Ageing is usually accompanied by a decline in immune and neuroendocrine functions. To elucidate the mechanisms underlying age-related immunosuppression, the functions and surface phenotypes of peritoneal cells in the monocyte/macrophage lineage from old mice were investigated. The role of glucocorticoids (GC) in the immunomodulation was also examined. Proliferative responses of spleen cells from control mice stimulated with concanavalin A (Con A) were significantly suppressed by adding peritoneal exudate cells from old mice. Flow cytometry analysis revealed that the proportion of MAC-1+ cells with a high density of type II or type III receptor for the Fc portion of IgG (Fc gamma RII/IIIbright cells) was increased markedly in the periotoneal exudate cells from old mice. The prominent suppressor activity for Con A responses of control spleen cells was found in the Fc gamma RII/IIIbright cells, whereas MAC-1+ cells with a low density of Fc gamma RII/III (Fc gamma RII/IIIdull cells) did not suppress the Con A responses. On the other hand, both the basal corticosterone concentrations in serum and the mRNA expression for GC receptor in peritoneal exudate cells increased significantly in old mice. Furthermore, the proportion of Fc gamma RII/IIIbright cells in peritoneal exudate cells from old mice was normalized on administration of the GC antagonist RU 38,486 (mifepristone). These results suggest that the increase in basal corticosterone concentrations in old mice induces the generation of Fc gamma RII/IIIbright suppressor cells, possibly leading to the immune-suppressive state.

Only gamma-chain T-cell receptor transcripts utilizing V-1 subgroup gene segments were found in peripheral blood lymphocytes from a patient with Omenn's syndrome. gamma-Chain T-cell receptor transcripts utilizing the V gamma 9 (V-II subgroup) gene segment were absent in peripheral blood lymphocytes from this patient. V gamma 9 J gamma 1.2 C gamma 1 rearrangements are those primarily found in peripheral blood lymphocytes (70 to 85%) from normal donors.

Immunoregulatory states in acute cold-stressed or cold-acclimatized mice were investigated. When male C57BL/6 mice were exposed to environmental temperature of 5 degrees for 24 hr (acute cold stress), the spleen cells showed depressed proliferative responses to stimulation with concanavalin A (Con A) or lipopolysaccharide (LPS) compared with control mice (reared at 25 degrees). The proportion of Thy-1.2+ cells increased significantly in spleens from these acute cold-stressed mice. The Con A responses of T-enriched cells from acute cold-stressed mice were restored to the normal level by adding plastic-adherent cells from control mice. Further, adherent cells from acute cold-stressed mice markedly suppressed the Con A responses of control spleen cells. Thus, the plastic-adherent cells appeared to be responsible for the suppressed Con A responses. On the other hand, proliferative responses to Con A or LPS were elevated in spleen cells from mice exposed to 5 degrees for 3 weeks (cold acclimatization). A significant decrease of Thy-1.2+ cells was detected in these spleens. It was shown that the enhanced proliferative responses were attributable to functional alterations of the plastic adherent cell population but not to those of lymphoid cell population. These findings indicate that the low or high responsiveness of spleen cells to Con A observed in cold-stressed or cold-acclimatized mice, respectively, may be due to a mechanism including the contrary modulations of functions of cells of mononuclear phagocyte lineage.

Addition of protoscoleces (PSC) of Echinococcus multiocularis suppressed proliferative responses in spleen cells stimulated with concanavalin A (Con A) on day 3 of culture. The suppression was not observed in the spleen cell population depleted of CD8+ cells but observed in the population depleted of CD4+ cells, suggesting the involvement of the CD8+ T cells in the apparent suppression. Indeed, flow cytometry analysis revealed that the proportion of CD8+ cells markedly increased in the Con A cultures containing PSC. Furthermore, when spleen cells were co-cultured with PSC alone, marked increase in the proportion of CD8+ cells as well as B220+ cells was observed. Addition of these increasing CD8+ cells suppressed the proliferative responses of fresh spleen cells stimulated with Con A. These findings suggest that the low responsiveness of spleen cells to Con A on day 3 of culture in the presence of PSC is attributable to an active suppressor mechanism including CD8+ T-suppressor cells generated by stimulation with PSC.

Summary A patient with apparent X-linked agammaglobulinaemia was found to be inordinately susceptible to anaphylactoid reactions to intramuscular injections of gammaglobulin. The patient was found also to have low levels of C1 esterase inhibitor (C1 INH). The possibility that the C1 INH deficiency and in this patient, whether genetic or acquired, fostered the susceptibility to the production of anaphylactoid reactions after gammaglobulin injections urges further studies of the association of C1 INH deficiency and anaphylactoid reactions to gammaglobulin injections. The possibility that C1 INH levels like C1q levels may be low in hypogammaglobulinaemic patients as a consequence of increased catabolism of this regulator of the complement system when IgG levels are low is considered.

Severe combined immunodeficiency disease (SCID) is a congenital disorder of severe B- and T-lymphocyte dysfunction in which several pathogenic mechanisms have been identified. The present study describes a female child with SCID who had a primary defect in the ability of T cells to secrete interleukin 2 (IL-2). B- and T-cell numbers were normal, but their functions were severely deficient. Mitogen and antigen-driven lymphoproliferative responses were diminished but were correctable in vitro with recombinant IL-2 (rIL-2). The patient's phytohemagglutinin-stimulated lymphocytes expressed IL-2 receptors normally. Despite the presence of the gene for IL-2, the patient's cells were grossly deficient in messenger RNA for IL-2 and endogenous IL-2 production. Pokeweed mitogen-driven B-cell differentiation was decreased and was not corrected by the addition of normal T cells to the B cells. Two attempts at immune reconstitution by haploidentical bone marrow transplantation failed. Therapy with rIL-2 (30,000 units/kg, given daily i.v.) resulted in marked clinical improvement as well in improved T-cell functions. The child, now 3 yr old, has been on rIL-2 therapy for 2 yr and receives rIL-2 (30,000 units/kg) three times weekly at home. This case study points to a new direction in the treatment of such disorders with rIL-2.

Two electrophoretic variants of murine complement component 3 (C3) were detected by using high-voltage electrophoresis of fresh mouse serum in agarose gels. Most of the inbred strains tested were homozygous for the S allele (for the slow-migrating variant); only four out of 46 strains had the alternative F allele (fast variant). Pen-bred Swiss-Webster animals belonged to one of three phenotypes--S, F, or SF--and the genes responsible for this variation segregated in a strictly Mendelian manner. In three such crosses, with 5* offspring, C3 segregated with H-2 in 46 instances, corresponding to a recombination frequency of approximately equal to 0.12.

To test the possibility that autoimmunity could be related to increased levels of anti-bovine antibodies and/or circulating immune complexes in selective IgA deficiency, we studied the sera of 30 consecutive patients for the quantitative level of antibody to bovine milk, the presence of antigen--antibody complexes and 10 selected autoantibodies. Higher titres of anti-milk antibody and circulating immune complexes were to be correlated with positive serological tests of autoimmunity in these patients, and rheumatoid arthritis (three cases) and neurological disease (four cases) were found in individuals who had both milk precipitins and circulating immune complexes. We suggest that the chronic excessive permeability of the gastrointestinal tract in selective IgA deficiency may permit the excessive absorption of many food proteins, leading to the formation of antigen--antibody complexes and autoimmunity.

The presence of Fc receptors, but not C3 receptors, on human and bovine spermatozoa was shown by sperm-induced rosette formation of antibody-treated ox erythrocytes and by immunofluorescence of aggregated human gammaglobulin (AHG) treated spermatozoa. Human and bovine spermatozoa bound AHG as well as circulating immune complexes (CIC) from human sera; monomeric IgG bound to these cells to a much lesser extent. Binding of AHG to the sperm surface also occurred when the AHG was suspended in either buffer or in serum that had been heated at 56 degrees C for 30 min. A bovine spermatozoa radioimmunoassay for the detection of immune complexes in human sera was developed and was shown to be comparable to the Raji cell radioimmunoassay (r = 0 . 81, P < 0 . 01) in the quantitation of soluble immune complexes from a variety of human sera. An enzyme-linked immunoassay was then developed, using bovine spermatozoa fixed to wells of microtitre plates, to simplify the immune complex detection assay. The technique provides an analysis that is rapid, does not require radioactive chemicals, expensive equipment, or cultured cells and yields values for immune complex determination both comparable and complementary to the Raji cell radioimmunoassay (r = 0 . 70, P < 0 . 01). The sperm assays detected soluble immune complexes in sera of vasectomized males, cancer patients and patients with autoimmune or dermatological diseases.

Spontaneous sperm agglutination was observed in the ejaculate of a 39-year-old male (patient Z). Using an enzyme-linked immunosorbent assay (ELISA) with spermatozoa fixed to wells of a microtitre plate it was determined that IgA was associated with patient Z's spermatozoa but not with a pooled sample of spermatozoa from fertile men Z's seminal fluid agglutinated human spermatozoa (titre 1:16). In contrast, no sperm-agglutinating or immobilizing activity was present in the patient's serum. Agglutination was prevented by preincubation of the seminal fluid with anti-IgA antibody but not with anti-IgA antibody. IgG from Z's seminal fluid readily bound to human spermatozoa, as determined by the ELISA assay. IgA from a pool of seminal fluids from fertile men or from 18 human sera bound to spermatozoa to a much lesser extent. By sucrose-gradient velocity centrifugation it was demonstrated that the anti-sperm IgA in patient Z's seminal fluid was 11S. Finally, antibody to secretory component removed the anti-sperm IgA from Z's seminal fluid. These data provide evidence that secretory IgA with anti-sperm activity is produced in the male genital tract. Furthermore, a new methodology is presented for the determination of antibodies to spermatozoa.

Unmeasurable total haemolytic complement (C) was observed in serum of a patient with untreated chronic lymphocytic leukaemia and recurrent non-hereditary angioedema. Analysis of C components immunochemically demonstrated a marked reduction of C1q and C1s inhibitor, undetectable C1r, C1s and an elevated B. Haemolytic C1, C4 and C2 were less than 5 percent of normal, functional C1s inhibitor was absent. Cryoglobulin and C1q precipitins were present in the serum. Of special interest was the presence of high levels of cold-reactive antilymphocyte antibody, determined by both C-dependent cytotoxicity and indirect immunofluorescence. The antibody exhibited specificities for both autologous lymphocytes and lymphocytes from normal donors; cytotoxic activity for autologous leukaemia cells was removed by absorption with normal isologous tonsil lymphocytes. Specific enrichment of this antibody relative to the serum level was demonstrated in the cryoglobulin and its isolated 19S fractions. Free lymphocyte surface antigen was also demonstrated by gel diffusion using specific rabbit antilymphocyte antiserum. These data strongly suggest the presence of pathogenetically significant circulating complexes of lymphocyte surface antigen and specific antibody in certain patients with CLL.

The effect of ultraviolet-inactivated feline leukaemia virus (UV-FeLV) on the development of feline immunoglobulin-secreting cells (ISC) was investigated using a reverse haemolytic plaque assay. Low concentrations of UV-FeLV at 2 X 10(-4) to 2.0 micrograms/ml stimulated the production of ISC. By contrast, the same concentration of UV-FeLV suppressed the development of pokeweed mitogen (PWM)-driven ISC. Maximum suppression of ISC occurred at 50 micrograms/ml of UV-FeLV. The generation of an interferon resistant to acid, heat and sodium dodecyl sulphate (SDS) in the media of lymphocyte cultures incubated with PWM was also significantly suppressed in the presence of 0.2 microgram/ml of UV-FeLV. These findings suggest that non-infectious viral particles appear to modify feline immunoglobulin and interferon-secreting systems.

An enzyme-linked immunosorbent assay (ELISA) for the quantitation of immune complexes in sera has been developed using Raji cells. In this assay the Raji cell-adherent complexes are detected by an alkaline phosphatase-conjugated rabbit anti-human IgG and the results are obtained by spectrophotometric measurements of the concentration of yellow colour produced after the enzyme's substrate, p-nitrophenyl phosphate, is added. This assay is as sensitive and reproducible as the Raji cell radioimmune assay using 125I, as it can detect as little as 16 micrograms of immune complex equivalent to heat-aggregated IgG/ml in serum samples. This method is inexpensive, convenient and most importantly, avoids the biohazards of using an isotope.

T cell lines or clones from two patients, one with a partial DiGeorge syndrome and one with severe common variable immunodeficiency expressed disulfide-linked gamma delta T cell antigen receptor (TCR) comprised of a gamma-chain polypeptide of 40-43 kD, and a delta-chain polypeptide of 37-40 kD. This gamma delta TCR appears to be similar to that found on T cell clones, and lines derived from peripheral blood lymphocytes from normal donors. Previous studies have shown that T cell lines derived from the peripheral blood of patients with immunodeficiency disorders express non-disulfide-linked gamma delta TCR. In contrast to the latter and coincident with findings in the present study, the vast majority of T cell lines and clones derived from the peripheral blood of normal donors express disulfide-linked gamma delta TCR.

Using isoelectric focusing in polyacrylamide gel and a hemolytic assay for development of patterns, extensive, structural polymorphism in human C8 has been delineated. Two alleles, C8A and C8B, have been identified in orientals, with gene frequencies of 0.655 and 0.345. In blacks, what appears to be a third common allele was found, so that frequencies were 0.692, 0.259, and 0.049 for C8A, C8B, and C8A1. In whites, C8A1 was rare with a frequency of 0.003, and frequencies for C8A and C8B were 0.649 and 0.349. Inheritance was autosomal codominant in family studies and the distribution of types in random unrelated populations fit the Hardy-Weinberg equilibrium in all groups. C8 allotypes have been determined for two previously studied families, each with a homozygous C8-deficient propositus. This study suggests that C8 deficiency is a silent or null allele of the C8 structural locus, and that half normal levels of C8 cannot be used as a single criterion for the establishment of heterozygous C8 deficiency. C8 allotypes, as well as 18 other autosomal markers, were also determined for 24 families. The C8 structural locus is not closely linked to these markers, including the human histocompatibility loci complex.

Serum levels of circulating immune complexes (CIC) assayed by the Raji cell radioimmunoassay, total haemolytic complement (TCH50), Clq and C3 were correlated with clinical stage, histological type, age, sex and treatment of eighty-six children with Hodgkin's disease over a period of 4 years. Most significant findings were the changes of levels of CIC, TCH50, Clq and C3 during disease activity and following treatment. Significant perturbations were also seen in association with relapse. Levels of C and CIC were significantly elevated (P less than 0.001) at the time of diagnosis prior to splenectomy and/or any treatment. In the group before treatment, 81 percent of CIC levels were above 16 micrograms/ml with a maximum value of 1120 micrograms/ml. During treatment 33 percent were still above normal with a maximum of 320 micrograms/ml. Within 1 year after cessation of treatment, 37 percent also remained above normal levels with a maximum of 240 micrograms/ml. At relapse prior to treatment, 63 percent were again elevated with a maximum of 1280 micrograms/ml. The most significant difference on TCH50 levels relates to treatment periods. Sera of patients with active disease who are previously untreated show elevation of TCH50 levels (P less than 0.001) (average 127 CH50 mu/ml. During and after treatment eht TCH50 levels drop to 96 and 102 CH50 mu/ml, as compared to normal control of 100 CH50 mu/ml. In sera of patients at the first, second or third relapse, the combined TCH50 levels are significantly different from controls and across treatment periods (P less than 0.005).

Purified protein A from Staphylococcus aureus Cowan I was injected intraperitoneally or was incorporated in filters ex vivo through which plasma from cats with feline leukemia virus (FeLV)-associated leukemia-lymphoma was passed. Before treatment, 65% of the FeLV-infected cats were anemic, and 70% were thrombocytopenic. Concomitant infections, or immune-mediated disease, was common. During treatment 50% of the cats with FeLV-associated disease improved objectively with normal posttreatment hematocrits, thrombocyte and leukocyte counts, disappearance of dysplastic hematologic elements, and correction of marrow dyscrasias. A 33% response to treatment occurred in cats with unequivocal manifestations of malignant disease and was characterized by reductions in tumor size and marrow and peripheral blood neoplastic cell populations. Clearance of FeLV viremia was documented in 28% of the treated cats. The several possible mechanisms by which treatment with staphylococcal protein A causes reduction in the extent of malignant disease are considered.
