The development of a rapid, precise and accurate analytical method for the determination of methylmercury in tuna fish samples is described. The method is based on the use of isotope dilution GC/MS with electron impact ionization, a widespread technique in routine testing laboratories. A certified spike containing 202Hg-enriched methylmercury was used for the isotope dilution of the samples. After extraction of the methylmercury from the sample, methylmercury was propylated using sodium tetrapropyl borate in SPME vials and the analytes were sampled from the headspace for 15 min. For isotope measurements, the molecular ion (MePrHg+) was used in the SIM mode. Five molecular ions were monitored, corresponding to the 198Hg, 199Hg, 200Hg, 201Hg and 202Hg isotopes. The detection at masses corresponding to 198Hg was used to correct for m + 1 contributions of 13C from the organic groups attached to the mercury atom on the 199Hg, 200Hg, 201Hg and 202Hg masses with simple mathematical equations, and the concentration of methylmercury was calculated on the basis of the corrected 200Hg/202Hg isotope ratio. The 202Hg-enriched methylmercury spike was applied, with satisfactory results, to the determination of methylmercury in the certified reference material BCR 464. The method was successfully applied to the determination of methylmercury in tuna fish samples, and the obtained results were included in the CCQM-P39 interlaboratory exercise, organized by the Institute for Reference Materials and Measurements (IRMM, Geel, Belgium) with excellent agreement between our results and the average obtained by the other participants. The development of a rapid, precise and accurate speciation method for the simultaneous determination of mono-, di- and tributyltin in environmental samples is described. The method is based on using isotope dilution gas chromatography/mass spectrometry (GC/MS) with electron ionization, a widely used technique in routine testing laboratories. A mixed spike containing 119Sn-enriched monobutyltin (MBT), dibutyltin (DBT) and tributyltin (TBT) was used for the isotope dilution of the samples. Five molecular ions were monitored for each analyte, corresponding to the 116Sn, 117Sn, 118Sn, 119Sn and 120Sn isotopes. The detection at masses corresponding to 116Sn and 117Sn were used to correct for m + 1 and m + 2 contributions of 13C from the organic groups attached to the tin atom on the 118Sn, 119Sn and 120Sn masses with simple mathematical equations and the concentrations of the butyltin compounds were calculated based on the corrected 118Sn/119Sn and 120Sn/119Sn isotope ratios. The 119Sn-enriched multispecies spike was applied with satisfactory results to the simultaneous determination of MBT, DBT and TBT in three certified reference materials: two sediments, PACS-2 and BCR 646, and the mussel tissue CRM 477. The method was compared with a previously published GC/inductively coupled plasma MS isotope dilution procedure, developed in our laboratory, by injecting the same samples into both instruments. Comparable analytical results in terms of precision and accuracy are demonstrated for both atomic and molecular mass spectrometric detectors. Thus, reliable quantitative organotin speciation analysis can be achieved using the more widespread and inexpensive GC/MS instrument. Copyright © 2004 John Wiley & Sons, Ltd.

The coupling of reversed-phase high-performance liquid chromatography (RP-HPLC) with inductively coupled plasma mass spectrometry (ICP-MS) using post-column isotope dilution analysis (ID) was applied to the study of cadmium binding to standard rabbit liver metallothionein isoform 1 (MT-1). The analytical performance of three different ICP mass spectrometers, time of flight (TOF), double focussing (DF) and quadrupole (Q), was compared according to their performance for cadmium detection during chromatographic separation. Chromatographic conditions were optimised on a Vydac C8 VHP 259 RP column using a water–methanol gradient. Seven different sub-isoforms were distinguished by using this column coupled to a UV–Vis spectrophotometer, which greatly improved the resolution power exhibited by conventional anion-exchange chromatography. The ICP-DF-MS instrument was seriously affected by the methanol gradient and, thereafter, showed the worst performance in all cases. In contrast, the ICP-TOF-MS was able to resist up to 50% methanol with no serious changes in sensitivity or plasma stability. This instrument exhibited the lowest detection limits. The ICP-Q-MS provided results in between the other two instruments. In all cases, the use of post-column isotope dilution analysis was able to correct for matrix effects and signal drift, allowing for accurate determination of total MT concentration and the actual distribution of Cd-binding sub-isoforms.

The use of an inductively coupled plasma mass spectrometer (ICP-MS) with a time-of-flight (TOF) analyser is illustrated for the simultaneous determination of seven elements forming hydrides (As, Bi, Ge, Hg, Sb, Se and Sn) using flow injection (FI) as the sample introduction system. The chemical conditions for an adequate simultaneous volatile species generation as well as the plasma parameters for simultaneous determination were evaluated. The compromise hydride generation conditions were: 1% NaBH4 and 0.7 M HNO3 final concentration, while an Ar carrier gas flow rate of 1.1 l min−1 provided optimum results for the transport of the volatile species to the ICP-MS. Analytical figures of merit were evaluated under these compromise conditions and compared with those obtained by continuous nebulisation of the same elements in the ICP-MS(TOF). Detection limits in the low pg ml−1 region, linearity over more than five orders of magnitude and repeatability of 2–5% were achieved with the proposed methodology. Application of such a strategy was demonstrated by the simultaneous multi-elemental determination of As, Bi, Ge, Hg, Sb, Se and Sn in spiked urine samples with recoveries better than 80%. For Se, accurate results were only obtained by avoiding volatile species generation, demonstrating the presence of selenium species other than Se(IV) in human urine. The accuracy of the method was also checked by analysing a certified urine sample (NIST SRM 2670) by FI-ICP-MS(TOF) with satisfactory results.

A procedure for Stable Isotope Dilution Analysis in molecular Mass Spectrometry which does not require a methodological calibration graph and can be applied in combination with minimal labelling has been evaluated. This alternative approach is based on the determination of the molar fractions for each pure isotope pattern (natural abundance or labelled) contributing to the isotope pattern observed in the mixture of natural abundance and labelled molecules by multiple linear regression. Two labelled compounds, (13)C(1)-labelled or (13)C(6)-labelled phenol, were compared to study the influence of the number of (13)C atoms in the labelled molecule. The procedure was evaluated by comparing the results obtained for the determination of phenol in NIST 1584 CRM by GC-EI-MS using the classical isotope dilution calibration procedure and the new procedure based on multiple linear regression of isotope patterns without a calibration graph. The results obtained using the proposed procedure agreed well with both the certified values and those obtained using the classical Isotope Dilution Mass Spectrometry (IDMS) calibration procedures. For the evaluated procedure, a full uncertainty budget determination has been developed taking into account all uncertainty sources, including those derived from the uncertainties in the isotope patterns of the natural and labelled compounds. The measurements with the (13)C(1)-labelled phenol provided lower propagated uncertainties in comparison to the use of (13)C(6)-labelled phenol.

The fragmentation patterns of butyltin compounds (mono-, di-, and tributyltin) in an electron impact ion source were studied using an isotope pattern reconstruction algorithm with emphasis on isotope ratio measurements from molecular clusters. For this purpose, standards of natural tin isotope abundance and a (119)Sn-enriched mixture of the three compounds were both ethylated and propylated using sodium tetraalkylborates. The corresponding mass spectra of the various tetraalkyltin compounds prepared were obtained by GC/MS after their extraction with hexane. The results showed that pure interference-free molecular clusters were obtained only for certain R(3)Sn(+) ions where no isobaric overlap with R(2)SnH(+) ions occurred (e.g. BuEt(2)Sn(+) overlaps with Bu(2)SnH(+)). These ions are ideal candidates for accurate Sn isotope ratio measurements, while isotope pattern perturbing interferences are observed for other molecular fragments down to Sn(.)(+). Isotope pattern reconstruction algorithm thus can be used as an analytical tool to ensure the absence of molecular interferences--a requirement for accurate isotope ratio measurements from molecular clusters. The relevance of these studies for the determination of butyltin compounds in environmental samples by isotope dilution GC/MS is also discussed.

cited By 12; International audience; Six metrology institutes (NMIs) representing at the Comité International des Poids et Mesures (CIPM) 4 Member States of the Metre Convention and 2 international organisations, and 8 "expert" laboratories selected outside CIPM have compared their capabilities to quantitatively measure methylmercury (MeHg) in a prepared tuna material containing approximately 4.3 mg kg-1 Hg. This comparison was the object of the CIPM-Comité Consultatif pour la Quantité de Matière (CCQM) Pilot Study 39, organised by the Institute for Reference Materials and Measurements (IRMM), from the European Commission - Joint Research Centre. Beside the test material itself, a bottle of the BCR-464 tuna Certified Reference Material (CRM) and an ampoule of IRMM-670, a 202Hg isotope enriched MeHg candidate isotopic CRM, were distributed to all participants, who were free to apply the measurement strategy of their choice. Four, including 1 NMI, relied on external calibration or the method of standard additions, whereas the other 10 implemented an isotope dilution mass spectrometry (IDMS) approach and chose to use the IRMM-670 for their measurements. Alkaline digestion at room temperature (with manual shaking) or high temperature (under sonication, oven or hot plate conditions) was employed by most participants, with hydrochloric acid leaching the second most popular choice. Alkylation (4 phenylations, 4 ethylations and 3 propylations) in the aqueous phase was preferred by a large majority over butylation by the Grignard reaction. All participants were requested to estimate the uncertainty associated with their results and 9 out of 14 stated relative combined uncertainties below 6% (k = 2). Despite this apparent consensus, the perception of which factor caused the largest contribution to this estimation differed among participants because of the differences in the analytical methodologies deployed but also because of wide differences of the concepts of uncertainty estimation. The 'mixture mode' (MM) median, calculated also from the measurement uncertainties stated by the participants, was 1.967 ± 0.204 × 10-5 mol kg -1 (95% confidence). Twelve of the results were re-grouped within a range of less than 0.3 × 10-5 mol kg-1 (MM median = 1.967 ± 0.162 × 10-5 mol kg-1, 95% confidence): they nearly all (1 exception) overlapped with each other within k = 2 stated uncertainties. For the other 2 results the uncertainty seemed to have been particularly underestimated as they lay, respectively, at more than 20% above and less than -40% below the overall average. The relative standard deviation of the results of 9 laboratories out of the 10 that applied IDMS was about 2.6%. It can be assumed from the degree of equivalence shown by 12 out of 14 study participants that, at present, laboratories worldwide are potentially able to supply accurate results for MeHg in fish-type matrices (containing about 2 × 10-5 mol kg-1) within ± 10% uncertainty. This encouraging outcome permitted scheduling of a follow-up CCQM-K43 key comparison for a lower MeHg content level in salmon tissues. © The Royal Society of Chemistry 2005.
