Abstract The study established experimental transects in undisturbed areas of the Caohai Nature Reserve in Weining, Guizhou Province. The study aims to examine complete successional transects in different landscapes: non-karst, karst, and vegetation restoration, using the spatiotemporal substitution method. It analyzes the distribution patterns of Total potassium (TK) and Avail potassium (AK) in the soil and employs a Generalized Linear Mixed Model (GLMM) to investigate the effects of geomorphology, soil aggregates, and their interactions on the changes in soil potassium(K) elements. Furthermore, it explores the synergistic effects of K cycling during the vegetation restoration process in karst soils. The findings indicated that: (1) In karst areas, the overall TK content was higher than in non-karst areas, while the AK content in the topsoil was higher in non-karst areas. The TK content in the karst areas tended to approach that of the vegetation restoration zone. (2) After restoration, the dominant plants changed from Gramineae to Gramineae + Asteraceae, and the vegetation richness increased, thereby improving the soil fertility and potassium fixation capacity. (3) The GLMM showed that the karst landscape had a significant effect on the TK and AK contents of surface soil. In the early stage of vegetation restoration, the joint effect of soil and vegetation affected the TK and AK contents in soil.

The polycyclic skeleton of tris(4,5-dehydro-2,3:6,7-dibenzotropone) is a key structural fragment in carbon schwarzites, a theoretical form of negatively curved carbon allotrope. This report presents a new synthesis of this compound using a Ni-mediated Yamamoto coupling reaction and structural analysis of it with X-ray crystallography. Interestingly, it is observed that tris(4,5-dehydro-2,3:6,7-dibenzotropone) crystallized from its solution in hexane resulting in colorless and yellow crystal polymorphs, where it adopts conformations of approximate Cs and C2 symmetry, respectively. Furthermore, expanding its π-skeleton through the Barton–Kellogg and Scholl reactions led to the successful synthesis of a curved polycyclic arene containing three heptagons and two pentagons.

Abstract Social adjustment is critical to preschool children’s overall development and well-being. Household chaos and emotion regulation are important influencing factors that affect young children’s social adjustment. Our study examines the interplay between household chaos and social adjustment among preschool children and investigates the mediating function of emotion regulation. Three parent report scales with sufficient reliability and validity were completed by parents from six kindergartens: the Confusion, Hubbub and Order Scale (CHAOS), Emotion Regulation Checklist (ERC), and Social Competence and Behavior Evaluation (SCBE-30). The results showed that household chaos can directly and indirectly predict social adjustment via the intermediary effect of emotion regulation. Understanding and addressing household chaos, as well as supporting children in developing emotion regulation skills, can play a significant role in promoting social adjustment and mental health in preschool children.

Abstract With the increase in high-rise buildings in cities, public flue exhaust systems have a direct impact on residential air quality and the living environment. Although existing studies have analyzed the problems in public flue exhaust systems through computational fluid dynamics (CFD) numerical simulations and experimental tests, these studies often lack an in-depth exploration of the specific impacts of individual components in the system. To solve this problem, this study not only thoroughly analyzes the key components in the public flue system, such as branch pipes, check valves, and tee pipes, but also develops a parametric public flue simulation system software based on C# and verifies the accuracy of the simulation through experiments. The study first determines the key parameters affecting the comprehensive resistance coefficient of the branch pipe, check valve, tee pipe, and other components through CFD simulation and experimental testing. Subsequently, a visualization program is developed using the C# language, which can quickly simulate and visualize the flow changes in the flue according to different building parameters such as the number of floors, height of floors, and size of the flue. The results confirm that the program can efficiently predict the flow distribution under different design options, providing a practical tool for the optimal design and performance evaluation of public flues, which is important for improving the air quality of the living environment.

Abstract All-optical diffractive neural networks, as analog artificial intelligence accelerators, leverage parallelism and analog computation for complex data processing. However, their low space transmission efficiency or large spatial dimensions hinder miniaturization and broader application. Here, we propose a terahertz spoof plasmonic neural network on a planar diffractive platform for direct multi-target recognition. Our approach employs a spoof surface plasmon polariton coupler array to construct a diffractive network layer, resulting in a compact, efficient, and easily integrable architecture. We designed three schemes: basis vector classification, multi-user recognition, and MNIST handwritten digit classification. Experimental results reveal that the terahertz spoof plasmonic neural network successfully classifies basis vectors, recognizes multi-user orientation information, and directly processes handwritten digits using a designed input framework comprising a metal grating array, transmitters, and receivers. This work broadens the application of terahertz plasmonic metamaterials, paving the way for terahertz on-chip integration, intelligent communication, and advanced computing systems.

Abstract As robots are increasingly participating in our daily lives, the quests to mimic human abilities have driven the advancements of robotic multimodal senses. However, current perceptual technologies still unsatisfied robotic needs for home tasks/environments, particularly facing great challenges in multisensory integration and fusion, rapid response capability, and highly sensitive perception. Here, we report a flexible tactile sensor utilizing thin-film thermistors to implement multimodal perceptions of pressure, temperature, matter thermal property, texture, and slippage. Notably, the tactile sensor is endowed with an ultrasensitive (0.05 mm/s) and ultrafast (4 ms) slip sensing that is indispensable for dexterous and reliable grasping control to avoid crushing fragile objects or dropping slippery objects. We further propose and develop a robotic tactile-visual fusion architecture that seamlessly encompasses multimodal sensations from the bottom level to robotic decision-making at the top level. A series of intelligent grasping strategies with rapid slip feedback control and a tactile-visual fusion recognition strategy ensure dexterous robotic grasping and accurate recognition of daily objects, handling various challenging tasks, for instance grabbing a paper cup containing liquid. Furthermore, we showcase a robotic desktop-cleaning task, the robot autonomously accomplishes multi-item sorting and cleaning desktop, demonstrating its promising potential for smart housekeeping.

Abstract Intrastromal cell therapy utilizing quiescent corneal stromal keratocytes (qCSKs) from human donor corneas emerges as a promising treatment for corneal opacities, aiming to overcome limitations of traditional surgeries by reducing procedural complexity and donor dependency. This investigation demonstrates the therapeutic efficacy of qCSKs in a male rat model of corneal stromal opacity, underscoring the significance of cell-delivery quality and keratocyte differentiation in mediating corneal opacity resolution and visual function recovery. Quiescent CSKs-treated rats display improvements in escape latency and efficiency compared to wounded, non-treated rats in a Morris water maze, demonstrating improved visual acuity, while stromal fibroblasts-treated rats do not. Advanced imaging, including multiphoton microscopy, small-angle X-ray scattering, and transmission electron microscopy, revealed that qCSK therapy replicates the native cornea’s collagen fibril morphometry, matrix order, and ultrastructural architecture. These findings, supported by the expression of keratan sulfate proteoglycans, validate qCSKs as a potential therapeutic solution for corneal opacities.

Abstract Molecular computing is an emerging paradigm that plays an essential role in data storage, bio-computation, and clinical diagnosis with the future trends of more efficient computing scheme, higher modularity with scaled-up circuity and stronger tolerance of corrupted inputs in a complex environment. Towards these goals, we construct a spatially localized, DNA integrated circuits-based classifier (DNA IC-CLA) that can perform neuromorphic architecture-based computation at a molecular level for medical diagnosis. The DNA-based classifier employs a two-dimensional DNA origami as the framework and localized processing modules as the in-frame computing core to execute arithmetic operations (e.g. multiplication, addition, subtraction) for efficient linear classification of complex patterns of miRNA inputs. We demonstrate that the DNA IC-CLA enables accurate cancer diagnosis in a faster (about 3 h) and more effective manner in synthetic and clinical samples compared to those of the traditional freely diffusible DNA circuits. We believe that this all-in-one DNA-based classifier can exhibit more applications in biocomputing in cells and medical diagnostics.

Abstract Phosphorescence is ubiquitous in heavy atom-containing organic phosphors, which attracts considerable attention in optoelectronics and bioelectronics. However, heavy atom-free organic materials with efficient phosphorescence are rare under ambient conditions. Herein, we report a series of adaptive host-guest materials derived from dibenzo-heterocyclic analogues, showing host-dependent color-tunable phosphorescence with phosphorescence efficiency of up to 98.9%. The adaptive structural deformation of the guests arises from the hyperconjugation, namely the n→π* interaction, enabling them to inhabit the cavity of host crystals in synergy with steric effects. Consequently, a perfect conformation match between host and guest molecules facilitates the suppression of triplet exciton dissipation, thereby boosting the phosphorescence of these adaptive materials. Moreover, we extend this strategy to a ternary host-guest system, yielding both excitation- and time-dependent phosphorescence with a phosphorescence efficiency of 92.0%. This principle provides a concise way for obtaining efficient and color-tunable phosphorescence, making a major step toward potential applications in optoelectronics.

Summary: High salt (HS) consumption is a risk factor for multiple autoimmune disorders via disturbing immune homeostasis. Nevertheless, the exact mechanisms by which HS exacerbates rheumatoid arthritis (RA) pathogenesis remain poorly defined. Herein, we found that heightened phosphorylation of PDPK1 and SGK1 upon HS exposure attenuated FoxO1 expression to enhance the glycolytic capacity of CD4 T cells, resulting in strengthened Th17 but compromised Treg program. GSK2334470 (GSK), a dual PDPK1/SGK1 inhibitor, effectively mitigated the HS-induced enhancement in glycolytic capacity and the overproduction of IL-17A. Therefore, administration of GSK markedly alleviated HS-exacerbated RA progression in collagen-induced arthritis (CIA) model. Collectively, our data indicate that HS consumption subverts Th17/Treg homeostasis through the PDPK1-SGK1-FoxO1 signaling, while GSK could be a viable drug against RA progression in clinical settings.

Abstract The detection of pathogens is critical for clinical diagnosis and public health surveillance. Detection is usually done with nucleic acid-based tests (NATs) and rapid antigen tests (e.g., lateral flow assays [LFAs]). Although NATs are more sensitive and specific, their use is often limited in resource-poor settings due to specialized requirements. To address this limitation, we developed a rapid DNA-RNA Hybrid Capture immunoassay (HC) that specifically detects RNA from pathogens. This assay utilizes a unique monoclonal antibody, S9.6, which binds DNA-RNA hybrids. Biotinylated single-stranded DNA probes are hybridized to target RNAs, followed by hybrid capture on streptavidin and detection with S9.6. The HC-ELISA assay can detect as few as 104 RNA molecules that are 2.2 kb in length. We also adapted this assay into a LFA format, where captured Bacillus anthracis rpoB RNA of 3.5 kb length was detectable from a bacterial load equivalent to 107 CFU per 100 mg of mouse tissue using either HC-ELISA or HC-LFA. Importantly, we also demonstrated the versatility of HC by detecting other pathogens, including SARS-CoV-2 and Toxoplasma gondii, showing its potential for broad pathogen detection. Notably, HC does not require amplification of the target nucleic acid and utilizes economical formats like ELISA and LFA, making it suitable for use in sentinel labs for pathogen detection or as a molecular tool in basic research laboratories. Our study highlights the potential of HC as a sensitive and versatile method for RNA-based pathogen detection.

Introduction: The Qinnan Depression in the Bohai Bay Basin is lowly explored, where hydrocarbon generation potential of source rocks is required detailed investigation to identify hydrocarbon exploration potential and direction.Methods: This study is based on organic geochemical analysis performed on the Paleogene source rocks. After that, the burial, thermal maturity, and hydrocarbon generation histories of the Paleogene source rocks at various sags were reconstructed on BasinMod software based on reasonable geological models and geological parameters derived from geological analogy.Results: Results show that the 3rd member (Es3) and 1st member (Es1) of the Eocene Shahejie Formation are high-quality source rocks with a mixture of type I and II kerogen, whose organic matter was originated from aquatic organisms under a reducing environment. The 3rd member (Ed3) of the Oligocene Dongying Formation are fair-quality source rocks with type II kerogen and are dominated by mixed organic matter depositing in a weakly reducing environment. Clear differences in hydrocarbon generation were observed in these three source rocks owing to differential subsidence and sedimentation among the Eastern, Southeastern, and Western sags. Source rocks at the Eastern Sag were maturated early and lasted for a long time, which were currently at late mature (1.0–1.3%Ro) to high mature (1.3–2.0%Ro) stages. Two hydrocarbon generation events occurred in the Oligocene and Miocene, respectively, with intensive hydrocarbon generation capacity. Source rocks at the Southeastern Sag were maturated late and lasted for a short period, which were currently at a medium mature (0.7–1.0%Ro) stage. The major hydrocarbon generation event had occurred since the late Miocene, with intensive hydrocarbon generation capacity. Source rocks at the Western Sag were at an early mature (0.5–0.7%Ro) stage and began to generate hydrocarbon in the Pliocene, with weak hydrocarbon generation capacity. The hydrocarbon generation capacity of the Paleogene source rocks is as follows: Es3>Es1>Ed3.Discussion: The Qinnan Depression has high oil and gas exploration potential, where exploration activities should focus on effective traps around depression and slope zones located at the Eastern and Southeastern sags.

Abstract Selective vascular access to the brain is desirable in metabolic tracer, pharmacological and other studies aimed to characterize neural properties in isolation from somatic influences from chest, abdomen or limbs. However, current methods for artificial control of cerebral circulation can abolish pulsatility-dependent vascular signaling or neural network phenomena such as the electrocorticogram even while preserving individual neuronal activity. Thus, we set out to mechanically render cerebral hemodynamics fully regulable to replicate or modify native pig brain perfusion. To this end, blood flow to the head was surgically separated from the systemic circulation and full extracorporeal pulsatile circulatory control (EPCC) was delivered via a modified aorta or brachiocephalic artery. This control relied on a computerized algorithm that maintained, for several hours, blood pressure, flow and pulsatility at near-native values individually measured before EPCC. Continuous electrocorticography and brain depth electrode recordings were used to evaluate brain activity relative to the standard offered by awake human electrocorticography. Under EPCC, this activity remained unaltered or minimally perturbed compared to the native circulation state, as did cerebral oxygenation, pressure, temperature and microscopic structure. Thus, our approach enables the study of neural activity and its circulatory manipulation in independence of most of the rest of the organism.

Abstract To analyze the relationship between the composition of urinary stones and various influencing factors in the Enshi region. We used FT-IR to examine the composition of 1092 stone samples. Combined with the relevant clinical materials, the data were analyzed using both one-dimensional statistical methods and multivariate statistical methods. The study included 1092 stone samples, classified as follows: 457 (41.8%) with a single component, 453 (41.5%) with two components, 149 (13.6%) with three components, and 33 (3.0%) with four components. Stones were categorized into five types: Calcium Oxalate (CaOx) (76.4%), carbapatite (CaP) (9.3%), Struvite (ST) (8.3%), Uric Acid (UA) (4.9%), and Others (1.0%). Age, gender, urinary tract infection (UTI), family history of urinary stones (FH), hyperuricemia (HUA) and stone location were significantly associated with stone type. Logistic regression revealed that females and UTI were relative risk factors for predicting CaP and ST, while FH and HUA were relative risk factors for predicting UA. Our study indicates that the overall composition of urinary tract stones in the Enshi region is consistent with that of the entire China. Additionally, the predisposing factors for stone formation vary in terms of gender, age, FH, UTI, hyperuricemia HUA, and stone location.

The diurnal variation of surface incident solar radiation (Rs) has a significant impact on the Earth’s climate. Satellite-retrieved Rs datasets display good spatial and temporal continuity compared with ground-based observations and, more importantly, have higher accuracy than reanalysis datasets. Facilitated by these advantages, many scholars have evaluated satellite-retrieved Rs, especially based on monthly and annual data. However, there is a lack of evaluation on an hourly scale, which has a profound impact on sea–air interactions, climate change, agriculture, and prognostic models. This study evaluates Himawari-8 and Clouds and the Earth’s Radiant Energy System Synoptic (CERES)-retrieved hourly Rs data covering 60°S–60°N and 80°E–160°W based on ground-based observations from the Baseline Surface Radiation Network (BSRN). Hourly Rs were first standardized to remove the diurnal and seasonal cycles. Furthermore, the sensitivities of satellite-retrieved Rs products to clouds, aerosols, and land cover types were explored. It was found that Himawari-8-retrieved Rs was better than CERES-retrieved Rs at 8:00–16:00 and worse at 7:00 and 17:00. Both satellites performed better at continental sites than at island/coastal sites. The diurnal variations of statistical parameters of Himawari-8 satellite-retrieved Rs were stronger than those of CERES. Relatively larger MABs in the case of stratus and stratocumulus were exhibited for both hourly products. Smaller MAB values were found for CERES covered by deep convection and cumulus clouds and for Himawari-8 covered by deep convection and nimbostratus clouds. Larger MAB values at evergreen broadleaf forest sites and smaller MAB values at open shrubland sites were found for both products. In addition, Rs retrieved by Himawari-8 was more sensitive to AOD at 10:00–16:00, while that retrieved by CERES was more sensitive to COD at 9:00–15:00. The CERES product showed larger sensitivity to COD (at 9:00–15:00) and AOD (at 7:00–10:00) than Himawari-8. This work helps data producers know how to improve their future products and helps data users be aware of the uncertainties that exist in hourly satellite-retrieved Rs data.

The signal blockage and multipath effects of the Global Navigation Satellite System (GNSS) caused by urban canyon scenarios have brought great technical challenges to the positioning and navigation of autonomous vehicles. In this paper, an improved factor graph optimization algorithm enhanced by a resilient noise model is proposed. The measurement noise is resilient and adjusted based on an approximate Gaussian distribution-based estimation. In estimating and adjusting the noise parameters of the measurement model, the error covariance matrix of the multi-sensor fusion positioning system is dynamically optimized to improve the system accuracy. Firstly, according to the approximate Gaussian statistical property of the GNSS/odometer velocity residual sequence, the measured data are divided into an approximate Gaussian fitting region and an approximate Gaussian convergence region. Secondly, the interval is divided according to the measured data, and the corresponding variational Bayesian network and Gaussian mixture model are used to estimate the innovation online. Further, the noise covariance matrix of the adaptive factor graph-based model is dynamically optimized using the estimated noise parameters. Finally, based on low-cost inertial navigation equipment, GNSS, odometer, and vision, the algorithm is implemented and verified using a simulation platform and real-vehicle road test. The experimental results show that in a complex urban road environment, compared with the traditional factor graph fusion localization algorithm, the maximum improvement in accuracy of the proposed algorithm can reach 65.63%, 39.52%, and 42.95% for heading, position, and velocity, respectively.

Abstract Due to its ability to adapt to a variety of electromagnetic (EM) environments, the sensing‐enabled metasurface has garnered significant attention. However, large‐scale EM‐field sensing to obtain more information is still very challenging. Here, an adaptive information metasurface is proposed to enable intelligent sensing and wave manipulating simultaneously or more specifically, to realize intelligent target localization and beam tracking adaptively. The metasurface is composed of an array of meta‐atoms, and each is loaded with two PIN diodes and a sensing‐channel structure, for polarization‐insensitive and programmable beamforming and sensing. By controlling the state of the PIN diode, the proposed meta‐atom has 1‐bit phase response in the designed frequency band, while the sensing loss keeps higher than ‐10 dB for both “ON” and “OFF” states. Hence there is nearly no interaction between the beamforming and sensing modes. Experiments are conducted to show multiple functions of the metasurface, including intelligent target sensing and self‐adaptive beamforming, and the measured results are in good agreement with the numerical simulations and theoretical calculations.

Abstract As the most aggressive tumor, TNM staging does not accurately identify patients with pancreatic cancer who are sensitive to therapy. This study aimed to identify associated risk factors and develop a nomogram to predict survival in pancreatic cancer surgery patients and to select the most appropriate comprehensive treatment regimen. First, the survival difference between radiotherapy and no radiotherapy was calculated based on propensity score matching (PSM). Cox regression was conducted to select the predictors of overall survival (OS). The model was constructed using seven variables: histologic type, grade, T stage, N stage, stage, chemotherapy and radiotherapy. All patients were classified into high- or low-risk groups based on the nomogram. The nomogram model for OS was established and showed good calibration and acceptable discrimination (C-index 0.721). Receiver operating characteristic curve (ROC) and DCA curves showed that nomograms had better predictive performance than TNM stage. Patients were divided into low-risk and high-risk groups according to nomogram scores. Radiotherapy is recommended for high-risk patients but not for low-risk patients. We have established a well-performing nomogram to effectively predict the prognosis of pancreatic cancer patients underlying surgery. Abstract To explore the clinical application value of optical coherence microscopy (OCM) in Hirschsprung’s disease. 109 HSCR patients were recuited in a Chinese hospital from January 2018 to July 2021. All the recruited patients underwent barium enema angiography preoperatively and the resected diseased intestinal tubes were evaluated intraoperatively. The OCM and the histopathological examination were performed successively on the surgical specimens, and the OCM images were compared with the relevant tissue sections to characterize different lesions. 10 non-HSCR fetal colorectal tissues at the same period were retained for OCM, the characteristics of which with and without HSCR under OCM imaging were analyzed. In the OCM images of in vitro tissue, it can be clearly observed that the scattering degree of HSCR narrow segment mucosal is high, glands and crypt structures are reduced or even atrophy, and the scattering degree of submucosal and intermuscular is low; In the dilated segment, the low scattering and high scattering are complex, and the muscle layer is obviously hypertrophy and structural disorder. Compared with the pathological findings, the OCM sensitivity, Kappa value, and AUC area reached 92.66%, 0.63, and 0.91, respectively. OCM can quickly and clearly display the structure of all layers of colorectal tissue, which is highly consistent with the corresponding histopathological examination results and has high sensitivity. which will provide a more reliable basis for OCM diagnosis of early HSCR, targeted biopsy and location of operative treatment, and has a certain potential for clinical application.

Abstract The non-coding RNA secondary structure largely determines its function. Hence, accuracy in structure acquisition is of great importance. Currently, this acquisition primarily relies on various computational methods. The prediction of the structures of long RNA sequences with high precision and reasonable computational cost remains challenging. Here, we propose a deep learning model, RNA-par, which could partition an RNA sequence into several independent fragments (i-fragments) based on its exterior loops. Each i-fragment secondary structure predicted individually could be further assembled to acquire the complete RNA secondary structure. In the examination of our independent test set, the average length of the predicted i-fragments was 453 nt, which was considerably shorter than that of complete RNA sequences (848 nt). The accuracy of the assembled structures was higher than that of the structures predicted directly using the state-of-the-art RNA secondary structure prediction methods. This proposed model could serve as a preprocessing step for RNA secondary structure prediction for enhancing the predictive performance (especially for long RNA sequences) and reducing the computational cost. In the future, predicting the secondary structure of long-sequence RNA with high accuracy can be enabled by developing a framework combining RNA-par with various existing RNA secondary structure prediction algorithms. Pachytene Piwi-interacting RNAs (piRNAs) expressed in mammalian germ lines are abundant, but their evolution and function are not fully understood. Here, the authors find that pachytene piRNA loci are hotspots of structural variation, which underlies rapid piRNA birth, divergence, and loss.

The authors present an intelligent metasurface system that uses a target detection algorithm combined with a depth camera, to automatically detect the position of moving targets and achieve real-time wireless communications. The system can operate for multiple targets in limited ambient light, outdoor and other realistic environments.

Considering that laser scan stations are expensive and heavy for the indoor environment reconstruction of unmanned systems, a low-cost and light reconstruction system is proposed in this research. The system consists of a cross-structured light visual (SLV) sensor and an inertial navigation system (INS). The cross-SLV sensor is used to scan the surroundings and to estimate the angle change between two adjacent measurements. To improve the robustness and accuracy of the angle measurement, a Kalman Filter (KF) with simple construction is designed to fuse the inertial information from the INS. The factors which influence ranging accuracy are analyzed and ranging experiments show that the SLV sensor has an accuracy of higher than 2% when the distance is less than 4 m. Then the reconstruction results of a kitchen and corridor show that the error of most points is less than 50 mm for either kitchen (94%) or corridor (85%), and the mean errors and standard deviations of kitchen and corridor are less than 20 mm and 30 mm, respectively.

Summary: Cytosine methylation is an important epigenetic modification involved in regulation of plant development. However, the epigenetic mechanisms governing peanut seed development remain unclear. Herein, we generated DNA methylation profiles of developmental seeds of peanut H2014 and its smaller seed mutant H1314 at 15 and 60 days after pegging (DAP, S1, S4). Accompanying seed development, globally elevated methylation was observed in both lines. The mutant had a higher methylation level of 31.1% than wild type at S4, and 27.1–35.9% of the differentially methylated regions (DMRs) between the two lines were distributed in promoter or genic regions at both stages. Integrated methylome and transcriptome analysis revealed important methylation variations closely associated with seed development. Furthermore, some genes showed significantly negative correlation of expression with the methylation level within promoter or gene body. The results provide insights into the roles of DNA methylation in peanut seed development.

Abstract In this study, a prospective study was conducted by using optical coherence tomography (OCT) in the in vivo detection of vulvar diseases. The clinical efficacy of the OCT we investigated in the detection of vulvar diseases, and the characteristics of the OCT images were defined. Overall, this study recruited 63 patients undergoing the colposcopy for vulvar lesions in three Chinese hospitals from December 20th, 2018 and September 24th, 2019. The colposcopy and the OCT examination were performed successively, and the OCT images were compared with the relevant tissue sections to characterize different lesions. The OCT diagnoses where categorized into 7 types, including normal and inflammatory vulva, condyloma acuminata, papilloma, lichen sclerosus, atrophic sclerosing lichen, fibrous epithelial polyp as well as cysts. The structural characteristics of the vulva tissue can be clearly observed in the OCT image, which are consistent with the characteristics of the tissue section. Compared with the pathological results, the sensitivity, specificity and accuracy of the OCT examination reached 83.82% (95% confidence interval, CI 72.5%–91.3%), 57.89% (95% CI 34.0%–78.9%) and 78.16%, respectively. The OCT is found with the advantages of being noninvasive, real-time and sensitive and with high resolution. It is of high significance to screening vulva diseases, and it is expected as one of the methods to clinically diagnose vulva diseases.

Summary: Acute kidney injury (AKI) is a serious disease with no effective treatment. Abnormal opening of mitochondrial permeability transition pore (MPTP) is an important pathological process in ischemia reperfusion injury (IRI), the key factor of AKI. It is essential to elucidate MPTP regulation mechanism. Here, we identified mitochondrial ribosomal protein L7/L12 (MRPL12) specifically binds to adenosine nucleotide translocase 3 (ANT3) under normal physiological conditions, stabilizes MPTP and maintains mitochondrial membrane homeostasis in renal tubular epithelial cells (TECs). During AKI, MRPL12 expression was significantly decreased in TECs, and MRPL12-ANT3 interaction was reduced, leading to ANT3 conformation change, MPTP abnormal opening, and cell apoptosis. Importantly, MRPL12 overexpression protected TECs from MPTP abnormal opening and apoptosis during hypoxia/reoxygenation (H/R). Our results suggest MRPL12-ANT3 axis involves in AKI by regulating MPTP, and MRPL12 could be potential intervention target for treatment of AKI.

Abstract With the overconsumption of disposable plastics, there is a considerable emphasis on the recycling of waste plastics to relieve the environmental, economic, and health‐related consequences. Here, a sulfur‐assisted pyrolysis strategy is demonstrated for versatile upcycling of plastics into high‐value carbons with an ultrahigh carbon‐atom recovery (up to 85%). During the pyrolysis process, the inexpensive elemental sulfur molecules are covalently bonded with polymer chains, and then thermally stable intermediates are produced via dehydrogenation and crosslinking, thereby inhibiting the decomposition of plastics into volatile small hydrocarbons. In this manner, the carbon products obtained from real‐world waste plastics exhibit sulfur‐rich skeletons with an enlarged interlayer distance, and demonstrate superior sodium storage performance. It is believed that the present results offer a new solution to alleviate plastic pollution and reduce the carbon footprint of plastic industry.

Abstract Deep coal seam mining often leads to water resource loss due to bedrock water entering the workings of the mine and is discharged adjacent to the mining area. Using the geological conditions of the Maiduoshan coal mine, this paper applied a physical simulation experiment. The specified rock above the coal seam was hydraulically fractured in advance to form a postmining grouted fracture network, followed by grouting to construct a flexible isolation layer that blocked the infiltration of groundwater from the aquifer into the water-conducting fracture zone. Stress sensors, flow sensors and strata displacement monitoring technology were deployed inside the experimental material to study the spatial distribution characteristics and evolution law of the water-conducting fracture zone in the overlying rocks. Analysis of the water-conducting fracture zone development law, stress variation, overburden evolution characteristics, fracturing and grouting sequence of the flexible isolation layer and the effect of postmining grouting on the water barrier was conducted. These experiments verified the feasibility of fracture and grouting of the flexible isolation layer. These research results will provide practical guidance for the transition from the current safe and efficient mining methods to safe and green mining methods of deep coal mining in the western mining areas of China.

There are great differences in oil and gas enrichment at different locations in petroliferous basins, and faults are an important factor affecting the differential enrichment of oil and gas. In this paper, taking the No. 2 structure in the Nanpu Sag, Bohai Bay Basin, East China, as an example, based on the analysis of fault geometry and kinematics characteristics, the determination of oil source faults, the analysis of fault trap origin and effectiveness, and the analysis of fault caprock combinations are carried out to study the differences and mechanisms of oil and gas enrichment east and west of the Nanpu No. 2 structure. The results show that the fault-source contact relationship affects the direction of oil and gas migration. The No. 2 structure in the Nanpu Sag has a dual-source hydrocarbon supply. The western structure supplies hydrocarbons from the north and east, with oil and gas migrating along the direction parallel and perpendicular to the fault strike. The east structure supplies hydrocarbons from the south and north, with oil and gas migrating along the direction perpendicular to the fault strike. The effectiveness of fault traps affects the scale of oil and gas accumulation. Most of the traps of the Nanpu No. 2 structure formed before hydrocarbon expulsion, which are all effective traps and are conducive to oil and gas accumulation. The relationship between fault and caprock determines the vertical accumulation layers of oil and gas: The faulted thickness of the east structure is large, the fault is vertically closed, and oil and gas are enriched mainly in the deep layer. The faulted thickness of the west structure is small, and oil and gas are enriched in both deep and shallow layers.

Bidirectional DNA replication complexes initiated from the same origin remain colocalized in a factory configuration for part or all their lifetimes. However, there is little evidence that sister replisomes are functionally interdependent, and the consequence of factory replication is unknown. Here, we investigated the functional relationship between sister replisomes in Escherichia coli, which naturally exhibits both factory and solitary configurations in the same replication cycle. Using an inducible transcription factor roadblocking system, we found that blocking one replisome caused a significant decrease in overall progression and velocity of the sister replisome. Remarkably, progression was impaired only if the block occurred while sister replisomes were still in a factory configuration – blocking one fork had no significant effect on the other replisome when sister replisomes were physically separate. Disruption of factory replication also led to increased fork stalling and requirement of fork restart mechanisms. These results suggest that physical association between sister replisomes is important for establishing an efficient and uninterrupted replication program. We discuss the implications of our findings on mechanisms of replication factory structure and function, and cellular strategies of replicating problematic DNA such as highly transcribed segments.

Abstract RNA-seq has matured and become an important tool for studying RNA biology. Here we compared two RNA-seq (MGI DNBSEQ and Illumina NextSeq 500) and two microarray platforms (GeneChip Human Transcriptome Array 2.0 and Illumina Expression BeadChip) in healthy individuals administered recombinant human erythropoietin for transcriptome-wide quantification of differential gene expression. The results show that total RNA DNB-seq generated a multitude of target genes compared to other platforms. Pathway enrichment analyses revealed genes correlate to not only erythropoiesis and oxygen transport but also a wide range of other functions, such as tissue protection and immune regulation. This study provides a knowledge base of genes relevant to EPO biology through cross-platform comparisons and validation.

Abstract Chinese prickly ash (Zanthoxylum) is extensively used as spice and traditional medicine in eastern Asian countries. Recently, an emergent yellow-flower disease (YFD) break out in green Chinese prickly ash (Zanthoxylum schinifolium, Qinghuajiao in Chinese) at Chongqing municipality, and then leads to a sharp reduction in the yield of Qinghuajiao, and thus results in great economic losses for farmers. To address the molecular response for the emergent YFD of Qinghuajiao, we analyzed the transcriptome of 12 samples including the leaves and inflorescences of asymptomatic and symptomatic plants from three different towns at Chongqing by high-throughput RNA-Seq technique. A total of 126,550 genes and 229,643 transcripts were obtained, and 21,054 unigenes were expressed in all 12 samples. There were 56 and 164 different expressed genes (DEGs) for the AL_vs_SL (asymptomatic leaf vs symptomatic leaf) and AF_vs_SF (asymptomatic flower vs symptomatic flower) groups, respectively. The results of KEGG analysis showed that the “phenylpropanoid biosynthesis” pathway that related to plant–pathogen interaction were found in AL_vs_SL and AF_vs_SF groups, and the “Plant–pathogen interaction” found in AF_vs_SF group, implying that this Qinghuajiao YFD might cause by plant pathogen. Interestingly, we detected 33 common unigenes for the 2 groups, and almost these unigenes were up-regulated in the symptomatic plants. Moreover, most of which were homologs to virus RNA, the components of viruses, implying that this YFD was related to virus. Our results provided a primary molecular basis for the prevention and treatment of YFD of Qinghuajiao trees.

Abstract Time-to-fall off an accelerating rotating rod (rotarod) is widely utilized to evaluate rodent motor performance. We reasoned that this simple outcome could be refined with additional measures explicit in the task (however inconspicuously) to examine what we call movement sub-structure. Our goal was to characterize normal variation or motor impairment more robustly than by using time-to-fall. We also hypothesized that measures (or features) early in the sub-structure could anticipate the learning expected of a mouse undergoing serial trials. Using normal untreated and baclofen-treated movement-impaired mice, we defined these features and automated their analysis using paw video-tracking in three consecutive trials, including paw location, speed, acceleration, variance and approximate entropy. Spectral arc length yielded speed and acceleration uniformity. We found that, in normal mice, paw movement smoothness inversely correlated with rotarod time-to-fall for the three trials. Greater approximate entropy in vertical movements, and opposite changes in horizontal movements, correlated with greater first-trial time-to-fall. First-trial horizontal approximate entropy in the first few seconds predicted subsequent time-to-fall. This allowed for the separation, after only one rotarod trial, of different-weight, untreated mouse groups, and for the detection of mice otherwise unimpaired after baclofen, which displayed a time-to-fall similar to control. A machine-learning support vector machine classifier corroborated these findings. In conclusion, time-to-fall off a rotarod correlated well with several measures, including some obtained during the first few seconds of a trial, and some responsive to learning over the first two trials, allowing for predictions or preemptive experimental manipulations before learning completion.

Gold nanoparticles (Au NPs) with enzyme-like activities are useful glucose oxidase mimics, but the insights into the mechanism of this reaction are limited. Here, the authors show that the process of glucose oxidation by Au NPs is analogous to the one catalysed by glucose oxidase, involving dehydrogenation and oxygen reduction to H2O2; and that other noble metal NPs also catalyse glucose dehydrogenation, but oxygen is preferably reduced to water.

The Local Climate Zone (LCZ) classification scheme is a vital method of building a category dataset for high-resolution urban land. For the development of urban meteorology, air pollution and related disciplines, the high-resolution classification data of urban buildings are very important. This study aims to create LCZ datasets with detailed architectural characteristics for major cities and urban agglomerations in China, and obtain more accurate results. We constructed 120 m resolution land use datasets for 63 cities (mainly provincial capitals, municipalities directly under the Central Government, important prefecture-level cities and special administrative regions) and 4 urban agglomerations in China based on the local climate zone (LCZ) classification scheme using the World Urban Database and Access Portal Tools method (WUDAPT). Nearly 100,000 samples were used, of which 76,000 training samples were used to provide spectral signatures and 23,000 validation samples were used to ensure accuracy assessments. Compared with similar studies, the LCZ datasets in this paper were generally of good quality, with an overall accuracy of 71–93% (mean 82%), an accuracy for built classifications of 57–83% (mean 72%), and an accuracy for natural classifications of 70–99% (mean 90%). In addition, 35% of 63 Chinese cities have construction areas of more than 5%, and the plateaus northwest of Chengdu and Chongqing are covered with snow all year round. Therefore, based on the original LCZ classification system, the construction area (LZC H) and the snow cover (LCZ I) were newly added as the basic classifications of urban LCZ classification in China. Detailed architectural features of cities and urban agglomerations in China are provided by the LCZ datasets in this study. It can be applied to fine numerical models of the meteorological and atmospheric environment and improve the prediction accuracy.

Surface incident solar radiation (Rs) plays an important role in climate change on Earth. Recently, the use of satellite-retrieved datasets to obtain global-scale Rs with high spatial and temporal resolutions has become an indispensable tool for research in related fields. Many studies were carried out for Rs evaluation based on the monthly satellite retrievals; however, few evaluations have been performed on their diurnal variation in Rs. This study used independently widely distributed ground-based data from the Baseline Surface Radiation Network (BSRN) to evaluate hourly Rs from the Clouds and the Earth’s Radiant Energy System Synoptic (CERES) SYN1deg–1Hour product through a detrended standardization process. Furthermore, we explored the influence of cloud cover and aerosols on the diurnal variation in Rs. We found that CERES-retrieved Rs performs better at midday than at 7:00–9:00 and 15:00–17:00. For spatial distribution, CERES-retrieved Rs performs better over the continent than over the island/coast and polar regions. The Bias, MAB and RMSE in CERES-retrieved Rs under clear-sky conditions are rather small, although the correlation coefficients are slightly lower than those under overcast-sky conditions from 9:00 to 15:00. In addition, the range in Rs bias caused by cloud cover is 1.97–5.38%, which is significantly larger than 0.31–2.52% by AOD.

Structured illumination microscopy is usually limited to 2 times spatial resolution improvement over the diffraction limit. Here, the authors introduce a metamaterial structure to generate speckle-like sub-diffraction limit illumination patterns in the near field, and achieve a 7-fold resolution improvement down to 40 nm.

Abstract Genetic factors are one of the most important causes of non-obstructive azoospermia (NOA). ESX1 is an X-linked testis-biased expressed gene, and a potential biomarker for testicular sperm retrieval in NOA patients, yet few systematic studies have investigated its association with NOA. Here, we performed selected exonic sequencing in a large cohort of Chinese males, and four novel missense mutations (including one compound mutation), one novel synonymous mutation of ESX1 unique to NOA patients were identified. We analyzed the effects of ESX1 mutations on cyclin A degradation and cell cycle progression by immunoprecipitation assay and flow cytometry, and found that the compound mutant p.[P365R; L366V] ESX1 compromised the stabilizing effect of ESX1 on polyubiquitinated cyclin A, thereby causing the failure of M phase arrest in cells. Further studies showed that the deleterious effect of the compound mutations on ESX1 protein function was attributed to p.P365R but not p.L366V alteration. The novel ESX1 mutation p.P365R might confer high risk for NOA in Han Chinese population, probably via affecting cell cycle control.

De novo DNA methylation has been associated with T cell exhaustion in cancer immunotherapy. Here the authors show that the pre-treatment of CD19 CAR-T cells with the DNA methyltransferase inhibitor decitabine limits exhaustion and confers enhanced proliferative, effector and memory properties upon antigen exposure, with improved tumor control.

Abstract Human–robot collaboration is playing more and more important roles in current deployments of robotic systems in our lives. Haptic perception and intelligent control are essential to ensure safety and efficiency of human–robot interaction. However, existing robotic sensory and control systems are deficient in terms of performance issues, complexity, and cost. Here, the authors report a multifunctional electronic skin (e‐skin) incorporating multiple perceptions with intelligent robotic control, by which robots can safely and dexterously interact with humans. The e‐skin with a simple and cost‐effective sensory structure has multimodal perceptions of proximity, temperature, contact force, and contact position with broad measuring range, high sensitivity, and fast response. The e‐skin is applied onto robots to accomplish obstacle avoidance, safe and dexterous human–robot interaction, smart teaching, and playing Tai‐Chi, which demonstrate a broad range of applications for intelligent robots equipped with e‐skins.

Abstract Conventional antiplatelet agents indiscriminately inhibit both thrombosis and hemostasis, and the increased bleeding risk thus hampers their use at more aggressive dosages to achieve adequate effect. Blocking integrin αIIbβ3 outside‐in signaling by separating the β3/Src interaction, yet to be proven in vivo, may nonetheless resolve this dilemma. Identification of a specific druggable target for this strategy remains a fundamental challenge as Src SH3 is known to be responsible for binding to not only integrin β3 but also the proteins containing the PXXP motif. In vitro and in vivo mutational analyses show that the residues, especially E97, in the RT loop of Src SH3 are critical for interacting with β3. DCDBS84, a small molecule resulting from structure‐based virtual screening, is structurally validated to be directed toward the projected target. It specifically disrupts β3/Src interaction without affecting canonical PXXP binding and thus inhibits the outside‐in signaling‐regulated platelet functions. Treatment of mice with DCDBS84 causes a profound inhibition of thrombosis, equivalent to that induced by extremely high doses of αIIbβ3 antagonist, but does not compromise primary hemostasis. Specific targets are revealed for a preferential inhibition of thrombosis that may lead to new classes of potent antithrombotics without hemorrhagic side effects.

High nutritional value and the development of efficient biotechnological methods of controlled production have made black porgy (Acanthopagrus schlegelii) an economically important fish in Chinese aquaculture in recent years. However, aquaculture production of the species faces multiple issues associated with reduced growth rate, low reproduction ability, and high mortality during production, which are associated with the species’ limited tolerance to low temperatures. To date, comprehensive information on the genetic-based mechanisms of cold tolerance and adaptation to low temperature in the species are still unavailable. In this study, the HiSeq™2500 (Illumina) sequencing platform was used to analyze the transcriptomic profile of the liver tissue in the black porgy subjected to different extents of cold shock, including a control temperature group (AS, T = 15 °C), an intermediate temperature group (AL1, T = 10 °C), and an acute low-temperature stress group (AL2, T = 5 °C). For this purpose, three standardized cDNA libraries of AS, AL1, and AL2 were established. We obtained 43,258,908, 48,239,072, and 38,983,833 clean reads from the AS group, AL1 group, and AL2 group, respectively. After pairwise comparison, 70 differentially expressed genes (DEGs) were identified in the examined fish groups. Among them, 60 genes were found to be significantly differentially expressed after trend analysis. GO annotation and enrichment results showed that they were mainly enriched into three categories: biological processes (12 subcategories), molecular functions (7 subcategories), and cellular components (7 subcategories). KEGG analysis results indicated that all significantly differentially expressed genes were annotated to 102 signaling pathways, including biological rhythm, cholesterol metabolism, glycerolipid metabolism, animal autophagy, FoxO signaling pathway, steroid biosynthesis, and regulation of adipocyte lipolysis and apoptosis. Four of them, namely: G6PC, GPX1, GCK, and HSPE1 were randomly selected for further qRT-PCR verification of data reliability obtained by RNA-Seq technology. In this study, we found that environmental acute cold stress mainly affected the black porgy’s biological processes related to metabolism, apoptosis, and signal transduction. The data that we have reported provides baseline information for further studies concerning the genetic responses of the black porgy under cold stress conditions, the improvement of its aquaculture production, and other economically important matters regarding their limited tolerance to cold shock.

With the popularization of humans working in tandem with robots and artificial intelligence (AI) by Industry 5.0, ultrasonic non-destructive testing (NDT)) technology has been increasingly used in quality inspections in the industry. As a crucial part of handling ultrasonic testing results–signal processing, the current approach focuses on professional training to perform signal discrimination but automatic and intelligent signal optimization and estimation lack systematic research. Though the automated and intelligent framework for ultrasonic echo signal processing has already exhibited essential research significance for diagnosing defect locations, the real-time applicability of the algorithm for the time-of-flight (ToF) estimation is rarely considered, which is a very important indicator for intelligent detection. This paper conducts a systematic comparison among different ToF algorithms for the first time and presents the auto-diagnosis of the ToF approach based on the Defect Peaks Tracking Model (DPTM). The proposed DPTM is used for ultrasonic echo signal processing and recognition for the first time. The DPTM using the Hilbert transform was verified to locate the defect with the size of 2–10 mm, in which the wavelet denoising method was adopted. With the designed mechanical fixture through 3D printing technology on the pipeline to inspect defects, the difficulty of collecting sufficient data could be conquered. The maximum auto-diagnosis error could be reduced to 0.25% and 1.25% for steel plate and pipeline under constant pressure, respectively, which were much smaller than those with the DPTM adopting the cross-correlation. The real-time auto-diagnosis identification feature of DPTM has the potential to be combined with AI in future work, such as machine learning and deep learning, to achieve more intelligent approaches for industrial health inspection.

Satellite precipitation products have been applied to many research fields due to their high spatial and temporal resolution. However, satellite inversion of precipitation is indirect, and different inversion algorithms limit the accuracy of the measurement results, which leads to great uncertainty. Therefore, it is of great significance to quantify and record the error characteristics of different satellite precipitation products for their better application in hydrology and other research fields. In this study, based on CN05.1, which is a set of site–based interpolation data, we evaluated the accuracies of the six satellite precipitation datasets (IMERG–E, IMERG–L, IMERG–F, GSMaP, CMORPH, and PERSIANN–CDR) at different temporal scales (daily, monthly, and yearly) in mainland China for the period from 2001 to 2015. The results were as follows: (1) In terms of mean precipitation, IMERG–F was superior to other data in all areas. IMERG products and PERANN–CDR performed better than other products at all scales and were more suitable for precipitation research in mainland China. Site correction can effectively improve the accuracy of product inversion, so IMERG–F was significantly better than IMERG–E and IMERG–L. (2) Except PERSIANN–CDR, all precipitation products underestimated precipitation in the range of 1–4 mm/day and had a high coincidence with CN05.1 in the range of 4–128 mm/day. (3) The performance of six types of satellite precipitation products in summer was better than that in winter. However, the error was larger in seasons with more precipitation. (4) In the Qinghai–Tibet Plateau, where there are few stations, the inversion of precipitation by satellite products is closer to the actual situation, which is noteworthy. These results help users understand the characteristics of these products and improve algorithms for future algorithm developers.

Abstract Next‐generation inter‐chip communication requires ultrafast ultra‐compact interconnects. Designer plasmonics offers a possible route towards this goal. Further development of the plasmonic technique to circuit applications requires the direct amplification of plasmonic signals on a compact platform. However, significant signal distortions and limited operational speeds prevent the application of traditional MOS‐based amplifiers to plasmonics. Up to day, the amplification of surface plasmons without phase distortion has remained a scientific challenge. In this work, the concept of parametric amplification (PA) is transplanted to the plasmonics and is realized experimentally an ultrathin reconfigurable PA using a spoof surface plasmon polariton (SSPP) waveguide integrated with tunable and nonlinear varactors. The measured parametric gain in the experiment can reach up to 9.14 dB within a short nonlinear propagation length, for example, six SSPP wavelengths, in excellent agreement with the theoretical prediction. By tuning the bias voltage of varactors, the phase‐matching condition can be precisely controlled over a broad frequency band, enabling the authors to realize the multi‐frequency PA of plasmonic signals. Measured phase responses confirm that the plasmonic parametric amplifier can significantly suppress the signal distortions as compared with the traditional MOS‐based amplifier, which is a property highly desired for ultrafast wireless communication systems and integrated circuits.

Summary: Aryl–heteroatom (C–X) bonds ubiquitously exist in organic, medicinal, and material chemistry, but a universal method to construct diverse C–X bonds is lacking. Here we report our discovery of a convenient and efficient approach to construct various C–X bonds using arylammonium salts as the substrate via an SNAr process. This strategy features mild reaction condition, no request of transition metal catalyst, and easy formation of various C–X bonds (C–S, C–Si, C–Sn, C–Ge, C–Se, C–N). Abstract Recently, programmable metamaterials or metasurfaces have been developed to dynamically edit electromagnetic waves for realizing different functions in the same platform. However, the proposed programmable metasurfaces can only control reflected or transmitted wavefronts in half‐space. Here, a “Janus” digital coding metasurface with the capabilities to program various electromagnetic functions in the reflected (with R‐codes) and transmitted (with T‐codes) waves simultaneously is presented. Three PIN diodes are employed to design the metaparticle, and the state of the PIN diodes can be switched to change the reflected and transmitted phases independently. Three schemes achieved by the proposed programmable metasurface are provided as illustrative examples, including anomalous deflections, beam focusing, and scattering reduction in the full space. As a proof‐of‐concept, a prototype composed of 10 × 20 metaparticles is fabricated and the measured results are in good agreement with the designs and numerical results, validating the full‐space modulations enabled by the programmable metasurface. It is expected that the new programmable metasurface can broaden the applications in stealth technologies, imaging systems, and the next generation of wireless communications.

Cardiac hypertrophy is a complex process induced by the activation of multiple signaling pathways. We previously reported that anacardic acid (AA), a histone acetyltransferase (HAT) inhibitor, attenuates phenylephrine (PE)-induced cardiac hypertrophy by downregulating histone H3 acetylation at lysine 9 (H3K9ac). Unfortunately, the related upstream signaling events remained unknown. The mitogen-activated protein kinase (MAPK) pathway is an important regulator of cardiac hypertrophy. In this study, we explored the role of JNK/MAPK signaling pathway in cardiac hypertrophy induced by PE. The mice cardiomyocyte hypertrophy model was successfully established by treating cells with PE in vitro. This study showed that p-JNK directly interacts with HATs (P300 and P300/CBP-associated factor, PCAF) and alters H3K9ac. In addition, both the JNK inhibitor SP600125 and the HAT inhibitor AA attenuated p-JNK overexpression and H3K9ac hyperacetylation by inhibiting P300 and PCAF during PE-induced cardiomyocyte hypertrophy. Moreover, we demonstrated that both SP600125 and AA attenuate the overexpression of cardiac hypertrophy-related genes (MEF2A, ANP, BNP, and β-MHC), preventing cardiomyocyte hypertrophy and dysfunction. These results revealed a novel mechanism through which AA might protect mice from PE-induced cardiomyocyte hypertrophy. In particular, AA inhibits the effects of JNK signaling on HATs-mediated histone acetylation, and could therefore be used to prevent and treat pathological cardiac hypertrophy.
