The aim of this study was to determine ecotoxicological parameters for biomonitoring of environmental risk of native soils from a ceramic industrial area that had been contaminated with cadmium (Cd) and chromium (Cr) by using the earthworm, Eisenia fetida. Initially, lab tests were conducted to compare earthworm (Eisenia fetida) growth, survival, morphology, behavior, and reproduction rates following exposure to six concentrations of contaminated soil at 0%, 6.25%, 12.5%, 25%, 50%, or 100% mixed in artificial soil and cow dung following a 28-d incubation period. The second experiment consisted of utilizing Eisenia fetida in a predetermined lowest observed effect concentration to measure heavy metals bioaccumulation from superficial soil collected from a ceramic industrial area following a 56-d exposure. Data demonstrated that in the lab earthworms maintained at 6.25% of contaminated soil, exhibited significant increase in mean weight, bioaccumulation of Cd and Cr associated with a significant decrease in the amount of Cd and Cr in the soil. At field testing, similar results that were observed as in the lab as evidenced by rise in mean weight, higher levels of Cd and Cr in the earthworm tissue accompanied by significant fall in soil levels of Cd and Cr. In conclusion, at tested relevant environmental concentrations, the use of Eisenia fetida for assessing ecotoxicological risk arising from contaminated soil due to ceramic industrial pollutant emissions was found to be an effective tool for biomonitoring program.

Sugarcane is cultivated in tropical countries for sugar and ethanol production. In Brazil, this culture is among the most profitable with a production of 658.7 million tons/harvest. Sugarcane filter cake (SCFC) is a waste rich in organic matter and micronutrients, but also contains toxic metals. As it has been used as fertilizer and there is not enough knowledge about its environmental impacts, this work assessed the genotoxicogenetic effects of raw SCFC and associations with soil and sugarcane bagasse (SCB), by Allium cepa tests. Six associations of SCFC + soil and five associations of SCFC + soil + SCB were tested at three moments of degradation: initial (T0), 3 and 6 months (T1 and T2). Genotoxicogenetic assays were performed with solid substrates of these associations and with their respective aqueous extracts. Chemical analysis showed a decrease in metals, total organic carbon and nitrogen after 6 months of biodegradation, complying with Brazilian laws. In general, the combination of SCFC + soil + SCB was better than using only SCFC. T0 solubilized samples of different associations composed by highest quantities of SCFC inhibited the MI and induced CA without presenting mutagenicity (except for 75%-SCFC + soil + SCB). T1 samples showed more cytotoxicity than T0 samples, and also presented genotoxic and mutagenic effects. Solid substrate and solubilized associations of SCFC + soil + SCB of T2 samples had no toxicity. These results suggest 6 months of biodegradation and the SCB adding as effective to reduce toxicogenetic effects induced by SCFC. The rivers from the region of Catalão, Southeast Goiás State, Brazil, are exposed to intense anthropogenic influences including agricultural activities, industry, and urban waste. The aim of this study was to determine the severity of water pollution by conducting an experiment involving in situ biomonitoring of water constituents on genotoxicity in fish inhabiting these sites. The genotoxicity of three sites of the region were analyzed utilizing the micronucleus (MN) test. It was of interest to determine whether there were differences between sampling sites such as urban perimeter, agriculture, and fertilizer industry in control, and monitored species including Astyanax fasciatus, Astyanax altiparanae, and Characidium fasciatum. Data demonstrated that the species at sites 1, 2, and 3 exhibited a marked increase in frequency of MN compared to fish from site 4. Significant elevation in frequency of MN occurred in erythrocytes of A. fasciatus and A. altiparanae at sites 1 and 2. At site 3 higher frequencies of MN were observed in C. fasciatum. MN induction in C. fasciatum was correlated with chromium levels in water and sediment, while A. fasciatum and A. altiparanae showed an association with zinc in water and sediment. Data suggest that benthic and nektonic fish species display different sensitivities in relation to anthropogenic contaminant influences.

The micronucleus (MN) test and analysis of heavy metal biological accumulation in Tradescantia pallida (T. pallida) were bioassays used to assess the genotoxic potential of emissions from a complex of ceramic industries into the atmosphere in a city in Brazil that is considered a national reference source for roof tile production. The ceramic industry emission-exposed T. pallida plants were biomonitored during the dry season, in June, July, and August 2013. In addition to the contaminated monitoring site, a reference site in a peri-urban area was utilized, for comparative purposes. Genotoxicity assessments were determined monthly, while heavy metal bioaccumulation was measured at the end of the total exposure period. The MN frequency was significantly greater in T. pallida plants exposed in the ceramic industry emission monitored area compared to the reference site, and highest MN rates were observed in July and August. With respect to heavy metal bioaccumulation in T. pallida leaves, cadmium (Cd) and chromium (Cr) concentrations were significantly higher in plants at the ceramic industry emission monitoring site. Thus, in relation to the parameters assessed, T. pallida was found to be sensitive to atmospheric contamination by heavy metals attributed to ceramic products emissions generated by the ceramic industry, confirming that this plant species may be employed as a reference organism in biomonitoring studies.

The potential impact of drill cuttings on the two deep water calcareous red algae Mesophyllum engelhartii and Lithothamnion sp. from the Peregrino oil field was assessed. Dispersion modelling of drill cuttings was performed for a two year period using measured oceanographic and discharge data with 24 h resolution. The model was also used to assess the impact on the two algae species using four species specific impact categories: No, minor, medium and severe impact. The corresponding intervals for photosynthetic efficiency (ΦPSIImax) and sediment coverage were obtained from exposure-response relationship for photosynthetic efficiency as function of sediment coverage for the two algae species. The temporal resolution enabled more accurate model predictions as short-term changes in discharges and environmental conditions could be detected. The assessment shows that there is a patchy risk for severe impact on the calcareous algae stretching across the transitional zone and into the calcareous algae bed at Peregrino., (Copyright © 2015 Elsevier Ltd. The problems that most affect the quality of the waters of rivers and lakes are associated with the discharges performed in these environments, mainly industrial and domestic effluents inappropriately treated or untreated. The comet assay is a sensitive tool and is recommended for studies of environmental biomonitoring, which aim to determine the genotoxicity potential of water pollutants. This study aimed to assess the genotoxic potential of the Ribeirão Tatu waters, region of Limeira, São Paulo (SP), by the comet assay with mammalian cells (hepatoma tissue culture (HTC)). Water samples were collected along the Ribeirão Tatu at three distinct periods: November 2008, February 2009 and August 2009, and five collection sites were established: P1, source of the stream; P2, site located downstream the urban perimeter of the municipality of Cordeirópolis and after receiving the pollution load of this city; P3, collection site located upstream the urban perimeter of the city of Limeira; P4, urban area of Limeira; and P5, rural area of Limeira, downstream the discharges of the city sewage. The results showed that for the November 2008 collection, there was no water sample-induced genotoxicity; for the February 2009 collection, the sites P1 and P2 were statistically significant in relation to the negative control (NC), and for the August 2009 collection, the site P5 was statistically significant. These results could be explained by the content of different metals during the different seasons that are under the influence of domestic, industrial and agricultural effluents and also due to the seasonality, since the water samples collected in the period of heavy rain (February 2009) presented a higher genotoxicity possibly due to the entrainment of contaminants into the bed of the stream promoted by the outflow of rainwaters. The comet assay showed to be a useful and sensitive tool in the evaluation of hydric resources impacted by pollutants of diverse origins, and a constant monitoring should be done in order to verify the influence of different factors (season, amount of contaminants) in the water quality.

In Brazil, only 20.2% of the municipalities have sewage collection and treatment. The use of medical diagnostic by radiographic processing generates effluents which may contain contaminants such as silver (Ag) that causes irreversible damage to health. They can also contaminate soil, water, and food if there is no treatment before disposal. This study aimed to identify and quantify the Ag concentration in radiographic film-washing water and fixer generated from radiographic processing in 12 Brazilian health institutions, in order to offer subsidies to the environmental and public health managers about this issue. The Ag values found in the washing water films samples ranged from 0.1 to 1,785.97 mg L(-1). In the fixer samples, the values ranged from 435.59 to 16,325.92 mg L(-1). These data are far above the values required by Brazilian legislation whose limit is 0.1 mg L(-1) for Ag in effluents released directly or indirectly into the environment. This research reveals that the disposal of Ag in this kind of effluent needs to be monitored and controlled by the Brazilian government in order to protect both the human and environment health from those contaminants.

Wastewater control at storage terminals of liquid chemical products in bulk is very difficult because of the variety of products handled in the facilities generating effluents of variable composition. The main objective of this work was to verify if the Vibrio fischeri acute toxicity test could be routinely included in the wastewater management of those facilities along with physical and chemical analysis in order to evaluate and improve the quality of the generated effluents. The study was performed in two phases before and after the implementation of better operational practices/treatment technologies. Chemical oxygen demand (COD) and toxicity of treated effluents did not correlate showing that effluents with low COD contain toxic substances and non-biodegradable organic matter, which may be not degraded when discharged into the aquatic environment. Segregation of influents or pre-treatment based on toxicity results and biodegradability index were implemented in the facilities generating significant improvements in the quality of final effluents with reduction of Biochemical oxygen demand (BOD) and toxicity. The integration of physical and chemical analysis with the V. fischeri toxicity test turned out to be an excellent tool for wastewater management in chemical terminals allowing rapid decision making for pollution control and prevention measures. The mutagenicity of acidic and organic extracts of surface soil under the influence of a coal-fired power plant was evaluated by Salmonella/microsome assay using strains TA97a, TA98 and TA100 in the absence and presence of exogenous metabolic systems (S9 mix). Additionally, strains YG1041 and YG1042 (sensitive to nitroderivatives) were used for the organic extracts. In general, the responses were higher in the organic extracts in the presence of S9 mix. The comparison between strains TA98 and TA100 and their derived strains YG1041 and YG1042, respectively, allowed the detection of the presence of nitro-aromatic compounds in some sampling areas, which was confirmed by chemical analysis. The interpretation of the set of mutagenesis data suggests that there are two important mutagenic compound dispersion routes in the area of study: frameshift mutagens were dispersed predominantly by runoff and leaching, while base-pair substitution mutagens were dispersed mainly by the atmosphere. This mutagenic damage might be attributed to the effects of several substances detected in the area, such as aliphatic hydrocarbons and the metals aluminum, cadmium, lead and iron.

Radioactive lightning rods were manufactured in Brazil until 1989, when the licenses for using radioactive sources in these products were lifted by the national nuclear authority. Since then, these rods have been replaced by the Franklin type and collected as radioactive waste. However, only 20% of the estimated total number of installed rods has been delivered to the Brazilian Nuclear Commission. This situation causes concern, since there is the possibility of the rods to be disposed as domestic waste. In Brazil, 64% of the municipal solid waste is disposed at garbage dumps without sufficient control. In addition, (241)Am, the radionuclide most commonly employed, is classified as a high-toxicity element, when incorporated. In the present study, (241)Am migration experiments were performed by means of a lysimeter system, in order to evaluate the risk of contamination caused by radioactive lightning rods disposed as common solid waste. (241)Am sources removed from lightning rods were placed inside lysimeters filled with organic waste that was collected at the restaurant of the Instituto de Pesquisas Energéticas e Nucleares. The generated leachate was periodically analyzed, and characteristics such as pH, redox potential, solid content and the concentration of the radioactive material were determined. The equivalent dose for members of the public was calculated considering ingestion of contaminated drinking water as the major path of exposure. Estimated doses were about 20-times below the effective dose limit of 1 mSv year(-1) for members of the public as recommended by the International Commission on Radiological Protection. This suggests the radiation risk caused by lightning rods disposed at uncontrolled garbage dumps to be low. It should be noted, however, that the number of investigated lightning rods was quite small. The results of this study might therefore not be entirely representative and should be interpreted with care. They provide, however, a very first basis for characterizing the transfer of (241)Am from lightning rods to the human food chain.

Saplings of Tibouchina pulchra and Psidium guajava, cultivated under standardized soil conditions, were placed in two sites at Cubatão (state of São Paulo, southeast Brazil) to study the effects of air pollution on growth, biomass allocation and foliar nitrogen and fluoride concentrations. Thirty-six potted plants were maintained over two periods of one year (Jul/00 to Jun/01; Dec/00 to Nov/01) at each of two experimental sites with distinct levels of air pollution: Pilões River Valley (PV) with vegetation virtually unaffected by air pollution; and Mogi River Valley (MV) severely affected by pollutants released mainly by chemical, fertilizer, iron and steel industries. For both species, saplings growing at MV showed alterations of growth and biomass allocation, as well as increased leaf concentrations of nitrogen and fluoride. Comparing both experimental periods, the one starting in winter (the driest season in Southeastern Brazil) seemed to affect the saplings more severely, the differences of the measured parameters between MV and PV being higher than in the second period. Multivariate analysis revealed two groups of data: one representing the MV and the other the PV saplings. For both species, saplings growing at MV showed differences in chemical composition, growth and biomass allocation, compared with the PV saplings. The results suggested that seasonal conditions of the first months of sapling exposure (summer or winter) modulate the intensity of responses to pollution stress.

The physico-chemical characteristics and the acute toxicity of several wastewater streams, generated in the industrial production of synthetic rubber, were determined. The acute toxicity was evaluated in bioassays using different organisms: Danio rerio (fish), Lactuca sativa (lettuce) and Brachionus calyciflorus (rotifer). The removal of toxicity attained in the industrial wastewater treatment plant was also determined upstream and downstream of the activated sludge process. The results obtained indicate that the critical streams in terms of acute toxicity are the effluents from the liquid polymer unit and the spent caustic butadiene washing stage. The biological treatment was able to partially remove the toxicity of the industrial wastewater. However, a residual toxicity level persisted in the biotreated wastewater. The results obtained with Lactuca sativa showed a high degree of reproducibility, using root length or germination index as evaluation parameters. The effect of volatile pollutants on the toxicity results obtained with lettuce seeds was assessed, using ethanol as a model compound. Modifications on the assay procedure were proposed. A strong correlation between the toxic responses of Lactuca sativa and Danio rerio was observed for most industrial effluent streams.

To verify whether dyes emitted within the discharge of a dye processing plant were contributing to the mutagenicity repeatedly found in the Cristais River, Sao Paulo, Brazil, we chemically characterized the following mutagenic samples: the treated industrial effluent, raw and treated water, and the sludge produced by a Drinking Water Treatment Plant (DWTP) located approximately 6 km from the industrial discharge. Considering that 20% of the dyes used for coloring activities might be lost to wastewaters and knowing that several dyes have mutagenic activity, we decided to analyze the samples for the presence of dyes. Thin layer chromatographic analysis indicated the presence of three prevalent dyes in all samples, except for the drinking water. This combination of dyes corresponded to a commercial product used by the industry, and it tested positive in the Salmonella assay. The structures of the dye components were determined using proton magnetic resonance and mass spectrometric (MS) methods, and the dyes were tested for mutagenicity. The blue component was identified as the C.I. Disperse Blue 373, the violet as C.I. Disperse Violet 93, and the orange as C.I. Disperse Orange 37. The dyes showed mutagenic responses of 6300, 4600, and 280 revertants/microg for YG1041 with S9 respectively. A bioassay-directed fractionation/chemical analysis showed that the C.I. Disperse Blue 373 contributed 55% of the mutagenic activity of the DWTP sludge. We showed that these dyes contributed to the mutagenic activity found in the Cristais River environmental samples analyzed and are indirectly affecting the quality of the related drinking water. Therefore, we believe that this type of discharge should be more thoroughly characterized chemically and toxicologically. Additionally, human and ecological risks associated with the release of dye processing plant effluents should be more fully investigated, especially where the resultant water is taken for human consumption.

Blood lead was determined in 7 adults, 2 children, 1 bull, 11 cows, and 3 calves living on agricultural land near an industrial waste site. An unexposed human control group was composed of 2 adults and 2 children. Mean blood lead from the exposed group was 4.6 micrograms/dl for adults, 5.1 micrograms/dl for children, 1.3 micrograms/dl for the bull, 1.2 micrograms/dl for cows, and 1.8 micrograms/dl for calves. Unexposed adults had 4.1 micrograms/dl and children had 3.7 micrograms/dl lead blood. The blood lead content did not suggest a health hazard from lead for either humans or animals.

The present study was carried out on the waters of the Caí River (Rio Grande do Sul, Brazil) in an area under the influence of a petrochemical industrial complex, as the continuation of a study in which the mutagenic activity of water samples was evaluated in the internal area of this complex. In the previous study, the release of inducing substances was detected, revealing the need for a full analysis of the real ecological impact of the industrial complex on the river. Water samples from different sites along the Caí River were subjected to the Ames test during a study of 20 months duration for the detection of possible mutagens. Strains TA100 and TA98 were used for initial sample screening in the presence and absence of the S9 mix at a standard dose of 2000 microliters/plate. When positive activity (values equal to twice the spontaneous mutation rate) and/or cytotoxic activity (cell survival below 60%) was detected, the dose-response relationship was studied. Thirty-four percent of the samples tested were mutagenic, with different values according to collection site. Of the total number of positive responses, 6% were obtained for samples collected at the blank site upstream from the area studied, 82% at sites closest to the industrial complex, and 12% in downstream areas. Strain TA98 was the most sensitive in assays with no metabolic activation. A low frequency of induction (2%) was observed for strain TA102. Application of the Ames test permitted the delimitation of three areas of influence of the petrochemical industrial complex, and the test proved to be adequate for the detection of contaminants from the petrochemical industry.
