Cystoisospora (syn. Isospora) suis is the causative agent of neonatal porcine coccidiosis and one of the main causes of diarrhoea in suckling piglets worldwide. Infection with porcine cytomegalovirus (PCMV, Suid herpesvirus 2) causes inclusion body rhinitis in pigs. In a Swiss pig herd (n=2 boars, 7 sows, 2 gilts, 18 finishing pigs, 30 fattening pigs, 54 suckling piglets), an outbreak of PCMV infection with high morbidity in all age categories, characterized by fever, anorexia, reduced general condition, respiratory signs and increased piglet mortality, was diagnosed by histopathology and molecular methods. Five fattening pigs (age~17weeks) additionally showed diarrhoea, not typical for PCMV infections, and one fattener had to be euthanized due to poor condition. Histopathologically, severe fibrinopurulent jejunoileitis with extensive atrophy and fusion of intestinal villi, loss of goblet cells and crypt abscesses associated to C. suis infection were present. In the liver, herpesvirus intranuclear inclusion bodies were observed and PCMV was confirmed by PCR/sequencing. No further infectious causes of diarrhoea (i.e. Rotavirus A; TGEV; PEDV; PCV-2; enterotoxigenic Escherichia coli or Lawsonia intracellularis) were detected in the euthanized fattener. Coproscopically, C. suis oocysts were identified in the faeces from further fatteners with diarrhoea. While C. suis usually produces disease only in suckling piglets, its association with severe intestinal lesions and diarrhoea in ~17-week-old fatteners was surprising. It is supposed that the underlying PCMV infection might have contributed to the presentation of clinical cystoisosporosis in fattening pigs. In order to estimate the diversity, clinical involvement and zoonotic potential of parasites in pigs submitted for diagnosis to the PathoPig project of the Swiss Federal Food Safety and Veterinary Office, faeces (n=125) from suckling piglets (n=39), weaners (n=60) and piglets beginning fattening (n=26) from 74 Swiss farms were examined by 3 coproscopical methods (i.e. sedimentation/zinc chloride-flotation; SAFC and Ziehl-Neelsen staining). Samples microscopically positive for Cryptosporidium were further tested by PCR/sequencing for species assessment. The most frequently detected parasite was Balantidium coli, a facultative pathogenic ciliate with zoonotic potential, in 5.1, 36.7 and 50.0% of suckling, weaners and fatteners and 43.2% of farms; however, no association with disease was observed. Isospora (syn. Cystoisospora) suis infections were detected in 13.3 and 11.1% of suckling piglets with and without diarrhoea, and in 10.0 and 13.3% of weaners and fatteners with diarrhoea, respectively, and were significant associated with emaciation. Cryptosporidium infections were detected in 10.3, 15.0 and 19.2% of sucklings, weaners and fatteners, respectively, and in 18.9% of the farms. Interestingly, two age-related species were identified: C. suis in younger piglets (2 to 6weeks) and C. scrofarum in older ones (6 to 17weeks). None of the pigs infected with C. scrofarum (n=8), but 3 of 4 piglets infected with C. suis (co-infection with I. suis in 2 cases) had diarrhoea. The zoonotic species C. parvum was not detected, nevertheless, sporadic cases of human infection with the porcine-adapted species have been reported. Isospora suis is an intestinal protozoan parasite in pigs. The 2-3 weeks old piglets are most often infected by I. suis because their immune system is not fully developed. The infection exhibits clinical features such as diarrhea and dehydration and seriously affects the economic interests of farmers. The traditional method of identifying I. suis relies on the detection of fecal oocysts, which depends heavily on the accumulation of experience. Thus, missed detection, and false alarms often occur during detection. With the development of molecular-based detection methods, development of a simple, convenient and more sensitive method for the detection of I. suis is an urgent need. In this study, based on the 18S rRNA gene sequence, a fluorescence -based real-time loop-mediated isothermal amplification (LAMP) assay was established for the detection of I. suis. The results showed that the assay is highly specific and sensitive, with a detection limit of 2.74 × 10(2) copies/μL recombinant plasmid of I. suis, corresponding to 1 fg/μL plasmid when converted to DNA concentration. The sensitivity is about 100 times higher than conventional PCR. Additionally, DNA extracted from a certain number of oocysts was used for detection, and it showed that the LAMP assay had a detection limit of 5 oocysts, lower than that of 13 oocysts of conventional PCR. The established LAMP assay overcomes the shortage of the traditional microscopy-based method, and provides a valuable way for molecular detection of I. suis., (Copyright © 2016 Elsevier Inc. Background: Diarrhea in piglets directly affects commercial swine production. The disease results from the interaction of pathogens with the host immune system and is also affected by management procedures. Several pathogenic agents such as Campylobacter spp., Clostridium perfringens, Escherichia coli, Salmonella spp., group A rotavirus (RV-A), coronaviruses (transmissible gastroenteritis virus; porcine epidemic diarrhea virus), as well as nematode and protozoan parasites, can be associated with disease cases., Results: All bacterial, viral, protozoan, and parasitic agents here investigated, with the exception of Salmonella spp. as well as both coronaviruses, were detected in varying proportions in piglet fecal samples, and positive animals were equally distributed between case and control groups. A statistically significant difference between case and control groups was found only for Cystoisospora suis (p = 0.034) and Eimeria spp. (p = 0.047). When co-infections were evaluated, a statistically significant difference was found only for C. perfringens β2 and C. suis (p = 0.014)., Conclusions: The presence of pathogens in piglets alone does not determine the occurrence of diarrhea episodes. Thus, the indiscriminate use of antibiotic and anthelminthic medication should be re-evaluated. This study also reinforces the importance of laboratory diagnosis and correct interpretation of results as well as the relevance of control and prophylactic measures.

To determine the prevalence of coccidian infection in suckling piglets in China, fecal samples from 779 litters of suckling piglets were collected on 80 different farms in 17 provinces from September 2009 to December 2010. These samples were examined through saturated saline flotation technique. The prevalences of coccidian infection ranged from 0 to 32.5% among different provinces and the average was 16.7% (130/779). The highest prevalence of 19.9% (69/346) was found in 8-14 day-old litters of suckling piglets. Seven coccidian species were detected in the positive litters of suckling piglets, including Isospora suis (63.9%), Eimeria debliecki (46.9%), Eimeria polita (19.2%), Eimeria suis (20.8%), Eimeria perminuta (13.9%), Eimeria scabra (4.6%), and Eimeria yanglingensis (1.5%). 55.4% of the positive litters of suckling piglet infected more than one coccidian species. Background: Piglet isosporosis is one of the most common parasitic diseases in modern pig production. To prevent clinical disease, prophylactic treatment of piglets with toltrazuril (BAYCOX® 5%, Bayer HealthCare, Animal Health, Monheim, Germany) is widely practiced in the past 20 years. There are only very few reports documenting the likely effect of managerial practices, such as hygiene measures, all-in-all-out management of farrowing facilities and piglet manipulations, and/or farm-specific environment - i.e. design and materials of the farrowing pen and room - in the risk of disease occurrence and transmission. Therefore, in this cross-sectional study, we identified litter- and herd-level factors associated with the odds and the level of Isospora suis oocyst excretion in nursing piglets of Greek farrow-to-finish pig herds. Faecal samples were collected from 314 liters of 55 randomly selected herds. Oocyst counts were determined by a modified McMaster technique and possible risk-factor data were collected through a questionnaire. In the analysis, we employed a two-part model that simultaneously assessed the odds and the level of oocyst excretion., Results: Factors associated with lower odds of oocyst excretion were: use of toltrazuril treatment, all-in all-out management of the farrowing rooms, no cross-fostering or fostering during the first 24 hours after farrowing, plastic flooring in the farrowing pens, farrowing rooms with more than fourteen farrowing pens and employment of more than two caretakers in the farrowing section. Factors associated with lower oocyst excretion level were: use of toltrazuril treatment and caretakers averting from entering into farrowing pens., Conclusion: Apart from prophylactic treatment with toltrazuril, the risk and the level of I. suis oocyst excretion from piglets in their second week of life, was associated with managerial and environmental factors. Changes in these factors, which may enhance prevention of piglet isosporosis - either alternatively or supplementary to medical control - are of increasing importance because of the likely development of resistant parasites under the currently widespread use of anticoccidial compounds.

An observational study was conducted to determine the prevalence of Isospora suis oocysts in fecal samples from suckling piglets in Ontario, and to evaluate the relationship between the presence of I. suis oocysts and diarrhea. Fifty farms and 709 litters of piglets were included in the study. Oocysts were detected on 70% of farms, with 187 litters infected. A litter of pigs that was positive for oocysts was significantly more likely to exhibit diarrhea than a litter that was negative [odds ratio (OR) = 4.0; 95% confidence interval (CI) = 2.8 to 5.8; P < 0.001]. Management and housing factors were examined with respect to risk factors for the presence of I. suis. Farms that did not use a detergent when cleaning farrowing crates were 10-times more likely to be positive for I. suis than those that used a detergent (P = 0.007). It was concluded that coccidiosis is a common problem on Ontario swine farms.

Porcine neonatal coccidiosis is caused by the protozoan Isospora suis and affects mainly piglets in the first three weeks of life. High morbidity with diarrhoea and reduced weight gain lead to economic losses, affecting pig-breeding worldwide. Infection causes damage of the mucosal surface in the jejunum and ileum and transient non-haemorrhagic diarrhoea. Secondary infections with other enteric pathogens may lead to increased mortality. Despite its economic and veterinary importance, the immunology of porcine isosporosis is still poorly understood. A striking feature of the infection is the rapidly increasing age resistance prohibiting the development of clinical disease in piglets older than 3-4 weeks irrespective of the immune status. It can be hypothesised that the development of the innate immune system in the first weeks of life and subsequently its interplay with the adaptive immune system is closely related to this phenomenon. Infections with I. suis induce migration of TcR-gammadelta(+) cells to the gut during primary infection and lead to induction of IFN-gamma production by TcR-gammadelta(+) cells and CD4(+) T-helper cells in blood and various lymphoid tissues. Like in other coccidial infections both innate as well as adaptive response mechanisms are activated during infection. They might be both not completely developed in the first weeks of life and therefore leaving a time frame for successful infection.

Data from 13 trials involving 124 suckling piglets experimentally infected with Isospora suis were evaluated for the effects of infection dose and age on the clinical and parasitological outcome of infection in four different models, infections with 1,000 oocysts on the 1(st) day of life (d.o.l.) (model 1; 9 piglets/3 litters), 1,000 oocysts on the 4(th) d.o.l. (model 2; 25 piglets/11 litters), 1,500 oocysts on the 4(th) d.o.l. (model 3; 40 piglets/20 litters) and 10,000 oocysts on the 4(th) d.o.l. (model 4; 50 animals/10 litters). Weights were determined on the day of birth and in weekly intervals. Faecal consistency and quantitative oocysts excretion were evaluated for 2 weeks starting 4 days after infection (d.p.i.). The weight gain depression was most noticeable in model 2 (infection on the 1(st) d.o.l.), where animals only gained 2.08 x their birth weight until the 22(nd) d.o.l., compared to 2.31-2.52 x in the other groups. This correlated with the occurrence of watery diarrhoea which was found in 37 % of the samples in the acute phase (4-11 d.p.i.) in model 2 but only in 12-20 % of the samples in the other models. Median oocyst excretion peaked earlier in the models with higher infection doses but reached the highest values in model 2 (early infection). As in previous studies, this cross-sectional analysis of a larger number of animals confirms the influence of age on the outcome of isosporosis in suckling piglets, stressing the need to control the infection at an early life phase.

The modification of flotation method for the examination of diarrhoeic piglet faeces for the detection of Isospora suis oocysts was elaborated. The method was based on removing fractions of fat from the sample of faeces by centrifugation with a 25% Percoll solution. The investigations were carried out in comparison to the McMaster method. From five variants of the Percoll flotation method, the best results were obtained when 2ml of flotation liquid per 1g of faeces were used. The limit of detection in the Percoll flotation method was 160 oocysts per 1g, and was better than with the McMaster method. The efficacy of the modified method was confirmed by results obtained in the examination of the I. suis infected piglets. From all faecal samples, positive samples in the Percoll flotation method were double the results than that of the routine method. Oocysts were first detected by the Percoll flotation method on day 4 post-invasion, i.e. one-day earlier than with the McMaster method. During the experiment (except for 3 days), the extensity of I. suis invasion in the litter examined by the Percoll flotation method was higher than that with the McMaster method. The obtained results show that the modified flotation method with the use of Percoll could be applied in the diagnostics of suckling piglet isosporosis.

Little is known about the prevalence of Isospora in domestic pigs in Western Australia. A total of 289 pig faecal samples were collected from pre- and post-weaned pigs and sows from 1 indoor and 3 outdoor piggeries located in the south-west region of Western Australia. Faecal samples were screened using a PCR-RFLP assay based on the ITS-1 rDNA locus. An overall prevalence of 10.4% (30/289) was identified. Isospora was detected in 16.3% (20/123) of pre-weaned animals and 6.4% (10/156) of post-weaned animals. PCR-RFLP analysis confirmed the presence of Isospora suis in 86.7% of the positive Isospora isolates. Isospora was significantly associated with diarrhea and the findings of this study suggest that management factors such as cleaning practices, flooring types and stocking densities need to be investigated in the porcine host to find new and improved measures for control.

The aim of this investigation was to determine the prevalence of coccidian infections in suckling piglets and sows in Poland. The research was carried out in 14 out of 16 Polish provinces in the years 2003-2005. The investigation was conducted on three types of farms: large farms (>100 sows), medium farms (25-100 sows) and small farms (<25 sows). Diarrhoea of unweaned piglets was observed on all the examined farms. Overall, 780 litters of suckling piglets from 104 farms and 267 mother sows were examined. The faeces were analyzed with the modified McMaster method. Isopsora suis was found in 217 (27.8%) litters from 70 (66.7%) farms. Eimeria spp. was detected only in 20 (2.6%) litters from 12 (11.5%) farms. On the large farms I. suis infection was detected in 31.7% of litters whereas Eimeria spp. in 1.4% of them. On the medium sized farms I. suis was found in 18.1% of litters and Eimeria spp. in 0.6%. On the small farms I. suis was detected in only 13.2% of litters, whereas Eimeria spp. in as many as 28.9%. I. suis and Eimeria spp. oocysts were found in 18 (6.7%) and 16 (6%) sows respectively. From 72 sows producing I. suis infected piglets only 12 (16.7%) shed I. suis oocysts and as little as 4 (5.6%) shed Eimeria oocysts. In the remaining 56 sows (77.8%) no cases of coccidian infections were detected. The results of this investigation demonstrate the high prevalence of I. suis in suckling piglets on the large swine farms in Poland.

A highly sensitive diagnostic method is of great importance for detection of I. suis (Isospora suis). In the present study KSFV (combined sedimentation-flotation method) using different flotation solutions (NaCl, ZnSO4, NaCl-sucrose) was investigated regarding sensitivity, costs and expenditure of human labour in comparison to FV (flotation method) using NaCl-sucrose solution and to AM (autofluorescence microscopy). Ninety faecal samples were examined. Using AM, oocysts were detected in 83.3% of the samples. In cases of KSFV and FV, respectively, 41.1% to 53.3% of samples were tested positive. The quantity of detected oocysts was markedly higher in AM than in KSFV or FV. Therefore AM is the most sensitive of the comprised methods. The expenditure of labour is lowest in AM, time exposure is lower in FV than in KSFV. For performance of AM a fluorescence microscope is required contrary to KSFV and FV.

Nationwide surveys for the occurrence of Isospora suis were carried out in Germany, Austria and Switzerland including a questionnaire regarding herd size, health status and management practices and a coccidiosis sampling kit for pooled faecal samples from litters of suckling piglets. A total of 184 veterinary practices participated in the survey and returned 1745 samples (331 kits) from 324 farms in the north (n = 98), south (n = 84), centre/east (n = 42) and west (n = 10) of Germany, Austria (n = 61) and Switzerland (n = 29) with larger farms in north and centre/east (average number of sows: 270 and 500) and smaller ones in the south (95), Austria (60) and Switzerland (43). Larger farms tended to have better hygienic standards (slatted floors, disinfection of the farrowing units). The majority of the participating farms (93.5%) reported problems with diarrhoea in piglets at 2-3 weeks of age, significantly associated (P < 0.001) with uneven weaning weights (94.9%). Toltrazuril (5%; Baycox) was used only rarely; however, in these farms unevenness of weaning weights was less frequently observed (P = 0.011). A 76.2% of the farms were positive for I. suis (samples contained mostly low or moderate oocyst numbers), especially in the south (P < 0.001). Oocysts were more frequently found in samples from farms with reported diarrhoea (P = 0.011), uneven weight gain (P = 0.019) or in herds of small size (P < 0.001). Disinfection, floor type or treatment with toltrazuril did not affect the frequency of observation of oocysts.

Parasites from swine faeces were examined for autofluorescence. Oocysts of Eimeria polita, E. scabra and Isospora suis, cysts of Balantidium coli and eggs of Oesophagostomum dentatum, Strongyloides ransomi and Trichuris suis (but not those of Ascaris suum) emitted light after excitation with UV light. I. suis oocyst counts in McMaster chambers utilising autofluorescence were compared to those from conventional bright field microscopy. Similarly, faecal smears containing I. suis were examined using the same techniques. Autofluorescence was superior to bright field microscopy in detecting oocysts after flotation and was highly significantly more sensitive when direct smears were examined.

Faecal samples from suckling (n = 205) and weaned piglets (n = 82) with diarrhoea from 24 farms in Southern Germany were examined for shedding of important metazoic parasitic, viral and bacterial pathogens using culture, microscopic and electronmicroscopic methods. Escherichia coli isolates were tested further for the enterotoxin genes est-Ia and elt-I by colony blot hybridization. Isospora suis was diagnosed in 26.9% and Cryptosporidium parvum in 1.4% of the piglets investigated. The proportion of coronavirus-positive animals was 13.4% and 4% were positive for rotavirus. It was found that 17.6% of the animals were infected with enterotoxigenic E. coli (ETEC; 10.1% ETEC-ST-Ia and 8.6% ETEC-LT-I, respectively). The occurrence of the pathogens was significantly associated with the age of the animals examined (P < 0.001). Isospora suis was predominantly isolated from suckling piglets (in the second and third week of life), while in weaned piglets (fourth week of life) rotavirus and ETEC were most prevalent. On 22 of the 24 piglet production farms examined at least one of the investigated pathogens was detected. Coronavirus was diagnosed in 66.7%, I. suis in 62.5%, rotavirus in 20.8% and C. parvum in 8.3% of the farms. These results underline the fact that despite the hygienic, technical and immune preventive efforts during the last years, enteropathogens are still common in German piglet production units.

The prevalence of Isospora suis and clinical signs of isosporosis were observed in 33 swine farms from 20 sites in the southeastern state of São Paulo, Brazil. The study was performed by collecting 177 faecal samples from nursing and weaned piglets. A history of clinical neonatal isosporosis, as well as the type of farrowing and nursery houses and the pig management in the farms were correlated to the prevalence of I. suis oocysts. Six faecal samples were collected in each of the farms (two from groups of 10- to 19-day-old piglets, two from groups of 20- to 29-day-old and another two from groups of 30- to 50-day-old pigs). Faecal consistency was also registered at the time of their collection. Chi-square tests were used for statistical analysis. Oocysts were more prevalent in farms with a history of neonatal isosporosis than in those without previous cases. Faecal consistency was not related to oocyst elimination. In farms with a history of clinical isosporosis, faecal samples from groups of 10- to 19-day-old piglets showed a higher prevalence of oocysts than the groups of other ages studied. There was no difference in the prevalence of oocysts between nursing and weaned piglets. Oocysts were more prevalent in faecal samples collected from dirty-cemented floors than from self-cleaning floors in the farrowing houses. Types of floor and pig management in nursery houses were not associated with the presence of oocysts in weaned pigs.

From July 1993 to June 1994 2036 fecal samples from 196 litters from 10 pig farms were collected. Three or four piglets from each litter and the respective sows were examined 3 times at 2, 3 and 4 weeks after farrowing. I. suis was detected in all 10 farms. The rate of infection of the litters was 62.2%. 71.6% of all litters with diarrheic problems were I. suis positive. The percentage of I. suis in the feces of piglets was 17.4% at the age of 2 weeks, 41.3% at the age of 3 weeks and 39.1% at the age of 4 weeks. It was also shown that I. suis occurs on any farm with any type of management system and at any time of the year. Fecal samples of sows were always Isospora negative. It is inferred that I. suis is a more important causative agent of porcine neonatal diarrhoea than considered so far. Therefore, further studies on epizootiological and therapeutical aspects and sanitation measures are necessary.

To determine the efficacy of toltrazuril as a prophylactic treatment for coccidiosis in piglets caused by Isospora suis (I suis), a single 1.0 mL dose of toltrazuril was administered orally to 1056 piglets between 3 and 6 days of age, in 5 piggeries. Prophylactic treatment of piglets reduced the occurrence of coccidiosis in litters from 71% to 22%. The number of antibacterial treatments given and the number of piglets affected per litter were also significantly reduced, resulting in some improvement in growth rates to weaning. The severity of diarrhoea was significantly reduced, as was the amount of oocyst excretion. The number of days that piglets excreted oocysts in the faeces was reduced from 4.9 days to 2.5 days. The detection of I suis in piglets with diarrhoea was reduced from 84% in the untreated piglets to 6% in the piglets given the prophylactic treatment.

Forty-seven piglets were inoculated with doses of 100 to 50,000 sporulated oocysts of Isospora suis. After 5-7 days oocysts were found in faeces. The patent period extended from 8 to 16 days. The shedding of oocysts showed a cyclic pattern with 2-3 peaks separated by intervals of approximately 5 days. Subpatent periods were often seen between the peaks. The level of oocyst shedding during the initial days of the patent period reflected, to some extent, the inoculation dose. However, a maximum of OPG at the 100,000 level was observed among one or more piglets from all groups, regardless of the inoculation dose. Among the majority of piglets inoculated with more than 100 oocysts, the highest OPG-figures were observed in the first peak of the cyclic pattern. Unlike this, the maximum of OPG was observed in the second peak of the cycle among 6 of the 7 piglets inoculated with 100 oocytes only. The triphasic pattern was most pronounced in the low dose group. The marked upscaling of oocyst production, as particularly registered in the low dosed groups, seams to explain at least part of the problems met under practical conditions, when trying to eliminate the transmission of oocytes between successive litters in the farrowing boxes. The cyclic excretion pattern and an apparent absence of autoinfection may indicate that the development of I. suis in the host includes several oocyte producing generations descending from the same initial infection. The presence of subpatent periods can probably explain the marked variation in OPG, as they are often recorded when examining faecal samples from piglets, even when the samples are originating from the same litter.

The aim of this study was to determine the cause and risk factors involved in preweaning piglet diarrhoea. Faecal samples from 2380 diarrhoeic piglets, 5 to 30 days of age, were examined for enteropathogens. Isospora suis oocysts were detected in 53.8% of samples, Escherichia coli in 18.2% and rotavirus in 16.9%. I suis had the widest distribution, being present on 70.9% of 151 piggeries. The onset of diarrhoea occurred mainly between 7 and 14 days (77.5%), peaking at 10 days. Records of 4086 litters from two intensive piggeries were analysed and showed no seasonal variation or effect of sow parity on the incidence of piglet diarrhoea. I suis was the most common enteropathogen associated with diarrhoea in piglets from 5 days of age until weaning.

Results of a program designed to control neonatal porcine coccidiosis on a total confinement, farrow-to-finish swine farm are reported. The control program consisted of washing, phenol disinfection, and steam cleaning of farrowing houses and treatment of sows with amprolium HCl before and after farrowing. Before initiation of the control program, 88.9% of the sows examined in the farrowing house were negative for coccidian oocysts, 9.9% were positive for Eimeria spp, and 1.2% were positive for Isospora suis. Most pigs nursing on sows before initiation of the control program had diarrhea at 5 to 10 days of age, which led to dehydration and weight loss. Morbidity was high, and mortality was moderate. Composite fecal samples from these litters were all positive (100%) for I suis. After initiation of the control program, 99.6% of the sows examined in the farrowing house were negative for coccidian oocysts and 0.4% were positive for Eimeria spp. Clinical signs of coccidiosis were rarely present in nursing pigs examined after the control program was initiated; however, I suis was still present in 19.8% of the composite fecal samples from pigs examined. An association between oocyst production in sows and I suis infections in pigs was not found in the present study. Oocysts of Eimeria spp were not found in the feces from the pigs.

Faecal samples from 232 domestic pigs raised on concrete, 98 free-ranging village pigs, and five wild boar showed 46.6 (108/232), 54 (53/98) and 80% (4/5) prevalence of coccidian oocysts, respectively. Eight species of Eimeria, and Isospora suis, were recovered. In their descending order of predominance in the pigs raised on concrete, the species of coccidia were E. debliecki (26.7%), E. scabra (22.4%), E. neodebliecki (19.8%), E. porci (15.5%), E. suis (11.6%), E. polita (8.6%), E. perminuta (7%), E. spinosa (5.6%) and I. suis (3.9%). The first five species listed above predominated in the village pigs as well. E. polita, E. spinosa and I. suis were not found in the wild boar. I. suis oocysts prevailed in 8.3% of the 36 sows on concrete, and in 11.1% (3/27) of those which were positive for coccidia. Isosporoid oocysts were absent in the village sows. Of the 125 less than 24-day-old piglets, 29.6% were diarrhoeic, and of these, 43.2% were positive for coccidia. Four of the 16 (25%) coccidia-positive, diarrhoeic piglets, and four of the 37 (10.8%) coccidia-positive non-diarrhoeic piglets shed I. suis oocysts, an observation which seems to weaken the present contention that I. suis is the primary causative agent of neonatal porcine coccidiosis. The highest mean number of oocysts per gram faeces (23,550) was recorded from the diarrhoeic farm piglets on concrete, and the lowest of 6,100 from the gestating farm sows. Mean opg data revealed very little significant quantitative variation between the corresponding age groups of the free-ranging village pigs and the commercially-farmed ones. One of the most interesting findings in the study was that the sows were more frequently infected than all other age groups.

A species of porcine coccidia, Isospora suis, when inoculated into piglets, produced diarrhea, dehydration, weight loss and death. Gross lesions were characterized by a fibrino-necrotic membrane within the jejunum and ileum. Villous atrophy and variable erosion, often with an adhered necrotic membrane, were seen microscopically. Asexual and sexual stages of I. suis were seen within the intestinal epithelium and oocysts were recovered from the feces. The severity of clinical disease and lesions were dose-related.

Coccidiosis is a disease of the young piglet due to infection with Isospora suis and is characterized by diarrhea which is nonresponsive to antibacterial therapy. There is variable morbidity and mortality. Piglets develop a more severe clinical illness and enteritis when infected with I. suis at one to three days of age than when infected at two weeks of age. Microscopic lesions range from villous atrophy and mild erosion to severe fibrinonecrotic enteritis.

In two studies carried out in 1974-75 on 10 eight and 25 two week old piglets obtained from a SPF-herd the endoparasitic status was determined. Samples of faeces were examined by microscopy which revealed the presence of Isospora suis, a finding that was quite unexpected in a SPF-herd. During days 17-23 pp. the 25 piglets were not found to excrete oocysts, the earliest findings of oocysts being made 24-35 days p.p. The 10 piglets being kept isolated in individual pens excreted oocysts up until 88 days p.p. The epidemiology of Isospora suis is discussed.

Oocysts of Isospora suis were not found in any of 77 fecal samples from sows on farms with a history of neonatal coccidiosis. Oocysts of Isospora suis were found in 1 of 172 fecal samples from farms without a history of neonatal coccidiosis. Oocysts of Eimeria spp were found in 81.8% of the sows from farms with a history of neonatal coccidiosis and in 94.8% of the sows from farms without a history of neonatal coccidiosis. Oocysts of Isospora spp from birds were encountered as a pseudoparasite in several fecal samples.

Serum proteins were evaluated by agarose electrophoresis at periodic intervals between postinoculation days (PID) 2 and 35 in 4 litters of pigs inoculated orally at 36 to 38 hours of age with 70,000 sporulated Isospora suis oocysts and in 4 litters of age-matched noninoculated control pigs. In inoculated pigs, clinical disease characterized by vomiting and diarrhea began at PID 3 to 4 and was resolved by PID 11. Isospora suis oocysts were detected in feces of inoculated pigs from PID 5 to 25, with peak numbers present on PID 5 to 6. Of 43 pigs inoculated with I suis oocysts, 4(9.3%) died of coccidiosis, and samples were not obtained for serum protein evaluation. Of 39 noninoculated control pigs, all remained clinically normal, and I suis oocysts were not detected in their feces. Serum protein fraction values in inoculated and control groups compared at each sample collection time did not differ significantly, except at PID 15, when beta-globulin values were lower in inoculated pigs (P = 0.02). At PID 35, total serum proteins, albumin, and alpha 1-, beta-, and gamma-globulin values of inoculated pigs were lower than those of controls, but there were too few pigs examined for definitive statistical analysis. Differences in trends over time were observed between inoculated and control groups for several serum protein fractions. In inoculated pigs, total proteins (P less than 0.1) and beta-globulins (P less than 0.01) decreased with time, whereas those of control pigs increased. Similar differences in trends were noticed for albumin and alpha 1-globulins, but these were not significant.(ABSTRACT TRUNCATED AT 250 WORDS)

Examinations of fecal specimens from swine of all ages maintained in a totally enclosed confinement facility in south Georgia were conducted in a 5-year survey (1977-1981) to determine the prevalence, intensity, and transmission patterns of intestinal nematode and protozoan parasites. Weaned pigs in the nursery had no detectable parasitic infections except sporadic Isospora suis in newly weaned pigs. Growing-finishing hogs had a low prevalence of Ascaris suum, Oesophagostomum spp. and Balantidium suis. Mean A. suum prevalence and mean eggs per gram feces (EPG) increased until about 200 days of age and then declined. Oesophagostomum spp. and B. suis prevalence and intensity tended to increase throughout life. Gilts (mean age 273 days) in the gestation unit had a higher (P less than 0.01) prevalence of A. suum and lower (P less than 0.01) prevalences of Oesophagostomum spp. and B. suis than sows (mean age 706 days). Similarly, A. suum EPG was higher (P less than 0.01) and Oesophagostomum spp. EPG and B. suis cysts per gram feces (CPG) were lower (P less than 0.01) in gilts than in sows. No evidence for a peri-parturient increase in parasite eggs or cysts was found. Instead, there was a higher prevalence of B. suis (P less than 0.01) in gestating than in lactating animals and more lactating sows than gestating sows were negative for parasites (P less than 0.01). Apparent differences due to location (gestation unit or farrowing unit) were largely attributable to age differences. Trichuris suis infections were rare and not shown to be established in the herd. Strongyloides ransomi in suckling piglets was controlled by anthelmintic therapy. I. suis occurred in piglets throughout the study but was never found in sows in the farrowing unit, either before or after birth of a subsequently infected litter of piglets. No parasites requiring intermediate hosts occurred.
