Background The Antarctic krill, Euphausia superba is a pelagic crustacean, abundant in high-density swarms (10 000 - 30 000 ind/m2) with a circumpolar distribution and a key role in the food web of the Southern Ocean. Only three EST derived microsatellite markers have been used in previous genetic studies, hence we developed additional highly polymorphic microsatellite markers to allow robust studies of the genetic variability and population differentiation within this species. Findings The microsatellite markers described here were obtained through an enriched genomic library, followed by 454 pyrosequencing. A total of 10 microsatellite markers were tested in 32 individuals from the Antarctic Peninsula. One of the tested loci was fixed for one allele while the other was variable. Of the remaining nine markers, seven showed no departure from Hardy-Weinberg equilibrium. The mean number of alleles was 14.9. Conclusions These markers open perspectives for population genetic studies of this species to unravel genetic structure, dispersal and population biology, vital information for future conservation. Chromophoric dissolved organic matter (CDOM), the optically active fraction of dissolved organic matter, is primarily generated by pelagic organisms in the open ocean. In this study, we experimentally determined the quantity and spectral quality of CDOM generated by bacterioplankton using two different substrates (with and without photoproducts) and by Antarctic krill Euphausia superba and evaluated their potential contributions to CDOM dynamics in the peninsular region of the Southern Ocean. CDOM was generated by bacteria in all experiments, and the presence of photoproducts influenced both the quantity and the spectral quality of the resultant CDOM. We confirmed a direct link between bacterial production and CDOM generation, which yielded in situ CDOM duplication times from 31 to 33 d. Antarctic krill as a direct source of CDOM was also confirmed experimentally. We estimated that CDOM generation by krill would lead to CDOM duplication times from 0.48 to 0.80 d within krill swarms. Our findings highlight the potential significance of bacteria and Antarctic krill swarms in the generation of CDOM and underscore the dynamic nature of CDOM in this area. [ABSTRACT FROM AUTHOR]

A set of eight large (20 m³) mesocosms were moored in Johnson's Dock (62°39.576'S, 60°22.408'W, Livingston Island, Antarctica) to experimentally generate a gradient of phytoplankton biomass and production in order to test the extent of coupling between bacteria (heterotrophic Bacteria and Archaea) and phytoplankton, as well as the role of bacterial losses to protist grazers. This was achieved by imposing four light levels (100%, 50%, 25%, and 10%) in the presence or absence of nutrient additions (0.1 mol NH4Cl, 0.1 mol F6Na2Si, and 0.01 mol KH2PO4 per day per mesocosm). The experimental treatments resulted in a broad range of chlorophyll a (Chl a) (0.31-93.5 µg Chl a L-1) and average primary production rates, while bacteria responded in a much narrower range of biomass (3-447 µg C L-1) and production (0.21-15.71 µg C L-1d-1). Results confirm that bacteria--chlorophyll and bacterial production--primary production relationships in the Southern Ocean differ from the typical relationships applicable to aquatic ecosystems elsewhere. Bacteria respond to phytoplankton blooms, but they respond so weakly that bacterial production represents a small percentage of primary production (1-10%). Although other mechanisms might also contribute to the weak bacterial response to phytoplankton blooms, we demonstrate that the reason for it is likely the tight control of bacterial populations by their predators. Protist grazers are able to sustain faster growth rates in the cold waters of the Southern Ocean than are bacteria, thereby preventing bacteria from responding to phytoplankton blooms more forcibly. Abstract: A multipurpose analyzer system based on sequential injection analysis (SIA) for the determination of dissolved oxygen (DO) in seawater is presented. Three operation modes were established and successfully applied onboard during a research cruise in the Southern ocean: 1st, in-line execution of the entire Winkler method including precipitation of manganese (II) hydroxide, fixation of DO, precipitate dissolution by confluent acidification, and spectrophotometric quantification of the generated iodine/tri-iodide (I2/I3 −), 2nd, spectrophotometric quantification of I2/I3 − in samples prepared according the classical Winkler protocol, and 3rd, accurate batch-wise titration of I2/I3 − with thiosulfate using one syringe pump of the analyzer as automatic burette. In the first mode, the zone stacking principle was applied to achieve high dispersion of the reagent solutions in the sample zone. Spectrophotometric detection was done at the isobestic wavelength 466nm of I2/I3 −. Highly reduced consumption of reagents and sample compared to the classical Winkler protocol, linear response up to 16mgL−1 DO, and an injection frequency of 30 per hour were achieved. It is noteworthy that for the offline protocol, sample metering and quantification with a potentiometric titrator lasts in general over 5min without counting sample fixation, incubation, and glassware cleaning. The modified SIMPLEX methodology was used for the simultaneous optimization of four volumetric and two chemical variables. Vertex calculation and consequent application including in-line preparation of one reagent was carried out in real-time using the software AutoAnalysis. The analytical system featured high signal stability, robustness, and a repeatability of 3% RSD (1st mode) and 0.8% (2nd mode) during shipboard application. [Copyright &y& Elsevier]
