Background: Cinmethylin, a pre‐emergence herbicide inhibiting fatty acid thioesterase activity, has recently been introduced to Australian cereal cropping for the control of Lolium rigidum Gaud. (annual ryegrass). To date, there have been no confirmed cases of cinmethylin resistance identified in this species, but some populations exhibit reduced sensitivity to this herbicide. To explore the mechanism which contributes to reduced sensitivity of annual ryegrass to cinmethylin, the extent and nature of cinmethylin metabolism, using carbon‐14 (14C)‐labelled herbicide, were analysed in three reduced‐sensitivity annual ryegrass populations, alongside a susceptible population and cinmethylin‐tolerant wheat as controls. Results: All samples showed the same metabolite profile, with the extent of production of a specific water‐soluble metabolite being correlated to the level of herbicide sensitivity. Application of the cytochrome P450 inhibitor phorate caused a decrease in water‐soluble metabolite production as well as seedling growth in the presence of cinmethylin, indicating that reduced cinmethylin sensitivity in annual ryegrass could be wholly or partially due to oxidative modification of cinmethylin. Conclusion: Because annual ryegrass has the potential to metabolize cinmethylin in the same way as wheat, careful stewardship is required to ensure the longevity of this herbicide. © 2022 The Authors. Lolium rigidum Gaud., a grass weed species infesting winter field crops, has evolved resistance to the largest number of herbicide modes of action. In this study, 140 field populations of L. rigidum were screened with 14 herbicide treatments. Herbicide resistance at the recommended label dosage of pre‐emergence (PRE), post‐emergence (POST) and binary herbicide mixtures was considered present when plant survival was ≥6%. Plant survival to four acetyl‐CoA carboxylase (ACCase) POST herbicides averaged across all populations was approximately 15%, indicating substantial herbicide resistance. In contrast, the mean survival to the PRE treatments was only 2%, reflecting effective control of L. rigidum. Herbicide mixtures were the most effective treatments, with a significantly lower resistance frequency than stand‐alone herbicides. For example, only 12% of the tested samples were resistant to the mixture of clethodim + butroxydim in comparison with 40% and 61% to either butroxydim or clethodim, respectively. Similarly, 8% of the samples were resistant to the mixture of trifluralin + prosulfocarb versus a much greater frequency of 36% and 51% resistance to prosulfocarb and trifluralin, respectively. Surprisingly, the binary mixtures of trifluralin + triallate or pyroxasulfone + triallate are not affected by resistance (presently) due to the greater efficacy than that of either stand‐alone herbicide. Thus, herbicide mixtures can delay the onset of resistance and mitigate the existing levels of herbicide resistance and cross‐resistance in L. rigidum. Systematic screenings of a large number of field populations could identify the most (cost‐) effective herbicide mixtures and foster their informed adoption on farm to mitigate the rapid evolution of weed resistance in lieu of expert assumptions or modelling simulations. BACKGROUND Resistance to the dinitroaniline herbicide trifluralin in Lolium rigidum (annual ryegrass) often is mediated by the enhanced capacity to metabolize the herbicide to less toxic polar conjugates and/or by functionally recessive target‐site mutations in α‐tubulin. RESULTS: In two L. rigidum populations possessing enhanced trifluralin metabolism, resistance was largely reversed by recurrent selection with the thiocarbamate herbicide prosulfocarb (i.e. plant survival was two‐ to >20‐fold lower). Their ability to metabolize trifluralin was significantly decreased (by ≈2.3‐fold) following recurrent prosulfocarb selection, to levels comparable to those observed in susceptible plants or when trifluralin metabolism was inhibited by treatment with the insecticide phorate. CONCLUSIONS: This study provides evidence that trait(s) enabling efficient trifluralin metabolism in L. rigidum are purged from the population under prosulfocarb recurrent selection. The level of trifluralin metabolism in vitro and its inhibition caused by phorate action on trifluralin‐metabolizing enzyme(s) is equivalent to the effect produced by prosulfocarb selection. The hypothetical link between the two phenomena is that the putative monooxygenase(s) conferring trifluralin metabolic resistance also mediate the activation of prosulfocarb to its toxic sulfoxide. Thus, we speculate that survival to prosulfocarb via a lack of metabolic herbicide activation, and survival to trifluralin conferred by enhanced herbicide metabolism, are mutually exclusive. BACKGROUND Multiple‐herbicide resistance in Lolium rigidum and other weed species is increasingly exerting pressure on herbicide discovery research for solutions against resistance‐prone weeds. In this study we investigate: (i) the responses of L. rigidum populations and wheat to the new herbicide cinmethylin in comparison with other pre‐emergence herbicides, (ii) the effect of seed burial depths on cinmethylin efficacy and crop selectivity, and (iii) the basis of cinmethylin selectivity in wheat. RESULTS: Cinmethylin at 400 g ha−1 controls herbicide‐susceptible and multiple‐resistant L. rigidum, with a reduction of >85% in plant emergence and 90% in aboveground biomass. Cinmethylin provides effective control of a large number of field populations of L. rigidum with evident resistance to trifluralin. When the wheat seed is buried ≥1 cm below the cinmethylin‐treated soil surface, the emergence of crop seedlings is not different from the untreated control. The organophosphate insecticide phorate synergizes cinmethylin toxicity in wheat, with an LD50 of 682 g ha−1 in the absence of phorate versus 109 g ha−1 in the presence of phorate (84% reduction). The synergistic effect of phorate with cinmethylin on herbicide‐susceptible L. rigidum appears smaller (a 44% reduction in the LD50 of cinmethylin). CONCLUSIONS: Cinmethylin is effective in controlling multiple‐resistant L. rigidum and appears safe for wheat when the seed is separated at depth from the herbicide applied to the soil surface. BACKGROUND Organophosphate insecticides can inhibit specific cytochrome P450 enzymes involved in metabolic herbicide resistance mechanisms, leading to synergistic interactions between the insecticide and the herbicide. In this study we report synergistic versus antagonistic interactions between the organophosphate insecticide phorate and five different herbicides observed in a population of multiple herbicide-resistant Lolium rigidum. RESULTS Phorate synergised with three different herbicide modes of action, enhancing the activity of the ALS inhibitor chlorsulfuron (60% LD50 reduction), the VLCFAE inhibitor pyroxasulfone (45% LD50 reduction) and the mitosis inhibitor trifluralin (70% LD50 reduction). Conversely, phorate antagonised the two thiocarbamate herbicides prosulfocarb and triallate with a 12-fold LD50 increase. CONCLUSION We report the selective reversal of P450-mediated metabolic multiple resistance to chlorsulfuron and trifluralin in the grass weed L. rigidum by synergistic interaction with the insecticide phorate, and discuss the putative mechanistic basis. Herbicides that act by inhibiting the biosynthesis of very-long-chain fatty acids ( VLCFAs) have been used to control grass weeds in major crops throughout the world for the past 60 years. VLCFA-inhibiting herbicides are generally highly selective in crops, induce similar symptoms in susceptible grasses and can be found within the herbicide groups classified by the HRAC as K3 and N. Even after many years of continuous use, only 12 grass weed species have evolved resistance to VLCFA-inhibiting herbicides. Here, the cases of resistance that have evolved in major grass weed species belonging to the Avena, Echinochloa and Lolium genera in three different agricultural systems are reviewed. In particular we explore the possible reasons why VLCFA herbicides have been slow to select resistant weeds, outline the herbicide mode of action and discuss the resistance mechanisms that are most likely to have been selected. © 2014 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Abstract: In two different locations of the Western Australian “wheatbelt”, Lolium rigidum (rigid ryegrass) seeds were collected from organic fields (no herbicide use) and neighbouring conventional fields (persistent herbicide use), the latter infested with herbicide-resistant plants, to investigate the occurrence of gene flow among field populations as revealed by herbicide resistance gene transfer. Herbicides targeting acetyl-CoA carboxylase (ACCase) or acetolactate-synthase (ALS) were used to detect herbicide-resistant plants. Overall, the frequency of plants resistant to ACCase- or ALS-inhibiting herbicides was, respectively, 21% and 74% in the conventional fields and 2% and 37% in neighbouring organic fields. Mutant, herbicide-resistant ACCase and ALS alleles were detected in 16% and 38% of plants from conventional fields and in 0.53% and 3.7% of plants from organic fields. Identical mutant, herbicide-resistant ALS haplotypes were detected both in conventional and organic fields, supporting the occurrence of gene flow between L. rigidum populations in different fields. Gene flow can thus substantially increase the frequency of herbicide-resistant plants in unselected L. rigidum populations. Although gene flow cannot be prevented, it can be limited or managed. Hygiene tactics such as clean crop seed, weed seed removal at harvest and seed destruction post-harvest should be considered in order to minimize gene transfer among farms. [Copyright &y& Elsevier]

This is the first report of low-dose glyphosate selection causing a shift towards paraquat resistance. Herbicide resistance in weed species is a serious threat to world agriculture. We report rapid resistance evolution in the genetically variable cross-pollinated grass weed Lolium rigidum when recurrently selected with low doses. Results show that an herbicide-susceptible L. rigidum population selected over three generations with below-label doses of glyphosate exhibited not only glyphosate resistance evolution but also a progressive and concomitant shift in sensitivity to the structurally unrelated herbicide paraquat. Thus, reduced paraquat sensitivity was a consequence of recurrent selection with glyphosate at low doses. In the three-time glyphosate-selected progeny, the estimated paraquat dose to cause 50% mortality (LD) was 4-fold greater than for the unselected susceptible parent. Studying the evolutionary outcomes of below-label herbicide dose selection can help prevent genetic changes in weed populations and sustain the efficacy of herbicides widely used in world agriculture. Gene flow promotes genetic exchange among plant populations mediating evolutionary dynamics; yet, the importance of gene flow at distance via pollen movement is poorly understood. A field experiment at the landscape level was conducted with Lolium rigidum herbicide-susceptible individuals (population VLR1) placed into an otherwise Lolium-free bushland environment at increasing distances from adjacent large commercial crop fields infested with herbicide-resistant L. rigidum. Herbicide resistance was used as a marker to quantify the distance and the rate of pollen-mediated gene flow. About 21,245 seeds were produced on the isolated, susceptible mother plants of which 3,303 seedlings were tested for herbicide resistance and 664 seedlings were found to be resistant. Pollen-mediated gene flow occurred at 3,000 m (maximum tested distance). Both Mendelian and molecular analyses (sequencing and CAPS markers) confirmed the introgression of herbicide resistance genes. This is the first documented case of long-distance gene flow in L. rigidum. The results are important for future modeling simulations of herbicide resistance evolution and subsequent mobility. The adoption of integrated agronomic strategies, the control of potential receptor plants on fields’ margins and conservative use of herbicides can be realistic options to minimize herbicide resistance spread. Overreliance on herbicides for weed control is conducive to the evolution of herbicide resistance. Lolium rigidum (annual ryegrass) is a species that is prone to evolve resistance to a wide range of herbicide modes of action. Rapid detection of herbicide-resistant weed populations in the field can aid farmers to optimize the use of effective herbicides for their control. The feasibility and utility of a rapid 7-d agar-based assay to reliably detect L. rigidum resistant to key pre- and post-emergence herbicides including clethodim, glyphosate, pyroxasulfone and trifluralin were investigated in three phases: correlation with traditional pot-based dose-response assays, effect of seed dormancy, and stability of herbicides in agar. Easy-to-interpret results were obtained using non-dormant seeds from susceptible and resistant populations, and resistance was detected similarly as pot-based assays. However, the test is not suitable for trifluralin because of instability in agar as measured over a 10-d period, as well as freshly-harvested seeds due to primary dormancy. This study demonstrates the utility of a portable and rapid assay that allows for on-farm testing of clethodim, glyphosate, and pyroxasulfone resistance in L. rigidum, thereby aiding the identification and implementation of effective herbicide control options.
