDD/Fs were estimated at the end of exposure, indicating the preferential accumulation of hexachlorinated congeners in most tissues. Updated assessment on transfer of polychlorinated dibenzo-p-dioxins and furans (PCDD/Fs) and dioxin-like polychlorinated biphenyls (DL-PCBs) from feed to food is essential for understanding human exposure risk. A controlled feeding experiment was conducted for laying hens to reevaluate the transfer characteristics of dioxin-like compounds from feed to egg. Two fortified diets (1.17 and 5.13 pg TEQ g -1 dry weight), made by blending with fly ash, were fed to laying hens for 14 days, followed by 28-days depletion. Levels of ∑TEQ PCDD/Fs+DL-PCBs in eggs rapidly increased once exposure started, reaching at 1.98 and 6.86 pg TEQ g -1 lw at the end of exposure for low- and high-exposure groups, respectively, and dropped to under the European legislation (maximum levels: 5.0 pg TEQ g -1 lw) after 28-days depletion. The quantitative depletions showed that the half-lives (T 1/2 ) of ∑TEQ PCDD/Fs in eggs were 23 and 14 days for low- and high-exposure groups, respectively. The depletion and accumulation rates of PCDD/Fs were in the range of 0.026-0.151 and 0.005-0.016 day -1 , respectively, representing that the T 1/2 of PCDD/Fs in eggs ranged from 5 to 27 days. The depletion kinetics of DL-PCBs was not significant in egg. The hens with higher laying rates exhibited shorter T 1/2 of PCDD/Fs, implying that increasing laying rate could expedite the depletion of PCDD/Fs in egg. The T 1/2 of PCDD/Fs in egg were negatively correlated with the chlorine number, indicating that lower chlorinated congeners tended to be retained in the egg. Transfer rates of PCDD/Fs were in the range of 4-19%, which were lower than the previous results. These results were attributed to short exposure time and low bioavailability of PCDD/Fs in fly ash. Polychlorinated biphenyls (PCBs) and dibenzo-p-dioxins and furans (PCDD/Fs) are notorious persistent organic pollutants (POPs), which may bioaccumulate through food chain and play detrimental effects to organisms even at trace levels. Quantification of PCBs and PCDD/Fs in biotic samples is a great challenge. In the present study, gas chromatography coupled to triple quadrupole mass spectrometry with an atmospheric pressure chemical ionization source (GC-(APCI)MS/MS) was studied for the isotope-dilution analysis of PCBs (mono-to deca-) and PCDD/Fs in Chinese mitten crab food webs. High-resolution mass spectrometry (HRMS) was applied for comparison. Light PCBs are compared between the two instruments for the first time. After optimization of instrument parameters, the RSDs of relative response factors of calibration curves were between 3.4% and 15.5% for PCBs and 1.7%-7.9% for PCDD/Fs. The limits of detection were between 0.021 and 0.150 pg/mL for PCBs and 0.051-0.237 pg/mL for PCDD/Fs. PCB concentrations in crab food web samples detected by GC-(APCI)MS/MS were well correlated with those detected by HRGC/HRMS. A DiCB, 3,3'-dichlorobiphenyl (PCB11), was the dominant PCB congener in aquatic food webs. Other MoCB and DiCB congeners were also widely identified; hence, low-weight PCB congeners should arouse more concern in the future. The presence of polychlorinated biphenyls (PCBs) in feed materials has caused great public concern because PCBs can accumulate in farmed animals, be transferred farm-to-fork and, ultimately, have a detrimental impact on human health. Recently, the occurrence of PCBs in marine environments has garnered scientific attention due to their high levels and potential reproductive threats to marine apex predators. Fishmeal is marine-origin feed material and is susceptible to PCB contamination from the aquatic trophic chain. The present study collected 102 fishmeal samples during 2012-2017 from major global fishmeal production areas (United States, Europe, China, South America and Southeast Asia). The levels of PCBs (26 congeners) were between 0.4 and 19.9 ng g -1 dw (mean: 1.94 ng g -1 dw), with a 75.3% contribution from indicator PCBs on a weight basis. Together with PCDD/Fs, 4.9% of fishmeal exceeded the maximum levels set by the European Commission for dioxin-like compounds (4.0 pg WHO-TEQ/g). The highest PCB levels were found in fishmeal from the U.S. (6.85 ng g -1 dw), which was nearly five times higher than the other four sampling areas. No clear time trends were found for PCBs in fishmeal during the sampling period. Predicted PCB concentrations in farmed fish via fishmeal consumption were between 1.24 and 2.76 ng g -1 dw, which was comparable to PCBs in market fish. When compared to other emerging POPs in the same batches of fishmeal, PCBs were still found to be an alarming class of POPs. Concentrations and patterns of 75 PCN congeners in feed raw materials of animal and plant origin were investigated. Six types of feed raw materials of animal origin and three types of feed raw materials of plant origin from China were collected in 2016. The total concentrations of PCNs in the collected materials ranged from 147 to 1009 ng kg-1, with the highest occurring in fish meal. The mean PCNs concentration in feed raw materials of animal origin (551 ng kg-1) was higher than in those of plant origin (294 ng kg-1). Additionally, lower chlorinated PCNs were the main homologues in raw feed materials, while Di-CNs were the predominant homologues in all samples (mean: 53%), followed by tri-CNs (mean: 28%). The most abundant congeners were CN5/7 and 24/14. Additionally, the toxicity equivalencies (TEQs) of PCNs in the feed raw materials ranged from 0.010 to 0.046 ng TEQ kg-1, with the highest TEQ concentrations of PCNs detected in gluten meal. Together, CN5/7, 66/67, 65/70, and 73 contributed approximately 64% of the total PCN TEQs in raw feed materials. Concentrations of polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and polychlorinated biphenyls (PCBs) in the feed raw materials were detected to compare the TEQ distribution of those dioxin-like compounds. Size-fractionated samples of urban particulate matter (PM; ≤1.0, 1.0-2.5, 2.5-10, and >10 μm) and gaseous samples were simultaneously obtained to study the distribution of polychlorinated biphenyls (PCBs) in the atmosphere in Beijing, China. Most recent investigations focused on the analysis of gaseous PCBs, and much less attention has been paid to the occurrence of PCBs among different PM fractions. In the present study, the gas-particle partitioning and size-specific distribution of PCBs in atmosphere were investigated. The total concentrations (gas + particle phase fractions) of Σ 12 dioxin-like PCBs, Σ 7 indicator PCBs, and ΣPCBs were 1.68, 42.1, and 345 pg/m 3 , respectively. PCBs were predominantly in the gas phase (86.8-99.0 % of the total concentrations). The gas-particle partition coefficients (K p ) of PCBs were found to be a significant linear correlated with the subcooled liquid vapor pressures (P L 0 ) (R 2  = 0.83, P < 0.01). The slope (m r ) implied that the gas-particle partitioning of PCBs was affected both by the mechanisms of adsorption and absorption. In addition, the concentrations of PCBs increased as the particle size decreased (>10, 2.5-10, 1.0-2.5, and ≤1.0 μm), with most of the PCBs contained in the fraction of ≤1.0 μm (53.4 % of the total particulate concentrations). Tetra-CBs were the main homolog in the air samples in the gas phase and PM fractions, followed by tri-CBs. This work will contribute to the knowledge of PCBs among different PM fractions and fill the gap of the size distribution of particle-bound dioxin-like PCBs in the air.

The concentrations of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), dioxin-like polychlorinated biphenyls (dl-PCBs), and polychlorinated naphthalenes (PCNs) were determined in workplace air from eight secondary nonferrous metal processing plants to investigate occupational exposure to these toxic compounds. The total estimated daily intakes of PCDD/Fs and dl-PCBs for workers by inhalation in the workplace were in the range of 0.15-9.91 and 0.13-8.59 pg of WHO-TEQ/kg of body weight (bw) for moderate and light activities, respectively. The daily inhalation doses for workers in the workplaces of three investigated plants exceeded the tolerable daily intake recommended by the World Health Organization. These results indicate that the risk of occupational exposure to dioxins by inhalation in the workplace of plants investigated was considerably high. For PCNs, the daily inhalation doses for workers in the workplace were in the range of 0.005-4.46 and 0.004-3.87 pg of TEQ/kg of bw for moderate and light activities, respectively, which were lower than those of dioxins. To identify the source of PCDD/Fs, PCBs, and PCNs in workplace air, their homologue profiles were compared with those in stack gas from the plants investigated. It was found that significant dioxin contamination in workplace air was mainly attributed to the emission of fugitive gas from smelting furnaces during reclamation processes.

In this study, field measurements were conducted to estimate and characterize the atmospheric emission levels and profiles of polychlorinated biphenyls (PCBs) from multiple industrial thermal processes. The emission levels and profiles of PCBs from five types of thermal processes at twenty-three plants were studied and compared with eight processes reported in our previous studies. Correlation analysis was preformed to identify a marker congener for emission of ΣPCB. A significant correlation was observed between congener CB-118 and ΣPCB (R(2)=0.65 and p<0.01), which suggests that CB-118 is a good marker congener for emission of ΣPCB. The profiles of PCBs emitted from the thirteen thermal processes were compared, and this information could be used for studying source-receptor relationships and identifying the specific sources of PCBs. To prioritize the sources for control, the concentrations of PCBs from thirteen industrial thermal sources were compared. The PCB concentrations from secondary zinc smelting and thermal wire reclamation were about one to three order magnitude higher than those of other sources, which suggests that these two sources be given priority in PCB source control. Finally, the atmospheric emission factors of PCBs from the thirteen industrial sources were summarized, and these data will be useful for developing an integrated emission inventory of PCBs., (Copyright © 2012 Elsevier Ltd. Polychlorinated naphthalenes (PCNs) are candidates for inclusion in the Stockholm Convention on persistent organic pollutants. PCNs are structurally and toxicologically similar to 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) and its analogues. Intake in food is considered to be an important human exposure pathway for PCNs. In this preliminary study, cheese and butter samples were analysed for PCNs, polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and polychlorinated biphenyls (PCBs) using an isotope dilution gas chromatography high-resolution mass spectrometry method. The aim of this study was to evaluate the PCN concentrations in the cheese and butter samples and to compare them with the PCDD, PCDF and PCB concentrations. The PCN concentrations were 5.6–103 pg g−1of wet weight in the seven cheese samples tested and 5.0–199 pg g−1of wet weight in the seven butter samples tested. The mass concentrations of lower chlorinated congeners were greater than those of the higher chlorinated congeners. Congeners of CN45/36, CN27/30 and CN33/34/37 were much more abundant than other congeners found in tetrachlorinated PCNs. Congeners of CN51, CN66/67 and CN73 were determined to be the predominant congeners in penta-, hexa- and heptachlorinated homologs, respectively. The PCNs contributed around 5% of the total PCN, PCDD, PCDF and PCB toxic equivalence (TEQ) values. CN73 was found to be the dominant PCN congener and contributed more than 40% to the PCN TEQ value. Congeners CN66/67, CN69 and CN63 were also found at relatively high levels. The PCB congener CB118 was the predominant congener (by mass-based concentration) of the 12 dioxin-like PCBs (dl-PCBs). The PCBs contributed 53.8% of the total TEQ, and congener CB126 contributed more than any other compound that was analysed to the total TEQ. Abstract: The formation and distribution of dioxin-like polychlorinated biphenyls (dl-PCBs) during cooking was investigated. Concentrations of dl-PCBs in liquid residues, cooked beef, and oil fumes generated during heating were determined by isotope dilution HRGC/HRMS. Although the levels of dl-PCBs in well-done beef were lower compared with those of raw beef, relatively high concentrations of dl-PCBs were detected in the oil fumes produced during heating. This suggests that dl-PCBs in raw beef may have volatilized into the oil fumes during the cooking process. Sucralose and chloropropanols contained in raw materials may have resulted in increased dl-PCB concentrations and the level of toxic equivalents (TEQ) in the oil fumes produced under high temperature during cooking. Concentrations of dl-PCBs did not vary greatly in cooked beef, except when higher levels of chloropropanols were presented in the uncooked raw materials. Results indicate that sucralose and chloropropanols may promote the formation of dl-PCBs during the cooking process. The newly produced dl-PCBs from raw beef cooking were mainly present in oil fumes, which gave rise to high levels of TEQ in oil fumes. [Copyright &y& Elsevier]

The evaluation of toxicity related to polychlorinated dibenzo- p -dioxins and furans (PCDD/Fs) and dioxin-like polychlorinated biphenyls (DL-PCBs) is crucial for a comprehensive risk assessment in real-world exposure scenarios. This study employed a controlled feeding experiment to investigate the metabolic effects of dioxin-like compounds (DLCs) on laying hens via feed exposure. Diets enriched with two concentrations (1.17 and 5.13 pg toxic equivalents (TEQ)/g dry weight (dw)) were administered over 14 days, followed by 28 days of clean feed. Metabolomics analyses of blood samples revealed significant metabolic variations between PCDD/Fs and DL-PCBs exposed groups and controls, reflecting the induced metabolic disruption. Distinct changes were observed in sphingosine, palmitoleic acid, linoleate, linolenic acid, taurocholic acid, indole acrylic acid, and dibutyl phthalate levels, implying possible connections between PCDD/Fs and DL-PCBs toxic effects and energy-neuronal imbalances, along with lipid accumulation and anomalous amino acid metabolism, impacting taurine metabolism. Moreover, we identified three differential endogenous metabolites—L-tryptophan, indole-3-acetaldehyde, and indole acrylic acid—as potential ligands for the aryl hydrocarbon receptor (AhR), suggesting their role in mediating PCDD/Fs and DL-PCBs toxicity. This comprehensive investigation provides novel insights into the metabolic alterations induced by PCDD/Fs and DL-PCBs in laying hens, thereby enhancing our ability to assess risks associated with their exposure in human populations.
