Background: Snakebite envenomation (SBE) and scorpion sting envenomation (SSE) are significant neglected tropical diseases that primarily affect impoverished communities in rural areas of developing nations. A lack of understanding about snake and scorpion species and their distribution exacerbates the disabilities and fatalities caused by SBE and SSE. In Sudan, particularly in regions affected by ongoing conflicts where healthcare resources are scarce, social media platforms offer a cost-effective approach to addressing public health challenges. Our aim in this study is to highlight the benefits of using social media for data collection and health promotion in such environments. Methods: We present a cost-effective communication and data collection strategy implemented at the Toxic Organisms Research Centre (TORC) of the University of Khartoum, focusing on a Facebook group, "Scorpions and Snakes of Sudan", as our primary social media platform. Additionally, we discuss the lessons learned and the initial impact of this strategy on enhancing population health literacy. Results: The group community is composed of ~ 5000 members from 14 countries. During the period from January 2023 to January 2024, we received 417 enquiries about snakes and scorpions belonging to 11 families and composed of 55 species. In addition, 53 other enquiries covered a range of organisms and their tracks (e.g., spiders, skinks, chameleons, foxes, sun spiders, centipedes, lizards, moth larvae, and insect tracks). The first photographic evidence of Malpolon monspessulanus in Sudan was via the group activities. The rare species Telescopus gezirae, the Blue Nile cat snake, is also documented via the group member's queries. Recognizing the evolving nature of social media use in public health, we also address the current limitations and evidence gaps that need to be addressed to effectively translate best practices into policy. Conclusion: In conclusion, utilizing Facebook as an institutional platform to share scientific information in simple Arabic language underscores the proactive roles that citizens, scientists, and public health stakeholders can play in leveraging social media for eHealth, eAwareness, and public health initiatives. This approach highlights the potential for collaborative efforts, particularly during crises, to maximize the benefits of social media in advancing public health. Due to the high reproductive investment, females are the decisive sex when making decisions about whether or not to reproduce. Generally, female scorpions display aggressive sexual behavior in response to male reproductive advances when they are not receptive. However, female receptivity can vary over time, depending on environmental factors favorable to reproduction Thus, the current study aimed to examine the correlation between the number of reproductive attempts by male Tityus pusillus scorpion and mating success over time, considering the female's behavioral response. Behavioral analyses were monthly conducted on 150 T. pusillus field-caught pairs over a five-month period. Our results indicate that despite females reacting aggressively to male advances, the persistence of the male has a positive effect in courtship probability. Furthermore, we observed variations in female behavior throughout the studied period, suggesting that female receptivity might be linked to the reproductive season. Background: Scorpion envenomation, a prevalent medical emergency in rural areas, demands immediate attention due to its potential severity. While ocular manifestations are uncommon, they can lead to significant complications such as corneal ulceration. We present a unique case of corneal ulceration subsequent to a yellow scorpion (Hemiscorpius lepturus) sting near the eye, a scenario not previously documented. Case presentation: A 34-year-old male sought medical care following a scorpion sting despite prior anti-venom treatment. Clinical examination revealed pronounced ocular inflammation, corneal stromal melting, and anterior chamber inflammation, with microbiological confirmation of Pseudomonas spp infection. Treatment comprised fortified ceftazidime and vancomycin eye drops, alongside topical corticosteroids, leading to visual and corneal healing. Conclusion: This case highlights the urgency of addressing scorpion envenomation and its potential for severe ocular complications, including corneal ulceration. Prompt diagnosis and targeted therapy with antibiotics and corticosteroids are crucial for favorable outcomes. Injury by scorpion sting envenoming is a serious health problem, especially in rural areas. This paper offers an inventory of the plants used for treating scorpion sting in Algerian Septentrional Sahara. An ethnobotanical survey was undertaken in El Oued, Ghardaïa and Ouargla. The interviews, using semi-structured interviews with 360 people of different ethnic groups, were analysed with quantitative tools: Relative frequency of citation (RFC), Use value (UV), Informant consensus factor (Fic), Jaccard index (JI) and Chi2 test. Results show that the local populations use 53 plants belonging to 34 families for the scorpion sting treatment; Asteraceae and Lamiaceae are dominant. The most used part of the plants was leaves (23 species) and the most used method of preparation was the paste (23.26%). The highest RFC and Fic were recorded for Retama raetam (RFC = 0.46; Fic = 0.69). Among 101 remedy recipes, 69.31% of recipes were used externally in the scorpion envenomation treatment. Insect‐specific neurotoxic peptides derived from the venoms of scorpions and spiders can cause acute paralysis and death when injected into insects, offering a promising insecticidal component for insect pest control. However, effective delivery systems are required to help neurotoxic peptides pass through the gut barrier into the hemolymph, where they can act. Here, we investigated the potential of a novel nanocarrier, Drosophila X virus‐like particle (DXV‐VLP), for delivering a neurotoxin from the scorpion Androctonus australis Hector (AaIT) against the invasive pest fruit fly, Drosophila suzukii. Our results show that the fusion proteins of DXV polyproteins with AaIT peptide at their C‐termini could be sufficiently produced in Lepidoptera Hi5 cells in a soluble form using the recombinant baculovirus expression system, and could self‐assemble into VLPs with similar particle morphology and size to authentic DXV virions. In addition, the AaIT peptides displayed on DXV‐VLPs retained their toxicity, as demonstrated in injection bioassays that resulted in severe mortality (72%) in adults after 72 h. When fed to adults, mild mortality was observed in the group treated with DXV‐AaIT (38%), while no mortality occurred in the group treated with AaIT peptide, thus indicating the significant role of DXV‐VLPs in delivering AaIT peptides. Overall, this proof‐of‐concept study demonstrates for the first time that VLPs can be exploited to enhance oral delivery of insect‐specific neurotoxic peptides in the context of pest control. Scorpion stings are the major public health problem related to arthropods in tropical and subtropical areas of Iran. The study aimed to determine the species diversity and geographical distribution of scorpions in southern regions of Kerman province, Iran. The descriptive cross-sectional study was carried out from March 2019 to November 2021 in the south of Kerman province. The specimens were collected by rock rolling method using long forceps during the day and UV light during the night. The specimens were put in containers containing 80% ethanol, transported to the laboratory, identified using valid taxonomic keys, and studied their sexes. A total of 126 scorpions were collected from 2 families, 7 genera, and 10 species including Hemiscorpius acanthocercus (2.2%), H. enischnochela (0.4%), Androctonus crassicauda (56.1%), Mesobuthus kirmanensis (29%), Odontobuthus baluchicus (3.5%), Hottentotta navidpouri (2.2%), Ho. sistanensis (2.2%), Orthochirus hormozganensis (1.3%), O. kucerai (0.9%) and Sassanidotus gracilis (1.3%). Mesobuthus kirmanensis were reported as dominant species. H. navidpouri, H. sistanensis, O. baluchicus, and O. hormozganensis are reported for the first time from Kerman province. Androctonus crassicauda, H. acanthocercus and H. enischnochela are known as dangerous species and could enhance the potential risk of scorpion stings deaths, especially in children. The fauna and geographical distribution of scorpions in the southeast of Iran showed the presence of suitable habitats for scorpions and can be a warning to the health care system. Urophonius trewanke sp. nov. is described from the Mauline Andean woods of northern Chilean Patagonia. This species belongs to the granulatus species group, which includes the most basal species within the genus. This species is only active in summer as in all species of its group. Over 400 million years old, scorpions represent an ancient group of arachnids and one of the first animals to adapt to life on land. Presently, the lack of available genomes within scorpions hinders research on their evolution. This study leverages ultralong nanopore sequencing and Pore-C to generate the first chromosome-level assembly and annotation for the desert hairy scorpion, Hadrurus arizonensis. The assembled genome is 2.23 Gb in size with an N50 of 280 Mb. Pore-C scaffolding reoriented 99.6% of bases into nine chromosomes and BUSCO identified 998 (98.6%) complete arthropod single copy orthologs. Repetitive elements represent 54.69% of the assembled bases, including 872,874 (29.39%) LINE elements. A total of 18,996 protein-coding genes and 75,256 transcripts were predicted, and extracted protein sequences yielded a BUSCO score of 97.2%. This is the first genome assembled and annotated within the family Hadruridae, representing a crucial resource for closing gaps in genomic knowledge of scorpions, resolving arachnid phylogeny, and advancing studies in comparative and functional genomics. Chelicerae, distinctive feeding appendages in chelicerates, such as spiders, scorpions, or horseshoe crabs, can be classified based on their orientation relative to the body axis simplified as either orthognathous (parallel) or labidognathous (inclined), exhibiting considerable diversity across various taxa. Among extinct chelicerates, sea scorpions belonging to the Pterygotidae represent the only chelicerates possessing markedly elongated chelicerae relative to body length. Despite various hypotheses regarding the potential ecological functions and feeding movements of these structures, no comprehensive 3D kinematic investigation has been conducted yet to test these ideas. In this study, we generated a comprehensive 3D model of the pterygotid Acutiramus, making the elongated right chelicera movable by equipping it with virtual joint axes for conducting Range of Motion analyses. Due to the absence in the fossil record of a clear indication of the chelicerae orientation and their potential lateral or ventral movements (vertical or horizontal insertion of joint axis 1), we explored the Range of Motion analyses under four distinct kinematic settings with two orientation modes (euthygnathous, klinogathous) analogous to the terminology of the terrestrial relatives. The most plausible kinematic setting involved euthygnathous chelicerae being folded ventrally over a horizontal joint axis. This configuration positioned the chelicera closest to the oral opening. Concerning the maximum excursion angle, our analysis revealed that the chela could open up to 70°, while it could be retracted against the basal element to a maximum of 145°. The maximum excursion in the proximal joint varied between 55° and 120° based on the insertion and orientation. Our findings underscore the utility of applying 3D kinematics to fossilized arthropods for addressing inquiries on functional ecology such as prey capture and handling, enabling insights into their possible behavioral patterns. Scorpion envenomation poses a global public health issue, with an estimated 1,500,000 cases worldwide annually resulting in 2600 deaths. North Africa, particularly Morocco, experiences severe envenomations, mainly attributed to Androctonus mauretanicus and Buthus occitanus in Morocco, and Buthus occitanus and Androctonus australis hector in Algeria and Tunisia, with case numbers often underestimated. Current treatment relies mainly on symptomatic approaches, except in Morocco, where management is limited to symptomatic treatment due to controversies regarding specific treatment. In Morocco, between 30,000 and 50,000 scorpion envenomation cases are reported annually, leading to hundreds of deaths, mainly among children. Controversies among clinicians persist regarding the appropriate course of action, often limiting treatments to symptomatic measures. The absence of a specific antivenom for the venoms of the most lethal scorpions further exacerbates the situation. This study aims to address this gap by developing a monovalent antivenom against the endemic and most dangerous scorpion, Androctonus mauretanicus. The antivenom was produced by immunizing albino rabbits with a mixture of Androctonus mauretanicus venom collected from high-risk areas in Morocco. Immunizations were performed by subcutaneous injections at multiple sites near the lymphatic system, following an immunization schedule. Production control of neutralizing antibody titers was conducted through immunodiffusion. Once a sufficient antibody titer was achieved, blood collection was performed, and the recovered plasma underwent affinity chromatography. The efficacy of purified IgG was evaluated by determining the ED50 in mice, complemented by histological and immunohistochemical studies on its ability to neutralize venom-induced tissue alterations and the neutralization of toxins bound to receptors in the studied organs. The monovalent antivenom demonstrated specificity against Androctonus mauretanicus venom and effective cross-protection against the venom of the scorpions Buthus occitanus and Androctonus australis hector, highly implicated in lethal envenomations in the Maghreb. This study shows that the developed monovalent antivenom exhibits notable efficacy against local scorpions and a surprising ability to neutralize the most lethal envenomations in North Africa. These results pave the way for a new, more specific, and promising therapeutic approach to countering severe scorpion envenomations, especially in Morocco, where specific treatment is lacking. Failure of sub-arachnoid block (SAB), due to resistance to bupivacaine after a recent scorpion sting can lead to multiple block attempts and subsequent conversion to general anaesthesia. We report this case series of 10 patients with successful SAB with newly launched 0.75% hyperbaric ropivacaine, in patients with recent scorpion sting. Thus, intrathecal hyperbaric ropivacaine may be considered as the local anaesthetic agent of choice in patients with scorpion sting to prevent failure of SAB. [ABSTRACT FROM AUTHOR]

New locality records for Mesobuthus mesopotamicus (Penther, 1912) are given from Iraq. The populations from southeastern Turkey, earlier identified as M. mesopotamicus by Kovařík et al. (2022) are examined and compared with the specimens from type locality of M. mesopotamicus. These populations are described as a new species, Mesobuthus faiki sp. n. Barbaracurus hofereki sp. n. from Djibouti is described and compared with other species of the genus. B. hofereki sp. n. is the only species of the genus with pedipalp movable finger with 8 rows of granules and pectinal tooth count 27-28 in female. Also, its pedipalp chela with very narrow manus (chela length/width ratio 6.07 in female), narrower than in all other African species of the genus. BeKm‐1 is a peptide toxin from scorpion venom that blocks the pore of the potassium channel hERG (Kv11.1) in the human heart. Although individual protein structures have been resolved, the structure of the complex between hERG and BeKm‐1 is unknown. Here, we used molecular dynamics and ensemble docking, guided by previous double‐mutant cycle analysis data, to obtain an in silico model of the hERG–BeKm‐1 complex. Adding to the previous mutagenesis study of BeKm‐1, our model uncovers the key role of residue Arg20, which forms three interactions (a salt bridge and hydrogen bonds) with the channel vestibule simultaneously. Replacement of this residue even by lysine weakens the interactions significantly. In accordance, the recombinantly produced BeKm‐1R20K mutant exhibited dramatically decreased activity on hERG. The troglomorphic scorpion genus Troglotayosicus Lourenço, 1981, occurs in hypogean and epigean habitats in the Andean and Amazonian rainforests of Colombia and Ecuador. The phylogenetic relationships among the species of Troglotayosicus are currently unknown. In the present contribution, a new species, Troglotayosicus akaido, sp. nov., is described from specimens collected in the leaf litter of a primary rainforest in the Colombian Amazon, near the border with Peru, raising the number of species in the genus to seven. The new species represents the easternmost record of the genus and further extends its distribution into the Amazon. Its phylogenetic position was tested in an analysis of all species of the genus and two outgroup taxa, scored for 131 morphological characters (16 new and 115 legacy; 104 binary and 27 multistate) analyzed with maximum likelihood under the MK model. Troglotayosicus was recovered as monophyletic and composed of two main clades. The morphological survey revealed that the ventral macrosetae of the leg telotarsi of the type species, Troglotayosicus vachoni Lourenço, 1981, are simple, subspiniform macrosetae, irregularly distributed, but not arranged into clusters nor forming elongated clusters of setae/spinules, as previously suggested. A distribution map and key to the identification of the species of Troglotayosicus are provided. Two new monotypic buthid genera are described: Sanaag gen. n. and Sahil gen. n. from Somaliland, both belonging to the 'Buthus' group. Sanaag gen. n. (type species Gint maidensis Kovařík et al., 2018) differs from Gint Kovařík et al., 2013 in the structure of its hemispermatophore, which has a large, tall, subtriangular hook-like basal lobe, and in the shape of its telson which is rather bulbous. Sahil gen. n. (type species Sahil elmii sp. n.) differs from all morphologically similar small-sized genera of the Horn of Africa in having the ventral aspect of cheliceral fixed finger with two denticles, and the movable finger of the pedipalp with 7 subrows of denticles. This study investigates the use of a saltwater (sodium chloride and water) solution as a phase change material (PCM) in a small fridge for storing scorpion antivenom in Sudan's northern state. The experimental results demonstrate the effectiveness of the PCM in maintaining suitable storage temperature which 3 ± 2°C. By adding salt to the water, the freezing point decreased, enabling the solution to absorb more heat energy because the heat capacity of the saltwater solution is higher than the saltwater ice phase. The base case maintained (2310 g of water only) an average temperature of 9.5°C, while Case 1 (35 g of salt in 2310 g of water) reached 7°C and Case 2 (70 g of salt in 2310 g of water) achieved 5°C. Increasing the amount of saltwater solution led to improved heat absorption. Case 5 (70 g of salt in 4950 g of water) achieved an optimal storage temperature of 4°C. The relationship between outside and inside temperatures showed stable maintenance after initial melting, with a slight increase of approximately 1.5°C. These findings demonstrate the potential of solar-powered fridge with PCM for reliable medication storage in remote areas. Scorpions (Arachnida: Scorpions) are one of the medically important oldest arthropods. They have been one of the humans' interests in terms of special morphology, painful stings, mortality, and behavioral attractions for many years. Diagnosis of scorpions is based on morphological characteristics and molecular methods. But, one of the most important and fundamental problems of identifying the Iranian scorpions is the lack of a comprehensive method. Due to the fact that a relatively long time has not passed since the classification of scorpions in Iran and there are no definitive divisions about the genera and species in the country, the current pictorial key was provided to identify the Iranian scorpions. In this study, various methods were used to catch the scorpions. For preparing the pictorial key, different genera of scorpions were collected from urban, rural, plain, and mountain areas in different parts of Iran as well as some archived specimens from the medical arachnid's Museum of Shiraz University of Medical Sciences were used from 2010 to 2020 collection boxes. The key presented in this study was prepared only based on morphological features. It was necessary to remind a few points about this key and its use. Considering the above-mentioned points, this key was provided for 16 genera, 64 species, and 3 families of Iranian scorpions, and the results of other researchers are included in it. The study of various aspects of scorpion life, especially faunistic studies has been recently considered in Iran. Various methods such as molecular methods and composition of scorpion venom can be used for classification, the most common of which are morphological features that have a long history. The overall assumption that scorpions are generalist predators is often based on conspicuous information from the literature. Here, we compiled a list of prey consumption by scorpions from different environments in Brazil to produce a documentation of predations by this taxon. This list is based on observations made under natural conditions in Atlantic Forest, Caatinga, and Cerrado formations. We compiled 135 predation instances including 11 scorpion species from field work through 14 years. The observed diet composition of the scorpions was mainly based on cockroaches, crickets, grasshoppers, spiders, and other scorpions. Such data highlights the generalist diet and cannibalism of scorpions with many cannibalistic events among the records of intraguild predation. Scorpions possess midventral touch/taste organs called pectines, which may be important for learning the nuances of the substrate during navigation as well as the detection of pheromones, spermatophores, and food. The pectines possess thousands of minute structures called peg sensilla that are responsive to both chemicals and mechanical deflection of the peg shaft. While much is known about the chemical responsiveness of the pegs, very little is known about their mechanosensory properties. Here we ask if the peg mechanosensory response is "all-or-nothing" or graded depending on the intensity of stimulation. We made electrophysiological recordings of neural activity from individual peg sensilla while deflecting the peg to elicit apparent mechanosensory responses. Our records show the presence of a rapid firing (.100 Hz), quickly adapting waveform that is indicative of a mechanoreceptor and appears to be independent of previously identified chemo-responsive cells. We tested mechanosensory response dynamics in two ways. The first test focused on a shorter-duration touch versus a longer-duration touch, while the second focused on a smaller deflection versus a larger deflection. Both pairs of stimulations (short vs long touch; small vs large touch) produced repeatable and statistically distinct responses in terms of spiking frequency. These results indicate the mechanosensory responses of peg sensilla are graded, which sheds light on the textural resolvability of the pectines and informs models of the type of information that scorpions obtain while assessing surfaces in their environment. A new record of an adult female of Parabuthus eritreaensis Kovařík, 2003 in Somaliland confirms true distribution of this species, already discussed in Kovařík et al. (2016: 19-21). Three new species are described from Somaliland, P. dorisae sp. n., P. evae sp. n., and P. quincyae sp. n.. The hemispermatophore of P. dorisae sp. n. is illustrated and described. In addition to the analyses of external morphology and hemispermatophore, we have provided descriptions of the karyotypes of P. dorisae sp. n. and P. quincyae sp. n. Despite the presence of multivalents (CVIII and CXIV), both species exhibit karyotypes with 2n=16 and chromosomes that gradually decrease in length, with the exception of the first chromosome, which is longer than the following chromosomes. This study reports, for the first time, scorpions belonging to the genus Centruroides Marx 1890 in Tlaxcala, Mexico. Three specimens of C. limpidus (Karsch 1879) were collected in urban areas of the state. The scorpion fauna of Tlaxcala remains understudied, possibly due to the low prevalence of scorpionism. The presence of C. limpidus indicates a potential public health concern in the state. Este estudio se reporta por primera vez escorpiones pertenecientes al género Centruroides Marx 1890 en Tlaxcala, México. Se colectaron tres especímenes de C. limpidus (Karsch 1879) en zonas urbanas del estado. La fauna de escorpiones de Tlaxcala permanece poco estudiada, posiblemente debido a la baja prevalencia del escorpionismo. Patterns of descriptions of new species of Mexican scorpions between 2000-2023 were analyzed. We based our study on the Zoological Record of Web of ScienceTM. We found 111 species in seven families described in 83 papers published in scientific journals from all over the world. Families with the most species described were Vaejovidae (52), Buthidae (26), Diplocentridae (23), Euscorpiidae (4), and Typhlochactas (3). Palaeozoic fossils of whip scorpions (Arachnida: Uropygi: Thelyphonida) are extremely rare, with only seven species of this age previously described. A new species of fossil whip scorpion, as well as the first ichnospecies assignable to this group, are described here from the Carboniferous Narragansett Basin of Massachusetts, USA. A body fossil from the Rhode Island Formation (Moscovian) is referred to as Parilisthelyphonus bryantae gen. nov. sp. nov.. At more than 34 mm long it represents both the largest known Palaeozoic whip scorpion and the first fossil arachnid found in the Rhode Island Formation in ~130 years of scrutiny. The whip scorpion trace fossil, comprising a full-body impression and associated tracks, is described from the subjacent Wamsutta Formation (Late Bashkirian) of Massachusetts, USA as I nmontibusichnus charleshenryturneri igen. nov. isp. nov.. With an estimated body length of less than 10 mm, the producer would be the smallest known Palaeozoic thelyphonid. These discoveries within the Narragansett Basin represent only the second site in the western hemisphere, in what was western Laurasia, to yield Palaeozoic whip scorpions. The Narragansett Basin is of significant Pangaean biogeographical importance among whip scorpion sites, being located between the westernmost Mazon Creek Lagerstätte of Illinois and the eastern assemblage of localities in Europe, and serves as an important new fossil calibration point for phylogenetic studies of this arachnid group. Scorpionism is a public health problem, especially in tropical regions. In Brazil, the prevalence of envenomation by scorpions is high, and the average national lethality is around 0.16 percent. The Tityus serrulatus scorpion is the primary species of medical importance. However, objective tools to predict and define the severity of these envenomations are lacking. This was an observational study conducted among patients aged 0–19 years with scorpionism. Patients were admitted to a reference hospital between December 2020 and May 2022. Point-of-care ultrasound was performed within 24 hours of the scorpion sting. Forty-nine patients were included, with a median age of 3.6 (interquartile range 2.3–5.3) years and a predominance of females (51 percent). Fifteen patients (30.6 percent) presented major life-threatening signs, 32 (65.3 percent) minor systemic manifestations, and two (4.1 percent) only local manifestations. Left ventricular dysfunction was identified in 13 patients (26.5 percent). Ten patients (20.4 percent) presented pattern B (visualization of three or more B lines in the evaluated quadrant) in at least one lung window. The sensitivity and specificity of cardiac and pulmonary ultrasound to identify the most severely ill patients were 86 percent and 94 percent, respectively. The changes found on point-of-care ultrasound were associated with life-threatening signs. All patients with class III envenomation were referred to the intensive care unit, showing the importance of early identification of this subgroup. The main limitations were the small sample size and the fact that admission to intensive care was not based on systematic criteria. Point-of-care ultrasound is able to identify early signs of pulmonary congestion and heart failure in scorpionism. It can be useful for the objective selection of patients who are at a higher risk of complications and death and who require intensive support; it may also be valuable for periodic reassessments. Scorpion venoms are known to contain over 100,000 biologically active constituents. However, only a few of them have been studied. The major constituents of venom are proteins and peptides, which exhibit various biological and pharmacological properties, including anticancer activities. In the current study, the venom of yellow scorpions (Buthus sindicus) found in Sindh, Pakistan, was extracted and evaluated for its anti-cancer and anti-inflammatory activities. The crude venom showed a dose dependent inhibition of phagocyte oxidative burst from human whole blood cells (28.3% inhibition at highest tested concentration of 300 μg/mL). In-vitro cytotoxicity of crude venom was evaluated against human prostrate (PC3), cervical (HeLa) and neuroblastoma (U87-MG) cell lines, along with cytotoxicity against normal human fibroblast (BJ) cells. Crude venom was cytotoxic to all cell lines, with prominent inhibitory effect on PC3 cells. Crude venom was fractionated through RP-UPLC, resulted in fifteen fractions, followed by evaluation of their anticancer potential. Among all, the fraction I significantly (P < 0.001) reduced the cell viability of all three cancer cell lines, and exhibited insignificant cytotoxicity against normal cell line. Furthermore, the apoptotic cell death pathway was evaluated for crude venom, and fraction I, in most sensitive cell line PC3, by using flow-cytometry analysis. Both crude venom and its fraction I caused a mitochondrial-mediated apoptosis in prostate cancer cells (PC3). To the best of our knowledge, this is the first report of the anticancer and anti-inflammatory activity of venom of Pakistani yellow scorpions. A new species, Scorpiops (Euscorpiops) krachan sp. nov., belonging to the family Scorpiopidae Kraepelin, 1905 is described based on three adult males and one adult female collected in the Kaeng Krachan National Park, Phetchaburi Province, Thailand. The new species presents most features exhibited by scorpions of the subgenus Euscorpiops and can be characterized notably by a very small size, a sexual dimorphism strongly marked with male pedipalps elongated, a distinct trichobothrial pattern and other morphological features. This new taxon may represent one endemic element for the scorpion fauna of Thailand. The genus Olivierus Farzanpay, 1987 in Xinjiang Uygur Autonomous Region, China, is revised based on recently collected topotypes and other populations from 12 localities. Brief differential diagnoses are provided, with colored illustrations and photos in vivo habitus, emphasizing the key characters. Chinese appellations, conservation status, and documentation of behavior and post-envenomation symptoms are also included. Only two species are now recognized for this genus in Xinjiang: O. longichelus (Sun & Zhu, 2010) and O. przewalskii (Birula, 1897), based on both morphological and molecular evidence. The two species exhibit extensive distribution in Xinjiang (China) while also occurring in adjacent countries. Three new synonyms are proposed: Mesobuthus bolensis Sun et al., 2010 = Olivierus longichelus (Sun & Zhu, 2010), syn. n.; Mesobuthus karshius Sun & Sun, 2011 = Olivierus longichelus (Sun & Zhu, 2010), syn. n.; Olivierus tarabaevi Fet et al., 2021 = Olivierus longichelus (Sun & Zhu, 2010), syn. n. Two species, Olivierus extremus (Werner, 1936) and O. hainanensis (Birula, 1904), are likely synonymous with O. martensii (Karsch, 1879). Urban landscapes restrain the distribution of forest‐dwelling species, which may be related to challenging conditions that impair body condition. The dynamics in urban areas lead to the simplification of communities that inhabit forest patches in cities with the turnover of sensitive species for opportunistic ones. In this study, we investigated the effect of urbanization on the body condition and diversity of scorpions at the landscape scale. Sampling was carried out in 10 forest patches in an urban matrix in Brazil, originally covered by a tropical rainforest. The surroundings of the landscape of each forest patch were characterized through the amount of forest, agriculture, and urban land cover. Individual body length, dry, lipid, and muscular masses were used as proxies of Tityus pusillus body condition. In total, 147 scorpions were collected, belonging to the species Ananteris mauryi, T. pusillus, T. stigmurus, and T. neglectus. Forest cover explained 28% of species variation. There was a positive relationship between forest cover and T. pusillus and A. mauryi abundances, while T. stigmurus was negatively affected by forest cover. Species richness and total scorpion abundance were not influenced by landscape metrics. In terms of body condition, only females of T. pusillus were affected by landscape variables, with individuals showing higher body mass with an increase in forest cover. Our results suggest that urban forests can support scorpion assemblages. However, there is a turnover in specialist forest species for opportunistic species. Scorpion sting envenomation, despite its high incidence and prevalence in marginalized areas, is not recognized as a neglected tropical disease (NTD) by the World Health Organization (WHO). However, scorpion sting envenomation meets all the criteria to be recognized as an NTD. Globally, there are more than 1.2 million reported cases and 3250 deaths each year, with Mexico having the highest number of cases. Scorpion sting envenomation has devastating health, social, and economic consequences, and an official designation as an NTD by the WHO could lead to improved information systems, surveillance, and intervention strategies. A new case of pedipalp regeneration is described and illustrated in a subadult female of Scorpio kruglovi Birula, 1910. A systematic understanding of the variation in species composition across time and space is urgently needed to protect threatened Brazilian forests.This study has disentangled the among‐site variation in beta diversity among scorpion assemblages from the Caatinga and Atlantic Forest. We calculated the relative contribution of species replacement and differences in species richness in explaining the beta diversity in each forest, correlating such metrics with environmental characteristics. Finally, we estimated the relative site‐specific contribution to identify potential environmental drivers determining unique combinations of species composition.The average among‐site dissimilarity in scorpion assemblages was higher in the Caatinga than that in the Atlantic Forest. The differences in species composition among Caatinga sites were mainly due to the spatial replacement of species. Differences in species richness at the local scale explained most of the variation in species composition among Atlantic Forest sites.Environmental variables related to topography and land use partially explained the variation in among‐site dissimilarity, species replacement, differences in species richness and the ecological uniqueness in terms of species composition in the Caatinga. Environmental variables describing topography and land use were also important in explaining differences in species richness and local contributions to the beta diversity among Atlantic Forest sites. The relative site‐specific contribution to beta diversity was higher in Caatinga sites located in ecotonal zones and Atlantic Forest sites situated within conservation reserves.Using scorpions as biological models, we have demonstrated that the species composition does not vary in a consistent way between different forest types. Sensilla on the dentate margins of scorpion pedipalp fingers, associated with denticle subrows or terminal denticles, were surveyed by white light, UV fluorescence and scanning electron microscopy. A total of 163 species belonging to 103 genera from 19 families were examined, representing all major phylogenetic lineages in the order. In the majority of species (141/152) with dentition organized into linear subrows of median denticles, a small socketed sensillum with a short shaft was observed at the proximal terminus of each subrow, termed a 'subrow proximal sensilla' (SPS). With few exceptions, SPS were non-fluorescent ('Type N') in buthids, and fluorescent ('Type F') in non-buthids. SPS were present in buthids, chactoid families, and vaejovids; they were absent in Pseudochactas, bothriurids and scorpionoid families. Subterminal sensilla (STS) were found proximal to terminal denticles. Buthid fingers typically had two pairs of Type N STS, a proximal filiform pair and a distal spatulate pair, herein termed 'Cruz-Armas sensilla' after their discoverers; in a minority of species, the spatulate pair was missing. Non-buthid fingers typically had a single Type F STS, similar to Type F SPS. Type N sensilla studied here may be mechanoreceptive 'macrosetae', and Type F sensilla chemo-/mechanoreceptive 'microsetae', both specialized for detecting close-contact stimuli. In order to correctly interpret fluorescence images of sensilla, we reviewed the optics of UV excitation and fluorescence emission in scorpion cuticle. We propose a dielectric waveguide model of the cuticle, in which fluorescence emitted in the hyaline exocuticle is trapped by total internal reflection, and is able to propagate tangential to the surface. We suggest that this phenomenon could enhance topographic contrast of small surface features in fluorescence imaging, and increase the efficiency of UV fluorescence detection by ocular or extraocular photoreceptors. We observed highly effective block of UV light by the cuticle, and strong UV attenuation by non-fluorescent endocuticle. In North Africa, scorpion stings pose an urgent public health problem, particularly for children with high morbidity and mortality rates. The main species implicated are the Androctonus mauretanicus (Am), Androctonus australis hector (Aah), and Buthus occitanus (Bo). Immunotherapy is the specific therapeutic approach aimed at directly neutralizing toxins, despite their severity and rapid diffusion. In the present study, we evaluate, histologically and immunohistologically, the neutralization potency of the selective antivenom produced against, among other species, the Am, Aah, and Bo at the level of the tissue alterations in Swiss mice, as experimental subjects. Firstly, the lethal doses 50 test was conducted to assess the venom's toxic activity, and then the median effective dose of the antivenom was determined against each venom. The histological and immunohistological analyses were performed by injecting the sublethal dose of venom, the complex venom and antivenom, or the antivenom 2 h following inoculation of venom. Our study revealed the highest toxicity of the Am, followed by the Aah and then the Bo venom. The neutralizing ability and effectiveness of the antivenom to completely or partially neutralize the tissular damages were demonstrated in all organs studied: brain, heart, lungs, liver, and kidneys. Our results highlighted the important cytoplasmic and membranous staining in the heart compared to the brain tissue for the three scorpion venoms. Therefore, the scorpionic antivenoms are able to reach their target even at the tissue level. The data on the taxonomy and distribution of scorpions and solifuges with the aim of evaluating the current state of their fauna in eastern Transcaucasia are reported. The taxonomic list of species was compiled using the materials obtained from the available literature sources, the data of faunistic studies, and the results of studying the collection of the Azerbaijan Institute of Zoology, Baku, and the author's own collections. Updated information on the taxonomy and distribution of scorpions and solifuges in eastern Transcaucasia is presented, with two new species of scorpions and three species of solifuges having been identified. To summarize, according to the 2022 data, the Scorpiones order in the studied region is represented by five species in three genera in a single family, while the Solifugae order, by 14 species in seven genera in five families. Scorpion venoms contain bioactive peptides and proteins. Some, can be used for pharmaceutical purposes. So, identification of venom proteins matters because, in addition to determining the function of the toxins can also be an excellent guide to developing new drugs. Here, we got transcriptome of venom glands from four Iranian scorpion species, including Hemsicorpius lepturus, Mesobuthus eupeus, Andructunus crassicuada, and Hottentotta saulcyi using cDNA library synthesis and high-throughput transcriptomic analysis of the venom glands. In a comparative way, we identified the cDNA encoding isoforms of subunits (alpha and beta) of BotLVP1/BmLVP1-like protein in the venom gland of three species except for H. lepturus. Characterization and structure determination of the LVP1_like proteins combined with gene map analysis provided evidence of the existence of some isoforms of LVP1_like proteins, encoded by genes with two exons and one intron, which can be classified in CSαβ superfamily in the venom gland of three Iranian scorpion species. According to the high similarity with BotLVP1 and BmLVP1, these proteins could also be potent to mediate cholesterol homeostasis. Background: Venom phospholipase D (PLDs), dermonecrotic toxins like, are the major molecules in the crude venom of scorpions, which are mainly responsible for lethality and dermonecrotic lesions during scorpion envenoming. The purpose of this study was fivefold: First, to identify transcripts coding for venom PLDs by transcriptomic analysis of the venom glands from Androctonus crassicauda, Hottentotta saulcyi, and Hemiscorpius lepturus; second, to classify them by sequence similarity to known PLDs and motif extraction method; third, to characterize scorpion PLDs; fourth to structural homology analysis with known dermonecrotic toxins; and fifth to investigate phylogenetic relationships of the PLD proteins. Results: We found that the venom gland of scorpions encodes two PLD isoforms: PLD1 ScoTox-beta and PLD2 ScoTox-alpha I. Two highly conserved regions shared by all PLD1s beta are GAN and HPCDC (HX2PCDC), and the most important conserved regions shared by all PLD2s alpha are two copies of the HKDG (HxKx4Dx6G) motif. We found that PLD1 beta is a 31–43 kDa acidic protein containing signal sequences, and PLD2 alpha is a 128 kDa basic protein without known signal sequences. The gene structures of PLD1 beta and PLD2 alpha contain 6 and 21 exons, respectively. Significant structural homology and similarities were found between the modeled PLD1 ScoTox-beta and the crystal structure of dermonecrotic toxins from Loxosceles intermedia. Conclusions: This is the first report on identifying PLDs from A. crassicauda and H. saulcyi venom glands. Our work provides valuable insights into the diversity of scorpion PLD genes and could be helpful in future studies on recombinant antivenoms production. [ABSTRACT FROM AUTHOR]

Scorpions are ancient and historically renowned for their potent venom. Traditionally, the systematics of this group of arthropods was supported by morphological characters, until recent phylogenomic analyses (using RNAseq data) revealed most of the higher‐level taxa to be non‐monophyletic. While these phylogenomic hypotheses are stable for almost all lineages, some nodes have been hard to resolve due to minimal taxonomic sampling (e.g. family Chactidae). In the same line, it has been shown that some nodes in the Arachnid Tree of Life show disagreement between hypotheses generated using transcritptomes and other genomic sources such as the ultraconserved elements (UCEs). Here, we compared the phylogenetic signal of transcriptomes vs. UCEs by retrieving UCEs from new and previously published scorpion transcriptomes and genomes, and reconstructed phylogenies using both datasets independently. We reexamined the monophyly and phylogenetic placement of Chactidae, sampling an additional chactid species using both datasets. Our results showed that both sets of genome‐scale datasets recovered highly similar topologies, with Chactidae rendered paraphyletic owing to the placement of Nullibrotheas allenii. As a first step toward redressing the systematics of Chactidae, we establish the family Anuroctonidae (new family) to accommodate the genus Anuroctonus. [ABSTRACT FROM AUTHOR]

Scorpion stings are a common medical emergency in many parts of the world. However, no statistical records are available about the incidence of scorpion sting envenomation and related mortality rates in the Sudan. This study aimed to describe the burden in the Sudan and to highlight the most affected states. We extracted retrospective hospital-based data on scorpions' stings envenomation for the years 2014–2018 from the annual statistical reports of the Federal Ministry of Health, Republic of the Sudan. Descriptive statistics were used to analyze the extracted data and elucidate the results. A total of 129,427 people were envenomed during 2014–2018 with a mean of 25,885 cases/year. More adults were stung compared to children. However, mortality was higher among children younger than 15 years old 4.7% (186/3974) than older victims 1% (56/5371). Envenomation was more frequent in males (56%) than females (44%). The Northern state reported the highest incidence of scorpion stings (344 per 100,000 population), followed by the River Nile state (240 per 100,000 population), and Khartoum state (174 per 100,000 population). Fatalities amongst hospitalized patients were 2.6% (242/9345) with the Northern state accounting for 34% (0.8/2.3) of total fatalities reported. [ABSTRACT FROM AUTHOR]

Scorpion envenomation is a common emergency department presentation in Arizona and antivenom is often administered. This study evaluates the safety of Centruroides (Scorpion) Immune F(ab')2 (Equine) administered as a single-vial-serial dosing strategy via slow intravenous push (IVP) over five minutes in the emergency department. This was a retrospective chart review of patients who received at least one vial of antivenom between April 1, 2019, and July 31, 2021. The primary outcome of this study was the rate of the serious antivenom administration-related adverse events. Twenty-five patients were included. One patient met criteria for a serious administration-related adverse event. No other serious administration, non-serious administration, or non-administration related adverse events were found. Neither grade of envenomation, repeat doses, nor total number of vials affected the rate of serious, non-serious, or non-administration-related adverse events. Venoms are primarily believed to evolve under strong diversifying selection resulting from persistent coevolution between predator and prey. Recent research has challenged this hypothesis, proposing that venoms from younger venomous lineages (e.g., snakes and cone snails) are governed predominantly by diversifying selection, while venoms from older venomous lineages (e.g., centipedes, scorpions, and spiders) are under stronger purifying selection. However, most research in older lineages has tested selection at more diverse phylogenetic scales. Although these tests are important for evaluating broad macroevolutionary trends underlying venom evolution, they are less equipped to detect species-level evolutionary trends, which likely have large impacts on venom variation seen at more diverse phylogenetic scales. To test for selection among closely related species from an older venomous lineage, we generated high-throughput venom-gland transcriptomes and venom proteomes for four populations of Giant Desert Hairy Scorpions (Hadrurus), including three Hadrurus arizonensis populations and one Hadrurus spadix population. We detected significant episodic and pervasive diversifying selection across a highly abundant toxin family that likely has a major role in venom function (α KTxs), providing a contrast to the stronger purifying selection identified from other studies on scorpion venoms. Conversely, we detected weak episodic diversifying and/or stronger purifying selection in four toxin families (non-disulfide bridged peptides, phospholipase A2s, scorpine-like antimicrobial peptides, and serine proteases), most of which were less abundant and likely have ancillary functional roles. Finally, although we detected several major toxin families at disproportionate transcriptomic and/or proteomic abundances, we did not identify significant sex-based variation in Hadrurus venoms. Background: Kerman Province is one of the endemic foci of scorpionism in southeastern Iran. Totally 17 species are reported for Kerman Province fauna. The current study seeks to determine the fauna of medically important scorpions in northern parts of this province. Methods: A descriptive cross-sectional study was conducted from January 2019 to March 2021. The specimens were captured from different areas, by rock-rolling during day and using Ultraviolet light at night. The specimens were preserved in ethanol (80%) and then identified using reliable identification keys for scorpions. Results: Totally, 499 specimens were captured from eight cities in northern Kerman. In total, 11 species belonging to three families were identified. Buthacus sp. is reported for the first time from Kerman Province. Buthacus sp., Hottentotta cf. juliae, and Hottentotta cf. schach, were the lowest in frequency of occurrence, against Mesobuthus navidpouri as the highest frequency. Conclusion: The results of this study are useful for the preparation of regional or monovalent antivenom for the Razi Vaccine and Serum Research Institute. Whilst undertaking curation in the collections of the Natural History Museum, London, the senior author came across a jar of scorpions marked 'scorpions with curled feet'. Further investigation showed this jar was full of scorpions with teratological anatomy. We hereby document and illustrate all of the specimens and discuss their respective anomalies. Furthermore, a comprehensive checklist of the literature on anomalies in scorpions, including giving the life stage and sex where known, is compiled for the benefit of future workers. A new species, Microbuthus saharicus sp. n., is described from the inland deserts of Mauritania; it represents the first exception to the general coastal distribution observed in the species of this genus. With this new description, the total number of known species in Africa is increased to five. The disrupted peri-Saharan pattern of distribution presented by the group is however confirmed. The diet of the Pharaoh Eagle-Owl, Bubo ascalaphus, was investigated based on 338 pellets collected from caves and underground caves in Ara'r region, northern Saudi Arabia. Small mammals constituted the highest number of consumed prey (75.75%), followed by arthropods (20%), birds (2.9%) and reptiles (1.26%). The Libyan Jird, Meriones libycus, was the most consumed rodent (26.46%) followed by Sundevall's Jird, Meriones crassus (20.47%), while the least were Cheesman Gerbil, Gerbillus cheesmani, and Wagner's Gerbil, Gerbillus dasyurus. At least three species of scorpions, Androctonus crassicauda, Compsbuthus sp. and Scorpio sp., and two species of reptiles (Ptyodactylus hasselquistii and Trapellus agnetae) were recovered. Study of owl pellet contents proved to be a valuable tool to study species composition in unexplored regions. Also, our findings substantiate the fact that the Pharaoh Eagle-Owl is an opportunistic species that adapts to available preys in its habitat. [ABSTRACT FROM AUTHOR]

Material belonging to a small collection of Moroccan arachnids housed in the Institute of Biodiversity and Ecosystem Research at the Bulgarian Academy of Sciences was examined and the results are presented here. A total of 28 species belonging to 16 families from three orders of arachnids were provided, as follows: Araneae (16 species, 12 families), Scorpiones (nine species, two families) and Solifugae (three species, two families). Five spider species, Adonea cf. fimbriata (Eresidae), Plexippus cf. clemens (Salticidae), Thanatus oblongiusculus (Philodromidae), Aelurillus aeruginosus (Salticidae) and Steatoda incomposita (Theridiidae), represent new or potential new records for the fauna of Morocco. An observation of phoresy by pseudoscorpions on a scorpion host is recorded for the first time worldwide. Pseudoscorpions of the endemic species Nannowithius wahrmani (Beier, 1963) (Pseudoscorpiones: Withiidae) were observed phoretic on the endemic scorpion species Birulatus israelensis Lourenço, 2002 (Scorpiones: Buthidae) in Israel. Spiders are among the animals evoking the highest fear and disgust and such a complex response might have been formed throughout human evolution. Ironically, most spiders do not present a serious threat, so the evolutionary explanation remains questionable. We suggest that other chelicerates, such as scorpions, have been potentially important in the formation and fixation of the spider-like category. In this eye-tracking study, we focused on the attentional, behavioral, and emotional response to images of spiders, scorpions, snakes, and crabs used as task-irrelevant distractors. Results show that spider-fearful subjects were selectively distracted by images of spiders and crabs. Interestingly, these stimuli were not rated as eliciting high fear contrary to the other animals. We hypothesize that spider-fearful participants might have mistaken crabs for spiders based on their shared physical characteristics. In contrast, subjects with no fear of spiders were the most distracted by snakes and scorpions which supports the view that scorpions as well as snakes are prioritized evolutionary relevant stimuli. We also found that the reaction time increased systematically with increasing subjective fear of spiders only when using spiders (and crabs to some extent) but not snakes and scorpions as distractors. The maximal pupil response covered not only the attentional and cognitive response but was also tightly correlated with the fear ratings of the picture stimuli. However, participants' fear of spiders did not affect individual reactions to scorpions measured by the maximal pupil response. We conclude that scorpions are evolutionary fear-relevant stimuli, however, the generalization between scorpions and spiders was not supported in spider-fearful participants. Herein a new species of Paruroctonus Werner, 1934 is described from alkali-sink habitats in the San Joaquin Desert of central California, Paruroctonus tulare sp. nov. It can be differentiated from other Paruroctonus by a combination of morphological features including scalloped pedipalp fingers in males, specific setal counts and morphometric ratios, and specific patterns of fuscous pigmentation. It also inhabits a unique distribution allopatric with all other Paruroctonus species except P. variabilis Hjelle, 1982. Photographs of a large series of live P. tulare sp. nov. from across their range and detailed images of several morphological features are provided, their distribution is modeled, a haplotype network is presented, and details about their habitat, ecology, and conservation are discussed.
