Ethnopharmacological Relevance: Saussurea obvallata (DC.) Edgew. is a traditional Tibetan medicine used for the treatment of inflammation-related diseases, but the scientific validation was very limited., Aim of the Study: This study aimed to rapid screen and targeted isolate cyclooxygenase-2 (COX-2) inhibitors from S. obvallata extract., Materials and Methods: An efficient ligand-fishing method based on affinity solid phase extraction (A-SPE) combining with HPLC was developed. The identified COX-2 inhibitors were separated using preparative liquid chromatography. In vitro COX-2 inhibition assays were employed to confirm the inhibitory activities of the isolated compounds. In addition, the effect of the isolated compounds on the production of prostaglandin E2 (PGE2) and the expression of COX-2 in LPS-induced RAW 264.7 were evaluated., Results: A total of four phenylpropanoids, isolariciresinol, syringaresinol, pinoresinol and balanophonin were targeted isolated as COX-2 inhibitors with IC 50 values of 36.4 ± 2.6 μM, 23.1 ± 1.8 μM, 3.6 ± 0.3 μM and 12.1 ± 0.9 μM, respectively. The isolated compounds significantly inhibited LPS-induced NO production in a dose-dependent manner. And, the results of the inhibitory effect on the release of PGE 2 and the expression of COX-2 in LPS-induced macrophages were consistent with A-SPE analysis., Conclusion: The present work demonstrated that the developed A-SPE-HPLC method could successfully targeted isolated COX-2 inhibitors from S. obvallata extract. And, the isolation results indicated that the therapeutic effect of S. obvallata on inflammation-related diseases was partly based on the COX-2 active ingredients., (Copyright © 2021. Pterocephalus hookeri , as a kind of popular traditional Tibetan medicine, is reputed to treat inflammatory related diseases. In the present work, a cyclooxygenase-2 functionalized affinity solid-phase extraction HPLC system was developed and combined with preparative-HPLC for rapidly screening and separating cyclooxygenase-2 ligand from P. hookeri extracts. Firstly, ligands of cyclooxygenase-2 were screened from extracts by affinity solid-phase extraction HPLC system. Then directed by the screening results, the recognized potential active compounds were targeted separated. As a result, the major cyclooxygenase-2 inhibitor of P. hookeri was obtained with a purity of >95%, which was identified as sylvestroside I. To test the accuracy of this method, the anti-inflammatory activity of sylvestroside I was inspected in lipopolysaccharide-induced RAW 264.7 cells. The results show that sylvestroside I significantly suppressed the release of prostaglandin E 2 with dose-dependent, which was in good agreement with the screening result of the affinity solid-phase method. This method of integration of screening and targeted separation proved to be very efficient for the recognition and isolation of cyclooxygenase-2 inhibitors from natural products.

The high-polar compounds from natural products are often used as medicines due to their good bioactivities. However, owing to the complexity and diversity of their structure, the separation of high-polar compounds is still a challenging work. For this, an efficient method for enrichment and separation of the high-polar compounds from Saussurea obvallata was developed. First, the target compounds were enriched from the total extract using a solid-phase extraction method. An offline two-dimensional liquid chromatography method was used for the separation of pure compounds from the enriched sample. After optimization of chromatographic conditions, high separation selectivity of target compounds was obtained on a polar-modified C18 column and a HILIC XAmide column. Hence, a two-dimensional reversed-phase × hydrophilic interaction liquid chromatography system was constructed and enlarged from the analytical level to the preparative level. In the first dimension, four fractions were obtained on the XCharge C18 column with a recovery rate of 71.2%. In the second-dimension preparation on the XAmide column, eight high-polar compounds with more than 96% purity were isolated. All compounds were isolated from Saussurea obvallata for the first time. Pterocephalus hookeri, a classical Tibetan herb, is mainly used to treat rheumatoid arthritis (RA) and contains various constituents potentially with cyclooxygenase-2 (COX-2) selective inhibition. A novel strategy for screening and target separating COX-2 inhibitors from the extracts of P. hookeri based on affinity solid-phase extraction (ASPE) column combined with preparative high-performance liquid chromatography (pre-HPLC) was successfully developed. The potential COX-2 inhibitors of P. hookeri were screened and recognized by the ASPE-HPLC system, which strategy is to analyze the compounds isolated by the ASPE column. Then, the active compounds were targeted separated by pre-HPLC according to real-time chromatograms. The control drugs celecoxib and glipizide were analyzed to verify the specificity and accuracy of the developed method. As a result, two pure compounds with COX-2 binding affinities were successfully separated from P. hookeri. They were characterized as swertisin and scopoletin using 1 H- and 13 C NMR spectroscopy, and the in vitro COX-2 inhibitory activities were verified. Compounds with COX-2 inhibitory activities could be screened and targeted separated from crude extracts by this strategy, which indicated that the proposed method was feasible, robust and effective for rapid separation of COX-2 inhibitors from natural products., (Copyright © 2021 Elsevier B.V. In this paper, the potential use of multiwalled carbon nanotubes (MWCNTs) as solid phase extraction (SPE) adsorbent was evaluated for preconcentration of linear alkylbenzene sulfonates (LAS) using ion-pair (IP)-SPE with tetrabutylammonium hydroxide (TBAH). The LAS homologues present in the aqueous sample were ion-paired with TBAH and the solution was passed through the MWCNT cartridges. The analytes retained in the cartridge were eluted with methanol and the concentrated methanol extract was analysed by HPLC-UV. In order to obtain the satisfactory recovery of LAS homologues, various parameters including the type and amount of the ion-pair reagents, the desorption and enrichment conditions such as the effect of eluent and its volume, pH, the flow rate, the ultrasonic time of sample, and the volume of sample solution were systematically optimized. Under the optimal conditions, LAS homologues could be easily extracted by the proposed SPE cartridge. The favorable limits of detection (LOD) for LAS homologues were in the range from 0.02 to 0.03 μg L(-1), and the relative standard deviations (RSDs) were 1.55-2.54% for 10 μg L(-1) LAS (n=6). The proposed method has been successfully applied for the analysis of LAS homologues in aqueous environmental samples. A comparison study with ion-pair solid extraction on MWCNTs, C8 and C18 as adsorbents for LAS demonstrated that ion pair-based solid extraction on MWCNTs adsorbent was advantageous over C8 and C18, the widely used traditional adsorbents., (Copyright © 2011 Elsevier B.V. The potential of eggshell membrane (ESM) as a novel solid-phase extraction bio-adsorbent was investigated in the present study. The ESM with a unique structure of intricate lattice network showed a predominant ability to capture linear alkylbenzene sulfonates (LAS) as a model of organic pollutants by the hydrophobic interactions between ESM and LAS molecular at pH very close to the isoelectric point of ESM, which was similar to the most widely used trapping mechanism for SPE. Under the optimal conditions, the breakthrough capacities of the ESM packed cartridge for C10-C13 LAS homologues were found to be 30, 53, 50, and 43microgg(-1), respectively. On the basis of high-performance liquid chromatography separation and UV detection of LAS homologues, the proposed system could respond down to 0.027ngmL(-1) of LAS with a linear calibration range from 0.2 to 100ngmL(-1), showing a good LAS enrichment ability of eggshell membrane biomaterial with high sensitivity, and could be successfully used for the detection of residual LAS in environmental water samples. The reproducibility among columns was satisfactory (RSD among columns is less than 10%). The eggshell membrane (ESM) contains several surface functional groups such as amines, amides and carboxylic groups with potential as SPE adsorbent for the retention of target species of interest. In this paper, the potential use of ESM, a typical biomaterial, as solid-phase extraction (SPE) adsorbent is evaluated for analysis of trace arsenic(V) in environmental water samples in combination with hydride generation atomic fluorescence spectrometry (HG-AFS). In order to obtain the satisfactory recovery of arsenic(V), various parameters including the desorption and enrichment conditions such as pH, the flow rate and the volume of sample solution, the amount of ESM and the content of sodium chloride were systematically optimized and the effects of co-existed ions were also investigated in detail. Under the optimal conditions, arsenic(V) could be easily extracted by the ESM packed cartridge and the breakthrough adsorption capacity was found to be 3.9 microg g(-1). The favorable limit of detection (LOD) for arsenic(V) was found to be 0.001 microg L(-1) with an enrichment factor of 33.3, and the relative standard deviations (R.S.Ds) was 2.1% for 0.6 microg L(-1) arsenic (n=11). The reproducibility among columns was satisfactory (R.S.D. among columns is less than 5%). The end functionalization of CNTs can introduce oxygen-containing negatively functional groups such as -COOH, -OH, or -CO on their surface site. If cationic surfactant such as cetyltrimethylammonium chloride (CTAC) was added to the functionalized CNTs, then interactions such as hydrophobic and ionic may lead to formation of hemimicelle/admicelle aggregates on the CNTs, a new kind of adsorbents, namely, the hemimicelle capped CMMWCNTs, is obtained. The application of the hemimicelle capped carbon nanotubes-based nanosized solid-phase extraction (SPE) adsorbents in environmental analysis is reported for the first time using arsenic as model target. The effect of adsorption and desorption conditions for arsenic including the amount of surfactant, initial pH of sample solution, the ultrasonic time of sample solution, the amount of electrolyte, flow rate, eluent and its amount were investigated and optimized prior to its determination by atomic fluorescence spectrophotometry (AFS). Arsenic can be quantitatively retained on the hemimicelle capped CMMWCNTs at pH 5-6 from sample volume up to 500 mL and then eluted completely with 2 molL(-1) HNO(3) in the presence of 10 mgL(-1) CTAC. The method detection limit for arsenic determination with AFS detection was 2ngL(-1), and the relative standard deviation (RSD, n=11) was 5.3% at the 0.5 microgL(-1) level. The recoveries of arsenic in the spiked environmental water samples ranged from 94% to 104.29% with 500 mL of water sample. The proposed method has been applied successfully to the analysis of arsenic in aqueous environmental samples, which demonstrates the hemimicelle capped CMMWCNTs can be an excellent SPE adsorbents for arsenic pretreatment and enrichment from real water samples.

Abstract: The potential use of cigarette filters (CFs) as solid-phase extraction (SPE) adsorbents for the preconcentration of six fluoroquinolones (FQs) antibacterial agents prior to liquid chromatography was examined in this paper. In order to find a suitable procedure for extraction of the target FQs in one single step, various parameters probably affecting the extraction efficiency including the eluent kind and volume, sample flow rate, pH, ion strength and sample volume were systematically optimized. Under the optimized conditions, the target FQs could be easily extracted by the proposed SPE cartridge. Combination of SPE with HPLC/UV provided detection limits for different FQs of 2–5ngL−1 when 500mL of water sample was processed. The precision of the method, expressed as relative standard deviation, ranged from 4.1 to 6.3% for 2.5μgL−1 FQs. The recoveries of FQs spiked in environmental water samples ranged from 76 to 112%. The results obtained from the proposed method demonstrated that CFs-based solid-phase extraction combined with HPLC/UV was suitable for analyzing fluoroquinolones in water samples at ngL−1 concentration level. An offline preparative two-dimensional reversed-phase liquid chromatography/ hydrophilic interaction liquid chromatography coupled with hydrophilic interaction solid-phase extraction method was developed for the preparative isolation of flavonoid glycosides from a crude sample of Sphaerophysa salsula. First, the non-flavonoids were removed using an XAmide solid-phase extraction cartridge. Based on the separation results of three different chromatographic stationary phases, the first-dimensional preparation was performed on an XAqua C18 prep column, and 15 fractions were obtained from the 5.2 g target sample. Then, three representative fractions were selected for additional purification on an XAmide preparative column to further isolate the flavonoid glycosides. In all, eight flavonoid glycosides were isolated in purities over 97%. The results demonstrated that the two-dimensional liquid chromatography method used in this study was effective for the preparative separation of flavonoid glycosides from Sphaerophysa salsula. Abstract: Carbon nanotubes (CNTs) are a kind of new carbon-based nano-materials which have drawn great attention in many application fields. The potential of multi-walled carbon nanotubes (MWNTs) as solid-phase extraction (SPE) adsorbents for the preconcentration of environmental pollutants has been investigated in recent years. The goal of this work was to investigate the feasibility of MWNTs used as SPE adsorbents to enrich dichlorodiphenyltrichloroethane (DDT) and its metabolites including 1,1-dichloro-2,2-bis-(4′-chlorophenyl)ethane (DDD) and 1,1-dichloro-2,2-bis-(4′-chlorophenyl)ethane (DDE) at trace level which are typical persistent organic pollutants in environment. Parameters that maybe influence the extraction efficiency such as the eluent volume, sample flow rate, sample pH and the sample volume were optimized in detail. The experimental results showed the excellent linear relationship between peak area and the concentration of DDT and its metabolites over the range of 0.2–60μgL−1, and the precisions (RSD) were 2.3–2.5% under the optimal conditions. The detection limits of proposed method could reach 4–13ngL−1 based on the ratio of chromatographic signal to base line noise (S/N=3). Satisfied results were achieved when the proposed method was applied to determine the four target compounds in realworld water samples with spiked recoveries over the range of 89.7–115.5%. All these facts indicated that MWCNTs as SPE packing materials coupled to HPLC was an excellent alternative for the routine analysis of DDT and its metabolites at trace level in environment.
