Staphylococcus aureus (Gram-positive) and Pseudomonas aeruginosa (Gram-negative) bacteria represent major infectious threats in the hospital environment due to their wide distribution, opportunistic behavior, and increasing antibiotic resistance. This study reports on the deposition of polyvinylpyrrolidone/antibiotic/isoflavonoid thin films by the matrix-assisted pulsed laser evaporation (MAPLE) method as anti-adhesion barrier coatings, on biomedical surfaces for improved resistance to microbial colonization. The thin films were characterized by Fourier transform infrared spectroscopy, infrared microscopy, and scanning electron microscopy. In vitro biological assay tests were performed to evaluate the influence of the thin films on the development of biofilms formed by Gram-positive and Gram-negative bacterial strains. In vitro biocompatibility tests were assessed on human endothelial cells examined for up to five days of incubation, via qualitative and quantitative methods. The results of this study revealed that the laser-fabricated coatings are biocompatible and resistant to microbial colonization and biofilm formation, making them successful candidates for biomedical devices and contact surfaces that would otherwise be amenable to contact transmission.

This work reports the non-surfactant templated synthesis and characterization of a new tyrosine-silica/antibiotics (TyR-SiO2/ATBs) nanocomposite, as well as both in vitro and in vivo cytotoxicity and antimicrobial activity against the microbial pathogen Staphylococcus aureus. The in vitro microbiological tests proved that the obtained nanobiostructure significantly enhance the antimicrobial activity of three commonly used antibiotics against S. aureus (i.e. erythromycin (ERI), gentamicin (GEN), and cloxacillin (CLO)) as revealed by the increased diameters of the growth inhibition zones and the decreased minimal inhibitory concentration values, as well as by the inhibitory effect of sub-lethal antibiotic concentrations on the ability of the respective pathogenic strains to adhere and colonize different substrata. These results, correlated with the lack of toxicity against mesenchymal stem cells along with an appropriate in vivo biodistribution highlight the promising therapeutic potential of this carrier that allows a decrease of the required active doses while significantly lessening the harmful side effects of the medication on the host organism., (Copyright © 2016 Elsevier B.V. In recent years, severe infections caused by the Gram positive pathogen Staphylococcus aureus have emerged both in community and clinical settings. The increased infection rates are explained by the versatility and resistance of this pathogen, which produces a wide arsenal of virulence factors, organize within difficult to eradicate biofilms and displays various resistance genes, leading current antibiotic therapies ineffective. Nanotechnological progress highlights the impact of magnetic nanoparticles in the current and future diagnosis and therapy procedures of the infectious diseases. These nanostructures could be used as efficient carriers for many natural and synthetic antimicrobials, which may be further utilized for the development of soluble anti-Staphylococcal formulations and improved surfaces and coatings of different uses, optimized to reduce attachment and biofilm formation.

Food safety and quality are major concerns in the food industry. Despite numerous studies, polyethylene remains one of the most used materials for packaging due to industry reluctance to invest in new technologies and equipment. Therefore, modifications to the current materials are easier to implement than adopting whole new solutions. Antibacterial activity can be induced in low-density polyethylene films only by adding antimicrobial agents. ZnO nanoparticles are well known for their strong antimicrobial activity, coupled with low toxicity and UV shielding capability. These characteristics recommend ZnO for the food industry. By incorporating such safe and dependable antimicrobial agents in the polyethylene matrix, we have obtained composite films able to inhibit microorganisms' growth that can be used as packaging materials. Here we report the obtaining of highly homogenous composite films with up to 5% ZnO by a melt mixing process at 150 °C for 10 min. The composite films present good transparency in the visible domain, permitting consumers to visualize the food, but have good UV barrier properties. The composite films exhibit good antimicrobial and antibiofilm activity from the lowest ZnO composition (1%), against both Gram-positive and Gram-negative bacterial strains. The homogenous dispersion of ZnO nanoparticles into the polyethylene matrix was assessed by Fourier transform infrared microscopy and scanning electron microscopy. The optimal mechanical barrier properties were obtained for composition with 3% ZnO. The thermal analysis indicates that the addition of ZnO nanoparticles has increased thermal stability by more than 100 °C. The UV-Vis spectra indicate a low transmittance in the UV domain, lower than 5%, making the films suitable for blocking photo-oxidation processes. Nowadays, the world is facing a serious crisis represented by the rapid emergence of resistant bacteria, which jeopardizes the efficacy of antibiotics. This crisis has been attributed to the overuse and misuse of antibiotics, as well as the cessation of new drug production by the pharmaceutical industry. Therefore, bacterial strains with resistance to multiple antibiotic classes have appeared, such as Staphylococcus aureus, Acinetobacter spp. and Pseudomonas aeruginosa. This review aims to provide an updated summary of the current approach to the treatment of infections due to resistant microorganisms, with a focus on the application of the antimicrobial effects of inorganic nanoparticles in combination with collagen to promote wound healing. The aim of this work was to obtain an antimicrobial coating (NanoAg) for polyester-nylon wound dressings (WDs) for reducing the risk of exogenous wound related infections. The as-prepared NanoAg-WDs were characterized by XRD (X-ray Diffraction), SEM (Scanning Electron Microscopy), TEM (Transmission Electron Microscopy), SAED (Selected Area Electron Diffraction) and IRM (InfraRed Microscopy). Biological characterization consisted of in vitro evaluation of the interaction with fibroblast cell cultures and in vivo biodistribution studies of AgNPs on mice models. Then, specimens of commercial WDs were immersed in a glucose and NaOH solution of silver nanoparticles, followed by the subsequent dropwise addition of AgNO3 solution. The antimicrobial efficiency of the NanoAg-WDs was assessed by in vitro qualitative and quantitative analyses on Staphylococcus aureus and Pseudomonas aeruginosa strains. The in vitro and in vivo studies demonstrated that the tested nanoparticles utilized to coat WDs have a good biocompatibility, allowing the normal development of cultured human cells and revealing a normal biodistribution within a mouse model, without toxic effects. The modified and viable cells count analyses proved that the modified WDs exhibit an improved inhibitory activity of microbial colonization, attachment and biofilm growth. The reported data recommend this type of coatings to obtain modified WDs with antibacterial properties, able to prevent the exogenous microbial contamination of the wound tissue, colonization and further biofilm development. We report on the synthesis and evaluation of biopolymeric spheres of poly(lactide-co-glycolide) containing different amounts of magnetite nanoparticles and Ibuprofen (PLGA-Fe3O4-IBUP), but also chitosan (PLGA-CS-Fe3O4-IBUP), to be considered as drug delivery systems. Besides morphological, structural, and compositional characterizations, the PLGA-Fe3O4-IBUP composite microspheres were subjected to drug release studies, performed both under biomimetically-simulated dynamic conditions and under external radiofrequency magnetic fields. The experimental data resulted by performing the drug release studies evidenced that PLGA-Fe3O4-IBUP microspheres with the lowest contents of Fe3O4 nanoparticles are optimal candidates for triggered drug release under external stimulation related to hyperthermia effect. The as-selected microspheres and their chitosan-containing counterparts were biologically assessed on macrophage cultures, being evaluated as biocompatible and bioactive materials that are able to promote cellular adhesion and proliferation. The composite biopolymeric spheres resulted in inhibited microbial growth and biofilm formation, as assessed against Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans microbial strains. Significantly improved antimicrobial effects were reported in the case of chitosan-containing biomaterials, regardless of the microorganisms' type. This study reports on the successful deposition of γ-cyclodextrin/usnic acid (γCD/UA) thin film by Matrix Assisted Pulsed Laser Evaporation (MAPLE) as anti-adherent coating on medical surfaces against microbial colonization. The obtained results demonstrate that these bioactive thin films inhibit Staphylococcus aureus biofilm formation at all stages, starting with their initiation. The antibiofilm effect was constant along the bacterial incubation time. Furthermore, the γCD/UA coatings show a great biocompatibility which means that this material is suitable for the development of modern medical devices with antimicrobial properties. Bacterial cellulose-silica hybrid composites were prepared starting from wet bacterial cellulose (BC) membranes using Stöber reaction. The structure and surface morphology of hybrid composites were examined by FTIR and SEM. The SEM pictures revealed that the silica particles are attached to BC fibrils and are well dispersed in the BC matrix. The influence of silica particles upon BC crystallinity was studied using XRD analysis. Thermogravimetric (TG) analysis showed that the composites are stable up to 300 °C. A Plackett-Burman design was applied in order to investigate the influence of process parameters upon silica particle sizes and silica content of BC-silica composites. The statistical model predicted that it is possible for silica particles size to vary the synthesis parameters in order to obtain silica particles deposed on BC membranes in the range from 34.5 to 500 nm, the significant parameters being ammonia concentration, reaction time and temperature. The silica content also varies depending on process parameters, the statistical model predicting that the most influential parameters are water-tetraethoxysilane (TEOS) ratio and reaction temperature. The antimicrobial behavior on Staphylococcus aureus of BC-silica composites functionalized with usnic acid (UA) was also studied, in order to create improved surfaces with antiadherence and anti-biofilm properties.
