Background: Microaggregates are suspected to protect soil organic carbon (SOC) from microbial decay, but its residence time is not well understood. Aims: We aimed at unraveling the relevance of microaggregates for C storage and testing the hypothesis that C in the interior of aggregates is older, compared to the exterior. Methods: We sampled soil under C3 vegetation and at a site where cropping shifted to C4 vegetation 36 years ago. We isolated free and macroaggregate‐occluded size fractions (250–53 µm) by wet sieving and ultrasound, manually isolated aggregates therefrom, and analyzed whether vegetation‐related differences in δ13C could be traced at the interior and exterior of microaggregate cross‐sections using elemental and laser ablation‐isotope ratio mass spectrometry. Results: Size fraction weights comprised <5% of microaggregates. Based on a source partitioning approach including C3‐ and C4‐derived C, we found mean residence times of SOC in occluded and free microaggregates of 62 and 105 years, respectively. Thus, C storage was longer than that in size fractions (35 years) and bulk soil (58 years). The small‐scale variability of δ13C within aggregate cross‐sections was considerable, both in C3 and C4 soil, yet without significant (p = 0.46) differences between interior and exterior locations. Conclusions: We conclude that microaggregates do not persist in an intact form in such a long‐term that systematic differences in δ13C patterns between exterior and interior parts can develop. Summary: Understanding P uptake in soil–plant systems requires suitable P tracers. The stable oxygen isotope ratio in phosphate (expressed as δ18OP) is an alternative to radioactive labelling, but the degree to which plants preserve the δ18OP value of the P source is unclear. We hypothesised that the source signal will be preserved in roots rather than shoots.In soil and hydroponic experiments with spring wheat (Triticum aestivum), we replaced irrigation water by 18O‐labelled water for up to 10 d. We extracted plant inorganic phosphates with trichloroacetic acid (TCA), assessed temporal dynamics of δ18OTCA‐P values after changing to 18O‐labelled water and combined the results with a mathematical model.Within 1 wk, full equilibration of δ18OTCA‐P values with the isotope value of the water in the growth medium occurred in shoots but not in roots. Model results further indicated that root δ18OTCA‐P values were affected by back transport of phosphate from shoots to roots, with a greater contribution of source P at higher temperatures when back transport was reduced.Root δ18OTCA‐P partially preserved the source signal, providing an indicator of P uptake sources. Liming is widely used to alleviate soil acidity in western and central Europe, but its role in the cycling of magnesium (Mg) in arable soil–plant systems is still ambiguous. Here, we systematically analysed Mg concentrations and the natural Mg stable isotope compositions (δ26Mg) of two Mg pools in soil profiles and a major crop (winter rye) in a long‐term German agricultural experimental field that has been managed with and without liming since 1923. The results showed that the δ26Mg signatures of the bulk soil Mg pool in the studied Albic Luvisol displayed limited variation with depth and between the liming treatments. In contrast, the exchangeable soil Mg pool exhibited an increase in δ26Mg values with depth down to 50 cm, which was more pronounced in the limed plots. We attributed this enrichment of light Mg isotopes in upper layers to the Mg addition from "Dolokorn 90" lime, as well as to the removal of heavy Mg isotopes by plant uptake. The subsequent use of a simple isotope‐mixing model suggested that only 25% of the remaining Mg in the soil exchangeable pool stemmed from the last liming practice. The other parts of the exchangeable soil Mg pool had either interacted with the bulk soil matrix or had been utilized by the plants. Almost 100 years of liming enhanced Mg uptake by the vegetation, probably via elevated Mg contents in the grain, and reflected by the stronger depletion of heavy Mg isotopes in the soil exchangeable Mg pool relative to non‐limed plots. Whole winter rye plants were enriched in heavy Mg isotopes but they displayed similar Mg isotope compositions among plant organs in all plots, indicating identical Mg uptake and translocation strategies in the different trials. Tracing the stable isotope compositions of soil and plant Mg thus opens novel opportunities to evaluate soil management impacts on the cycling and fate of this essential nutrient in agricultural systems. Aims: Magnesium (Mg) deficiency is detrimental to plant growth. However, how plants respond to Mg deficiency via regulation of Mg uptake and allocation is yet not fully understood. In this study, we tested whether Mg isotope compositions (δ26Mg) associated with Mg mass balance of the plants could be used as an indicator to trace Mg uptake and subsequent translocation processes under sufficient and low-Mg supply conditions. We aimed at using stable isotope fractionation as a novel proxy for nutrient uptake and cycling in plants. Methods: We grew wheat plants (Triticum aestivum) in a greenhouse under control (1 mM Mg) and low-Mg supply (0.05 mM Mg) conditions, respectively. The Mg concentrations and isotope compositions in roots, stems, leaves and spikes/grains at different growth stages were analyzed. Results: Wheat plants were systematically enriched in heavy Mg isotopes relative to the nutrient solution regardless of Mg supply conditions. With crop growth, the δ26Mg of the whole plants, as well as each plant organ, gradually shifted towards higher values in the control. However, the δ26Mg value of the whole plants in the low-Mg supply did not vary significantly. In addition, the wheat stems and spikes showed continuous enrichment of lighter Mg isotopes in the low-Mg supply than those in the control. Conclusions: As reflected from Mg isotope compositions, the Mg supply in the growth media could affect the Mg uptake and subsequent translocation processes in plants. Changes in δ26Mg indicated that wheat plants likely regulated their Mg uptake strategy by switching between active and passive pathways during their life cycle. When Mg supply was low, a more negative δ26Mg value of the spikes suggested a potentially enhanced remobilization of Mg from leaves to spikes. Microaggregates (<250 µm) are key structural subunits of soils. However, their formation processes, rates, and transformation with time are poorly understood. We took advantage of multiple isotope labelling of potential organic gluing agents and inorganic building units to unravel their role in soil aggregation processes being initiated with and without plant growth. We added 13C-labelled extracellular polymeric substances (EPS), 15N-labelled bacteria, 57Fe-labelled goethite, and 29Si-labelled montmorillonite to fine soil <250 µm of an Ap horizon from a Stagnic Luvisol, which was planted with Festuca heteromalla or kept bare in a climate chamber. Samples were taken after 4, 12, and 30 weeks, and separated into free (f) and occluded (o) microaggregates of different sizes (<20 µm, 53–20 µm, 250–53 µm), and in stable macroaggregates (>250 µm) that resisted 60 J mL−1 ultrasonic dispersion. Afterwards, we assessed the C, N, Fe, and Si stable isotope composition in each size fraction. After four weeks we found a rapid build-up of stable macroaggregates comprising almost 50 % of soil mass in the treatment with plants and respective soil rooting, but only 5 % when plants were absent. The formation of these stable macroaggregates proceeded with time. Soil organic carbon (SOC) contents were elevated by 15 % in the large macroaggregates induced by plant growth. However, the recovery of EPS-derived 13C was below 20 % after 4 weeks, indicating rapid turnover in treatments both with and without plants. The remaining EPS-derived C was mainly found in macroaggregates when plants were present and in the occluded small microaggregates (<20 µm) when plants were absent. The excess of bacterial 15N closely followed the pattern of EPS-derived 13C (R2 = 0.72). In contrast to the organic gluing agents, the goethite-57Fe and montmorillonite-29Si were relatively equally distributed across all size fractions. Overall, microaggregates were formed within weeks. Roots enforced this process by stabilizing microaggregates within stable macroaggregates. As time proceeded the labelled organic components decomposed, while the labelled secondary oxides and clay minerals increasingly contributed to aggregate stabilization and turnover at the scale of months and beyond. The impact of termites on nutrient cycling and tropical soil formation depends on their feeding habits and related material transformation. The identification of food sources, however, is difficult, because they are variable and changed by termite activity and nest construction. Here, we related the sources and alteration of organic matter in nests from seven different termite genera and feeding habits in the Terra Firme rainforests to the properties of potential food sources soil, wood, and microepiphytes. Chemical analyses comprised isotopic composition of C and N, cellulosic (CPS), non-cellulosic (NCPS), and N-containing saccharides, and molecular composition screening using pyrolysis-field ionization mass spectrometry (Py-FIMS). The isotopic analysis revealed higher soil δ13C (-27.4‰) and δ15N (6.6‰) values in nests of wood feeding Nasutitermes and Cornitermes than in wood samples (δ13C = -29.1‰, δ15N = 3.4‰), reflecting stable-isotope enrichment with organic matter alterations during or after nest construction. This result was confirmed by elevated NCPS:CPS ratios, indicating a preferential cellulose decomposition in the nests. High portions of muramic acid (MurAc) pointed to the participation of bacteria in the transformation processes. Non-metric multidimensional scaling (MDS) revealed increasing geophagy in the sequence Termes < Embiratermes < Anoplotermes and increasing xylophagy for Cornitermes < Nasutitermes., and that the nest material of Constrictotermes was similar to the microepiphytes sample, confirming the report that Constrictotermes belongs to the microepiphyte-feeders. We therewith document that nest chemistry of rainforest termites shows variations and evidence of modification by microbial processes, but nevertheless it primarily reflects the trophic niches of the constructors. Leaf area index ( LAI), a measure of canopy density, is a key variable for modelling and understanding primary productivity, and also water use and energy exchange in forest ecosystems. However, LAI varies considerably with phenology and disturbance patterns, so alternative approaches to quantifying stand-level processes should be considered. The carbon isotope composition of soil organic matter (δ13 CSOM) provides a time-integrated, productivity-weighted measure of physiological and stand-level processes, reflecting biomass deposition from seasonal to decadal time scales., Our primary aim was to explore how well LAI correlates with δ13 CSOM across biomes., Using a global data set spanning large environmental gradients in tropical, temperate and boreal forest and woodland, we assess the strength of the correlation between LAI and δ13 CSOM; we also assess climatic variables derived from the World Clim database., We found that LAI was strongly correlated with δ13 CSOM, but was also correlated with Mean Temperature of the Wettest Quarter, Mean Precipitation of Warmest Quarter and Annual Solar Radiation across and within biomes., Synthesis. Our results demonstrate that δ13 CSOM values can provide spatially explicit estimates of leaf area index ( LAI) and could therefore serve as a surrogate for productivity and water use. While δ13 CSOM has traditionally been used to reconstruct the relative abundance of C3 versus C4 species, the results of this study demonstrate that within stable C3- or C4-dominated biomes, δ13 CSOM can provide additional insights. Reseeding grasses into degraded cropland and using it as permanent secondary pasture may combat desertification and promote sequestration of soil organic carbon (SOC). Our objective was to assess the contribution of lignin and grass-derived carbon to the rates of SOC accrual upon such land-use change. We sampled secondary pastures of different age (1 to 31 years old), established on former degraded cropland at three agro-ecosystems in the semi-arid Highveld of South Africa; old degraded croplands and primary grassland used for cattle grazing served as controls. Topsoil samples (0–5 and 5–10 cm) were characterized by their natural 13 C and 15 N abundances and by their contents of lignin-derived phenols (VSC) after alkaline CuO oxidation. The results showed that the δ 13 C signature in secondary pasture soil was variable and not a sensitive indicator of land restoration in these sites. Soil δ 15 N values declined from 8 to 6‰ with increased duration of secondary pasture use and approached the values of the primary grassland, suggesting that N-use efficiency improved. The VSC contents increased in the surface soils and reached a new steady-state equilibrium after 38 years. Lignin sequestration was accelerated relative to total SOC in the 0–5 cm soil depth but delayed in the 5–10 cm soil depth, reflecting different amounts of depth-dependent inputs of plant debris into the pastures. Even if absolute VSC contents did not reach the respective concentrations in the primary grassland, the contribution of VSC to total SOC (g VSC kg − 1 C) was similar to that of the primary grassland. Decreasing ratios of syringyl to vanillyl structural units reflected that after 8–19 years all remaining lignin derived primarily from the new grassland vegetation. Lignin enhances SOC accumulation only in the very surface soil (0–5 cm) but not below this depth, rendering the restoration of subsoil C stocks difficult in these ecosystems. [ABSTRACT FROM AUTHOR]

Agricultural production in Central Europe increasingly suffers from extreme drought events. Improving root access to nutrient and water resources in the subsoil below the plow layer is a potential option to maintain productivity during dry summers. Here, we tested a strip-wise subsoil amelioration method that combines subsoil loosening with organic matter incorporation into the subsoil (biowaste or green waste compost) and compared it with a treatment of only subsoil loosening and a non-ameliorated control. A field experiment with randomized block design was conducted on a Luvisol with an argic horizon (Bt), with a rotation of spring barley and winter wheat. In the first two years after amelioration, we monitored soil physico-chemical parameters, microbial biomass, and shoot and root growth at anthesis as well as harvested grain yield and quality. Subsoil loosening with organic matter incorporation significantly decreased soil bulk density at the depth of compost incorporation when biowaste compost was used, but not when green waste compost had been incorporated. Nutrient stocks, nutrient availability and microbial biomass were not consistently affected by the subsoil amelioration. Nevertheless, the incorporation of organic material, especially biowaste compost, significantly increased root growth into the subsoil and subsequently significantly enhanced crop nutrient uptake, biomass and grain yield production. Green waste compost incorporation had less pronounced effects, with an increase in grain yield only in the second year after amelioration. Differences in crop development could not be explained by any single soil parameter, suggesting that it was rather a combined effect of loosened subsoil and better supply of subsoil resources that resulted in an increase in subsoil root length density and subsequently led to better crop performance. • Only loosening of subsoil did not affect soil bulk density and nutrient availability. • Root growth into subsoil was enhanced after loosening with compost addition. • Also yields after subsoil loosening were increased with compost addition. A sustainable use of soil resources is urgently required to cope with the increasing demand for agricultural products during climate change. To inspire farmers on new soil cultivation methods like subsoil management requires not only yield measurements but also nutrient use efficiency measurements for which analytical tools are still missing. Stable isotopes of the macronutrient magnesium (Mg) are a potential novel subsoil management evaluation tool in agronomy and soil/plant sciences because its isotope ratios shift considerably during Mg uptake by crops. The feasibility of Mg stable isotopes was first demonstrated conceptually by simulating subsoil management on soils with low, middle, and high inventories of bioavailable Mg and crop plants typically cultivated in Germany. This simulation showed that the magnitude of Mg isotope shifts among crops and the exchangeable fraction of Mg in soil is resolvable from the long-term external precision of Mg isotope analyses only if three conditions are met. First, the crop uptake-related Mg isotope fractionation factor should be at the upper end of hitherto published fractionation factors. Second, a high Mg uptake flux of crop plants (e.g., sugar beets) is matched by a low Mg supply from the exchangeable fraction in soil (e.g., sandy soils). Third, subsoil management causes a considerable deepening of the rooting system (e.g., flipping the topsoil root cluster below 30 cm depth). If these conditions are met, Mg stable isotopes can be used in a qualitative manner to identify the main Mg uptake depth, and in a quantitative manner by calculating the Mg use efficiency, defined here as the ratio of Mg uptake versus Mg supply, solely from Mg isotope ratios. This concept was tested for Alfisols on field trials by conducting deep loosening with and without the incorporation of compost. Magnesium isotope shifts in crops and the exchangeable fraction of Mg in soil were mostly unresolvable from the long-term external precision of Mg isotope analyses, which positively tested the Mg isotope concept for well nurtured soils. However, systematic Mg isotope shifts among bulk crops cultivated on and beside a melioration strip were found and attributed to the uplift of isotopically distinct compost-derived Mg on the melioration strip and root restricting layers beside the melioration strip. Given that the Mg isotope composition of the exchangeable fraction barely varies with depth, field-based crop uptake-related 'apparent' Mg isotope fractionation factors of winter wheat and spring barley could be determined, which differed from one another (Δ26Mg wheat-rem. exch. = 0.63 ± 0.05‰, Δ26Mg barley-rem. exch. = 0.37 ± 0.12‰). Nonetheless, the quantitative approach of Mg isotopes was violated when calcareous fertilizer was applied to the field as differences in the isotope-derived Mg use efficiency could be attributed to the uneven distribution of lime-derived Mg with depth. Using Mg stable isotopes as a new geochemical routine for agronomy and soil/plant sciences requires future work focussing on isotope fractionation factors related to crop uptake and intra-plant translocation of Mg – which may depend on species, environmental conditions, and nutrient status – to allow minimally invasive sampling of the soil-plant system and to reduce sample sets. • A conceptual Mg isotope model was established and positively tested in the field. • Mg stable isotopes are a powerful novel tool for subsoil management evaluation. • Mg use efficiency of crops can solely be quantified from Mg stable isotopes. • Lime application limits Mg use efficiency determination solely from Mg isotopes. • A criteria catalogue ensures a successful application of Mg isotopes in agronomy.
