In this study, two types of scaffolds were fabricated by 3D printing. Morphology, physical properties, biochemical were evaluated. Cell morphology and distribution were observed. It was found that the silk fibroin/collagen (SF/C) scaffold-based material had significantly higher values than the silk fibroin/chitosan (SF/CS) scaffold-based material. Hematoxylin and eosin staining of the scaffolds revealed that the number of cells in the SF/C scaffold was higher. Cells grew well inside the SF/C scaffold as measured by scanning electron microscope. Reverse Transcriotion-Polymerase Chain Reaction (RT-PCR) and Western blot showed that type II collagen and Sox9 can be found in SF/C scaffold. In this study, the performances of different preparation methods of the scaffolds were analyzed for chondrocyte tissue engineering. Silk fibroin/collagen (SF/C) was fabricated using a vacuum freeze-dried technique and by 3D printing. The porosity, water absorption expansion rates, mechanical properties, and pore sizes of the resulting materials were evaluated. The proliferation and metabolism of the cells was detected at different time points using an MTT assay. Cell morphologies and distributions were observed by histological analysis and scanning electron microscopy (SEM). The porosity, water absorption expansion rate, and Young’s modulus of the material obtained via 3D printing were significantly higher than those obtained by the freeze-dried method, while the pore size did not differ significantly between the two methods. MTT assay results showed that the metabolism of cells seeded on the 3D printed scaffolds was more viable than the metabolism on the freeze-dried material. H&E staining of the scaffolds revealed that the number of cells in the 3D printed scaffold was higher in comparison to a similar measurement on the freeze-dried material. Consequently, stem cells grew well inside the 3D printed scaffolds, as measured by SEM, while the internal structure of the freeze-dried scaffold was disordered. Introduction: In this study, the silk fibroin blended constructs were produced, scaffold performances of different kinds of scaffold were analyzed, and the better type for tissue engineering was optimized. Methods: The silk fibroin/collagen (SF/C) and silk fibroin/chitosan (SF/CS) were made using a freeze-drying technique, porosity, water absorption expansion rate, mechanical properties and pore size of different scaffold was detected. Bone marrow mesenchymal stem cells (BMSCs) of 4-week-old male Wistar rats were separated by density gradient centrifugation, third generation BMSCs were seeded onto scaffolds, cultured 14 days, proliferation and metabolize of cells were detected in different time using the thiazolyl blue tetrazolium bromide (MTT) assay method, and cell morphology and distribution were observed by histological analysis and scanning electron microscopy (SEM). Results: Porosity, water absorption expansion rate and Young's modulus of SF/C were significantly higher than SF/CS ( p < 0.05); pore size of SF/C and SF/CS was 103 ± 12 and 76 ± 11 μm and had no significant differences between two types ( p > 0.05); MTT results showed that the metabolism of cells in the SF/C was better than SF/CS; after cultivation for 14 days, in the inner zone of scaffolds, cells staining were little or absent from SF/CS, lots of cells staining were existing in SF/C; pore size was consistent, holes communicated with each other better, stem cells grew well inside the scaffolds, extended fully and secreted much extracellular matrix under SEM in SF/C scaffold; internal structure of SF/CS was disorder, holes size were not consistent, and did not communicated with each other and cells were partly dead. Conclusion: Compared with SF/CS, SF/C scaffold showed better porosity, water absorption expansion rate, elasticity modulus and pore size, cells grow well inside the scaffolds, and was more suitable for tissue engineering. The most prevalent oral and maxillofacial cancer is oral squamous cell carcinoma (OSCC). Patient survival is compromised by relapse due to post-operative tumor remnants and significant mucosal defects. Photothermal therapy (PTT) is used to ablate local malignancies, but the capacity of tumor cells to resist it is correlated with the overexpression of heat shock protein 70 (HSP70). In this work, PTT and tissue engineering scaffold were rationally integrated to construct a Ti 3 C 2 MXene, collagen, silk fibroin and quercetin composite scaffold (M-CSQ scaffold), a 3D printed biomaterial that simultaneously kill OSCC cells and promote the regeneration of the mucosal defects. The M-CSQ scaffolds were prepared by cryogenic 3D printing and freeze-drying techniques with sufficient pores that allow an abundant cell migration and provide a proliferation space. Ti 3 C 2 MXene has excellent photothermal conversion ability and stability. Quercetin targeted HSP70 to decrease its expression in OSCC cells, consequently weakening their resistance to high temperature and enhancing the effect of PTT. The M-CSQ scaffold effectively killed OSCC cells in vitro and inhibited tumor growth in vivo. [Display omitted] The tissue engineering is a promising method for cartilage regeneration studies. The rough in vivo initial repair environment of articular cartilage is a challenge for the development of cartilage tissue engineering. Hereon, in this study, a silk fibroin/collagen type II scaffold was designed. The optimal mechanical environment for chondrocyte/scaffold complex in vitro maturation culture was determined and in vitro cultured mature artificial cartilage was evaluated for in vivo defect repair. Our research shows that the silk fibroin/collagen type II scaffold has good performance and can meet the special requirements of cartilage repair. 10 % compressive strain is the optimal compressive loading in vitro. Under optimal compression loading in vitro , chondrocytes can proliferate and grow rapidly on the scaffold to achieve better "maturation". The in vitro cultured chondrocyte/scaffold complex effectively improves the cartilage repair effect after implantation. The compressive elastic modulus of the new tissue reached 0.682 ± 0.010 MPa after 12 weeks of repair (0.714 ± 0.011 MPa for the surrounding host cartilage). Therefore, this study not only enhance the effect of cartilage repair but also provides a promising strategy for mechanical stimulation to promote functional in situ regeneration of articular cartilage. As a promising alternative to autologous nerve grafts, tissue-engineered nerve grafts have been extensively studied as a way to bridge peripheral nerve defects and guide nerve regeneration. The main difference between autogenous nerve grafts and tissue-engineered nerve grafts is the regenerative microenvironment formed by the grafts. If an appropriate regenerative microenvironment is provided, the repair of a peripheral nerve is feasible. In this study, to mimic the body’s natural regenerative microenvironment closely, we co-cultured Schwann cells (SCs) and adipose-derived stem cells (ADSCs) as seed cells and introduced them into a silk fibroin (SF)/collagen scaffold to construct a tissue-engineered nerve conduit (TENC). Twelve weeks after the three different grafts (plain SF/collagen scaffold, TENC, and autograft) were transplanted to bridge 1-cm long sciatic nerve defects in rats, a series of electrophysiological examinations and morphological analyses were performed to evaluate the effect of the tissue-engineered nerve grafts on peripheral nerve regeneration. The regenerative outcomes showed that the effect of treatment with TENCs was similar to that with autologous nerve grafts but superior to that with plain SF/collagen scaffolds. Meanwhile, no experimental animals had inflammation around the grafts. Based on this evidence, our findings suggest that the TENC we developed could improve the regenerative microenvironment and accelerate nerve regeneration compared to plain SF/collagen and may serve as a promising strategy for peripheral nerve repair.
