Tomato (Solanum lycopersicum L.) is among the most important vegetable crops worldwide. Early blight disease, caused by Alternaria solani, is a destructive foliar disease of tomato and other Solanaceae species. Herein, we investigated the in vitro antifungal properties of gallic acid and two of its derivatives (syringic and pyrogallic acids) against A. solani during 2019 and 2020 seasons. The physiological and biochemical effects of these compounds on infected tomato plants were also investigated using the whole plant bioassay. The in vitro investigation showed that all tested compounds showed fungistatic action and inhibited the mycelial radial growth of A. solani in a dose-dependent manner. In two separate pot-experiments, those compounds efficiently suppressed the development of the disease symptoms and area under disease progress curve (AUDPC), without any phytotoxic effects on the treated tomato plants. Additionally, all tested compounds positively enhanced the biochemical traits of treated plants including the chlorophyll content, the total soluble phenolics, the total soluble flavonoids, and the enzymatic activities of catalase, peroxidase, and polyphenol oxidase during 2019 and 2020 seasons. Moreover, the treatment with gallic acid and its derivatives significantly increased all yield components of A. solani-infected tomato plants such as the total number of flowers and fruits, and the fruit yield for each tomato plant in both experiments. Considering the fungitoxicity of phenolic acids against A. solani with no phytotoxicity on treated tomato plants, we believe that gallic acid and its derivatives might be a sustainable eco-friendly control strategy to reduce the usage of chemical fungicides partially or entirely against A. solani particularly, and fungal diseases in general.

Domates fidelerinde önemli verim kayıplarına neden olan erken yanıklık etmeni Alternaria solani (Ell. & Mart.) L.R. Jones and Grout toprakta uzun süre canlı kalabilmesi ve geniş konukçu çevresine sahip olmasından dolayı rotasyonla kontrolü zor olan bir etmendir. Bitki hastalıkları ile mücadelede kullanılan kimyasal ilaçların ise çevre, insan ve hayvan sağlığı üzerindeki olumsuz etkileri, dayanıklılık ve kalıntı probleminin ortaya çıkmasına ilişkin kaygılar, bilim adamlarını kimyasal mücadeleye alternatif yeni, daha ekonomik ve etkin mücadele yöntemleri keşfetme arayışına itmiştir. Bu bağlamda akla gelen yaklaşımlardan birisi de mikrobiyal biyoajanların hastalık ve zararlıların kontrolünde kullanılmasıdır.Bu nedenle, patojene karşı biyolojik mücadele ajanlarının belirlenerek etkin kullanımının sağlanması oldukça önemlidir. Bu çalışmada; domates fidelerinde hastalık yapan erken yanıklık hastalığına karşı biyolojik mücadele ajanı olabilecek fungus ve bakteri izolatları belirlenmiştir. Çalışmada daha önce yapılan farklı çalışmalarda; yabani ve kültür bitkilerinin toprak altı veya toprak üstü aksamlarından izole edilerek, Microbial Identification System (MIS)'ine göre tanılanmış olan (3 adedi Bacillus subtilis, 1 adedi B. pumilus, 1 adedi Pseudomonas fluorescens, 2 adedi Pantoea agglomerans, 2 adedi B. megaterium, 1 adedi B. cereus, 1 adedi Paenibacillus polymxa, 1 adedi B. sphaericus, 1 adet B. thuringiensis türlerine ait izolatlar) toplam 13 adet biyoajan bakteri ve yine daha önce yapılan çalışmalarda etkililiği farklı bitki patojenlerine test edilerek moleküler sistemde tanılanmış olan Tricoderma harzianum türüne ait toplam 2 fungus izolatı kullanılmıştır. Bu bakterilerin ve fungusların patojene karşı etkililiği in vitro şartlarda test edilmiş ve en etkili sonuç veren 6 bakteri ve 1 fungus izolatının etkililiği in vivo şartlarda da belirlenmiştir. Bu amaçla 3 tekerrürlü olarak kurulan saksı denemelerinde 3'er saksıya sadece patojen fungus izolatı, 3'er saksıya sadece biyoajan bakteri ve biyoajan fungus izolatı ve 3'er saksıya da patojen fungus ve biyoajan bakteri ve biyoajan fungus izolatı birlikte uygulanmıştır. Yapılan çalışmanın sonucunda B. amyloliquefaciens türüne ait 2 izolatın ve T. harzianum'un 1 izolatının Alternaria solani'ye karşı mücadelede in vivo şartlardada etkili olduğu tespit edilmiştir. Ayrıca T. harzianum izolatının bitki gelişimi üzerinde olumlu etki yaptığı da belirlenmiştir. Sonuç olarak; en etkili bulunan 2 bakteri ve 1 fungus izolatının domates yetiştiriciliğinde A. solani' nin kontrolünde biyokontrol ajanı olarak kullanılabileceği belirlenmiştir. Concerns about the adverse effects of environment, human and animal health, the endurance and the problem of residuals on chemical used in the controling against plant diseases have prompted scientists to seek new, more economical and effective ways of alternative methods to chemical control. One approach that comes to mind in this context is the use of microbial bioassays to control diseases and pests. For this reason, it is very vital to decide the biological controling agents against the pathogen and to offer effective uses. This study tries to determine of fungus and bacterial isolates that can be as biological control agents against. This study used the data in the previous studies; a total of 13 bioassay bacteria identified according to the Microbial Identification System (MIS), isolated from underground or overland parts of wild and cultivated plants, and also tested for effectiveness in previously conducted studies on different plant pathogens and detected in the molecular system a total of 2 isolates of Trichoderma harzianum were used. The effectiveness of these bacteria and fungi against pathogens has been tested in vitro conditions and the effectiveness of 6 most effective bacteria and 1 fungus is determined in vivo conditions. For this purpose, 3 potted plants were constructed with 3 repetitions, 3 pots were isolated only as pathogenic fungus isolate, 3 pots were only bioassay bacteria and bioassay fungus isolates and 3 pots were pathogenic fungus and bioassay bacteria and bioassay fungus isolate. As a result of the study, 2 isolates belonging to the Bacillus amyloliquefaciens strain and 1 isolate of T. harzianum has been found effective against A. solani in vivo. It has also been determined that T. harzianum isolate has a positive effect on plant growth. As a result; it is identified that the application of 2 bacteria, 1 fungus isolate, which is most effective, can be used as a biocontrol agent in the control of A. solani in tomato production. 83

The 22 isolates of them were tested for their sensitivities to spesific mode of action 6 fungicides. A.solani is a heterocaryotic organism and İt has high sporulation ability. Therefore, intensive and misuse pesticide applications could give rise to development of less sensitivity to fungicides. In our tests, sensitivities of the isolates to azoxystrobin, iprodion, imazaliL, prochloraz, fenhexamid and myclobutanil were studied, and in conclusion, imazalil was found the most effective fungicide to A.solani, and prochloraz and myclobutanil also were effective to the A.solani isolates, respectively. 39 ÖZET Bu çalışmada V. psalliotae ' nin A. solanı ile biyolojik savaşımda kullanılma olanakları dünyada ilk kez incelenmiştir. Önce antagonistic A.solani sporlarını parazitlediği ortaya konmuş, daha sonra saksı ve sera denemelerinde antagonist uygulamasının hastalığa karşı sırasıyla %100 ve %43.4 koruyuculuk sağladığı bulunmuştur. Çalışmanın bir başka bölümünde V. psalliotae' nin A.solani ile kimyasal savaşımda kullanılan Mancozeb, Iprodion, Bakı roksiklorür ve Chlorthalonil'in farklı dozlarına (10, 30, 100, 300 ppm) karşı in vitro duyarlılığı araştırılmıştır. Hesaplanan ED__ değerleri Mancozeb,` Iprodion, Bakı roksiklorür için > 300 50 ppm ve Iprodion için 180 ppm'dir. Sonuçlar, antagonistin sadece Iprodion' a duyarlı sayılabileceğini, diğer fungisidlerle birlikte ise entegre savaşım olanakları açısından incelenmesi gerektiğini ortaya koymaktadır. Ayrıca, gerek in vitro, gerekse saksı denemeleri için yeterli miktarda A.solani sporunun elde edilmesi için literatürde önerilen yöntemler denenmiş ve başarılı olmadıkları anlaşılmıştır. Bu ihtiyaçtan doğan arayış sonucunda SHAHIN ve SHEPARD (1979) yönteminden yola çıkarak şekersiz Y-ortamından ve soğuk stresinden yararlanılan yeni ve başarılı bir yöntem geliş tirilmiş tir. V. psalliotae ' nin kitle halinde üretilrr^si amacıyla patates sakkaroz sıvı ortamı (PS) seçilmiştir. Çünkü fungus bu ortamda PSA ortamına göre daha hızlı ve yoğun gelişmektedir. Ayrıca bu ortamda agar bulunmadığı için, antagonistin üretim maliyeti hiçbir kimyasal preparatla kıyaslanmayacak kadar da düşük olmaktadır. 40 SUMMARY In this study, the possibility of using Verticillium psalliotae in the biological control of Alternaria solani was investigated for the first time. Initially, it was clearly demonstrated by light microscopy that the antagonist could parasitize the host conidia. In the further pot and greenhouse experiments it was demonstrated that the application of V. psalliotae protected tomato plants by 100 and 43.6 %, respectively. In an additional investigation performed in vitro to gain information on the sensitivity of V. psalliotae towards the different doses (10, 30, 100 and 300 ppm) of some fungicides commonly used in the chemical control of early blight, LD values were calculated as > 300 ppm for Mancozeb, Copperoxychlorid and Chlorthalonil, and 180 ppm for Iprodion. Our data indicated that the antagonist can be assumed as having some sensitivity only against Iprodion. Further studies are needed to reveal the possibilities of using V. psalliotae and the fungicides in an integrated control program against early blight of tomato. In addition, some methods suggested at the literature were tested to obtain enough amounts of conidium of A. solani for both in vitro and pot experiments and resulted in failure. Therefore, based on the method by SHAHIN and SHEPARD (1979), a new and41 more successful method was developed to stimulate the sporulation °* A. solani in culture using a Y-medium without sugar and exposing the fungus to cold stress. Potato saccharose liquid medium (PS) was observed to be the most prefarable for the mass production of V. psalliotae. 48

105 ÖZET Altemaria solani (Ellis and Martin) Sorauer'in neden olduğu Erken Yanıklık hastalığı domateslerde hemen her yıl görülen en önemli yaprak hastalıklarından biridir. Bu hastalık son yıllarda Ege, Akdeniz ve Marmara bölgesindeki açık domates ekim alanları ve özellikle seralarda ciddi za rarlara neden olmaktadır. Bu çalışmada özellikle A.solani'nin kimyasal savaşım olanaklarını araştırmak gayesi güdülmüş, ayrıca etmenin invitro'da sporulasyonu, morfolojik ve mikros kobik özellikleri, çeşit reaksiyonlarına yönelik araştırma larda yapılmıştır. İnvitro'da A.solani'nin en iyi sporulasyonu, V-8, V-88 veya domates suyu agar besiyerinde bir dizi sıcaklık ve ışık koşulları altında, 23 C'de 6 gün karanlık, 2 6°C'de yakın ultraviyole ışığında 12 saat aydınlık, 18 C'de en az 12 saat karanlık inkubasyonu sonucu elde edildi. 1986-1989 yıllarında Ege, Akdeniz ve Marmara böl gelerindeki seralardan ve açık domates ekim alanlarından 164 adet Altemaria spp. izolatı elde edildi. Süpermarmande domates çeşidi ile yürütülen patojenisite denemelerinde 97 izolat patojen, kalan 67' si saprofit bulundu. Elde edilen bu patojen izolatların sporlarıma kapasitesi, suda çözünebilir pigment oluşumu, havai miselyum rengi ve yapısı gibi bazı kültürel özellikleri, virülensleri belirlendi, izolatların havai miselyum renkleri yeşil, gri ve kahverenginin çeşitli tonlarında olmak üzere 6 grupta toplandı. Havai miselyum yapısı ise izolatların %48'inde pamuk ben zeri, %39'unda kadife benzeri, %13'ünde de keçe benzeri bir yapıya sahipti. İzolatların %73'ü sarıdan kırmızıya kadar değişen renklerde suda çözünebilir pigment oluştururken geride kalan %2 7' sinin pigment oluşturmadığı gözlendi.106 4 5 97 izolatın V-8 besiyerinde spor verme kapasitesi lü -2x10 spor /ml arasında değişti. Ayrıca bu çalışmada yukarıdaki farklı kültürel özellikleri temsil eden 22 a. solanı ' izolatının her birinde ayrı ayrı spor boyutları ve bölme sayıları hesaplandı. Buna göre; sporların ortalama olarak 14.3-23.4 ym ende, 58.9-108.5 ym boyda olduğu, boy+kuyruk uzunluğunun 14.2-299.6 ym arasında değiştiği, bölme sayısı nın ortalama 8-15, kuyruk sayısının da 1-3 arasında olduğu görüldü. Saksı koşullarında 31 domates çeşidinin sera ve açık a. solani izolatlanna karşı reaksiyonlarının belirlendiği denemelerde, domates çeşitlerinin farklı a. solanı izolatlanna karşı genelde farklı duyarlılık gösterdiği görül dü. Sırasıyla B-2274, C.Mejorado, Eurovite ve Petomech domates çeşitleri hem sera hem açık izolatına daha az duyarlı bulundu. A. solani'ye kimyasal savaşımda en etkili fungisidleri belirlemek gayesiyle; sera ve açık alanlardan elde edilen yaklaşık 60 a. solanı izolatı fungisid içeren ve içermeyen PDA besiyerinde miseliyal gelişim ve spor çimlenmesi denemeleriyle 12 fungiside (maneb, zineb, mancozeb, propineb, thiram, metiram, captan, dichlofluanid, chlorothalonil, vinelozolin, iprodion ve prochloraz)' karşı testlendi. Miseliyal gelişimle ilgili olarak ED5Q değerleri ve spor çimlenmesi içinde MIC değerleri hesaplandı. İnvitro'da yürütü len bu denemelerde prochloraz, dichlof luanid, iprodion, maneb ve mancozeb' in A. solani'ye en etkili fungisidler olduğu saptandı. Ayrıca, fungisitlere duyarlılığı azalmış olarak bulunan 11 a. solanı izolatında duyarlılık azalışının kalıcılığı ve bu izolatların büyüme hızları belirlendi. Fungisid içermeyen PDA besiyerinde geriye 10 transfer sonrasında 11 izolat için saptanan ED5Q değerleri, izolatların genelde fungisidlere duyarlı olduğunu göstermekle kalmadı, zineb,107 mancozeb, propineb ve vinclozolin'e duyarlılığı azalmış olan bazı izolatlara da rastlandı. Bu denemelerde duyarlılık azalışı ile doğaya uyum arasında belirgin bir ilişki olmadığı gözlendi. Saksı denemeleri, dichlofluanid, iprodion ve maneb'in uygulama dozunun yarısında bile a. Solani'yi etkin bir şekilde kontrol edebileceğini, buna karşın mancozeb' in bu fungisidlere oranla daha az etkili olduğunu ortaya koydu. 108 SUMMARY Early Blight caused by Altemaria soiani (Ellis and Martin) Sorauer is one of the most important annually- occurring foliage diseases of tomotoes. In Ege, Akdeniz and Marmara regions in recent years, this disease has caused serious damages especially in greenhouses, and open tomoto growing areas. This study has been undertaken to investigate the possibilities of chemical control of Early Blight in particular. In addition, sporulation of A. soiani in vitro, morphological and microskobic features of A. soiani isolates, cultivar reactions to a. soiani has been searched. Successive incubation under the following light and temperature conditions -6 days at 23 C in dark, 12 hours at 26°C under near ultraviolet light, at least 12 hours at 18°C in dark-using V-8, V-88 or tomato juice agar gave the best result for inducing sporulation of A. soiani in vitro. In 1986-1989, 164 Altemaria spp. isolates were obtained from greenhouses and open tomato growing areas in Ege, Akdeniz and Marmara regions. While 97 isolates were found to be pathogen on tomato plants (ev. Süpermarmande) the remainder 67 was saprophyt. Some cultural features such as texture and pigmentation of aerial, mycelium, ability of water soluble pigmentation and sporulative capacity, and virulence of 97 A. soiani isolates were examined. In 97 isolates, 6 different shades of green, grey and brown pigmentation were noticed. Cotton like texture of aerial mycelium was 48% of isolates, velwet-like (39%) and felt-like (13%). 73 per cent of isolates produced water soluble pigment differing from yellow to red, while no pigmentation occurred in the remaining 27%. The capacity 4 5 for sporulation in 97 isolates varied from 10 to 2 x 10 spore/ml on V-8 juice agar. Dimensions and number of cell109 walls in a populations of spores each of 22 representative A.solani isolates were also measured. The average dimensions of different isolates varied from 14.3 to 23.4 ym for width, from 58.9 - 108.5 ym for length, from 14.2-299.6 ym for body beak length. The number of cell walls was 8-15, the number of beaks 1-3 on an average. In pot experiments, thirty one tomato varieties tested for reaction to greenhouse and field isolates of A.solani, varying levels of sensitivity to different A.so lani isolates were commonly observed among tomato varieties. Some tomoto cultivars were found to be less sensitive to both isolates, in the following descending order: B-2274, C.Mejorado, Eurovite, Petomech. To find out the most effective fungicides for the chemical control of A.solani, some 60 A.solani isolates obtained from greenhouse and field were tested for their response to 12 fungcides (maneb, zineb, mancozeb, propineb, thiram, metiram, çaptan, dichlofulanid,chlorothalonil, vinclozolin iprodion and prochloraz) with linear growth and spore germination experiments on PDA with and without fungicide. The EDj-0 values for growth inhibition and the MIC values for spore germination were calculated. The results of these i«. vitro studies showed that prochloraz, dichlof luanid, iprodion, maneb and mancozeb were the most effective fungicides. Furthermore, growth rate and the stability of 11 A.solani isolates showing decreased sensitivity to fungicides were also determined. The EDgQ values of 11 iso lates after 10 passages on unamended PDA indicated that most isolates were sensitive to fungicides in general. However, some isolates turned out to be reduced sensitivity to zineb, mancozeb, propineb and vinclozolin. In these assays, no significant correlations were observed between decrease in sensitivity and fitness.no Pot tests proved that dichlofluanid, iprodion and maneb could effectively control A. solani, even if these fungicides were used half of their recommended dosages, while mancozeb was slightly less effective than the other three. 132
