Objectives : Ischemic stroke is the main neurological cause of acquired incapability in adults and a prominent cause of mortality. Several association studies have been conducted to explore the role of candidate genes in this neurological condition. Methods : In the present study, we aimed at identification of association between Glutamate Metabotropic Receptor 7 ( GRM7 ) and risk of ischemic stroke in Iranian population. Two intronic variants within this gene (rs6782011 and rs779867) were genotyped in 318 sporadic ischemic stroke cases and 300 unrelated, healthy controls individuals. Results : No significant difference was found in allele, genotype or haplotype frequencies of these SNPs between cases and controls after correction for multiple comparisons. Conclusion : Consequently, the assessed GRM7 variants are not implicated in risk of ischemic stroke in Iranian population.

L-glutamate is the chief excitatory neurotransmitter in the central nervous system (CNS) which activates metabotropic receptors including the metabotropic glutamate receptor GRM7. Single nucleotide polymorphisms (SNPs) within GRM7 gene have been associated with several psychiatric conditions. In the present study, we assessed association between two GRM7 SNPs (rs6782011 and rs779867) and two neuropsychiatric disorders including attention deficit hyperactive disorder (ADHD) and mood disorders. There were no significant differences in genotype, allele and haplotypes frequencies of the rs6782011 and rs779867 between bipolar disorder 1 (BPD1) patients and controls. The CC genotype of the rs6782011 was significantly associated with BPD2 in recessive model (OR (95% CI) = 1.78 (1.09-2.91), adjusted P value = 0.04) and with ADHD in dominant and co-dominant models (OR (95% CI) = 1.98 (1.11-3.53), adjusted P value = 0.04; OR (95% CI) = 2.27 (1.23-4.17), adjusted P value = 0.04 respectively). The C G haplotype (rs6782011 and rs779867 respectively) was more prevalent among both BPD2 patients (OR (95%CI) = 2.03 (1.36-3.01), adjusted P value = 0.002) and MDD patients (OR (95%CI) = 2.08 (1.37-3.16), adjusted P value = 0.002) compared with controls. The role of metabotropic glutamate receptors in the pathogenesis of schizophrenia or response to antipsychotic treatment has been proposed previously. The aim of the current study was to investigate the associations between two intronic variants within GRM7 gene (rs6782011 and rs779867) and schizophrenia in Iranian population. These two single nucleotide polymorphisms (SNPs) were genotyped in 273 schizophrenic patients and 300 age and sex-matched normal controls. The frequency of A allele of the rs779867 was significantly lower in the schizophrenic patients compared with healthy subjects (OR (95% CI) = 0.71 (0.56-0.89), adjusted P value = 0.008). This SNP was associated with schizophrenia in co-dominant and dominant models (adjusted P values of 0.03 and 0.02 respectively). However, there was no difference in allele and genotype frequencies of the rs6782011 SNP between cases and controls. Consequently, the results of current study further highlight the participation of GRM7 in the pathogenesis of schizophrenia.

Glutamate excitotoxicity has been previously associated with development of multiple sclerosis (MS). The metabotropic glutamate receptor 7 (GRM7) gene is a G protein-coupled receptor that suppresses the cyclic AMP cascade after activation by glutamate. Single nucleotide polymorphisms (SNPs) within this gene have been reported to be associated with neuropsychiatric disorders. In the present study, we assessed associations between two intronic variants of GRM7 (rs6782011 and rs779867) and risk of MS in an Iranian population. The rs779867 was associated with MS risk in recessive model (OR (95% CI) = 0.67 (0.48-0.94)), P-value = 0.02, adjusted P-value = 0.04). There was no significant difference in allele and genotype frequencies of rs6782011 between cases and controls. None of the estimated haplotype blocks of rs6782011 and rs779867 were associated with MS risk in the assessed population. The current study provides some evidence for participation of GRM7 in the pathogenesis of MS and warrants further studies in larger sample sizes., (Copyright © 2019 Elsevier B.V. Autism spectrum disorder (ASD) as a synaptopathy is revealed to be pertained to aberrant glutamatergic neurotransmission. Glutamate receptor, metabotropic 7 (GRM7), a receptor coding gene of this pathway, is a new candidate gene for autism. The aim of this study was to examine if there is a relationship between genetic variants rs779867 and rs6782011 of GRM7 with ASD. The present research was designed as a population-based, case-control study including 518 ASD patients versus 472 control individuals. The results showed that the frequency of rs779867 G/G genotype was significantly higher in ASD patients compared to healthy controls (P = 0.0001). Also, the G allele of this SNP was found to be significantly more frequent in the patients than control group (P = 0.0001). Haplotype analysis exhibited significant association of two estimated block of rs6782011/rs779867 in ASD patients versus control group. We found higher significant frequency of GT haplotype and lower frequencies of AT and AC haplotypes in the patients group compared to healthy controls (P = 0.001, P = 0.006, and P = 0.05, respectively). Our study indicated that the rs779867 polymorphism is associated with ASD; thus, results of this study provide supportive evidence of association of the GRM7 gene with ASD.
