Objectives: Fluorescence spectroscopy of human urine is a method with the potential to gain importance as a diagnostic tool in the medical field, e.g., for measuring Coproporphyrin III (CPIII) as an indicator of cancer and acute types of porphyria. Food can change human urine's color, which could influence the urine fluorescence spectrum and the detection of CPIII in urine. To determine if there is a noticeable influence on the urine fluorescence spectrum or on the detection of CPIII in urine, 16 vitamin supplements, and three food items were tested. Such investigation may also prevent false interpretation of measured data., Methods: Urine samples were collected before and after (overnight, ca. 8 h) intake of each test substance. Samples were investigated by fluorescence spectrum analysis. At excitation wavelengths from 300 to 500 nm and emission wavelengths from 400 to 700 nm excitation-emission-matrices were measured. Data obtained from urine before intake were compared to the data from overnight urine. Furthermore, the investigation of any interference with the CPIII concentration was performed at an excitation wavelength of 407 ± 3 nm and emission wavelengths of 490-800 nm., Results: Only vitamin B2, but none of the other tested substances, showed noticeable influence on the urine fluorescence spectrum. None of the tested substances showed noticeable interference with the recovery rate of CPIII., Conclusions: The correct interpretation of measured data by fluorescence spectroscopy is possible with the exception if vitamin B2 supplementation was performed; thus, the consumption of vitamin B2 supplements before fluorescence testing of the patient's urine should be avoided and/or must be requested. The main sources of vitamins, which are essential substances, are mainly aliment products, foods for special dietary uses and dietary supplements. Therefore, the study of the native content of vitamins in aliment foods has always been of interest. For the chromatographic separation of vitamins, rather versatile C18 columns are used as a stationary phase, which allow one to obtain reliable results using UV detection for vitamin-enriched foods, dietary supplements and vitamin premixes. However, for unfortified foods, this stationary phase in a UV detection system does not give acceptable results. The aim of the work was to develop a technique for the chromatographic separation of vitamins B1 and B2 in unfortified foods using a diode array detector. Material and methods . To prepare samples of foods, concentrated acid hydrolysis (1.0 g of sample and 4 ml of 0.1 N hydrochloric acid) was carried out in a water bath for 30 min at a temperature of 95 °C, followed by enzymatic hydrolysis and degreasing. Further studies of the samples were carried out on an Agilent Technologies 1100 chromatographic system with diode array detection. For the determination of vitamin B1, a Poroshell 120 Hilic column 4.6×150 mm, grain size 2.7 μm was used. As eluent A, a 10 mM aqueous solution of ammonium acetate with 0.5% acetic acid was used, eluent B was acetonitrile (gradient elution: 0-2 min - 90% B, 8-12 min - 50% B, 14-18 min - 90% B). To determine vitamin B2, a C18 Poroshell column 4.6×250 mm, grain size 5 μm was used. As eluent A, a classical phosphate buffer with pH 2.5 was used, eluent B - acetonitrile (gradient elution: 0-5 min - 0% B, 5-15 min - 90% B, 15-22 min - 90% B, 22-24 min - 0% B, 24-27 min - 0% B). Vitamin B1 was detected at a wavelength of 270 nm, vitamin B2 at 450 nm. Under selected conditions, good retention and efficient separation of vitamins B1 (over 16,000 theoretical plates) and B2 (over 20,000 theoretical plates) was observed. Results . It was demonstrated that the HPLC method with diode array detection can be used to quantify the native content of vitamins B1 and B2 in products with a complex food matrix. For the selective determination of these vitamins, a complex of chromatographic conditions is optimal: reverse-phase HPLC for vitamin B2 and hydrophilic interaction chromatography for vitamin B1. A suitable sample preparation of food products for the content of vitamins B1 and B2 under selected chromatographic conditions is concentrated acid-enzymatic hydrolysis. The limit of quantitation for vitamins B1 and B2 was 40 μg/100 g. Comparison of the enzymatic activity of amylorizin and thermostable α-amylase showed that during long-term hydrolysis for 16 hours (37 °С) with amylorizin, the degree of vitamin extraction was two fold higher than during hydrolysis (95 °С, 1 h) with α-amylase. Conclusion . Background and Aim: Stroke represents a significant public health challenge, necessitating the exploration of preventive measures. This network meta‐analysis aimed to assess the efficacy of different vitamin treatments compared to a placebo in preventing stroke. Methods: A systematic electronic search in databases including PubMed, EmBASE, Web of Science, clinicaltrials.gov, and Google Scholar until 31 May 2023 was conducted, to identify published studies investigating the association between vitamin intake and the risk of stroke. Pooled risk ratio (RR) with 95% confidence intervals (CIs) was calculated using a frequentist network meta‐analysis. Furthermore, we ranked vitamins based on p‐scores, facilitating a comparative assessment of their effectiveness in preventing stroke. Results: A total of 56 studies, including 17 randomized controlled trials (RCTs) and 39 cohort studies were analyzed. Direct estimates obtained from network meta‐analysis, we found that vitamin A (RR:.81 [.72–.91]), vitamin B‐complex (RR:.85 [.74–.97]), vitamin B6 (RR: 79 [.68–.92]), folate (RR:.86 [.75–.97]), vitamin C (RR:.77 [.70–.85]) and vitamin D (RR:.73 [.64–.83]) were significantly associated with a decreased stroke risk. However, no significant association was observed for vitamin B2, vitamin B12, and vitamin E. Subsequent to network meta‐analysis, vitamins were ranked in decreasing order of their efficacy in stroke prevention based on p‐score, with vitamin D (p‐score =.91), vitamin C (p‐score =.79), vitamin B6 (p‐score =.70), vitamin A (p‐score =.65), vitamin B‐complex (p‐score =.53), folate (p‐score =.49), vitamin B2 (p‐score =.39), vitamin E (p‐score =.28), vitamin B12 (.13) and placebo (.10). Conclusion: Our study has established noteworthy connections between vitamin A, vitamin B‐complex, vitamin B6, folate, vitamin C, and vitamin D in the realm of stroke prevention. These findings add substantial weight to the accumulating evidence supporting the potential advantages of vitamin interventions in mitigating the risk of stroke. However, to solidify and validate these observations, additional research is imperative. Well‐designed clinical trials or cohort studies are needed to further explore these associations and formulate clear guidelines for incorporating vitamin supplementation into effective stroke prevention strategies. This study aimed to characterise the market‐driven fortification of vitamins and minerals in packaged foods targeted at children in Brazil. We analysed 535 food labels using data collected in a census‐type method (n = 5620) of food labels in a Brazilian supermarket in 2013. Micronutrients declared in nutrition claims and the ingredients list (synthetic compounds) were considered to be added for commercial purposes. Analysis of the ingredients list and nutrition claims showed that market‐driven fortification of vitamins and minerals was present in 27.1% of foods. The main vitamins and minerals were vitamins A, B complex, C, D, calcium, iron and zinc. The food groups 'Milk and dairy products' and 'Sugars, sugary foods and snacks' had the highest frequencies of micronutrients declared in the ingredients list. Calcium, iron, phosphorus, zinc and all vitamins, except B7, were found to be added for commercial purposes. Micronutrients were found to be commonly added to packaged foods as a marketing strategy directed at parents and their children. Future studies should assess the amount of vitamins and minerals added to packaged foods targeted at children and whether intakes of vitamins and minerals in children are potentially excessive. Heavy metals have negative impacts on human health including red blood cells hemolysis and increases oxidation processes, leading to many health issues. In this study, several blood samples were collected from people who work in North Oil Company, Kirkuk, Iraq. In addition to people who live, work away from oil fields as the control group. This investigation looks at the impact of various concentrations of cadmium and / or lead individually and combined (Cd:Pb) on erythrocytes hemolysis. The findings demonstrated that hemolysis produced by the (100 mM) were (72.0, 88.80, and 96.90%) for (Cd, Pb, and Cd: Pb) respectively. Vitamins (B2 and C) were applied with different concentrations (0.1, 0.2, 0.3, 0.4, and 0.5 mol/L). to investigate the reduction of hemolysis percentage. The findings showed that both vitamins have positive effect in reducing the hemolysis percentage. Vitamin C has even more significant impact in reducing the hemolysis compared to Vitamin B2. In conclusion, cadmium and lead can cause erythrocyte hemolysis individually or combined, additionally, vitamins C and B2 were proved to in reducing hemolysis effectively. [ABSTRACT FROM AUTHOR]

Sorghum and orange‐fleshed sweet potato (OFSP) flours were blended to produce composite flours at eight different ratios of 90:10, 80:20, 70:30, 60:40, 50:50, 40:60, 30:70, and 20:80, respectively, whereas 100% sorghumflour was used as control. The physicochemical compositions, nutritional and functional properties, as well as color attributes of the composite flour blends were evaluated. The acquired data were analyzed using ANOVA, and the means were separated using the Duncan multiple range test. Significant differences (p <.05) were observed in the physicochemical and nutritional properties of the flour blends. The protein levels in the composite flour decreased as the proportion of OFSP flour increased. However, the levels of vitamins, particularly vitamins A and C contents of the composite flours increased with higher proportions of OFSP, ranging from 0.27 and 1.74 mg/100 g in sample S100 to 2.13 and 2.12 mg/100 g in sample S20O80, respectively. In contrast, an increase in the percentage of OFSP flour resulted in a decrease in the contents of vitamin B‐complex, particularly vitamins B2 and B6. These values decreased slightly from 0.19 and 1.98 mg/100 g in sample S100 to 0.16 and 0.03 mg/100 g in sample S20O80, respectively. Furthermore, as the proportion of OFSP flour increased, there was a reduction in the calcium levels from 17.39 mg/100 g in the 100% sorghum sample to 13.52 mg/100 g in the S20O80 sample. However, no particular trend was observed in, magnesium, iron, and phosphorus levels. Sample S50O50 had the highest percentage of essential and conditional amino acids, except for cysteine, valine, and phenylalanine. The findings also revealed significant variations (p <.05) in the composite flour samples' functional properties and color measurements. Highly valued for their nutritional benefits, sprouts are characterized by high water content, which promotes microbial proliferation, potentially leading to toxicity and a reduced shelf life. To address this challenge, the present study explores the application of a novel drying–texturizing approach, named IVDV (Intensification of Vaporization by Decompression to the Vacuum), to sprouts. This technique would enable faster drying of the sprouts and better preservation of their nutritional content, compared to traditional hot-air drying. Using Response Surface Methodology, optimal IVDV parameters (saturated steam pressure P, processing time t, and water content W) for wheat sprouts were defined, with a focus on preserving vitamins, proteins, and lipids, and optimizing the expansion ratio. This optimization process identified optimal experimental conditions at 5.5 bars, a duration of 15 s, and 8.8% d.b. water content. Under these conditions, the use of IVDV endowed the expanded sprouts with a crunchier and more friable texture. In addition, it significantly improved the preservation of vitamins B6 and E by 412% and 42%, respectively, compared to traditional mild hot-air drying, without significantly affecting vitamin B2, proteins, and lipids. When combined with conventional hot-air drying, IVDV not only enhanced the preservation of the sprouts' nutritional content but also reduced drying time and energy consumption. Summary: Sickle cell disease (SCD) is associated with high rates of undernutrition and stunting. Undernutrition in combination with chronic haemolysis may lead to deficiencies in micronutrients necessary for erythropoiesis. Here we examined selected levels of ferritin, vitamins B2, B6, B9 and B12, and vitamin C that were measured in blood samples from 820 SCD patients from Tanzania with no history of hospital admission, infections or painful episodes in the previous 30 days. We studied children (0–8 years), early adolescents (9–14 years), late adolescents (15–17 years) and adults (≥18 years). Severely low levels of vitamin B12 were observed across the four age groups. Despite the lowered vitamin B12 concentrations, total homocysteine concentrations were normal across both genders in all age groups. We found no significant gender‐related differences between the other measured micronutrients. In this large SCD population, spanning the whole life cycle, a low level of vitamin B12 was consistently found across both genders and all age groups. Given the pivotal role of vitamin B12 in cellular metabolism, particularly in erythropoiesis, more studies are required to unravel how to better detect clinically relevant vitamin B12 deficiency among SCD patients, and thus to identify more precisely those who need supplementation of vitamin B12. Notwithstanding the increased interest in wild edible plants, little is known on how some domestic thermal processes can affect their content. The aim of this study was to investigate the amounts of minerals, B1 and B2 vitamins, tocols, and carotenoids in raw, boiled, and steamed wild edible plants, namely, Sonchus asper (L.) Hill s.l., Sonchus oleraceus L., Cichorium intybus L., and Beta vulgaris L. var cicla. All vegetables were confirmed as high sources of lutein (from 6 to 9 mg/100 g) and β-carotene (from 2 to 5 mg/100 g). Quite high amounts of violaxanthin and neoxanthin were found. Alfa-tocopherol and γ-tocopherol were the main tocols, with same contents in raw and processed vegetables (about 2.5 mg/100 g). The most abundant macro element and trace element were, respectively, potassium and iron. B1 and B2 vitamins were found in low amounts in almost all plants, with the exception of thiamine in Beta vulgaris (about 1.6 mg/100 g). Boiling led to a significant loss of minerals (up to 60%) and B-group vitamins (up to 100%), while, among carotenoids, it only affected violaxanthin levels (up to 90%). Steamed vegetables showed only a slight reduction, about 20%, in β-carotene and lutein, with a marked decrease in violaxanthin and neoxanthin. Ferroptosis featured by iron‐dependent lipid peroxidation plays a critical role in tumor regression, offering an alternative therapeutic target for the treatment of aggressive triple‐negative breast cancer. However, the traditional delivery strategy to enrich iron deposition is limited in producing a surplus of lipid oxidation because of the aggressive antioxidant response established in solid tumors. Here, vitamin B2‐ferric chloride nanocomplex self‐assembled through the coordination of ferric iron and hydroxy groups to simultaneously promote iron accumulation and oxidative stress is employed. It is observed that the self‐assembly nanostructures with appropriate size enable effective delivery of the therapeutic agents to the tumor site and cancer cells. More importantly, sonosensitizer vitamin B2 and ferric chloride cooperatively facilitate the production of reactive oxygen species and amplify nanocatalytic therapy. To further disrupt the antioxidant system, metformin with solute carrier family 7 member 11 inhibitory activity is used to synergistically boost ferroptosis of tumor cells. The combinational ferroptosis promoter exhibits augmented nanocatalytic antitumor efficacy in 4T1 tumor‐bearing mice with negligible adverse effects. A creatively designed novel two-step enhancement technique is presented in which B vitamin molecules are dynamically adsorbed onto the surface of silver nanoparticles by sodium borohydride, followed by local plasmon resonance in the presence of cations (calcium ions), ultimately achieving synergistic chemical and physical enhancement on the same molecule and constructing a "surface hot spots" two-step enhancement platform for vitamin detection. The Raman signal of the promoted vitamin molecule is enhanced by nine orders of magnitude. In a subsequent study it was observed that the vitamin B2 molecules were in a near-vertical image on the surface of the silver nanoparticles, which may also contribute to the Raman signal enhancement. Combined with deep learning techniques, the method has been successfully applied to the detection of B vitamins in body fluids. As an accurate, rapid, reproducible, non-invasive, and versatile assay platform, it holds great promise for the intelligent identification of trace B molecules in food, pharmaceuticals, and the human body. Beyond their nutritional benefits, vitamins could decrease the risk of chronic diseases due to their potent antioxidant capacity. The present work is aimed at reviewing the state of the art regarding (1) the vitamins involved in oxidative stress prevention in accordance with the requirements established by the European Food Safety Authority (EFSA) and (2) the foods of plant origin that are sources of those vitamins and have potential benefits against oxidative stress in humans. According to the European regulations based on EFSA scientific evidence, riboflavin, vitamin C, and vitamin E are those vitamins subjected to the approved health claim "contribute to the protection of cells from oxidative stress". Scientific studies conducted in humans with some natural food sources of riboflavin (almonds, wheat germ, mushrooms, oat bran), vitamin C (guava, kale, black currant, Brussels sprouts, broccoli, orange), and vitamin E (hazelnuts, almonds, peanuts, pistachio nuts, extra virgin olive oil, dates, rye) have been performed and published in the literature. However, no food of plant origin has obtained a favorable EFSA opinion to substantiate the approval of health claims related to its potential properties related to oxidative stress prevention. Further studies (concretely, well-controlled human intervention studies) must be carried out in accordance with EFSA requirements to provide the highest level of scientific evidence that could demonstrate the potential relationship between foods of plant origin and antioxidant capacity. This review could be useful for the scientific community to study the application of health claims referring to the antioxidant capacity potentially exerted by foods of plant origin. This study assessed the effect of the steamed and boiling cooking methods on the vitamin content of six staple green vegetables in Nigeria. Vitamin levels in fresh, steamed and boiled leafy vegetables were determined using spectrophotometric and titrimetric methods. The results of vitamin analysis showed that fresh vegetable of Amaranthus hybridus contained the largest amount of ascorbic acid, thiamin and niacin (358.40 mg/100 g, 0.09 mg/100 g and 0.08 mg/100 g), while Talinum triangulare had the highest level of riboflavin (0.18 mg/100 g). The steamed Amaranthus hybridus contained the highest levels of ascorbic acid, thiamin and niacin (322.56 mg/100 g, 0.08 mg/100 g and 0.73 mg/100 g), while steamed leaves of Talinum triangulare had the highest quantity of riboflavin (0.16 mg/100 g). The boiled Amaranthus hybridus had the highest amount of ascorbic acid, thiamin and niacin (197.12 mg/100 g, 0.05 mg/100 g and 0.41 mg/100 g), whereas the boiled Talinum triangulare had the highest level of riboflavin (0.09 mg/100 g). The percentage of vitamin loss in steamed leafy vegetables varies from 9 to 11 %, 9 to 17 %, 9 to 13 % and 9 to 12 % in ascorbic acid, thiamin, riboflavin and niacin respectively. Almost 50 % of the essential vitamins were lost in boiled leafy vegetables. "Fluorescent detection of vitamin B2 (VB2)" is a commonly used experiment in undergraduate students' instrumental analysis course; however, it provides limited experimental content and inadequate training for students. Spinach is a green vegetable with relatively high VB2 content, but the blanching treatment, which is used to remove oxalic acid, inevitably results in the partial loss of VB2. Herein, we demonstrate that fluorescent analysis can be used to determine the amount of VB2 released from spinach during blanching treatment. Based on this, we propose a teaching reform idea of adding measurement of the time-dependent VB2 loss rate during blanching to undergraduate experimental education. This would not only greatly enrich the original experimental content, but also make the experiment more interesting and relevant to human life, and improve students' ability to apply what they have learned to practical problems. Background: The quest for organic agriculture has necessitated the search for natural products which are abundant in phytochemicals and have significant therapeutic effects. This study was carried out to investigate the nutritional and ethno-medicinal potentials of egg-lime-molasses mixture (ELM). Methods: Fresh eggs were placed in a bowl after which 1 liter of lime juice and 500 g of molasses were added into the same bowl, then it was covered tightly and left for 10 days at temperature of 27°C and relative humidity of 61%. At the end of 10 days, the egg shells were dissolved into solution and then blended together. The mixture was then analyzed for proximate composition, phytochemicals, minerals and vitamins. The analyses were investigated in accordance with standard procedures. Result: Proximate analysis of ELM indicated the presence of moisture content (19.60%), crude protein (15.20%), lipids (5.50%), ash (14.60%), crude fibre (9.60%), carbohydrates (35.20%), fatty acid (4.40%) and energy (1060.30 Kcal/100 kg). Phytochemical screening revealed that ELM contained alkaloids, flavonoids, glycosides, saponin, steroids, phenols, terpenoides, tannin and antraquinones. Quantitative evaluation of the phytochemicals showed that ELM contained alkaloids (8.46 mg/100 g), flavonoids (2.30 mg/100 g), glycosides (0.08 mg/100 g), Saponin (5.25 mg/100 g), steroids (0.22 mg/l00 g), phenols (0.09 mg/100 g), terpenoides (0.56 mg/100 g), tannin (8.34 mg/100 g) and antraquinones (1.60 mg/100 g) and the vitamin constituents are Vitamin A (3.20 mg/100 g), Vitamin B1(280 mg/100 g), Vitamin B2 (880 mg/100 g), Vitamin B3 (340 mg/100 g), Vitamin C (15.40 mg/100 g) and Vitamin E (0.015 mg/100 g). Mineral analysis showed that it contained calcium (29.95%), magnesium (4.08%), potassium (23.20%), sodium (0.38%), phosphorus (6.90%), chlorine (0.30%), manganese (1.44 ppm), iron (3.60 ppm), aluminum (5.35%), titanium (2.10 ppm) and silicon (22.70 ppm). It was concluded that ELM is rich in various nutrients and phytochemicals conferring it the ability to perform multiple biological activities and as a natural alternative to antibiotics especially in monogastric animal production. THE local strain Lentinula edodes (OM432157), which was discovered for the first time in the Iraqi environment, was cultivated in the laboratory using nine treatments based on sawdust as the main substrate. A comparative study was done on the nutritional value of the local strain L. edodes (OM432157) in the wild and the cultivated one. It was observed that the highest growth rate of the fungus strain under study was in T8 (1.8 cm) and the lowest growth rate in T5 1.1 (cm). Fungal tissue growth in treatments (T4, and T5) were required (29 days), while in treatment T8 it was required (26 days), and in other remaining treatments it was accomplished in 25 days. The time required for pin heads to form in T5 was faster (17 days), followed by T8 (18 days) and T9 (19 days). The water-soluble vitamins (C, B1, B2) and the fat-soluble vitamins (E, K, D, A) were analyzed by HPLC. It was found that the cultivated mushrooms were a good source of vitamin B2 (6.9mg/100g dw) while wild mushrooms were a source of vitamin B1 (33.3mg/100g dw). The local strain of the cultivated and wild shiitake mushrooms also contained all fat-soluble vitamins, and the highest content of vitamin A was recorded in the cultivated type (19.9mg/100g dw) and the wild (103mg/100g.dw). These results are the first in Iraq, through which we found that the local strain L. edodes (OM432157), has high nutritional value and can be considered ideal supplements for many low-vitamin food items in our diets. Despite being present in minimal amounts, vitamin B2 (VB2), vitamin C (VC), and vitamin B6 (VB6) each play indispensable roles in human metabolisms. Given that VB2, VC, and VB6 cannot be synthesized by the human body, detections of these three vitamins both in fermentation liquid where vitamins are industrially manufactured and in human serum where vitamin concentrations could be clinically controlled are of significant importance. Here, a nanoporous gold (NPAu) modified screen-printed electrode (NPAu/SPE) was fabricated to detect VB2, VC, and VB6 based on NPAu's electro-oxidation towards vitamins. Owing to the wide separation of peak potentials among VB2, VC, and VB6, the simultaneous detection of these three vitamins was achieved by the NPAu/SPE within a potential range from −0.8 V to 0.8 V. The achieved limits of detection (LOD) for VB2, VC, and VB6 were 0.46, 6.44, and 1.92 μM, with sensitivities of 68.58, 4.77, and 15.94 μA/μM, respectively. Subsequent reliability experiments suggested that the NPAu/SPE exhibited solid anti-interference capability and repeatability. Additionally, the real-sample detection of the NPAu/SPE towards VB2, VC, and VB6 was achieved both in human serum and in fermentation liquid with comparable accuracy (the recovery rates were from 89.8% to 111.7%) as high-performance liquid chromatography (HPLC). Moreover, the portable NPAu/SPE showed comparable performance in terms of the LOD and linear dynamic range when compared to glassy carbon electrodes (GCE) limited to laboratory detection. The proposed NPAu/SPE possesses various advantageous properties including portability, easy fabrication, high sensitivity, and cost-efficiency, making it a potential candidate for clinical and industrial multi-vitamins analysis. Coenzymes are important for all classes of enzymatic reactions and essential for cellular metabolism. Most coenzymes are synthesized from dedicated precursors, also referred to as vitamins, which prototrophic bacteria can either produce themselves from simpler substrates or take up from the environment. The extent to which prototrophs use supplied vitamins and whether externally available vitamins affect the size of intracellular coenzyme pools and control endogenous vitamin synthesis is currently largely unknown. Here, we studied coenzyme pool sizes and vitamin incorporation into coenzymes during growth on different carbon sources and vitamin supplementation regimes using metabolomics approaches. We found that the model bacterium Escherichia coli incorporated pyridoxal, niacin, and pantothenate into pyridoxal 50-phosphate, NAD, and coenzyme A (CoA), respectively. In contrast, riboflavin was not taken up and was produced exclusively endogenously. Coenzyme pools were mostly homeostatic and not affected by externally supplied precursors. Remarkably, we found that pantothenate is not incorporated into CoA as such but is first degraded to pantoate and β- alanine and then rebuilt. This pattern was conserved in various bacterial isolates, suggesting a preference for β-alanine over pantothenate utilization in CoA synthesis. Finally, we found that the endogenous synthesis of coenzyme precursors remains active when vitamins are supplied, which is consistent with described expression data of genes for enzymes involved in coenzyme biosynthesis under these conditions. Continued production of endogenous coenzymes may ensure rapid synthesis of the mature coenzyme under changing environmental conditions, protect against coenzyme limitation, and explain vitamin availability in naturally oligotrophic environments. Polymorphisms in folate cycle genes (methylene tetrahydrofolate reductase – MTHFR, methionine synthase reductase – MTRR and methionine synthase – MTR) and associated hyperhomocysteinemia and folate deficiency play an important role among multiple genetic causes early pregnancy loss. The results of studies examining the influence of polymorphic variants of the folate cycle genes in women with pregnancy losses are quite contradictory. It has been proven that prescribing folic acid preparations or in combination with other vitamins and trace elements at the stage of preparation for pregnancy and during pregnancy significantly reduces the risk of miscarriage and pregnancy loss, is an important prevention of defects in the development of the nervous system of the fetus and disorders of the autistic spectrum in newborns and children. Adenomyosis remains a complex and urgent problem in obstetrics and gynecology, as today there is convincing evidence of the negative impact of adenomyosis both on fertility and on the course and outcomes of pregnancy. The objective: to study the prevalence of folate cycle gene polymorphisms and to assess the effectiveness of vitamin-mineral complex use in the pre-gravid preparation taking into account the genetic peculiarities of folate metabolism in women with early pregnancy loss (EPL) and adenomyosis. Materials and methods. Data from the examination at the stage of pre-gravid preparation of 40 women with early pregnancy loss – EPL (5–13 weeks of gestation) and adenomyosis, which were included in the main group, and 34 women with adenomyosis without a history of EPL (control group) were analyzed. In 73,5% of women of the control group, the C/C genotype of the MTHFR 677 C>T gene prevailed, in women with EPL and adenomyosis, a significant increase in the frequency (50%) of the heterozygous polymorphism for this C/T allele of the MTHFR 677 gene and a decrease in the frequency of detection of neutral A/A allele and increased frequency (80%) of MTHFR 1298 gene allele polymorphism (A/C and C/C). In women with EPL and adenomyosis, a significant increase in homocysteine (12,8±2,3мμmol/l) and a deficiency of folic acid (5,0±0,9 ng/ml) in blood serum compared to reference values were found. Conclusions. The use of vitamin-mineral complex, which includes betaine (200 mg), cystine (200 mg), zinc (10 mg), niacin (16 mg), folic acid (in the form of 5-MTHF-glucosamin – 400 mcg), vitamin B12 (in the form of methylcobalamin – 2.5 mcg), vitamin B6 (1.4 mg) and riboflavin (1.4 mg), 1 tablet 1-2 times a day for 3 months for the pre-gravid preparation leaded to significant decrease of homocysteine concentration, normalization of folic acid concentration. Purpose: This study's purpose was to identify the relationship between daily vitamin intake and blood glucose in cancer patients undergoing chemotherapy and identify factors affecting blood glucose. Methods: This descriptive study included 134 cancer patients undergoing chemotherapy at a university hospital. Data were analyzed with descriptive statistics, independent T-test, one-way analysis of variance, Pearson's correlation coefficients, and stepwise multiple regression analysis using SPSS/WIN 27.0 version. Results: The average blood glucose of the subjects was 128.13±37.34 mg/dL and 30.6% of the subjects had readings of over 140 mg/dL. Blood glucose varied significantly by sex, age, education level, exercise frequency, and comorbidity. Vitamins A, E, and C, thiamin, riboflavin, niacin, vitamin B6, folate, and biotin were negatively correlated with blood glucose. Factors affecting blood glucose were exercising 7 times a week (β-.61, p<.001), 4 to 6 times a week (β=-.41, p<.001), 1 to 3 times a week (β=-.38, p<.001), age (β=.18, p=.016), and vitamin A intake (β=-.16, p<.043), with a total explanatory power of approximately 31.5%. Conclusion: Nurses should provide patients undergoing chemotherapy with lifestyle interventions including exercise at least 3 days a week and appropriate vitamin A intake, especially for older patients, to prevent hyperglycemia. Purpose: This study aims to describe micronutrient intake according to food processing degree and to investigate the association between the dietary share of ultra-processed foods and micronutrient inadequacies in a representative sample of Portuguese adult and elderly individuals. Methods: Cross-sectional data from the National Food, Nutrition and Physical Activity Survey (2015/2016) were used. Food consumption data were collected through two 24-h food recalls, and food items were classified according to the NOVA system. Linear regression models were used to assess the association between the micronutrient density and the quintiles of ultra-processed food consumption—crude and adjusted. Negative Binomial regressions were performed to measure the prevalence ratio of micronutrient inadequacy according to ultra-processed food quintiles. Results: For adults, all evaluated vitamins had significantly lower content in the fraction of ultra-processed foods compared to unprocessed or minimally processed foods, except vitamin B2. For the elderly, out of ten evaluated vitamins, seven presented significantly less content in ultra-processed foods compared to non-processed ones. The higher energy contribution of ultra-processed foods in adults was associated with a lower density of vitamins and minerals. This association was not observed in the elderly. For adults, compared with the first quintile of ultra-processed food consumption, the fifth quintile was positively associated with inadequate intakes of vitamin B6 (PR 1.51), vitamin C (PR 1.32), folate (PR 1.14), magnesium (PR 1.21), zinc (PR 1.33), and potassium (PR 1.19). Nutritional supplements produced from soy and being rich in protein, fat and many other valuable substances have become a promising source of fortification of bakery products. This study aimed to develop a technology for producing wheat bread enriched with a protein-vitamin-mineral ingredient (PVMI) obtained from germinated soybeans. When kneading the dough, PVMI was added together with wheat flour and other dry components. Mathematical modeling of the wheat bread sensory evaluation showed the optimal content of PVMI in the recipe was 20% of the weight of wheat flour. As a result of the correlation-regression analysis, the most rational production characteristics for obtaining bread enriched with BVMI have been established. The amount of protein in the bread sample increased by 71.9%, dietary fiber - by 40.9%, vitamin E - by 34.7%, vitamin B1 - by 190%, vitamin B2 - by 80%, choline - by 186%. The fat quantity was 4.1 times more. At the same time, the total carbohydrate content decreased by 18.1% compared to the control sample. Effects of holding time before cooling, cooling method, and light or dark refrigerated storage on the stability of vitamin A, vitamin C, thiamine, riboflavin, and folate were investigated in fortified and unfortified soymilk. Vitamin C loss (6%) and mild vitamin A isomerization occurred when soymilk was held hot after fortification. Cooling bottled soymilk at ambient temperature or in an ice‐water bath did not affect any vitamins. Loss of riboflavin (18%) and vitamin A isomerization occurred during 12 days of light‐exposed refrigerated storage, in contrast to no vitamin degradation during dark refrigerated storage. A sensory panel of youth and children indicated no significant preferences between fortified and unfortified soymilk except for color, where the lighter‐colored unfortified soymilk was preferred. Acceptable vitamin stability and sensory characteristics can be achieved in fortified soymilk produced in small‐scale batch processes with appropriate management of production and storage conditions. Multivitamin tablet certified reference material (CRM, 108-10-019) was developed for the analysis of seven water-soluble vitamins, including thiamine, riboflavin, nicotinamide, pantothenic acid, pyridoxine, biotin, and folic acid. The CRM was prepared in powder form by grinding multivitamin tablets and then mixing, sieving, and bottling the powder. For the certification of each water-soluble vitamin, the isotope dilution mass spectrometry based on the liquid chromatography was applied. The methods for each analyte were validated by confirming the repeatability and reproducibility and by comparing with other CRMs. The property values and uncertainties for the vitamins were determined with 10 units from sample stored at − 20 °C. The homogeneity of each certified component was also examined in the range of 0.48–2.2%. All certified values for the seven water-soluble vitamins were stable for 3 or 6 years after the initial certification under storage conditions at − 20 °C. For fat-soluble vitamins, including retinol, α-tocopherol, cholecalciferol, and phylloquinone, two expert laboratories participated in analyses based on official methods, and the mean values of the reported results were assigned as reference values. The multivitamin tablet CRM (108-10-019) will be useful for validating analytical methods and for ensuring the quality of results for vitamin analysis in multivitamin tablets or similar products. Riboflavin deficiency produces severe peripheral neve demyelination in young, rapidly growing chickens. While this naturally-occurring vitamin B2 deficiency can cause a debilitating peripheral neuropathy, and mortality, in poultry flocks, it can also be a useful experimental animal model to study the pathogenesis of reliably reproducible peripheral nerve demyelination. Moreover, restitution of normal riboflavin levels in deficient birds results in brisk remyelination. It is the only acquired, primary, demyelinating tomaculous neuropathy described to date in animals. The only other substance that causes peripheral nerve demyelination similar to avian riboflavin deficiency is tellurium and the pathologic features of the peripheral neuropathy produced by this developmental neurotoxin in weanling rats are also described. Riboflavin (vitamin B2) is one of the essential vitamins that the human body needs to maintain normal metabolism. Its biosynthesis has become one of the successful models for gradual replacement of traditional chemical production routes. B. subtilis is characterized by its short fermentation time and high yield, which shows a huge competitive advantage in microbial fermentation for production of riboflavin. This review summarized the advancements of regulation on riboflavin production as well as the synthesis of two precursors of ribulose-5-phosphate riboflavin (Ru5P) and guanosine 5′-triphosphate (GTP) in B. subtilis. Plasma activated water (PAW) recently received much attention as an alternative food preservation method. However, its effects on food quality are still scarce. This study evaluates the effect of PAW processing time on bioactive compounds of rocket-salad leaves including: 18 phenolic compounds, vitamin C, riboflavin, nicotinic acid, and nicotinamide. Moreover, the impact of PAW on both antioxidant (DPPH) and peroxidase (POD) activities was also investigated. This was performed using HPLC-DAD, HPLC-MS/MS, and spectrophotometric analysis. All treatments induced non-significant increases in total phenolic contents. However, depending on processing time, significant increases or decreases of individual phenolic compounds were observed. PAW-10 and -20 increased the ascorbic acid content to 382.76 and 363.14 mg/100 g, respectively, compared to control (337.73 mg/100 g). Riboflavin and nicotinic acid contents were increased significantly in PAW-20 (0.53 and 1.26 mg/100), compared to control (0.32 and 0.61 mg/100 g, respectively). However, nicotinamide showed non-significant increase in all treatments. Antioxidant activity improved significantly only in PAW-20, while peroxidase activity was reduced up to 36% in the longest treatment. In conclusion, PAW treatment could be an effective technique for rocket decontamination since it positively influenced the quality of rocket, improving the retention of polyphenols and vitamins. Helichrysum odoratissimum (L.) Sweet (Asteraceae) is a perennial shrub used in South African folk medicine to combat human diseases such as diabetes, coughs, asthma, and diarrhoea, and for wound treatment. This study evaluated the leaf and stem of H. odoratissimum for their nutritive and anti-nutritive qualities using the standard methods of the Association of Official Analytical Chemists (AOAC), atomic absorption spectrometry (AAS) and colorimetry. Crude fat and protein were high in the leaf (7.61% and 7.82% DW), but low in the stem (2.25% and 3.4%), respectively. The neutral detergent fibre (NDF) content was significantly higher in the stem (68.5%) compared to the leaf (43.6%), while non-fibre carbohydrate (NFC) in the leaf (24.93%) was higher than in the stem (15.67%). Mineral analysis showed that both the leaf and stem of H. odoratissimum are good sources of potassium, calcium, sodium, magnesium, phosphorus and iron. Appreciable amounts of vitamins B2, C and E in the leaf and stem supports the folkloric utility of the plant in the traditional treatment of wounds, coughs and colds. A tolerable amount of phytate in both leaf (0.36%) and stem (1.06%) might be a factor for safer consumption of the plant as food and medicine. Keroplatus is a genus of fungus gnats family Keroplatidae (Diptera, Bibionomorpha). Larvae of some species emit a constant blue light from the body. The bioluminescence of Keroplatidae is one of the least studied of all terrestrial insects and very few facts are known to date of its biology and biochemistry. Here we report the high level of riboflavin in Keroplatus testaceus larvae, a fluorescent compound that might be relative to its bioluminescent system. Extracellular hydrolysis of flavin-adenine dinucleotide (FAD) and flavin mononucleotide (FMN) to riboflavin is thought to be important for cellular uptake of vitamin B2 because FAD and FMN are hydrophilic and do not pass the plasma membrane. However, it is not clear whether FAD and FMN are hydrolyzed by cell surface enzymes for vitamin B2 uptake. Here, we show that in human cells, FAD, a major form of vitamin B2 in plasma, is hydrolyzed by CD73 (also called ecto-5' nucleotidase) to FMN. Then, FMN is hydrolyzed by alkaline phosphatase to riboflavin, which is efficiently imported into cells. We determined that this two-step hydrolysis process is impaired on the surface of glycosylphosphatidylinositol (GPI)- deficient cells due to the lack of these GPI-anchored enzymes. During culture of GPI-deficient cells with FAD or FMN, we found that hydrolysis of these forms of vitamin B2 was impaired, and intracellular levels of vitamin B2 were significantly decreased compared with those in GPI-restored cells, leading to decreased formation of vitamin B2-dependent pyridoxal 50 -phosphate and mitochondrial dysfunction. Collectively, these results suggest that inefficient uptake of vitamin B2 might account for mitochondrial dysfunction seen in some cases of inherited GPI deficiency. [ABSTRACT FROM AUTHOR]

Food fortification is one strategy that has been used to overcome micronutrient deficiencies among vulnerable populations. Maize, a common staple food in Kenya, has been used as a suitable fortification vehicle. However, several factors, including storage conditions, impact micronutrient stability in fortified maize flour. This study aimed to assess the influence of storage condition on the retention of retinol and B-vitamins in selected commercial fortified maize flour. Fresh samples of fortified maize flours from two brands (codedXX1 and XY2) were sampled from the manufacturers at the point of production. The storage stability of retinol and B-vitamins in the two brands (XX1 and XY2) was monitored for 6 months at 25°C/75% relative humidity and 35°C/83% relative humidity. Retinol and thiamine were the least stable vitamins in both flour brands, while riboflavin and folate were relatively stable. Niacin was the most stable vitamin. Retinol was the least stable vitamin for brand XXI at both 25 °C/75% RH and 35 °C/83% RH, followed by thiamine, riboflavin, folate, and niacin. However, brand XY2 showed that under both storage conditions, thiamine was the least stable vitamin, followed by retinol, riboflavin, folate, and niacin. Vitamin retention was higher in samples stored at a lower temperature and relative humidity (25°C/75% RH) than in samples stored at higher temperature and relative humidity (35°C/83% RH) for both brands. In conclusion, thiamine and retinol were generally more susceptible to storage losses. Although the vitamin content in the flour samples decreased during storage, the changes in both storage conditions (except for riboflavin) and both brands were not significantly different. Background The generation of the active form of vitamin B-6, pyridoxal 5′-phosphate (PLP), in tissues is dependent upon riboflavin as flavin mononucleotide, but whether this interaction is important for maintaining vitamin B-6 status is unclear. Objective To investigate vitamin B-6 and riboflavin status, their metabolic interaction, and relationship with methylenetetrahydrofolate reductase (MTHFR) genotype in adulthood. Methods Data from 5612 adults aged 18–102 y were drawn from the Irish National Adult Nutrition Survey (NANS; population-based sample) and the Trinity-Ulster Department of Agriculture (TUDA) and Genovit cohorts (volunteer samples). Plasma PLP and erythrocyte glutathione reductase activation coefficient (EGRac), as a functional indicator of riboflavin, were determined. Results Older (≥65 y) compared with younger (<65 y) adults had significantly lower PLP concentrations (P  < 0.001). A stepwise decrease in plasma PLP was observed across riboflavin categories, from optimal (EGRac ≤1.26), to suboptimal (EGRac: 1.27–1.39), to deficient (EGRac ≥1.40) status, an effect most pronounced in older adults (mean ± SEM: 76.4 ± 0.9 vs 65.0 ± 1.1 vs 55.4 ± 1.2 nmol/L; P  < 0.001). In individuals with the variant MTHFR 677TT genotype combined with riboflavin deficiency, compared with non-TT (CC/CT) genotype participants with sufficient riboflavin, we observed PLP concentrations of 52.1 ± 2.9 compared with 76.8 ±0.7 nmol/L (P  < 0.001). In participants with available dietary data (i.e. NANS cohort, n = 936), PLP was associated with vitamin B-6 intake (nonstandardized regression coefficient β: 2.49; 95% CI 1.75, 3.24; P  < 0.001), supplement use (β: 81.72; 95% CI: 66.01, 97.43; P  < 0.001), fortified food (β: 12.49; 95% CI: 2.08, 22.91; P  = 0.019), and EGRac (β: –65.81; 95% CI: –99.08, –32.54; P  < 0.001), along with BMI (β: –1.81; 95% CI: –3.31, –0.30; P  = 0.019). Conclusions These results are consistent with the known metabolic dependency of PLP on flavin mononucleotide (FMN) and suggest that riboflavin may be the limiting nutrient for maintaining vitamin B-6 status, particularly in individuals with the MTHFR 677TT genotype. Randomized trials are necessary to investigate the PLP response to riboflavin intervention within the dietary range.
