In recent days, there has been a noticeable surge in demand for high-quality rice. However, the influences of salinity on the quality and starch properties of high-quality rice remain unclear. Three high-quality rice cultivars (Nanjing 9108, Nanjing 5055, and Nanjing 46) were studied to investigate the responses of grain quality to salt stress. There were three treatments, including a control zero salt level (0 g·kg−1, CK), and two salt levels of 0.1 g·kg−1 (0.1% salt stress, T1) and 0.2 g·kg−1 (0.2% salt stress, T2). The study involved the assessment of the appearance, milling, cooking, and eating qualities of rice. We also conducted an analysis of pasting properties, an evaluation of starch thermal properties, and an examination of the fine structure of amylopectin. The findings suggest that as the level of salt stress increases, the yield of rice gradually declines, which is primarily due to a significant reduction in the total spikelet number and the ratio of filled grains. Compared with CK treatment, the appearance and milling quality of rice were significantly improved within the T1 treatment. In addition, the protein concentration and amylose concentration were significantly decreased, the gel consistency was significantly increased, and the cooking and eating qualities were improved. In terms of starch properties, the peak viscosity, breakdown value, infrared ratio (1022/995), and short-chain-length amylopectin ratio increased significantly, while the setback value, pasting temperature, gelatinization enthalpy, relative crystallinity, and infrared ratio (1045/1022) decreased significantly. When comparing T2 with CK, the appearance quality and cooking and eating quality had deteriorated, and the milling quality was improved. The changes in the structural and physicochemical properties of starch were opposite to those in the comparison between the T1 treatment and the CK group. Accordingly, we propose that moderate salt stress has the potential to enhance rice quality, even though there may be a slight decrease in yield. Since CEQs are quantitative traits and challenging for measurement, efforts have mainly focused on two major genes, Wx and SSIIa. Chalkiness and floury endosperm significantly affect the eating quality of rice, leading to noticeable changes in CEQ. Due to the easily observable phenotype of floury endosperm, cloning single gene mutations that cause floury endosperm and evaluating changes in CEQs indirectly facilitate the exploration of the minor genes controlling CEQ. In this study, eight mutants with different degrees of floury endosperm, generated through ethylmethane sulfonate (EMS) mutagenesis, were analyzed. These mutants exhibited wide variation in starch morphology and CEQs. Particularly, the z2 mutant showed spherical starch granules significantly increased rapid visco analyzer (RVA) indexes and urea swelling, while the z4 mutant displayed extremely sharp starch granules and significantly decreased RVA indexes and urea swelling compared to the wild type. Additionally, these mutants still maintained correlations with certain RVA profiles, suggesting that the genes PUL, which affect these indexes, may not undergo mutation. Cloning these mutated genes in the future, especially in z2 and z4, will enhance the genetic network of rice eating quality and hold significant importance for molecular marker-assisted breeding to improve rice quality. Abstract Background Endosperm starch provides prime energy for cereal seedling growth. Cereal endosperm with repression of starch branching enzyme (SBE) has been widely studied for its high resistant starch content and health benefit. However, in barley and maize, the repression of SBE changes starch component and amylopectin structure which affects grain germination and seedling establishment. A high resistant starch rice line (TRS) has been developed through inhibiting SBEI/IIb, and its starch has very high resistance to in vitro hydrolysis and digestion. However, it is unclear whether the starch resists in situ degradation in seed and influences seedling growth after grain germination. Results In this study, TRS and its wild-type rice cultivar Te-qing (TQ) were used to investigate the seedling growth, starch property changes, and in situ starch degradation during seedling growth. The slow degradation of starch in TRS seed restrained the seedling growth. The starch components including amylose and amylopectin were simultaneously degraded in TQ seeds during seedling growth, but in TRS seeds, the amylose was degraded faster than amylopectin and the amylopectin long branch-chains with B-type crystallinity had high resistance to in situ degradation. TQ starch was gradually degraded from the proximal to distal region of embryo and from the outer to inner in endosperm. However, TRS endosperm contained polygonal, aggregate, elongated and hollow starch from inner to outer. The polygonal starch similar to TQ starch was completely degraded, and the other starches with long branch-chains of amylopectin and B-type crystallinity were degraded faster at the early stage of seedling growth but had high resistance to in situ degradation during TRS seedling growth. Conclusions The B-type crystallinity and long branch-chains of amylopectin in TRS seed had high resistance to in situ degradation, which inhibited TRS seedling growth.

The difference in grain yield between superior grains (SG) on the upper part and inferior grains (IG) on the lower part of the same panicle was widely reported. To date, variations in rice taste quality between SG and IG and the related starch physicochemical properties remained poorly understood. Here, rice cultivars with different taste quality (NT, normal taste; GT, good taste) were grown to investigate the mechanism underlying taste difference between SG and IG and the correlation between cooked rice taste and starch properties. In this study, the taste value of GT rice was 32.2% higher than that of NT rice across the cultivars. The GT rice comprised a series of typical taste qualities of larger stickiness, smaller hardness, lower apparent amylose content (AAC), and lower protein content (PC). The taste quality differed among rice grains on the same panicle; SG achieved 21.9% and 17.0% higher taste value than IG in GT rice and NT rice, respectively. The higher taste value in SG was owing to the larger stickiness and lower PC. Meanwhile, SG of GT rice achieved the lowest PC (8.2%) and gluten content (5.6%), which might indicate a better health value. Additionally, larger and smoother granules, more fa (DP < 12), lower crystallinity, and larger 1045/1022 cm−1 ratios were found in SG starch compared to IG starch. These led to a weaker swelling power and lower gelatinization enthalpy in SG starch, while gelatinization temperature and retrogression enthalpy were the opposite. Moreover, SG starch exhibited higher storage modulus, loss modulus, slowly digestible starch contents, and resistant starch contents than IG. Our results revealed a great difference in taste quality between SG and IG in rice. The larger and smoother starch granules and shorter chain length could increase the ordered structure of starch, thus improving swelling power, gelatinization properties, and rheological characteristics and facilitating better taste quality of SG over IG. Besides, the lower PC (especially gluten content), higher slowly digestible starch, and higher resistant starch content indicated a more promising health value of SG in the food industry. Physically modified rice starch by ball-mill is important for making good quality bread, noodles, and several processed foods. The production of rice flour was done by ball-mill in an industrial scale. The effects of ball-mill treatment on the physicochemical properties of rice starch were investigated. The gel pasting properties of Joseongheugchal employed to ball-mill treatment showed that pasting time was 0.44 min shorter than that of the control and it also decreased peak viscosity (PKV), hot peak viscosity (HPV), and cold paste viscosity (CPV). In Hongjinju rice, RVA (rapid visco analyzer) pasting trace was characterized with high peak viscosity (3,448 RVU: rapid visco unit), and cold paste viscosity (2,975 RVU), also with high breakdown viscosity (1,597 RVU). The onset, peak, and conclusion gelatinization temperatures of ball-milled Josengheugchal rice starch were 57.4, 64.2, and 76.5°C, respectively, which were significantly higher than those of the control. The parameters of DSC (Differential scanning calorimetry) of ball-milled Hongjinju rice were 57.9, 65.1, and 77.3°C, respectively. The mean particle size of starch in both Josengheugchal and Hongjinju by ball-milling was decreased. The distribution of mean particle size in ball-milled rice starch was smaller in Josengheugchal than in Hongjinju compared to the native starch. As rice grains were ball-milled, the regions of amorphism were becoming larger and larger at the expense of crystalline regions, and diffraction peak patterns were decreased commonly in two colored rice cultivars. ABSTRACT The grain quality of rice has recently attracted a lot of attention around the world, including Africa. Rice germplasm in African gene banks has not been adequately characterized for its cooking and eating qualities which are mainly controlled by starch physicochemical properties. The aim of this study was to characterize two groups of rice germplasm from Africa and the U.S. for starch properties including gelatinization temperature (GT), apparent amylose content (AAC), and paste viscosity (RVA), and to determine molecular marker associations with these traits. Wide diversity was found for all traits studied. Variation in these traits was significantly associated with molecular markers for the alk and waxy genes which encodes soluble SS II ( SSIIa) and granule bound synthase ( GBSS) enzymes, respectively. Based on four previously reported waxy SNP haplotypes, 93.1 and 75.9% of the variation in AAC was explained for the U.S. and African germplasm, respectively. The classification of the genotypes by their SNP haplotypes helped to clarify the relationship between AAC and peak viscosity. The characterization of individual lines using physicochemical properties and functional markers, and the determination of marker-trait associations will facilitate the breeding of rice for grain quality in Africa and elsewhere. High temperature stress during ripening increases the frequency of chalky grains, resulting in a lower market value for rice (Oryza sativa L.). Changes in starch properties and the accumulation pattern of storage proteins are proposed to be related to the occurrence of chalky grains. This study investigated changes in the accumulation of key storage compounds in the grains of Japanese highly palatable rice cultivars, subjected to high temperature stress when grown in a growth chamber and the field. The 13 kDa prolamin content was significantly reduced in a highly heat-sensitive cultivar, Tsukushiroman, whereas the 13 kDa prolamin content was not affected in a heat-tolerant cultivar, Genkitsukushi, even in a high temperature chamber condition (31/26 °C day/night), when compared with the control chamber condition (26/21 °C day/night) for both genotypes. In addition, grains grown in field conditions revealed that severely chalky grains had less 13 kDa prolamin than perfect grains in all five genotypes. Changes in amylose content and the distribution of amylopectin chain lengths did not explain the difference in grain appearance both for chamber and field experiments. The mechanism responsible for the effects of endogenous lipids on starch functionality is not fully understood. In this study, a high-lipid rice mutant (GZ1), a low apparent amylose content (AAC) mutant (GZ93) and parent rice (R7954) were employed to study the effect of the removal of starch lipids (SL) and non-starch lipids (NSL) on the physicochemical properties of starch and resistant starch (RS) and the rice gel texture. GZ1 had a lipid content of 2.52%, of which 61.1% was NSL. Removal of NSL only had a slight impact on the AAC, RS content, gelatinization temperature, swelling power and paste viscosity, but further removal of SL significantly increased the AAC and RS contents. For R7954 and GZ93, removal of SL decreased T p , ΔH, PKV, CPV, and BDV but significantly increased the swelling power (SP). In GZI, the removal of SL resulted in significant increases in ΔH, PKV, HPV, CPV, SBV and the hardness of the gel texture. These results suggested that SLs had a much greater influence on the starch physicochemical properties and gel texture than NSLs. The results may be helpful in designing breeding programs for new rice varieties with better functionality.
