Glufosinate is an important and widely used non-selective herbicide active on a wide range of plant species. Evolution of resistance to glufosinate in weedy plant species (including the global weed Eleusine indica) is underway. Here, we established the molecular basis of target site glufosinate resistance in Eleusine indica. Full-length E. indica glutamine synthetase (GS) iso-genes (EiGS1-1 , 1-2 , 1-3 , and EiGS2) were cloned, and expression of EiGS1-1 and EiGS1-2 was higher than that of EiGS2. A novel point mutation resulting in a Ser59Gly substitution in EiGS1-1 was identified in glufosinate-resistant plants. Rice calli and seedlings transformed with the mutant EiGS1-1 gene were resistant to glufosinate. Purified mutant EiGS1-1 expressed in yeast was more tolerant to glufosinate than the wild-type variant. These transgenic results correlate with a more glufosinate-resistant GS in the crude tissue extract of resistant versus susceptible E. indica plants. Structural modelling of the mutant EiGS1-1 revealed that Ser59 is not directly involved in glufosinate binding but is in contact with some important binding residues (e.g. Glu297) and especially with Asp56 that forms an intratoroidal contact interface. Importantly, the same Ser59Gly mutation was also found in geographically isolated glufosinate-resistant populations from Malaysia and China, suggesting parallel evolution of this resistance mutation. The introduction of harvest weed seed control (HWSC) techniques and associated machinery has enabled the routine use of an alternative weed control technology at a novel weed control timing in global grain cropping fields. Driven by the significant threat of widespread populations of annual ryegrass (Lolium rigidum) with multiple-herbicide resistance, in the 1990s Australian growers and researchers developed techniques to target, at grain harvest, the seed production of annual ryegrass and other important weed species. The HWSC approach to weed management is now routinely used by a majority of Australian grain producers as an integral component of effective weed control programs. Here we detail the development and introduction of current HWSC systems and describe their efficacy in Australian grain production systems. The use of HWSC has likely contributed to lower annual ryegrass population densities and thus mitigated the impacts of herbicide resistance as well as slowing further evolution of resistance. In addition, low weed densities enable the introduction of site-specific weed control technologies and the opportunity to target specific in-crop weeds with non-selective alternative weed control techniques. With an awareness of the evolutionary potential of weed species to adapt to all forms of weed control, there is an understanding that HWSC treatments need to be judiciously used in grain cropping systems to ensure their ongoing efficacy. The successful use of Australian developed HWSC systems has attracted global interest and there is now a considerable international research effort aimed at introducing this alternative weed control approach and timing into the world's major cropping systems. BACKGROUND: Relatively new herbicides that target 4‐hydroxyphenylpyruvate dioxygenase (HPPD) are now available for use on the world's great grain crops (rice, wheat, corn and soybean) and for other uses. With widespread and persistent use of HPPD‐inhibiting herbicides, the evolution of HPPD‐inhibiting herbicide resistant weeds is inevitable. Currently, resistance to HPPD‐inhibiting herbicides is known in two weed species, waterhemp and Palmer amaranth. Here, we report a HPPD‐inhibiting herbicide resistant wild radish population from the Western Australia grain belt. This population was not selected with HPPD‐inhibiting herbicides, rather it evolved resistance to earlier used herbicides with different modes of action and exhibits cross‐resistance to HPPD‐inhibiting herbicides. RESULTS: Dose–response experiments showed the resistant (R) population exhibits 4 to 6.5‐fold resistance to the HPPD‐inhibiting herbicides mesotrione, tembotrione and isoxaflutole, compared to the susceptible (S) population. This resistance is not target‐site based as cloning of full coding sequences of the HPPD genes from S and R plants did not reveal resistance‐endowing single nucleotide polymorphisms. The HPPD gene expression levels are similar in S and R plants. In addition, no differences in [14C]‐mesotrione uptake and translocation were observed in the S and R plants. However, the time required for R plants to metabolise 50% [14C]‐mesotrione is 7.7‐fold faster than for the S plants. CONCLUSION: We confirm resistance to HPPD‐inhibiting herbicides exists in a population of the economically damaging global weed wild radish. Echinochloa crus-galli is a major weed in rice fields in China, and quinclorac has been long used for its control. Over-reliance of quinclorac has resulted in quinclorac resistance in E. crus-galli. Two resistant (R) E. crus-galli populations from Hunan, China were confirmed to be at least 78-fold more resistant to quinclorac than the susceptible (S) population. No difference in foliar uptake of 14C-labelled quinclorac was detected between the R and S plants. However, a higher level of 14C translocation and a lower level of quinclorac metabolism were found in the R plants. Basal and induced expression levels of β-cyanoalanine synthase (β-CAS) gene and β-CAS activity were not significantly different between the R and S plants. However, the induction expression of 1-aminocyclopropane-1-carboxylic acid oxidase (ACO1) gene by quinclorac treatment was evident in the S plants but not in the R plants. Quinclorac resistance in the two resistant E. crus-galli populations was not likely to be related to foliar uptake, translocation or metabolism of quinclorac, nor to cyanide detoxification via β-CAS. A novel glyphosate resistance double point mutation (T102I/P106S, TIPS) in the 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) gene has been recently identified for the first time only in the weed species Eleusine indica. Quantification of plant resistance cost associated with the TIPS and the often reported glyphosate resistance single P106S mutation was performed. A significant resistance cost (50% in seed number currency) associated with the homozygous TIPS but not the homozygous P106S EPSPS variant was identified in E. indica plants. The resistance cost associated with the TIPS mutation escalated to 85% in plants under resource competition with rice crops. The resistance cost was not detected in nonhomozygous TIPS plants denoting the recessive nature of the cost associated with the TIPS allele. An excess of 11-fold more shikimate and sixfold more quinate in the shikimate pathway was detected in TIPS plants in the absence of glyphosate treatment compared to wild type, whereas no changes in these compounds were observed in P106S plants when compared to wild type. TIPS plants show altered metabolite levels in several other metabolic pathways that may account for the expression of the observed resistance cost. [ABSTRACT FROM AUTHOR]

BACKGROUND Weeds can be a greater constraint to crop production than animal pests and pathogens. Pre-emergence herbicides are crucial in many cropping systems to control weeds that have evolved resistance to selective post-emergence herbicides. In this study we assessed the potential to evolve resistance to the pre-emergence herbicides prosulfocarb + S-metolachlor or pyroxasulfone in 50 individual field Lolium rigidum populations collected in a random survey in Western Australia prior to commercialisation of these pre-emergence herbicides. RESULTS This study shows for the first time that in randomly collected L. rigidum field populations the selection with either prosulfocarb + S-metolachlor or pyroxasulfone can result in concomitant evolution of resistance to both prosulfocarb + S-metolachlor and pyroxasulfone after three generations. CONCLUSIONS In the major weed L. rigidum, traits conferring resistance to new herbicides can be present before herbicide commercialisation. Proactive and multidisciplinary research (evolutionary ecology, modelling and molecular biology) is required to detect and analyse resistant populations before they can appear in the field. Several studies show that evolved cross-resistance in weeds is complex and often unpredictable. Avena is a problematic weed of cropping regions of southern Australia and many areas of the world. In 2010, a random survey was conducted across 14 million hectares of the Western Australian grain belt to monitor the change in herbicide resistance levels by comparing resistance frequency results with a survey conducted in 2005. Screening Avena populations with herbicides commonly used to control this weed revealed that 48% of Avena populations displayed resistance to the commonly used acetyl-Co A carboxylase-inhibiting herbicides, which was lower than that found in 2005 (71%). The broad-spectrum herbicides glyphosate and paraquat provided good control of all Avena populations. Resistance to acetolactate synthase-inhibiting herbicides and to flamprop were detected for the first time in Western Australia in this survey. Raphanus raphanistrum is a problematic weed, which has become increasingly difficult to control in Australian cropping regions. In 2010, a random survey was conducted across 14 million ha of the Western Australian grain belt to establish the frequency of herbicide resistance in R. raphanistrum and to monitor the change in resistance levels by comparing results with a previous survey in 2003. Screening R. raphanistrum populations with herbicides commonly used to control this weed revealed that most populations (84%) contained individual plants resistant to the acetolactate synthase-inhibiting herbicide chlorsulfuron, whereas 49% of populations also had plants resistant to the imidazolinone herbicides. Resistance to other mode of action herbicides (2,4-D (76%) and diflufenican (49%)) was also common. Glyphosate, atrazine and pyrasulfotole + bromoxynil remained effective on most R. raphanistrum populations. These results demonstrate that resistance to some herbicides has increased significantly since 2003 when the values were 54% for chlorsulfuron and 60% for 2,4-D; therefore, a wide range of weed management options that target all phases of the cropping program are needed to sustain these cropping systems in the future. Random surveys conducted in Western Australia have shown that herbicide resistance in major crop weeds is common and increasing. The latest survey on resistance in Raphanus raphanistrum revealed that 84% of populations were resistant to the ALS-inhibiting herbicides, 76% were resistant to synthetic auxins, and 49% to PDS inhibitors. Glyphosate is the most important and widely used herbicide in world agriculture. Intensive glyphosate selection has resulted in the widespread evolution of glyphosate-resistant weed populations, threatening the sustainability of this valuable once-in-a-century agrochemical. Field-evolved glyphosate resistance due to known resistance mechanisms is generally low to modest. Here, working with a highly glyphosate-resistant Eleusine indica population, we identified a double amino acid substitution (T102I + P106S [TIPS]) in the 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) gene in glyphosate-resistant individuals. This TIPS mutation recreates the biotechnology-engineered commercial first generation glyphosate-tolerant EPSPS in corn (Zea mays) and now in other crops. In E. indica, the naturally evolved TIPS mutants are highly (more than 180-fold) resistant to glyphosate compared with the wild type and more resistant (more than 32-fold) than the previously known P106S mutants. The E. indica TIPS EPSPS showed very high-level (2,647-fold) in vitro resistance to glyphosate relative to the wild type and is more resistant (600-fold) than the P106S variant. The evolution of the TIPS mutation in crop fields under glyphosate selection is likely a sequential event, with the P106S mutation being selected first and fixed, followed by the T102I mutation to create the highly resistant TIPS EPSPS. The sequential evolution of the TIPS mutation endowing high-level glyphosate resistance is an important mechanism by which plants adapt to intense herbicide selection and a dramatic example of evolution in action. Twenty-two amino acid substitutions at seven conserved amino acid residues in the acetohydroxyacid synthase (AHAS) gene have been identified to date that confer target-site resistance to AHAS-inhibiting herbicides in biotypes of field-evolved resistant weed species. However, the effect of resistance mutations on AHAS functionality and plant growth has been investigated for only a very few mutations. This research investigates the effect of various AHAS resistance mutations in Lolium rigidum on AHAS functionality and plant growth. The enzyme kinetics of AHAS from five purified L. rigidum populations, each homozygous for the resistance mutations Pro-197-Ala, Pro-197-Arg, Pro-197-Gln, Pro-197-Ser or Trp-574-Leu, were characterized and the pleiotropic effect of three mutations on plant growth was assessed via relative growth rate analysis. All these resistance mutations endowed a herbicide-resistant AHAS and most resulted in higher extractable AHAS activity, with no-to-minor changes in AHAS kinetics. The Pro-197-Arg mutation slightly (but significantly) increased the Km for pyruvate and remarkably increased sensitivity to feedback inhibition by branched chain amino acids. Whereas the Pro-197-Ser and Trp-574-Leu mutations exhibited no significant effects on plant growth, the Pro-197-Arg mutation resulted in lower growth rates. It is clear that, at least in L. rigidum, these five AHAS resistance mutations have no major impact on AHAS functionality and hence probably no plant resistance costs. These results, in part, explain why so many Pro-197 AHAS resistance mutations in AHAS have evolved and why the Pro-197-Ser and the Trp-574-Leu AHAS resistance mutations are frequently found in many weed species. The article presents a discussion on glyphosate-resistant weeds. Glyphosate is a herbicide used around the world to control weeds since 1996. With the increase in the planting of glyphosate-resistant crops in the U. S. came the growth of glyphosate-resistant weeds including Ambrosia artemissifolia L. and Amaranthus palmeri S Watson. Glyphosate-resistant weeds were also found in Argentina and Brazil. The article mentions that glyphosate-resistant weeds are expected to evolve with continued planting of glyphosate-resistant crops. It also discusses the importance of diversifying weed management for the continued use of glyphosate as herbicide.

This article reports on glyphosate, a highly effective broad-spectrum herbicide. It discusses that glyphosate has become the dominant herbicide worldwide because it is very toxicologically and environmentally safe. Glyphosate is the only herbicide that targets 5-enolpyruvyl-shikimate-3-phosphate synthase and it translocates well. Its cost has dropped dramatically after glyphosate became a generic compound. However, the use of this herbicide is being threatened by the evolution of glyphosate-resistant weeds.

In Australian grain-growing regions, the evolution of weeds resistant to multiple herbicides threatens the sustainability of existing cropping systems. As a result, herbicide susceptibility is considered to be a potentially exhaustible resource. Based on a survey of 132 Western Australian grain growers, perceptions of various factors relating to the herbicide resource were measured. These include the rate of resistance evolution, cost of resistance, the future availability of replacement herbicides, reductions in the level of resistance in a population over time, and mobility of resistance. Growers were found to have a generally high level of awareness of herbicide resistance and consider it to have a high potential cost. However, many perceive that the stock of effective herbicides is likely to be renewed in the short term, either through new herbicide development or resistance reversion. The RIM (resistance and integrated management) model is presented. RIM represents a wide diversity of herbicide and non-herbicide options for management of Australia''s most important crop weed, Lolium rigidum, in the context of the non-irrigated extensive farming system of southern Australia. Enterprise choices in the model include cereals, lupins, canola and three types of pastures for grazing by sheep. Users of RIM may specify the enterprise sequence and any feasible combination of the 35 weed treatment options each year over 10 or 20 years. Weed treatment options include selective herbicides (11), non-selective herbicides (5), non-chemical treatments (16) and user-defined treatments (3). The model represents weed and seed bank population dynamics, weed-crop competition, weed treatment impacts (including phytotoxicity), agronomic details, and financial details.
