Geopropolis is a product containing wax, plant resin, and soil particles. It is elaborated by stingless bees of tribe Meliponini. Methanol extracts of sample of geopropolis produced by Scaptotrigona postica (“mandaguari”) in the state of Rio Grande do Norte (RN, northeast Brazil) were analyzed for the determination of standard parameters (total phenols, total flavonoids, and radical scavenging activity) and chemical characterization by HPLC-DAD-MS/MS analysis. The sample analyzed has high contents of total phenols and flavonoids, as well as high antioxidant activity. The constituents characterized were mainly flavonols, such as quercetin methyl ethers, and methoxychalcones. Such chemical profile is similar to the composition of a green propolis from the same area of RN, which is produced by Africanized Apis mellifera, using shoot apices of Mimosa tenuiflora, popularly known as “jurema-preta.” This finding provides evidence that “mandaguari” geopropolis and honeybee propolis have the same botanical origin in RN. The sharing of a plant resin source by phylogenetically distant bees (Apinae and Meliponinae) suggests that bee genetic factors play little role in the choice of plants for resin collection and that the availability of potential botanical sources plays a decisive role. Background: The Amazon is the largest rainforest in the world and is home to a rich biodiversity of medicinal plants. Several of these plants are used by the local population for the treatment of diseases, many of those with probable anti-inflammatory effect. The aim of the present investigation was to evaluate the in vitro antioxidant and anti-peroxidases potential of the ethanol extracts of five plants from the Brazilian Amazon (Byrsonima japurensis, Calycophyllum spruceanum, Maytenus guyanensis, Passiflora nitida and Ptychopetalum olacoides). Methods: DPPH, ABTS, superoxide anion radical, singlet oxygen and the ß-carotene bleaching methods were employed for characterization of free radical scavenging activity. Also, total polyphenols were determined. Antioxidant activities were evaluated using murine fibroblast NIH3T3 cell. Inhibition of HRP and MPO were evaluated using amplex red® as susbtract. Results: The stem bark extracts of C. spruceanum and M. guyanensis provided the highest free radical scavenging activities. C. spruceanum exhibited IC50 = 7.5 ± 0.9, 5.0 ± 0.1, 18.2 ± 3.0 and 92.4 ± 24.8 µg/mL for DPPH*, ABTS+*,O2-* and ¹O2 assays, respectively. P. olacoides and C. spruceanum extracts also inhibited free radicals formation in the cell-based assay. At a concentration of 100 µg/mL, the extracts of C. spruceanum, B. japurensis inhibited horseradish peroxidase by 62 and 50 %, respectively. C. spruceanum, M. guyanensis, B. japurensis also inhibited myeloperoxidase in 72, 67 and 56 %, respectively. Conclusions: This work supports the folk use these species that inhibited peroxidases and exhibited significant free radical scavenging and antioxidant activities what can be related to treatment of inflammation. The purpose of this study was to evaluate the bioactive compounds and antioxidant activity of extracts from aracá ( Psidium cattleianum ), butiá ( Butia eriospatha ), and pitanga ( Eugenia uniflora ) fruits with different flesh colors (i.e., purple, red, and orange), and blackberries ( Rubussp. ;cv.Xavante and Cherokee)collected in the southern region of Brazil.Thecontentof ascorbic acid, total carotenoids, and phenolics were determined. The profile of the phenolic compounds wasassessedbyhigh-performance liquid chromatography combined with diode array detection (HPLC-DAD). The antioxidant activity was determined using the ferric- reducing antioxidant power (FRAP) assay, 2,2-diphenyl-2-picrylhydrazyl hydrate (DPPH) assay, total reactive antioxidant potential(TRAP) assay,and total antioxidantreactivity (TAR) assay. The Xavante blackberry and purple-fleshed pitanga showed the highest total phenolic content [816.50 mg gallic acid equivalents (GAE)/100g and 799.80 mg GAE/100g, respectively]. The aracá and red-fleshed pitanga showed the highest carotenoid content (6.27 ug β -caro- tene/g and 5.86 ug β-carotene/g, respectively). The fruits contained several phenolic com- pounds such as quercetin derivatives, quercitrin, isoquercitrin, and cyanidin derivatives, which may contribute differentially to the antioxidant capacity. The highest scavenging activityintheDPPHassaywasfoundforpurple-fleshedpitanga(IC50 36.78mg/L),blackberries [IC50 44.70 (Xavante) and IC50 78.25 mg/L (Cherokee)], and aracá (IC50 48.05 mg/L), which also showed the highest FRAP, followed by orange- and red-fleshed pitanga. The jaboticaba tree, Plinia trunciflora (O. Berg) Kausel, is popularly named “jabuticabeira” in Brazil and is used in folk medicine to treat diabetes and chronic inflammation of the tonsils, but studies evaluating the central effects of this species are limited. This study evaluated the antidepressant-like and antioxidant effects of P. trunciflora (PT) aqueous extract, in which five different anthocyanins were identified. PT showed significant ferric-reduction power and DPPH radical scavenging activity in vitro and reduced lipid peroxidation both in vitro and ex vivo. At the behavioural level, PT (400 and 800 mg/kg, i.p.) dose-dependently reduced immobility time in the tail suspension test in Swiss male mice. The identification of bioactive compounds accompanied by the in vitro and ex vivo antioxidant activity of PT suggests that these activities might be related to the antidepressant-like activity of P. trunciflora. The main antioxidant compounds (lycopene, β-carotene, total phenolic, and vitamin C) and total antioxidant activity in eight dried tomatoes marketed in Brazil were determined. The total antioxidant activity was determined by the DPPH radical scavenging method and the β-carotene/linoleic acid system in both hydrophilic and hydrophobic extracts. The contents of the antioxidant compounds varied significantly among the products analyzed. The hydrophobic extracts showed higher antioxidant activity with the β-carotene/linoleic acid system while the hydrophilic extracts showed higher antioxidant activity with the DPPH method. Despite differences found, the dried tomatoes can be considered products with high contents of antioxidant compounds with high antioxidant activity. [ABSTRACT FROM PUBLISHER]

Artepillin C (3,5-diprenyl-4-hydroxycinnamic acid) is the main bioactive component of Brazilian green propolis. The aim of this study was to investigate the anti-inflammatory effect of artepillin C on LPS + IFN-γ- or PMA-stimulated RAW264.7 macrophages. The cell viability was evaluated by MTT and LDH assays. The radical scavenging ability was determined using DPPH• and ABTS•+. ROS and RNS generation was analyzed by chemiluminescence. NO concentration was detected by the Griess reaction. The release of various cytokines by activated RAW264.7 cells was measured in the culture supernatants using a multiplex bead array system based on xMAP technology. NF-κB activity was confirmed by the ELISA-based TransAMNF-κB kit. At the tested concentrations, the compound did not decrease the cell viability and did not cause the cytotoxicity. Artepillin C exerted strong antioxidant activity, significantly inhibited the production of ROS, RNS, NO, and cytokine IL-1β, IL-3, IL-4, IL-5, IL-9, IL-12p40, IL-13, IL-17, TNF-α, G-CSF, GM-CSF, MCP-1, MIP-1α, MIP-1β, RANTES, and KC, and markedly blocked NF-κB expression in stimulated RAW264.7 macrophages. Our findings provide new insights for understanding the mechanism involved in the anti-inflammatory effect of artepillin C and support the application of Brazilian green propolis in complementary and alternative medicine. Background: Annona dioica St. Hill (Annonacaeae) is a Brazilian plant used in folk medicine for the treatment of several types of rheumatisms and diarrhoea. The focus of this work was to evaluate the in vitro antiproliferative and antioxidant activity and the in vivo hypoglycaemic and anti-inflammatory activity of A. dioica and identify the principal constituents of this plant. Methods: The crude methanol extract (EAD) and hexane (HF), chloroform (CF), ethyl acetate (EAF) and hydromethanol fractions (HMF) were evaluated for free radical scavenging activity using the DPPH assay. The EAD and EAF were assayed for hypoglycaemic activity in rats. The EAD was tested in an antiproliferation assay and for anti-inflammatory effects in paw oedema, in addition to myeloperoxidase activity induced by carrageenan (Cg) in mice. The EAF was assayed using chromatographic methods. Results: The fractionation of the EAF through chromatographic methods identified derivatives of the flavonoids quercetin and kaempferol. Among all the tested fractions, the ethyl acetate and hydromethanol fractions were the most potent, exhibiting an IC50 of 8.53 and 10.57 μg/mL, respectively, which is comparable to that of the commercial antioxidant butylated hydroxytoluene (BHT). The oral administration of the EAD (100 mg/kg) and EAF (15 mg/kg) inhibited the increase of glucose levels, resulting in a hypoglycaemic effect. The EAD (30 to 300 mg/kg) exhibited an anti-oedematogenic effect in Cg-induced paw oedema in a time- and dose-dependent manner. The results showed a reduction of MPO activity by A. dioica 6 h after the induction of paw oedema at all doses tested with maximal inhibition at 300 mg/kg. Conclusions: Our results reveal for the first time that compounds contained in the A. dioica leaves exert antiinflammatory, hypoglycaemic, antiproliferative, and antioxidant effects. Vegetables are widely consumed in Brazil and exported to several countries. This study was performed to evaluate the phenolic content and antioxidant activity of vegetables commonly consumed in Brazil using five different methods, namely DPPH and ABTS free radical, β-carotene bleaching, reduction of Fe3+ (FRAP), oxidative stability in Rancimat, and the chemical composition using gas chromatography-mass spectrometry (GC-MS). The content of phenolic compounds ranged from 1.2 mg GA/g (carrot) to 16.9 mg GA/g (lettuce). Vegetables presenting the highest antioxidant activity were lettuce (77.2 µmol Trolox/g DPPH·; 447.1 µmol F2+/g FRAP), turmeric (118.6 µmol Trolox/g ABTS·+; 92.8% β-carotene), watercress and broccoli (protective factor 1.29--Rancimat method). Artichoke, spinach, broccoli, and asparagus also showed considerable antioxidant activity. The most frequent phenolic compounds identified by GC-MS were ferulic, caffeic, p-coumaric, 2-dihydroxybenzoic, 2,5-dihydroxybenzoic acids, and quercetin. • The native fruits such as feijoa and cherry are a source of phenolic compounds. • Phenolic compounds of fruits and its hydroethanolic extracts were analyzed by LC–MS/MS. • More than 20 phenolic compounds were identified among the samples analyzed. • Three phenolic compounds were reported for the first time in this study for feijoa. • The extracts were potent scavenger of against hydroxyl, peroxyl and ABTS radical. Acca sellowiana (feijoa) and Eugenia involucrata (cherry) are fruits species of Brazilian biodiversity (Myrtaceae family). In this study, a sampling process was used with three different harvesting sites. The composition of phenolic compounds of these fruits was determined by HPLC–DAD–MS/MS. Moreover, the antioxidant capacity of hydroethanolic extracts against hydrogen peroxide (H 2 O 2), hydroxyl (OH), peroxyl (ROO–) and ABTS radicals was evaluated. Thirty and twenty-seven phenolic compounds were identified in feijoa and cherry, respectively. The major phenolic compounds found were pedunculagin isomer (5040.87, 3443.66 and 1324.95 μg·g−1) in feijoa and procyanidin (1406.54, 1888.00 and 1380.64 μg·g−1) in cherry. Hydroethanolic extract of these fruits was a potent scavenger of free radicals and excellent source of phenolic compounds. In hydroethanolic extracts of feijoa, the phenolic content increased by around 50%, while in the cherry the content was similar to that found in the fruit. For ORAC method, sample 2 of feijoa and cherry showed values of 383 and 126 µM·TE·g−1, respectively, featuring the highest antioxidant capacity. This study is the first to report the identification of castalagin, catechin and epicatechin in feijoa, and rutin in cherry. Arrabidaea chica (Brazilian name = pariri) is a plant species that belongs to the Bignoniaceae family, occurring in tropical America and widespread in the Amazonian region of Brazil. In this study, the phenolic compound and carotenoid profiles of A. chica leaves were determined by HPLC-DAD-MS. Scutellarin was identified as the main phenolic compound (15,147.22 μg/g, dry basis, d.b.) and lutein (204.28 μg/g, d.b.), β-carotene (129.5 μg/g, d.b.) and α-carotene (79.86 μg/g, d.b.) as the major carotenoids. Moreover, A. chica leaves presented 152.7 μg/g of ascorbic acid (fresh weight). For the first time, the carotenoid profile and ascorbic acid contents were reported for A. chica leaves. Regarding the antioxidant capacity, A. chica extract was able to scavenge ABTS radical (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)), and for the first time, the protective effect against singlet oxygen ( 1 O 2 ) was reported. The A. chica extract was efficient as ABTS radical scavenger (86.81 μM Trolox/g of fresh leaves) and inhibited the degradation of tryptophan by 1 O 2 in a concentration-dependent manner with an IC 50 at 177 μg/mL. Species of Eugenia have been used as an antidiabetic natural source. Chemical, antioxidant and antiglycant screening of extracts from pedra-ume caá (Eugenia punicifolia) fruits were performed. 1 H NMR assisted by non-supervised chemometric methods were employed for the evaluation of the chemical profiles which were distinguished according to the color of fruit maturation stages, as well as for pulp and seed fruit. Furthermore, 1 H NMR fingerprint analysis of the crude extract allowed the identification of quercitrin and myricitrin, beside other nine compounds. The extracts of the yellow (YP) and green (GP) pulps presented higher antiglycant and antioxidant activities. Fresh juice from E. punicifolia was encapsulated in microcapsules produced with dextrose equivalent (DE) of 10, 20 or 30 as wall materials for the maintainment of their antioxidant and antiglycant properties. The more efficient retention of the bioactive compounds was found using the DE30. The Encapsulation Efficiency (EE) and the Retention Efficiency (RE) of this system was found around 89.7% and 97.6%, respectively. In addition, NMR spectra revealed the presence of flavonoids O-glycosylated (quercitrin and myricitrin) which might be related to the antiglycant and antioxidant activities. The YP presented larger content of quercitrin (117.6 ± 0.4 mg per each 100 g of fresh fruit).
