Cold, dry, and nutrient-poor, the McMurdo Dry Valleys of Antarctica are among the most extreme terrestrial environments on Earth. Numerous studies have described microbial communities of low elevation soils and streams below glaciers, while less is known about microbial communities in higher elevation soils above glaciers. We characterized microbial life in four landscape features (habitats) of a mountain in Taylor Valley. These habitats varied significantly in soil moisture and include moist soils of a (1) lateral glacial moraine, (2) gully that terminates at the moraine, and very dry soils on (3) a southeastern slope and (4) dry sites near the gully. Using rRNA gene PCR amplicon sequencing of Bacteria and Archaea (16S SSU) and eukaryotes (18S SSU), we found that all habitat types harbored significantly different bacterial and eukaryotic communities and that these differences were most apparent when comparing habitats that had macroscopically visible soil crusts (gully and moraine) to habitats with no visible crusts (near gully and slope). These differences were driven by a relative predominance of Actinobacteria and a Colpodella sp. in non-crust habitats, and by phototrophic bacteria and eukaryotes (e.g., a moss) and predators (e.g., tardigrades) in habitats with biological soil crusts (gully and moraine). The gully and moraine also had significantly higher 16S and 18S ESV richness than the other two habitat types. We further found that many of the phototrophic bacteria and eukaryotes of the gully and moraine share high sequence identity with phototrophs from moist and wet areas elsewhere in the Dry Valleys and other cold desert ecosystems. These include a Moss (Bryum sp.), several algae (e.g., a Chlorococcum sp.) and cyanobacteria (e.g., Nostoc and Phormidium spp.). Overall, the results reported here broaden the diversity of habitat types that have been studied in the Dry Valleys of Antarctica and suggest future avenues of research to more definitively understand the biogeography and factors controlling microbial diversity in this unique ecosystem. Cyanobacteria are generally thought to be responsible for primary production and nitrogen fixation in the microbial communities that dominate Antarctic ecosystems. Recent studies of bacterial communities in terrestrial Antarctica, however, have shown that Cyanobacteria are sometimes only scarcely present, suggesting that other bacteria presumably take over their role as primary producers and diazotrophs. The diversity of key genes in these processes was studied in surface samples from the Sør Rondane Mountains, Dronning Maud Land, using clone libraries of the large subunit of ribulose-1,5-biphosphate carboxylase/oxygenase (RuBisCO) genes ( cbbL, cbbM) and dinitrogenase-reductase ( nifH) genes. We recovered a large diversity of non-cyanobacterial cbbL type IC in addition to cyanobacterial type IB, suggesting that non-cyanobacterial autotrophs may contribute to primary production. The nifH diversity recovered was predominantly related to Cyanobacteria, particularly members of the Nostocales. We also investigated the occurrence of proteorhodopsin and anoxygenic phototrophy as mechanisms for non-Cyanobacteria to exploit solar energy. While proteorhodopsin genes were not detected, a large diversity of genes coding for the light and medium subunits of the type 2 phototrophic reaction center ( pufLM) was observed, suggesting for the first time, that the aerobic photoheterotrophic lifestyle may be important in oligotrophic high-altitude ice-free terrestrial Antarctic habitats. According to paleontological data, large-scale, global glaciations occurred repeatedly over the history of Earth. Such glaciations unavoidably resulted in a sharp decrease in the number of habitats suitable for development of phototrophic microorganisms, which mostly require sunlight. Molecular data indicate that some groups of Antarctic phototrophic microorganisms emerged before formation of the Antarctic ice sheet (30-45 Ma) and probably survived the subsequent glaciation maxima. Analysis of the data on the present-day occurrence of phototrophs in Antarctica revealed the presence of liquid water in a habitat to be the key factor for survival of phototrophic communities. At negative air temperatures, this requirement may be met either in warmer microniches (0 to 30°C and higher) or in brines with freezing temperatures below zero (to −52°C). Development of phototrophic communities in such habitats may indicate the theoretical possibility of existence of the niches favoring growth of phototrophic microorganisms during Precambrian global glaciations. Non-photochemical fluorescence quenching (NPQ) is mainly associated with the transthylakoid proton gradient (ΔpH) and xanthophyll cycle. However, the exact mechanism of NPQ is different in different oxygenic photosynthetic organisms. In this study, several inhibitors were used to study NPQ kinetics in the sea ice alga Chlamydomonas sp. ICE-L and to determine the functions of ΔpH and the xanthophyll cycle in the NPQ process. NHCl and nigericin, uncouplers of ΔpH, inhibited NPQ completely and zeaxanthin (Z) was not detected in 1 mM NHCl-treated samples. Moreover, Z and NPQ were increased in the samples containing N,N'-dicyclohexyl-carbodiimide (DCCD) under low light conditions. We conclude that ΔpH plays a major role in NPQ, and activation of the xanthophyll cycle is related to ΔpH. In dithiothreitol (DTT)-treated samples, no Z was observed and NPQ decreased. NPQ was completely inhibited when NHCl was added suggesting that part of the NPQ process is related to the xanthophyll cycle and the remainder depends on ΔpH. Moreover, lutein and β-carotene were also essential for NPQ. These results indicate that NPQ in the sea ice alga Chlamydomonas sp. ICE-L is mainly dependent on ΔpH which affects the protonation of PSII proteins and de-epoxidation of the xanthophyll cycle, and the transthylakoid proton gradient alone can induce NPQ. Aquatic mosses in the genera Bryum and Leptobryum form unique tower-like 'moss pillars' underwater in some Antarctic lakes, in association with algae and cyanobacteria. These are communities with a two-layer structure comprising an oxidative exterior and reductive interior. Although habitats and photosynthetic properties of moss pillars have been reported, microfloral composition of the two-layer structure has not been described. Here we report fatty acid analysis of one moss pillar and molecular phylogenetic analysis, based on the 16S rRNA gene, of this and one other moss pillar. Cluster analysis of the phospholipid fatty acid composition showed three groups corresponding to the exterior, upper interior, and lower interior of the pillar. This suggested that species composition differed by section, with the exterior dominated by photosynthetic organisms such as mosses, algae, and cyanobacteria, the upper interior primarily containing gram-positive bacteria and anaerobic sulfate-reducing bacteria, and the lower interior dominated by gram-negative bacteria. Molecular phylogenetic analysis revealed that Proteobacteria dominate the moss pillar as a whole; cyanobacteria were found on the exterior and the gram-positive obligate anaerobe Clostridium in the interior, while gram-positive sulfate-reducing bacteria were present in the lowest part of the interior. Nitrogen-fixing bacteria and denitrifying bacteria were found in all sections. Thus, fatty acid analysis and genetic analysis showed similar patterns. These findings suggest that microorganisms of different phylogenetic groups inhabit different sections of a single moss pillar and form a microbial community that performs biogeochemical cycling to establish and maintain a structure in an oxidation-reduction gradient between exterior and interior. High-elevation valleys in dry areas of the Himalayas are among the most extreme, yet least explored environments on Earth. These barren, rocky valleys are subjected to year-round temperature fluctuations across the freezing point and very low availability of water and nutrients, causing previous workers to hypothesize that no photoautotrophic life (primary producers) exists in these locations. However, there has been no work using modern biogeochemical or culture-independent molecular methods to test the hypothesis that photoautotrophs are absent from high Himalayan soil systems. Here, we show that although microbial biomass levels are as low as those of the Dry Valleys of Antarctica, there are abundant microbial photoautotrophs, displaying unexpected phylogenetic diversity, in barren soils from just below the permanent ice line of the central Himalayas. Furthermore, we discovered that one of the dominant algal clades from the high Himalayas also contains the dominant algae in culture-independent surveys of both soil and ice samples from the Dry Valleys of Antarctica, revealing an unexpected link between these environmentally similar but geographically very distant systems. Phylogenetic and biogeographic analyses demonstrated that although this algal clade is globally distributed to other high-altitude and high-latitude soils, it shows significant genetic isolation by geographical distance patterns, indicating local adaptation and perhaps speciation in each region. Our results are the first to demonstrate the remarkable similarities of microbial life of arid soils of Antarctica and the high Himalayas. Our findings are a starting point for future comparative studies of the dry valleys of the Himalayas and Antarctica that will yield new insights into the cold and dry limits to life on Earth. Photosynthetic dinoflagellates contain a diverse collection of plastid types, a situation believed to have arisen from multiple endosymbiotic events. In addition, a number of heterotrophic (phagotrophic) dinoflagellates possess the ability to acquire chloroplasts temporarily by engulfing algae and retaining their chloroplasts in a functional state. These latter relationships typically last from a few days to weeks, at which point the chloroplasts lose function, are digested and replaced with newly acquired plastids. A novel and abundant dinoflagellate related to the icthyotoxic genera Karenia and Karlodinium was recently discovered by us in the Ross Sea, Antarctica. Sequencing of its plastid small subunit ribosomal gene indicated that it did not share evolutionary history with the plastids of Karenia or Karlodinium, but was closely related to the free-living haptophyte Phaeocystis antarctica, a species that often dominates phytoplankton blooms in the Ross Sea. Chloroplast uptake was observed to occur rapidly (within 2 days), with retention in cultures being long-lived (several months) but not permanent. The dinoflagellate was also incapable of growing indefinitely in continuous darkness with algae as prey. Our findings may indicate an emerging endosymbiotic event yielding a dinoflagellate that is presently neither purely phototrophic nor purely heterotrophic, but occupies a niche juxtaposed between these contrasting nutritional modes. A sediment core from Progress Lake, one of the oldest lacustrine sequences in East Antarctica, contains distinct zones dating from a previous interglacial (most likely Marine Isotope Stage 5e, c. 125-115 kyr BP) and the present interglacial (Marine Isotope Stage I), separated by a transition zone representing when the lake became sub-glacial. Profiles of fossil pigments, determined using high performance liquid chromatography and liquid chromatography-tandem mass spectrometry, show distinct differences in the photoautotrophic community during these two interglacial periods. The first was dominated by algae and purple phototrophic bacteria, with periods of photic zone euxinia indicated by pigments from anoxygenic phototrophic bacteria. Specific chlorophyll a derivatives reveal periods when grazing pressure impacted significantly on the phytoplankton community. The virtual absence of pigments in the transition zone reflects severe restriction of photoautotrophic activity, consistent with the lake having become sub-glacial. Retreat of snow and ice in the late Holocene (3345 14C yr BP) allowed establishment of a less diverse primary producer community, restricted to algae and cyanobacteria. Grazers were severely restricted and oxidative transformation was more important than during the previous interglacial. The pigment data provide a unique and detailed insight in to the evolution of the lake ecology over an interglacial-glacial-interglacial transition and strong evidence that the Marine Isotope Stage 5e interglacial in this region of coastal East Antarctica was several degrees warmer than at present. At Cape Evans on Ross Island, Antarctica, the rhodophyte Phyllophora antarctica is the dominant primary producer in terms of biomass from 10 to >30 m depth. The vast majority of Phyllophora occurs as accumulations of unattached plants. Whilst decomposition and incorporation of macroalgal drift material into the food web is rapid in temperate ecosystems, we predicted these processes to be slow in Antarctica. We address the functional role of macroalgal detritus in fuelling the biodiversity of benthic communities at Cape Evans during the summers of 2001 and 2002. Specifically we (a) describe the distribution and biomass of attached and drift algae, (b) assess the photosynthetic capacity and degradation of drift accumulations using in situ fluorometry, (c) assess the effect of patches of drift Phyllophora on underlying macrofaunal communities, and, (d) use stable isotopes to investigate the possible uptake of Phyllophora by macrofauna. We found drift Phyllophora accumulations throughout the depth range investigated (3–31 m), with peak biomasses of 140±30 g dwt m-2 in the 15–25 m depth strata. At this depth stratum Phyllophora was a conspicuous habitat element with the % cover on the seafloor averaging 30%. While initially the drift algal accumulations appeared in good health we measured significant declines in photosynthetic capacity between years suggesting ongoing, albeit slow, degradation of the drift algal accumulations. Our results demonstrate that Phyllophora drift accumulations have a structuring role on soft-sediment communities, which increases in strength with the gradual degradation of the algae. The longevity of Phyllophora is enhanced by secondary metabolites, which serve as protection against grazers, and their extreme shade adaptation. However, our carbon and nitrogen stable isotope data of polychaetes and amphipods associated with Phyllophora suggest that macroalgal detritus enters the food web, and although this process is slow, Phyllophora accumulations might serve to dampen the seasonality in food supply providing higher trophic levels with a more constant food source. Although anoxygenic photosynthesis is thought to play an important role in the primary productivity of permanently frozen lakes in the Antarctic dry valleys, the bacterial communities responsible for this metabolism remain uncharacterized. Here we report the composition and activity of phototrophic purple bacteria in Lake Fryxell, Antarctica, as determined by analysis of a photosynthesis-specific gene, pufM. The results revealed an extensive diversity and highly stratified distribution of purple nonsulfur bacteria in Lake Fryxell and showed which phylotypes produced pufM transcripts in situ. Enrichment cultures for purple bacteria yielded two morphotypes, each with a pufM signature identical to signatures detected by environmental screening. The isolates also contained gas vesicles, buoyancy structures previously unknown in purple nonsulfur bacteria, that may be necessary for these organisms to position themselves at specific depths within the nearly freezing water column. Oscillatorian cyanobacteria dominate benthic microbial mat communities in many polar freshwater ecosystems. Capable of growth at low temperatures, all benthic polar oscillatorians characterized to date are psychrotolerant (growth optima > 15° C) as opposed to psychrophilic (growth optima ≤ 15° C). Here, psychrophilic oscillatorians isolated from meltwater ponds on Antarctica's McMurdo Ice Shelf are described. Growth and photosynthetic rates were investigated at multiple temperatures, and compared with those of a psychrotolerant isolate from the same region. Two isolates showed a growth maximum at 8° C, with rates of 0.12 and 0.08 doublings·d-1, respectively. Neither displayed detectable growth at 24° C. The psychrotolerant isolate showed almost imperceptible growth at 4° C and a rate of 0.9 doublings·d-1 at its optimal temperature of ∼23° C. In both photosynthesis versus irradiance and photosynthesis versus temperature experiments, exponentially growing cultures were acclimated for 14 days at 3, 8, 12, 20, and 24° C under saturating light intensity, and [14C] photoincorporation rates were measured. Psychrophilic isolates acclimated at 8° C showed greatest photosynthetic rates; those acclimated at 3° C were capable of active photosynthesis, but photoincorporation was not detected in cells acclimated at 20 and 24° C, because these isolates were not viable after 14 days at those temperatures. The psychrotolerant isolate, conversely, displayed maximum photosynthetic rates at 24° C, though photoincorporation was actively occurring at 3° C. Within acclimation temperature treatments, short-term photosynthetic rates increased with increasing incubation temperature for both psychrophilic and psychrotolerant isolates. The article discusses living organisms and their food webs that exist in the coldest and most remote polar lakes on Earth. Free water on or under ice sheets or glaciers contains numerous species, most of which are microorganisms that exist in ecosystems thought to be indicative of climate change. Polar lakes in the Antarctic are populated by life forms such as cryptophytes, which are photosynthetic flagellates, and mixotrophic ciliates such as Mesodinium rubrum. The author also examines viral life in these lakes and discusses ways in which glaciers facilitate the existence of mini-lakes called cryoconites.
