Fiber-based materials have emerged as a promising option to increase the efficiency of water treatment plants while reducing their environmental impacts, notably by reducing the use of unsustainable chemicals and the size of the settling tank. Cellulose fiber-based super-bridging agents are sustainable, reusable, and versatile materials that considerably improve floc separation in conventional settling tanks or via alternative screening separation methods. In this study, the effectiveness of fiber-based materials for wastewater treatment was evaluated at lab-scale (0.25 L) and at pilot-scale (20 L) for two separation methods, namely settling and screening. For the fiber-based method, the performance of floc separation during settling was slightly affected by an 80x upscaling factor. A small decrease in turbidity removal from 93 and 86 % was observed for the jar and pilot tests, respectively. By contrast, the turbidity removal of the conventional treatment, i.e., no fibers with a settling separation, was largely affected by the upscaling with turbidity removals of 84 and 49 % for jar and pilot tests, respectively. Therefore, results are suggesting that fiber-based super-bridging agents could be implemented in full-scale water treatment plants. Moreover, the tested fibers increase the robustness of treatment by providing better floc removal than conventional treatment under several challenging conditions such as low settling time and screening with coarse screen mesh size. Furthermore, at both lab-scale and pilot-scale, the use of fiber-based materials reduced the demand for coagulant and flocculant, potentially lowering the operational costs of water treatment plants and reducing the accumulation of metal-based coagulants and synthetic polymers in sludge. Acute toxicity tests using the model organism Daphnia magna show that the cellulose fibers introduce insignificant toxicity at the optimized fiber concentration. Although dedicated mechanistic studies are required at various scales to understand in detail the influence of fibers on water treatment (coagulation/flocculation time, floc formation, floc size distribution velocity gradient, etc.), the efficacy and scalability of the fiber-based approach, along with its minimal environmental impact, position it as a viable and sustainable option for existing and future wastewater treatment plants., Competing Interests: Declaration of competing interest M. Single-use face masks can release microfibres upon exposure to environmental conditions. This study investigates the number of microfibres released in the presence and absence of UV irradiation and mechanical friction and the removal of the released microfibres in a simulated conventional wastewater treatment process. UV exposure results in a four-fold increase in the number of microfibres released from new masks and used masks resulting in ~2400 microfibres/mask and ~1100 microfibres/mask, respectively. Application of mechanical friction to the UV-exposed new and used masks further increases the number of released microfibres per mask. In a simulated coagulation/flocculation process, the removals of microfibers originating from new masks and used masks are 79% and 91%, respectively. XPS analysis reveals that the silica content of the used masks is 240% higher than that of new masks, which could explain the higher removal efficiency of microfibers from used masks. FTIR analysis of the masks after UV exposure shows carbonyl indices of 0.73 ± 0.70 and 0.27 ± 0.10 for the microfibres from used and new masks, respectively. The efficacy of plastic particle removal by municipal water treatment plants is currently uncertain, and the mechanisms involved in microplastic (MP) coagulation and flocculation have only been superficially investigated. The removal of pristine versus weathered plastic debris and the impact of plastic particle size on removal remain largely unexplored. In this study, coagulation, flocculation, and settling performances were investigated using pristine and weathered MPs (polyethylene (PE) and polystyrene (PS) microspheres, and polyester (PEST) fibers). Weathering processes that changed the surface chemistry and roughness of MPs impacted MP affinity for coagulants and flocculants. A quartz crystal microbalance with dissipation monitoring was used to identify the mechanisms involved during MP coagulation and flocculation. Measured deposition rates confirmed the relatively low affinity between plastic surfaces and aluminum-based coagulants compared to cationic polyacrylamide (PAM). In every case examined, coagulant efficiency increased when the plastic surface was weathered. Removals of 97 and 99% were measured for PEST and weathered PE, respectively. Larger pristine PE MPs were the most resistant to coagulation and flocculation, with 82% removal observed even under enhanced coagulation conditions. By understanding the interaction mechanisms, the removal of weathered MPs was optimized. Finally, this study explored the use of settled water turbidity as a possible indicator of MP removal.

Self-assembly of graphene oxide (GO) nanosheets into porous 3D sponges is a promising approach to exploit their capacity to adsorb contaminants while facilitating the recovery of the nanosheets from treated water. Yet, forming mechanically robust sponges with suitable adsorption properties presents a significant challenge. Ultra-strong and highly porous 3D sponges are formed using GO, vitamin C (VC), and cellulose nanocrystals (CNCs) - natural nanorods isolated from wood pulp. CNCs provide a robust scaffold for the partially reduced GO (rGO) nanosheets resulting in an exceptionally stiff nanohybrid. The concentration of VC as a reducing agent plays a critical role in tailoring the pore architecture of the sponges. By using excess amounts of VC, a unique hierarchical pore structure is achieved, where VC grains act as soft templates for forming millimeter-sized pores, the walls of which are also porous and comprised of micron-sized pores. The unique hierarchical pore structure ensures the interconnectivity of pores even at the core of large sponges as evidenced by micro and nano X-ray computed tomography. The unique pore architecture translates into an exceptional specific surface area for adsorption of a wide range of contaminants, such as dyes, heavy metals, pharmaceuticals and cyanotoxin from water.

Electrochemical disinfection has been shown to be an efficient method with a shortrequired contact time for treatment of drinking water supplies, industrial raw water supplies, liquid foodstuffs, and wastewater effluents. In the present work, the electrochemical disinfection of saline water contaminated with bacteria was investigated in chloride-containing solutions using Sb-doped Sn 80% -W 20% -oxide anodes. The influence of current density, bacterial load, initial chloride concentration, solution pH, and the type of bacteria (E. coli D21, E. coli O157:H7, and E. faecalis) on disinfection efficacy was systematically examined. The impact of natural organic matter and a radical scavenger on the disinfection process was also examined. The electrochemical system was highly effective in bacterial inactivation for a 0.1 M NaCl solution contaminated with ∼10 7  CFU/mL bacteria by applying a current density ≥1 mA/cm 2 through the cell.100% inactivation of E. coli D21 was achieved with a contact time of less than 60 s and power consumption of 48 Wh/m 3 , by applying a current density of 6 mA/cm 2 in a 0.1 M NaCl solution contaminated with ∼10 7 CFU/mL. Reactive chlorine species as well as reactive oxygen species (e.g. hydroxyl radicals) generated in situ during the electrochemical process were determined to be responsible for inactivation of bacteria., (Copyright © 2017 Elsevier Ltd. Attached growth biological treatment systems are a promising solution to ammonia removal in cold-temperature climates. Environmental scanning electron microscopy (ESEM) and confocal laser scanning microscopy in combination with fluorescent in situ hybridization (FISH) was used to investigate the effects of 4 months of exposure to 4 degrees C on nitrifying biofilm and biomass. These molecular and microscopic methods were modified to minimize loss of mass and distortion of in situ perspectives. Environmental scanning electron microscopy revealed that nitrifying biofilm did not exhibit significant changes in volume with exposure to 4 degrees C. Confocal laser scanning microscopy in combination with FISH showed that the number of ammonia-oxidizing bacteria (AOB) cells present in the biofilm was statistically consistent during exposure to 4 degrees C. The RNA content of AOB cells remained sufficient for FISH enumeration. The number of nitrite-oxidizing bacteria cells remained steady during exposure to 4 degrees C; however, the RNA content of the cells appeared to decrease with exposure to 4 degrees C, thereby preventing their enumeration using FISH.

Increased popularity of attached-growth wastewater treatment systems (e.g. biological aerated filtration processes-BAF) has created the need for a rapid and reliable method of characterizing biofilms. In addition to the mass of the biofilm that may serve as a control parameter for attached-growth treatment systems, the nitrogen content of the biofilm is also of great interest with increasingly strict nitrogen removal guidelines. Existing methods that may be used to analyse biofilms in such processes involve complex sample preparation and microbiological expertise that limit their application in many biofilm wastewater treatment studies and at existing treatment facilities as a feasible method of monitoring the biofilm. This paper describes a simple technical procedure that enables biofilm samples attached to polystyrene beads to be characterized in terms of the biofilm mass and the nitrogen content of the biofilm. The proposed protocol incorporates an agitation procedure that demonstrates 99.9% removal of the biofilm from polystyrene beads; a modified TSS procedure that measures the removed biofilm mass; and subsequently a modified total Kjeldahl nitrogen (TKN) procedure that enables the nitrogen content of the biofilm to be measured directly on the filter. Moreover, this protocol allows numerous beads to be analysed with limited manipulation and without the loss of critical mass.

Owing to its widespread occurrence in drinking water supplies and its significant resistance to environmental stresses, Cryptosporidium parvum is regarded as one of the most important waterborne microbial parasites. Accordingly, a substantial research effort has been aimed at elucidating the physical, chemical and biological factors controlling the transport and removal of Cryptosporidium oocysts in natural subsurface environments and drinking water treatment facilities. In this review, a multi-scale approach is taken to discuss the current state-of-knowledge on Cryptosporidium-sand interactions at a nano-scale, bench-scale and field-scale relevant to water treatment operations. Studies conducted at the nano-scale and bench-scale illustrate how techniques based on the principles of colloid and surface chemistry are providing new insights about oocyst-sand interactions during transport of Cryptosporidium oocysts in granular porous media. Specifically, atomic force microscopy and impinging jet experiments reveal the importance of oocyst surface biomolecules in controlling Cryptosporidium/sand interactions by a mechanism of steric hindrance. Traditional bench-scale column transport studies conducted over a broad range of experimental conditions highlight the role of physicochemical filtration and physical straining in the removal of oocysts from the pore fluid. Such experiments have also been used to evaluate the influence of biofilms formed on grain surfaces and the presence of natural organic matter on oocyst-sand interactions. Whilst filtration studies conducted at the plant-scale have been useful for evaluating the effectiveness of various materials as surrogates for Cryptosporidium oocysts, at this macro-scale, little could be learnt about the fundamental mechanisms controlling oocyst-sand interactions. This review of the literature on Cryptosporidium-sand interactions at different length scales points to the importance of combining studies at the plant-scale with well-controlled investigations conducted at the nano- and bench-scales. Furthermore, because oocyst surface properties play an important role in controlling the extent of interaction with sand surfaces, a thorough discussion of Cryptosporidium oocyst characteristics and electrical properties is presented.

The transport and filtration behavior of Cryptosporidium parvum oocysts in columns packed with quartz sand was systematically examined under repulsive electrostatic conditions. An increase in solution ionic strength resulted in greater oocyst deposition rates despite theoretical predictions of a significant electrostatic energy barrier to deposition. Relatively high deposition rates obtained with both oocysts and polystyrene latex particles of comparable size at low ionic strength (1 mM) suggest that a physical mechanism may play a key role in oocyst removal. Supporting experiments conducted with latex particles of varying sizes, under very low ionic strength conditions where physicochemical filtration is negligible, clearly indicated that physical straining is an important capture mechanism. The results of this study indicate that irregularity of sand grain shape (verified by SEM imaging) contributes considerably to the straining potential of the porous medium. Hence, both straining and physicochemical filtration are expected to control the removal of C. parvum oocysts in settings typical of riverbank filtration, soil infiltration, and slow sand filtration. Because classic colloid filtration theory does not account for removal by straining, these observations have important implications with respect to predictions of oocyst transport.

Abstract: Bacteriophages are bacterial viruses with unique characteristics that make them excellent surrogates for mammalian pathogenic viruses in environmental studies. Simple and reliable methodologies for isolation, detection, characterization and enumeration of somatic and F-specific bacteriophage are available in the literature. Limited information or methods are available for producing high-titer purified phage suspensions for studying microbial transport and survival in natural and engineered environments. This deficiency arises because most research on the production of high-titer phage suspensions was completed over half a century ago and more recent advances on these methods have not been compiled in a single publication. We present a review of the available methods and new data on the propagation, concentration and purification of two bacteriophage host systems (somatic PRD1/Salmonella thyphimurium and F-specific PR772/Escherichia coli) that are commonly utilized in laboratory and field-scale assessments of subsurface microbial transport and survival. The focus of the present study is to recommend the approach(es) that will ensure maximum bacteriophage yields while optimizing suspension purification (i.e. avoiding modification of surface charge of the phage capsids and/or inadvertent introduction of dissolved organic matter to the study system).
