Quiescence (G0) maintenance and exit are crucial for tissue homeostasis and regeneration in mammals. Here, we show that methyl-CpG binding protein 2 (Mecp2) expression is cell cycle-dependent and negatively regulates quiescence exit in cultured cells and in an injury-induced liver regeneration mouse model. Specifically, acute reduction of Mecp2 is required for efficient quiescence exit as deletion of Mecp2 accelerates, while overexpression of Mecp2 delays quiescence exit, and forced expression of Mecp2 after Mecp2 conditional knockout rescues cell cycle reentry. The E3 ligase Nedd4 mediates the ubiquitination and degradation of Mecp2, and thus facilitates quiescence exit. A genome-wide study uncovered the dual role of Mecp2 in preventing quiescence exit by transcriptionally activating metabolic genes while repressing proliferation-associated genes. Particularly disruption of two nuclear receptors, Rara or Nr1h3, accelerates quiescence exit, mimicking the Mecp2 depletion phenotype. Our studies unravel a previously unrecognized role for Mecp2 as an essential regulator of quiescence exit and tissue regeneration.

In light of the limitation that passive suspension can only provide vibration isolation within a specific range, a magnetorheological suspension in extrusion mode was developed. The reliability of structural parameters was ensured through theoretical analysis and numerical simulation, building upon traditional hydraulic suspension. A model linking static stiffness to the diameter of the upper extrusion plate, as well as the heights of the upper and lower liquid chambers, was established using Simulink as an evaluation index. The static stiffness performance of the magnetorheological suspension was then optimized using this model. Results indicate that while meeting the static stiffness requirements, the optimized Magnetorheological Suspension demonstrated a 29.22% increase in static stiffness (approximately 57.71 N/mm) compared to its previous state, validating the effectiveness of stiffness optimization for this system.

Aiming at the problem of high vibration and high wear of centrifugal pump tongue, this paper proposes a double-tongue volute structure. Under the condition of ensuring the reliability of CFD results, the influence of various combinations of tongue and volute base circle on the turbulent kinetic energy of centrifugal pump and the radial force of impeller is explored. The traditional single-tongue volute centrifugal pump is compared with various characteristic indexes, and the unsteady numerical calculation is carried out based on different working conditions. It is concluded that the double-tongue volute structure can improve the pressure fluctuation at the monitoring points near the tongue. The results show that the double-tongue volute structure can improve the static pressure gradient and velocity gradient of the middle section of the centrifugal pump and reduce the maximum turbulent kinetic energy value at the tongue under small flow conditions. When the working condition is 1.0 Q, the radial force of the impeller of the C-type double-tongue volute reaches the minimum value of 3.03 N, which can effectively balance part of the radial force.

The 3D quantitative analysis of facial morphology is of importance in plastic surgery (PS), which could help surgeons design appropriate procedures before conducting the surgery. We propose a system to simulate and guide the shaping effect analysis, which could produce a similar but more harmonious face simulation. To this end, first, the depth camera based on structured light coding is employed for facial 3D data acquisition, from which the point cloud data of multiple facial perspectives could be obtained. Next, the cascade regression tree algorithm is used to extract the esthetic key points of the face model and to calculate the facial features composed of the key points, such as the nose, chin, and eyes. Quantitative facial esthetic indexes are offered to doctors to simulate PS. Afterward, we exploit a face mesh metamorphosis based on finite elements. We design several morphing operators, including augmentation, cutting, and lacerating. Finally, the regional deformation is detected, and the operation effect is quantitatively evaluated by registering the 3D scanning model before and after the operation. The test of our proposed system and the simulation of PS operations find that the measurement error of facial geometric features is 0.458 mm, and the area is 0.65 mm2. The ratings of the simulation outcomes provided by panels of PS prove that the system is effective. The manipulated 3D faces are deemed more beautiful compared to the original faces respecting the beauty canons such as facial symmetry and the golden ratio. The proposed algorithm could generate realistic visual effects of PS simulation. It could thus assist the preoperative planning of facial PS.

Abstract Background Colorectal carcinoma evolves through a multitude of molecular events including somatic mutations, epigenetic alterations, and aberrant protein expression, influenced by host immune reactions. One way to interrogate the complex carcinogenic process and interactions between aberrant events is to model a biomarker correlation network. Such a network analysis integrates multidimensional tumor biomarker data to identify key molecular events and pathways that are central to an underlying biological process. Due to embryological, physiological, and microbial differences, proximal and distal colorectal cancers have distinct sets of molecular pathological signatures. Given these differences, we hypothesized that a biomarker correlation network might vary by tumor location. Results We performed network analyses of 54 biomarkers, including major mutational events, microsatellite instability (MSI), epigenetic features, protein expression status, and immune reactions using data from 1380 colorectal cancer cases: 690 cases with proximal colon cancer and 690 cases with distal colorectal cancer matched by age and sex. Edges were defined by statistically significant correlations between biomarkers using Spearman correlation analyses. Large-scale genomic characterization of tumors from prospective cohort studies may yield new insights into cancer pathogenesis. We performed whole-exome sequencing of 619 incident colorectal cancers (CRCs) and integrated the results with tumor immunity, pathology, and survival data. We identified recurrently mutated genes in CRC, such as BCL9L, RBM10, CTCF, and KLF5, that were not previously appreciated in this disease. Furthermore, we investigated the genomic correlates of immune-cell infiltration and found that higher neoantigen load was positively associated with overall lymphocytic infiltration, tumor-infiltrating lymphocytes (TILs), memory T cells, and CRC-specific survival. The association with TILs was evident even within microsatellite-stable tumors. We also found positive selection of mutations in HLA genes and other components of the antigen-processing machinery in TIL-rich tumors. These results may inform immunotherapeutic approaches in CRC. More generally, this study demonstrates a framework for future integrative molecular epidemiology research in colorectal and other malignancies.

The environmental pollution generated by electronic waste (e-waste), waste-gas, and wastewater restricts the sustainable development of society. Environmental-friendly electronics made of degradable, resorbable, and compatible thin-film materials were utilized and explored, which was beneficial for e-waste dissolution and sustainable development. In this paper, we present a literature review about the development of various degradable and disposable thin-films for electronic applications. The corresponding preparation methods were simply reviewed and one of the most exciting and promising methods was discussed: Printing electronics technology. After a short introduction, detailed applications in the environment sensors and eco-friendly devices based on these degradable and compatible thin-films were mainly reviewed, finalizing with the main conclusions and promising perspectives. Furthermore, the future on these upcoming environmental-friendly electronic devices are proposed and prospected, especially on resistive switching devices, showing great potential applications in artificial intelligence (AI) and the Internet of Thing (IoT). These resistive switching devices combine the functions of storage and computations, which can complement the off-shelf computing based on the von Neumann architecture and advance the development of the AI.
