Mit einer polyvalenten Peroxidase wurde ein Schlüsselenzym des Lignocelluloseabbaus aus Kulturen des Basidiomyceten Pleurotus sapidus chromatographisch gereinigt. Nach einer dreistufigen Reinigung wurde das Enzym mit einem Reinigungsfaktor von 130 aus dem Überstand einer Submerskultur isoliert. Die Aktivitätsoptima wurden für die Substrate b,b-Carotin, ABTS, Syringol und Veratrylalkohol ebenso bestimmt wie die Wechselzahlen und katalytischen Konstanten. In Zusammenarbeit mit der Firma Artes Biotechnology GmbH wurde ein Hansenula polymorpha Stamm generiert, welcher die für die polyvalente Peroxidase aus Pleurotus sapidus kodierende cDNA enthielt. Das Enzym wurde heterolog exprimiert und die Aktivitätsoptima des rekombinanten Enzyms ermittelt. Anschließend wurde der Abbau der Substrate Rapsstroh und „Rückstand einer Biogasanlage“ durch den Basidiomyceten Pleurotus sapidus untersucht. Dazu wurde die Gesamtheit der sekretierten Enzyme (das Sekretom) von Pleurotus sapidus in Submers- und Emerskulturen analysiert. Photometrisch bestimmt wurden die Aktivitätsverläufe von Laccasen, Esterasen, Lipasen, Cellulasen, Xylanasen und Peroxidasen, insbesondere von Manganperoxidasen und polyvalenten Peroxidasen. Das Sekretom von Pleurotus sapidus wurde anschließend mittels zweidimensionaler Elektrophorese getrennt, massenspektrometrisch identifiziert und bioinformatisch ausgewertet. Verschiedene Substrate wurden mit gereinigten Enzympräparationen und Kulturüberständen von Pleurotus sapidus umgesetzt. Hierbei wurde die Umsetzung von Ligninmodellsubstanzen wie zum Beispiel Coniferyl- und Veratrylalkohol, aber auch von Lignin organosolv und feingemahlenem Rapsstroh durch die gereinigte polyvalente Peroxidase aus Pleurotus sapidus gezeigt. Ebenso wurde ein Umsatz von feingemahlenem Rapsstroh mit Pleurotus sapidus Submerskulturen nachgewiesen. Die Versuche wurden analytisch durch photometrische Bestimmungen (Glucose- und Phenolgehalt), Messung der Partikelgrößenverteilungen und HPLC-/HPSEC-Messungen ausgewertet. After three steps, the enzyme was isolated with a purification factor of 130. Optimal reaction conditions were determined together with turnover numbers and product formation rates for the substrates: b,b-carotene, ABTS, syringol, and veratryl alcohol. In cooperation with Artes® Biotochnology, a Hansenula polymorpha strain was generated containing the cDNA encoding the versatile peroxidase of Pleurotus sapidus. The enzyme was expressed heterologously, and pH and temperature optima were determined for the recombinant enzyme. The degradation of the lignocelluloses rape straw and residues from a biogas plant by Pleurotus sapidus were studied. Therefore, the entirety of enzymes (“the secretome“) secreted by Pleurotus sapidus in submerged and surface cultures were analysed. The activities of laccases, esterases, lipases, cellulases, xylanases, and peroxidases, in particular manganese peroxidases and versatile peroxidases were calculated by photometric assays. The secretome of Pleurotus sapidus was separated by two-dimensional electrophoresis and identified by mass spectrometry. A bioinformatic analysis of the data was performed. In-vitro digestions of various substrates were carried out with the isolated versatile peroxidase and culture supernatants of Pleurotus sapidus. The conversion of various low molecular weight lignin model substances like coniferyl alcohol or veratryl alcohol, finely ground rape straw and lignin organosolv were investigated. The conversion of finely ground rape straw was demonstrated with submerged cultures of Pleurotus sapidus. The product analysis was performed by photometric assays, determination of particle size distribution, and HPLC-/HPSEC analysis.
