The  Escherichia coli  ŽP strain (ŽP) was constructed based on the known probiotic  E. coli  strain Nissle 1917. It was genetically modified to carry the colicin E7 synthesis gene encoding DNase on a conjugative plasmid and the colicin E7 immunity gene in the chromosome. The aim of this study was to evaluate the effects of the daily ŽP per oral administration (5 × 10 8 or 5 × 10 10 CFU per bird) on the growth performance, hematological, biochemical, histological parameters, gut microbiota, and nonspecific immunity of the 4-24 days old broilers. The ŽP administration increased the abundance of genera Bacillus, Butyrivibrio , and Clostridium  and did not influence the weight gain of 4-16 days old broilers. The biochemical parameters were within normal ranges for poultry in experimental and control groups. The ŽP administration had no effect on the erythrocyte numbers, hemoglobin and immunoglobulin Y concentrations, but significantly increased the serum lysozyme concentration, leukocyte numbers, and reactive oxygen species production by phagocytes compared with the control group. It did not cause inflammatory changes in intestinal mucosa, Peyer's patches, and spleen. Thus, the ŽP had no detrimental effects on broiler health and could be an efficient probiotic for the broiler colibacillosis prophylaxis.

A probiotic is considered a live microbial feed supplement that has beneficial effects on the host. In this study, the probiotic property by which Enterococcus faecium HS-08 strengthens the immune system was investigated. Using a murine model, we evaluated the abilities of this strain to increase intestinal short-chain fatty acid contents and to induce the production of mucosal immunoglobulin A (IgA), which are crucial for mucosal immune systems. Various amounts (0%, 0.0038%, 0.038%, or 0.38%) of strain HS-08 cells were administered to BALB/cAJcl mice, which resulted in a dose-dependent increase of fecal IgA levels. A qRT-PCR analysis of Peyer's patch cells revealed that the gene expression of retinal-dehydrogenase, interleukin 6, B-cell-activating factor, and a proliferation-inducing ligand were increased, which leads to IgA secretion via a T-cell-independent mechanism. The administration of 0.038% and 0.38% of strain HS-08 cells also increased fecal acetate levels, which plays an important role for maintaining immune functions. This cecal floral analysis and the stability of strain HS-08 against gastrointestinal digestion suggest that this strain can inhabit the host intestine. In conclusion, the administration of E. faecium HS-08 increased intestinal acetate levels and enhanced IgA secretion, which may result in strengthening of the mucosal immune system.

Much is made of the need to translate scientific research into improved care of people or other life forms. Grant applications invariably start with the extent of a problem and end by claiming that their work will or could result in making an impact. In truth, very few projects ever lead to translation at the level of the host, nor was that really their intent. For those who are focused on applied science, there are many ways to reach the desired goal, sometimes through serendipity or by logical stepwise progress. The following paper will provide personal insight into the stages, pitfalls, and ultimate assessment of relevance in the context of using probiotic lactobacilli for human health and other applications.

The gut microbiota is increasingly considered as a symbiotic partner in the maintenance of good health. Metagenomic approaches could help to discover how the complex gut microbial ecosystem participates in the control of the host's brain development and function, and could be relevant for future therapeutic developments, such as probiotics, prebiotics and nutritional approaches for psychiatric disorders. Previous reviews focused on the effects of microbiota on the central nervous system in in vitro and animal studies. Objective: To define the terms of use of vaccines, probiotics, and cranberry in urology., Materials and Methods: A literature search was conducted on MEDLINE for all these treatments used in urology. Products for bacterial interference such as probiotics are also used, their properties are described. Actinomycetes, as the main producers of a variety of active metabolites and enzymes, have received extensive attention in aquaculture. An actinomycete had been isolated from Haizhou Bay and been identified by morphological analysis and 16S rRNA sequencing. Results showed that the actinomycete had the highest similarity (99.65%) with Nocardiopsis sp. and was named strain E10. Strain E10 produced an exo-enzyme with proteinase effect that showed strong antibacterial activity against Vibrio anguillarum. This strain E10 was tested as a probiotic for the culture of Exopalaemon carinicauda. After having been fed with a diet supplemented with strain E10 for 30 days, the final body weight (Wt), weight gain rate (WGR), special growth rate (SGR), survival rate (SR) and digestive enzymes (chymotrypsin, α -amylase and lipase) of E. carinicauda in all experimental groups showed no significant differences (P > 0.05). However, the antioxidant enzymes (total superoxide dismutase, catalase and peroxidase) and the non-specific immune-related enzymes (acid phosphatase, alkaline phosphatase and lysozyme) in the treatment groups increased significantly (P < 0.05) compared with the control group. The cumulative mortality of the treatment groups (35.56% in 5 × 109 CFU/100 g) was significantly lower than that of the control treatment (73.33%) (P < 0.05) post challenge with V. anguillarum for 14 days. The results indicated that strain E10 had no significant effect on the growth, but a significant improvement on the immunity of E. carinicauda. The present study aims to evaluate the influence of a hypercaloric diet (HC) diet on the inflammatory and metabolic parameters of mice and the protective effect of probiotics, as well as the possible effects on the offspring. The control groups (C, CP, and CPP) received the normocaloric diet – Nuvilab Quimtia® – 3.86 kcal/kg. The hypercalorie groups (HC, HCP and HCPP) received the diet accordingly – PragSoluções® – 5.98 kcal/kg. The control pregnant probiotic (CPP) and the hypercaloric pregnant probiotic group (HCPP) received the corresponding diet plus probiotics during pregnancy. We carried out metabolic and biometric parameters, inflammation markers, and histological analyses of the liver. An increase in the following parameters was found in the F0: adiposity, leptin, energy efficiency coefficient, lower insulin decay rate, and hepatic lipidosis. Interestingly, there was no deposition of fat in the liver of the group fed with hypercaloric died and treated with probiotics, thus not differing from the control-diet group. Maternal overeating promoted altered metabolic parameters of both dams and offspring, demonstrating the metabolic effects of a hypercaloric diet on the F1; conversely, probiotics had beneficial effects in the F1. Deleterious effects of a fructose- and lipid-rich diet may be transmitted to the F1; however, probiotic supplementation seems to counteract these consequences. [ABSTRACT FROM AUTHOR]

The aim of this work was to characterize the functional properties of Enterococcus faecium strains identified after isolation from human faeces. Of these isolates, strain R13 showed the best resistance to low pH, bile salts, and survival in the simulated in vitro digestion assay, and demonstrated an important level of adhesion to hexadecane as a potential probiotic candidate. Analysis of the antibiotic resistance of E. faecium strains indicated that in general these isolates were sensitive to the tested antibiotics and no strain appeared to be resistant to vancomycin. Examination of the virulence determinants for E. faecium strains demonstrated that all strains contained the virulence genes common in gut- and food-originated enterococci, and strain R13 harboured the lowest number of virulence genes. Additionally, no strain contained the genes related to cytolysin metabolism and showed hemolytic activity. The antimicrobial role of E. faecium strains was tested against several pathogens, in which different levels of inhibitory effects were observed, and strain R13 was inhibitory to all tested pathogens. PCR screening of genes encoding enterocin A and B indicated the presence of these genes in E. faecium strains. Preliminary characterization of bacteriocins revealed that their activity was lost after proteolytic enzyme treatments, but no alteration in antimicrobial activity was observed at different pHs (3.5 to 9.5) and after heat treatments. In conclusion, this study revealed the functional characteristics of E. faecium R13 as a gut isolate, and this strain could be developed as a new probiotic after further tests. In the present study, we examined the adherence of indigenous vaginal bacteria, probiotic strains, and metritis pathogens to mucus collected from different parts of the reproductive tracts of heifers and cows and compared their adherence with the bacterial adherence to mucus collected from the stomach and large intestine of pigs. Most of the vaginal strains adhered to mucus collected from different parts of the reproductive tract and strongly adhered to gastric mucus, with the exception of Lactobacillus buchneri 24S8. Only Lactobacillus mucosae 29S8, Enterococcus faecium E21, and E. faecium EAC adhered to colonic mucus. Probiotic strains adhered strongly to mucus collected from the reproductive tract and gastric mucus but did not adhere to colonic mucus. Pathogenic strains were adherent to vaginal, uterine horn, and gastric mucus, except Escherichia coli O8:K88ab:H9 (65), Fusobacterium necrophorum, and Gardnerella vaginalis, which adhered to uterine cervix mucus. Only Kocuria kristinae and G. vaginalis adhered to uterine body mucus; E. coli O149:K88ac (EC) adhered to colonic mucus. The strains did not exhibit host specificity but rather strain specificity. The ability to adhere to mucus was a characteristic unique to each strain. To our knowledge, this is the first report regarding in vitro adherence of GRAS (Generally Regarded As Safe) lactobacilli isolated from different sources to mucus collected from different parts of the reproductive tract. Next-generation sequencing is revealing the complex interactive networks of host-bacteria interactions, as it is now possible to screen in detail the microbiota harbored by a host. This study investigated the influence of a probiotic treatment on the survival and microbiota of brook charr ( Salvelinus fontinalis), focusing on its disturbance of the natural microbiota (dysbiosis). The results indicated that an indigenous probiotic strain (identified as Rhodococcus sp.) colonized neither the fish skin mucus nor the water following the probiotic treatment. Instead, the probiotic strain was detected only in the biofilm of the test tank. Nevertheless, a substantial beneficial effect of the probiotic treatment was observed: the population of the pathogen Flavobacterium psychrophilum decreased in the treated tank water. This study clearly shows that the indigenous strain chosen for the probiotic treatment did not disturb the natural fish skin mucus microbiota but acted directly through the production system to control the growth of the pathogen and, as a consequence, to enhance fish survival. The canine-derived strain Lactobacillus fermentum CCM 7421 has been demonstrated to exert certain health benefits as a probiotic in dogs. Synbiotic combinations are widely used but are rarely studied in dogs. In this study the prebiotic inulin in combination with L. fermentum CCM 7421 was tested for its effects on faecal microbial populations, faecal characteristics, and blood biochemistry in canine experiments. Healthy adult dogs ( n = 36) were randomly assigned to 1 of 3 treatment groups (12 dogs/treatment): ( i) the control group (C), ( ii) the probiotic group (LF group: L. fermentum CCM 7421, 108 CFU/mL of Ringer buffer, 0.1 mL/kg of body mass), and ( iii) the synbiotic group (LF+I group: L. fermentum CCM 7421 + inulin (I; Raftifeed IPS) added as 1% of diet). The experiment lasted for 7 weeks with a 2-week treatment period. We detected a significant increase of lactic acid bacteria (LF versus C, day 7; LF versus C and LF versus LF+I, days 28 and 49), a decrease of clostridia (LF versus C, day 14), a lower pH value (LF versus LF+I, day 28), and a higher ammonia concentration (LF versus LF+I, days 14 and 49) in faecal samples. The synbiotic LF+I combination did not intensify the probiotic L. fermentum CCM 7421 efficacy, but its slight laxative effect can be useful to prevent constipation, e.g., in senior dogs. The taxonomic position and biological activities of two Bacillus strains used in veterinary probiotics were studied in this work. These microorganisms inhibit growth of a broad spectrum of pathogenic cultures. They synthesize proteolytic enzymes and other biologically active metabolites, and to some extent, supplement each other with probiotic activities. It is not clear whether these versatile activities are properties of individual strains or bacterial taxa as whole. 16S rRNA comparisons were conducted and illustrated the relatedness of these strains to Bacillus amyloliquefaciens. Their cell wall fatty acid contents were consequently analysed and specified a relation to the ' Bacillus velezensis' ecomorph. On account of the previous observations, a simple method of 16S rRNA profiling by polymorphic nucleotides was proposed to determine a group of organisms closely related to ' B. velezensis' and B. amyloliquefaciens subsp. plantarum, for they are biologically active strains suitable for use in biotechnology. A bacteriocin producer strain MC13 was isolated from the gut of (grey mullet) and identified as . The bacteriocin of E. faecium MC13 was purified to homogeneity, as confirmed by Tricine sodium dodecyl sulphate - polyacrylamide gel electrophoresis (SDS-PAGE). Reverse-phase high-performance liquid chromatography (HPLC) analysis showed a single active fraction eluted at 26 min, and matrix-assisted laser desorption ionization time of flight (MALDI-TOF) mass spectrometry analysis showed the molecular mass to be 2.148 kDa. The clear zone in native PAGE corresponding to enterocin MC13 band further substantiated its molecular mass. A dialyzed sample (semicrude preparation) of enterocin MC13 was broad spectrum in its action and inhibited important seafood-borne pathogens: , , and . This antibacterial substance was sensitive to proteolytic enzymes: trypsin, protease, and chymotrypsin but insensitive to catalase and lipase, confirming that inhibition was due to the proteinaceous molecule, i.e., bacteriocin, and not due to hydrogen peroxide. Enterocin MC13 tolerated heat treatment (up to 90 °C for 20 min). Enterococcus faecium MC13 was effective in bile salt tolerance, acid tolerance, and adhesion to the HT-29 cell line. These properties reveal the potential of E. faecium MC13 to be a probiotic bacterium. Enterococcus faecium MC13 could be used as potential fish probiotic against pathogens such as V. parahaemolyticus, , and in fisheries. Strains of Butyrivibrio are principal cellulytic bacteria in the rumen of the High Arctic Svalbard reindeer (). According to phylogenetic analysis based on 16S rRNA gene sequencing, Butyrivibrio can be divided into three subgroups within the Clostridia class of the phylum Firmicutes, but the current phenotypic and genotypic differentiation within the family Lachnospiraceae is insufficient. This current study describes the sequence diversity of the 16S-23S rRNA intergenic transcribed spacer (ITS) region of Butyrivibrio isolates from reindeer. A total of 17 different ITS sequences with sizes between 449 and 784 nt were obtained. Genes encoding tRNAIle and tRNAAla were identified in four of the sequences. Phylogenetic neighbor-joining trees were constructed based on the ITS sequence and compared with a phylogenetic neighbor-joining tree based on 16S rRNA gene sequences previously obtained for the same isolates. These comparisons indicated a better differentiation between strains in the ITS sequence than the 16S rRNA gene based tree. Through this study, a better means for identifying and tracking fibrolytic and potentially probiotic Butyrivibrio strains in reindeer and other ruminants has been provided. [ABSTRACT FROM AUTHOR]

Abstract: The intestinal microbiota displays dramatic changes in the ICU patient, with a loss of the entire beneficial lactic bacterial flora. In situations of hyperinflammation followed by immune paresis that are due to the acute disease but also to chronic diseases and drugs used, it is crucial to reestablish inflammatory and immune homeostasis, and microbiota manipulations with probiotics or synbiotics is an instrument of choice. It is not recommended to give any ICU patient any probiotic or synbiotic, for fear of inefficacy or even increased mortality. However, there is strong evidence in elective surgery, polytrauma and acute pancreatitis for well-defined formulations of bacterial strains and fibers. The inflammatory response is immediate and therefore administration of probiotics or synbiotics in these situations must also be immediate. [Copyright &y& Elsevier]

The discovery of several healthy beneficial effects of the consumption of dairy products fermented with some bacterial strains led to the investigation of the functional properties of these microorganisms. One of the most studied properties is the cholesterol-lowering activity of bacteria with probiotic characteristics, mostly isolated from human gut. In this work, eight Lactobacillus plantarum and five Lactobacillus paracasei strains isolated from cheese were studied in vitro for their cholesterol-lowering action and their acid and bile tolerance. The ability of these strains to remove cholesterol was assessed in de Man, Rogosa and Sharpe (MRS) medium, supplied with cholesterol, and in ultra-high temperature (UHT) whole homogenized milk. Among all tested strains, two L. plantarum and three L. paracasei strains gave rise to a significant reduction of the cholesterol level inMRSbroth; in particular, L. plantarum strains lowered the cholesterol content by an average of 19.4%, whereas L. paracasei strains lowered by an average of 6.8%. The two L. plantarum strains possessing the highest cholesterol-lowering activity in MRS broth were also tested in milk. Results showed that L. plantarum strains maintained this activity because after 24 h the cholesterol decrease ranged from about 5.0% to 8.2% without significant variations between the two strains. Results from the binding assay suggested that cholesterol was mainly removed through the adsorption on the cell wall. Data from acid and bile tolerance assays showed that the L. plantarum dairy isolates were able to maintain viability at pH 2 and to grow in a medium with bile salts, and therefore were regarded as probiotics or dairy starters for new probiotic or functional food production. Bacterial vaginosis (BV) is the most prevalent vaginal infection worldwide and is characterized by depletion of the indigenous lactobacilli. Antimicrobial therapy is often ineffective. We hypothesized that probiotic Lactobacillus rhamnosus GR-1 and Lactobacillus reuteri RC-14 might provide an adjunct to antimicrobial treatment and improve cure rates. Sixty-four Brazilian women diagnosed with BV were randomly assigned to receive a single dose of tinidazole (2 g) supplemented with either 2 placebo capsules or 2 capsules containing L. rhamnosus GR-1 and L. reuteri RC-14 every morning for the following 4 weeks. At the end of treatment (day 28), the probiotic group had a significantly higher cure rate of BV (87.5%) than the placebo group (50.0%) (p = 0.001). In addition, according to the Gram-stain Nugent score, more women were assessed with “normal” vaginal microbiota in the probiotic group (75.0% vs. 34.4% in the placebo group; p = 0.011). The objectives of this study were to demonstrate the gastro-intestinal tolerance and the capacity to modulate the intestinal microbiota of some Lactobacillus acidophilus, L. casei and L. rhamnosus strains with high probiotic potential. L. acidophilus and L. casei mixture that is used to produce probiotic fermented milk has been evaluated for its acid resistance and bile salts tolerance and compared to other lactic acid bacteria (LAB). The commercial culture and the three strains of L. rhamnosus exhibited complete tolerance to acid for pH ⩾ 2.5. The minimal inhibitory concentration of the bile salts mixture was 50 g·L -1 for all bacteria. The impact of the ingestion of the novel probiotic on the fecal microbiota was evaluated in vivo using healthy C57Bl/6 mice. Fecal samples were analyzed for the microbiota enumeration using selective plating. Fecal analysis showed an increase of total culturable LAB and a decrease in Staphylococcus spp. population in the LAB-treated mice indicating that these cultures could improve the intestinal health. Also, reduction in fecal Enterobacteriaceae was noticed following mice gavage with L. rhamnosus ATCC 9595 while a higher enumeration was measured for L. rhamnosus RW-9595M, an exopolysaccharideoverproducing mutant. Consumption of dairy products containing viable probiotic strains has increased dramatically in recent years, owing to general health claims associated therewith. This trend has boosted diversification of the portfolio of said products, including whey cheese matrices. However, taking into account the relatively poor organoleptic and textural features of these matrices, improvement is in order via incorporation of selected additives, provided that viability of the strains is duly assayed. Lactobacillus paracasei LAFTI® L26 was accordingly incorporated into whey protein solid matrices, in the presence of several additives aimed at enhancing their organoleptic appeal and textural performance. These matrices were produced from a combination of either ovine or bovine whey (or a mixture thereof) with ovine milk, and were inoculated at 10% (v/v) with the probiotic strain. Sugar, sugar and aloe vera, sugar and chocolate, and sugar and jam were further added, and the resulting products were then stored at 7 ?C for 21 d. In general, viable cell numbers remained high in all experimental matrices throughout storage. Despite the observed low extents of breakdown, proteolytic activities by the end of storage were higher in matrices containing jam. Furthermore, L. paracasei partially converted lactose into lactic acid in these matrices. Additives enhanced the organoleptic features of whey cheeses, and produced different textural patterns. The higher sensory scores were attained by matrices containing sugar: sugar and aloe vera received the best scores by 3 d of storage, but these scores decreased as storage time elapsed. Lactobacillus casei DN-114 001 is a probiotic strain able to interact with the immune system and to interfere with gastrointestinal pathogens. The derived strain DN-114 001Rif was studied during its transit through the upper and distal intestine of human volunteers. Seven volunteers participated in the study, which involved intestinal intubation to sample ileal contents and collection of fecal samples, with a wash-out period of 8 days between the 2 steps. The retrieval of the probiotic was analyzed in the ileum every 2 h for 8 h following the ingestion of one dose of the test product and in the feces prior to, during, and after daily consumption of the test product for 8 days. Persistence of the probiotic amplifiable DNA was assessed using temporal temperature gradient gel electrophoresis and real-time PCR. Fluorescent in situ hybridization allowed analysis of the composition of the dominant digestive microbiota. The ingestion of L. casei DN-114 001Rif led to a significant and transient increase of its amplifiable DNA in ileal and fecal samples. This is related to a high stability in the composition of dominant groups of the gut microbiota. Data from ileal samples are scarce and our study confirms the potentiality for interaction between probiotics and the human immune system. Lactobacillus casei DN-114 001 is a probiotic strain able to interact with the immune system and to interfere with gastrointestinal pathogens. The derived strain DN-114 001Rif was studied during its transit through the upper and distal intestine of human volunteers. Seven volunteers participated in the study, which involved intestinal intubation to sample ileal contents and collection of fecal samples, with a wash-out period of 8 days between the 2 steps. The retrieval of the probiotic was analyzed in the ileum every 2 h for 8 h following the ingestion of one dose of the test product and in the feces prior to, during, and after daily consumption of the test product for 8 days. Abstract: Coccidiosis is the major parasitic disease of poultry. In this study, the role of the commercial probiotic MitoMax® which contains Pediococcus acidilactici and Saccharomyces boulardii was evaluated by measuring body weight gain, fecal oocyst shedding, and serum antibody responses as an alternative control method of prophylactic drug against coccidiosis. Day-old broiler chicks were fed regular or probiotic diets supplemented with MitoMax at 0.01%, 0.1%, or 1.0% of diet, and challenged 2 weeks later with 5000 oocysts of either Eimeria acervulina (EA) or Eimeria tenella (ET). Birds fed 1.0% or 0.1% MitoMax-supplemented diets in EA- or ET-infected groups shed less (P<0.05) oocysts than control-infected chickens. Also, chickens fed 0.1% MitoMax-supplemented diet and infected with EA exhibited higher (P<0.001) serum Eimeria-specific antibodies than other groups. These results demonstrate that MitoMax may enhance the resistance of birds against coccidiosis by enhancing humoral immunity when included at ⩾0.1% of the broiler diet. [Copyright &y& Elsevier]

Abstract: Besides assuring growth and development in early infancy, human breast milk plays a key role in preventing infections, in particular by favouring the growth of bifidobacteria. The development of infant formulas containing probiotic agents consequently represents a significant advance in the provision of an optimal alternative to breast feeding when this is not feasible. A further concern is to avoid an excessively high protein content relative to breast milk, potentially favouring obesity in later life. Objectives: The objective of this open Multicenter comparative study was to demonstrate that a new infant formula, characterized by a reduced protein content and the presence of a probiotic agent, was as effective as conventional formulas in assuring growth during the first months of life. Population and methods: The study population comprised full-term healthy infants with an age at inclusion of 1-28 days, whose mothers no longer wished to continue breast feeding or were unable to breast feed. The test formula (GE), enriched with Bifidobacterium lactis, had a protein content of 1.5 g/100 ml. The primary end point was daily weight gain from day 0 to day 90. Secondary end points were daily increase in height and cranial circumference, overall and gastrointestinal tolerance and formula acceptability. A total of 203 infants were included in the analysis, 117 receiving GE and 86 the formula habitually prescribed by the paediatrician. Results: Daily gains in weight, height and cranial circumference did not differ significantly between the groups. The rates of overall and gastrointestinal adverse events were comparable in the two groups and the acceptability of GE was consistently good, similar to that of the standard formulas. Conclusion: The results of this study demonstrated that the new probiotic infant formula GE has equivalent efficacy to conventional formulas in achieving growth in early infancy, and is well tolerated.
