Helicobacter pylori is a microorganism that infects 60% of the population and is considered the main cause of atrophic gastritis, gastric and duodenal ulcers, and gastric cancer. Different emerging pathogens have been found in drinking water and their presence is considered to be an important public health problem. For this reason, it is necessary to carry out the validation of reliable technologies for this type of pathogens and evaluate their performance. This paper reports, for the first time, H. pylori reduction in a drinking water pilot plant of two slow sand filters (SSF). Inlet water was taken from a gravel filtration system of a rural water supply in Colombia and then inoculated with viable cells of H. pylori. By determining the Genomic Units (GU) through quantitative Polymerase Chain Reaction (qPCR), the concentration of GU/sample was measured. In the inlet water amplification for SSF1 and SSF2 were 5.13 × 10 2  ± 4.48 × 10 2 and 6.59 × 10 2  ± 7.32 × 10 2 , respectively, while for the treated water they were 7.0 ± 5.6 and 2.05 × 10 1  ± 2.9 × 10 1 GU/sample for SSF1 and SSF2, respectively. The SSF pilot plant reached up to 3 log reduction units of H. pylori; therefore, since there is not an H. pylori contamination indicator and its periodic monitoring is financially complicated, the SSF could guarantee the drinking water quality necessity that exists in rural areas and small municipalities in developing countries, where infection rates and prevalence of this pathogen are high., (© 2024. Tap water contamination is a growing concern in healthcare facilities, and despite chlorination, tap water in these facilities contains several pathogenic microorganisms causing healthcare-associated waterborne infections or nosocomial outbreaks. Shower units are particularly prone to contamination as they are conducive for bacterial growth and can even produce bioaerosols containing pathogenic bacteria. Shower units coupled with point-of-use (POU) water filters are a simple and safe option; however, their efficacy has been under-reported. Therefore, we determined the efficacy of showerheads attached with a POU filter capsule in preventing infections in our hospital. We investigated the presence of pathogenic bacteria in water sampled from three shower units. After replacing the original shower units with new ones incorporated with a sterile-grade water filter capsule (0.2 µm; QPoint™), the water samples were analyzed for up to 2 months. The POU filters removed several pathogenic bacteria (Mycobacterium, Pseudomonas, Stenotrophomonas, Aeromonas, and Klebsiella spp.). Filter effectiveness depends on regional water quality and we believe that effective tap water treatment combined with the use of POU filters (introduced at a reasonable cost in healthcare facilities) can considerably minimize waterborne diseases in hospitals and improve patient care.

The use of surface and recycled water sources for irrigation can reduce demand on critical groundwater resources. Treatment or mitigation may be necessary for the use of these alternative water sources in order to reduce risk associated with microbial pathogens present in the water. In this study, the efficacy of a zero-valent iron (ZVI) sand filter was assessed for the reduction of Listeria monocytogenes and Escherichia coli in surface water. Water recovered from an agricultural pond was inoculated with E. coli TVS353 and an environmental L. monocytogenes isolate at 7 Log 10 CFU/mL and horizontally filtered over a six-month period through a PVC pipe filter, filled with 35%:65% (volume:volume) ZVI:sand or sand alone. Filtered water was used to irrigate lettuce and bacterial persistence on lettuce leaves was determined for 7 days post-irrigation. Both ZVI:sand-filtered water and sand-filtered water contained significantly (p < 0.005) lower levels of E. coli and L. monocytogenes compared to initial unfiltered inoculated water. Population reductions of E. coli and L. monocytogenes were comparable after sand filtration. However, ZVI:sand filtration resulted in significantly greater population reductions of L. monocytogenes (P < 0.05) compared to E. coli. Populations of E. coli on leaves of lettuce plants irrigated with ZVI:sand-filtered water were not significantly lower than populations on plants irrigated with sand-filtered irrigation water over the 7-day period. However, populations of L. monocytogenes on lettuce leaves irrigated with ZVI-treated water were significantly lower than counts on plants irrigated with sand-filtered irrigation water on days 3 and 4 post irrigation (p = 0.052 and p = 0.042 for days 3 and 4, respectively. The differences observed in reductions of L. monocytogenes and E. coli by ZVI filtration is due to the differing effect that ZVI disruption has on Gram-positive and Gram-negative cell walls and membranes. In biologically active filters (BAFs), microorganisms acclimated on the media surface are the key players responsible for removing organic water contaminants. In this study, next generation sequencing by Illumina MiSeq was used to characterize the microbial community structures in the influent, effluent, and media of a set of bench-scale BAFs that have been demonstrated with high removal efficiency (>75%) of 16 contaminants of emerging concern (CECs), which include a variety of pharmaceuticals (e.g., sulfamethoxazole and ibuprofen), X-ray contrast agent (i.e., iopromide), and pesticides (e.g., atrazine) that are prevalently found in municipal waste streams. Proteobacteria and Planctomycetes were the most abundant phyla in filter media, while the influent and effluent samples were dominated by Proteobacteria, Actinobacteria, and Chlamydiae. Factorial and principal component analysis revealed microbial structures in the media were significantly affected by the operation conditions, including media type (GAC versus dual media anthracite sand), EBCT (10 versus 18 min), and pre-ozonation. Detrended correspondence analysis demonstrated media materials predominantly governed the structures of the acclimated biofilm in BAFs as they provide direct attachment surface. This is in line with the higher microbial activity and better treatment performance exhibited by GAC BAFs compared to the dual media BAFs, corroborating the importance of filter media selection to promote the acclimation of active and robust biofilm for efficient CEC removal. Principal component analysis revealed the significant influence from ozonation, which does not only break down CECs, but also stimulates microbes that grow on the ozonation products. Partial canonical correlation analysis further proved the shaping of biofilm communities on the BAF media is more associated with media type and ozonation compared to EBCT. Putative CEC degraders are predicted based on their dominance in the media and degradation capabilities reported in previous literature. Waterborne pathogens are a global concern for public health worldwide. Despite continuing efforts to maintain water safety, water quality is still affected by deterioration and pollution. Legionella pneumophila colonizes man-made water systems and can infect humans causing Legionnaire's disease (LD), pneumonia. The prevention of LD is a public health issue and requires specific systems to control and detect these microorganisms. Culture plate is the only technique currently approved, but requires more than 10 days to obtain results. A rapid test that inform in hours about the presence of Legionella pneumophila in water samples will improve the control of this pathogen colonization. In order to control colonization by L. pneumophila we developed a membrane filter method to capture and immunodetect this microorganism in water samples. This membrane filter is used to retain the bacteria using a nitrocellulose disc inside a home-made cartridge. Subsequently we perform the immunodetection of the bacteria retained in the nitrocellulose (blocking, antibody incubation, washings and developing). On comparing our test with the gold-standard, the most important finding is the considerably reduction in time maintaining the same detection limit. This rapid test is easily automated for L. pneumophila detection allowing a comprehensive surveillance of L. pneumophila in water facilities and reducing the variability in the analyses due to the low need for manipulation. Moreover, corrective measures may be applied the same day of the analysis. This paper presents a new model to simulate long-term microbial removal in stormwater biofilters. The water flow module uses a 'three-bucket' approach to describe the flow processes in biofilters, while the microbial quality module employs the one-dimensional advection-dispersion equation to represent microbial transport and fate under different design and operational conditions. Three governing processes for microbial removal, adsorption, desorption and die-off, are included; temperature is also incorporated as a key factor for die-off. The model was tested using long term monitoring data collected from laboratory columns in which five different biofilter configurations were studied over a period of 44weeks. A multi-objective calibration with the balance of instantaneous ponding levels and event outflow volumes was implemented on the water flow module, and the Nash-Sutcliffe Efficiency (E) values ranged from 0.82 to 0.95. The microbial quality module was tested using the effluent Escherichia coli concentration data, and the E values obtained for different configurations were between 0.46 and 0.68. The optimized parameter values agreed with those presented in literature. However, sensitivity analyses suggested that the model's prediction was not sensitive to all parameters, the explanation for which was hypothesized to be data paucity rather than model structural uncertainties. Model validation was also conducted by splitting the data into calibration and validation datasets. The application of ozone-biological activated carbon (O 3 -BAC) as an advanced treatment method in drinking water treatment plants (DWTPs) can help to remove organic micropollutants and further decrease the dissolved organic carbon (DOC) level in finished water. With the increase attention to microbial safety of drinking water, a pre-positioned O 3 -BAC followed by a sand filter has been implanted into DWTP located in Shanghai, China to increase the biostability of effluents. The results showed that BAC had high removal efficiencies of UV 254 , DOC and disinfection by-product formation potential (DBPFP). The removal efficiencies between pre- and post-positioned BAC filtrations were similar. Based on the analyses of fluorescence excitation-emission matrix spectrophotometry (FEEM), the generation and leakage of soluble microbial products (SMPs) were found in both two BAC filtrations on account of the increased fluorescence intensities and fluorescence regional integration (FRI) distribution of protein-like organics, as well as the enhanced biological index (BIX). The leakage of SMPs produced by metabolism of microbes during BAC process resulted in increased DBPFP yield and carcinogenic factor per unit of DOC (CF/DOC). Although BAC filtration reduced the DBPFP and CF, there still was high health risk of effluents for the production of SMPs. Therefore, the health risks for SMPs generated by BAC filtration in drinking water advanced treatment process should be addressed, especially with that at high temperature., (Copyright © 2018 Elsevier Ltd. Both river bank filtration and dead-end sand filtration are becoming increasingly applied in rural areas to improve the quality of fecally contaminated water. To evaluate the capacity of both treatments to remove E. coli, fecal streptococci, and somatic and K13-phages, this study investigates their concentrations in diluted wastewater after short-distance tangential sand filtration and dead-end sand filtration. Bacteria were almost undetectable in both systems after 60 cm depth, and at a pore-water velocity of 1 m/d. Both phages underwent removal of 2.5 logs by tangential filtration, whereas dead-end filtration removed 5.1 logs and 3.9 logs of K13-phages and somatic phages, respectively. After discounting removal by the schmutzdecke, observed only in the dead-end filtration, both systems removed phages similarly. It is concluded that short-distance river bank filtration, alone, does not meet WHO requirements for drinking water. However, the concomitant reduction of suspended solids renders the filtered water amenable to further treatment steps.

The recovery of Mycobacterium avium subspecies paratuberculosis (Map) from the environment can be a laborious process - owing to Map being fastidious, its low number, and also high numbers of other microbial populations in such settings. Protocols i.e. filtration, decontamination and modified elution were devised to recover Map from spiked water sediments. Three culture media: Herrold's Egg Yolk Media (HEYM), Middlebrook 7H10 (M-7H10) and Bactec 12B were then employed to grow the organism following its elution. In the sterile sediment samples the recovery of Map was significant between the time of exposure for each of HEYM and M-7H10, and insignificant between both media (P < 0.05). However, in the non-sterile sediment samples, the HEYM grew other background microflora including moulds at all the times of exposure whilst 4 h followed by M-7H10 culture yielded Map colonies without any background microflora. Using sterile samples only for the Bactec 12B, the recovery of Map decreased as time of exposure increased. Based on these findings, M-7H10 should be considered for the recovery of Map from the natural environment including water sediments where the recovery of diverse microbial species remains a challenge., Significance of the Study: Map is a robust pathogen that abides in the environment. In water treatment operations, Map associates with floccules and other particulate matter including sediments. It is also a fastidious organism, and its detection and recovery from the water environment is a laborious process and can be misleading within the abundance of other mycobacterial species owing to their close resemblance in phylogenetic traits. In the absence of a reliable recovery method, Map continues to pose public health risks through biofilm in household water tanks, hence the need for the development of a reliable recovery protocol to monitor the presence of Map in water systems in order to curtail its public health risks., (Copyright © 2018 Elsevier B.V. This study examined the efficacy of reovirus concentration from large volumes of water using two positively charged filters: Zeta Plus 1MDS and NanoCeram. The results indicated that an average of 61 and 81% of input reoviruses were effectively recovered, respectively, from recycled water and tap water using NanoCeram filtration.

The existence of pathogenic bacteria in drinking water has been a threat to the safety of human well-being. Traditional methods to detect bacteria are standard plate counts or rapid methods such as ELISA and PCR. However, those methods can be time-consuming or require pre-enrichment when detecting low concentrations of bacteria. Filter membrane has been used for bacteria collection and culturing when monitoring environmental water samples. In this study, we applied surface enhanced Raman spectroscopy (SERS) to rapidly screen bacteria cells on a filter membrane based on the unique signal from an indicator molecule, 4-mercaptophenylboronic acid (4-mpba), that can specifically bind to the surface of bacteria through diol group in its structure and give off consistent SERS signal. With a pore size of 0.22 μm, the filter membrane functions as both a concentrating device and a separation mechanism that eliminates molecules smaller than the pore size. With SERS mapping, 4-mpba gives characteristic signal, indicating the presence of bacteria, whereas no 4-mpba signal was observed in the absence of bacteria. The developed method can detect Escherichia coli, Salmonella enterica, and Listeria monocytogenes on a filter membrane non-selectively in 80 min. Application of the method in pond water was demonstrated. Using in-field bioreactors, we investigated the influence of exogenous microorganisms in groundwater planktonic and biofilm microbial communities as part of the Integrated Field Research Challenge (IFRC). After an acclimation period with source groundwater, bioreactors received either filtered (0.22 μM filter) or unfiltered well groundwater in triplicate and communities were tracked routinely for 23 days after filtration was initiated. To address geochemical influences, the planktonic phase was assayed periodically for protein, organic acids, physico-/geochemical measurements and bacterial community (via 16S rRNA gene sequencing), while biofilms (i.e. microbial growth on sediment coupons) were targeted for bacterial community composition at the completion of the experiment (23 d). Based on Bray-Curtis distance, planktonic bacterial community composition varied temporally and between treatments (filtered, unfiltered bioreactors). Notably, filtration led to an increase in the dominant genus, Zoogloea relative abundance over time within the planktonic community, while remaining relatively constant when unfiltered. At day 23, biofilm communities were more taxonomically and phylogenetically diverse and substantially different from planktonic bacterial communities; however, the biofilm bacterial communities were similar regardless of filtration. These results suggest that although planktonic communities were sensitive to groundwater filtration, bacterial biofilm communities were stable and resistant to filtration. Bioreactors are useful tools in addressing questions pertaining to microbial community assembly and succession. These data provide a first step in understanding how an extrinsic factor, such as a groundwater inoculation and flux of microbial colonizers, impact how microbial communities assemble in environmental systems.

Little is known about the forces that determine the assembly of diverse bacterial communities inhabiting drinking water treatment filters and how this affects drinking water quality. Two contrasting ecological theories can help to understand how natural microbial communities assemble; niche theory and neutral theory, where environmental deterministic factors or stochastic factors predominate respectively. This study investigates the development of the microbial community on two common contrasting filter materials (quartz sand and granular activated carbon-GAC), to elucidate the main factors governing their assembly, through the evaluation of environmental (i.e. filter medium type) and stochastic forces (random deaths, births and immigration). Laboratory-scale filter columns were used to mimic a rapid gravity filter; the microbiome of the filter materials, and of the filter influent and effluent, was characterised using next generation 16S rRNA gene amplicon sequencing and flow-cytometry. Chemical parameters (i.e. dissolved organic carbon, trihalomethanes formation) were also monitored to assess the final effluent quality. The filter communities seemed to be strongly assembled by selection rather than neutral processes, with only 28% of those OTUs shared with the source water detected on the filter medium following predictions using a neutral community model. GAC hosted a phylogenetically more diverse community than sand. The two filter media communities seeded the effluent water, triggering differences in both water quality and community composition of the effluents. Overall, GAC proved to be better than sand in controlling microbial growth, by promoting higher bacterial decay rates and hosting less bacterial cells, and showed better performance for putative pathogen control by leaking less Legionella cells into the effluent water., (Copyright © 2017 The Authors. Environmental surveillance of waterborne pathogens is vital for monitoring the spread of diseases, and electropositive filters are frequently used for sampling wastewater and wastewater-impacted surface water. Viruses adsorbed to electropositive filters require elution prior to detection or quantification. Elution is typically facilitated by a peristaltic pump, although this requires a significant startup cost and does not include biosafety or cross-contamination considerations. These factors may pose a barrier for low-resource laboratories that aim to conduct environmental surveillance of viruses. The objective of this study was to develop a biologically enclosed, manually powered, low-cost device for effectively eluting from electropositive ViroCap™ virus filters. The elution device described here utilizes a non-electric bilge pump, instead of an electric peristaltic pump or a positive pressure vessel. The elution device also fully encloses liquids and aerosols that could contain biological organisms, thereby increasing biosafety. Moreover, all elution device components that are used in the biosafety cabinet are autoclavable, reducing cross-contamination potential. This device reduces costs of materials while maintaining convenience in terms of size and weight. With this new device, there is little sample volume loss due to device inefficiency, similar virus yields were demonstrated during seeded studies with poliovirus type 1, and the time to elute filters is similar to that required with the peristaltic pump. The efforts described here resulted in a novel, low-cost, manually powered elution device that can facilitate environmental surveillance of pathogens through effective virus recovery from ViroCap filters while maintaining the potential for adaptability to other cartridge filters.

Bacteriophages are widely used to the treatment of drug-resistant bacteria and the improvement of food safety through bacterial lysis. However, the limited investigations on bacteriophage restrict their further application. In this study, a novel and highly efficient method was developed for isolating bacteriophage from water based on the electropositive silica gel particles (ESPs) method. To optimize the ESPs method, we evaluated the eluent type, flow rate, pH, temperature, and inoculation concentration of bacteriophage using bacteriophage f2. The quantitative detection reported that the recovery of the ESPs method reached over 90%. The qualitative detection demonstrated that the ESPs method effectively isolated 70% of extremely low-concentration bacteriophage (10 0 PFU/100L). Based on the host bacteria composed of 33 standard strains and 10 isolated strains, the bacteriophages in 18 water samples collected from the three sites in the Tianjin Haihe River Basin were isolated by the ESPs and traditional methods. Results showed that the ESPs method was significantly superior to the traditional method. The ESPs method isolated 32 strains of bacteriophage, whereas the traditional method isolated 15 strains. The sample isolation efficiency and bacteriophage isolation efficiency of the ESPs method were 3.28 and 2.13 times higher than those of the traditional method. Legionella pneumophila is a ubiquitous microorganism widely distributed in aquatic environments and can cause Legionellosis in humans. A promising approach to detect viable cells in water samples involves the use of quantitative polymerase chain reaction (qPCR) in combination with photoactivatable DNA intercalator propidium monoazide (PMA). However, the PMA efficiency could be different depending on the experimental conditions used. The aim of this study was to compare two PMA exposure protocols: (A) directly on the membrane filter or (B) in liquid after filter washing. The overall PMA-induced qPCR means reductions in heat-killed L. pneumophila cells were 2.42 and 1.91 log units for exposure protocols A and B, respectively. A comparison between the results obtained reveals that filter exposure allows a higher PMA-qPCR signal reduction to be reached, mainly at low concentrations ( p < 0.05). This confirms the potential use of this method to quantify L. pneumophila in water with low contamination.

Scientists recently reported the unexpected detection of unknown or poorly studied bacterial diversity in groundwater. The ability to uncover this neglected biodiversity mainly derives from technical improvements, and the term "microbial dark matter" was used to group taxa poorly investigated and not necessarily monophyletic. We focused on such under-investigated microbial dark matter of drinking water treatment plant from groundwater, across carbon filters, to post-chlorination. We tackled this topic using an integrated approach where the efficacy of stringent water filtration (10000 MWCO) in recovering even the smallest environmental microorganisms was coupled with high-throughput DNA sequencing to depict an informative spectrum of the neglected microbial diversity. Our results revealed that the composition of bacterial communities varies across the plant system: Parcubacteria (OD1) superphylum is found mainly in treated water, while groundwater has the highest heterogeneity, encompassing non-OD1 candidate phyla (Microgenomates, Saccharibacteria, Dependentiae, OP3, OP1, BRC1, WS3). Carbon filters probably act as substrate for microorganism growth and contribute to seeding water downstream, since chlorination does not modify the incoming bacterial community. New questions arise about the role of microbial dark matter in drinking water. Indeed, our results suggest that these bacteria might play a central role in the microbial dynamics of drinking water.

The recovery of Campylobacter species from food and environmental sources is challenging due to the slow growth of these bacteria and the need to suppress competing organisms during the isolation procedures. The addition of multiple selective antimicrobials to growth media can negatively impact recovery of some Campylobacter spp. Here, we describe our current method for the isolation of thermotolerant Campylobacter species, mainly C. jejuni and C. coli, from food and environmental samples. We emphasize the use of membrane filtration during plating for the specific isolation of Campylobacter spp. and a reduced use of antimicrobial supplements throughout the whole isolation process.

A consecutive centrifugal method was developed to concentrate enteric viruses from water. Using five selected human enteric viruses, the recovery rates were evaluated and compared with results from NanoCeram filtration methods. The highest recovery rate was achieved for coxsackievirus (78.11 %, 42.97-101.39), and the lowest rate was for adenovirus (32.84 %, 19.68-66.20). In comparison with NanoCeram disc filtration with beef-extract flocculation (BE), the recovery rate was increased for all viruses. The rate of increase varied from a low of 8.24 % (astroviruses) to a high of 24.22 % (noroviruses). The NanoCeram filtration with BE was further modified by NaPP buffer plus one-step centrifugation using a Centricon plus-70 device. The modified method further increased the concentrations of viruses in the sample. Virus concentration was increased to 19.45 ± 7.19-fold for rotavirus, 19.40 ± 6.54-fold for norovirus, 16.10 ± 7.61-fold for coxsackievirus, 12.80 ± 3.00-fold for astrovirus and 11.97 ± 6.94-fold for adenovirus compared to the NanoCeram filtration with BE. Subsequent cell culture showed that the infectivity of the viruses was not altered by any of the three methods. Three methods will provide testing labs with choices for cost-effective approaches to concentrate viruses from water.

Verocytotoxigenic Escherichia coli (VTEC) is associated with a wide spectrum of disease from mild self-limiting diarrhoea to haemolytic uremic syndrome. Contaminated drinking water is accepted as an important route of transmission in Ireland as elsewhere however established methods for detection of VTEC in drinking water have limitations. We describe a sensitive and rapid method for detection of VTEC from large volumes (20 to 30L) of drinking water based on filtration, enrichment culture of filters and real-time PCR detection of VTEC virulence and O antigen determinants from enrichments. The use of porous ceramic filters is promoted globally for household water treatment, but these filters are ineffective in removing viruses from water. In order to increase virus removal, we combine a promising natural coagulant, chitosan, as a pretreatment for ceramic water filters (CWFs) and evaluate the performance of this dual barrier water treatment system. Chitosan is a non-toxic and biodegradable organic polymer derived by simple chemical treatments from chitin, a major source of which is the leftover shells of crustacean seafoods, such as shrimp, prawns, crabs, and lobsters. To determine the effectiveness of chitosan, model test water was contaminated with Escherichia coli K011 and coliphage MS2 as a model enteric bacterium and virus, respectively. Kaolinite clay was used to model turbidity. Coagulation effectiveness of three types of modified chitosans was determine at various doses ranging from 5 to 30 mg/L, followed by flocculation and sedimentation. The pre-treated supernatant water was then decanted into the CWF for further treatment by filtration. There were appreciable microbial removals by chitosan HCl, acetate, and lactate pretreatment followed by CWF treatment, with mean reductions (95% CI) between 4.7 (± 1.56) and 7.5 (± 0.02) log10 for Escherichia coli, and between 2.8 (± 0.10) and 4.5 (± 1.04) log10 for MS2. Turbidity reduction with chitosan treatment and filtration consistently resulted in turbidities < 1 NTU, which meet turbidity standards of the US EPA and guidance by the World Health Organization (WHO). According to WHO health-based microbial removal targets for household water treatment technology, chitosan coagulation achieved health protective targets for both viruses and bacteria. Therefore, the results of this study support the use of chitosan to improve household drinking water filtration processes by increasing virus and bacteria reductions.

Water monitoring is essential to ensure safe drinking water for consumers. However existing methods have several drawbacks, particularly with regard to the poor recovery of Cryptosporidium due to the inability to efficiently elute Cryptosporidium oocysts during the established detection process used by water utilities. Thus the development of new inexpensive materials that could be incorporated into the concentration and release stage that would control Cryptosporidium oocysts adhesion would be beneficial. Here we describe improved filter performance following dip-coating of the filters with a "bioactive" polyacrylate. Slow sand filtration (SSF) is an effective low-tech water treatment method for pathogen and particle removal. Yet despite its application for centuries, it has been uncertain to which extent pathogenic microbes are removed by mechanical filtration or due to ecological interactions such as grazing and competition for nutrients. In this study, we quantified the removal of bacterial faecal indicators, Escherichia coli and Enterococcus faecalis, from secondary effluent of a wastewater treatment plant and analysed the microbial community composition in compartments of laboratory model SSF columns. The columns were packed with different sand grain sizes and eliminated 1.6-2.3 log units of faecal indicators, which translated into effluents of bathing water quality according to the EU directive (<500 colony forming units of E. coli per 100 ml) for columns with small grain size. Most of that removal occurred in the upper filter area, the Schmutzdecke. Within that same zone, total bacterial numbers increased however, thus suggesting a specific elimination of the faecal indicators. The analysis of the microbial communities also revealed that some taxa were removed more from the wastewater than others. These results accentuate the contribution of biological mechanisms to water purification in SSF.

The production of a drinking water that meets current aesthetic, microbiological and chemical standards, generally requires a combination of mechanical purification and disinfection in a multi-component treatment chain. Treatment choices and optimisation of water processing is best informed by using markers (including microbiological parameters) which indicate how each stage contributes to the production of the potable water. The present study combines culture-based and a number of culture-independent analyses to indicate what is happening at each stage of a state-of-the-art water treatment chain at Vansjø near the city of Moss in Norway. We show that particularly clarification with flotation and post-chlorination have profound and positive effects on water quality with respect to the removal and inactivation of microbes. Post-chlorination achieved better disinfection of the water than UV-treatment and was of paramount importance, as the penultimate step filtration through granular activated shed microbes to the water. Cloning and sequencing showed that some clones present in the raw water were detected at all stages in the treatment process, perhaps providing examples of microbes breaching physically all barriers in the treatment process. The biosand filter (BSF) is an intermittently operated, household-scale slow sand filter for which little data are available on the effect of sand composition on treatment performance. Therefore, bench-scale columns were prepared according to the then-current (2006-2007) guidance on BSF design and run in parallel to conduct two microbial challenge experiments of eight-week duration. Triplicate columns were loaded with Accusand silica or crushed granite to compare virus and E. coli reduction performance. Bench-scale experiments provided confirmation that increased schmutzdecke growth, as indicated by decline in filtration rate, is the primary factor causing increased E. coli reductions of up to 5-log10. However, reductions of challenge viruses improved only modestly with increased schmutzdecke growth. Filter media type (Accusand silica vs. crushed granite) did not influence reduction of E. coli bacteria. The granite media without backwashing yielded superior virus reductions when compared to Accusand. However, for columns in which the granite media was first backwashed (to yield a more consistent distribution of grains and remove the finest size fraction), virus reductions were not significantly greater than in columns with Accusand media. It was postulated that a decline in surface area with backwashing decreased the sites and surface area available for virus sorption and/or biofilm growth and thus decreased the extent of virus reduction. Additionally, backwashing caused preferential flow paths and deviation from plug flow; backwashing is not part of standard BSF field preparation and is not recommended for BSF column studies. Overall, virus reductions were modest and did not meet the 5- or 3-log10 World Health Organization performance targets.

Biosand filters in the Artibonite Valley of Haiti, the epicenter of the cholera epidemic that began in October 2010, were tested for total coliform and Vibrio cholerae removal efficiencies. While coliform are often used as an indicator organism for pathogenic bacteria, a correlation has never been established linking the concentration of coliform and V. cholerae, the causative agent for cholera. Hence, a method for field enumeration of V. cholerae was developed and tested. To this end, a plate count test utilizing membrane filtration technique was developed to measure viable V. cholerae cell concentration in the field. Method accuracy was confirmed by comparing plate count concentrations to microscopic counts. Additionally, biosand filters were sampled and removal efficiencies of V. cholerae and coliform bacteria compared. The correlation between removal efficiency and time in operation, biofilm ("schmutzdecke") composition, and idle time was also investigated. The plate count method for V. cholerae was found to accurately reflect microscope counts and was shown to be effective in the field. Overall, coliform concentration was not an appropriate indicator of V. cholerae concentration. In 90% of the influent samples from the study, coliform underestimated V. cholerae concentration (n = 26). Furthermore, coliform removal efficiency was higher than for V. cholerae hence providing a conservative measurement. Finally, time in operation and idle time were found to be important parameters controlling performance. Overall, this method shows promise for field applications and should be expanded to additional studies to confirm its efficacy to test for V. cholerae in various source waters.

Two biofilters were constructed using biological activated carbon (BAC) and nitrosamine-containing water from two drinking water treatment plants. The microbiome of each biofilter was characterized by 454 high-throughput pyrosequencing, and one nitrosamine-reducing bacterium was isolated. The results showed that nitrosamines changed the relative abundance at both the phylum and class levels, and the new genera were observed in the microbial communities of the two BAC filters after cultivation. As such, the genus Rhodococcus, which includes many nitrosamine-reducing strains reported in previous studies, was only detected in the BAC2 filter after cultivation. These findings indicate that nitrosamines can significantly affect the genus level in the microbial communities. Furthermore, the isolated bacterial culture Rhodococcus cercidiphylli A41 AS-1 exhibited the ability to reduce five nitrosamines (N-nitrosodimethylamine, N-nitrosodiethylamine, N-nitrosodi-n-propylamine, N-nitrosopyrrolidine, and N-nitrosodi-n-butylamine) with removal ratios that ranged from 38.1% to 85.4%. The isolate exhibited a better biodegradation ability with nitrosamine as the carbon source when compared with nitrosamine as the nitrogen source. Large sample volumes are traditionally required for the analysis of waterborne pathogens. The need for large volumes greatly limits the number of samples that can be processed. The aims of this study were to compare extraction and detection procedures for quantifying protozoan parasites and viruses from small volumes of marine water. The intent was to evaluate a logistically simpler method of sample collection and processing that would facilitate direct pathogen measures as part of routine monitoring programs. Samples were collected simultaneously using a bilayer device with protozoa capture by size (top filter) and viruses capture by charge (bottom filter). Protozoan detection technologies utilized for recovery of Cryptosporidium spp. and Giardia spp. were qPCR and the more traditional immunomagnetic separation-IFA-microscopy, while virus (poliovirus) detection was based upon qPCR versus plaque assay. Filters were eluted using reagents consistent with the downstream detection technologies. Results showed higher mean recoveries using traditional detection methods over qPCR for Cryptosporidium (91% vs. 45%) and poliovirus (67% vs. 55%) whereas for Giardia the qPCR-based methods were characterized by higher mean recoveries (41% vs. 28%). Overall mean recoveries are considered high for all detection technologies. Results suggest that simultaneous filtration may be suitable for isolating different classes of pathogens from small marine water volumes. More research is needed to evaluate the suitability of this method for detecting pathogens at low ambient concentration levels.

In an attempt to obtain a conservative estimate of virus removal during slow sand and river bank filtration, a somatic phage was isolated with slow decay and poor adsorption to coarse sand. We continuously fed a phage suspension to a 7-m infiltration path and measured the phage removal. In a second set of experiments, we fed the phage suspension to 1-m long columns run at different pore water velocities. Using the data obtained, a mathematical model was constructed describing removal vs. pore water velocity (PWV), assuming different statistical distributions of the adsorption coefficient λ. The bimodal distribution best fit the results for PWVs higher than 1 m/d. It predicted a removal of approximately 4 log10 after 50 days infiltration at 1 m/d. At PWVs below 1 m/d the model underestimated removal. Sand-bound phages dissociated slowly into the liquid phase, with a detachment constant kdet of 2.6 × 10⁻⁵. This low kdet suggests that river bank filtration plants should be intermittently operated when viral overload is suspected, e.g. during flooding events or at high water-marks in rivers, in order for viruses to become soil-associated during the periods of standstill. Resuming filtration will allow only a very slow virus release from the soil.

Unlabelled: A modification of the UK reference and ISO 14189 TSCA medium for the enumeration of Clostridium perfringens from water coupled with a membrane filter transfer technique for testing for production of acid phosphatase was evaluated. The new tryptose cycloserine agar (TCA) medium, which lacks sodium metabisulphite but contains sodium pyruvate to improve recovery, allows the isolation and confirmation of Cl. perfringens within 18-24 h of sample processing. Data from a multilaboratory study analysed according to ISO 17994 showed that TCA was equivalent to TSCA for the enumeration of Cl. perfringens. The identification of acid phosphatase-negative isolates revealed a false-negative rate for the TCA method of 0.8%. The TCA membrane filter transfer procedure provides confirmed Cl. perfringens counts in half the time of the TSCA method and is simple to undertake., Significance and Impact of the Study: The testing of drinking water for Clostridium perfringens is a regulatory parameter in Europe and the UK. Current UK and ISO methods employ membrane filtration (MF) and TSCA medium followed by subculture and confirmation of isolates by testing for acid phosphatase. This takes 48 h. We present here the results of a multilaboratory evaluation of a MF method that features a simplified isolation medium (TCA) and a membrane transfer procedure for the acid phosphatase test resulting in confirmed results being available in 18-24 h. In order to study the chemical composition of aquatic microbes it is necessary to obtain completely separated fractions of subpopulations. Size separation by filtration is usually unsuccessful because the smaller group of organisms contaminates the larger fractions due to being trapped on filter surfaces of nominally much larger pore sizes. Here we demonstrate that a simple sucrose density separation method allowed us to separate microorganisms of even subtle size differences and to determine their bulk biochemical composition (proteins, polysaccharides+nucleic acids, and lipids). Both autotrophs and heterotrophs (through anaplerotic pathways) were labeled with (14)C-bicarbonate for biochemical fractionation. We provided proof of concept that eukaryotic microbes could be cleanly separated from prokaryotes in cultures and in field samples, enabling detection of differences in their biochemical makeup. We explored methodological issues regarding separation mechanisms, fixation, and pre-concentration via tangential flow filtration of oligotrophic marine waters where abundances of microorganisms are comparably low. By selecting an appropriate centrifugal force, two processes (i.e., isopycnal and rate-zonal separation) can be exploited simultaneously resulting in finely-separated density fractions, which also resulted in size separation. EPA Method 1615 was developed with a goal of providing a standard method for measuring enteroviruses and noroviruses in environmental and drinking waters. The standardized sampling component of the method concentrates viruses that may be present in water by passage of a minimum specified volume of water through an electropositive cartridge filter. The minimum specified volumes for surface and finished/ground water are 300 L and 1,500 L, respectively. A major method limitation is the tendency for the filters to clog before meeting the sample volume requirement. Studies using two different, but equivalent, cartridge filter options showed that filter clogging was a problem with 10% of the samples with one of the filter types compared to 6% with the other filter type. Clogging tends to increase with turbidity, but cannot be predicted based on turbidity measurements only. From a cost standpoint one of the filter options is preferable over the other, but the water quality and experience with the water system to be sampled should be taken into consideration in making filter selections.

Small-scale concentration of viruses (sample volumes 1-10 L, here simulated with spiked 100 ml water samples) is an efficient, cost-effective way to identify optimal parameters for virus concentration. Viruses can be concentrated from water using filtration (electropositive, electronegative, glass wool or size exclusion), followed by secondary concentration with beef extract to release viruses from filter surfaces, and finally tertiary concentration resulting in a 5-30 ml volume virus concentrate. In order to identify optimal concentration procedures, two different electropositive filters were evaluated (a glass/cellulose filter [1MDS] and a nano-alumina/glass filter [NanoCeram]), as well as different secondary concentration techniques; the celite technique where three different celite particle sizes were evaluated (fine, medium and large) followed by comparing this technique with that of the established organic flocculation method. Various elution additives were also evaluated for their ability to enhance the release of adenovirus (AdV) particles from filter surfaces. Fine particle celite recovered similar levels of AdV40 and 41 to that of the established organic flocculation method when viral spikes were added during secondary concentration. The glass/cellulose filter recovered higher levels of both, AdV40 and 41, compared to that of a nano-alumina/glass fiber filter. Although not statistically significant, the addition of 0.1% sodium polyphosphate amended beef extract eluant recovered 10% more AdV particles compared to unamended beef extract.

The global need to improve bacterial detection in liquid media has motivated multidisciplinary research efforts toward developing new approaches that overcome the shortcomings of traditional techniques. We recently proposed the use of oligomers of acylated lysyls (OAKs) in their resin-linked form (ROAKs) for the efficient, robust, and inexpensive filtration of bacteria. Here, to investigate the potential for the use of ROAKs in downstream applications, we first examined the capacity of ROAKs to capture bacteria as a function of environmental conditions and structure-activity relationships (SARs). We next assessed their ability to release the captured bacteria and then combined both abilities to improve real-time PCR outcomes. ROAKs were able to deplete liquid samples of bacterial content after incubation or continuous flow, illustrating the efficient capture of different bacterial species under a wide range of ionic strength and pH conditions. We also show circumstances for the significant release of captured bacteria, live or dead, for further analysis. Finally, the SAR study revealed a shorter ROAK derivative exhibiting a capture capacity similar to that of the parent construct but the increased recovery of ROAK-bound bacteria, enabling improvement of the detection sensitivity by 20-fold. Introduction and Objective: Mycobacteria have been isolated from almost all types of natural waters, as well as from man-made water distribution systems. Detection of mycobacteria using PCR has been described in different types of water; however, currently, there is no standardised protocol for the processing of large volumes of water., Material and Methods: In the present study, different filtering methods are tested and optimised for tap or river water filtration up to 10 L, as well as filter processing and DNA isolation using four commercially available kits., Results: The PowerWater DNA isolation kit (MoBio, USA), together with a kit used for soil and other environmental samples (PowerSoil DNA isolation kit, MoBio), had the highest efficiency. Filtration of 10 L of water and elution of the filter in PBS with the addition of 0.05% of Tween 80 is suggested., Conclusions: The described protocol for filter elution is recommended, and the use of the PowerWater DNA isolation kit for the highest mycobacterial DNA yield from water samples. The described protocol is suitable for parallel detection of mycobacteria using cultivation.

Background: Opportunistic waterborne pathogens (eg, Legionella, Pseudomonas) may persist in water distribution systems despite municipal chlorination and secondary disinfection and can cause health care-acquired infections. Point-of-use (POU) filtration can limit exposure to pathogens; however, their short maximum lifetime and membrane clogging have limited their use., Methods: A new faucet filter rated at 62 days was evaluated at a cancer center in Northwestern Pennsylvania. Five sinks were equipped with filters, and 5 sinks served as controls. Hot water was collected weekly for 17 weeks and cultured for Legionella, Pseudomonas, and total bacteria., Results: Legionella was removed from all filtered samples for 12 weeks. One colony was recovered from 1 site at 13 weeks; however, subsequent tests were negative through 17 weeks of testing. Total bacteria were excluded for the first 2 weeks, followed by an average of 1.86 log reduction in total bacteria compared with controls. No Pseudomonas was recovered from filtered or control faucets., Conclusion: This next generation faucet filter eliminated Legionella beyond the 62 day manufacturers' recommended maximum duration of use. These new POU filters will require fewer change-outs than standard filters and could be a cost-effective method for preventing exposure to Legionella and other opportunistic waterborne pathogens in hospitals with high-risk patients., (Copyright © 2014 Association for Professionals in Infection Control and Epidemiology, Inc. The fecal coliform, Escherichia coli, is a historical organism for the detection of fecal pollution in water supplies. The presence of E. coli indicates a potential contamination of the water supply by other more hazardous human pathogens. In order to accurately determine the presence and degree of fecal contamination, it is important that standard methods approved by the US Environmental Protection Agency are designed to determine the presence of E. coli in a water supply, and distinguish E. coli from other coliform bacteria (e.g. Citrobacter, Klebsiella and Enterobacter). These genera of bacteria are present not only in fecal matter, but also in soil and runoff water and are not good indicators of fecal contamination. There is also ambiguity in determining a positive result for fecal coliforms on M-FC filters by a blue colony. When all variations of blue, including light blue or glossy blue, were examined, confirmation methods agreed with the positive M-FC result less often than when colonies that the technician would merely call "blue", with no descriptors, were examined. Approximately 48 % of M-FC positive colonies were found to be E. coli with 4 methylumbelliferyl-β-D-glucuronide (MUG), and only 23 % of samples producing a positive result on M-FC media were found to be E. coli using API-20E test strips and current API-20E profiles. The majority of other M-FC blue colonies were found to be Klebsiella or were unidentifiable with current API-20E profiles. Two positive M-FC colonies were found to be Kluyvera with API-20E, both of which cleaved MUG and produced fluorescence under UV light, a characteristic used to differentiate E. coli from other fecal coliforms.

Anodic aluminum oxide (AAO) filters have high porosity and can be manufactured with a pore size that is small enough to quantitatively capture viruses. These properties make the filters potentially useful for harvesting total microbial communities from water samples for molecular analyses, but their performance for nucleic acid extraction has not been systematically or quantitatively evaluated. In this study, we characterized the flux of water through commercially produced nanoporous (0.02 μm) AAO filters (Anotop; Whatman) and used isolates (a virus, a bacterium, and a protist) and natural seawater samples to test variables that we expected would influence the efficiency with which nucleic acids are recovered from the filters. Extraction chemistry had a significant effect on DNA yield, and back flushing the filters during extraction was found to improve yields of high-molecular-weight DNA. Using the back-flush protocol, the mass of DNA recovered from microorganisms collected on AAO filters was ≥ 100% of that extracted from pellets of cells and viruses and 94% ± 9% of that obtained by direct extraction of a liquid bacterial culture. The latter is a minimum estimate of the relative recovery of microbial DNA, since liquid cultures include dissolved nucleic acids that are retained inefficiently by the filter. In conclusion, we demonstrate that nucleic acids can be extracted from microorganisms on AAO filters with an efficiency similar to that achievable by direct extraction of microbes in suspension or in pellets. Exposure to various infectious viruses in environmental drinking water can constitute a public health risk. However, it is difficult to detect viruses in water due to their low concentration. In this study, we have developed a novel filter cartridge system containing electropositive granule media (EGM). Viruses present in large volumes of environmental samples were adsorbed onto the EGM, and then recovered by elution and poly(ethylene glycol) (PEG) concentration. To evaluate the system's efficiency in viral recovery, poliovirus (PV-1), a surrogate for enteric viruses, was used to artificially contaminate river water samples which were then assayed by quantitative real-time PCR. To optimize the concentration procedure, the eluent type, water flow rate and properties (e.g., pH, bacterial, and viral loads), were evaluated. The highest virus recovery was obtained by pumping river water at a flow rate of 300 mL/min and then pushing 3 L of an eluent containing 3× broth [1.5% (w/v) NaCl, 3% (w/v) tryptone, 1.5% (w/v) beef powder] with 0.05 mol/L glycine through the filter. Using this procedure, the recovery efficiencies of PV-1 from 10 to 100 L of spiked river water were up to 99%. In addition, this method is virus load and pH dependent. Virus recovery was maximal at a load of between 10(3.5) and 10(5.5) TCID50 and a pH ranging from 5 to 7. The bacterial load in the water has no effect on virus recovery. Different types of viruses and surface water were tested to validate the system's applicability. Results revealed that the EGM filter cartridge was able to concentrate PV-1, human adenoviruses (HAdVs) and noroviruses (HuNoVs) with high efficiency from river, lake, and reservoir water. Furthermore, it showed more efficient recovery than glass wool and 1MDS filters. These data suggest that this system provides rapid and efficient virus recovery from a large volume of natural surface water and, as such, could be a useful tool in revealing the presence of viruses in surface water.

Waterborne transmission plays an essential role in the transmission and spread of avian influenza viruses. The abundance of influenza viruses in environmental water is usually extremely low and viruses or viral genomes can hardly be detected by conventional reverse transcription (RT-) PCR without concentration. In the present study, an electropositive filter membrane was used to concentrate influenza viruses from water sample, in addition, a glass fiber filter has been used prior to positive charged membrane for the prefiltration. Unlike the traditional adsorption-elution method, Trizol-LS reagent was used to lyse the viruses attached directly to the electropositive filter membrane and the influenza virus genomic RNA was extracted, followed by RT-PCR analysis. The method established in this study could improve the efficiency of the conventional RT-PCR technique used to detect the M, NP, and HA genes of influenza virus in natural water samples. Standard filtration methods have been characterized by poor recoveries when processing large-volume samples of environmental water. A method to pre-remove particulates present in turbid waters would be necessary to enhance recovery of protozoan oocysts. Particulate separation can be achieved by the proposed multiplex particle refining (MPR) system. This system employs multiple counter-flow microfiltration units that are arranged into a cascade-like structure. By use of this design, the target oocysts are pre-concentrated from environmental waters. The performance of the MPR system was investigated using 10-L deionized water and surface water spiked with 100 Cryptosporidium parvum oocysts. A recovery rate of around 85% was obtained for spiked river water. The water samples were processed using high flow rate and a simple filtration protocol. Further experiments were conducted using the MPR as a pre-filter for five commercially available filters. The recovery rates were two- to threefold higher employing the pre-filter than using the filters alone. The merit of the refining system to use different numbers of counter-flow units led to superior oocyst recovery rate for the Filta-Max and Envirochek HV filters, which are approved by the US Environmental Protection Agency. This work demonstrates a feasible tool for improved filtration performance in environmental waters.

The present study focuses on the effect of filter media (riverbed gravel, mud balls, coal and glass balls) for the removal of pathogens during vermifiltration of domestic wastewater. This novel study was conducted for the first time on different vermifilters (VFs) with different media, and pathogen removal was extensively investigated for a period of 3 months. Results indicated that vermifilter with riverbed material (VFR) showed maximum biochemical oxygen demand and chemical oxygen demand removal of 76 and 67%, respectively and vermifilter with mud balls (VFM) showed maximum log removal of total coliforms (2.8), fecal coliforms (2.7), fecal streptococci (2.2), Salmonella (2.1) and Escherichia coli (2.1). The area-based bacterial removal rate constant (k) was found in the range of 2.96-6.68 m/d, which is very high in comparison to the reported values. The population of total heterotrophic bacteria and total fungi was found to be 2-log (99%) higher in VFR and VFM, as compared to other media. The growth rate of Eisenia fetida was higher (42% increase in biomass) in glass balls vermifilter (VFG), and also the abrasions on the body wall of earthworms in VFG showed fewer injuries. Overall, the results of the study described the importance and role of each filter medium.

Effective household water treatment can improve drinking water quality and prevent disease if used correctly and consistently over time. One year after completion of a randomized controlled study of water filters among households in Zambia with children < 2 years old and mothers who were human immunodeficiency virus (HIV)-positive, we conducted a follow-up study to assess use and performance of new filters distributed at the conclusion of the study; 90% of participating households met the criteria for current users, and 75% of participating households had stored water with lower levels of fecal contamination than source water. Microbiologically, the filters continued to perform well, removing an average of 99.0% of fecal indicator bacteria. Although this study provides some encouraging evidence about the potential to maintain high uptake and filter performance, even in the absence of regular household visits, additional research is necessary to assess whether these results can be achieved over longer periods and with larger populations.

Unlabelled: The registered infection and outbreaks of epidemic tend to monitor potential reservoirs of Legionella infection. According to the Act of 29 March 2007 on the requirements for the quality of water intended for human consumption are required to test for the presence and number of Legionella in the water system of hospitals. In case of detection of L. pneumophila serogroup 1 (SG 1) or increased above normal number other serogroups of bacteria it is necessary to eradicate these bacteria from the water system. The aim of this study was to assess the degree of contamination of the water supply system of selected public buildings and analyze the effectiveness of disinfection methods for the elimination of L. pneumophila in hot water systems., Material and Methods: The materials for this study were hot and cold water samples which were collected from the water supply system of 23 different objects. Enumeration of Legionella bacteria in water samples was determined by membrane filtration (FM) and/or by surface inoculation methods according to the standards: PN-ISO 11731: 2002: "The quality of the water. Detection and enumeration of Legionella" and PN-EN ISO 11731-2: 2008: "Water quality--Detection and enumeration of Legionella--Part 2: Methodology of membrane filtration for water with a small number of bacteria"., Results: L. pneumophila was present in 164 samples of hot water, which accounted for 76.99%. In all tested water samples L. pneumophila SG 2-14 strains were detected. The most virulent strain--L. pneumophila SG 1 was not detected. In examined 23 objects in 12 of L. pneumophila exceed acceptable levels > 100 CFU/100 ml., Conclusions: The presence of L. pneumophila SG 2-14 demonstrated in all examined objects, indicating the risk of infection, and the need for permanent monitoring of the water system supply. The thermal disinfection is the most common, inexpensive, and effective method of control of L. pneumophila used in examined objects, but does not eliminate bacterial biofilm. Disinfection using the filters stopped of L. pneumophila, and was the method of complementary thermal disinfection. Chlorine dioxide is a very effective biocide for large numbers of L. pneumophila in water systems.

The effects of Tween-20, a non-ionic surfactant, and Zn (II) on microbial activity and removal performance for ethylbenzene in a biotrickling filter (BTF) were evaluated. Batch experiments were conducted to evaluate the surfactant and Zn (II) at various concentrations for their toxicity to microorganisms, and results indicated that Tween-20 was beneficial to microbial activity at all the tested concentration, while Zn (II) affected adversely when the concentration overpassed 5.0mgL(-1). Then effects of the two additives on removal efficiency of ethylbenzene were evaluated in a BTF at an empty-bed retention time of 30s and an ethylbenzene concentration of 1100mgm(-3). Results showed that the optimal concentrations of Tween-20 and Zn (II) were about 12 and 1.0mgL(-1), respectively. Compared to the results when neither of the two additives was added, Tween-20 improved ethylbenzene removal efficiency from 67% to 86% at the optimal condition, while on that basis, Zn (II) just increased the removal efficiency from 86% to 90%. In iron and manganese-containing groundwater treatment for drinking water production, biological filter is an effective process to remove such pollutants. Until now the exact microbial mechanism of iron and manganese removal, especially coupled with other pollutants, such as ammonia, has not been clearly understood. To assess this issue, the performance of a full-scale biofilter located in Harbin, China was monitored over four months. Microbial populations in the biofilter were investigated using T-RFLP and clone library technique. Results suggested that Gallionella, Leptothrix, Nitrospira, Hyphomicrobium and Pseudomonas are dominant in the biofilter and play major roles in the removal of iron, manganese and ammonia. The spatial distribution of microbial populations along the depth of the biofilter demonstrated the stratification of the removal of iron, manganese and ammonia. Additionally, the absence of ammonia-oxidizing bacteria in the biofilter implicated that ammonia-oxidizing archaea might be responsible for the oxidation of ammonia to nitrite., (Crown Copyright © 2013. Water and wastewater filtration systems often house pathogenic bacteria, which must be removed to ensure clean, safe water. Here, we determine the persistence of the model bacterium Pseudomonas aeruginosa in two types of filtration systems, and use P. aeruginosa bacteriophages to determine their ability to selectively remove P. aeruginosa. These systems used beds of either anthracite or granular activated carbon (GAC), which were operated at an empty bed contact time (EBCT) of 45 min. The clean bed filtration systems were loaded with an instantaneous dose of P. aeruginosa at a total cell number of 2.3 (± 0.1 [standard deviation]) × 10(7) cells. An immediate dose of P. aeruginosa phages (1 mL of phage stock at the concentration of 2.7 × 10(7) PFU (Plaque Forming Units)/mL) resulted in a reduction of 50% (± 9%) and >99.9% in the effluent P. aeruginosa concentrations in the clean anthracite and GAC filters, respectively. To further evaluate the effects of P. aeruginosa phages, synthetic stormwater was run through anthracite and GAC biofilters where mixed-culture biofilms were present. Eighty five days after an instantaneous dose of P. aeruginosa (2.3 × 10(7) cells per filter) on day 1, 7.5 (± 2.8) × 10(7) and 1.1 (± 0.5) × 10(7) P. aeruginosa cells/g filter media were detected in the top layer (close to the influent port) of the anthracite and GAC biofilters, respectively, demonstrating the growth and persistence of pathogenic bacteria in the biofilters. A subsequent 1-h dose of phages, at the concentration of 5.1 × 10(6) PFU/mL and flow rate of 1.6 mL/min, removed the P. aeruginosa inside the GAC biofilters and the anthracite biofilters by 70% (± 5%) and 56% (± 1%), respectively, with no P. aeruginosa detected in the effluent, while not affecting ammonia oxidation or the ammonia-oxidizing bacterial community inside the biofilters. These results suggest that phage treatment can selectively remove pathogenic bacteria with minimal impact on beneficial organisms from attached growth systems for effluent quality improvement., (Copyright © 2013 Elsevier Ltd. This paper describes a method to detect the presence of bacteria in aqueous samples, based on the capture of bacteria on a syringe filter, and the infection of targeted bacterial species with a bacteriophage (phage). The use of phage as a reagent provides two opportunities for signal amplification: (i) the replication of phage inside a live bacterial host and (ii) the delivery and expression of the complementing gene that turns on enzymatic activity and produces a colored or fluorescent product. Here we demonstrate a phage-based amplification scheme with an M13KE phage that delivers a small peptide motif to an F(+), α-complementing strain of Escherichia coli K12, which expresses the ω-domain of β-galactosidase (β-gal). The result of this complementation-an active form of β-gal-was detected colorimetrically, and the high level of expression of the ω-domain of β-gal in the model K12 strains allowed us to detect, on average, five colony-forming units (CFUs) of this strain in 1 L of water with an overnight culture-based assay. We also detected 50 CFUs of the model K12 strain in 1 L of water (or 10 mL of orange juice, or 10 mL of skim milk) in less than 4 h with a solution-based assay with visual readout. The solution-based assay does not require specialized equipment or access to a laboratory, and is more rapid than existing tests that are suitable for use at the point of access. This method could potentially be extended to detect many different bacteria with bacteriophages that deliver genes encoding a full-length enzyme that is not natively expressed in the target bacteria.

Since the beginning of environmental virology in the mid-twentieth century, a key challenge to scientists in the environmental field has been how to collect, isolate and detect pathogenic viruses from water that is used for drinking and/or recreational purposes. Early studies investigated different types of membrane filters, with more sophisticated technologies being developed more recently. The purpose of this study was to look at the current state of the science of methods for the concentration of viruses from water. Several technologies were reviewed, and associated data were included in a meta-analysis which showed that electronegative filters, electropositive filters and ultrafilters are comparable in performance and that significant differences in recovery are due to virus type rather than filter type, water matrix or sample volume. This information is useful, as it will help to determine which method(s) should be used, particularly if there is a specific viral type being targeted for a particular study. In addition, it will be helpful when sampling different environmental water matrices and/or when budget allowance must be taken into consideration.
