A mycological survey was carried out on 115 samples of whole dried black pepper seeds, from two main production regions of Brazil (Pará and Espírito Santo). A high incidence of contamination was verified in both regions when 99.1% of the samples showed filamentous fungi contamination. A total of 497 species of nine different genera were isolated (Aspergillus, Eurotium, Rhizopus, Penicillium, Curvularia, Cladosporium, Absidia, Emericella and Paecilomyces). The genus Aspergillus was the predominant (53.5%) followed by species from the Eurotium genus (24.5%). Eurotium chevalieri (16.4%) was the most predominant species followed by A. flavus (14.6%) present on 55 samples of black pepper (47.8%) analysed. Twenty-five samples (21.7%) were contaminated with aflatoxigenic strains of A. flavus and A. parasiticus . In relation to the types of aflatoxins produced by mycotoxigenic strains, it was observed that 25 strains (44.6%) of 56 isolated of A. flavus produced aflatoxins. From 12 samples, A. ochraceus species were isolated in low frequency (3.5%). Two strains of A. ochraceus from 16 isolated were producers of ochratoxin A. With respect to the aflatoxins and ochratoxin A natural contamination, none of the samples presented detectable levels of these mycotoxins using thin-layer chromatographic analysis. Abstract: In Brazil, commercial feedstuffs are an important component in modern animal husbandry, but there is no information available about fungal contamination and ochratoxin A (OTA) production. The aims of this study were to determine the mycoflora incidence in poultry feeds and evaluate OTA production. In addition, the ability to produce OTA by several Aspergillus and Penicillium species was investigated. A total of 96 samples of poultry feeds were collected from four factories in Rio de Janeiro. Samples were examined for total moulds, for Aspergillus and Penicillium spp. occurrence and for their relative densities on dichloran rose bengal chloramphenicol and dichloran 18% glycerol media. The capacity to produce ochratoxin A by selected Aspergillus and Penicillium species was determined by HPLC. Total mould counts were generally higher than 1×105 CFUml−1. Aspergillus and Penicillium species were isolated in the highest numbers. Aspergillus flovus and Penicillium citrinum were the most prevalent species. There was a high percentage of potential OTA producers (46%). The amount of OTA produced on this substrate was enough to cause adverse effects in animals. Several strains isolated from poultry feeds were able to produce high levels of OTA on chloramphenicol yeast medium. OTA in raw materials needs to be surveyed and storage practices must be investigated to determine occurrence and establish the livestock toxicological risk in poultry feed. [Copyright &y& Elsevier]

This study dealt with the influence of the temperature on the bacterial dynamics of two spontaneously fermented wheat sourdoughs, propagated at 21 ± 1 °C (SD1) and 30 ± 1 °C (SD2), during nine backslopping steps (BS1 to BS9). Proteobacteria was the only phylum found in flour. Escherichia hermannii was predominant, followed by Kosakonia cowanii, besides species belonging to the genera Pantoea and Pseudomonas. After one step of propagation, Clostridium and Bacillus cereus group became predominant. Lactobacillus curvatus was found at low relative abundance. For the second backslopping step , Clostridium was flanked by L. curvatus and Lactobacillus farciminis. From BS4 (6th day) onward, lactic acid bacteria (LAB) became predominant. L. farciminis overcame L. curvatus and remained dominant until the end of propagations for both sourdoughs. At 21 °C, Bacillus , Clostridium , Pseudomonas, and Enterobacteriaceae were gradually inhibited. At the end of propagation, SD1 harbored only LAB. Otherwise, the temperature of 30 °C favored the persistence of atypical bacteria in SD2, as Pseudomonas and Enterobacteriaceae. Therefore, the temperature of 21 °C was more suitable for sourdough propagation in Brazil. This study enhanced the knowledge of temperature's influence on microbial assembly and contributed to the elucidation of sourdough microbial communities in Brazil. • Spontaneously sourdoughs were propagated at 21 ± 1 °C and 30 ± 1 °C. • Lactic acid bacteria became dominant after the fourth step of propagation. • Lactobacillus farciminis was predominant in sourdoughs. • Temperature variation changed the subdominant microbial communities. • Propagation at 30 °C favored the persistence of enterobacteria in sourdough. A survey for the detection of enterotoxigenic Escherichia coli (ETEC), rotavirus and enterotoxigenic Clostridium perfringens in diarrheic stools of children up to 2 years old was carried out in the region of Campinas, SP, Brazil. Twenty-seven (20.45%) faecal specimens were positive for ETEC. From these samples 41 strains of ETEC were isolated from which 40 produced only thermolabile (LT) enterotoxin, as detected by a modified radial immune haemolysis test. Among the 183 faecal specimens examined for the detection of rotavirus, 29 (15.84%) were positive when examined by polyacrilamide gel electrophoresis (PAGE) and immunoenzymatic assay (EIA) being 15 (51.7%), derived from stools collected from winter months. All strains of rotavirus belonged to group A and through the PAGE technique, it was observed that the most frequent (9 strains) electrophoretype, according to the adopted classification, was Ib, IIc, IIIb, IVa. Only 113 fecal specimens were examined for the presence of enterotoxigenic C. perfringens. For the detection of enterotoxin in culture supernatants the reverse passive haemagglutination and intravenous inoculation of mice were used. Twelve (10.61%) enterotoxigenic C. perfringens strains were found. Taking into consideration these findings the authors call the attention of the relative value of conventional coprocultures for diagnostic purposes, pointing out the important of establishing simplified methods which would render easier, the detection and identification of the groups of enteropathogenic agents studied in this research.

Viruses similar to the bisegmented double-stranded (ds) RNA picobirnaviruses described in human faeces and the intestinal contents of Oryzomys nigripes rats and guinea pigs were isolated from the faeces of pigs taken from several areas in the state of Sao Paulo, Brazil. Samples were collected from 912 pigs of several breeds, aged nine to 61 days, and assayed by polyacrylamide gel electrophoresis with silver staining and a combined enzyme immunoassay for rotavirus and adenovirus, using the simian rotavirus SA11 as control. Electrophoretic profiles resembling the bisegmented dsRNA viruses were detected in 106 pigs with 15.3 per cent occurring in animals with diarrhoea compared to 9.6 per cent in animals without diarrhoea.

Rotaviruses were detected by polyacrylamide gel electrophoresis (PAGE) in 11 (84.6%) of 13 faecal specimens from neonatal piglets with acute diarrhoea in a piggery near the city of Campinas, State of São Paulo, Brazil. An immunoenzimatic assay for group A rotavirus (IEA-A) was positive in ten of the samples, all of which showed a PAGE profile typical of that group. Another sample was showed a group B profile in PAGE. An immunoenzimatic assay specific for group B (IEA-B) for this faecal sample was positive, confirming the PAGE results.
