We report a kinetic and mechanistic study on the title reactions, in which 1 O 2 was generated by the reaction of H 2 O 2 with Cl 2 and bubbled into an aqueous solution of guanine and 9-methylguanine (9MG) at different pH values. Oxidation kinetics and product branching ratios were measured using online electrospray ionization mass spectrometry coupled with absorption and emission spectrophotometry, and product structures were determined by collision-induced dissociation (CID) tandem mass spectrometry. Experiments revealed strong pH dependence of the reactions. The oxidation of guanine is noticeable only in basic solution, while the oxidation of 9MG is weak in acidic solution, increases in neutral solution, and becomes intensive in basic solution. 5-Guanidinohydantoin (Gh) and spiroiminodihydantoin (Sp) were detected as the major oxidation products of guanine and 9MG, and Sp became dominant in basic solution. A reaction intermediate was captured in mass spectra, and assigned to gem-diol on the basis of CID measurements. This intermediate served as the precursor for the formation of Gh. After taking into account solution compositions at each pH, first-order oxidation rate constants were extracted for individual species: that is, 3.2-3.6 × 10 7 M -1 s -1 for deprotonated guanine, and 1.2 × 10 6 and 4.6-4.9 × 10 7 M -1 s -1 for neutral and deprotonated 9MG, respectively. Guided by approximately spin-projected density-functional-theory-calculated reaction potential energy surfaces, the kinetics for the initial 1 O 2 addition to guanine and 9MG was evaluated using transition state theory (TST). The comparison between TST modeling and experiment confirms that 1 O 2 addition is rate-limiting for oxidation, which forms endoperoxide and peroxide intermediates as determined in previous measurements of the same systems in the gas phase.

Mutagenicity of singlet O2 is due to its oxidatively generated damage to the guanine nucleobases of DNA. Oxidation of neutral guanosine has been assumed to be initiated by the formation of a transient 4,8-endoperoxide via a Diels-Alder cycloaddition of singlet O2. Protonation and deprotonation of guanosine represent another factor related to DNA damage and repair. Herein, 9-methylguanine was utilized as a model substrate to mimic the correlation between singlet O2 oxidation of the nucleoside and its ionization states, both in the absence and in the presence of water ligands. We used guided-ion-beam scattering tandem mass spectrometry to detect and quantify transient intermediates at room temperature. To provide a reliable description of reaction potential surfaces, different levels of theory including restricted and unrestricted density functional theory, CCSD(T), MP2, and multi-reference CASSCF and CASMP2 were applied. By means of molecular potential, kinetic and direct dynamics simulations, two reaction pathways were identified and neither follows the mechanism for neutral guanosine. Singlet O2 oxidation of protonated 9-methylguanine begins by a concerted cycloaddition; but it is mediated by a 5,8-endoperoxide. By contrast, a concerted cycloaddition does not occur for deprotonated 9-methylguanine. The latter involves a stepwise addition starting with the formation of an 8-peroxide, which subsequently evolves to a 4,8-endoperoxide. This dichotomy implies that acidic and basic media may lead to different chemistries for guanosine oxidation in aqueous solutions, starting from initial stage. The comparison with oxidation of protonated/deprotonated guanine illustrates the different mechanisms and products and particularly the suppressed oxidizability of 9-methylguanine vs. free guanine.

The chemistry of singlet O2 toward the guanine base of DNA is highly relevant to DNA lesion, mutation, cell death, and pathological conditions. Corroborated by results from potential energy surface exploration, kinetic modeling, and dynamics simulations, various aspects of endoperoxide formation and transformation (including its dependence on guanine ionization and hydration states, as well as on collision energy) were determined. We report a reaction apparatus developed to monitor singlet oxygen ((1)O2) reactions in solution using on-line ESI mass spectrometry and spectroscopy measurements. (1)O2 was generated in the gas phase by the reaction of H2O2 with Cl2, detected by its emission at 1270 nm, and bubbled into aqueous solution continuously. (1)O2 concentrations in solution were linearly related to the emission intensities of airborne (1)O2, and their absolute scales were established based on a calibration using 9,10-anthracene dipropionate dianion as an (1)O2 trapping agent. Products from (1)O2 oxidation were monitored by UV-Vis absorption and positive/negative ESI mass spectra, and product structures were elucidated using collision-induced dissociation-tandem mass spectrometry. To suppress electrical discharge in negative ESI of aqueous solution, methanol was added to electrospray via in-spray solution mixing using theta-glass ESI emitters. Capitalizing on this apparatus, the reaction of (1)O2 with methionine was investigated. We have identified methionine oxidation intermediates and products at different pH, and measured reaction rate constants. (1)O2 oxidation of methionine is mediated by persulfoxide in both acidic and basic solutions. Persulfoxide continues to react with another methionine, yielding methionine sulfoxide as end-product albeit with a much lower reaction rate in basic solution. Density functional theory was used to explore reaction potential energy surfaces and establish kinetic models, with solvation effects simulated using the polarized continuum model. Combined with our previous study of gas-phase methionine ions with (1)O2, evolution of methionine oxidation pathways at different ionization states and in different media is described.

We report an in-depth study on the gas-phase reactions of singlet O2[a(1)Δg] with methionine (Met) at different ionization and hydration states (including deprotonated [Met - H](-), hydrated deprotonated [Met - H](-)(H2O)1,2, and hydrated protonated MetH(+)(H2O)1,2), using guided-ion-beam scattering mass spectrometry. The measurements include the effects of collision energy (Ecol) on reaction cross sections over a center-of-mass Ecol range from 0.05 to 1.0 eV. The aim of this study is to probe the influences of Met ionization and hydration on its oxidation mechanism and dynamics. Density functional theory calculations, Rice-Ramsperger-Kassel-Marcus modeling, and quasi-classical, direct dynamics trajectory simulations were performed to examine the properties of various complexes and transition states that might be important along reaction coordinates, probe reaction potential energy surfaces, and to establish the atomic-level mechanism for the Met oxidation process. No oxidation products were observed for the reaction of [Met - H](-) with (1)O2 due to the high-energy barriers located in the product channels for this system. However, this nonreactive property was altered by the microsolvation of [Met - H](-); as a result, hydroperoxides were captured as the oxidation products for [Met - H](-)(H2O)1,2 + (1)O2. For the reaction of MetH(+)(H2O)1,2 + (1)O2, besides formation of hydroperoxides, an H2O2 elimination channel was observed. The latter channel is similar to what was found in the reaction of dehydrated MetH(+) with (1)O2 (J. Phys. Chem. B 2011, 115, 2671). The reactions of hydrated protonated and deprotonated Met are all inhibited by Ecol, becoming negligible at Ecol ≥ 0.5 eV. The kinetic and dynamical consequences of microsolvation on Met oxidation and their biological implications are discussed.

It has been proposed (J. Phys. Chem. B 2011, 115, 2671) that the ammonium group is involved in the gas-phase reaction of protonated methionine (MetH(+)) with singlet oxygen (1)O2, yielding hydrogen peroxide and a dehydro compound of MetH(+) where the -NH3(+) transforms into cyclic -NH2-. For the work reported, the gas-phase reaction of protonated N-acetylmethionine (Ac-MetH(+)) with (1)O2 was examined, including the measurements of reaction products and cross sections over a center-of-mass collision energy (Ecol) range from 0.05 to 1.0 eV using a guided-ion-beam apparatus. The aim is to probe how the acetylation of the ammonium group affects the oxidation chemistry of the ensuing Ac-MetH(+). Properties of intermediates, transition states, and products along the reaction coordinate were explored using density functional theory calculations and Rice-Ramsperger-Kassel-Marcus (RRKM) modeling. Direct dynamics trajectory simulations were carried out at Ecol of 0.05 and 0.1 eV using the B3LYP/4-31G(d) level of theory. In contrast to the highly efficient reaction of MetH(+) + (1)O2, the reaction of Ac-MetH(+) + (1)O2 is extremely inefficient, despite there being exoergic pathways. Two product channels were observed, corresponding to transfer of two H atoms from Ac-MetH(+) to (1)O2 (H2T), and methyl elimination (ME) from a sulfone intermediate complex. Both channels are inhibited by collision energies, becoming negligible at Ecol > 0.2 eV. Analysis of RRKM and trajectory results suggests that a complex-mediated mechanism might be involved at very low Ecol, but direct, nonreactive collisions prevail over the entire Ecol range and physical quenching of (1)O2 occurs during the early stage of collisions.

We present a study on the reactions of singlet oxygen O2[a(1)Δg] with hydrated protonated and deprotonated cysteine (Cys) in the gas phase, including measurements of the effects of collision energy (E(col)) and hydration number on reaction cross sections over a center-of-mass E(col) range from 0.05 to 1.0 eV. The aim is to probe how successive addition of water molecules changes the oxidation chemistry of Cys in the gas phase. Hydrated clusters, generated by electrospray ionization, have structures of HSCH2CH(NH3(+))CO2H(H2O)(1,2) and HSCH2CH(NH2)CO2(-)(H2O)(1,2) for protonated and deprotonated forms, respectively. In contrast to (1)O2 reactions with dehydrated protonated/deprotonated Cys of which hydroperoxide products all decomposed, reactions with hydrated protonated/deprotonated Cys yielded stable hydroperoxide products, analogous to photooxidation reaction of Cys in solution. We investigated the number of water ligands necessary to produce a stable hydroperoxide, and found that a single water molecule suffices--that is, to relax nascent, energized hydroperoxide in the hydrated cluster by elimination of water. Hydrated protonated Cys shows higher reaction efficiency than the hydrated deprotonated one, particularly with the addition of the second water ligand. Reactions of hydrated protonated/deprotonated Cys are suppressed by E(col), becoming negligible at E(col) ≥ 0.5 eV. Density functional theory calculations were used to locate reaction coordinates for these systems. Quasi-classical, direct dynamics trajectory simulations were performed for HSCH2CH(NH3(+))CO2H(H2O) + (1)O2 at the B3LYP/4-31G(d) level of theory. Analysis of trajectories highlights the importance of complex mediation in the early stages of the reaction, and illustrates that water can catalyze proton transfer within the hydrated complex.

We present a study on the gas-phase reaction of deprotonated cysteine with the lowest electronically excited state of molecular oxygen O2[a(1)Δg], including the measurement of the effects of collision energy (E(col)) on reaction cross sections over a center-of-mass E(col) range from 0.1 to 1.0 eV. Deprotonated cysteine was generated using electrospray ionization, and has a carboxylate anionic structure (HSCH2CH(NH2)CO2(-)) in the gas phase. Three product ion channels were observed. The dissociation of HSCH2CH(NH2)CO2(-) to NH2CH2CO2(-) and neutral CH2S has the largest cross section over the entire E(col) range. This product channel is driven by the electronic excitation energy of (1)O2 (the so-called dissociative excitation transfer), and is strongly suppressed by E(col). Two minor channels correspond to the formation of HSCH2C(NH)CO2(-) + H2O2 via abstraction of two hydrogen atoms from HSCH2CH(NH2)CO2(-) by (1)O2, and the formation of OSCH2CH(NH2)CO2(-) radical via elimination of ·OH from an intermediate complex, respectively. Density functional theory calculations were used to locate various complexes, transition states, and products. Quasi-classical direct dynamics trajectory simulations were carried out at E(col) = 0.2 eV using the B3LYP/4-31G(d) level of theory. Trajectory results were used to guide the construction of a reaction coordinate, discriminate between different mechanisms, and provide additional mechanistic insights. Analysis of trajectories highlights the importance of complex mediation at the early stages of all reactions, and suggests a partially concerted mechanism for H2O2 elimination.

The reactions of deprotonated tyrosine ([Tyr-H](-)) and tryptophan ([Trp-H](-)) with the lowest electronically excited state of molecular oxygen O(2)[a(1)Δ(g)] have been studied in the gas phase, including the measurement of the effects of collision energy (E(col)) on reaction cross sections over a center-of-mass E(col) range from 0.05 to 1.0 eV. [Tyr-H](-) and [Trp-H](-) were generated using electrospray ionization, and both have a pure carboxylate anion structure in the gas phase. Density functional theory calculations and RRKM modeling were used to examine properties of various complexes, transition states, and products that might be important along the reaction coordinate. It was found that deprotonation of Tyr and Trp results in a large effect on their (1)O(2)-mediated oxidation. For [Tyr-H](-), the reaction corresponds to the formation of a hydroperoxide intermediate, followed by intramolecular H transfer and subsequent dissociation to product ion 4-(2-aminovinyl)phenolate, and neutral H(2)O(2) and CO(2). Despite that the reaction is 1.83 eV exothermic, the reaction cross section shows a threshold-like behavior at low E(col) and increases with increasing E(col), suggesting that the reaction bears an activation barrier above the reactants. Quasi-classical, direct dynamics trajectory simulations were carried out for [Tyr-H](-) + (1)O(2) at E(col) = 0.75 eV, using B3LYP/4-31G* level of theory. Trajectories demonstrated the intermediacy of complexes at the early stage of the reaction. A similar product channel was observed in the reaction of [Trp-H](-) with (1)O(2), yielding product ion 3-(2-aminovinyl)indol-1-ide, H(2)O(2) and CO(2). However, the reaction cross section of [Trp-H](-) is strongly suppressed by E(col) and becoming negligible at E(col) > 1.0 eV, indicating that this reaction proceeds without energy barriers above the reactants.

Electronic structure calculations were used to examine properties of complexes, transition states and products that might be important along the reaction coordinate. For CysH(+) + (1)O(2), the product channel corresponds to C(α)-C(β) bond rupture of a hydroperoxide intermediate CysOOH(+) accompanied by intramolecular H atom transfer, and subsequent dissociation to H(2)NCHCO(2)H(+), CH(3)SH and ground triplet state O(2). The reaction is driven by the electronic excitation energy of (1)O(2), the so-called dissociative excitation energy transfer. Quasi-classical direct dynamics trajectory simulations were calculated for CysH(+) + (1)O(2) at E(col) = 0.2 and 0.3 eV, using the B3LYP/6-21G method. Most trajectories formed intermediate complexes with significant lifetime, implying the importance of complex formation at the early stage of the reaction. Dissociative excitation energy transfer was also observed in the reaction of TrpH(+) with (1)O(2), leading to dissociation of a TrpOOH(+) intermediate. In contrast to CysOOH(+), TrpOOH(+) dissociates into a glycine molecule and charged indole side chain in addition to ground-state O(2) because this product charge state is energetically favorable. The reactions of CysH(+) + (1)O(2) and TrpH(+) + (1)O(2) present similar E(col) dependence, i.e., strongly suppressed by collision energy and becoming negligible at E(col) > 0.5 eV. This is consistent with a complex-mediated mechanism where a long-lived complex is critical for converting the electronic energy of (1)O(2) to the form of internal energy needed to drive complex dissociation.

The reaction of protonated methionine with the lowest electronically excited state of molecular oxygen O(2)(a(1)Δ(g)) was studied in a guided ion beam apparatus, including the measurement of reaction cross sections over a center-of-mass collision energy (E(col)) range of 0.1-2.0 eV. A series of electronic structure and RRKM calculations were used to examine the properties of various complexes and transition states that might be important along the reaction coordinate. Only one product channel is observed, corresponding to generation of hydrogen peroxide via transfer of two hydrogen atoms (H2T) from protonated methionine to singlet oxygen. At low collision energies, the reaction approaches the collision limit and may be mediated by intermediate complexes. The reaction shows strong inhibition by collision energy, and becomes negligible at E(col) > 1.25 eV. A large set of quasi-classical direct dynamics trajectory simulations were calculated at the B3LYP/6-21G level of theory. Trajectories reproduced experimental results and provided insight into the mechanistic origin of the H2T reaction, how the reaction probability varies with impact parameter, and the suppressing effect of collision energy. Analysis of the trajectories shows that at E(col) = 1.0 eV the reaction is mediated by a precursor and/or hydroperoxide complex, and is sharply orientation-dependent. Only 20% of collisions have favorable reactant orientations at the collision point, and of those, less than half form precursor and hydroperoxide complexes which eventually lead to reaction. The narrow range of reactive collision orientations, together with physical quenching of (1)O(2) via intersystem crossing between singlet and triplet electronic states, may account for the low reaction efficiency observed at high E(col)., (© 2011 American Chemical Society)

Reaction of protonated tyrosine with the lowest electronically excited singlet state of molecular oxygen, (1)O(2) (a(1)Delta(g)), is reported over the center-of-mass collision energy (E(col)) range from 0.1 to 3.0 eV, using an electrospray-ionization, guided-ion-beam scattering instrument, in conjunction with ab initio electronic structure calculations and direct dynamics trajectory simulations. Only one product channel is observed, corresponding to generation of hydrogen peroxide via transfer of two hydrogen atoms from protonated tyrosine. Despite being exoergic, the reaction is in competition with physical quenching of (1)O(2) and is very inefficient. At low E(col), the reaction may be mediated by intermediate complexes and shows strong inhibition by collision energy. At high E(col), the reaction efficiency drops to approximately 1% and starts to have contribution from a direct mechanism. Quasi-classical trajectory simulations were performed to probe the mechanism at high collision energies. Analysis of trajectories shows that, at E(col) of 3.0 eV, a small fraction of hydrogen peroxide (25%) is produced via a direct, concerted mechanism where two hydrogen atoms are transferred simultaneously, but most hydrogen peroxide (75%) is formed by dissociation of hydroperoxide intermediates. According to ab initio calculations and trajectory simulations, collisions also lead to formation of various endoperoxides, and dissociation of endoperoxides may play a role in physical quenching of (1)O(2). The apparatus and experimental techniques are described in detail.

8‐Oxo‐2′‐deoxyguanosine (OG) is the most common DNA lesion. Notably, OG becomes more susceptible to oxidative damage than the undamaged nucleoside, forming mutagenic products in vivo. Herein the reactions of singlet O2 with the radical cations of 8‐oxo‐2′‐deoxyguanosine (OG.+) and 9‐methyl‐8‐oxoguanine (9MOG.+) were investigated using ion‐molecule scattering mass spectrometry, from which barrierless, exothermic O2‐addition products were detected for both reaction systems. Corroborated by static reaction potential energy surface constructed using multi‐reference CASPT2 theory and molecular dynamics simulated in the presence of the reactants′ kinetic and internal energies, the C5‐terminal O2‐addition was pinpointed as the most probable reaction pathway. By elucidating the reaction mechanism, kinetics and dynamics, and reaction products and energetics, this work constitutes the first report unraveling the synergetic damage of OG by ionizing radiation and singlet O2.
