Abstract Digital twins represent a key technology for precision health. Medical digital twins consist of computational models that represent the health state of individual patients over time, enabling optimal therapeutics and forecasting patient prognosis. Many health conditions involve the immune system, so it is crucial to include its key features when designing medical digital twins. The immune response is complex and varies across diseases and patients, and its modelling requires the collective expertise of the clinical, immunology, and computational modelling communities. This review outlines the initial progress on immune digital twins and the various initiatives to facilitate communication between interdisciplinary communities. We also outline the crucial aspects of an immune digital twin design and the prerequisites for its implementation in the clinic. We propose some initial use cases that could serve as “proof of concept” regarding the utility of immune digital technology, focusing on diseases with a very different immune response across spatial and temporal scales (minutes, days, months, years). Lastly, we discuss the use of digital twins in drug discovery and point out emerging challenges that the scientific community needs to collectively overcome to make immune digital twins a reality.

The moth genus Plutella Schrank, 1802 (Lepidoptera, Plutellidae) includes 26 described species. In addition to the diamondback moth, P. xylostella (Linnaeus, 1758), which is an important and widely distributed pest of cruciferous crops, only two species have been previously recorded in Chile, both with distribution ranges restricted to the southern temperate rainforests.Plutella copaquillaensis sp. nov. is described and illustrated, based on adults reared from larvae collected on Neuontobotrys lanata (Walp.) Al-Shehbaz (Brassicaceae) in the arid highlands of the Andes of northern Chile. The new species resembles P. xylostella in wing pattern, but clearly differs in genitalia morphology.

As queer and trans scientists, we face varied and systemic barriers to our professional success, resulting in our relative absence from faculty ranks at many institutions. In this Perspective, we call for a change in faculty hiring practices and present concrete guidance to make it a more inclusive process.

In this paper, it is revealed that random exploration and attraction of the best (REAB) are two underlying procedures in many swarm intelligence algorithms. This is particularly shown in two of the most known swarm algorithms: the particle swarm optimization (PSO) and gray wolf optimizer (GWO) algorithms. From this observation, it is here proposed that instead of building algorithms based on a narrative derived from observing some animal behavior, it is more convenient to focus on algorithms that perform REAB procedures; that is, to build algorithms to make a wide and efficient explorations of the search space and then gradually make that the best-evaluated search agent to attract the rest of the swarm. Following this general idea, two REAB-based algorithms are proposed; one derived from the PSO and one derived from the GWO, called REAB-PSO and REAB-GWO, respectively. To easily and succinctly express both algorithms, variable-sized open balls are employed. A comparison of proposed procedures in this paper and the original PSO and GWO using a controller tuning problem as a test bench show a significant improvement of the REAB-based algorithms over their original counterparts. Ideas here exposed can be used to derive new swarm intelligence algorithms.

Remote laboratories are essential in addressing access and quality challenges in technical education. They enable students from various locations to engage with real equipment, overcome geographic and economic constraints, and provide solutions during crises, such as pandemics, when in-person learning is limited. As a key element of Education 4.0, remote labs promote technical skill development, enhance engineering education, and support diverse learning approaches. This study presents a remote laboratory based on Field Programmable Gate Arrays (FPGAs), developed using a waterfall methodology integrating IoT and Cloud Computing technologies to facilitate close interaction between hardware and software. The lab focuses on controlling DC, servo, and stepper motors, allowing students to apply theoretical concepts such as digital signals, pulse-width modulation (PWM), and data representation in bits in a practical setting. The testing phase involved 50 robotics and mechatronics engineering students who participated in hands-on sessions for one month, followed by a structured survey evaluating their experience, interaction, and the educational relevance of the platform. The survey shows high student satisfaction, highlighting the platform’s strengths and identifying areas for improvement. The results also underscore the system’s potential to significantly enhance the educational experience in remote environments, aligning with the United Nations Sustainable Development Goals (SDGs).

This study explores the development of biocompatible scaffolds for bone regeneration, utilizing polylactic acid (PLA) combined with calcium phosphate as a pH buffer and diatomaceous earth as a biocompatibilizer. These materials were extruded and 3D-printed to enhance cell adhesion and biodegradability after enough cell growth. The biocompatibility of the resulting composites, with different proportions of the components and sterilization methods, was tested according to the ISO 10993 protocol. The optimal performance, with nearly zero cytotoxicity, was observed with 20 PLA/1 CP/1 DE mass ratios and gamma sterilization. Tension analysis and scanning electron microscopy (SEM) were applied to the 3D-printed composites, which were also analyzed by differential scanning calorimetry (DSC) to understand the origin of the tension properties better, which were comparable to those of cancellous bone. Degradation tests under physiological conditions for 13 weeks showed no significant mass loss. Furthermore, it was observed that cell adhesion, viability, proliferation, and osteoconduction are possible in the scaffolds studied, opening opportunities for future studies to substantiate the use of 3D-printed silica-filled composites as an alternative to homologous implants for various bone regeneration applications.

Background/Objectives: Diabetic foot ulcers (DFUs) are a common and severe complication of diabetic patients, with significant global prevalence and associated health burdens, including high recurrence rates, lower-limb amputations, and substantial associated economic costs. This study aimed to understand the user needs of healthcare professionals treating diabetic foot ulcers for newly developed material technologies. Methods: An open-ended questionnaire was used to identify user needs, identify the limitations of current treatments, and determine the specific requirements for ideal treatment. This information was used to develop a list of key considerations for creating innovative material technologies to improve diabetic wound treatment results. Results: Most respondents indicated that they followed published treatment guidelines for DFUs but noted that treatment often required a case-specific approach. Antibiotics and surgical debridement were commonly used for infection control. The participants showed a strong preference for wound dressings with lasting antibacterial properties. Respondents identified ideal properties for new products, including ease of use, enhanced antibacterial properties, affordability, and targeted biological activity. The respondents also highlighted the importance of a holistic approach to DFU management, integrating product development with comprehensive care strategies and patient education. Conclusions: This study highlights the complexity of DFU care, emphasizing that no single product can address all treatment needs. Future materials could focus on combination therapies and specific use cases. Additionally, understanding global variations in treatment practices and educating users on the proper application of newly developed material technologies is crucial for improving the management of DFUs and patient outcomes.

The micromoth fauna of the arid environments of the western slopes of central Andes remains poorly explored. Plants native to this area host overlooked species. A survey for micromoth larvae on the Chilean endemic Stevia philippiana Hieron. (Asteraceae) was performed.The first records of plume moths (Lepidoptera, Pterophoridae) associated with S. philippiana are provided. Adults of Adaina jobimi Vargas, 2020 and a new species of Oidaematophorus Wallengren, 1862 were reared from larvae collected on inflorescences and leaves, respectively. Oidaematophorus andresi sp. n. is described and illustrated. A phylogenetic analysis of mitochondrial DNA sequences clustered each of the two plume moths with the type species of its respective genus. These records expand the host plant range of A. jobimi and add a second species of Oidaematophorus to the Chilean fauna of plume moths.

Background: The temperature upshift has been widely used as an induction system to produce recombinant proteins (RPs). However, thermoinduction could affect bacterial metabolism, RP production, and RP aggregation. Understanding the structure and functionality of those aggregates, known as inclusion bodies (IBs), is a research area of interest in bioprocesses being scarcely studied under thermoinduction. Here, we describe the effect of the thermoinduction (39°C or 42°C) on the production of the recombinant human granulocyte–macrophage colony-stimulating factor (rHuGM-CSF) using Escherichia coli W3110 under the system λpL/cI857. Results: Results indicated that at 39°C, the production of biomass was almost doubled as well as the acetate accumulation compared to 42°C. Cultures thermoinduced at 42°C improved 1.5-fold the total protein over biomass yield and 1.25-fold the RP over total protein yield. Furthermore, 42°C accelerated the onset of IB formation, changing its architecture. Additionally, IBs formed at 42°C were less soluble and presented higher disorderly structures compared with IBs formed at 39°C, enriched in α-helix and amyloidal-like structures. Conclusions: This study highlights the observation that IBs attain different architecture in response to small changes in environmental conditions, such as the induction temperature, being this helpful information to improve thermoinduced bioprocesses.How to cite: Restrepo-Pineda S, Rosiles-Becerril D, Vargas-Castillo AB, et al. El propósito de este documento es proveer información general del estado actual del conocimiento del caracol cola de cuerno (Macrochlamys indica Benson) así como recomendaciones para su detección y monitoreo en los viveros. El caracol cola de cuerno fue detectado en agosto del 2020 en el condado de Miami-Dade (Talamas 2020). Este caracol es considerado como una plaga de importancia cuarentenaria en los Estados Unidos debido a que se puede convertir en una plaga agrícola y es potencialmente hospedero de nematodos parásitos de importancia médica (Grewal et al. 2003; Cowie et al. 2009; Jayashankar y Murthy 2015). Un programa de monitoreo, control y erradicación del caracol cola de cuerno fue implementado por la División de Industria Vegetal del Departamento de Agricultura y Servicios al Consumidor del estado de Florida (FDACS-DPI, por sus siglas en inglés). En la actualidad el programa de erradicación ha sido suspendido por parte de FDACS-DPI.

El ácaro de la erinosis del lichi es una plaga nativa de Asia que ha sido reportada en varios países. En Brasil ha causado pérdidas y ha incrementado los costos de producción en un 20%. También en India se ha reportado una reducción del 80% en el rendimiento. En 2018, el LEM fue encontrado en el condado de Lee, Florida y luego en otros del centro y el sur del estado. En consecuencia, la División de Industria Vegetal del Departamento de Agricultura y Servicios al Consumidor del estado de Florida estableció un programa de erradicación y cuarentena en el condado de Lee. Esta publicación brinda un perfil a profundidad del ácaro de la erinosis del lichi y está dirigida a personas interesadas con conocimiento de biología, así como a una audiencia académica.

The development of multicistronic vectors enabling differential transgene expression is a goal of gene therapy and poses a significant engineering challenge. Current approaches rely on the insertion of long regulatory sequences that occupy valuable space in vectors, which have a finite and limited packaging capacity. Here we describe a simple method of achieving differential transgene expression by inserting stop codons and translational readthrough motifs (TRMs) to suppress stop codon termination. TRMs reduced downstream transgene expression ∼sixfold to ∼140-fold, depending on the combination of stop codon and TRM used. We show that a TRM can facilitate the controlled secretion of the highly potent cytokine IL-12 at therapeutically beneficial levels in an aggressive immunocompetent mouse melanoma model to prevent tumor growth. Given their compact size (6 bp) and ease of introduction, we envisage that TRMs will be widely adopted in recombinant DNA engineering to facilitate differential transgene expression.

El picudo del botón del hibisco es una plaga originaria de México y de Texas, visto en Florida por primera vez en 2017. El incremento de las poblaciones del picudo entre 2019 y 2020 impactó negativamente la industria del hibisco en Florida, resultando en grandes pérdidas económicas (el estado lidera la producción nacional). Esta plaga está regulada por la División de Industria Vegetal del Departamento de Agricultura y Servicios al Consumidor, que establece que cualquier vivero que lo detecte debe seguir un acuerdo para reducir probabilidades de dispersión. El propósito de este documento es proveer información acerca de esta importante plaga a productores de viveros y al público interesado.

Background: Parkinson’s disease (PD) is associated with progressive neuronal damage and dysfunction. Oxidative stress helps to regulate neurodegenerative and neuronal dysfunction. Natural compounds could attenuate oxidative stress in a variety of neurological disorders. B. juncea is a rich source of antioxidants. The present study aimed to evaluate the therapeutic potential of B. juncea leaves for the treatment of PD by applying behavioral, in vivo and in silico studies. For in vivo studies rats were divided into six groups (n = 6). Group-I served as normal control (vehicle control). Group-II was disease control (haloperidol 1 mg/kg). Group-III was kept as a standard group (L-Dopa 100 mg/kg + carbidopa 25 mg/kg). Groups (IV–VI) were the treatment groups, receiving extract at 200-, 400- and 600 mg/kg doses respectively, for 21 days orally. Results: In vivo study results showed that the extract was found to improve muscles strength, motor coordination, and balance in PD. These behavioral outcomes were consistent with the recovery of endogenous antioxidant defence in biochemical analysis which was further corroborated with histopathological ameliorations. Dopamine levels increased and monoamine oxidase B (MAO-B) levels decreased dose-dependently in the brain during the study. Herein, we performed molecular docking analysis of the proposed extracted phytochemicals has explained that four putative phytochemicals (sinapic acid, rutin, ferulic acid, and caffeic acid) have presented very good results in terms of protein-ligand binding interactions as well as absorption, distribution, metabolism, excretion & toxicity (ADMET) profile estimations. Conclusion: The undertaken study concluded the anti-Parkinson activity of B. juncea and further suggests developments on its isolated compounds in PD therapeutics.

Chira River is located on the north coast of Peru. The lack of knowledge of the ichthyofauna from coastal drainages from Peru is noteworthy. The aim of this study is to characterize the ichthyofauna along the Chira River basin in terms of diversity and altitudinal range distribution. The material examined belongs to the Ichthyological collection of the Natural History Museum (MUSM). The diversity of fishes is composed of 27 species belonging to 19 families and ten orders. Siluriformes and Characiformes were the most diverse, consisting of 22% (six species) for each one. Five species are new records for the Chira River (three natives and two non-natives). Six marine species and seven exotic species were recorded as well. The altitudinal distribution patterns for all species were registered. This study increases the known diversity of freshwater fishes from Pacific Drainage Rivers in Peru, and it could be used for management and conservation plans.

ABSTRACT Academic conferences are integral to the dissemination of novel research findings and discussion of pioneering ideas across all postsecondary disciplines. For some participants, these environments are spaces to develop new collaborations, research projects, and social bonds; however, for others, conferences can be a place of marginalization and outright hostility. To assess how diverse individuals experience conference spaces, we interpreted results from a conference climate survey filled out by 198 of 482 registrants of the Society for the Advancement of Biology Education Research (SABER) West 2021 conference. Analysis of the survey data was conducted by six biology education researchers, who in addition to raising conference participant voices, provide insights, and next steps whose implementation can promote greater participant equity, representation, and engagement in future science, technology, engineering, and math (STEM) education conferences specifically and potentially all academic conference spaces more broadly.

Biogeographical studies have traditionally focused on readily visible organisms, but recent technological advances are enabling analyses of the large-scale distribution of microscopic organisms, whose biogeographical patterns have long been debated. Here we assessed the global structure of plankton geography and its relation to the biological, chemical, and physical context of the ocean (the ‘seascape’) by analyzing metagenomes of plankton communities sampled across oceans during the Tara Oceans expedition, in light of environmental data and ocean current transport. Using a consistent approach across organismal sizes that provides unprecedented resolution to measure changes in genomic composition between communities, we report a pan-ocean, size-dependent plankton biogeography overlying regional heterogeneity. We found robust evidence for a basin-scale impact of transport by ocean currents on plankton biogeography, and on a characteristic timescale of community dynamics going beyond simple seasonality or life history transitions of plankton.

Infections and disease caused by the obligate human pathogen Bordetella pertussis (Bp) are increasing, despite widespread vaccinations. The current acellular pertussis vaccines remain ineffective against nasopharyngeal colonization, carriage, and transmission. In this work, we tested the hypothesis that Bordetella polysaccharide (Bps), a member of the poly-β-1,6-N-acetyl-D-glucosamine (PNAG/PGA) family of polysaccharides promotes respiratory tract colonization of Bp by resisting killing by antimicrobial peptides (AMPs). Genetic deletion of the bpsA-D locus, as well as treatment with the specific glycoside hydrolase Dispersin B, increased susceptibility to AMP-mediated killing. Bps was found to be both cell surface-associated and released during laboratory growth and mouse infections. Addition of bacterial supernatants containing Bps and purified Bps increased B. pertussis resistance to AMPs. By utilizing ELISA, immunoblot and flow cytometry assays, we show that Bps functions as a dual surface shield and decoy. Co-inoculation of C57BL/6J mice with a Bps-proficient strain enhanced respiratory tract survival of the Bps-deficient strain. In combination, the presented results highlight the critical role of Bps as a central driver of B. pertussis pathogenesis. Heterologous production of Bps in a non-pathogenic E. coli K12 strain increased AMP resistance in vitro, and augmented bacterial survival and pathology in the mouse respiratory tract. These studies can serve as a foundation for other PNAG/PGA polysaccharides and for the development of an effective Bp vaccine that includes Bps.

Abstract Background Olive tree (Olea europaea L. subsp. europaea, Oleaceae) has been the most emblematic perennial crop for Mediterranean countries since its domestication around 6000 years ago in the Levant. Two taxonomic varieties are currently recognized: cultivated (var. europaea) and wild (var. sylvestris) trees. However, it remains unclear whether olive cultivars derive from a single initial domestication event followed by secondary diversification, or whether cultivated lineages are the result of more than a single, independent primary domestication event. To shed light into the recent evolution and domestication of the olive tree, here we analyze a group of newly sequenced and available genomes using a phylogenomics and population genomics framework. Results We improved the assembly and annotation of the reference genome, newly sequenced the genomes of twelve individuals: ten var. europaea, one var. sylvestris, and one outgroup taxon (subsp. cuspidata)—and assembled a dataset comprising whole genome data from 46 var. europaea and 10 var. sylvestris. Phylogenomic and population structure analyses support a continuous process of olive tree domestication, involving a major domestication event, followed by recurrent independent genetic admixture events with wild populations across the Mediterranean Basin. Cultivated olives exhibit only slightly lower levels of genetic diversity than wild forms, which can be partially explained by the occurrence of a mild population bottleneck 3000–14,000 years ago during the primary domestication period, followed by recurrent introgression from wild populations. Genes associated with stress response and developmental processes were positively selected in cultivars, but we did not find evidence that genes involved in fruit size or oil content were under positive selection. This suggests that complex selective processes other than directional selection of a few genes are in place. Conclusions Altogether, our results suggest that a primary domestication area in the eastern Mediterranean basin was followed by numerous secondary events across most countries of southern Europe and northern Africa, often involving genetic admixture with genetically rich wild populations, particularly from the western Mediterranean Basin.

Introducción. El hambre es uno de los principales referentes en la política mundial de seguridad alimentaria. Sin embargo, cada año mueren miles de personas a causa de la falta de ingesta de alimentos. Objetivo. Evaluar el número de fallecimientos por desnutrición en América del Sur, en un periodo correspondiente a los últimos veinte años de información. Materiales y métodos. Se empleó el modelo lineal general, incorporando la prueba de Tukey con base en un nivel de significancia estadístico del 5%. Adicionalmente, se llevó a cabo la técnica multivariada clúster. Resultados. El mayor número de muertes por desnutrición ocurrió en los niños menores de cuatro años y en adultos mayores. El mayor porcentaje de muertes por desnutrición en niños menores de un año se presentó en Venezuela (23,9% mujeres; 25,4% hombres). Chile tiene el menor porcentaje de muertes por desnutrición en menores de un año (0,4% mujeres; 0,5% hombres). Conclusión. A pesar de que los países de América del Sur presentan gran disponibilidad de alimentos, las muertes por hambre siguen siendo prevalentes en parte de la región.

The recent global Zika epidemics have revealed the significant threat that mosquito-borne viruses pose. There are currently no effective vaccines or prophylactics to prevent Zika virus (ZIKV) infection. Limiting exposure to infected mosquitoes is the best way to reduce disease incidence. Recent studies have focused on targeting mosquito reproduction and immune responses to reduce transmission. Previous work has evaluated the effect of insulin signaling on antiviral JAK/STAT and RNAi in vector mosquitoes. Specifically, insulin-fed mosquitoes resulted in reduced virus replication in an RNAi-independent, ERK-mediated JAK/STAT-dependent mechanism. In this work, we demonstrate that targeting insulin signaling through the repurposing of small molecule drugs results in the activation of both RNAi and JAK/STAT antiviral pathways. ZIKV-infected Aedes aegypti were fed blood containing demethylasterriquinone B1 (DMAQ-B1), a potent insulin mimetic, in combination with AKT inhibitor VIII. Activation of this coordinated response additively reduced ZIKV levels in Aedes aegypti. This effect included a quantitatively greater reduction in salivary gland ZIKV levels up to 11 d post-bloodmeal ingestion, relative to single pathway activation. Together, our study indicates the potential for field delivery of these small molecules to substantially reduce virus transmission from mosquito to human. As infections like Zika virus are becoming more burdensome and prevalent, understanding how to control this family of viruses in the insect vector is an important issue in public health.

Mechanical stress is known to fuel several hallmarks of cancer, ranging from genome instability to uncontrolled proliferation or invasion. Cancer cells are constantly challenged by mechanical stresses not only in the primary tumour but also during metastasis. However, this latter has seldom been studied with regards to mechanobiology, in particular resistance to anoikis, a cell death programme triggered by loss of cell adhesion. Here, we show in vitro that migrating breast cancer cells develop resistance to anoikis following their passage through microporous membranes mimicking confined migration (CM), a mechanical constriction that cancer cells encounter during metastasis. This CM-induced resistance was mediated by Inhibitory of Apoptosis Proteins, and sensitivity to anoikis could be restored after their inhibition using second mitochondria-derived activator of caspase (SMAC) mimetics. Anoikis-resistant mechanically stressed cancer cells displayed enhanced cell motility and evasion from natural killer cell-mediated immune surveillance, as well as a marked advantage to form lung metastatic lesions in mice. Our findings reveal that CM increases the metastatic potential of breast cancer cells.

This study presents an extensive review of published and unpublished occurrence records of fish species in the Loreto department. Located in the northeast of the country, Loreto is the most geographically extensive region in the Peruvian territory. Despite the increase in fish collections in Loreto in recent years, the ichthyofauna of this department needs to be more documented. Based on a database of scientific collections and bibliographic information, an updated checklist of the freshwater fishes from Loreto is presented. The results reveal a rich and diversified ichthyofauna, with 873 species distributed in 331 genera, 50 families and 15 orders. The main groups are Characiformes (42.6%), Siluriformes (34.8%), Gymnotiformes (8.6%) and Cichliformes (7.4%). Part of the ichthyofauna has restricted distribution for Loreto (4.7%). In addition, 9.0% of species from Loreto are used in fisheries. Meanwhile, 219 species (25%) were categorized according to the IUCN criteria where only six species (0.7%) are currently considered threatened species (CR, EN or VU). The results presented in this work indicate that this department needs more studies to know the biodiversity of fish, likewise, the information presented constitutes a contribution to the knowledge of fish diversity that would support environmental management actions and decision-making aimed at conserving one of the most diverse departments of Peru.

The Patch-seq approach is a powerful variation of the patch-clamp technique that allows for the combined electrophysiological, morphological, and transcriptomic characterization of individual neurons. To generate Patch-seq datasets at scale, we identified and refined key factors that contribute to the efficient collection of high-quality data. We developed patch-clamp electrophysiology software with analysis functions specifically designed to automate acquisition with online quality control. We recognized the importance of extracting the nucleus for transcriptomic success and maximizing membrane integrity during nucleus extraction for morphology success. The protocol is generalizable to different species and brain regions, as demonstrated by capturing multimodal data from human and macaque brain slices. The protocol, analysis and acquisition software are compiled at https://githubcom/AllenInstitute/patchseqtools. This resource can be used by individual labs to generate data across diverse mammalian species and that is compatible with large publicly available Patch-seq datasets.

Mango pericarp powders (Mangifera indica L.) of Ataulfo, Keitt and Tommy Atkins cultivars were obtained by dehydration and grinding. The content of moisture, ash, fat, dietary fiber, antioxidant capacity and total phenols was evaluated. The cultivar with functional ingredient potential was Ataulfo with antioxidant capacity of 34,811 µmol ET g-1 and 7578 mg EAG 100 g-1 total phenol, and dietary fiber ≈ 20 %. All three cultivars can be considered as ingredients for adding or enriching fiber in food formulations.

Abstract Background The Hemiptera (aphids, cicadas, and true bugs) are a key insect order, with high diversity for feeding ecology and excellent experimental tractability for molecular genetics. Building upon recent sequencing of hemipteran pests such as phloem-feeding aphids and blood-feeding bed bugs, we present the genome sequence and comparative analyses centered on the milkweed bug Oncopeltus fasciatus, a seed feeder of the family Lygaeidae. Results The 926-Mb Oncopeltus genome is well represented by the current assembly and official gene set. We use our genomic and RNA-seq data not only to characterize the protein-coding gene repertoire and perform isoform-specific RNAi, but also to elucidate patterns of molecular evolution and physiology. We find ongoing, lineage-specific expansion and diversification of repressive C2H2 zinc finger proteins. The discovery of intron gain and turnover specific to the Hemiptera also prompted the evaluation of lineage and genome size as predictors of gene structure evolution. Furthermore, we identify enzymatic gains and losses that correlate with feeding biology, particularly for reductions associated with derived, fluid nutrition feeding. Conclusions With the milkweed bug, we now have a critical mass of sequenced species for a hemimetabolous insect order and close outgroup to the Holometabola, substantially improving the diversity of insect genomics. We thereby define commonalities among the Hemiptera and delve into how hemipteran genomes reflect distinct feeding ecologies. Given Oncopeltus’s strength as an experimental model, these new sequence resources bolster the foundation for molecular research and highlight technical considerations for the analysis of medium-sized invertebrate genomes.

This article analyzes the implementation of a circular economy model for the management of reusable solid waste in the Colombian municipalities of Arbeláez (province of Cundinamarca) and Tibasosa (province of Boyacá). The analysis is conducted using M-GRCT, a circular economy decision support model for the design of recyclable waste management systems in low-income municipalities. The model allows for performing calculations on a set of two scenarios integrating a sociocultural dynamics assessment—this being a characteristic feature of this type of municipalities. Results show that both the linear and circular models of waste management are economically viable. However, the particular conditions of each municipality, the tariff system, the number of subscribers and the variations in costs and inflation in each municipality affect the results of economic viability. In addition, the waste production scale and the volumes of recoverable waste also affect the results. All these factors are reflected in the scenarios analyzed. In terms of economic viability, the circular model presents better results in Arbeláez, while in the municipality of Tibasosa, the best results are obtained with a linear economy approach.

Inflammatory bowel disease (IBD) is a chronic recurrent inflammatory disease with unknown etiology. Dextran sulfate sodium (DSS) induced colitis is a widely used mouse model in IBD research. DSS colitis involves activation of the submucosal immune system and can be used to study IBD-like disease characteristics in acute, chronic, remission and transition phases. Insight into colon inflammatory parameters is needed to understand potentially irreversible adaptations to the chronification of colitis, determining the baseline and impact of further inflammatory episodes. We performed analyses of non-invasive and invasive colitis parameters in acute, chronic and remission phases of the DSS colitis in C57BL/6 mice. Non-invasive colitis parameters poorly reflected inflammatory aspects of colitis in chronic remission phase. We found invasive inflammatory parameters, positively linked to repeated DSS-episodes, such as specific colon weight, inflamed colon area, spleen weight, absolute cell numbers of CD4+ and CD8+ T cells as well as B cells, blood IFN-γ level, colonic chemokines BLC and MDC as well as the prevalence of Turicibacter species in feces. Moreover, microbial Lactobacillus species decreased with chronification of disease. Our data point out indicative parameters of recurrent gut inflammation in context of DSS colitis.

Mexico is one of the richest countries in amphibian species (420 spp.), with a high level of endemism (69%). The order Anura represents the most diverse and widespread of the three extant amphibian orders (257 spp.). The anurofauna of Mexico’s tropical dry forest ecosystem host a high proportion of the species and endemism registered in the country. In terms of conservation, both dry forests and amphibians are at risk due to climate change because it is expected that as the temperature becomes higher and precipitation decreases, this vegetation type may experience water stress. We applied the MaxEnt algorithm to estimate the potential current and future (year 2070) geographic distribution patterns of 95 endemic Mexican anuran species inhabiting the country’s tropical dry forests by considering two representative concentration pathway scenarios (RCP4.5/RCP8.5) and analyzed the potential distributional pattern changes. The results indicated that overall, species would experience enough of a significant warming effect to cause a reduction in the original distribution area, with 44% of species losing an average of 50% of their original range (9 spp. in threatened category); additionally, 22% of the species in the dry forest ecosystem will experience an average increase of almost 50% in their original area, two species will lose more than 80% of their range, and one will disappear.

Diagnosis of Toxoplasma gondii acute infection was first attempted by detection of specific IgM antibodies, as for other infectious diseases. However, it was noted that this immunoglobulin declines slowly and may last for months or even years. Apart from the diagnostic problem imposed on clinical management, this phenomenon called our attention due to the underlying phenomena that may be causing it. We performed a systematic comparison of reports studying IgM antibody kinetics, and the data from the papers were used to construct comparative plots and other graph types. It became clear that this phenomenon is quite generalized, and it may also occur in animals. Moreover, this is not a technical issue, although some tests make more evident the prolonged IgM decay than others. We further investigated biological reasons for its occurrence, i.e., infection dynamics (micro-reactivation–encystment, reinfection and reactivation), parasite strain relevance, as well as host innate, natural B cell responses and Ig class-switch problems inflicted by the parasite. The outcomes of these inquiries are presented and discussed herein.

Radiotherapy treatments are carried out using computerized axial tomography. In radiation therapy planning, the radiation oncologist must do a manual segmentation of volumes of interest to delineate the organs that should be irradiated. This way of carrying out the process generates long execution times and introduces a subjective component. In this study, a contour-propagation algorithm is formulated to automate the segmentation, based on elastic registration or nonrigid demon registration. A heuristic algorithm to find the parameters that optimize the registration is also proposed. The parameters found along with the contour-propagation algorithm are able to estimate contours of scans with Dice similarity coefficients (DSC) greater than 0.92 and maintain stability with B-spline registration, which takes in the parameters found as input. The study allows for validating the results using the correlation coefficient (CC) to compare the similarity between the voxels’ gray-scale intensity of the estimated tomography and the original tomography, obtaining values greater than 0.96. These values were validated under medical criteria and applied to liver and breast CT scans, indicating good performance for radiation therapy planning.

Cell entry of enveloped viruses relies on the fusion between the viral and plasma or endosomal membranes, through a mechanism that is triggered by a cellular signal. Here we used a combination of computational and experimental approaches to unravel the main determinants of hepatitis B virus (HBV) membrane fusion process. We discovered that ERp57 is a host factor critically involved in triggering HBV fusion and infection. Then, through modeling approaches, we uncovered a putative allosteric cross-strand disulfide (CSD) bond in the HBV S glycoprotein and we demonstrate that its stabilization could prevent membrane fusion. Finally, we identified and characterized a potential fusion peptide in the preS1 domain of the HBV L glycoprotein. These results underscore a membrane fusion mechanism that could be triggered by ERp57, allowing a thiol/disulfide exchange reaction to occur and regulate isomerization of a critical CSD, which ultimately leads to the exposition of the fusion peptide.

Tuberculosis (TB) is a leading cause of death globally. Understanding the population dynamics of TB’s causative agent Mycobacterium tuberculosis complex (Mtbc) in-host is vital for understanding the efficacy of antibiotic treatment. We use longitudinally collected clinical Mtbc isolates that underwent Whole-Genome Sequencing from the sputa of 200 patients to investigate Mtbc diversity during the course of active TB disease after excluding 107 cases suspected of reinfection, mixed infection or contamination. Of the 178/200 patients with persistent clonal infection >2 months, 27 developed new resistance mutations between sampling with 20/27 occurring in patients with pre-existing resistance. Low abundance resistance variants at a purity of ≥19% in the first isolate predict fixation in the subsequent sample. We identify significant in-host variation in 27 genes, including antibiotic resistance genes, metabolic genes and genes known to modulate host innate immunity and confirm several to be under positive selection by assessing phylogenetic convergence across a genetically diverse sample of 20,352 isolates.

(1) Background: Epidemiological studies have identified associations between fine particulate matter (PM2.5) and ozone exposure with cardiovascular disease; however, studies linking ambient air pollution and premature coronary artery disease (pCAD) in Latin America are non-existing. (2) Methods: Our study was a case–control analysis nested in the Genetics of Atherosclerotic Disease (GEA) Mexican study. We included 1615 participants (869 controls and 746 patients with pCAD), recruited at the Instituto Nacional de Cardiología Ignacio Chávez from June 2008 to January 2013. We defined pCAD as history of myocardial infarction, angioplasty, revascularization surgery or coronary stenosis > 50% diagnosed before age 55 in men and age 65 in women. Controls were healthy individuals without personal or family history of pCAD and with coronary artery calcification equal to zero. Hourly measurements of ozone and PM2.5 from the Atmospheric Monitoring System in Mexico City (SIMAT in Spanish; Sistema de Monitero Atmosférico de la Ciudad de México) were used to calculate annual exposure to ozone and PM2.5 in the study participants. (3) Results: Each ppb increase in ozone at 1-year, 2-year, 3-year and 5-year averages was significantly associated with increased odds (OR = 1.10; 95% CI: 1.03–1.18; OR = 1.17; 95% CI: 1.05–1.30; OR = 1.18; 95% CI: 1.05–1.33, and OR = 1.13; 95% CI: 1.04–1.23, respectively) of pCAD. We observed higher risk of pCAD for each 5 µg/m3 increase only for the 5-year average of PM2.5 exposure (OR = 2.75; 95% CI: 1.47–5.16), compared to controls. (4) Conclusions: Ozone exposure at different time points and PM2.5 exposure at 5 years were associated with increased odds of pCAD. Our results highlight the importance of reducing long-term exposure to ambient air pollution levels to reduce the burden of cardiovascular disease in Mexico City and other metropolitan areas.

Due to the increased incidence of fungal infections and the emergence of antifungal resistance mainly by Candida species, the need for safe and effective novel therapies is imperative. Consequently, plants and herbs are a powerful source to combat infections. Here, we evaluated the anti-Candida potential of an ethanolic extract from Piper nigrum. The phytochemical analysis of P. nigrum revealed bioactive compounds such as alkaloids, terpenoids, and tannis. Our results showed that P. nigrum extract suppressed the virulence factors of C. albicans strains, including hyphae formation in both liquid and solid media, reduced secretion of phospholipases/proteinases, and affected biofilm formation. Furthermore, the P. nigrum extract showed no hemolytic effect in vitro and exhibited reduced cytotoxicity on Vero cells and G. mellonella larvae at concentrations that inhibited hyphae and biofilm in C. albicans. Moreover, the extract demonstrated antifungal activity against C. auris strains. In conclusion, the P. nigrum extract affected the growth and morphogenesis of Candida (even in resistant strains), demonstrating that this plant has an anti-candida activity and represents a promising resource for discovering novel antifungal compounds.

This work describes the design and implementation of a Zero-Length Column system to measure: diffusion coefficients, adsorption isotherm parameters of pure components and mixtures. In addition, a graphical user interface (GUI) was developed in LabVIEW for the semi-automatic operation of the system. The system is novel because it integrates all the aforementioned functionalities without using mass spectrometry. Two adsorbents, zeolite 5A and Basolite® C300 (Copper benzene-1,3,5-tricarboxylate) and two adsorbates methane and ethane were used to perform the validation of adsorption and diffusion experiments. The Henry constants and diffusion coefficients obtained reproduce those previously reported. The combination of the experimental setup and the GUI significantly reduce the amount of sample and measurement time needed in the characterization of the molecular sieves by conventional volumetric and gravimetric systems. The proposed system is relatively inexpensive, robust, easy to build, and capable of reproducing the results of other techniques.

Blockchain is one of the latest innovations that is increasingly attracting the attention of various stakeholders in different fields, including the education sector. This is primarily due to its attractive features, such as decentralization, transparency, traceability, security, and reliability. Despite its advantages, blockchain still faces several challenges, and the acceptance rate of this technology is still low. Thus, the purpose of this study was to conduct a review of published articles that have discussed the challenges of adopting blockchain in the education sector. The review contained scientific papers published from 2017 to 2022 and, from the screened records, 32 articles were analyzed in full-text form. In this review, 14 challenges were reported and classified, based on the technology-organization-environment (TOE) framework. In addition, this review showed that organizational and environmental barriers received little attention in the literature, compared to technological barriers.

Chemotherapy is commonly used for cancer treatment, however the lack of selectivity on healthy cells and the development of resistance phenomena are the major issues. A better understanding of cancer genetics helped the development of new targeted anticancer treatments, which permit drug delivery with high specificity and lower toxicity. Moreover, the multi-target drug design concept represents the current trend for future drug research and development. Starting from good results previously obtained by our research group on the resveratrol (RSV) phenylacetamide derivative 2, which displayed an interesting anti-inflammatory and anti-proliferative activity towards the breast cancer cells MCF-7 and MDA-MB-231, we identified other features, as the ability to perturb the cytoskeleton dynamics and interfere with the migration and metastatic processes. In vitro and in silico studies demonstrate that the derivative 2 is a tubulin and actin polymerization inhibitor and an actin depolymerization promotor. In addition, it interferes with the metastatic potential in both the breast cancer cells, inhibiting the in vitro cell migration and decreasing the spheroids number. These promising results demonstrate that the RSV phenylacetamide derivative 2 could be an important starting point in the discovery and development of safer and more efficacy multi-targeted agents.

Goal-directed behaviors are essential for normal function and significantly impaired in neuropsychiatric disorders. Despite extensive associations between genetic mutations and these disorders, the molecular contributions to goal-directed dysfunction remain unclear. We examined mice with constitutive and brain region-specific mutations in Neurexin1α, a neuropsychiatric disease-associated synaptic molecule, in value-based choice paradigms. We found Neurexin1α knockouts exhibited reduced selection of beneficial outcomes and impaired avoidance of costlier options. Reinforcement modeling suggested that this was driven by deficits in updating and representation of value. Disruption of Neurexin1α within telencephalic excitatory projection neurons, but not thalamic neurons, recapitulated choice abnormalities of global Neurexin1α knockouts. Furthermore, this selective forebrain excitatory knockout of Neurexin1α perturbed value-modulated neural signals within striatum, a central node in feedback-based reinforcement learning. By relating deficits in value-based decision-making to region-specific Nrxn1α disruption and changes in value-modulated neural activity, we reveal potential neural substrates for the pathophysiology of neuropsychiatric disease-associated cognitive dysfunction.

Synucleinopathies are neurological disorders associated with α-synuclein overexpression and aggregation. While it is well-established that overexpression of wild type α-synuclein (α-syn-140) leads to cellular toxicity and neurodegeneration, much less is known about other naturally occurring α-synuclein splice isoforms. In this study we provide the first detailed examination of the synaptic effects caused by one of these splice isoforms, α-synuclein-112 (α-syn-112). α-Syn-112 is produced by an in-frame excision of exon 5, resulting in deletion of amino acids 103–130 in the C-terminal region. α-Syn-112 is upregulated in the substantia nigra, frontal cortex, and cerebellum of parkinsonian brains and higher expression levels are correlated with susceptibility to Parkinson’s disease (PD), dementia with Lewy bodies (DLB), and multiple systems atrophy (MSA). We report here that α-syn-112 binds strongly to anionic phospholipids when presented in highly curved liposomes, similar to α-syn-140. However, α-syn-112 bound significantly stronger to all phospholipids tested, including the phosphoinositides. α-Syn-112 also dimerized and trimerized on isolated synaptic membranes, while α-syn-140 remained largely monomeric. When introduced acutely to lamprey synapses, α-syn-112 robustly inhibited synaptic vesicle recycling. Interestingly, α-syn-112 produced effects on the plasma membrane and clathrin-mediated synaptic vesicle endocytosis that were phenotypically intermediate between those caused by monomeric and dimeric α-syn-140. These findings indicate that α-syn-112 exhibits enhanced phospholipid binding and oligomerization in vitro and consequently interferes with synaptic vesicle recycling in vivo in ways that are consistent with its biochemical properties. This study provides additional evidence suggesting that impaired vesicle endocytosis is a cellular target of excess α-synuclein and advances our understanding of potential mechanisms underlying disease pathogenesis in the synucleinopathies.

Arbuscular mycorrhizal fungi (AMF) colonization, sampled at 32–50 days post-inoculation (dpi), was significantly reduced in suppressor of prosystemin-mediated responses2 (spr2) mutant tomato plants impaired in the ω−3 FATTY ACID DESATURASE7 (FAD7) gene that limits the generation of linolenic acid and, consequently, the wound-responsive jasmonic acid (JA) burst. Contrary to wild-type (WT) plants, JA levels in root and leaves of spr2 mutants remained unchanged in response to AMF colonization, further supporting its regulatory role in the AM symbiosis. Decreased AMF colonization in spr2 plants was also linked to alterations associated with a disrupted FAD7 function, such as enhanced salicylic acid (SA) levels and SA-related defense gene expression and a reduction in fatty acid content in both mycorrhizal spr2 roots and leaves. Transcriptomic data revealed that lower mycorrhizal colonization efficiency in spr2 mutants coincided with the modified expression of key genes controlling gibberellin and ethylene signaling, brassinosteroid, ethylene, apocarotenoid and phenylpropanoid synthesis, and the wound response. Targeted metabolomic analysis, performed at 45 dpi, revealed augmented contents of L-threonic acid and DL-malic acid in colonized spr2 roots which suggested unfavorable conditions for AMF colonization. Additionally, time- and genotype-dependent changes in root steroid glycoalkaloid levels, including tomatine, suggested that these metabolites might positively regulate the AM symbiosis in tomato. Untargeted metabolomic analysis demonstrated that the tomato root metabolomes were distinctly affected by genotype, mycorrhizal colonization and colonization time. In conclusion, reduced AMF colonization efficiency in spr2 mutants is probably caused by multiple and interconnected JA-dependent and independent gene expression and metabolomic alterations.

Although the number of known tardigrade taxa in Mexico has increased significantly in the last ten years, the knowledge of their diversity faces challenges, as more than half of the Mexican territory has no records of this phylum. Thus, we developed a strategy to provide a present and future scenario for understanding the Mexican biodiversity of Tardigrada, described the distribution patterns of the current recorded species, calculated the estimated richness, and the estimated taxonomic effort needed to complete the national inventory. We obtained 474 records of 105 taxa, belonging to 42 genera and 75 species, distributed in 12 of the 14 biogeographical provinces of Mexico. We found that 54.72% of the species are present in more than three world regions and 3.79% of species that have been recorded only in Mexican provinces. Distribution patterns could be recognized for 11 species, two of which have a Nearctic distribution, seven are Neotropical and two are distributed in both regions. The Mexican biogeographical provinces with the greatest diversity of tardigrades, both at specific and generic level, were the Transmexican Volcanic Belt (TVBP) and the Sierras Madre Oriental (SMOrP) and Sierra Madre Occidental (SMOcP), which have been previously identified as particularly species-rich regions. Diversity estimation methods predict that more than 290 species of tardigrades could be found in Mexico.

It is demonstrated that data augmentation is a promising approach to reduce the size of the captured dataset required for training automatic road surface classifiers. The context is on-board systems for autonomous or semi-autonomous driving assistance: automatic power-assisted steering. Evidence is obtained by extensive experiments involving multiple captures from a 10-channel multisensor deployment: three channels from the accelerometer (acceleration in the X, Y, and Z axes); three microphone channels; two speed channels; and the torque and position of the handwheel. These captures were made under different settings: three worm-gear interface configurations; hands on or off the wheel; vehicle speed (constant speed of 10, 15, 20, 30 km/h, or accelerating from 0 to 30 km/h); and road surface (smooth flat asphalt, stripes, or cobblestones). It has been demonstrated in the experiments that data augmentation allows a reduction by an approximate factor of 1.5 in the size of the captured training dataset.

The damage caused by chronic alcohol consumption is frequently reported, but the effects caused by weekend recreational consumption, which is much more frequent than even daily consumption, have not, to our knowledge, been reported. The metabolism of ethanol, once consumed, starts from the mouth, and the biotransformation process follows different routes. In this study, the effect of weekend alcohol consumption on the oral cavity was observed. Methods: Thirty male and female rats were divided into six groups (n = 5), with control groups (male/female) and groups administered with 5% and 40% ethanol solution ad libitum consumption 2 days a week for 3 months. After treatment, the animals were sacrificed, an incisional slice of the cheek and back of the tongue was obtained, and the tissues were processed according to the histological technique and routine staining (hematoxylin-eosin, H&E). Samples were observed using light microscopy. Results: Histological changes were observed in samples of tongue and cheek mucosa including different levels of keratinization of the surface layer. Epithelial dysplasia, acanthosis, and chronic inflammation were also observed. The vascularization level also increased because of the ethanol-induced damage. The results were very similar between female and male groups. Conclusion: Weekend alcohol consumption for a period of 3 months causes oral-cavity tissue alterations that could contribute to tumor growth and the development of cancer in the oral cavity.

Male and female adults of Ypsolopha moltenii sp. n. (Lepidoptera, Ypsolophidae) are described and illustrated from the western slopes of the Andes of northern Chile. Larvae of Y. moltenii feed on inflorescences of the native shrub Adesmia verrucosa Meyen (Fabaceae). This finding represent the first species of Ypsolopha Latreille, 1796 described from mainland Chile and the first species of this genus with larvae feeding on the highly diverse South American genus Adesmia.

En Colombia los estudios sobre la avifauna en humedales interiores son escasos. En el caso del departamento del Tolima no se cuenta con información detallada, por lo cual se requieren trabajos que provean datos sobre este tópico. Por tal motivo, este estudio tiene como objetivo caracterizar la comunidad de aves asociada a los humedales de zonas bajas del departamento. Para esto, se estudiaron 13 humedales ubicados por debajo de 1000 m s.n.m. Se registraron 147 especies, distribuidas en 44 familias y 18 órdenes. Los humedales con mayor número de especies fueron Toqui Toqui (78) y El Edén (65), mientras que Chicoalí (22), El Samán y El Gavilán (27, cada uno) registraron el menor número; sin embargo, no se encontraron diferencias significativas en la composición de la avifauna de los humedales evaluados. Predominaron las especies de las categorías ecológicas para aves II (35 %) y III (38 %) y en menor medida fueron registradas las especies pertenecientes a la categoría Ia (1 %). A pesar de la alta intervención a la que están sometidos los humedales del bosque seco tropical en el Tolima, estos ecosistemas aún sustentan un número importante de especies.

Many crustacean species are obligate associates of pocilloporid corals, where they feed, reproduce, and find shelter. However, these coral-associated crustaceans have been poorly studied in the eastern tropical Pacific. Determining the crustacean richness and taxonomic distinctness could help in comparing different coral reefs and the potential effects of degradation. This study evaluated the spatio–temporal variation of the taxonomic diversity and distinctness of coral-associated crustaceans in four ecosystems of the Central Mexican Pacific (CMP) with different conditions and coral cover. In all ecosystems, 48 quadrants were sampled during the summer and winter for two years. A total of 12,647 individuals belonging to 88 species, 43 genera, and 21 families were recorded. The sampling effort yielded 79.6% of the expected species richness in the study area. Species rarity had 19% singletons, 4% doubletons, 22% unique, and 9% duplicate species; two species represented new records for the Mexican Pacific, and six were new to the CMP. This study recorded most of the symbiotic crustacean species in pocilloporid corals previously reported in the CMP. The taxonomic diversity and distinctness differed significantly between coral ecosystems and seasons, which was also visualized by nMDS ordination, showing an evident spatio–temporal variation in the taxonomic beta diversity.

Agricultural systems have increased in extension and intensity worldwide, altering vertebrate functional diversity, ecosystem functioning, and ecosystemic services. However, the effects of open monoculture crops on bird functional diversity remain little explored, particularly in highly biodiverse regions such as the tropical Andes. We aim to assess the functional diversity differences of bird guilds between monoculture crops (coffee, cocoa, and citrus) and secondary forests. We use four functional diversity indices (Rao Q, Functional Richness, Functional Evenness, and Functional Divergence) related to relevant morphological, life history, and behavioral traits. We find significant differences in functional diversity between agroecosystem and forest habitats. Particularly, bird functional diversity is quite homogeneous among crop types. Functional traits related to locomotion (body weight, wing-chord length, and tail length), nest type (closed), and foraging strata (canopy and understory) are dominant at the agroecosystems. The bird assemblages found at the agroecosystems are more homogeneous in terms of functional diversity than those found at the secondary forests, as a result of crop structure and management. We recommend promoting more diverse agroecosystems to enhance bird functional diversity and reduce their effects on biodiversity.

For centuries, indigenous sheep have been bred in extensive low-input systems in Midwestern Brazil. The hypothesis of this study was the assumption that phenotypic evaluation of indigenous livestock may drive the establishment of breed standards and official breed recognition, and, therefore, promote more local business opportunities. On the basis of more integrative and participatory theoretical background to applying any decision based on phenotype, we designed this research to determine the most typical and unusual phenotypes of Pantaneiro sheep. Pantaneiro ewes (281 ewes from five conservation units in five counties) were evaluated, bearing in mind both conservation and development. Descriptive statistics were used to classify ewes into typical, intermediate, and unusual phenotypes. Chi-squared tests for association were performed to test if morphological variation in the different sampling sites occurred randomly (p > 0.05) or not (p < 0.05). Some results suggest some sort of diversifying selection pressure, i.e., distinct preferences among keepers. We observed considerable morphologic variation among ewes, but it was straightforward to determine the predominant phenotypes. The study evokes integrative agrobiodiversity by putting phenotypical characterization of indigenous livestock into perspective. Discussions coming from this study may support innovative governance and participative decision-making, allied with strategies that value the socioeconomic, biocultural, and adaptive aspects of indigenous livestock in tropical regions and developing countries. This is a challenge for government, rare-breed keepers, value-chain actors, and civil society.
