Our study aimed to compare the anatomical features of the eyeball, lens, and retina between the two raptor birds of different visual active clock hours: the diurnal common kestrel (Falco tinnunculus rupicilaeformis) and the nocturnal little owl (Athene noctua glaux) using gross, morphometric analysis, histological, and scanning and transmission electron microscopy techniques. The semi-spherical eyeball of the kestrel had less convexity on the anterior surface than on the posterior surface; meanwhile, it was relatively larger in the owl. There is a relationship between the corneal diameter (CD) and the eye axial length (AL). There were significant differences in the retinal layer thickness between the two raptors, in which the diurnal kestrel had a thinner pigmented epithelium and photoreceptor layers compared to the nocturnal owl. Moreover, the inner nuclear and outer nuclear, inner plexiform, and outer plexiform layers in the diurnal kestrel were larger than those in the nocturnal owl. The differences in the pigmented epithelium layer lead to the higher visual acuity and better color vision of the diurnal kestrel compared to the nocturnal owl. The photoreceptor layer in diurnal kestrel was composed of single large and double cones, which are composed of chief cones and accessory cones; meanwhile, the photoreceptor layer in nocturnal owl had only single elongated rods. We concluded that the lens and retina of the two raptors revealed great variations in reflecting the adaptation of each bird to different modes of life. The statistical analysis found a strong positive correlation between the axial length of the eye and the corneal diameter in both birds, as indicated by the Pearson correlation coefficient., Competing Interests: Declarations. A rodent eyeball is a powerful tool for researching the pathomechanisms of many ophthalmic diseases, such as glaucoma, hypertensive retinopathy, and many more. Preclinical experiments enable researchers to examine the efficacy of novel drugs, develop new methods of treatment, or seek new pathomechanisms involved in the disease's onset or progression. A histological examination provides a lot of information necessary to assess the effects of the conducted experiments and can reveal degeneration, tissue remodeling, infiltration, and many other pathologies. In clinical research, there is rarely any chance of obtaining eye tissue suitable for a histological examination, which is why researchers should take advantage of the opportunity offered by the examination of eyeballs from rodents. This manuscript presents a protocol for the histological preparation of rodent eyeballs' sections. The procedure is presented for the eyeballs of mice and rats and has the following steps: (i) harvesting the eyeball, (ii) preserving the eyeball for further analysis, (iii) processing the tissue in paraffin, (iv) preparing slides, (v) staining with hematoxylin and eosin, (vi) assessing the tissue under a light microscope. With the proposed method, the retina can be easily visualized and assessed in detail.

The retina has a very high energy demand but lacks an internal blood supply in most vertebrates. Here we explore the hypothesis that oxygen diffusion limited the evolution of retinal morphology by reconstructing the evolution of retinal thickness and the various mechanisms for retinal oxygen supply, including capillarization and acid-induced haemoglobin oxygen unloading. We show that a common ancestor of bony fishes likely had a thin retina without additional retinal oxygen supply mechanisms and that three different types of retinal capillaries were gained and lost independently multiple times during the radiation of vertebrates, and that these were invariably associated with parallel changes in retinal thickness. Functionally distinct retinal ganglion cells have density and size gradients across the mouse retina, and some degenerative eye diseases follow topographic-specific gradients of cell death. Hence, the anatomical orientation of the retina with respect to the orbit and head is important for understanding the functional anatomy of the retina in both health and disease. However, different research groups use different anatomical landmarks to determine retinal orientation (dorsal, ventral, temporal, nasal poles). Variations in the accuracy and reliability in marking these landmarks during dissection may lead to discrepancies in the identification and reporting of retinal topography. The goal of this study was to compare the accuracy and reliability of the canthus, rectus muscle, and choroid fissure landmarks in reporting retinal orientation. The retinal relieving cut angle made from each landmark during dissection was calculated based on its relationship to the opsin transition zone (OTZ), determined via a custom MATLAB script that aligns retinas from immunostained s-opsin. The choroid fissure and rectus muscle landmarks were the most accurate and reliable, while burn marks using the canthus as a reference were the least. These values were used to build an anatomical map that plots various ocular landmarks in relationship to one another, to the horizontal semicircular canals, to lambda-bregma, and to the earth's horizon. Based on anatomical measurements of refractive structures in the eye, the positions of focused images were computed for several groups of semiaquatic mammals: rodents, a nonpinniped semiaquatic carnivore (the sea otter), and pinniped carnivores (seals, sea lions, and the walrus). In semiaquatic rodents, eye optics enable emmetropia in the air but cause substantial hypermetropia in the water. In semiaquatic carnivores, there are several mechanisms for amphibious vision that focus images on the retina in both air and water. These mechanisms include the potential for a substantial change in the lens shape of sea otters and the presence of the corneal emmetropic window in pinnipeds. Elephants are precocial mammals that are relatively mature as newborns and mobile shortly after birth. To determine whether the retina of newborn elephants is capable of supporting the mobility of elephant calves, we compared the retinal structures of 2 newborn elephants (1 African and 1 Asian) and 2 adult animals of both species by immunohistochemical and morphometric methods. For the first time, we present here a comprehensive qualitative and quantitative characterization of the cellular composition of the newborn and the adult retinas of 2 elephant species. We found that the retina of elephants is relatively mature at birth. All retinal layers were well discernible, and various retinal cell types were detected in the newborns, including Müller glial cells (expressing glutamine synthetase and cellular retinal binding protein; CRALBP), cone photoreceptors (expressing S-opsin or M/L-opsin), protein kinase Cα-expressing bipolar cells, tyrosine hydroxylase-, choline acetyltransferase (ChAT)-, calbindin-, and calretinin-expressing amacrine cells, and calbindin-expressing horizontal cells. The retina of newborn elephants contains discrete horizontal cells which coexpress ChAT, calbindin, and calretinin. While the overall structure of the retina is very similar between newborn and adult elephants, various parameters change after birth. The postnatal thickening of the retinal ganglion cell axons and the increase in ganglion cell soma size are explained by the increase in body size after birth, and the decreases in the densities of neuronal and glial cells are explained by the postnatal expansion of the retinal surface area. The expression of glutamine synthetase and CRALBP in the Müller cells of newborn elephants suggests that the cells are already capable of supporting the activities of photoreceptors and neurons. As a peculiarity, the elephant retina contains both normally located and displaced giant ganglion cells, with single cells reaching a diameter of more than 50 µm in adults and therefore being almost in the range of giant retinal ganglion cells found in aquatic mammals. Some of these ganglion cells are displaced into the inner nuclear layer, a unique feature of terrestrial mammals. Background: Optical coherence tomography (OCT), can be used in clinical practice to provide high resolution cross-sectional images of the retina, optic disc and macula structure. These measurements can be useful for early detection, diagnosis, monitoring and treatment guidance for retinal diseases. Therefore, repeatability of measurements in OCT is of great importance., Methods: Macula and optic disc parameters from the right eye of 30 healthy subjects were obtained twice with the Canon OCT-HS100 and Zeiss Cirrus HD-OCT 5000. Repeatability was evaluated by use of the coefficient of repeatability (CR) and the coefficient of repeatability as a percentage of the mean (CR%), and the obtained measurements were compared between the instruments., Results: CR% of optic disc parameters ranged between 0.90 and 22.22% and 0.00 and 16.00% with the Canon and Zeiss OCT respectively. For macular parameters CR% ranged between 0.62 and 2.81% and 0.99 and 1.81% with the Canon and Zeiss OCT respectively. No statistical difference could be found when comparing the CR of all macular and disc measurements between the instruments. Compared to our previously published data repeatability has significantly improved with the inclusion of automatic tracking systems with both the Canon and Zeiss OCT., Conclusion: Automatic tracking function improves repeatability in both Canon OCT-HS100 and Zeiss Cirrus HD-OCT 5000. However, measurements generated by the two instruments are still not interchangeable.

For compass orientation many insects rely on the pattern of sky polarization, but some species also exploit the sky chromatic contrast. Desert locusts, Schistocerca gregaria, detect polarized light through a specialized dorsal rim area (DRA) in their compound eye. To better understand retinal mechanisms underlying visual navigation, we compared opsin expression, spectral and polarization sensitivities and response-stimulus intensity functions in the DRA and main retina of the locust. In addition to previously characterized opsins of long-wavelength-absorbing (Lo1) and blue-absorbing visual pigments (Lo2), we identified an opsin of an ultraviolet-absorbing visual pigment (LoUV). DRA photoreceptors exclusively expressed Lo2, had peak spectral sensitivities at 441 nm and showed high polarization sensitivity (PS 1.3-31.7). In contrast, ommatidia in the main eye co-expressed Lo1 and Lo2 in five photoreceptors, expressed Lo1 in two proximal photoreceptors, and Lo2 or LoUV in one distal photoreceptor. Correspondingly, we found broadband blue- and green-peaking spectral sensitivities in the main eye and one narrowly tuned UV peaking receptor. Polarization sensitivity in the main retina was low (PS 1.3-3.8). V-log I functions in the DRA were steeper than in the main retina, supporting a role in polarization vision. Desert locusts occur as two morphs, a day-active gregarious and a night-active solitarious form. In solitarious locusts, sensitivities in the main retina were generally shifted to longer wavelengths, particularly in ventral eye regions, supporting a nocturnal lifestyle at low light levels. We present a new technique for simulating retinal image formation by tracing a large number of rays from objects in three dimensions as they pass through the optic apparatus of the eye to objects. Simulating human optics is useful for understanding basic questions of vision science and for studying vision defects and their corrections. Because of the complexity of computing such simulations accurately, most previous efforts used simplified analytical models of the normal eye. This makes them less effective in modeling vision disorders associated with abnormal shapes of the ocular structures which are hard to be precisely represented by analytical surfaces. We have developed a computer simulator that can simulate ocular structures of arbitrary shapes, for instance represented by polygon meshes. Topographic and geometric measurements of the cornea, lens, and retina from keratometer or medical imaging data can be integrated for individualized examination. We utilize parallel processing using modern Graphics Processing Units (GPUs) to efficiently compute retinal images by tracing millions of rays. A stable retinal image can be generated within minutes. We simulated depth-of-field, accommodation, chromatic aberrations, as well as astigmatism and correction. Vultures are highly reliant on their sensory systems for the rapid detection and localization of carrion before other scavengers can exploit the resource. In this study, we compared eye morphology and retinal topography in two species of New World vultures (Cathartidae), turkey vultures (Cathartes aura), with a highly developed olfactory sense, and black vultures (Coragyps atratus), with a less developed sense of olfaction. We found that eye size relative to body mass was the same in both species, but that black vultures have larger corneas relative to eye size than turkey vultures. However, the overall retinal topography, the total number of cells in the retinal ganglion cell layer, peak and average cell densities, cell soma area frequency distributions, and the theoretical peak anatomical spatial resolving power were the same in both species. This suggests that the visual systems of these two species are similar and that vision plays an equally important role in the biology of both species, despite the apparently greater reliance on olfaction for finding carrion in turkey vultures., (Copyright © 2013 Wiley Periodicals, Inc.)

Despite the large body of literature on ecomorphological adaptations to foraging in waterfowl, little attention has been paid to their sensory systems, especially vision. Here, we compare eye shape and retinal topography across 12 species representing 4 different foraging modes. Eye shape was significantly different among foraging modes, with diving and pursuit-diving species having relatively smaller corneal diameters compared to non-diving species. This may be associated with differences in ambient light intensity while foraging or an ability to tightly constrict the pupil in divers in order to facilitate underwater vision. Retinal topography was similar across all species, consisting of an oblique visual streak, a central area of peak cell density, and no discernible fovea. Because the bill faces downwards when the head is held in the normal posture in waterfowl, the visual streak will be held horizontally, allowing the horizon to be sampled with higher visual acuity. Estimates of spatial resolving power were similar among species with only the Canada goose having a higher spatial resolution. Overall, we found no evidence of ecomorphological adaptations to different foraging modes in the retinal ganglion cell layer in waterfowl. Rather, retinal topography in these birds seems to reflect the 'openness' of their habitats.

Retinal topography, cell density and sizes of ganglion cells in the killer whale (Orcinus orca) were analyzed in retinal whole mounts stained with cresyl violet. A distinctive feature of the killer whale's retina is the large size of ganglion cells and low cell density compared to terrestrial mammals. The ganglion cell diameter ranged from 8 to 100 µm, with the majority of cells within a range of 20-40 µm. The topographic distribution of ganglion cells displayed two spots of high cell density located in the temporal and nasal quadrants, 20 mm from the optic disk. The high-density areas were connected by a horizontal belt-like area passing below the optic disk of the retina. Peak cell densities in these areas were evaluated. Mean peak cell densities were 334 and 288 cells/mm(2) in the temporal and nasal high-density areas, respectively. With a posterior nodal distance of 19.5 mm, these high-density data predict a retinal resolution of 9.6' (3.1 cycles/deg.) and 12.6' (2.4 cycles/deg.) in the temporal and nasal areas, respectively, in water., (Copyright © 2012 S. A three-beam spectral domain optical coherence tomography system (OCT) whose center wavelength is 840 nm was developed. The three beams focus on fundus 3.1 mm apart from each other and are detected by a single line sensor. The distance between the beams is fixed and the beams scan a total area of 10×10  mm² while keeping this separation during three-dimensional (3-D) measurement. The line rate of the sensor is 70 kHz, therefore the total speed is equivalent to 210k A-scans per second in this system. A 1000(x)×500(z)×250(y) voxel volumetric 3D OCT data set can be acquired within 2 s. Images of a model eye, a healthy human eye and a diseased eye taken by this system are shown and evaluated. The image quality of one B-Scan is as good as an image from a single-beam OCT. Adjustment among the beams is solved by additional signal processing using a model eye. A multi-beam OCT has the potential not only for high speed imaging but also functional imaging although problems such as compensation among the beams and motion artifacts must be solved.

Eye morphology and the retinal topography of animals that live in either 'open' (e.g., grassland) or 'enclosed' (e.g., forest) terrestrial habitats show common adaptations to constraints imposed by these different habitat types. Although relationships between habitat and the visual system are well documented in most vertebrates, relatively few studies have examined this relationship in birds. Here, we compare eye shape and retinal topography across seven species from the family Phasianidae (Galliformes) that are diurnally active in either open or enclosed habitats. Species from enclosed habitats have significantly larger corneal diameters, relative to transverse diameters, than species from open habitats, which we predict serves to enhance visual sensitivity. Retinal topography, however, was similar across all seven species and consisted of a centrally positioned area centralis and a weak horizontal visual streak, with no discernible fovea. In the Japanese quail (Coturnix japonica), there was also a dorso-temporal extension of increased neuron density and, in some specimens, a putative area dorsalis. The total number of neurons in the retinal ganglion cell layer was correlated with retinal whole-mount area. Average and peak neuron densities were similar across species, with the exception of the Japanese quail, which had greater average and peak densities. Peak anatomical spatial resolving power was also similar among species, ranging from approximately 10-13 cycles/°. Overall, the pattern of retinal topography we found in phasianids is associated with ground-foraging in birds and presumably facilitates the identification of small food items on the ground as well as other visually guided behaviors, irrespective of habitat type.

The modular visual system of jumping spiders (Salticidae) divides characteristics such as high spatial acuity and wide-field motion detection between different pairs of eyes. A large pair of telescope-like anterior-median (AM) eyes is supported by 2-3 pairs of 'secondary' eyes, which provide almost 360 degrees of visual coverage at lower resolution. The AM retinae are moveable and can be pointed at stimuli within their range of motion, but salticids have to turn to bring targets into this frontal zone in the first place. We describe how the front-facing pair of secondary eyes (anterior lateral, AL) mediates this through a series of whole-body 'tracking saccades' in response to computer-generated stimuli. We investigated the 'response area' of the AL eyes and show a clear correspondence between the physical margins of the retina and stimulus position at the onset of the first saccade. Saccade frequency is maximal at the margin of AL and AM fields of view. Furthermore, spiders markedly increase the velocity with which higher magnitude tracking saccades are carried out. This has the effect that the time during which vision is impaired due to motion blur is kept at an almost constant low level, even during saccades of large magnitude.

Purpose:   We provide an account of the relationships between eye shape, retinal shape and peripheral refraction., Recent Findings:   We discuss how eye and retinal shapes may be described as conicoids, and we describe an axis and section reference system for determining shapes. Explanations are given of how patterns of retinal expansion during the development of myopia may contribute to changing patterns of peripheral refraction, and how pre-existing retinal shape might contribute to the development of myopia. Direct and indirect techniques for determining eye and retinal shape are described, and results are discussed. There is reasonable consistency in the literature of eye length increasing at a greater rate than height and width as the degree of myopia increases, so that eyes may be described as changing from oblate/spherical shapes to prolate shapes. However, one study indicates that the retina itself, while showing the same trend, remains oblate in shape for most eyes (discounting high myopia). Eye shape and retinal shape are not the same and merely describing an eye shape as being prolate or oblate is insufficient without some understanding of the parameters contributing to this; in myopia a prolate eye shape is likely to involve both a steepening retina near the posterior pole combined with a flattening (or a reduction in steepening compared with an emmetrope) away from the pole., Summary:   In the recent literature, eye and/or retinal shape have often been inferred from peripheral refraction, and, to a lesser extent, vice versa. Because both the eye's optics and the retinal shape contribute to the peripheral refraction, and there is large variation in the latter, this inference should be made cautiously. Recently retinal shape has been measured independent of optical methods using magnetic resonance imaging. Spontaneous activity is thought to regulate synaptogenesis in many parts of the developing nervous system. In vivo evidence for this regulation, however, is scarce and comes almost exclusively from experiments in which normal activity was reduced or blocked completely. Thus, whether spontaneous activity itself promotes synaptogenesis or plays a purely permissive role remains uncertain. In addition, how activity influences synapse dynamics to shape connectivity and whether its effects among neurons are uniform or cell-type-dependent is unclear. In mice lacking the cone-rod homeobox gene (Crx), photoreceptors fail to establish normal connections with bipolar cells (BCs). Here, we find that retinal ganglion cells (RGCs) in Crx⁻/⁻ mice become rhythmically hyperactive around the time of eye opening as a result of increased spontaneous glutamate release from BCs. This elevated neurotransmission enhances synaptogenesis between BCs and RGCs, without altering the overall circuit architecture. Using live imaging, we discover that spontaneous activity selectively regulates the rate of synapse formation, not elimination, in this circuit. Reconstructions of the connectivity patterns of three BC types with a shared RGC target further revealed that neurotransmission specifically promotes the formation of multisynaptic appositions from one BC type without affecting the maintenance or elimination of connections from the other two. Although hyperactivity in Crx⁻/⁻ mice persists, synapse numbers do not increase beyond 4 weeks of age, suggesting closure of a critical period for synaptic refinement in the inner retina. Interestingly, despite their hyperactivity, RGC axons maintain normal eye-specific territories and cell-type-specific layers in the dorsal lateral geniculate nucleus.

The four-eyed fish, Anableps anableps, has eyes with unusual morphological adaptations for simultaneous vision above and below water. The retina, for example, is divided such that one region receives light from the aerial field and the other from the aquatic field. To understand better the adaptive value of this partitioned retina, we characterized photoreceptor distribution using in situ hybridization. Cones expressing sws1, sws2b and rh2-2 (i.e. UV, and short wavelength-sensitive) opsins were found throughout the retina, whereas cones expressing rh2-1 (middle wavelength-sensitive) were largely limited to the ventral retina and those expressing lws (long wavelength-sensitive) opsins were only expressed in the dorsal retina. We next asked when this pattern evolved relative to the 'four-eyed' morphology. We characterized opsin expression in Jenynsia onca, a member of the sister genus to Anableps with typical teleost eye morphology. In J. onca, sws1, sws2b, rh2-2 and rh2-1 opsins were expressed throughout the retina; while lws opsins were not expressed in the ventral retina. Thus, the change that coincides with the evolution of unusual anablepid eye morphology is the loss of rh2-1 expression in the dorsal retina, probably to accommodate increased lws opsin expression. The retinal area that samples aerial light appears not to have changed with respect to photoreceptor transcription.

The next-generation retinal prostheses feature high image resolution and chronic implantation. These features demand the delivery of power as high as 100 mW to be wireless and efficient. A common solution is the 2-coil inductive power link, used by current retinal prostheses. This power link tends to include a larger-size extraocular receiver coil coupled to the external transmitter coil, and the receiver coil is connected to the intraocular electrodes through a trans-sclera trans-choroid cable. In the long-term implantation of the device, the cable may cause hypotony (low intraocular pressure) and infection. However, when a 2-coil system is constructed from a small-size intraocular receiver coil, the efficiency drops drastically which may induce over heat dissipation and electromagnetic field exposure. Our previous 2-coil system achieved only 7% power transfer. This paper presents a fully intraocular and highly efficient wireless power transfer system, by introducing another inductive coupling link to bypass the trans-sclera trans-choroid cable. With the specific equivalent load of our customized 512-electrode stimulator, the current 3-coil inductive link was measured to have the overall power transfer efficiency around 36%, with 1-inch separation in saline. The high efficiency will favorably reduce the heat dissipation and electromagnetic field exposure to surrounding human tissues. The effect of the eyeball rotation on the power transfer efficiency was investigated as well. The efficiency can still maintain 14.7% with left and right deflection of 30 degree during normal use. The surgical procedure for the coils' implantation into the porcine eye was also demonstrated.

The compound eye of the fruit fly, Drosophila melanogaster, has for decades been used extensively to study a number of critical developmental processes including tissue development, pattern formation, cell fate specification, and planar cell polarity. To a lesser degree it has been used to examine the cell cycle and tissue proliferation. Discovering the mechanisms that balance tissue growth and cell death in developing epithelia has traditionally been the realm of those using the wing disc. However, over the last decade a series of observations has demonstrated that the eye is a suitable and maybe even preferable tissue for studying tissue growth. The purpose of this study is to identify evolutionary origin and fate of anatomic features of the duck-billed platypus eye. Eyes from the duck-billed platypus and four key evolutionary basal vertebrates (Pacific hagfish, north hemisphere sea lamprey, and Australian and South American lungfishes) were prepared for light microscopy. In addition to a standard panel of stains, tissues were immunostained against a variety of rod and cone opsins. Finally, published opsin sequences of platypus and several other vertebrate species were aligned and compared with immunohistochemical results. A complete scleral cartilage similar to that seen in birds, reptiles and amphibians encloses the platypus eye. This feature is present in sharks and rays, and in extant relatives of tetrapods, the lungfishes. The choroid lacks a tapetum. The retina is largely avascular and is rod-dominated, with a minority of red- and blue- cone immunoreactive photoreceptors. Like marsupials and many nonmammalian vertebrates, cones contain clear inner segment droplets. Double cones were present, a feature not found in eutherian mammals or marsupials. Evaluation of opsins indicates that red and blue immunoreactive cone opsins, but not rhodopsin, are present in the most basal of the extant species examined, the Pacific hagfish. Rhodopsin appears in the Australian and South American lungfishes, establishing emergence of this pigment in an extant relative of tetrapods. Unlike eyes of eutherian mammals, the platypus eye has retained morphologic features present in early tetrapods such as amphibians and their evolutionarily basal sister group, the lungfishes. These include scleral cartilage, double cones and cone droplets. We examined differently aged postnatal (P) rats for the distribution and uptake of iron in the eye with the main emphasis on iron uptake in the retina. The concentration of iron in the eye was 48 μg/g in rats aged one postnatal day (P1). Then concentration fell to approximately 12 μg/g at P30 and rose to 35 μg/g at P70. Perls' stain which labels both ferrous and ferric iron was found to exhibit a weak labeling in the retina at P1 contrasted by a robust labeling of macrophages found in the choroid of the retina. In older aged rats, the labeling of cells of the retina was much more intense and confined to cells widespread in the layers of the retina. In both P16 and adult rats injected intravenously with [(59)Fe-(125)I]transferrin, the uptake of (59)Fe, estimated as the volume of distribution, was significantly higher than that of [(125)I]transferrin, and uptake of both compounds was higher than that of simultaneously injected [(131)I]albumin. In the P16 rat, the uptake of (59)Fe expressed as the volume of distribution, V(D), rose linearly reaching approximately 2500 nl at 60 min. In the adult rat, the uptake of (59)Fe was of the same magnitude. Comparing P1 and P16 rats, the uptake of (59)Fe, [(125)I]transferrin and [(131)I]albumin was higher at P1 in both eyeball and retina. Emulsion autoradiography of retinas from P16 and adult rats injected with [(55)Fe]transferrin into the vitreous body showed uptake mainly in photoreceptor cells and retinal ganglion cells. Adult rats injected into the vitreous body with [(59)Fe]transferrin showed anterograde axonal transport from the retina into the optic nerve, optic tract, and superior colliculus. Immunoprecipitates of homogenates of the optic nerve revealed that (59)Fe was precipitable with an antibody raised against ferritin, indicative of detachment of iron from transferrin within the axons of the retinal ganglion cells. The data demonstrate an age-dependent but continuous iron uptake by the retina, and are indicative of anterograde axonal transport of transferrin by retinal ganglion cells., (Copyright © 2010 Elsevier Inc. Shallow-water zooplanktivorous fish rely on their vision for foraging. In shallow water, feeding efficiency decreases in dim light and thus the fish cease foraging at crepuscular hours. Creatures living in the lower parts of their depth ranges are expected to be exposed to limited light levels for longer hours. However, observations of the zooplanktivore Dascyllus marginatus showed little change in foraging duration down to 40m deep. We asked whether the visual system's functionality changes with depth along the depth range of this damselfish; we examined eye and retina anatomy for changes in visual acuity and light sensitivity and used the optomotor response to test for spatial and temporal light summation. We found only minor changes in the anatomy of the eye that are not expected to affect visual sensitivity or acuity. However, behavioural experiments showed that the deeper water fish's test performance exceeded those of fish in shallow water under lower light levels. We found that deeper water fish responded to the optomotor test at lower light levels and also had more discriminating visual acuity in low light, which can increase their potential reactive distance. The plastic adaptive ability of the visual system to low light levels may explain the fish's ability to inhabit deeper reef habitats and thus expand their depth range limits., (Copyright © 2010 Elsevier Inc. Retinal topographies of some cell types and distribution of the tapetum lucidum in the sheep's eye were investigated in this study. The tapetum was observed macroscopically in the fundus. The topographical distributions of retinal ganglion cells (RGCs), cones, and rods were simultaneously analyzed in retinal whole mounts stained with cresyl violet. Short-wavelength-sensitive (S) cones were immunocytochemically identified in retinal whole mounts. The tapetum was located dorsal to the optic disc, with the nasal part elongated horizontally and the temporal part expanded dorsally. RGCs were distributed densely in the area centralis, horizontal visual streak, and anakatabatic area. The highest density in the area centralis was approximately 18,000 RGCs/mm(2). Cones showed high density in the horizontal area crossing the optic disc and dorsotemporal area, whereas rods showed high density in the horizontal area, which was greater in height than the horizontal area of high cone density. S cones showed high density in the dorsotemporal retina. The rod/cone ratios were high horizontally in the dorsal retina to the optic disc, with a mean value of 11:1. The cone/RGC and rod/RGC ratios were lower in the horizontal and dorsotemporal retina, and the rod/cone/RGC ratio was lowest in the area centralis (9:1:1). The retinal topographies and distribution of the tapetum were specialized in the horizontal and dorsotemporal fundus. This suggests that sheep have better visual acuity in horizontal and anteroinferior visual fields and that this specialization is related to the visual ecology of sheep., (Copyright 2010 Wiley-Liss, Inc.)

During early postnatal development, dendrites of retinal ganglion cells (RGCs) extend and branch in the inner plexiform layer to establish the adult level of stratification, pattern of branching, and coverage. Many studies have described the branching patterns, transient features, and regulatory factors of stratification of the RGCs. The rate of RGC dendritic field (DF) expansion relative to the growing retina has not been systematically investigated. In this study, we used two methods to examine the relative expansion of RGC DFs. First, we measured the size of RGC DFs and the diameters of the eyeballs at several postnatal stages. We compared the measurements with the RGC DF sizes calculated from difference of the eyeball sizes based on a linear expansion assumption. Second, we used the number of cholinergic amacrine cells (SACs) circumscribed by the DFs of RGCs at corresponding time points as an internal ruler to assess the size of DFs. We found most RGCs exhibit a phase of faster expansion relative to the retina between postnatal day 8 (P8) and P13, followed by a phase of retraction between P13 and adulthood. The morphological alpha cells showed the faster growing phase but not the retraction phase, whereas the morphological ON-OFF direction selective ganglion cells expanded in the same pace as the growing retina. These findings indicate different RGCs show different modes of growth, whereas most subtypes exhibit a fast expansion followed by a retraction phase to reach the adult size.

Purpose: To determine the dependency of straylight on ocular biometry., Methods: This prospective study included 518 eyes of 277 volunteers of diverse ethnic backgrounds with healthy eyes of various iris colors. The eyes had retinal straylight tested using a commercial psychophysical device. Ocular axial length and refraction were also measured with an ocular biometer and an autorefractometer, respectively., Results: The measured retinal straylight was validated by comparing data with the age model described in the literature as log(s) = P(1) + log[1 + (age/65)(4)], where P(1) is the logarithm of the average straylight for the eyes of a newborn. The data agreed well with this model, although P(1) was slightly higher (0.931 vs. 0.87). When this model was subtracted from the measured straylight values, a quadratic increase was found in the function of axial length, L: log(s) = 0.931 + log[1 + (age/65)(4)] + (0.01089L(2) - 0.4820L + 5.330). A similar model was defined for the spherical equivalent refraction SE. This corresponds to an increasing amount of straylight for increasing degrees of myopia. No correlation was found with keratometry and corneal astigmatism or with iris color., Conclusions: Retinal straylight increases not only with age, but also with axial length. Further study is needed to identify the cause of this dependency.

Purpose: To demonstrate the three-dimensional macular thickness distribution in normal subjects by spectral domain optical coherence tomography (SD-OCT) and evaluate its association with sex, age, and axial length., Methods: Mean regional retinal thickness measurements on the Early Treatment Diabetic Retinopathy Study (ETDRS) layout were obtained by three-dimensional raster scanning (6 x 6 mm) using SD-OCT in 248 normal eyes of 248 Japanese subjects., Results: Mean foveal thickness was 222 +/- 19 microm; it was significantly greater in men (226 +/- 19 microm) than in women (218 +/- 18 microm; P = 0.002) and did not correlate with age in either sex. Mean sectoral retinal thickness was also significantly greater in the men than in the women in all the quadrants of the inner ring (1-3 mm; P < 0.001 and P = 0.001-0.007) and in the temporal quadrant of the outer ring (3-6 mm; P < 0.001). The retinal thicknesses of each of the ETDRS sectors did not correlate significantly with axial length after adjustment for age in either sex. Retinal thickness in six of the eight sectors in the inner and outer rings showed a negative correlation with age after adjustment for axial length in the men (P < 0.001 and P = 0.001-0.018), whereas no correlation with age was observed in the women., Conclusions: SD-OCT demonstrated the three-dimensional macular thickness distribution in normal eyes. Macular thickness varied significantly with sex and age. These variables should be considered while evaluating macular thickness.

Birds that forage on the ground have been studied extensively in relation to behavioral trade-offs between foraging and scanning for predators; however, we know little about the topography of their retinas, which can influence how they gather visual information. We characterized the density of retinal ganglion cells across the retina and estimated visual acuity of four Passeriformes (European starling Sturnus vulgaris, brown-headed cowbird Molothrus ater, house sparrow Passer domesticus, house finch Carpodacus mexicanus) and one Columbiforme (mourning dove Zenaida macroura) that forage on the ground. We used cresyl violet to stain retinal ganglion cells and estimated visual acuity based on cell density and eye size. All species contained a single area centralis, where cell densities were >20,000 cells/mm(2). The proportion of the retina that fell in each of five cell density ranges varied between species. European starlings and house finches had the largest area of high cell density, mourning doves had the smallest. The largest proportion of the retina (>35%) of brown-headed cowbird and house sparrow was in the second-lowest cell density range. Considering the 25th percentile of highest cell densities, house finches and European starlings showed the highest cell densities and mourning doves the lowest. Estimated visual acuity increased from house finch, house sparrow, brown-headed cowbird, European starling to mourning dove, and was associated with both retinal area and cell density. Our findings suggest that these ground foragers do not have highly specialized retinas in relation to other types of foragers (e.g. tree foragers), probably because foraging on seeds and insects from the ground is not as visually demanding; however, the studied species showed variability in retinal topography that may be related to foraging techniques, eye size constraints, and size of the area centralis., (Copyright 2010 S. Several visual traits have previously been assessed in elasmobranchs; however, few studies have examined and compared multiple visual attributes within a particular genus. The primary advantage of studying closely related species is that any differences between them are more likely to reflect functional ecological adaptations rather than the effects of phylogenetic separation. In this study, the visual capabilities of 4 wobbegong shark species, which vary in life-history and/or habitat, were examined: the western wobbegong (Orectolobus hutchinsi), the spotted wobbegong (O. maculatus), the ornate wobbegong (O. ornatus) and the dwarf spotted wobbegong (O. parvimaculatus). The retinae of all 4 wobbegong species are duplex; rod and cone photoreceptors can be distinguished easily on the basis of morphology. Some variation in relative eye size exists, with O. parvimaculatus possessing the largest eyes. The topographic distribution of cells within the ganglion cell layer of O. hutchinsi reveals a weakly elongated central visual streak of increased cell density, mediating a higher spatial resolving power of 2.06 cycles deg⁻¹ in the frontal visual field. Retinal topography of O. maculatus and O. parvimaculatus is similar, with both possessing a dorsal horizontal streak facilitating an increased spatial resolving power of 3.51 cycles deg⁻¹ and 3.91 cycles deg⁻¹, respectively, in the lower visual field. O. parvimaculatus also possesses an area of increased cell density in the naso-ventral region of the retina, mediating acute vision in the upper caudal region of the visual field. While all 4 species have visual systems optimised for increased visual sensitivity, O. maculatus and O. parvimaculatus appear to be particularly well suited to activity under low light conditions., (Copyright © 2010 S. The eyes of three adult male African elephants were examined, the retinas were whole-mounted, stained and analyzed to determine visual acuity. A range of small to large ganglion cell types were observed across the retinas. We observed three regions of high ganglion cell density, one in the upper temporal quadrant, a visual or horizontal streak and a smaller region at the nasal end of the horizontal streak. The peak density of ganglion cells observed was 5,280/mm(2), and our calculations indicate that the elephant has a maximal visual acuity of between 13.16 and 14.37 cycles/degree. We observed a heterogeneous structure of a tapetum lucidum, the cells of which were found to be most strongly aggregated behind the temporal and nasal densities of retinal ganglion cells. The strength of the tapetum lucidum was weaker posterior to the density of ganglion cells forming the horizontal streak. The morphology of the elephant eye appears to be such that it reflects: (1) the importance of trunk-eye co-ordination for feeding; (2) the importance of 24-hour vigilance for either predators or conspecifics, and (3) the arrhythmic nature of the daily activity of this animal, being useful both diurnally and nocturnally., (Copyright 2010 S. Melanopsin, an opsin protein expressed in mammalian retinal ganglion cells (RGCs), makes them responsive to light. Such photosensitive RGCs form the retinohypothalamic tract (RHT) that provides signals to the suprachiasmatic nucleus (SCN), the master regulator of circadian rhythms. The SCN is adjusted daily to the environmental day/night cycle by signal inputs incoming from the RHT. In the present work we have studied, using immunohistochemistry techniques, the types and number of cells which expressed melanopsin during the postnatal development of pigmented C3H/He mice maintained in a standard daily cycle (12-h light/12-h dark). Our results clearly show for the first time that the retina maintains a rather constant number of melanopsin-expressing RGCs from the first postnatal day and, thus, demonstrate that no loss of these photosensitive cells occurs during postnatal development. This supports the general idea that the non-image-forming system, in which these cells are involved, is functional at the very early postnatal stage.

Vertebrate eyes are of the simple or camera type with a single optical system that creates an image on the retina in the back of the eye. There, the visual information is encoded as nervous signals by photoreceptors, processed by retinal neurons, and then sent to the brain via the optic nerve. Surprisingly at first sight, the retinal neurons are located between the lens and the light-sensitive parts of the photoreceptors. The tissue scatters some light, which leads to loss of light and image blur. The inverted retina has, therefore, long been regarded as inferior. Here, we provide evidence that the inverted retina actually is a superior space-saving solution, especially in small eyes. The inverted retina has most likely facilitated the evolution of image-forming eyes in vertebrates, and it still benefits especially small and highly visual species.

Pluripotent cells such as embryonic stem (ES) and induced pluripotent stem (iPS) cells are the starting point from which to generate organ specific cell types. For example, converting pluripotent cells to retinal cells could provide an opportunity to treat retinal injuries and degenerations. In this study, we used an in vivo strategy to determine if functional retinas could be generated from a defined population of pluripotent Xenopus laevis cells. Animal pole cells isolated from blastula stage embryos are pluripotent. Untreated, these cells formed only epidermis, when transplanted to either the flank or eye field. In contrast, misexpression of seven transcription factors induced the formation of retinal cell types. Induced retinal cells were committed to a retinal lineage as they formed eyes when transplanted to the flanks of developing embryos. When the endogenous eye field was replaced with induced retinal cells, they formed eyes that were molecularly, anatomically, and electrophysiologically similar to normal eyes. Importantly, induced eyes could guide a vision-based behavior. These results suggest the fate of pluripotent cells may be purposely altered to generate multipotent retinal progenitor cells, which differentiate into functional retinal cell classes and form a neural circuitry sufficient for vision., Competing Interests: The authors have declared that no competing interests exist.

Conserved developmental programs, such as the order of neurogenesis in the mammalian eye, suggest the presence of useful features for evolutionary stability and variability. The owl monkey, Aotus azarae, has developed a fully nocturnal retina in recent evolution. Description and quantification of cell cycle kinetics show that embryonic cytogenesis is extended in Aotus compared with the diurnal New World monkey Cebus apella. Combined with the conserved mammalian pattern of retinal cell specification, this single change in retinal progenitor cell proliferation can produce the multiple alterations of the nocturnal retina, including coordinated reduction in cone and ganglion cell numbers, increase in rod and rod bipolar numbers, and potentially loss of the fovea.

The visual system in humans is considered the gateway to the world and plays a principal role in the plethora of sensory, perceptual and cognitive processes. It is therefore not surprising that quality of vision is tied to quality of life . Despite widespread clinical and basic research surrounding the causes of visual disorders, many forms of visual impairments, such as retinitis pigmentosa and macular degeneration, lack effective treatments. Non-human primates have the closest general features of eye development to that of humans. Not only do they have a similar vascular anatomy, but amongst other mammals, primates have the unique characteristic of having a region in the temporal retina specialized for high visual acuity, the fovea(1). Here we describe a general technique for dissecting the primate retina to provide tissue for retinal histology, immunohistochemistry, laser capture microdissection, as well as light and electron microscopy. With the extended use of the non-human primate as a translational model, our hope is that improved understanding of the retina will provide insights into effective approaches towards attenuating or reversing the negative impact of visual disorders on the quality of life of affected individuals.

In mammals, a limited set of homeobox-containing transcription factors are expressed in the presumptive eye field and required to initiate eye development. How these factors interact together at the genetic and molecular level to coordinate this developmental process is poorly understood. We found that the Lhx2 and Pax6 transcription factors operate in a concerted manner during retinal development to promote transcriptional activation of the Six6 homeobox-gene in primitive and mature retinal progenitors. Lhx2 demarcates the presumptive retina field at the neural plate stage and Lhx2 inactivation delays initiation of Rx, Six3 and Pax6 expression in this domain. The later expressed Six6 is properly activated in the pituitary/hypothalamic axis of Lhx2(-/-) embryos, but expression fails to be initiated in the optic vesicle. Lhx2 and Pax6 associate with the chromatin at several regions of Six6 in vivo and cooperate for trans-activation of Six6 regulatory elements in vitro. In retinal progenitor/stem cells, both Lhx2 and Pax6 are genetically required for proper Six6 expression and forced co-expression of Lhx2 and Pax6 can synergistically trans-activate the Six6 locus. Our work reveals how two master regulators of eye development coordinate their action to sequentially promote tissue-specific transcriptional initiation and full activation of a retinal determinant gene.

The development of the eye of the oviparous brown banded bamboo shark, Chiloscyllium punctatum, was monitored from egg deposition through adulthood. The order and timing of retinal cell differentiation were assessed by light and transmission electron microscopy. As in other vertebrates, the ganglion cells are the first to differentiate, in this case by 81 days post-egg deposition (dpd). The order then deviates from what is typically quoted for vertebrates, with the Müller and amacrine cells differentiating morphologically around the same time, followed by the bipolar cells (101 dpd) and finally the horizontal cells and photoreceptors (124 dpd). The neural retina is fully differentiated and synaptic connections are formed approximately 1 month prior to hatching, which occurs at about 158 dpd. The mature retina is duplex, with a peak rod to cone ratio of approximately 12:1. The eye and lens of C. punctatum continue to grow throughout life and become less aspherical with growth; the equatorial (nasotemporal) lens diameter is 12% larger than the axial (anterior-posterior) lens diameter in embryos and 8% larger in adults. Access to developmental stages and the protracted gestational period of C. punctatum make it a highly valuable model for developmental studies of the visual system. This study also provides an evolutionary perspective on retinal neurogenesis in an elasmobranch.

Wave aberrations in the human eye are usually known with respect to the ideal spherical wavefront in the exit pupil. Using Kirchhoff's diffraction theory, we have derived a diffraction integral to compute the optical field on the retina from the wave aberration data. We have proposed a numerical algorithm based on the Stamnes-Spjelkavik-Pedersen (SSP) method to solve that integral. We have shown which approximations are admissible to reduce the complexity of the diffraction integral. In addition, we have compared our results with those of the conventional procedure used to compute intensities on the retina. We have found significant differences between our results and the conventional ones.

We use the whole-cell patch clamp technique to study the synaptic circuitry that underlies visual information processing in the retina. In this video, we will guide you through the process of performing whole-cell recordings of light evoked currents of individual cells in the retinal slice preparation. We use the aquatic tiger salamander as an animal model. We begin by describing the dissection of the eye and show how slices are mounted for electrophysiological recordings. Once the slice is placed in the recording chamber, we demonstrate how to perform whole-cell voltage clamp recordings. We then project visual stimuli onto the photoreceptors in the slice to elicit light-evoked current responses. During the recording we perfuse the slice with pharmacological agents, whereby an 8-channel perfusion system allows us to quickly switch between different agents. The retinal slice preparation is widely used for patch clamp recordings in the retina, in particular to study amacrine or bipolar cells, which are not accessible in a whole-mount preparation.

Objectives: To determine the vitreous volume, and retinal and lens wet weights in male and female Göttingen minipigs of different age groups., Procedures: Vitreous, lens and retina were isolated from fresh minipig eyes. Vitreous volume, lens and retina weight were measured and allometric scaling was utilized to predict these parameters., Results: There were no gender differences in body weights or retina and lens weights in the three age groups examined in this study. For vitreous volumes, females had a significantly larger volume (approximately 10%-12%) than males in the 4-6 and 6-8 month, but not in the 8-11-month group. The mean body weight increased from 12.2 +/- 2.6 kg (4-6 months) to 19.4 +/- 4.0 kg (8-11 months). In the same period, the mean vitreous volume increased from 2.00 +/- 0.28 to 2.67 +/- 0.31 mL, while the mean retinal and lens weights increased from 114 +/- 22 to 126 +/- 17 mg, and from 298 +/- 26 to 392 +/- 15 mg, respectively. Allometric analysis between six species for vitreous volume and four species for lens weight covering a weight range of 260-fold was not able to estimate the vitreous volume, but it did predict the lens weight in 8-11 months minipigs., Conclusions: Overall these measurements add important context to intraocular pharmacology studies, and will help in the improved design and interpretation of such experiments.

Vertebrate eye development has been an excellent model system to investigate basic concepts of developmental biology ranging from mechanisms of tissue induction to the complex patterning and bidimensional orientation of the highly specialized retina. Recent advances have shed light on the interplay between numerous transcriptional networks and growth factors that are involved in the specific stages of retinogenesis, optic nerve formation, and topographic mapping. In this review, we summarize this recent progress on the molecular mechanisms underlying the development of the eye, visual system, and embryonic tumors that arise in the optic system.

Purpose: To investigate the effect of shorter axial length on the laser spot size and laser energy during photodynamic therapy (PDT) in monkeys., Design: Experimental study with four rhesus monkeys., Methods: PDT was performed on the normal retina of monkeys whose ocular axial lengths are shorter (19.55 to 20.25 mm) than that of humans. After the PDT, the eyes were enucleated, and the diameter of the irradiated laser spot was measured with a microcaliper., Results: The area of actual laser spot was only 0.56 to 0.61 times of the planned area, which indicated that the laser energy/area was 1.64 to 1.78 times more intense than planned initially., Conclusion: These results are the in vivo demonstration that the diameter of PDT laser spot is smaller for eyes with shorter axial lengths.

Optical coherence tomography (OCT) poor mapping resolution has been pointed out as the biggest disadvantage of this technique when compared to others, e.g., retinal thickness analyzer. In this work we were able to solve this problem by developing an atlas of macular thickness of the human retina into which OCT scans were thereafter registered. This atlas is used to allow registering OCT scans from the Fast Macular Protocol, thus bringing OCT scans into the atlas coordinates, therefore correcting for misfixations, while simultaneously allowing to perform OCT inter-scan registration. From this initial registration, we were able to compute a thickness map into which Fast RNFL Protocol scans were merged, thus allowing for increased OCT mapping resolution.

Sensory axons are targeted to modality-specific nuclei in the thalamus. Retinal ganglion cell axons project retinotopically to their principal thalamic target, the dorsal lateral geniculate nucleus (LGd), in a pattern likely dictated by the expression of molecular gradients in the LGd. Deafferenting the auditory thalamus induces retinal axons to innervate the medial geniculate nucleus (MGN). These retino-MGN projections also show retinotopic organization. Here we show that ephrin-A2 and -A5, which are expressed in similar gradients in the MGN and LGd, can be used to pattern novel retinal projections in the MGN. As in the LGd, retinal axons from each eye terminate in discrete eye-specific zones in the MGN of rewired wild-type and ephrin-A2/A5 knockout mice. However, ipsilateral eye axons, which arise from retinal regions of high EphA5 receptor expression and represent central visual field, terminate in markedly different ways in the two mice. In rewired wild-type mice, ipsilateral axons specifically avoid areas of high ephrin expression in the MGN. In rewired ephrin knockout mice, ipsilateral projections shift in location and spread more broadly, leading to an expanded representation of the ipsilateral eye in the MGN. Similarly, ipsilateral projections to the LGd in ephrin knockout mice are shifted and are more widespread than in the LGd of wild-type mice. In the MGN, as in the LGd, terminations from the two eyes show little overlap even in the knockout mice, suggesting that local interocular segregation occurs regardless of other patterning determinants. Our data demonstrate that graded topographic labels, such as the ephrins, can serve to shape multiple related aspects of afferent patterning, including topographic mapping and the extent and spread of eye-specific projections. Background: Optical coherence tomography (OCT) acquires cross-sectional retinal images with high resolution using low-coherence interferometry. Few studies have studied the effect of demographic data and ocular parameters that may affect central retinal thickness. In this study, these factors were used as parameters to analyse if any significant relationship exists with central retinal thickness., Methods: Volunteers with a best-corrected visual acuity of 6/12 or better and no evidence of ocular abnormalities or interventions were recruited from October 2001 to March 2003. Body mass index (BMI), autorefraction, and keratometry recordings were measured, followed by applanation tonometry and A-scan ultrasonography. The central retinal thickness of the right eye was analysed using a scan length of 3 cm. Another 25 eyes were selected for interobserver reproducibility., Results: In all, 117 normal subjects (60 male and 57 female subjects) were recruited. The mean thickness of the central retina with a diameter of 1 mm was 203+/-23 microm for male and 189+/-20 microm for female subjects. Age, intraocular pressure, and keratometric readings were not significantly correlated with central retinal thickness. Using multiple regression, gender, BMI, axial length, and signal-to-noise ratio (P<0.05) were significantly associated with the central retinal thickness. The intraclass correlation coefficient was 0.98 for interobserver reproducibility., Conclusion: OCT has a high interobserver reproducibility. The male gender, larger BMI, and longer axial length are associated with a significantly thicker central retina and these parameters should be considered for assessing retinal thickening and baseline comparisons in future studies.

The mab-21 gene was first identified because of its requirement for ray identity specification in Caenorhabditis elegans. It is now known to constitute a family of genes that are highly conserved from vertebrates to invertebrates, and two homologues Mab21l1 and Mab21l2 have been identified in many species. Here we describe the generation of Mab21l2-deficient mice, which have defects in eye and body wall formation. The mutant mouse eye has a rudimentary retina, as a result of insufficient invagination of the optic vesicle due to deficient proliferation, causing the absence of lens. The defects in optic vesicle development correlate with reduced expression of Chx10, which is also required for retina development; Rx, Lhx2, and Pax6 expression is not significantly affected. We conclude that Mab21l2 expression is essential for optic vesicle growth and formation of the optic cup, its absence causing reduced expression of Chx10. Mutant mice also display abnormal extrusion of abdominal organs, defects in ventral body wall formation, resulting in death in utero at mid-gestational stage. Our results reveal that Mab21l2 plays crucial roles in retina and in ventral body wall formation.

Various parameters of the eye dimension and structure have been compared in 15 species of cardinal fish (Apogonidae), including both nocturnal and diurnal forms, mostly inhabiting rocky habitats in tropical and subtropical regions. In general, in the nocturnal forms the eye and retina are larger than in the diurnal fish of similar dimensions. In the nocturnal species, eye diameter to body length is ca. 12-13%, whereas in the diurnal species it is less than 10%. Retina size in adult fish of the various species varies from 20 mm(2) to 183 mm(2). Cytological examination of the studied retinas revealed that they are composes, additional to rods (20-40 microm), of both bulbous and slender double cones, as well as single cones. These cones form a mosaic comprising four double with one single in the center, a pattern that is less prominent at the periphery of the retina and more so in the fundus. The rod ellipsoids reveal normal mitochondria, whereas the cones bear ellipsoids featuring opaque and unusual, ellipsosome-like mitochondria. The number of rods in a retinas varies from 15 to 128 million, and the number of cones from 460,000 to 5,673,000. As revealed in cardinal fish of similar dimensions, the number of visual cells found in the retina is much higher in the nocturnal than in the diurnal species. We have determined the presence of spectral cone types, and the population densities of cones and rods, in subterranean mole-rats of the rodent family Bathyergidae, for which light and vision seems of little importance. Most mammals have two spectral cone types, a majority of middle- to long-wave-sensitive (L-) cones, and a minority of short-wave-sensitive (S-)cones. We were interested to see whether the subterranean bathyergids show the same pattern. In three species, Ansell's mole-rat Cryptomys anselli, the giant mole-rat Cryptomys mechowi and the naked mole-rat Heterocephalus glaber, spectral cone types and rods were assessed immunocytochemically with opsin-specific antibodies. All three species had rod-dominated retinae but possessed significant cone populations. A quantitative assessment in C. anselli and C. mechowi revealed surprisingly low photoreceptor densities of 100 000-150 000/mm(2), and high cone proportions, approximately 10% (8000-15 000/mm(2)). In all three species, the vast majority of the cones were strongly S-opsin-immunoreactive; L-opsin immunoreactivity was much fainter. In C. anselli, approximately 20% of the cones showed exclusive S-opsin label, approximately 10% exclusive L-opsin label and approximately 70% strong S-opsin and faint L-opsin double label (potential dual-pigment cones). This is the first observation in any mammal of an S-opsin dominance and low levels of L-opsin across the entire retina. It contrasts starkly with the situation in the muroid blind mole-rat Spalax ehrenbergi, which has been reported to possess L-opsin but no S-opsin. Evidently, within rodents an adaptation to subterranean life is compatible with very different spectral cone properties.

The vertebrate nervous system contains an immense diversity of distinct cellular components that are organized into precise spatial patterns. The importance of accurate neuronal architecture is particularly obvious in the retina, where it is necessary for the formation of visual images. The retina is structured in a distinct layered pattern that is remarkably conserved in evolution, including phyla as diverse as primates and teleost fish. Genetic analysis in zebrafish reveals mechanisms that are essential for the formation of this architecture.

Oilbirds (Steatornis caripensis) breed in the total darkness of caves and forage at night on fruits. Common pauraques (Nyctidromus albicollis) are crepuscular and nocturnal foragers on flying insects. We examined if their retinal structure and function can be correlated with their types and periods of activity. Electroretinograms (ERGs) were obtained from anesthetized birds in photopic and scotopic conditions to a wide range of light intensities, following which the retinas were processed for histological analysis. Retinal sensitivity is higher in oilbirds than in common pauraques. Under scotopic conditions with maximum flash luminance, the average (+/- 95% CI) b-wave amplitude of oilbirds is double that of common pauraques (500.4 +/- 49.8 and 245.4 +/- 40.9 microV, respectively) but, under photopic conditions, the results are the reverse (common pauraque: 69.4 +/- 18.1; oilbird: 23.0 +/- 4.4 microV). On the other hand, the retina of both species is highly rod-dominated, but rods are highly more numerous in oilbirds than in common pauraques (rods:cones ratio: 123:1 and 5:1, respectively). In oilbirds, rods are largely thinner and their outer segments are 1.0 microm in diameter and 18.6 microm in length. They are distributed over various levels in the photoreceptor layers, an arrangement known for deep-sea fishes, but so far unknown for birds. In common pauraques, rods are patchily distributed and their outer segments are 4.0 microm in diameter and 53 microm in length. The oilbirds rod thinness allows more rods per area unit, and thus to catch more photons per area unit under darkness, while the low cone number suggests that the species has poor daytime vision, which concurs with the species cavernicolous daytime habits. The lower rod number of common pauraques, compared to oilbirds, appears counterbalanced by their patchiness and longer and thicker outer segments to provide high retinal sensitivity. In addition, common pauraques also have a tapetum. These features, combined with a higher proportion of cones, show that common pauraques are well equipped for crepuscular and nocturnal foraging on flying insects in an open environment., (Copyright 2004 S.
