In the present article we report a new hybrid microfluidic device (hyFlow) comprising a disposable paper electrode and a three-dimensional (3D) printed plastic chip for the electrochemical detection of a magnetic bead-silver nanoparticle (MB-AgNP) bioconjugate. This hybrid device evolved due to the difficulty of incorporating micron-scale MBs into paper-only fluidic devices. Specifically, paper fluidic devices can entrap MB-containing conjugates within their cellulose or nitrocellulose fiber matrix. The hyFlow system was designed to minimize such issues and transport MB conjugates more efficiently to the electrochemical detection zone of the device. The hyFlow system retains the benefit of fluid transport by pressure-driven flow, however, no pump is required for its operation. The hyFlow device is capable of detecting either pre-formed MB-AgNP conjugates or conjugates formed in-situ formation. The detection limit of AgNPs using this device is 12 pM, which represents just 22 AgNPs per MB.

Pulsed dye laser irradiation in the wavelength range of 585 to 600nm is currently the gold standard for treatment of port-wine stains (PWSs). However, this treatment method is often ineffective for deeply seated blood vessels and in individuals with moderate to heavy pigmentation. Use of optical particles doped with the FDA-approved near-infrared (NIR) absorber, indocyanine green (ICG), can potentially provide an effective method to overcome these limitations. Herein, we theoretically investigate the effectiveness of particles derived from erythrocytes, which contain ICG, in mediating photothermal destruction of PWS blood vessels. We refer to these particles as NIR erythrocyte-derived transducers (NETs). Our theoretical model consists of a Monte Carlo algorithm to estimate the volumetric energy deposition, a finite elements approach to solve the heat diffusion equation, and a damage integral based on an Arrhenius relationship to quantify tissue damage. The model geometries include simulated PWS blood vessels as well as actual human PWS blood vessels plexus obtained by the optical coherence tomography. Our simulation results indicate that blood vessels containing micron- or nano-sized NETs and irradiated at 755nm have higher levels of photothermal damage as compared to blood vessels without NETs irradiated at 585nm. Blood vessels containing micron-sized NETs also showed higher photothermal damage than blood vessels containing nano-sized NETs. The theoretical model presented can be used in guiding the fabrication of NETs with patient-specific optical properties to allow for personalized treatment based on the depth and size of blood vessels as well as the pigmentation of the individual's skin.

High re-excision rates in breast-conserving surgery call for a new intraoperative approach to the lumpectomy margin evaluation problem. The unique intraoperative imaging system, presented here, demonstrated the capability of photoacoustic tomography (PAT) to deliver optical sensitivity and specificity, along with over 2-cm imaging depth, in a clinical setting. The system enabled the evaluation of tumor extent, shape, morphology, and position within lumpectomy specimens measuring up to 11 cm in diameter. The investigation included all major breast cancer-related lesions, such as invasive ductal carcinoma (IDC), multifocal IDC, ductal carcinoma in situ and combinations of these variants. Coregistration with established ultrasound (US) technology, as well as comparison to specimen radiography, validated the performance of PAT, which appeared to facilitate better tumor visualization. Contrary to expected PA contrast mechanisms, PAT images of hemoglobin distribution correlated poorly with US-determined tumor location, while hypointense regions in lipid-weighted PAT images were in better agreement with US.

We demonstrate the use of patterned aerosol adhesives to construct both planar and nonplanar 3D paper microfluidic devices. By spraying an aerosol adhesive through a metal stencil, the overall amount of adhesive used in assembling paper microfluidic devices can be significantly reduced. We show on a simple 4-layer planar paper microfluidic device that the optimal adhesive application technique and device construction style depends heavily on desired performance characteristics. By moderately increasing the overall area of a device, it is possible to dramatically decrease the wicking time and increase device success rates while also reducing the amount of adhesive required to keep the device together. Such adhesive application also causes the adhesive to form semi-permanent bonds instead of permanent bonds between paper layers, enabling single-use devices to be non-destructively disassembled after use. Nonplanar 3D origami devices also benefit from the semi-permanent bonds during folding, as it reduces the likelihood that unrelated faces may accidently stick together. Like planar devices, nonplanar structures see reduced wicking times with patterned adhesive application vs uniformly applied adhesive.

A new model for simulating non-Brownian flexible fibres suspended in a Newtonian fluid has been developed. Special attention has been given to include realistic flow conditions found in the industrial papermaking process in the key features of the model; it is the intention of the author to employ the model in simulations of the forming section of the paper machine in future studies. The model considers inert fibres of various shapes and finite stiffness, interacting with each other through normal, frictional and lubrication forces, and with the surrounding fluid medium through hydrodynamic forces. Fibre-fluid interactions in the non-creeping flow regime are taken into account, and the two-way coupling between the solids and the fluid phase is included by enforcing momentum conservation between phases. The incompressible three-dimensional Navier-Stokes equations are employed to model the motion of the fluid medium. The validity of the model has been tested by comparing simulation results with experimental data from the literature. It was demonstrated that the model predicts the motion of isolated fibres in shear flow over a wide range of fibre flexibilities. It was also shown that the model predicts details of the orientation distribution of multiple straight, rigid fibres in a sheared suspension. Model predictions of the viscosity and first normal stress difference were in good agreement with experimental data found in the literature. Since the model is based solely on first-principles physics, quantitative predictions could be made without any parameter fitting. En ny modell för simulering av rörelserna hos icke-Brownska böjliga fibrer dispergerade i en Newtonsk vätska har utvecklats. Eftersom det är författarens avsikt att modellen skall kunna tillämpas vid simulering av arkformning under de förhållanden som råder i en modern pappersmaskin, har särskilt omsorg givits till att inkludera motsvarande flödesvillkor i modellens giltighetsområde. Modellen hanterar fibrer av varierande form, massa och styvhet, som växelverkar sinsemellan via normal-, friktions- och smörjkrafter. Deras växelverkan med den omgivande vätskan sker via hydrodynamiska krafter vid finita Reynolds-tal. Den så kallade tvåvägskopplingen mellan fibrerna och vätskefasen har tagits i beaktande genom att kräva att rörelsemängden bevaras vid interaktionen mellan faserna. Vidare har Navier-Stokes ekvationer för inkompressibla vätskor använts för att beskriva mediets rörelser. Modellens giltighet har undersökts genom att jämföra resultat från simuleringar med experimentella data från litteraturen. Det har påvisats att modellen förutsäger rörelsen hos ensamma fibrer i ett skjuvflöde, för vitt skilda fiberflexibiliteter. Det visades också att modellen förutsäger detaljer hos fiberorienteringsdistributionen i suspensioner utsatta för skjuvflöde. Det kunde också konstateras att modellens förutsägelser av fibersuspensioners viskositet och första normalspänningsdifferens under skjuvning väl överensstämde med experimentella data i litteraturen. Kvantitativa förutsägelser har kunnat göras utan någon parameteranpassning, då modellen bygger uteslutande på väletablerade fysikaliska samband inom klassisk mekanik och strömningslära.
