In recent years, transgenic technology has developed rapidly, but the risk of the environmental release of transgenic organisms is still a key issue. Research on the impact on biodiversity is an effective way to objectively evaluate the risk. By taking transgenic maize HGK60 with insect-resistant gene Cry1Ah and common maize Zheng 58 as control, a 2-year experiment of arthropod community biodiversity in fields of them were studied using three methods.in 2019 and 2020. The results showed that a total of 124 species and 38537 individuals were observed from the experiment, belonging to 11 orders and 40 families. There was no significant difference in the individual number and species number of herbivorous, predatory and parasitic groups in the two kinds of maize in two years. Only the individual number of HGK60 was significantly higher than that of common maize Zheng 58 at heading stage in 2019. And the percentages of individual number and species number in different groups were basically the same in the two kinds of maize at each stage in two years. Analyses of Richness index, Shannon-Wiener diversity index, Dominance index and Evenness index showed no significant difference between the two kinds of maize in two years. The similarity coefficient of the arthropod community suggested that the arthropod community composition of HGK60 was similar to that of common maize Zheng 58. Furthermore, HGK60 had no significant effect on the relative stability of the arthropod community. These results indicated that despite the presence of a relatively minor difference in arthropod community between the two kinds of maize, the planting of HGK60 had little effect on arthropod community biodiversity. Endophytic bacteria isolated from non-transgenic and transgenic Roundup Ready® glyphosate-resistant (GR) soybean plants were investigated to analyze the correspondence between phenotypic and genotypic characteristics and to determine whether or not the strains could be grouped based on the source of isolation in transgenic or non-transgenic plants, respectively. Most of the strains recovered from GR plants have shown the ability for plant growth promotion (PGP) by means of IAA production and inorganic phosphate solubilization, and 100% of the strains showed great motility (swarm or swim); in addition, 90% of the strains were able to metabolize the majority of carbon sources tested. GR soybean fields showed higher endophytes abundance than non-transgenic; however, analyzing the phylogenetic trees constructed using the partial 16SrRNA gene sequences, higher diversity was observed in non-transgenic soybean fields. Overall the majority of isolated endophytes could utilize multiple patterns of carbon sources and express resistance to antibiotics, while isolates varied widely in the PGP ability. The greater pattern and frequency of utilization of carbon sources and frequency and intensity of antibiotic resistance compared with PGP ability within the soybean endophytes community suggest that carbon sources metabolism and antibiotic resistance confer a greater relative fitness benefit than PGP ability. In conclusion, cluster analysis of the phenotypes and 16SrRNA gene sequences reveals lack of correspondence between the pattern of bacterial isolates and the transgenic character of plants, and the heterogeneity of clustering suggested that various adaptive processes, such as stress response, could have contributed to generate phenotypic variability to enhance endophytes overall fitness. Cry protein expressing insect-resistant trait is mostly deployed to control major devastating pests and minimize reliance on the conventional pesticides. However, the ethical and environmental issues are the major constraints in their acceptance, and consequently, the cultivation of genetically modified (GM) crops has invited intense debate. Since root exudates of Bacillus thuringiensis (Bt) crops harbor the insecticidal protein, there is a growing concern about the release and accumulation of soil-adsorbed Cry proteins and their impact on non-target microorganisms and soil microbial processes. This review pertains to reports from the laboratory studies and field trials to assess the Bt toxin proteins in soil microbes and the processes determining the soil quality in conjunction with the existing hypothesis and molecular approaches to elucidate the risk posed by the GM crops. Ecological perturbations hinder the risk aspect of soil microbiota in response to GM crops. Therefore, extensive research based on in vivo and interpretation of results using high-throughput techniques such as NGS on risk assessment are imperative to evaluate the impact of Bt crops to resolve the controversy related to their commercialization. The cultivation of genetically modified plants (GMP) has raised concerns regarding the plants’ ecological safety. A greenhouse experiment was conducted to assess the impact of five seasons of continuous Bt (Bacillus thuringiensis) maize cultivation on the colonisation and community structure of the non-target organisms arbuscular mycorrhizal fungi (AMF) in the maize roots, bulk soils and rhizospheric soils using the terminal restriction fragment length polymorphism (T-RFLP) analysis of the 28S ribosomal DNA and sequencing methods. AMF colonisation was significantly higher in the two Bt maize lines that express Cry1Ab, 5422Bt1 (event Bt11) and 5422CBCL (MON810) than in the non-Bt isoline 5422. No significant differences were observed in the diversity of the AMF community between the roots, bulk soils and rhizospheric soils of the Bt and non-Bt maize cultivars. The AMF genus Glomus was dominant in most of the samples, as detected by DNA sequencing. A clustering analysis based on the DNA sequence data suggested that the sample types (i.e., the samples from the roots, bulk soils or rhizospheric soils) might have greater influence on the AMF community phylotypes than the maize cultivars. The advent of genetic engineering has revolutionized agriculture remarkably with the development of superior insect-resistant crop varieties harboring resistance against insect pests. Bacillus thuringiensis (Bt) has been used as a main source for insect-resistant genes. In addition to Bt endotoxins, various plant lectins and other non-Bt genes from different sources have also been introduced in crop plants of economic importance. The insect-resistant crops have made a huge economic impact worldwide since their commercial release. The cultivation of insect-resistant cultivars has resulted both in increased crop productivity and in decreased environmental pollution. Although insect-resistant crops have been allowed to be commercialized following proper biosafety guidelines and procedures, still these crops face many challenges in order to be fully adopted and accepted. The degradation kinetics of Bt proteins, horizontal and vertical gene flow, effects on nontarget insects or organisms, antibiotic resistance, and some other unintended effects have been noted and discussed. Although no concrete evidence regarding any significant hazard of genetically engineered crops has been presented so far, the debate still remains intense. Impartial and professionally competent regulatory mechanisms for the evaluation of insect-resistant and other transgenic crops must be fully functionalized. The first part of this review focuses the development of different insect-resistant crops and various strategies adapted to delay resistance development in insect pests, while the second part addresses the challenges and future prospects of insect-resistant crops. The use of genetically modified (GM) plants in agriculture has been a topic in public debate for over a decade. Despite their potential to increase yields, there may be unintended negative side-effects of GM plants on soil micro-organisms that are essential for functioning of agro-ecosystems. Fungi are important soil organisms and can have beneficial or harmful effects on plants. Their benefits to agro-ecosystems come from their activities as free-living saprobes breaking down soil organic matter thereby releasing nutrients to the crops, as well as from mutualistic interactions. On the other hand, soil-borne plant pathogenic fungi can cause severe damage in crops. Understanding of the impact of GM plants on the dynamics and functioning of soil fungi is essential to evaluate the possible risks of introduction of GM plants for ecosystem functioning. In recent years, over 50 studies have addressed the effects of various GM traits in crops on soil fungal community structure and function. These studies showed that GM crops can have positive, negative, or neutral effects on both free-living and plant-associated soil fungi. The observed discrepancy in results of these studies is discussed. This is done by highlighting a number of case studies. New methods developed in recent years have enabled microbial ecologists to get a better picture on the functioning and assembly of soil fungal communities. This review presents and discusses two of the most promising methods which are also readily usable in risk assessment of GM plants on soil fungi and that could help answer remaining key questions in the field. A laboratory soil degradation study was conducted to determine the biodegradation potential of a DvSnf7 dsRNA transcript derived from a Monsanto genetically modified (GM) maize product that confers resistance to corn rootworm (CRW; Diabrotica spp.). This study provides new information to improve the environmental assessment of dsRNAs that become pesticidal through an RNAi process. Three agricultural soils differing in their physicochemical characteristics were obtained from the U.S., Illinois (IL; silt loam), Missouri (MO; loamy sand) and North Dakota (ND; clay loam), and exposed to the target dsRNA by incorporating insect-protected maize biomass and purified (in vitro-transcribed) DvSnf7 RNA into soil. The GM and control (non-GM maize) materials were added to each soil and incubated at ca. 22°C for 48 hours (h). Samples were collected at 12 time intervals during the incubation period, extracted, and analyzed using QuantiGene molecular analysis and insect bioassay methods. The DT50 (half-life) values for DvSnf7 RNA in IL, MO, and ND soils were 19, 28, and 15 h based on QuantiGene, and 18, 29, and 14 h based on insect bioassay, respectively. Furthermore, the DT90 (time to 90% degradation) values for DvSnf7 RNA in all three soils were <35 h. These results indicate that DvSnf7 RNA was degraded and biological activity was undetectable within approximately 2 days after application to soil, regardless of texture, pH, clay content and other soil differences. Furthermore, soil-incorporated DvSnf7 RNA was non-detectable in soil after 48 h, as measured by QuantiGene, at levels ranging more than two orders of magnitude (0.3, 1.5, 7.5 and 37.5 µg RNA/g soil). Results from this study indicate that the DvSnf7 dsRNA is unlikely to persist or accumulate in the environment. Furthermore, the rapid degradation of DvSnf7 dsRNA provides a basis to define relevant exposure scenarios for future RNA-based agricultural products. [ABSTRACT FROM AUTHOR]

Genetically modified (GM) maize containing genes from the soil bacterium Bacillus thuringiensis (Bt) was cultivated on 29% of the total maize production area worldwide in 2009. Most studies to date compare Bt-maize varieties with their near isogenic lines; however, there is little information on the variability of conventional maize breeding lines and how the effects of Bt varieties are ranked within. In our study on the potential risks of Bt-maize varieties, we analyzed tissue quality and compared the effects of ten conventional and GM maize varieties on soil microbiological properties in a replicated climate chamber experiment. All maize varieties were cultivated twice in the same soil microcosm. Shoot yields and soluble C in leaf tissue of Bt varieties were higher than the ones of non- Bt. Soil dehydrogenase activity was reduced by 5% under Bt varieties compared to non- Bt, while most of the other soil microbial properties (soil microbial biomass, basal respiration) showed no significant differences between Bt and non- Bt varieties. The leaves and roots of one Bt variety were decomposed to a greater extent than the ones of its near isogenic line; the conventional breeding lines also showed higher values. Changes in crop and soil parameters were found when comparing the first and the second crops, but the effects of repeated cropping were the same for all tested varieties. Transgenic Bt cotton expresses the Cry1Ac protein from Bacillus thuringiensis, which could influence the plant's capability to host endophytic fungi. The diversity of endophytic fungi in leaves, stems and roots from transgenic (Bt) and its isoline (non-Bt) cotton was evaluated during different plant developmental stages to investigate possible non-target effects of genetically modified cotton on endophytic fungal communities. A total of 17 genera of endophytic fungi were isolated. The most frequently isolated species were Phomopsis archeri from leaves and stems and Phoma destructiva from roots. While the Bt modification had no effect on endophytes, the cotton tissue and the plant developmental stage significantly influenced the diversity and composition of the fungal community. Abstract  The rhizosphere is a niche exploited by a wide variety of bacteria. The expression of heterologous genes by plants might become a factor affecting the structure of bacterial communities in the rhizosphere. In a greenhouse experiment, the bacterial community associated to transgenic eucalyptus, carrying the Lhcb1-2 genes from pea (responsible for a higher photosynthetic capacity), was evaluated. The culturable bacterial community associated to transgenic and wild type plants were not different in density, and the Amplified Ribosomal DNA Restriction Analysis (ARDRA) typing of 124 strains revealed dominant ribotypes representing the bacterial orders Burkholderiales, Rhizobiales, and Actinomycetales, the families Xanthomonadaceae, and Bacillaceae, and the genus Mycobacterium. Principal Component Analysis based on the fingerprints obtained by culture-independent Denaturing Gradient Gel Electrophoresis analysis revealed that Alphaproteobacteria, Betaproteobacteria and Actinobacteria communities responded differently to plant genotypes. Abstract  The impact of multiple-year (0–5 years) cultivation of transgenic Bacillus thuringiensis (Bt) cotton on the functional bacterial populations in rhizosphere soil was investigated. The transgenic Bt + CpTI cotton line SGK321 and a non-Bt cotton line Shiyuan321 were planted in four fields in which Bt cotton had been continuously cultivated for 0, 1, 3, and 5 years. Rhizosphere soil samples were collected at the seedling, squaring, flower and boll, and boll-opening stages of cotton. Numbers of bacteria involved in nitrogen-fixing, organic phosphate-dissolving, inorganic phosphate-dissolving, and potassium-dissolving were measured with cultivation-dependent approaches. The data presented here showed no consistent statistically significant differences in the numbers of different groups of functional bacteria between rhizosphere soil of Bt and non-Bt cotton in the same field, and no obvious trends in the numbers of the various group of functional bacteria with the increasing duration of Bt cotton cultivation. These studies suggest that multiple-year cultivation of transgenic Bt cotton may not affect the functional bacterial populations in rhizosphere soil. [ABSTRACT FROM AUTHOR]

The introduction of genetically modified (GM) plants in agroecosystems raises concern about possible effects on nontarget species. The impact of a tomato line transformed for constitutive expression of tobacco β-1,3-glucanase and chitinase on indigenous nonpathogenic fungi was investigated. In greenhouse experiments, no significant differences were found in the colonization by arbuscular mycorrhizal fungi. Diversity indices computed from over 20 500 colonies of culturable rhizosphere and phyllosphere saprotrophic microfungi, assigned to 165 species (plus>80 sterile morphotypes), showed no significant differences between GM and wild-type plants. Differences were found by discriminant analysis in both the rhizosphere and the phyllosphere, but such effects were minor compared with those linked to different plant growth stages. Abstract  Pink-pigmented facultative methylotrophs (PPFMs) are one of the beneficial proteobacteria commonly found in phyllosphere, rhizosphere and as endophytes in cotton. To assess the impact of transgenic Bt-cotton on changes in the diversity and community profile of PPFMs by comparing with its non-transgenic cotton, a polyphasic approach including differential carbon-substrate utilization profiling and DNA fingerprinting techniques like ARDRA, RISA, BOX-PCR and ERIC-PCR were studied. PPFMs from phyllosphere, rhizoplane and internal tissues of the stem of both Bt-cotton and non-Bt-cotton were isolated and analysed in this study. All the results suggested that the diversity richness of PPFMs present in the phyllosphere, rhizoplane and internal tissues did not differ between Bt- and non-Bt-cotton. Proteinase inhibitors (PI) are present in plant tissues, especially in seeds, and act as a defense mechanism against herbivores and pathogens. Serine PI from soybean such as Bowman-Birk (BBPI) and Kunitz have been used to enhance resistance of sugarcane varieties to the sugarcane borer Diatraea saccharalis (Fabricius) (Lepidoptera: Crambidae), the major pest of this crop. The use of these genetically-modified plants (GM) expressing PI requires knowledge of its sustainability and environmental safety, determining the stability of the introduced characteristic and its effects on non-target organisms. The objective of this study was to evaluate direct effects of ingestion of semi-purified and purified soybean PI and GM sugarcane plants on the soil-dwelling mite Scheloribates praeincisus (Berlese) (Acari: Oribatida). This mite is abundant in agricultural soils and participates in the process of organic matter decomposition; for this reason it will be exposed to PI by feeding on GM plant debris. Eggs of S. praeincisus were isolated and after larvae emerged, immatures were fed milled sugarcane leaves added to semi-purified or purified PI (Kunitz and BBPI) or immatures were fed GM sugarcane varieties expressing Kunitz and BBPI type PI or the untransformed near isogenic parental line variety as a control. Developmental time (larva-adult) and survival of S. praeincisus was evaluated. Neither Kunitz nor BBPI affected S. praeincisus survival. On the other hand, ingestion of semi-purified and purified Kunitz inhibitor diminished duration of S. praeincisus immature stages. Ingestion of GM senescent leaves did not have an effect on S. praeincisus immature developmental time and survival, compared to ingestion of leaves from the isogenic parental plants. These results indicate that cultivation of these transgenic sugarcane plants is safe for the non-target species S. praeincisus. [ABSTRACT FROM AUTHOR]

Soil properties, microbial communities, and enzyme activities were studied in soil planted with transgenic or nontransgenic papaya under field conditions. The transgenic papaya contained a replicase (RP) mutant gene of the papaya ringspot virus (PRSV), which conferred resistance to the virus, the neomycin phosphotransferase II (NPT II) marker gene, which conferred Km resistance, and a cauliflower mosaic virus 35S promoter (CaMV 35S). There were significant differences ( P < 0.05) in the total number of colony forming units (CFUs) of bacteria, actinomycetes, and fungi between soils planted with RP-transgenic and nontransgenic plants; total CFUs of bacteria, actinomycetes, and fungi in soil planted with transgenic papaya were significantly higher by 0.43, 0.80, and 0.46 times, respectively. Significantly higher ( P < 0.05) CFUs of bacteria, actinomycetes, and fungi resistant to kanamycin (Km) were present in soils planted with the transgenic papaya than in those planted with nontransgenic papaya. Resistance quotients (CFU in the presence of a chemical relative to that without) of Km-resistant bacteria, actinomycetes and fungi were higher in soil planted with transgenic papaya, and the resistance quotients of Km-resistant bacteria, actinomycetes, and fungi in soils planted with transgenic papaya increased statistically significantly ( P<0.05) from 1.5 to 2.5, from 1.2 to 2.6, and from 0.9 to 2.8 times, respectively. Soils planted with transgenic papaya had significantly higher enzyme activities of arylsulfatases (+5.4 times), alkaline phosphatases (+0.5 time), invertase (+0.5 time) and phosphodiesterases (+0.2 time), but lower enzyme activities of proteases (−2.1 times), polyphenol oxidases (−1.4 times), urease (−0.2 time) than the soils planted with nontransgenic papaya. Our results suggest that transgenic papaya could alter chemical properties, enzyme activities, and microbial communities in soil. [ABSTRACT FROM AUTHOR]
