We isolated and characterized the community of cultivable fungi associated with marine macroalgae present in the Magellan sub-Antarctic straits and the South Shetland Islands, Maritime Antarctica, and evaluated their production of bioactive metabolites. A total of 201 filamentous fungal isolates were obtained. The genera Antarctomyces, Pseudogymnoascus, Microdochium, Trichoderma, Cladosporium, Penicillium, Neoascochyta, Entomortierella and Linnemannia were associated with Antarctic macroalgae, with Neoascochyta paspali being the most abundant taxon. In contrast, 12 taxa representing Cadophora, Microdochium, Penicillium, Pseudogymnoascus were associated with macroalgae from the Magellan sub-Antarctic, with Penicillium dominating the assemblages. The diversity indices of the fungal communities associated with macroalgae in the two regions were similar. Among 177 fungal extracts assessed for metabolite production, 31 (17.5%) showed strong phytotoxic activity and 17 (9.6%) showed anti-Trypanosoma cruzi activity. Penicillium showed the highest phytotoxic and anti-Trypanosoma activity values. The detection of taxa in common between the polar and cold temperate zones reinforces the need for further investigations of the distribution of species in these distinct ecoregions. The detection of bioactive extracts produced particularly by Penicillium representatives reinforces the potential to obtain active molecules that can be explored as natural products or as sources of bioactive compounds with application in agriculture and biomedicine., (© 2024. Gram-positive, aerobic, motile, rod-shaped, mesophilic epiphytic bacterium Planomicrobium okeanokoites was isolated from the surface of endemic species Himantothallus grandifolius in Larsemann Hills, Eastern Antarctica. The diversity of epiphytic bacterial communities living on marine algae remains primarily unexplored; virtually no reports from Antarctic seaweeds. The present study used morpho-molecular approaches for the macroalgae and epiphytic bacterium characterization. Phylogenetic analysis was performed using mitochondrial genome encoded COX1 gene; chloroplast genome encodes rbcL; nuclear genome encoded large subunit ribosomal RNA gene (LSU rRNA) for Himantothallus grandifolius and ribosomal encoded 16S rRNA for Planomicrobium okeanokoites. Morphological and molecular data revealed that the isolate is identified as Himantothallus grandifolius, which belongs to Family Desmarestiaceae of Order Desmarestiales in Class Phaeophyceae showing 99.8% similarity to the sequences of Himantothallus grandifolius, from King George Island, Antarctica (HE866853). The isolated bacterial strain was identified on the basis of chemotaxonomic, morpho-phylogenetic, and biochemical assays. A phylogenetic study based on 16S rRNA gene sequences revealed that the epiphytic bacterial strain SLA-357 was closest related to the Planomicrobium okeanokoites showing 98.7% sequence similarity. The study revealed the first report of this species from the Southern Hemisphere to date. Also, there has been no report regarding the association between the Planomicrobium okeanokoites and Himantothallus grandifolius; however, there are some reports on this bacterium isolated from sediments, soils, and lakes from Northern Hemisphere. A Gram-stain-negative, pink-pigmented, rod-shaped, non-flagellated, aerobic bacterium, designated strain SM1701 T , was isolated from a rotten seaweed collected off Fildes Peninsula, King George Island, West Antarctica. The strain grew at 4-30 °C, pH 6.0-8.0 and with 0.5-5 % (w/v) NaCl. It hydrolysed gelatin and Tweens (40, 60 and 80), but did not reduce nitrates to nitrites. The major cellular fatty acids of strain SM1701 T were iso-C 15 : 0 , iso-C 15 : 1 G, iso-C 16 : 1 G, C 16 : 0 and iso-C 17 : 0 3-OH. Polar lipids included phosphatidylethanolamine, one unidentified aminolipid, two unidentified glycolipids and one unidentified aminoglycolipid. The major respiratory quinone was MK-7. The genomic DNA G+C content of strain SM1701 T was 34.1 mol%. It showed high 16S rRNA gene sequence similarities to Crocinitomix algicola (93.8 %) and Crocinitomix catalasitica (92.5 %) and less than 91 % sequence similarities to other known members in the family Crocinitomicaceae . Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain SM1701 T constituted a distinct lineage within the family Crocinitomicaceae . The phylogenetic trees based on concatenated 261 protein sequences from genome sequences showed that strain SM1701 T occupied a branch separated from those of known genera in the family of Crocinitomicaceae , indicating it may belong to a new genus. On the basis of the polyphasic characterization of strain SM1701 T in this study, it is considered to represent a novel species in a new genus in the family Crocinitomicaceae , for which the name Putridiphycobacter roseus gen. nov., sp. nov. is proposed. The type strain is SM1701 T (=KCTC 62302 T =NBRC 113201 T =CGMCC 1.16510 T ).

The establishment of diatom communities depends on environmental factors such as the type of substrate and geographic conditions that influence the dispersal processes of these organisms. The main goal of this study was to evaluate the similarity between diatom communities associated with the macroalgae Prasiola crispa (Lightfoot) Kützing in relation to spatial distance from six sampled sites located in the South Shetland Islands, Maritime Antarctica. The diatom flora associated with Prasiola crispa was represented by 23 species distributed in 15 genera. Pinnularia australoschoenfelderi Zidarova, Kopalová & Van de Vijver, Luticola austroatlantica Van de Vijver, Kopalová, S.A.Spaulding & Esposito, Luticola amoena Van der Vijver, Kopalová, Zidarova & Levkov, Pinnularia austroshetlandica (Carlson) Cleve-Euler and Psammothidium papilio (D.E. Kellogg et al.) Kopalová & Zidarova were the most abundant species in our samples, together they represented 68% of the total number of individuals collected. There was great similarity and abundance of the diatom communites among the sampled points, which resulted in the absence of a linear relationship pattern with distance between sampling points. We conclude that distance was not a factor of differentiation of Antarctic diatom communities associated with terrestrial green macroalgae. Little is known about the diversity and roles of Gram-positive and pigmented bacteria in Antarctic environments, especially those associated with marine macroorganisms. This work is the first study about the diversity and antimicrobial activity of culturable pigmented Gram-positive bacteria associated with marine Antarctic macroalgae. A total of 31 pigmented Gram-positive strains were isolated from the surface of six species of macroalgae collected in the King George Island, South Shetland Islands. On the basis of 16S rRNA gene sequence similarities ≥99%, 18 phylotypes were defined, which were clustered into 11 genera of Actinobacteria (Agrococcus, Arthrobacter, Brachybacterium, Citricoccus, Kocuria, Labedella, Microbacterium, Micrococcus, Rhodococcus, Salinibacterium and Sanguibacter) and one genus of the Firmicutes (Staphylococcus). It was found that five isolates displayed antimicrobial activity against a set of macroalgae-associated bacteria. The active isolates were phylogenetically related to Agrococcus baldri, Brachybacterium rhamnosum, Citricoccus zhacaiensis and Kocuria palustris. The results indicate that a diverse community of pigmented Gram-positive bacteria is associated with Antartic macroalgae and suggest its potential as a promising source of antimicrobial and pigmented natural compounds., (© FEMS 2015.
