The aim of this study was to obtain baseline data for heavy metal(loids) concentrations of rice paddy fields to evaluate the impacts of soil metal(loids) concentrations on quality of rice, and to identify sources of metal(loids) pollution of paddy fields in Malawi. In total, 66 soil samples were collected from 22 different smallholder rice farmers' paddy fields (sites) in Malawi. Concentrations of metal(loids) (arsenic (As), cadmium (Cd), cobalt (Co), chromium (Cr), manganese (Mn), lead (Pb), uranium (U), and gallium (Ga)) were measured using inductively coupled plasma mass spectrometry (ICP-MS) after acid block digestion with a mixture of concentrated nitric acid (70% HNO3) and hydrogen peroxide (30% H₂O₂). Measured soil metal(loids) concentrations were compared with Soil UK CLEA soil metal(loids) guidelines, maximum allowable limits (MAL) recommended by the World Health Organization (WHO), the Chinese Environmental Quality Standards (CEQS) for soil metal(loids) concentrations in agricultural soils, and the normally reported soil metal(loids) concentrations in agricultural fields worldwide. Results indicated that mean soil As (2.2 mg As kg−1), Cd (0.044 mg Cd kg−1), Pb (11 mg Pb kg−1), Co (14 ± 6 mg Co kg−1), Mn (601 mg Mn kg−1), U (2.02 mg U kg−1), and Ga (24 mg Ga kg−1) concentrations were at least three times lower than the respective guidelines and MAL recommended by WHO, UK CLEA, and CEQS (20 mg As kg−1, 0.3 mg Cd kg−1, 32 mg Pb kg−1, 50 mg Co kg−1, and 2000 mg Mn kg−1). The values obtained in this study were also within the normally reported metal(loid) concentrations for unpolluted agricultural soils worldwide. However, the mean Cr concentration (78.0 mg Cr kg−1) obtained in this study was higher than that reported for agricultural soils in China (27 ± 5 mg Cr kg−1; range: 22–39 mg Cr kg−1) but was within the normally reported Cr concentrations (1–100 mg Cr kg−1) for unpolluted soils. These findings suggest that metal(loids) concentrations of Malawian rice paddies pose no threat to production to elevated metal(loids) accumulation in rice, and that rice paddies should be safeguarded from contamination. However, further research is required to investigate the impacts of factors such as source of irrigation of water, water management regimes, soil cation exchange capacity, organic manure/composts amendments, and the application of inorganic fertilizers on uptake, transfer, and translocation of soil metal(loids) to various parts of rice plants, even when the soil metal(loids) concentrations are low. Rice (Oryza sativa) is the staple food for over half the world's population yet may represent a significant dietary source of morganic arsenic (As), a nonthreshold, class 1 human carcinogen. Rice grain As is dominated by the inorganic species, and the organic species dimethylarsinic acid (DMA). To investigate how As species are unloaded into grain rice, panicles were excised during grain filling and hydroponically pulsed with arsenite, arsenate, glutathione-complexed As, or DMA. Total As concentrations in flag leaf, grain, and husk, were quantified by inductively coupled plasma mass spectroscopy and As speciation in the fresh grain was determined by x-ray absorption near-edge spectroscopy. The roles of phloem and xylem transport were investigated by applying a ± stem-girdling treatment to a second set of panicles, limiting phioem transport to the grain in panicles pulsed with arsenite or DMA. The results demonstrate that DMA is translocated to the rice grain with over an order magnitude greater efficiency than inorganic species and is more mobile than arsenite in both the phloem and the xylem. Phloem transport accounted for 90% of arsenite, and 55% of DMA, transport to the grain. Synchrotron x-ray fluorescence mapping and fluorescence microtomography revealed marked differences in the pattern of As unloading into the grain between DMA and arsenite-challenged grain. Arsenite was retained in the ovular vascular trace and DMA dispersed throughout the external grain parts and into the endosperm. Inorganic arsenic is a chronic exposure carcinogen. Analysis of UK baby rice revealed a median inorganic arsenic content (n =17) of 0.11mg/kg. By plotting inorganic arsenic against total arsenic, it was found that inorganic concentrations increased linearly up to 0.25mg/kg total arsenic, then plateaued at 0.16mg/kg at higher total arsenic concentrations. Inorganic arsenic intake by babies (4–12 months) was considered with respect to current dietary ingestion regulations. It was found that 35% of the baby rice samples analysed would be illegal for sale in China which has regulatory limit of 0.15mg/kg inorganic arsenic. EU and US food regulations on arsenic are non-existent. When baby inorganic arsenic intake from rice was considered, median consumption (expressed as μg/kg/d) was higher than drinking water maximum exposures predicted for adults in these regions when water intake was expressed on a bodyweight basis. A commercial arsenic field kit designed to measure inorganic arsenic (iAs) in water was modified into a field deployable method (FDM) to measure iAs in rice. While the method has been validated to give precise and accurate results in the laboratory, its on-site field performance has not been evaluated. This study was designed to test the method on-site in Malawi in order to evaluate its accuracy and precision in determination of iAs on-site by comparing with a validated reference method and giving original data on inorganic arsenic in Malawian rice and rice-based products. The method was validated by using the established laboratory-based HPLC-ICPMS. Statistical tests indicated there were no significant differences between on-site and laboratory iAs measurements determined using the FDM (p = 0.263, ά = 0.05) and between on-site measurements and measurements determined using HPLC-ICP-MS (p = 0.299, ά = 0.05). It has previously been shown that across different arsenic (As) soil environments, a decrease in grain selenium (Se), zinc (Zn), and nickel (Ni) concentrations is associated with an increase in grain As. In this study we aim to determine if there is a genetic element for this observation or if it is driven by the soil As environment. To determine the genetic and environmental effect on grain element composition, multielement analysis using ICP-MS was performed on rice grain from a range of rice cultivars grown in 4 different field sites (2 in Bangladesh and 2 in West Bengal). At all four sites a negative correlation was observed between grain As and grain Ni, while at three of the four sites a negative correlation was observed between grain As and grain Se and grain copper (Cu). For manganese, Ni, Cu, and Se there was also a significant genetic interaction with grain arsenic indicating some cultivars are more strongly affected by arsenic than others. [ABSTRACT FROM AUTHOR]

The concentration of arsenic (As) in rice grains has been identified as a risk to human health. The high proportion of inorganic species of As (As,) is of particular concern as it is a nonthreshold, class 1 human carcinogen. To be able to breed rice with low grain As, an understanding of genetic variation and the effect of different environments on genetic variation is needed. In this study, 13 cultivars grown at two field sites each in Bangladesh, India, and China are evaluated for grain As. There was a significant site, genotype, and site by genotype interaction for total grain As. Correlations were observed only between sites in Bangladesh and India, not between countries or within the Chinese sites. For seven cultivars the As was speciated which revealed significant effects of site, genotype, and site by genotype interaction for percentage Asi. Breeding low grain An cultivars that will have consistently low grain As and low Asi, over multiple environments using traditional breeding approaches maybe difficult. although CT9993-5-10-1-M, Lemont, Azucena, and Te-qing in general had low grain As across the field sites. An extensive data set of total arsenic analysis for 901 polished (white) grain samples, originating from 10 countries from 4 continents, was compiled. The samples represented the baseline (i.e., not specifically collected from arsenic contaminated areas), and all were for market sale in major conurbations. Median total arsenic contents of rice varied 7-fold, with Egypt (0.04 mg/kg) and India (0.07 mg/kg) having the lowest arsenic content while the U.S. (0.25 mg/kg) and France (0.28 mg/kg) had the highest content Global distribution of total arsenic in rice was modeled by weighting each country's arsenic distribution by that country's contribution to global production. A subset of 63 samples from Bangladesh, China, India, Italy, and the U.S. was analyzed for arsenic species. The relationship between inorganic arsenic content versus total arsenic content significantly differed among countries, with Bangladesh and India having the steepest slope in linear regression, and the U.S. having the shallowest slope. Rice has been demonstrated to be one of the major contributors to arsenic (As) in human diets in addition to drinking water, but little is known about rice products as an additional source of As exposure. Rice products were analyzed for total As and a subset of samples were measured for arsenic speciation using high performance liquid chromatography interfaced with inductively coupled plasma-mass spectrometry (HPLC–ICP-MS). A wide range of rice products had total and inorganic arsenic levels that typified those found in rice grain including, crisped rice, puffed rice, rice crackers, rice noodles and a range of Japanese rice condiments as well as rice products targeted at the macrobiotic, vegan, lactose intolerant and gluten intolerance food market. Most As in rice products are inorganic As (75.2–90.1%). This study provides a wider appreciation of how inorganic arsenic derived from rice products enters the human diet. [Copyright &y& Elsevier]
