Soil microbiome responses to short-term nitrogen (N) inputs remain uncertain when compared with previous research that has focused on long-term fertilization responses. Here, we examined soil bacterial/archaeal and fungal communities pre- and post-N fertilization in an 8 year-old switchgrass field, in which twenty-four plots received N fertilization at three levels (0, 100, and 200 kg N ha-1 as NH4NO3) for the first time since planting. Soils were collected at two depths, 0–5 and 5–15 cm, for DNA extraction and amplicon sequencing of 16S rRNA genes and ITS regions for assessment of microbial community composition. Baseline assessments prior to fertilization revealed no significant pre-existing divergence in either bacterial/archaeal or fungal communities across plots. The one-time N fertilizations increased switchgrass yields and tissue N content, and the added N was nearly completely removed from the soil of fertilized plots by the end of the growing season. Both bacterial/archaeal and fungal communities showed large spatial (by depth) and temporal variation (by season) within each plot, accounting for 17 and 12–22% of the variation as calculated from the Sq. root of PERMANOVA tests for bacterial/archaeal and fungal community composition, respectively. While N fertilization effects accounted for only ~4% of overall variation, some specific microbial groups, including the bacterial genus Pseudonocardia and the fungal genus Archaeorhizomyces, were notably repressed by fertilization at 200 kg N ha-1. Bacterial groups varied with both depth in the soil profile and time of sampling, while temporal variability shaped the fungal community more significantly than vertical heterogeneity in the soil. These results suggest that short-term effects of N fertilization are significant but subtle, and other sources of variation will need to be carefully accounted for study designs including multiple intra-annual sampling dates, rather than one-time “snapshot” analyses that are common in the literature. To find an electrical conductivity (EC) in the nutrient solution used for pakchoi (Brassica campestris L. ssp. Chinensis) cultivation that optimizes the plant’s physiology, growth, and quality, we conducted an experiment with eight EC treatments (from EC0 to EC9.6) in a hydroponic production system (i.e. soilless culture) under greenhouse condition in Shanghai, China. Plants biomass production, leaf photosynthesis, vegetable quality variables, tissue nitrate and nitrite contents, and antioxidant enzyme activities were measured. The results showed that very high (EC9.6) or low EC (EC0-0.6) treatments clearly decreased plants fresh weight (FW) and dry weight (DW), leaf size, leaf water content, leaf net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), and taste score. Nitrite content, and antioxidant enzyme activities were low in medium EC treatments (EC1.8 and EC2.4), but high in very high or low EC treatments. Leaf relative chlorophyll, ascorbic acid, and nitrate contents increased gradually from low EC to high EC treatments, while crude fiber and soluble sugar contents decreased. Based on growth and quality criteria, the optimal EC treatment would be EC1.8 or EC2.4 for pakchoi in the hydroponic production system. Too high or too low EC would induce nutrient stress, enhance plant antioxidant enzyme activities, and suppress pakchoi growth and quality. [ABSTRACT FROM AUTHOR]

Climate changes, including chronic changes in precipitation amounts, will influence plant physiology and growth. However, such precipitation effects on switchgrass, a major bioenergy crop, have not been well investigated. We conducted a two-year precipitation simulation experiment using large pots (95 L) in an environmentally controlled greenhouse in Nashville, TN. Five precipitation treatments (ambient precipitation, and -50%, -33%, +33%, and +50% of ambient) were applied in a randomized complete block design with lowland "Alamo" switchgrass plants one year after they were established from tillers. The growing season progression of leaf physiology, tiller number, height, and aboveground biomass were determined each growing season. Precipitation treatments significantly affected leaf physiology, growth, and aboveground biomass. The photosynthetic rates in the wet (+50% and +33%) treatments were significantly enhanced by 15.9% and 8.1%, respectively, than the ambient treatment. Both leaf biomass and plant height were largely increased, resulting in dramatically increases in aboveground biomass by 56.5% and 49.6% in the +50% and +33% treatments, respectively. Compared to the ambient treatment, the drought (-33% and -50%) treatments did not influence leaf physiology, but the -50% treatment significantly reduced leaf biomass by 37.8%, plant height by 16.3%, and aboveground biomass by 38.9%. This study demonstrated that while switchgrass in general is a drought tolerant grass, severe drought significantly reduces Alamo’s growth and biomass, and that high precipitation stimulates its photosynthesis and growth. Heat shock is considered an abiotic stress for plant growth, but the effects of heat shock on physiological responses of cucumber plant leaves with and without downy mildew disease are still not clear. In this study, cucumber seedlings were exposed to heat shock in greenhouses, and the responses of photosynthetic properties, carbohydrate metabolism, antioxidant enzyme activity, osmolytes, and disease severity index of leaves with or without the downy mildew disease were measured. Results showed that heat shock significantly decreased the net photosynthetic rate, actual photochemical efficiency, photochemical quenching coefficient, and starch content. Heat shock caused an increase in the stomatal conductance, transpiration rate, antioxidant enzyme activities, total soluble sugar content, sucrose content, soluble protein content and proline content for both healthy leaves and downy mildew infected leaves. These results demonstrate that heat shock activated the transpiration pathway to protect the photosystem from damage due to excess energy in cucumber leaves. Potential resistance mechanisms of plants exposed to heat stress may involve higher osmotic regulation capacity related to an increase of total accumulations of soluble sugar, proline and soluble protein, as well as higher antioxidant enzymes activity in stressed leaves. Heat shock reduced downy mildew disease severity index by more than 50%, and clearly alleviated downy mildew development in the greenhouses. Background: A three-year field experiment was conducted to examine the responses of corn yield and soil nitrous oxide (N2O) emission to various management practices in middle Tennessee. Methodology/Principal Findings: The management practices include no-tillage + regular applications of urea ammonium nitrate (NT-URAN); no-tillage + regular applications of URAN + denitrification inhibitor (NT-inhibitor); no-tillage + regular applications of URAN + biochar (NT-biochar); no-tillage + 20% applications of URAN + chicken litter (NT-litter), no-tillage + split applications of URAN (NT-split); and conventional tillage + regular applications of URAN as a control (CT-URAN). Fertilizer equivalent to 217 kg N ha-1 was applied to each of the experimental plots. Results showed that no-tillage (NT-URAN) significantly increased corn yield by 28% over the conventional tillage (CT-URAN) due to soil water conservation. The management practices significantly altered soil N2O emission, with the highest in the CT-URAN (0.48 mg N2O m-2 h-1) and the lowest in the NT-inhibitor (0.20 mg N2O m-2 h-1) and NT-biochar (0.16 mg N2O m-2 h-1) treatments. Significant exponential relationships between soil N2O emission and water filled pore space were revealed in all treatments. However, variations in soil N2O emission among the treatments were positively correlated with the moisture sensitivity of soil N2O emission that likely reflects an interactive effect between soil properties and WFPS. Conclusion/Significance: Our results indicated that improved fertilizer and soil management have the potential to maintain highly productive corn yield while reducing greenhouse gas emissions. Spatial and temporal patterns of carbon (C) storage in forest ecosystems significantly affect the terrestrial C budget, but such patterns are unclear in the forests in Hainan Province, the largest tropical island in China. Here, we estimated the spatial and temporal patterns of C storage from 1993–2008 in Hainan's forest ecosystems by combining our measured data with four consecutive national forest inventories data. Forest coverage increased from 20.7% in the 1950s to 56.4% in the 2010s. The average C density of 163.7 Mg C/ha in Hainan's forest ecosystems in this study was slightly higher than that of China's mainland forests, but was remarkably lower than that in the tropical forests worldwide. Total forest ecosystem C storage in Hainan increased from 109.51 Tg in 1993 to 279.17 Tg in 2008. Soil C accounted for more than 70% of total forest ecosystem C. The spatial distribution of forest C storage in Hainan was uneven, reflecting differences in land use change and forest management. The potential carbon sequestration of forest ecosystems was 77.3 Tg C if all forested lands were restored to natural tropical forests. To increase the C sequestration potential on Hainan Island, future forest management should focus on the conservation of natural forests, selection of tree species, planting of understory species, and implementation of sustainable practices. For the purposes of forest restoration, carbon (C) fixation, and economic improvement, eucalyptus (Eucalyptus urophylla) has been widely planted in South China. The understory of eucalyptus plantations is often occupied by a dense community of the fern Dicranopteris dichotoma, which intercepts tree canopy leaf litter before it reaches the ground. To understand the effects of this interception of litterfall on C cycling in eucalyptus plantations, we quantified the mass of intercepted litter and the influences of litterfall interception on litter decomposition and soil respiration. The total mass of E. urophylla litterfall collected on the understory was similar to that collected by the traditional litter trap method. All of the eucalyptus litterfall is intercepted by the D. dichotoma canopy. Of the litterfall that was intercepted by D. dichotoma, 20–40% and 60–80% was intercepted by the top (50–100 cm) and bottom (0–50 cm) of the understory canopy, respectively. Intercepted litterfall decomposed faster at the bottom of understory canopy (at the base of the plants) than at the top, and decomposition was slower on the soil surface in the absence of understory than on any location in the understory canopy. Soil respiration was highest when both the understory and litter were present and was lowest when both the understory and litter were absent. These results indicate that litterfall interception changed carbon flow between aboveground and belowground through litter decomposition and soil respiration, which changed carbon cycling in eucalyptus plantations. The temperature sensitivity (Q10) of soil heterotrophic respiration (Rh) is an important ecological model parameter and may vary with temperature and moisture. While Q10 generally decreases with increasing temperature, the moisture effects on Q10 have been controversial. To address this, we conducted a 90-day laboratory incubation experiment using a subtropical forest soil with a full factorial combination of five moisture levels (20%, 40%, 60%, 80%, and 100% water holding capacity - WHC) and five temperature levels (10, 17, 24, 31, and 38°C). Under each moisture treatment, Rh was measured several times for each temperature treatment to derive Q10 based on the exponential relationships between Rh and temperature. Microbial biomass carbon (MBC), microbial community structure and soil nutrients were also measured several times to detect their potential contributions to the moisture-induced Q10 variation. We found that Q10 was significantly lower at lower moisture levels (60%, 40% and 20% WHC) than at higher moisture level (80% WHC) during the early stage of the incubation, but became significantly higher at 20%WHC than at 60% WHC and not significantly different from the other three moisture levels during the late stage of incubation. In contrast, soil Rh had the highest value at 60% WHC and the lowest at 20% WHC throughout the whole incubation period. Variations of Q10 were significantly associated with MBC during the early stages of incubation, but with the fungi-to-bacteria ratio during the later stages, suggesting that changes in microbial biomass and community structure are related to the moisture-induced Q10 changes. This study implies that global warming’s impacts on soil CO2 emission may depend upon soil moisture conditions. Based on the isometric hypothesis, belowground plant biomass (MB) should scale isometrically with aboveground biomass (MA) and the scaling exponent should not vary with environmental factors. We tested this hypothesis using a large forest biomass database collected in China. Allometric scaling functions relating MB and MA were developed for the entire database and for different groups based on tree age, diameter at breast height, height, latitude, longitude or elevation. To investigate whether the scaling exponent is independent of these biotic and abiotic factors, we analyzed the relationship between the scaling exponent and these factors. Overall MB was significantly related to MA with a scaling exponent of 0.964. The scaling exponent of the allometric function did not vary with tree age, density, latitude, or longitude, but varied with diameter at breast height, height, and elevation. The mean of the scaling exponent over all groups was 0.986. Among 57 scaling relationships developed, 26 of the scaling exponents were not significantly different from 1. Our results generally support the isometric hypothesis. MB scaled near isometrically with MA and the scaling exponent did not vary with tree age, density, latitude, or longitude, but increased with tree size and elevation. While fitting a single allometric scaling relationship may be adequate, the estimation of MB from MA could be improved with size-specific scaling relationships. [ABSTRACT FROM AUTHOR]

Establishing seedlings in subtropical plantations is very important for forest health, succession and management. Information on seedling nutrient concentrations is essential for both the selection of suitable indigenous tree species to accelerate succession of the established plantation and sustainable forest management. In this study, we investigated the concentrations of nitrogen ([N]), phosphorus ([P]), and N∶P ratio in leaves, stems and roots of seedlings of three indigenous tree species (Castanopsis chinensis, Michelia chapensis and Psychotria rubra) transplanted with removing or retaining understory vegetation and litter at two typical subtropical forest plantations (Eucalyptus plantation and native species plantation). We also measured the relative growth rate (RGR) of seedling height, and developed the relationships between RGR and leaf [N], [P] and N∶P ratio. Results showed that treatments of understory vegetation and associated litter (i.e. removal or retained) generally had no significant effects on leaf [N], [P], N∶P ratio and RGR of the transplanted tree seedlings for the experimental period. But among different species, there were significant differences in nutrient concentrations. M. chapensis and P. rubra had higher [N] and [P] compared to C. chinensis. [N] and [P] also varied among different plant tissues with much higher values in leaves than in roots for all indigenous species. RGR of indigenous tree seedlings was mostly positively correlated with leaf [N] and [P], but negatively correlated with leaf N∶P ratio. Considering the low [P] and high N∶P ratio observed in the introduced indigenous tree seedlings, we propose that the current experimental plantations might be P limited for plant growth.
