Switchgrass (SG) is considered a model bioenergy crop and a warm‐season perennial grass (WSPG) that traditionally served as forage feedstock in the United States. To avoid the sole dependence on SG for bioenergy production, evaluation of other crops to diversify the pool of feedstock is needed. We conducted a 3‐year field experiment evaluating eastern gamagrass (GG), another WSPG, as complementary feedstock to SG in one‐ and two‐cut systems, with or without intercropping with crimson clover or hairy vetch, and under different nitrogen (N) application rates. Our results showed that GG generally produced lower biomass (by 29.5%), theoretical ethanol potential (TEP, by 2.8%), and theoretical ethanol yield (TEY, by 32.9%) than corresponding SG under the same conditions. However, forage quality measures, namely acid detergent fiber (ADF), crude protein (CP), and elements P, K, Ca, and Mg were significantly higher in GG than those in SG. Nitrogen fertilizer significantly enhanced biomass (by 1.54 Mg ha−1), lignin content (by 2.10 g kg−1), and TEY (787.12 L ha−1) in the WSPGs compared to unfertilized treatments. Intercropping with crimson clover or hairy vetch did not significantly increase biomass of the WSPGs, or TEP and TEY in unfertilized plots. Nitrogen (N) fertilization significantly affects soil extracellular oxidases, agents responsible for decomposition of slow turnover and recalcitrant soil organic carbon (SOC; e.g., lignin), and consequently influences soil carbon sequestration capacity. However, it remains unclear how soil oxidases mediate SOC sequestration under N fertilization, and whether these effects co‐vary with plant type (e.g., bioenergy crop species). Using a spatially explicit design and intensive soil sampling strategy under three fertilization treatments in switchgrass (SG: Panicum virgatum L.) and gamagrass (GG: Tripsacum dactyloides L.) croplands, we quantified the activities of polyphenolic oxidase (PHO), peroxidase (PER), and their sum associated with recalcitrant C acquisition (OX). The fertilization treatments included no N fertilizer input (NN), low N input (LN: 84 kg N ha−1 year−1 in urea), and high N input (HN: 168 kg N ha−1 year−1 in urea). Besides correlations between soil oxidases and SOC (formerly published), both descriptive and geostatistical approaches were applied to evaluate the effects of N fertilization and crop type on soil oxidases activities and their spatial distributions. Results showed significantly negative correlations between soil oxidase activities and SOC across all treatments. The negative relationship of soil oxidases and SOC was also evident under N fertilization. First, LN significantly depressed oxidases in both mean activities and spatial heterogeneity, which corresponded to increased SOC in SG (though by 5.4%). LN slightly influenced oxidases activities and their spatial heterogeneity, consistent with insignificant changes of SOC in GG. Second, HN showed trends of decrease in soil oxidase activities, which aligned with the significantly enhanced SOC in both croplands. Overall, this study demonstrated that soil oxidase activities acted as sensitive and negative mediators of SOC sequestration in bioenergy croplands and optimizing fertilizer use particularly in switchgrass cropland can improve for both carbon sequestration and environmental benefit. Nitrogen (N) fertilization affects bioenergy crop growth and productivity and consequently carbon (C) and N contents in soil, it however remains unclear whether N fertilization and crop type individually or interactively influence soil organic carbon (SOC) and total N (TN). In a three-year long fertilization experiment in switchgrass (SG: Panicum virgatum L.) and gamagrass (GG: Tripsacum dactyloides L.) croplands in Middle Tennessee USA, soil samples (0–15cm) were collected in plots with no N input (NN), low N input (LN: 84 kg N ha-1 yr-1 in urea) and high N input (HN: 168 kg N ha-1 yr-1 in urea). Besides SOC and TN, the aboveground plant biomass was also quantified. In addition to a summary of published root morphology data based on a separated mesocosm experiment, the root leachable dissolved organic matter (DOM) of both crops was also measured using archived samples. Results showed no significant interaction of N fertilization and crop type on SOC, TN or plant aboveground biomass (ABG). Relative to NN, HN (not LN) significantly increased SOC and TN in both crops. Though SG showed a 15–68% significantly higher ABG than GG, GG showed a 9.3–12% significantly higher SOC and TN than SG. The positive linear relationships of SOC or TN with ABG were identified for SG. However, GG showed structurally more complex and less readily decomposed root DOM, a larger root volume, total root length and surface area than SG. Collectively, these suggested that intensive N fertilization could increase C and N stocks in bioenergy cropland soils but these effects may be more likely mediated by the aboveground biomass in SG and root chemistry and morphology in GG. Nitrogen (N) fertilizers can potentially alter spatial distribution of soil organic carbon (SOC) and total nitrogen (TN) concentrations in croplands such as switchgrass (SG: Panicum virgatum L.) and gamagrass (GG: Tripsacum dactyloides L.), but it remains unclear whether these effects are the same between crops and under different rates of fertilization. 13C and 15N are two important proxy measures of soil biogeochemistry, but they were rarely examined as to their spatial distributions in soil. Based on a three-year long fertilization experiment in Middle Tennessee, USA, the top mineral horizon soils (0–15 cm) were collected using a spatially explicit design within two 15-m2 plots under three fertilization treatments in SG and GG croplands. A total of 288 samples were collected based on 12 plots and 24 samples in each plot. The fertilization treatments were no N input (NN), low N input (LN: 84 kg N ha−1 in urea) and high N input (HN: 168 kg N ha−1 in urea). The SOC, TN, SOC/TN (C: N), δ13C and δ15N were quantified and their within-plot variations and spatial distributions were achieved via descriptive and geostatistical methods. Results showed that SG generally displayed 10~120% higher plot-level variations in all variables than GG, and the plot-level variations were 20~77% higher in NN plots than LN and HN plots in SG but they were comparable in unfertilized and fertilized plots in GG. Relative to NN, LN and HN showed more significant surface trends and spatial structures in SOC and TN in both croplands, and the fertilization effect appeared more pronounced in SG. Spatial patterns in C: N, δ13C and δ15N were comparable among different fertilization treatments in both croplands. The descending within-plot variations were also identified among variables (SOC > TN > δ15N > C: N > δ13C). This study demonstrated that N fertilizations generally reduced the plot-level variance and simultaneously re-established spatial structures of SOC and TN in bioenergy croplands, which little varied with fertilization rate but was more responsive in switchgrass cropland. Soil microbiome responses to short-term nitrogen (N) inputs remain uncertain when compared with previous research that has focused on long-term fertilization responses. Here, we examined soil bacterial/archaeal and fungal communities pre- and post-N fertilization in an 8 year-old switchgrass field, in which twenty-four plots received N fertilization at three levels (0, 100, and 200 kg N ha-1 as NH4NO3) for the first time since planting. Soils were collected at two depths, 0–5 and 5–15 cm, for DNA extraction and amplicon sequencing of 16S rRNA genes and ITS regions for assessment of microbial community composition. Baseline assessments prior to fertilization revealed no significant pre-existing divergence in either bacterial/archaeal or fungal communities across plots. The one-time N fertilizations increased switchgrass yields and tissue N content, and the added N was nearly completely removed from the soil of fertilized plots by the end of the growing season. Both bacterial/archaeal and fungal communities showed large spatial (by depth) and temporal variation (by season) within each plot, accounting for 17 and 12–22% of the variation as calculated from the Sq. root of PERMANOVA tests for bacterial/archaeal and fungal community composition, respectively. While N fertilization effects accounted for only ~4% of overall variation, some specific microbial groups, including the bacterial genus Pseudonocardia and the fungal genus Archaeorhizomyces, were notably repressed by fertilization at 200 kg N ha-1. Bacterial groups varied with both depth in the soil profile and time of sampling, while temporal variability shaped the fungal community more significantly than vertical heterogeneity in the soil. These results suggest that short-term effects of N fertilization are significant but subtle, and other sources of variation will need to be carefully accounted for study designs including multiple intra-annual sampling dates, rather than one-time “snapshot” analyses that are common in the literature. Abstract: The mechanistic understanding of warming and nitrogen (N) fertilization, alone or in combination, on microbially mediated decomposition is limited. In this study, soil samples were collected from previously harvested switchgrass (Panicum virgatum L.) plots that had been treated with high N fertilizer (HN: 67 kg N ha−1) and those that had received no N fertilizer (NN) over a 3‐year period. The samples were incubated for 180 days at 15 °C and 20 °C, during which heterotrophic respiration, δ13C of CO2, microbial biomass (MB), specific soil respiration rate (Rs: respiration per unit of microbial biomass), and exoenzyme activities were quantified at 10 different collections time. Employing switchgrass tissues (referred to as litter) with naturally abundant 13C allowed us to partition CO2 respiration derived from soil and amended litter. Cumulative soil respiration increased significantly by 16.4% and 4.2% under warming and N fertilization, respectively. Respiration derived from soil was elevated significantly with warming, while oxidase, the agent for recalcitrant soil substrate decomposition, was not significantly affected by warming. Warming, however, significantly enhanced MB and Rs indicating a decrease in microbial growth efficiency (MGE). On the contrary, respiration derived from amended litter was elevated with N fertilization, which was consistent with the significantly elevated hydrolase. N fertilization, however, had little effect on MB and Rs, suggesting little change in microbial physiology. Temperature and N fertilization showed minimal interactive effects likely due to little differences in soil N availability between NN and HN samples, which is partly attributable to switchgrass biomass N accumulation (equivalent to ~53% of fertilizer N). Overall, the differential individual effects of warming and N fertilization may be driven by physiological adaptation and stimulated exoenzyme kinetics, respectively. Climate changes, including chronic changes in precipitation amounts, will influence plant physiology and growth. However, such precipitation effects on switchgrass, a major bioenergy crop, have not been well investigated. We conducted a two-year precipitation simulation experiment using large pots (95 L) in an environmentally controlled greenhouse in Nashville, TN. Five precipitation treatments (ambient precipitation, and -50%, -33%, +33%, and +50% of ambient) were applied in a randomized complete block design with lowland "Alamo" switchgrass plants one year after they were established from tillers. The growing season progression of leaf physiology, tiller number, height, and aboveground biomass were determined each growing season. Precipitation treatments significantly affected leaf physiology, growth, and aboveground biomass. The photosynthetic rates in the wet (+50% and +33%) treatments were significantly enhanced by 15.9% and 8.1%, respectively, than the ambient treatment. Both leaf biomass and plant height were largely increased, resulting in dramatically increases in aboveground biomass by 56.5% and 49.6% in the +50% and +33% treatments, respectively. Compared to the ambient treatment, the drought (-33% and -50%) treatments did not influence leaf physiology, but the -50% treatment significantly reduced leaf biomass by 37.8%, plant height by 16.3%, and aboveground biomass by 38.9%. This study demonstrated that while switchgrass in general is a drought tolerant grass, severe drought significantly reduces Alamo’s growth and biomass, and that high precipitation stimulates its photosynthesis and growth. Aims: The objective of this study was to investigate the effects of the precipitation changes on soil, microbial and root respirations of switchgrass soils, and the relationships between soil respiration and plant growth, soil moisture and temperature. Methods: A mesocosm experiment was conducted with five precipitation treatments over two years in a greenhouse in Nashville, Tennessee. The treatments included ambient precipitation, −50%, −33%, +33% and +50% of ambient precipitation. Soil, microbial, and root respirations were quantified during the growing seasons. Results: Mean soil and root respirations in the +50% treatment were the highest (2.48 and 0.93 μmol CO m s, respectively) among all treatments. Soil microbial respiration contributed more to soil respiration, and had higher precipitation sensitivity mostly than root respiration. Increases in precipitation mostly enhanced microbial respiration while decreases in precipitation reduced both microbial and root respirations. Across precipitation treatments, soil respiration was significantly influenced by soil moisture, soil temperature, and aboveground biomass. Conclusions: Our results showed that microbial respiration was more sensitive to precipitation changes, and precipitation regulated the response of soil respiration to soil temperature. Extracellular glycosidases in soil, produced by microorganisms, act as major agents for decomposing labile soil organic carbon (e.g., cellulose). Soil extracellular glycosidases are significantly affected by nitrogen (N) fertilization but fertilization effects on spatial distributions of soil glycosidases have not been well addressed. Whether the effects of N fertilization vary with bioenergy crop species also remains unclear. Based on a 3-year fertilization experiment in Middle Tennessee, USA, a total of 288 soil samples in topsoil (0–15 cm) were collected from two 15 m2 plots under three fertilization treatments in switchgrass (SG: Panicum virgatum L.) and gamagrass (GG: Tripsacum dactyloides L.) using a spatially explicit design. Four glycosidases, α-glucosidase (AG), β-glucosidase (BG), β-xylosidase (BX), cellobiohydrolase (CBH), and their sum associated with C acquisition (Cacq) were quantified. The three fertilization treatments were no N input (NN), low N input (LN: 84 kg N ha−1 year−1 in urea) and high N input (HN: 168 kg N ha−1 year−1 in urea). The descriptive and geostatistical approaches were used to evaluate their central tendency and spatial heterogeneity. Results showed significant interactive effects of N fertilization and crop type on BX such that LN and HN significantly enhanced BX by 14% and 44% in SG, respectively. The significant effect of crop type was identified and glycosidase activities were 15–39% higher in GG than those in SG except AG. Within-plot variances of glycosidases appeared higher in SG than GG but little differed with N fertilization due to large plot-plot variation. Spatial patterns were generally more evident in LN or HN plots than NN plots for BG in SG and CBH in GG. This study suggested that N fertilization elevated central tendency and spatial heterogeneity of glycosidase activities in surficial soil horizons and these effects however varied with crop and enzyme types. Application of nitrogen (N) fertilizer plus biochar may increase crop yield, but how biochar will interact with N fertilization to affect bioenergy crop switchgrass physiology, biomass, and soil CO2 emission (i.e., soil respiration) from switchgrass fields remains unclear. Here, we assessed this issue by conducting a field experiment near Nashville TN with two levels of biochar treatment (a control without biochar addition and biochar addition of 9 Mg ha−1), and four N fertilization levels (0 kg N ha−1, 17 kg N ha−1, 34 kg N ha−1, and 67 kg N ha−1, labeled as ON, LN, MN, and HN, respectively). Results showed that both biochar addition and N fertilization did not influence switchgrass leaf photosynthesis and biomass, but biochar addition enhanced leaf transpiration, and reduced water use efficiency. Soil respiration was reduced by biochar addition, but significantly enhanced by N fertilization. Biochar and N fertilization interactively influenced soil respiration and seasonal variation of soil respiration was mostly controlled by soil temperature. Our results indicated that switchgrass can maintain high productivity without much N input, at least for several years. The findings from this study are useful to optimize N fertilization and biochar addition in the switchgrass fields for maintaining relatively high productive switchgrass biomass while reducing soil CO2 emission. Precipitation changes altered soil heterotrophic respiration, but the underlying microbial mechanisms remain rarely studied. This study conducted three-year switchgrass (Panicum virgatum L.) mesocosm experiment to investigate soil heterotrophic respiratory responses to altered precipitation. Five treatments were considered, including ambient precipitation (P0), two wet treatments (P+33 and P+50: 33% and 50% enhancement relative to P0), and two drought treatments (P-33 and P-50: 33% and 50% reduction relative to P0). The plant's aboveground biomass (AGB), soil organic carbon (SOC), total nitrogen (TN), microbial biomass carbon (MBC), heterotrophic respiration (R s), biomass-specific respiration (R ss : respiration per unit of microbial biomass as a reciprocal index of microbial growth efficiency), and extracellular enzymes activities (EEAs) were quantified in soil samples (0–15 cm). Despite significantly different soil moisture contents among treatments, results showed no impact of precipitation treatments on SOC and TN. Increasing precipitation had no effect, but decreasing precipitation significantly reduced plant AGB. Relative to P0, P+33 significantly increased R s by more than 3-fold and caused no changes in MBC, leading to significantly higher R ss (P < 0.05). P+33 also significantly increased hydrolytic enzyme activities associated with labile carbon acquisition (C acq) by 115%. The only significant effect of drought treatments was the decreased β - d -cellobiosidase (CBH) and peroxidase (PEO) under P-33. Nonparametric analyses corroborated the strong influences of moisture and CBH on the enhanced precipitation, which stimulated soil respiratory carbon loss, likely driven by both elevated hydrolase activities and reduced microbial growth efficiency. However, the less sensitive drought effects suggested potential microbial tolerance to water deficiency despite depressed plant growth. Switchgrass ( Panicum virgatum L.) is widely selected as a model feedstock for sustainable replacement of fossil fuels and climate change mitigation. However, how climate changes, such as altered precipitation (PPT), will influence switchgrass growth and soil carbon storage potential have not been well investigated. We conducted a two-year PPT manipulation experiment with five treatments: −50%, −33%, +0%, +33%, and +50% of ambient PPT, in an “Alamo” switchgrass field in Nashville, TN. Switchgrass aboveground net primary production (ANPP), leaf gas exchange, and soil respiration (SR) were determined each growing season. Data collected from this study was then used to test whether switchgrass ANPP responds to PPT changes in a double asymmetry pattern as framed by Knapp et al. (2017), and whether it is held true for other ecosystem processes such as SR. Results showed that the wet (+33%, and +50%) treatments had little effects on ANPP and leaf gas exchange compared to the ambient precipitation treatment, regardless of fertilization or not. The −33% treatment did not change ANPP and leaf photosynthesis, but significantly decreased transpiration and enhanced water use efficiency (WUE). Only the −50% treatment significantly decreased ANPP and LAI, without changing leaf photosynthesis. SR generally decreased under the drought treatments and increased under the wet treatments, while there was no significant difference between the two drought treatments or between the two wet treatments. Our results demonstrate that switchgrass ANPP responded in a single negative asymmetry model to PPT changes probably due to relative high PPT in the region. However, even in such a mesic ecosystem, SR responded strongly to PPT changes in an “S” curve model, suggesting that future climate changes may have greater but more complex effects on switchgrass belowground than aboveground processes. The contrasting models for switchgrass ANPP and SR in response to PPT indicate that extreme wet or dry PPT conditions may shift ecosystem from carbon accumulation toward debt, and in turn provide government and policy makers with useful information for sustainable management of switchgrass.
