Laser speckle contrast imaging (LSCI) enables video rate imaging of blood flow. However, its relation to tissue blood perfusion is nonlinear and depends strongly on exposure time. By contrast, the perfusion estimate from the slower laser Doppler flowmetry (LDF) technique has a relationship to blood perfusion that is better understood. Multiexposure LSCI (MELSCI) enables a perfusion estimate closer to the actual perfusion than that using a single exposure time. We present and evaluate a method that utilizes contrasts from seven exposure times between 1 and 64 ms to calculate a perfusion estimate that resembles the perfusion estimate from LDF. The method is based on artificial neural networks (ANN) for fast and accurate processing of MELSCI contrasts to perfusion. The networks are trained using modeling of Doppler histograms and speckle contrasts from tissue models. The importance of accounting for noise is demonstrated. Results show that by using ANN, MELSCI data can be processed to LDF perfusion with high accuracy, with a correlation coefficient R = 1.000 for noise-free data, R = 0.993 when a moderate degree of noise is present, and R = 0.995 for in vivo data from an occlusion-release experiment.

Phthalocyanine-BODIPY dye (BODIPY = boron dipyrromethene) was synthesized, fully characterized, and used for molecular recognition of CYFRA 21-1, a lung cancer biomarker, from whole blood samples. Thin films of three magnesium oxides ((MgO) n , where n = 8, 9, or 10)) were deposited on a paper substrate, and they were immersed in a solution of phthalocyanine-BODIPY dye (1.17 × 10−3 mol/L) for the design of stochastic sensors. Limits of determination of picograms per milliliter magnitude order were recorded for the proposed stochastic sensors. CYFRA 21-1 was reliably identified and determined with recoveries higher than 95% and RSD lower than 1% in whole blood samples.

The analysis of the dynamic behaviour of genome-scale models of metabolism (GEMs) currently presents considerable challenges because of the difficulties of simulating such large and complex networks. Bacterial GEMs can comprise about 5000 reactions and metabolites, and encode a huge variety of growth conditions; such models cannot be used without sophisticated tool support. This article is intended to aid modellers, both specialist and non-specialist in computerized methods, to identify and apply a suitable combination of tools for the dynamic behaviour analysis of large-scale metabolic designs. We describe a methodology and related workflow based on publicly available tools to profile and analyse whole-genome-scale biochemical models. We use an efficient approximative stochastic simulation method to overcome problems associated with the dynamic simulation of GEMs. In addition, we apply simulative model checking using temporal logic property libraries, clustering and data analysis, over time series of reaction rates and metabolite concentrations. We extend this to consider the evolution of reaction-oriented properties of subnets over time, including dead subnets and functional subsystems. This enables the generation of abstract views of the behaviour of these models, which can be large—up to whole genome in size—and therefore impractical to analyse informally by eye. We demonstrate our methodology by applying it to a reduced model of the whole-genome metabolism of Escherichia coli K-12 under different growth conditions. The overall context of our work is in the area of model-based design methods for metabolic engineering and synthetic biology.

In this study, we investigated the statistical relationship between nanostructure variations of carbon nanotube buckypaper-polymer (BPP) composites and their resulting elastic properties. A statistical simulation was developed to predict the elastic properties of a single-layer BPP lamina and extrapolated to the resultant bulk composite part. The stochastic characteristics of BPP composite nanostructure were quantified from experimental observations and used to generate the input for each simulation set performed. The Mori-Tanaka method was used to calculate the stiffness tensor within the buckypaper-polymer region, and a Monte-Carlo simulation was applied to generate the probability distribution for the effective stiffness tensor within each BPP lamina. Classical laminate theory was then employed to predict the effective elastic response for a multi-layered BPP composite laminate. The theoretical predictions were compared with experimental data, and the resulting trends for the effective tensile modulus between experimental and theoretical corresponded well with each other.

This article proposes an Enhanced Memetic Differential Evolution (EMDE) for designing digital filters which aim at detecting defects of the paper produced during an industrial process. Defect detection is handled by means of two Gabor filters and their design is performed by the EMDE. The EMDE is a novel adaptive evolutionary algorithm which combines the powerful explorative features of Differential Evolution with the exploitative features of three local search algorithms employing different pivot rules and neighborhood generating functions. These local search algorithms are the Hooke Jeeves Algorithm, a Stochastic Local Search, and Simulated Annealing. The local search algorithms are adaptively coordinated by means of a control parameter that measures fitness distribution among individuals of the population and a novel probabilistic scheme. Numerical results confirm that Differential Evolution is an efficient evolutionary framework for the image processing problem under investigation and show that the EMDE performs well. As a matter of fact, the application of the EMDE leads to a design of an efficiently tailored filter. A comparison with various popular metaheuristics proves the effectiveness of the EMDE in terms of convergence speed, stagnation prevention, and capability in detecting solutions having high performance.

This paper presents a biologically inspired, hardware-realisable spiking neuron model, which we call the Temporal Noisy-Leaky Integrator (TNLI). The dynamic applications of the model as well as its applications in Computational Neuroscience are demonstrated and a learning algorithm based on postsynaptic delays is proposed. The TNLI incorporates temporal dynamics at the neuron level by modelling both the temporal summation of dendritic postsynaptic currents which have controlled delay and duration and the decay of the somatic potential due to its membrane leak. Moreover, the TNLI models the stochastic neurotransmitter release by real neuron synapses (with probabilistic RAMs at each input) and the firing times including the refractory period and action potential repolarisation. The temporal features of the TNLI make it suitable for use in dynamic time-dependent tasks like its application as a motion and velocity detector system presented in this paper. This is done by modelling the experimental velocity selectivity curve of the motion sensitive H1 neuron of the visual system of the fly. This application of the TNLI indicates its potential applications in artificial vision systems for robots. It is also demonstrated that Hebbian-based learning can be applied in the TNLI for postsynaptic delay training based on coincidence detection, in such a way that an arbitrary temporal pattern can be detected and recognised. The paper also demonstrates that the TNLI can be used to control the firing variability through inhibition with 80% inhibition to concurrent excitation, firing at high rates is nearly consistent with a Poisson-type firing variability observed in cortical neurons. It is also shown with the TNLI, that the gain of the neuron (slope of its transfer function) can be controlled by the balance between inhibition and excitation, the gain being a decreasing function of the proportion of inhibitory inputs. Finally, in the case of perfect balance between inhibition and excitation, i.e. where the average input current is zero, the neuron can still fire as a result of membrane potential fluctuations. The firing rate is then determined by the average input firing rate. Overall this work illustrates how a hardware-realisable neuron model can capitalise on the unique computational capabilities of biological neurons. © 2002 Elsevier Science Ltd.
