The objective of this paper was to develop a mechanistic-based framework to explicitly incorporate the factors controlling the bioavailability, toxicodynamics and mode of action to enhance predictive ability of arsenic (As) toxicity to protect the health of farmed tilapia Oreochromis mossambicus. We linked the biotic ligand model and damage assessment model to develop a toxicokinetic model for elucidating the site-specific temporal changes of As bioavailability and to characterize how the fish regulate the metal toxicity. We built a bioavailability-mode of action-based growth toxicity model by linking a bioenergetic growth model and damage assessment model to predict how the As affects on the tilapia growth in the entire life span in site-specific field ecosystems. Here we show that the proposed model well describes the water-chemistry-dependent toxicokinetics and toxicodynamics variations of As to tilapia. We selected two local tilapia farms with different water chemistries located at southwestern Taiwan coast region to implement the proposed algorithm to predict the risk of As exposure. Results indicate that the growth toxicity of O. mossambicus in Taihsi is more sensitive than that in Peimen. We found that the effect of ion competition on the As bioavailability and their ecotoxicological effects on tilapia are more obvious in Taihsi comparing with that in Peimen. We suggested that the proposed bioavailability- and mode of action-based framework can be used to capture the biological response and regulation of tilapia to As exposures. It is applicable for a site-specific and long-term ecotoxicological risk assessment.

Fluctuation exposure of trace metal copper (Cu) is ubiquitous in aquatic environments. The purpose of this study was to investigate the impacts of chronically pulsed exposure on biodynamics and subcellular partitioning of Cu in freshwater tilapia ( Oreochromis mossambicus). Long-term 28-day pulsed Cu exposure experiments were performed to explore subcellular partitioning and toxicokinetics/toxicodynamics of Cu in tilapia. Subcellular partitioning linking with a metal influx scheme was used to estimate detoxification and elimination rates. A biotic ligand model-based damage assessment model was used to take into account environmental effects and biological mechanisms of Cu toxicity. We demonstrated that the probability causing 50% of susceptibility risk in response to pulse Cu exposure in generic Taiwan aquaculture ponds was ~33% of Cu in adverse physiologically associated, metabolically active pool, implicating no significant susceptibility risk for tilapia. We suggest that our integrated ecotoxicological models linking chronic exposure measurements with subcellular partitioning can facilitate a risk assessment framework that provides a predictive tool for preventive susceptibility reduction strategies for freshwater fish exposed to pulse metal stressors. The accumulation of pollutants in the semi-enclosed waters of ports has long been a concern. This study assessed the pollution status, sources, and toxicity risks of 16 polycyclic aromatic hydrocarbons (PAHs) in surface sediments of 7 major ports in Taiwan. Total PAHs concentrations in sediments ranged between 8.4 and 572.5 ng/g dw, with an average of 112.4 ± 136.5 ng/g dw. The 3- and 4-ring PAHs (63 %) were the major constituents of PAHs in the sediments. Diagnostic ratios and positive matrix factorization analyses indicated that PAHs in sediments were mainly contributed by biomass combustion (45.0 %), coal combustion (31.5 %), and vehicle emissions and related fossil fuels (23.5 %). The results of ecological risk assessment showed a low–medium risk of PAHs in the sediments outside the port, whereas most of the sediments within the port presented a medium–high risk. An assessment of the possible human health risks indicated that PAHs were present at acceptable levels. For many decades, the traditional polyculture systems have contributed to increasing productivity throughout the Taiwan regions. It has been recognized that arsenic (As)-contaminated groundwater used for aquaculture in the southwestern coastal region of Taiwan is likely to pose a health threat to fish and humans. The purpose of this study was to assess farmed milkfish and tilapia exposure risk to As in a polyculture system using a model-based risk assessment framework. A first-order three-compartment model was used to simulate arsenic accumulation in fish and sediment appraised with the field-observed data. We constructed dose-response profiles obtained from acute toxicity bioassays to assess milkfish and tilapia exposure risk during different growing seasons. A probabilistic risk model was used to estimate the potential exposure risk. We showed that As accumulations in milkfish and tilapia were higher in summer than in fall. We found that there was a 20% probability for milkfish exceeding ~2% mortality in summer. However, waterborne As is not likely to pose a mortality risk for tilapia. Our results also revealed that tilapia benefited milkfish by reducing As concentrations in the water, indicating that tilapia can be biocontrol agents in the milkfish-tilapia polyculture systems. We suggest that the present mechanistic assessment framework can be used to assess exposure risk to environmental pollutants in polyculture systems and provide an appropriate exposure analysis to improve decision-making in the design and development of effective aquaculture systems. Abstract: The role of cadmium (Cd) bioaccessibility in risk assessment is less well studied. The aim of this study was to assess human health risk to Cd through inhalation and seafood consumption by incorporating bioaccessibility. The relationships between trophically available Cd and bioaccessibility were constructed based on available experimental data. We estimated Cd concentrations in human urine and blood via daily intake from seafood consumption and inhalation based on a physiologically-based pharmacokinetic (PBPK) model. A Hill-based dose–response model was used to assess human renal dysfunction and peripheral arterial disease risks for long-term Cd exposure. Here we showed that fish had higher bioaccessibility (∼83.7%) than that of shellfish (∼73.2%) for human ingestion. Our results indicated that glomerular and tubular damage among different genders and smokers ranged from 18.03 to 18.18%. Our analysis showed that nonsmokers had 50% probability of peripheral arterial disease level exceeding from 3.28 to 8.80%. Smoking populations had 2–3 folds higher morbidity risk of peripheral arterial disease than those of nonsmokers. Our study concluded that the adverse effects of Cd exposure are exacerbated when high seafood consumption coincides with cigarette smoking. It has been proposed that irreversible responses of organisms exposed to contaminants are due to a systems-level feedback. Here we tested this hypothesis by reanalyzing the published data on toxicokinetics and survival probability based on a systems-level threshold damage model (TDM) incorporating with a positive damage feedback to explore the steady-state response and dynamic behavior of damage for tilapia and freshwater clam exposed to waterborne arsenic (As). We found that ultrasensitivity appeared in As-tilapia and freshwater clam systems with Hill coefficient n ≥ 4, indicating that the positive damage feedback mechanism has been triggered. We confirmed that damage can trigger a positive feedback loop that together with As stressor increases irreversibility. This study also showed that TDM with positive feedback gave a much better predictability than that of TDM at As concentrations ranging from 100 to 500 mg l for freshwater clam, whereas for tilapia, two models had nearly same performance on predictability. We suggested that mortality-time profile derived Hill coefficient could be used as a new risk indicator to assess the survival probability for species exposed to waterborne metals. We anticipated that the proposed toxicokinetics/toxicodynamics with a positive damage feedback may facilitate our understanding and manipulation of complex mechanisms of metal susceptibility among species and improve current risk assessment strategies. The purpose of this article is to provide a risk-based predictive model to assess the impact of false mussel Mytilopsis sallei invasions on hard clam Meretrix lusoria farms in the southwestern region of Taiwan. The actual spread of invasive false mussel was predicted by using analytical models based on advection-diffusion and gravity models. The proportion of hard clam colonized and infestation by false mussel were used to characterize risk estimates. A mortality model was parameterized to assess hard clam mortality risk characterized by false mussel density and infestation intensity. The published data were reanalyzed to parameterize a predictive threshold model described by a cumulative Weibull distribution function that can be used to estimate the exceeding thresholds of proportion of hard clam colonized and infestation. Results indicated that the infestation thresholds were 2–17 ind clam−1 for adult hard clams, whereas 4 ind clam−1 for nursery hard clams. The average colonization thresholds were estimated to be 81–89% for cultivated and nursery hard clam farms, respectively. Our results indicated that false mussel density and infestation, which caused 50% hard clam mortality, were estimated to be 2,812 ind m−2 and 31 ind clam−1, respectively. This study further indicated that hard clam farms that are close to the coastal area have at least 50% probability for 43% mortality caused by infestation. This study highlighted that a probabilistic risk-based framework characterized by probability distributions and risk curves is an effective representation of scientific assessments for farmed hard clam in response to the nonnative false mussel invasion. The level and distribution of 16 USEPA Polycyclic aromatic hydrocarbon (PAHs) in aquaculture farmed fish (Mugil cephalus and Oreochromis mossambicus) and shellfish (Corbicula fluminea Formosa and Meretrix lusoria) were determined in Taiwan and then assessed cancer and non-cancer risks for those consuming these kinds of seafood. Results indicated that C. fluminea Formosa accumulated the highest average concentration of total PAHs (43.0 ± 11.3 ng/g wet weight) while M. lusoria contained the lowest concentration (20.0 ± 5.8 ng/g) among all species. The low-molecular-weight PAHs were dominant for both fish and shellfish, which consistent with other studies. Notably, parts of high-molecular-weight PAHs were found in shellfish whereas that was little in fish. The calculated hazard quotients (HQ) of all PAHs were smaller than 1 and the incremental lifetime cancer risks (ILCR) for Benzo[ a ]pyrene were below 1 × 10−5, suggesting that PAHs in the collected seafood could pose a low hazard to residents. Although the results indicated that the studied seafood is safe for human consumption, children and seniors post relatively higher risks, suggesting that it needs to continue monitoring and control the PAHs concentration in seafood and the associated environments. Focusing on 27 rare filter-feeding megamouth sharks (Megachasma pelagios) captured as a by-catch of drift gillnet fishery in the Pacific Ocean to the east of Taiwan, this study analyzes the concentrations of 24 elements in their muscle, discusses the bioaccumulation of each element and the correlation between different elements, and assesses the potential health risks of consuming megamouth shark muscle. Among the 24 elements, mean concentrations of Ga, Ag, Li, Bi, Hg, Co, and Cd were relatively low ranging from 10−3 to 10−1 mg/kg, those of Pb, Ba, Mn, Ni, As, Cr, B, Sr, Cu, and Zn ranged from 10−1–101 mg/kg, and those of Fe, Ca, Al, K, Mg, Ti, and Na were relatively high ranging from 101 to 103 mg/kg. The toxic element content index was most significantly correlated with the concentration of Cu. Hence, this study recommends that the concentration of Cu could be used as an indicator of metal accumulation in megamouth shark muscle. The log bioconcentration factor (BCF) ranged from less than 0 to 7.85 in shark muscle. For elements with a concentration of less than 100 μg/L in seawater, the log BCF was inversely proportional to their concentration in seawater. According to the correlation analysis, the accumulation of elements in muscle of megamouth sharks is primarily affected by the concentrations of dissolved elements in seawater, except that the accumulation of Hg, As, Cu, Ti, Al, and Fe appears to be mainly affected by feeding behaviors. The assessment of the health risk of consuming megamouth shark muscle showed that its total hazard index was greater than 1. This suggests that the long-term or high-frequency consumption of megamouth shark muscle may cause health hazards due to the accumulation of trace elements, particularly those with a large contribution of health risk, including As, Hg, and Cu. Water, suspended particulate matter (SPM), and sediment samples were collected from Salt River in Taiwan and analyzed the concentrations of 16 types of polycyclic aromatic hydrocarbons (PAHs). The analysis results were used to examine the distribution, source, partition behavior, and potential ecological risks of PAHs in the estuarine water systems. The mean concentration of total PAHs in water, SPM, and sediment samples was 0.485–10.2 μg/L, 26.7–169 mg/kg dw, and 0.343–29.4 mg/kg dw, respectively. The highest concentration was found at the river mouth and decreased toward the river and sea with the tide. The distribution of the diagnostic ratios of PAHs showed that the combustion of coal and petroleum products are the main sources of PAHs in Salt River. The in site organic carbon normalized partition coefficients for SPM-water (K' oc(SPM W)) and sediment-water (K' oc(Sed W)) were 2.8–4.5 and 4.6–6.0 (log units), respectively, increasing with the number of rings in PAHs. The values log K' oc(Sed W) and log K' oc(SPM-W) showed a significant linear correlation with their octanol-water partition coefficients (p < 0.01), and their slopes were 0.427 and 0.316, respectively. The fugacity fraction was used to evaluate the exchange of PAHs in water-SPM-sediment systems. Results showed that in SPM, 2–4-ring PAHs tend to be released into water, whereas 5–6-ring PAHs in water tend to be adsorbed onto SPM. The exchange of PAHs between water and sediment occurs in the direction of adsorption onto sediment from water. The assessment of the mean risk quotient, total toxicity equivalence, and mean effect range-median quotient of PAHs showed that the PAHs in the water and SPM of Salt River may have moderate to high ecological risk. In sediment, PAHs in the lower reaches and estuary may pose moderate to high ecological risk, whereas PAHs in the middle and upper reaches show low to moderate ecological risk. Abstract: The purpose of this study is to conduct a long-term site-specific risk assessment for zinc (Zn) susceptibility of bivalves, green mussel Perna viridis and hard clam Ruditapes philippinarum, based on published experimental data by linking the biologically-based damage assessment model with the subcellular partitioning concept. A comprehensive risk modeling framework was developed to predict susceptibility probability of two bivalve species exposed to waterborne Zn. The results indicated that P. viridis accumulates more Zn toxicity, whereas both toxic potency and the recovery rate of Zn are higher for R. philippinarum. We found that negative linear correlations exist in elimination–recovery and elimination–detoxification relationships, whereas a positive linear correlation was observed in recovery–detoxification relationships for bivalves exposed to waterborne Zn. Simulation results showed that the spatial differences of susceptibility primarily resulted from the variation of waterborne Zn concentration under field conditions. We found that R. philippinarum is more susceptible of Zn than P. viridis under the same exposure condition. Results also suggested that Zn posed no significant susceptibility risk to two bivalve species in Taiwan. Abstract: Exposures to carcinogenic polycyclic aromatic hydrocarbons (PAHs) have been linked to human lung cancer. The purpose of this study was to assess lung cancer risk caused by inhalation exposure to nano/ultrafine particle-bound PAHs at the population level in Taiwan appraised with recent published data. A human respiratory tract model was linked with a physiologically based pharmacokinetic model to estimate deposition fraction and internal organic-specific PAHs doses. A probabilistic risk assessment framework was developed to estimate potential lung cancer risk. We reanalyzed particle size distribution, total-PAHs, particle-bound benzo(a)pyrene (B[a]P) and PM concentrations. A dose–response profile describing the relationships between external B[a]P concentration and lung cancer risk response was constructed based on population attributable fraction (PAF). We found that 90% probability lung cancer risks ranged from 10−5 to 10−4 for traffic-related nano and ultrafine particle-bound PAHs, indicating a potential lung cancer risk. The particle size-specific PAF-based excess annual lung cancer incidence rate due to PAHs exposure was estimated to be less than 1 per 100,000 population, indicating a mild risk factor for lung cancer. We concluded that probabilistic risk assessment linked PAF for limiting cumulative PAHs emissions to reduce lung cancer risk plays a prominent role in future government risk assessment program. Abstract: Endotoxin, a component of gram-negative bacterial cell walls, is a proinflammatory agent that induces local and systemic inflammatory responses in normal subjects which can contribute to the risk of developing asthma and chronic obstructive lung diseases. A probabilistic approach linking models of exposure, internal dosimetry, and health effects were carried out to quantitatively assess the potential inhalation risk of airborne endotoxin for workers in cotton textile plants. Combining empirical data and modeling results, we show that the half-maximum effects of the endotoxin dose (ED50) were estimated to be 3.3×105 (95% confidence interval (CI): 1.9–14.7×105) endotoxin units (EU) for the blood C-reactive protein (CRP) concentration, 1.1×105 (95% CI: 0.6–1.7×105)EU for the blood polymorphonuclear neutrophil (PMN) count, and 1.5×105 (95% CI: 0.4–2.5×105)EU for the sputum PMN count. Our study offers a risk-management framework for discussing future establishment of limits for respiratory exposure to airborne endotoxin for workers in cotton textile plants. Abstract: Endotoxin, a component of gram-negative bacterial cell walls, is a pro-inflammatory agent that induces local and systemic inflammatory responses in normal subjects which can contribute to the risk of developing asthma and chronic obstructive lung diseases. A probabilistic approach linking models of exposure, internal dosimetry, and health effects was carried out to quantitatively assess the potential inhalation risk of airborne endotoxin in homes during the winter and summer seasons. Combining empirical data and modeling results, we show that the half-maximum effect of the endotoxin dose (ED50) was estimated to be 707.9 (95% confidence interval (CI): 308.8–1287.0) endotoxin units (EU) for body temperature change, 481.8 (95% CI: 333.2–630.3) EU for elevation of neutrophils, and 1174.5 (95% CI: 816.0–1532.9) EU for elevation of the cytokine, interleukin-6. Our study also suggests that airborne endotoxin in homes may pose potential risks, and a higher risk for elevation of neutrophils and cytokine interleukin-6 appeared in winter season than in summer.
