Alternaria is an important toxigenic fungal genus endangering the quality of cereal grains. No data are available in the literature regarding the composition of Alternaria species in wheat from the main wheat‐cultivating regions of China. In this study, a total of 282 strains were isolated from wheat kernels collected in various provinces. Phylogenetic characterization was performed using two informative loci, ASA10 and ASA19, demonstrating that all strains belonged to A. alternata. A low molecular diversity was observed, which did not appear to be influenced by geographic origin. The secondary metabolite profile of the strains was evaluated in rice culture. Altertoxin I was synthesized at a high concentration by the majority of the strains, while the accumulation of altenuene was lower in both frequency and level than that of alternariol and its derivative. A. alternata did not cause readily observable symptoms on wheat spikes after spraying of the conidia at anthesis, but a higher quantity of altertoxin I and Alternaria DNA were detected in inoculated grains. Artificial inoculations of apple fruit showed that all examined strains were capable of causing typical rot symptoms and producing all four toxins to a certain degree. The widespread occurrence of A. alternata on wheat grains and its ability to synthesize various toxic metabolites may pose a serious hazard for both growers and consumers. [ABSTRACT FROM AUTHOR]

BACKGROUND Fusarium head blight (FHB) is one of the disasters that seriously harm wheat and other small grain crops. It causes spoilage and mildew of the grain leading to a significant decline in the yield and quality of the grain. This research aimed to isolate antagonistic bacteria to purify antifungal proteins. A strain was isolated from the rhizosphere of healthy wheat in a wheat field affected by a severe FHB epidemic. This isolated strain was tentatively identified as Paenibacillus polymyxa 7F1, which displayed a strong inhibitory effect against several other pathogens. One novel antifungal protein was purified from the P. polymyxa 7F1 and successfully expressed. RESULTS: A crude culture of P. polymyxa 7F1 demonstrated antifungal activity that was stable at a temperature range of 60–90 °C and a pH range of 2.6–9.0. However, the antifungal activity of the P. polymyxa 7F1 was inhibited with proteinase K, trypsin, and neutral protease treatment. A 36 kDa protein with broad‐spectrum antifungal activity was purified from the P. polymyxa 7F1. A glycosyl hydrolase domain was identified from this protein through liquid chromatography–mass spectrometry (LC–MS) analysis. A recombinant plasmid pET32a(+)/36kd for prokaryotic expression was constructed, and the renatured p36kd protein demonstrated similar antifungal activity to the 36 kDa protein purified from the P. polymyxa 7F1. CONCLUSION: A novel antifungal protein produced by P. polymyxa 7F1 was purified and expressed. The recombinant protein showed good antifungal activity as the novel purified protein. Deoxynivalenol (DON) is commonly found in wheat and wheat-derived foods, posing a threat to human health. Biodegradation is an efficient and eco-friendly measure for mycotoxin detoxification. Understanding the mechanism of DON biodegradation is hence of great importance. Herein, we report the application of metabolomics methods for the analysis of DON degradation by a bacterial consortium isolated from wheat leaves collected in Jiangsu Province. Metabolomics analysis combined with a nuclear magnetic resonance analysis revealed the main degradation product, 3-keto-DON, and a minor degradation product, 3-epi-DON. Further study illustrated that DON underwent a two-step epimerization through the intermediate 3-keto-DON. Sequencing analysis of the 16S rRNA metagenome of the microorganismal community suggested that the abundance of three bacterial genera, Achromobacter, Sphingopyxis, and Sphingomonas, substantially increased during the coculture of bacterial consortium and DON. Further investigation revealed that Devosia sp. might be responsible for the epimerization of 3-keto-DON. These findings shed light on the catabolic pathways of DON during biodegradation and illustrate the potential of using metabolomics approaches in biodegradation studies. The work describes an opto-sensor for the highly sensitive detection of the zearalenone (ZON) in wheat. A molecularly imprinted polymer layer was coated onto the surface of a luminescent metal–organic framework with cyclodo-decanyl-2,4-dihydroxybenzoate (CDHB), an analog of ZON, chosen as the template and 3-aminopropyltriethoxysilane (APTES) as the functional monomer. The obtained MOF@MIP (MMIP) was fully characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), N2 adsorption–desorption measurements, and Fourier transform infrared spectroscopy (FT-IR). Under optimal conditions, the relative fluorescence intensity of MMIP decreased linearly with a concentration of ZON ranging from 0.05–1.0 mg L−1, with a detection limit of 0.018 mg L−1 and a good recovery of 95.53–98.01% (RSD ＜ 4.7%). A colloidal gold (ICS) test was developed for rapid detection of zearalenone (ZEN) in wheat samples. The mAb against ZEN was prepared in our laboratory and labelled with colloidal gold as a probe for the ICS test. The conditions were optimized and 30 nm colloidal gold nanoparticles were chosen for optimal performance. Millipore 135 was chosen as the NC membrane for its level of sensitivity. The optimum amount of coated antigen ZEN-OVA and anti-ZEN mAb was 0.5 mg/mL and 8 μg/mL, respectively. The ICS test, which has a detection limit of 15 ng/mL for ZEN, could be completed in 5 min. Analysis of ZEN in 202 wheat samples over three consecutive years revealed that data obtained from the ICS test were in a good agreement with LC-MS/MS data. 3-isopropylmalate dehydrogenase (IPMD) encoded by LEU2 is a key enzyme in leucine (Leu) biosynthetic pathway. Analysis of the genome sequence of Fusarium graminearum revealed two paralogous LEU2 genes (designated as FgLEU2A and FgLEU2B) in this fungus and the deduced amino acid sequences of FgLeu2A and FgLeu2B share 45% identity. Targeted disruption of individual FgLEU2A/B gene in F. graminearum assigned a more crucial role of FgLeu2A in Leu biosynthesis as disruption of FgLEU2A resulted in mutant (ΔFgLeu2A-10) that was Leu-auxotrophic and could not grow in minimal medium limited for amino acids, whereas FgLEU2B deletion mutant ΔFgLeu2B-2 was morphologically indistinguishable from the wild type strain PH-1. The growth defects of ΔFgLeu2A-10 could be overcome by exogenous addition of Leu at 0.25 mM. Double deletion of FgLEU2A and FgLEU2B (ΔFgLeu2AB-8) caused a more severe Leu-auxotrophic phenotype as the concentration of Leu exogenously added to medium to rescue the growth defect of ΔFgLeu2AB-8 should be raised to 1.25 mM, indicating a less important but nonnegligible role of FgLeu2B in Leu biosynthesis. Disturb of Leu biosynthesis caused by FgLEU2A deletion leads to slower growth rate, reduced aerial hyphal formation and red pigmentation on PDA plates and completely blocked conidial production and germination. All of the defects above could be overcome by Leu addition or complementation of the full-length FgLEU2A gene. ΔFgLeu2A-10 also showed significantly increased sensitivity to osmotic and oxidative stresses. Pathogenicity assay results showed that virulence of mutants lacking FgLEU2A were dramatically impaired on wheat heads and non-host cherry tomatoes. Additionally, a low level of deoxynivalenol (DON) production of ΔFgLeu2A-10 and ΔFgLeu2AB-8 in wheat kernels was also detected. Taken together, results of this study indicated a crucial role of FgLeu2A and a less important role of FgLeu2B in Leu biosynthesis and fungal infection-related morphogenesis in F. graminearum and FgLeu2A may serve as a potential target for novel antifungal development. BACKGROUND: The Fusarium graminearum species complex infects several cereals and causes the reduction of grain yield and quality. Many factors influence the extent of Fusariuminfection andmycotoxin levels. Such factors include crop rotation. In the present study, we explored the effect of rice ormaize as former crops onmycotoxin accumulation in wheat grains. RESULTS: More than 97% of samples were contaminated with deoxynivalenol (DON). DON concentrations in wheat grains from rice and maize rotation fields were 884.37 and 235.78 μgkg-1. Zearalenone (ZEN) was detected in 45% of samples which were mainly collected from maize-wheat rotation systems. Fusarium strains were isolated and more F. graminearum sensu stricto (s. str.) isolateswere cultured fromwheat samples obtained frommaize rotationfields.DONlevels producedby Fusariumisolates from rice rotation fields were higher than those of samples from maize rotation fields. CONCLUSIONS: Rice-wheat rotation favours DON accumulation, while more ZEN contamination may occur in maize-wheat rotation models. Fusarium head blight (FHB) resistant line Soru#1 was hybridized with the German cultivar Naxos to generate 131 recombinant inbred lines for QTL mapping. The population was phenotyped for FHB and associated traits in spray inoculated experiments in El Batán (Mexico), spawn inoculated experiments in Ås (Norway) and point inoculated experiments in Nanjing (China), with two field trials at each location. Genotyping was performed with the Illumina iSelect 90K SNP wheat chip, along with a few SSR and STS markers. A major QTL for FHB after spray and spawn inoculation was detected on 2DLc, explaining 15–22% of the phenotypic variation in different experiments. This QTL remained significant after correction for days to heading (DH) and plant height (PH), while another QTL for FHB detected at the Vrn-A1 locus on 5AL almost disappeared after correction for DH and PH. Minor QTL were detected on chromosomes 2AS, 2DL, 4AL, 4DS and 5DL. In point inoculated experiments, QTL on 2DS, 3AS, 4AL and 5AL were identified in single environments. The mechanism of resistance of Soru#1 to FHB was mainly of Type I for resistance to initial infection, conditioned by the major QTL on 2DLc and minor ones that often coincided with QTL for DH, PH and anther extrusion (AE). This indicates that phenological and morphological traits and flowering biology play important roles in resistance/escape of FHB. The transgenic wheat line N12-1 containing the WYMV-Nib8 gene was obtained previously through particle bombardment, and it can effectively control the wheat yellow mosaic virus (WYMV) disease transmitted by Polymyxa graminis at turngreen stage. Due to insertion of an exogenous gene, the transcriptome of wheat may be altered and affect root exudates. Thus, it is important to investigate the potential environmental risk of transgenic wheat before commercial release because of potential undesirable ecological side effects. Our 2-year study at two different experimental locations was performed to analyze the impact of transgenic wheat N12-1 on bacterial and fungal community diversity in rhizosphere soil using polymerase chain reaction-denaturing gel gradient electrophoresis (PCR-DGGE) at four growth stages (seeding stage, turngreen stage, grain-filling stage, and maturing stage). We also explored the activities of urease, sucrase and dehydrogenase in rhizosphere soil. The results showed that there was little difference in bacterial and fungal community diversity in rhizosphere soil between N12-1 and its recipient Y158 by comparing Shannon's, Simpson's diversity index and evenness (except at one or two growth stages). Regarding enzyme activity, only one significant difference was found during the maturing stage at Xinxiang in 2011 for dehydrogenase. Significant growth stage variation was observed during 2 years at two experimental locations for both soil microbial community diversity and enzyme activity. Analysis of bands from the gel for fungal community diversity showed that the majority of fungi were uncultured. The results of this study suggested that virus-resistant transgenic wheat had no adverse impact on microbial community diversity and enzyme activity in rhizosphere soil during 2 continuous years at two different experimental locations. Wheat Fusarium head blight (FHB), caused by Fusarium species, is a widespread and destructive fungal disease. In addition to the substantial yield and revenue losses, diseased grains are often contaminated with Fusarium mycotoxins, making them unsuitable for human consumption or use as animal feed. As a vital food and feed ingredient in China, the quality and safety of wheat and its products have gained growing attention from consumers, producers, scientists, and policymakers. This review supplies detailed data about the occurrence of Fusarium toxins and related intoxications from the 1980s to the present. Despite the serious situation of toxin contamination in wheat, the concentration of toxins in flour is usually lower than that in raw materials, and food-poisoning incidents have been considerably reduced. Much work has been conducted on every phase of toxin production and wheat circulation by scientific researchers. Regulations for maximum contamination limits have been established in recent years and play a substantial role in ensuring the stability of the national economy and people's livelihoods. Fusarium crown rot of wheat is a serious fungal disease that occurs worldwide. The disease has been emerging in the major wheat-growing areas in China since 2010. Fusarium pseudogramineaum is the predominant causative pathogen of crown rot of wheat in China. The 14α-demethylation inhibitor (DMI) fungicide metconazole has been shown to be effective against Fusarium spp., but little is known about its specific activity against F. pseudogramineaum. Metconazole exhibited strong antifungal activities against all thirty-nine F. pseudogramineaum strains collected from the major wheat-growing areas in China. Metconazole inhibited mycelial growth and conidial germ tube elongation of F. pseudograminearum. Metconazole treatment significantly reduced the production of major toxins and the expression levels of toxin biosynthesis genes. Genome-wide transcriptional profiling of F. pseudograminearum in response to metconazole indicated that the expression of genes involved in ergosterol biosynthesis, including fungicide target genes (cyp51 genes), was significantly induced by metconazole. Nine ATP-binding cassette (ABC) transporter-encoding genes were significantly expressed in response to metconazole treatment. Reduced ergosterol production and antioxidant enzyme activities were observed after metconazole treatment. Greenhouse experiments indicated a significant reduction in crown rot occurrence in wheat after seed treatment with metconazole. This study evaluated the potential of metconazole to manage wheat crown rot and provides information to understand its antifungal activities and mechanism of action against F. pseudograminearum. The present study was performed to identify prevailing Fusarium species and the environmental factors affecting their frequencies and the contamination of grain with major mycotoxins in Jiangsu province. The precipitation levels were 184.2 mm, 156.4 mm, and 245.8 mm in the years 2013–2015, respectively, and the temperature fluctuated by an average of 10.6 ± 7.2 °C in 2013, 10.9 ± 7.2 °C in 2014, and 10.6 ± 6.3 °C in 2015. Co-occurrence of deoxynivalenol (DON), 3-acetyldeoxynivalenol (3ADON), and 15-acetyldeoxynivalenol (15ADON) were observed in wheat. The average concentrations of DON were 879.3 ± 1127.8, 627.8 ± 640.5, and 1628.6 ± 2,168.0 μg/kg in 2013–2015, respectively. The average concentrations of 3ADON were 43.5 ± 59.0, 71.2 ± 102.5, and 33.5 ± 111.9 μg/kg in 2013–2015, respectively. We found that the average concentration of DON in wheat was positively correlated with precipitation ( r = 0.998, p < 0.01), and 3ADON was negatively correlated with precipitation ( r = − 0.887, p < 0.05). However, there was no correlation between precipitation and 15ADON or nivalenol (NIV). The differences in temperature were not as significant as the differences in rainfall amount over a short time period. Therefore, there were no correlations between temperature and the concentrations of trichothecenes, excluding 3ADON ( r = 0.996, p < 0.01). Our data indicated that Fusarium asiaticum is the primary pathogenic fungus prevalent in the Fusarium head blight disease nursery. The trichothecene chemotype composition differed between Fusarium graminearum sensu stricto (s. str.) and F. asiaticum isolates. The 3ADON chemotype was found only among strains of F. asiaticum . The NIV chemotype was not observed among strains of F. graminearum , while the 15ADON chemotype represented 100% of the F. graminearum strains collected. The results of this study indicated no correlations between environmental conditions and the species or genetic chemotype composition of pathogens in Jiangsu province in 2013–2015. The present study aimed to evaluate the feasibility of using near-infrared hyperspectral imaging (NIR-HSI) and chemometrics for quantifying deoxynivalenol (DON) in individual wheat kernels. In total, 120 wheat kernels of severely damaged kernels, moderately damaged kernels and asymptomatic kernels (SDKs, MDKs and AKs, respectively) were collected, and the DON content in the individual wheat kernels was analyzed by HPLC-MS/MS. Partial least squares (PLS), support vector machine (SVM) and local PLS based on global PLS scores (LPLS-S) algorithms were employed for building quantification models of DON. The results showed that SDKs and MDKs might contain low or no DON, while AKs could have a high DON content. Comparing the three modeling strategies, LPLS-S using mixed spectra achieved the best performance for kernels with RMSEP of 40.25 mg/kg and RPD of 2.24, which confirmed that NIR-HSI could be a feasible method for monitoring DON in individual kernels and removing highly contaminated kernels prior to food chain entry. • The impact of the kernel orientation (crease facing down or up) on the model results was investigated. • Wheat kernels were classified according to the degree of Fusarium infection by NIR-HSI and PLS-DA. • NIR-HSI combined with chemometrics was employed to quantify DON content in individual kernels.
